Brassinosteroids have been extensively used to overcome various abiotic stresses. But its role in combined stress of salt and excess copper remains unexplored. Seeds of two cultivars (Rocket and Jumbo) of Cucumis sativus were grown in sand amended with copper (100 mg?kg?1), and developed seedlings were exposed to salt stress in the form of NaCl (150 mM) at the 30-day stage of growth for 3 days. These seedlings were subsequently sprayed with 0 or 0.01 μM of 24-epibrassinolide (EBL) at the 35-day stage. The plants exposed to NaCl and Cu in combination exhibited a significant decline in fresh and dry mass of plant, chlorophyll content, activities of carbonic anhydrase, net photosynthetic rate and maximum quantum yield of the PSII primary photochemistry followed by NaCl and Cu stress alone, more severely in Jumbo than in Rocket. However, the follow-up treatment with EBL to the stressed and nonstressed plant improved growth, chlorophyll content, carbonic anhydrase activity and photosynthetic efficiency, and further enhanced the activity of various antioxidant enzymes viz. catalase, peroxidase and superoxide dismutase and content of proline at the 40-day stage of growth, and the response of the hormone was more effective in Rocket than in Jumbo. The elevated level of antioxidant enzymes as well as proline could have conferred tolerance to the NaCl- and/or Cu-stressed plants resulting in improved growth, water relations and photosynthetic attributes. Furthermore, antioxidant enzyme activity and proline content were more enhanced in Rocket than in Jumbo cultivar.  相似文献

为了解海南省昌江县石碌镇钴铜矿尾矿库环境特点,分别采集了矿砂、背景土壤、废水、纳污水体和植物进行分析。结果表明,钴铜矿尾矿库的土壤受As和Cu污染严重,周边土壤受到不同程度的重金属污染;废水经过尾矿库沉淀处理后,SS、As、Cu、Co、Fe等监测指标均有所降低,尾矿库起到沉淀吸附处理作用;纳污河流石碌河水质良好;尾矿库内自然生长的菖蒲和水竹均受到重金属不同程度污染,尾矿库内的甘蔗中Cr、Pb含量均超过食品中污染物限量标准,表明尾矿库内不宜种植可食用作物。  相似文献

Two different types of hydroponic cultures, "water culture" and "perlite system", were compared using white lupin plants (Lupinus albus L., cv. Marta) under different Cd treatments: 0, 0.2, 0.6, 2, 4, 6, 13, 20, 40 and 60 microM (water culture) and 0.2, 2, 20, 60 and 150 microM (moistened perlite). Fresh weight, shoot and root length, and total Cd concentration in the plants were measured. Moreover, a batch experiment was carried out to study the ability of perlite to adsorb and desorb Cd from nutrient solution. Lupin plants under Cd treatments in "water culture" showed a higher growth inhibition than those grown on perlite. A high positive correlation between Cd concentration in the plant and Cd supply was obtained regardless of the substrate used. Moreover, a high positive correlation between Cd doses with the "perlite system" and their equivalent Cd doses estimated for the "water culture" system was observed. Thus, the "water culture-equivalent" Cd doses were 14 times lower than the Cd doses in the perlite system. On the other hand, desorbed Cd concentrations were calculated giving values 12 times lower than the tested Cd doses.  相似文献

A sensitive, accurate and rapid analysis of major nutrients in aquatic systems is essential for monitoring and maintaining healthy aquatic environments. In particular, monitoring ammonium (NH(4)(+)) concentrations is necessary for maintenance of many fish stocks, while accurate monitoring and regulation of ammonium, orthophosphate (PO(4)(3-)), silicate (Si(OH)(4)) and nitrate (NO(3)(-)) concentrations are required for regulating algae production. Monitoring of wastewater streams is also required for many aquaculture, municipal and industrial wastewater facilities to comply with local, state or federal water quality effluent regulations. Traditional methods for quantifying these nutrient concentrations often require laborious techniques or expensive specialized equipment making these analyses difficult. Here we present four alternative microcolorimetric assays that are based on a standard 96-well microplate format and microplate reader that simplify the quantification of each of these nutrients. Each method uses small sample volumes (200 μL), has a detection limit ≤ 1 μM in freshwater and ≤ 2 μM in saltwater, precision of at least 8% and compares favorably with standard analytical procedures. Routine use of these techniques in the laboratory and at an aquaculture facility to monitor nutrient concentrations associated with microalgae growth demonstrates that they are rapid, accurate and highly reproducible among different users. These techniques offer an alternative to standard nutrient analyses and because they are based on the standard 96-well format, they significantly decrease the cost and time of processing while maintaining high precision and sensitivity.  相似文献

