Gas chromatography coupled to low-resolution mass spectrometry with electron capture negative ionization as detection mode (GC-LRMS (ECNI)) has been compared to gas chromatography coupled to high-resolution mass spectrometry using electron ionization as detection mode (GC-HRMS (EI)) for determination of polybrominated diphenyl ethers (PBDEs) in biological samples. Extracts of 5.0 g plasma, serum and milk samples were analyzed using both methods. The GC-LRMS (ECNI) and GC-HRMS (EI) systems were found to be equally well suited for determination of PBDEs in the biological samples, as well as in standard solutions, with respect to response, detection limits and repeatability at the pg-level. The estimated limits of detection (LOD) in milk extracts ranged from 0.3-0.6 pg PBDE/g milk and 0.4-0.7 pg PBDE/g milk, for the GC-LRMS (ECNI) and GC-HRMS (EI) systems, respectively. The method repeatability including sample preparation was in the range 4.7-8.4% and 0.6-10% relative standard deviation (RSD) for the GC-LRMS (ECNI) and GC-HRMS (EI) systems, respectively.  相似文献

The risks to Indo-Pacific Humpback Dolphins and Finless Porpoises associated with polychlorinated biphenyls (PCBs) were assessed. Stomach contents from twelve stranded Humpback Dolphins and sixteen stranded Finless Porpoises were collected. Concentrations of total and isomer-specific PCBs in the stomach contents were determined using dual-column gas chromatography equipped with electron capture detectors (GC-ECD). Risks due to the PCBs were assessed in three scenarios, based on total PCBs (summation of 41 PCB congeners), total toxicity equivalency (TEQs) and PCB 118, using the toxicity reference values (TRVs) as the threshold effects benchmarks. The calculated risk quotients (RQs) showed that risks due to PCBs were generally low or negligible. Specifically, RQs from total TEQs and total PCBs for Finless Porpoises are below one, suggesting that PCBs should be a low risk for the Finless Porpoise in Hong Kong waters. However, the Humpback Dolphin has RQs larger than 1 for total TEQs and total PCBs when the 95th percentile data were used in the evaluation. This indicates that further investigation may be needed to examine more closely the potential impact of toxic contaminants in the habitat of the Humpback Dolphin.  相似文献

Breast milk samples from 40 first-time mothers from the Pacific Northwest of the US and Canada were analyzed for polybrominated diphenyl ethers (PBDEs) and polychlorinated biphenyls (PCBs). Total PBDEs (summation operator PBDEs), calculated by summing values for the 12 PBDEs congeners analyzed, ranged from 6 to 321 ppb (lipid weight) (mean=96 ppb; median=50 ppb). In approximately 40% of the women (15/40), summation operator PBDEs>100 ppb lw in their milk, and four samples had levels >250 ppb lw. PBDE 47 was the dominant congener in most samples, whereas PBDE 153 was predominant in a few (3/40). summation operator PCBs were calculated by summing values for the 82 PCB congeners analyzed, and ranged from 49 to 415 ppb (lipid weight) (mean=147 ppb; median=126 ppb). approximately 30% of the mothers (13/40) have summation operator PBDEs> summation operator PCBs in their milk samples, and approximately 65% (25/40) have BDE 47>PCB 153 in breast milk samples, with BDE 47 averaging 3-fold greater levels than PCB 153. Clearly, the lower brominated PBDEs are surpassing PCBs as a major environmental concern in North America, and are likely affecting significant portions of the populations in these regions. PBDEs have become a major persistent organic pollutant. However, there are no positive correlations between levels of summation operator PBDEs and summation operator PCBs, or between levels of PBDE 47 and PCB 153, suggesting there may be some differences in exposure pathways for PBDEs and PCBs in humans.  相似文献