Four crop plants Oryza sativa (rice), Solanum melongena (brinjal), Spinacea oleracea (spinach) and Raphanus sativus (radish) were grown to study the impact of secondary treated municipal waste water irrigation. These plants were grown in three plots each of 0.5 ha, and irrigated with secondary treated waste water from a sewage treatment plant. Sludge from the same sewage treatment plant was applied as manure. Cultivated plants were analyzed for accumulation of heavy metals and pesticides. Results revealed the accumulation of six heavy metals cadmium (Cd), chromium (Cr), iron (Fe), copper (Cu), nickel (Ni), and zinc (Zn) as well as two pesticides [1,1-bis(p-chlorophenyl)-2,2,2-trichloroethane; DDT] and benzene hexa chloride (BHC). Order of the plants for the extent of bioaccumulation was S. oleracea > R. sativus > S. melongena > O. sativa. The study has shown the secondary treated waste water can be a source of contamination to the soil and plants.  相似文献

The metal accumulation potential of Bacopa monnieri L. was assessed under simulated laboratory conditions. This study was carried out in mixed metals (Cu, Cd, Pb, Cr, Mn) condition and repeated exposures in artificial contaminated soil. The growing shoots were planted in garden subsoil containing 3, 16, 32, 64, 160 M each of the above metals. After 8 weeks, plants were refeeded to three times higher concentrations of each metal than initially used to assess the maximum accumulating potential of the metals. The accumulation of the metals by the root and shoot was concentration and duration dependent. The metal accumulation was considerably higher in the fine root than in the shoot and showed the following order : Mn > Cr > Cu > Cd > Pb. The plants showed high tolerance to the metals as no visible phytotoxic symptom was produced after 8 weeks. However, as a result of combined metal toxicity, chlorophyll content was reduced by 62% after 12 weeks. The highest metal concentration was lethal to the plant at 16 weeks. In view of their high tolerance, the plants of B. monnieri may be considered for the amelioration of industrially-polluted wetlands experiencing regular flushing of wastewaters.  相似文献

The preparation and application of a practical electrochemical sensor for environmental monitoring and assessment of heavy metal ions in samples is a subject of considerable interest. In this paper, a carbon paste electrode modified with maize tassel for the determination of Cu(II) has been proposed. Scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FTIR) were used to study morphology and identify the functional groups on the modified electrode, respectively. First, Cu(II) was adsorbed on the carbon paste electrode surface at open circuit and voltammetric techniques were used to investigate the electrochemical performances of the sensor. The electrochemical sensor showed an excellent electrocatalytic activity towards Cu(II) at pH 5.0 and by increasing the amount of maize tassel biomass, a maximum response at 1:2.5 (maize tassel:carbon paste; w/w) was obtained. The electrocatalytic redox current of Cu(II) showed a linear response in the range (1.23 μM to 0.4 mM) with the correlation coefficient of 0.9980. The limit of detection and current–concentration sensitivity were calculated to be 0.13 (±0.01) μM and 0.012 (±0.001) μA/μM, respectively. The sensor gave good recovery of Cu(II) in the range from 96.0 to 98.0 % when applied to water samples.  相似文献