The efficiency of supercritical fluid extraction for the determination of 12 polychlorinated biphenyls from algae samples is compared to Soxhlet extraction. Analytical detection limits for the individual congeners ranged from 0.62 microgl(-1) to 19 microgl(-1). Recovery was tested for both methods using standard addition procedure. At maximum spike level of concentration, the mean recoveries were not significantly different (P>0.05) of all PCBs studied, with the exception of PCBs 28, 52, 77 and 169. Method precision for Soxhlet extraction (< or =3.9%) was slightly better than for SFE (< or =9.2%). Although both methods yield comparable results, SFE offers the advantage of detecting all PCBs studied at lower concentrations, reducing extraction time, and reducing the amount of solvents needed. The optimized methods were applied to the analysis of three real seaweed samples, except for PCB101 the concentrations of all PCBs were low or below the detection limits. The levels of PCB101 found in sample 1 were 6.6+/-0.54 ng g(-1) d.w., in sample 2 the levels were 8.2+/-0.86 ng g(-1) d.w. and in sample 3 they were 7.7+/-0.08 ng g(-1) d.w.  相似文献

Since the CALUX (Chemically Activated LUciferase gene eXpression) bioassay is a fast, sensitive and inexpensive tool for the analysis of a high number of samples, validation of new methods is urgently needed. In this study, a new method for the analysis of PCDD/Fs and dioxin-like PCBs in atmospheric deposition samples with the CALUX bioassay was developed, optimized and validated. The method consists of 4 steps: filtration, extraction, clean up and bioassay analysis. To avoid the use of large amounts of toxic solvents, new techniques were used for filtration and extraction: a C18 filter was used instead of a liquid/liquid extraction and an Accelerated Solvent Extractor (ASE) was used instead of the traditional soxhlet extraction. After pre-oxidation of the sample extract, clean up was done using a multi-layer silica gel column coupled to a carbon column. The PCDD/F and PCB fractions were finally analyzed with the H1L7.5c1 and/or the H1L6.1c3 mouse hepatoma cell lines. The limit of quantification was 1.4 pg CALUX-BEQ m−2 d−1 for the PCBs and 5.6 pg CALUX-BEQ m−2 d−1 for the PCDD/Fs, when using the new sensitive H1L7.5c1 cell line. The GC-HRMS recovery for all PCDD/F congeners was between 55% and 112%, with a mean recovery of 90%. CALUX recoveries of spiked procedural blanks were between the accepted ranges of 80-120%. Repeatability and reproducibility were satisfactory and no interferences from metals were detected. The first results from the Flemish measurement program showed good correlation between CALUX and GC-HRMS.  相似文献

A solid phase extraction (SPE) method using pyrenebutyric acid-bonded silica (PYB) as sorbent was developed to determine 23 polychlorinated biphenyls (PCBs) in sewage water by gas chromatography-mass spectrometry (GC-MS). Factors were optimized in SPE procedures including elution solvent, pH, and cartridge burden. The recoveries of 23 PCB congeners were 69.44-111.91% under optimized conditions. Comparisons were also conducted among PYB-SPE, C(18)-SPE and United States Environmental Protection Agency 608 (USEPA608) methods in the analysis of PCBs in sewage water samples. The results showed that the performance of PYB-SPE method was similar with USEPA608 method and better than C(18)-SPE method. Both PYB-SPE and USEPA608 methods were then employed to analyze PCBs in real spiked sewage water samples. The recoveries of PCB congeners determined by PYB-SPE method ranged from 70.6% to 92.4% in real spiked sewage water samples which were identified to be in accordance with USEPA608 method. Limits of detection (LOD) were in the range of 0.06-0.22ngL(-1) for PCB congeners. The optimized PYB-SPE method was successfully applied to the determination of PCBs in sewage water samples.  相似文献

The disposal and dumping of toxic waste is a matter of growing concern in developing countries, including South Africa. Frequently these countries do not possess access to gas chromatography-high resolution mass spectrometry (GC-HRMS) for the determination of persistent organic pollutants (POPs). This publication describes an alternative approach to the investigation of toxic waste using comprehensive gas chromatography coupled to time of flight mass spectrometry (GC × GC-TOFMS). The technology permits both comprehensive screening of toxic samples for numerous classes of organic pollutants and also quantitative analysis for the individual compounds. This paper describes the use of this technique by analysing samples obtained from a hazardous waste treatment facility in South Africa. After sampling and extraction the samples were analysed for polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs) and four dioxin-like non-ortho substituted polychlorinated biphenyls (PCBs). The quantitative values, as well as detection limits, obtained using the GC × GC-TOFMS methodology compares well with those obtained using GC-HRMS; the accepted benchmark technology for this analysis. Although GC × GC-TOFMS is not a target compound analytical technique (as is GC-HRMS), it is possible to obtain information on numerous other classes of organic pollutants present in the samples in one analytical run. This is not possible with GC-HRMS. Several different column combinations have been investigated for handling very complex waste samples and suggestions are presented for the most suitable combination.  相似文献

Persistent organic pollutants (POPs), organochlorine pesticides and polychlorinated biphenyls (PCBs), listed as per the Stockholm Convention (alpha -HCH, beta -HCH, gamma -HCH, p,p'-DDT, o,p'-DDT, p,p'-DDD, p,p'-DDE, aldrin, endrin, dieldrin, PCBs 28, 52, 118, 138, 153, and 180), were analyzed in municipal solid waste (MSW) compost samples from three different Brazilian composting plants located in three S?o Paulo State cities: Araras, Araraquara and S?o Paulo (Vila Leopoldinha). Quantitative and qualitative analyses were carried out using gas chromatography electron capture detection (GC-ECD) and gas chromatography mass spectrometry (GC-MS) (Ion Trap, electron impact ionization), respectively. The samples were analyzed in triplicate and the target POPs were not detected by GC-ECD. Twelve pollutants were identified in two samples when qualitative analysis (GC-MS) was used (beta -HCH, gamma -HCH, p,p'-DDT, o,p'-DDT, p,p'-DDD, and p,p'-DDE, PCBs 28, 118, 138, 153 and 180). The composting process has advantages such as urban solid waste reduction and landfill life-span increase, however the MSW compost quality, which can be utilized for agricultural purposes, should be evaluated and be controlled. This kind of study is the first step in making available information to answer questions regarding MSW compost for sustainable agricultural use, such as the pollutants accumulation in soil and in groundwater, and plants uptake.  相似文献

A method based on gas chromatography electron capture detection (GC-ECD) for the analysis of chlorinated paraffins (CPs) in biological samples has been investigated. The method includes photolytic destruction of halogenated aromatic compounds, such as PCBs, to eliminate some of the interferences in the analysis of CPs in environmental samples. Gel permeation chromatography was used to isolate CPs from the interfering components of Toxaphene and chlordane after the photolysis. GC-ECD gave a detection limit of 20 ng CPs/g fresh muscle tissue. The recovery of CPs from a spiked moose liver sample was estimated to 94%.  相似文献

Swine sewage could be source of nutrients and pollutants. This work estimates the environmental risk in nine samples from different farm treatment systems based on the evaluation of their effects in Daphnia magna acute test, and on the assessment of Cu, Zn and ammonia as main contributors. NH3 and Cu were responsible for LC50 results (1-5% of dilution). Organic compounds were quantified through several extraction methods (SPMDs, SPE and solvent extraction). A more exhaustive extraction was performed in an additional sample, which showed indole and phenol recoveries much higher than the previous ones. This method also includes PCBs (430 ppb) and fatty acids (approximately =150 ppm) quantification.  相似文献