The concentrations of Cd, Cu, Pb, and Zn in sediments, water, and different plant organs of six aquatic vascular plant species, Ceratophyllum demersum L. Echinochloa pyramidalis (Lam.) Hitchc. & Chase; Eichhornia crassipes (Mart.) Solms-Laub; Myriophyllum spicatum L.; Phragmites australis (Cav.) Trin. ex Steud; and Typha domingensis (Pers.) Poir. ex Steud, growing naturally in the Nile system (Sohag Governorate), were investigated. The aim was to define which species and which plant organs exhibit the greatest accumulation and evaluate whether these species could be usefully employed in biomonitoring and phytoremediation programs. The recorded metals in water samples were above the standard levels of both US Environmental Protection Agency and Egyptian Environmental Affairs Agency except for Pb. The concentrations of heavy metals in water, sediments, and plants possess the same trend: Zn > Cu > Pb > Cd which reflects the biomonitoring potentialities of the investigated plant species. Generally, the variation of heavy element concentrations in water and sediments in relation to site and season, as assessed by two-way repeated measured ANOVA, was significant (p < 0.05). However, insignificant variations were observed in the concentrations of Pb and Cd in sediments in relation to season and of Cu and Zn in relation to site. Results also showed that the selectivity of the heavy elements for the investigated plants varied significantly (p < 0.05) with species variation. The accumulation capability of the investigated species could be arranged according to this pattern: C. demersum > E. crassipes > M. spicatum > E. pyramidalis > T. domingensis > P. australis. On the basis of the element concentrations, roots of all the studied species contain higher concentrations of Cu and Zn than shoots while leaves usually acquire the highest concentrations of Pb. Cd concentrations among different plant organs are comparable except in M. spicatum where the highest Cd concentrations were recorded in the leaves. Our results also demonstrated that all the studied species can accumulate more than 1,450-fold the concentration of the investigated heavy elements in water rendering them of interest for use in phytoremediation studies of polluted waters. Given the absence of systematic water quality monitoring, heavy elements in plants, rather than sediments, provide a cost-effective means for assessing heavy element accumulation in aquatic systems during plant organ lifespan.  相似文献

Plots of Arabidopsis thaliana L. (Heynh.) plants were grown in two hydroponic systems, one using a solid rooting support media (vermiculite) and the other a flowing solution culture. Both were tested with various concentrations of bromacil (5-bromo-3-sec-butyl-6-methyluracil), dichlobenil (2,6-dichlorobenzonitrile), copper sulfate (CuSO4·5H2O) and 3-aminotriazole. The effects on biomass production were compared. Plants treated with bromacil at concentrations up to 0.05 mg l–1 were similarly reduced in growth in both culture methods. However, the toxic effects of dichlobenil, copper sulfate and 3-aminotriazole were different for both culture methods. Dichlobenil decreased plant growth in the solution culture at lower concentrations than in the vermiculite system. All aspects of growth were suppressed in the solution culture while the vegetative weight was unaffected in the vermiculite culture. Plants treated with 20 mg l–1 copper sulfate in solution culture yielded mean seed weight of 1.2 g/plot compared to 3.1 g/plot when treated with the same concentration in the double-plot system. Plants treated with 0.5 mg l–1 3-aminotriazole in solution culture produced no seed, while plants with the same nominal concentration in the double-pot method produced a mean seed weight of 6.5 g/plot. Solution culture testing was generally more sensitive and since roots were bathed directly in a solution of known pH, nutrient level and chemical concentration, the test conditions were more accurately defined.  相似文献