Polychlorinated naphthalenes (PCNs), short-chain chlorinated paraffins (SCCPs) and polychlorinated biphenyls (PCBs) were analysed in marine sediment samples collected from the coastal area of Barcelona (Spain) and near of a submarine emissary coming from a waste water treatment plant located at the mouth of the Besòs River (Barcelona). An integrated sample treatment based on Soxhlet extraction followed by a simple clean-up with Florisil and graphitized carbon cartridge was employed. Gas chromatography coupled to ion-trap tandem mass spectrometry (GC-MS/MS) and gas chromatography-mass spectrometry in electron capture negative ionization mode, were used for PCN and SCCP determinations, respectively, while for PCB analysis gas chromatography with electron capture detection (GC-ECD) was used. The method developed provided low limits of detection (0.001-0.003 ng g(-1) dry weight (dw) for PCNs, 1.8 ng g(-1) for SCCPs and 0.006-0.014 ng g(-1)dw for PCBs) and good run-to-run precisions (lower than RSD 8%) for the analysis of sediment samples. Concentration levels ranging from 0.17 to 3.27 ng g(-1)dw for PCNs, between 0.21 and 1.17 microg g(-1)dw for SCCPs, and from 2.33 to 44.00 ng g(-1) (dw) for PCBs, were found in the coastal sediments, while for samples collected near to the submarine emissary higher levels (from 2.02 to 6.56 ng g(-1)dw for PCNs, between 1.25 and 2.09 microg g(-1)dw for SCCPs and from 22.34 to 37.74 ng g(-1)dw for PCBs) were obtained. The results obtained provide new data about the occurrence of PCN and SCCP in the coastal area of Barcelona.  相似文献

Concentrations of polychlorinated biphenyls (PCBs) were determined in wild mussels (Mytilus galloprovincialis) from seven areas of the N-NW Spanish coast, during the period 1991-2009. The studied area is comprised of highly productive ecosystems which support important commercial fishing and shellfishing activities. The seven PCBs indicator congeners recommended by ICES (IUPAC nos. 28, 52, 101, 118, 138, 153 and 180), were analysed by gas chromatography with an electron capture detector (GC-ECD). Lowest average concentrations of PCBs were recorded in Arousa (1.04 μg kg(-1) wet weight), a low populated area with no industry; while the highest levels were found in areas near cities and/or showing a high industrial activity such as: Bilbao Ziérbena (139 μg kg(-1) ww) or A Coru?a (55 μg kg(-1) ww). The hexachlorinated congeners CB153 and CB138 dominated the profiles in all wild mussel populations, although a geographical pattern was found in which the relative contributions of the lower chlorinated congeners were higher in the west coast (Rías of Vigo, Pontevedra and Arousa), which may reflect the occurrence of recent inputs of those compounds in the area. Finally, the application of the Mann-Kendall's test demonstrate the existence of a significant decrease in PCBs levels since 1991 up to 1996 in the studied area, whereas either a slow decrease or stable concentrations were registered over the past decade.  相似文献

This study was designed to investigate the transportation, distribution, and bioaccumulation of PCBs in various environmental media and compartments using an ecologically simulated growth chamber. Spatial and temporal trends of PCBs in the growth chamber were discussed. The release of PCB congeners in soil was affected by the amount of rainfall with the transporting direction moving away from PCBs contaminated point. Two pathways of PCBs accumulation in plants were the uptake of roots and the deposition on shoots/leaves. There were 29 PCB congeners been found in the lalang grass. Higher concentrations of lower chlorinated PCBs were identified than higher chlorinated PCBs because of relatively higher vapor pressure for lower chlorinated congeners. After 10months of monitoring, PCBs were detected in water samples which were contributed by the release of PCBs from leached soil. Analysis of sediment showed that the percentages of low- and mid-chlorinated biphenyls were decreased 1% and 13%, respectively compared to the increase (14%) of high-chlorinated biphenyls. The increase of high-chlorinated PCBs was possibly caused by their low hydrophilicity which had resulted higher adsorption rate in sediment. All of five species of fish had been found significant amount of PCBs accumulation ( summation operatorPCBs: 21.7-102.1 microg/g-lipid). The concentrations of PCBs in fish were varied significantly among species. The range of bioaccumulation factors (BAFs) among different species of fish could be as much as 5 times depending on the consumption habits of fish. The mass balance of PCBs distribution in the growth chamber was also discussed.  相似文献