The applicability of the Phytotoxkit microbiotest for toxicity assessment of sediments in eutrophic freshwater ecosystems was evaluated. Sediments were collected from Turawa dam reservoir (southwestern Poland) which, for years, has been subjected to a marked nutrient enrichment and heavy metal contamination. The test plant species were exposed to whole sediments, solid phases of sediments, and pore waters. Phytotoxicity was estimated on the basis of seed germination and root elongation measurements, combined into an overall germination index (GI). For pore waters, the majority of GI values were not statistically different from the controls, which was consistent with chemical data. For solid phases and whole sediments, GI values showed diversified effects ranging from growth stimulation to growth inhibition. The results obtained vary depending on the plant species and the type of sediment samples. Generally, tests with solid phases of sediments showed phytostimulation, suggesting that higher amount of nutrients adsorbed on organic matter-rich sediments might conceal the inhibitory impact of heavy metals (Cd, Cr, Cu, Mn, Ni, Pb, and Zn). However, this beneficial impact is indicative of a significant nutrient load and, with respect to aquatic ecosystems, its potential resuspention that might accelerate the reservoir eutrophication. Under appropriate conditions heavy metals exerted stronger negative impact on plants. Tests with whole sediments had a higher acidity (pH?5.85) and showed adverse effects, though plant responses vary from inhibition (Lepidium sativum) to stimulation (Sorghum saccharatum). The study demonstrated variability in toxicity of contaminated nutrient-rich sediments as well as effectiveness and usefulness of the Phytotoxkit microbiotest as a practical and reliable tool for evaluation of the hazard of eutrophic ecosystems to higher plants.  相似文献

This study assessed the heavy metal (Cr, Mn, Ni, Cu, Zn, and Pb) uptake and its effect on biochemical parameters in Paspalum distichum, a wetland plant. Sludge collected from Bhalswa waste dump, New Delhi, was used as heavy metal source and dosed in different proportions viz. 20%, 40%, 60%, and 80% to the garden soil. The plants accumulated metals mostly in belowground organs. The metal accumulation followed the order: Cr>Mn>Cu>Zn>Ni>Pb. The range of heavy metal concentration in tissue of belowground organs after 180 days of growth was 1,778.65–4,288.01 ppm Cr, 828.11–1,360 ppm Mn, 236.52–330.07 ppm Ni, 155.79–282.35 ppm Cu, 27.05–91.16 ppm Zn, and 27.09–50.87 ppm Pb. The biochemical parameters viz. chlorophyll and protein contents and peroxidase (POD) activity exhibited no considerable adverse effect indicating the plants’ tolerance towards heavy metals. The high POD activity and synthesis of new protein bands at high sludge-dosed plants were also in support of this fact.  相似文献

Plants of L. leucocephala were grown in 100%soil (as control), 100% fly-ash and fly-ash amendedwith 50% press mud for 80 days, and analysed withrespect to plant growth, elemental composition andphysiological changes in different parts of the plant.The results revealed that amending fly-ash with pressmud enhanced plant growth as well as otherphysiological responses such as chlorophyll, protein,in vivo nitrate reductase activity compared to100% fly-ash treated plants. The elements Fe, Zn, Cuand Mn accumulated in larger quantities in plantsgrown in 100% fly-ash, and followed the order ofaccumulation Fe > Zn > Cu > Mn. Grapevines grown on acid soils with low fertility in southern Brazil are treated with intense foliar applications of copper (Cu) fungicides, resulting in an increased level of Cu in the soil and increased toxicity. The present study evaluated the accumulation and bioavailability of Cu, and soils with varying levels of Cu from the main producing regions of southern Brazil were collected. The forms of Cu present in the soil were assessed using chemical extractants; additionally, oat cultivation was performed, reflecting the use of the plant as an indicator of Cu bioavailability. Cu accumulated in the topsoil, mainly in bioavailable forms, and there was also an increase of Cu up to a depth of 0.4 m. Cu was primarily found in the mineral fraction, with apparent saturation of the soil organic matter functional groups. Inceptisol and Alfisol soils with a long history of cupric fungicide application were found to have levels of Cu toxic to oat plants. Furthermore, accumulated copper in Alfisol soil from the Campanha Gaúcha region of the state of Rio Grande do Sul had higher bioavailability compared to Cu accumulated in Inceptisol soil from the Serra Gaúcha region. In addition, the copper concentration in roots was found to serve as an indicator of Cu bioavailability in soil, but not of copper phytotoxicity in plants.  相似文献

对洪泽湖溧河洼区域的水生植物进行了Cu、Zn、Pb、Cr和Cd等重金属元素的污染调查与监测分析,结果表明:水生植物对重金属元素的吸收与积累反映了环境中的重金属污染水平,不同水生植物对各种重金属元素的吸收富集状况具有相对一致性,即Zn>Cu>Cr>Pb>Cd。水生植物对各种重金属元素的平均富集系数大小顺序为:Cd>Cu>Zn>Cr>Pb,这与各元素迁移性强弱的顺序也是相一致的,Cd、Cu、Zn等各元素较易为植物所吸收,而Pb的移动性较差。大部分水生植物根部的重金属含量比茎叶部分高。研究表明:可以从中筛选出具有高富集作用的植物,作为修复水体或土壤重金属污染的实验植物,为植物修复作用的研究提供参考。  相似文献

Regarding the hypothesis that neonicotinoid insecticides used for seed coating of agricultural crops - mainly corn, sunflower and seed rape - are related to the extensive death of honey bees, the phenomenon of corn seedling guttation has been recently considered as a possible route of exposure of bees to these systemic insecticides. In the present study, guttation drops of corn plants obtained from commercial seeds coated with thiamethoxam, clothianidin, imidacloprid and fipronil have been analyzed by an optimized fast UHPLC-DAD procedure showing excellent detection limits and accuracy, both adequate for the purpose. The young plants grown both in pots - in greenhouse - and in open field from coated seeds, produced guttation solutions containing high levels of the neonicotinoid insecticides (up to 346 mg L(-1) for imidacloprid, 102 mg L(-1) for clothianidin and 146 mg L(-1) for thiamethoxam). These concentration levels may represent lethal doses for bees that use guttation drops as a source of water. The neonicotinoid concentrations in guttation drops progressively decrease during the first 10-15 days after the emergence of the plant from the soil. Otherwise fipronil, which is a non-systemic phenylpyrazole insecticide, was never detected into guttation drops. Current results confirm that the physiological fluids of the corn plant can effectively transfer neonicotinoid insecticides from the seed onto the surface of the leaves, where guttation drops may expose bees and other insects to elevated doses of neurotoxic insecticides.  相似文献

Calendula alata plants were tested for their potential to remove stable cesium and lead from solutions in a 15-day period. The plants were grown hydroponically and placed in solutions containing CsCl and Pb(C?H?O?)? at different concentrations (0.6, 2 and 5 mg l?1). When plants were incubated in CsCl solutions 46.84 ± 2.12%, 41.35 ± 1.59%, and 52.06 ± 1.02% cesium was found to be remediated after 15 days. Moreover, more than 99% lead was removed from the Pb(C?H?O?)? solution in all three concentrations after 15 days during the same period. When both CsCl and Pb(C?H?O?)? were supplemented together in the solution, 9.92 ± 1.22%, 45.56 ± 3.52%, and 46.16 ± 1.48% cesium and 95.30 ± 0.72%, 96.64 ± 0.30%, and 99.02 ± 0.04% lead were removed after 15 days. The present study suggests that hydroponically grown C. alata could be used as a potential candidate plant for phytoremediation of cesium and lead from solutions; however, plants were found to be more efficient for the remediation of lead than cesium.  相似文献

The present experiment was done to evaluate the impact of ambient air pollution on carrot (Dacus carotavar. Pusa Kesar) plants using open top chambers (OTCs) ventilated with ambient (NFCs) or charcoal filtered air (FCs) at a suburban site of Varanasi, India. Various morphological, physiological and biochemical characteristics of the plants were studied at different growth stages. Air monitoring data clearly showed high concentrations of SO2, NO2and O3in the ambient air of study site. SO2and NO2concentrations were higher during early growth stages of carrot, whereas O3concentration was highest during later growth stages. Filtration of air has caused significant reductions in all the three pollutant concentrations in FCs as compared to NFCs.Plants growing in FCs showed significantly higher photosynthetic rate, stomatal conductance, water use efficiency and variable fluorescence as compared to plants growing in NFCs. Protein content also showed a similar pattern, however, lipid peroxidation, ascorbic acid content and peroxidase activity were higher in plants growing in NFCs as compared to FCs. Shoot length, number of leaves per plant, leaf area and root and shoot weight increased significantly upon filtration of ambient air. Total nitrogen decreased significantly in root, but increased significantly in shoot of plants grown in NFCs. Total P, Mg, Ca and K contents decreased significantly in plants grown in NFCs as compared to FCs. The individual pollutant concentrations were below threshold for plant injury, but the combined effect of all the three seems to act synergistically in causing greater adverse impact on dry weight and physiology of carrot plants. The study clearly indicates that air pollutants are high enough in the ambient air to cause significant unfavorable impact on carrot plants. The work further supports the usefulness of OTCs for assessing air pollution damage under field conditions in developing countries.  相似文献

The distribution of 14 elements (both essential and non-essential) in the Hass and Fuerte cultivars of avocados grown at six different sites in KwaZulu-Natal, South Africa, was investigated. Soils from the different sites were concurrently analysed for elemental concentration (both total and exchangeable), pH, organic matter and cation exchange capacity. In both varieties of the fruit, concentrations of the elements Cd, Co, Cr, Pb and Se were extremely low with the other elements being in decreasing order of Mg > Ca > Fe > Al > Zn > Mn > Cu > Ni > As. Nutritionally, avocados were found to be a good dietary source of the micronutrients Cu and Mn. In soil, Pb concentrations indicated enrichment (positive geoaccumuluation indices) but this did not influence uptake of the metal by the plant. Statistical analysis was done to evaluate the impact of soil quality parameters on the nutrient composition of the fruits. This analysis indicated the prevalence of complex metal interactions at the soil–plant interface that influenced their uptake by the plant. However, the plant invariably controlled metal uptake according to metabolic needs as evidenced by their accumulation and exclusion.  相似文献

Field experiments were conducted to study the impact of metal accumulation on malondialdehyde (MDA), cysteine and non-protein thiol (NPSH) contents in the plants of Prosopis juliflora grown on the fly ash (FA) amended with soil, blue green algae (BGA) biofertilizer, farm yard manure, press mud and Rhizobium inoculation. The analysis of data revealed that the level of MDA, cysteine and NPSH was higher in the roots of the plant than leaves, which was found positively correlated with metal accumulation. An increase of 361.14, 64.25 and 305.62% in MDA, cysteine and NPSH contents, respectively was observed after 45 days in the roots of the plants grown in 100% FA as compared to 100% garden soil (GS). The level of MDA, cysteine and NPSH was found less in the plants grown on various amendments of FA showing ameliorating effect on the toxicity induced due to the accumulation of metals. The decrease in MDA, cysteine and NPSH contents was higher in Rhizobium-inoculated plants as compared to uninoculated plants grown on 100% FA. The results showed a high tolerance potential of the plant, which is further increased by inoculating the plant with FA-tolerant Rhizobium showing feasibility of using P. juliflora in environmental monitoring of FA landfills.  相似文献

To assess metal mobility in pruning waste and biosolids compost (pH?6.9 and total concentration of metals in milligram per kilogram of Cd 1.9, Cu 132, Fe 8,513, Mn 192, Pb 81, and Zn 313), shrubs species Atriplex halimus and Rosmarinus officinalis were transplanted in this substrate and irrigated with citric acid (4 g?L?1, pH?2.9) and nutrient solution daily for 60 days. Citric acid significantly increased the concentrations of soluble Mn and Fe in the nutrient substrate solution measured by suction probes, while other metals did not vary in concentration (Cu and Zn) or were not observed at detectable levels (Cd and Pb). In plants, citric acid significantly increased the concentrations of Cu (2.7?±?0.1–3.3?±?0.1 mg?kg?1), Fe (49.2?±?5.2–76.8?±?6.8 mg?kg?1), and Mn (7.2?±?1.1–11.4?±?0.7 mg?kg?1) in leaves of R. officinalis, whereas the concentration of only Mn (25.4?±?0.3–42.2?±?2.9 mg?kg?1) was increased in A. halimus. Increasing Fe and Mn solubility by citric acid addition indicates the possibility of using it to improve plant nutrition. The mobility of metals in this substrate was influenced for the concentration of the metal, the degree of humification of organic matter and its high Fe content.  相似文献