Determination of polychlorobiphenyls in Venice Lagoon sediments   总被引:1,自引:0,他引:1  
Polychlorinated biphenyl (PCBs) congeners were measured in 14 sediment samples collected at five sites in the Venice Lagoon selected on the basis of a supposed diversity of inputs and of pollution levels. The PCBs were determined after a sonication assisted extraction by gas chromatography mass spectrometry (GCMS). The sum of the congener concentrations determined in the samples range from 4.05 to 239.15 ng/g on a dry weight basis. The concentration values are very variable both comparing the sites and comparing different samples from the same site and can be connected to the sand percentage in the sediment samples. The correlation matrix obtained from data matrix using the congener concentrations as variables shows high values, mostly between 0.964 and 1, suggesting that the pattern of the PCB congeners and the ratios among the concentrations in Venice Lagoon sediments are more or less constant; the samples differ only in the concentration level.  相似文献

Polychlorinated biphenyls (PCBs) were determined in a aquatic community from Los Padres Lake, Argentina. Twenty four PCB congeners from tri- to octa-chlorinated isomers were detected and quantified using conventional gas chromatography with electron capture detector (GC-ECD). The aim of this study was to investigate the concentrations of PCBs in freshwater organisms from a shallow lake of Argentina. Stems of bulrush (Schoenoplectus californicus), whole tissues of false loosestrife (Ludwigia sp.) and grass shrimp (Palaemonetes argentinus), and liver, gonads, muscle and mesenteric fat (if present) of fish species (Rhamdia sapo) and (Oligosarcus jenynsi) were analyzed. Two areas were selected to macrophytes sampling: the input area, main PCB source of the lake (Station 1), and the output area, a potential anoxic zone (Station 2). Macrophytes from Station 1 bioconcentrated higher total PCB levels than Station 2, showing that the former have received PCBs washed down from upstream areas. Penta- and hexa-congeners were enriched relative to other congeners in animal biota and macrophytes from Station 1, consistent with commercial mixture of Aroclor 1254 used in this region. In bulrush from Station 2 a predominance of tri- and tetra-chlorinated congeners was observed. Grass shrimp showed the lowest PCB values among animal biota. PCB concentrations in fish tissues varied with the species and the gonadal development. Mesenteric fat, only present in post-spawning organisms of R. sapo, had the highest values of PCBs relative to other tissues. A clearance of total PCBs in ovaries of post-spawning females of R. sapo was observed, but not in testes. O. jenynsi/P. argentinus biomagnification factor (BMF) had a mean value of 18.7. Congeners 44, 52 and 151, showed the highest BMF values, being 64, 66 and 62, respectively. These values would be a consequence of the low depuration rate of 44 and 52 congeners with orthochlorine substitution conducted by O. jenynsi and the high depuration rate of congener 151, which lacks 4 4- chlorine substitution, carried out by grass shrimp. Although the most of congeners have been biomagnified, they did not clearly displayed a concomitantly increasing with log Kow.  相似文献

MDGC-ECD procedures have been used to provide insight into the compositional complexity of some of the specific peaks or clusters observed in the gas chromatographic analysis of a technical toxaphene standard, with reference to individual toxaphene congeners (Parlar # components) that are flow commercially available. These investigations have focussed initially upon those peaks and clusters recently identified (Shoeib. M., Brice, K.A., Hoff, R., 1999. Chemosphere 39, 849-871) as dominant constituents of background ambient air. Multiple electron-capturing components have been found to be present in all the species studied: the available individual toxaphene congeners have been matched against these components where possible. In similar fashion, the responses obtained in equivalent gas chromatographic elution windows from the analysis of typical processed air sample extracts have been investigated, with the results showing clear differences relative to the patterns found in the technical toxaphene standard. In most cases, the air sample shows reduced complexity with fewer components present in the cluster. Also, the presence of interfering responses (due to PCBs and other organochlorines) is quite apparent and significant, showing that major problems and errors could arise when using single-column GC-ECD procedures for quantitation of toxaphene in environmental samples. The presence of certain of the Parlar species in the air samples has been confirmed and in most cases these represent the dominant toxaphene component found in the targeted cluster. Furthermore, the persistence of certain congeners in the atmospheric samples appears to be strongly dependent upon chemical structure, since the congeners in question possess an alternating exo-endo chlorine substitution pattern around the six-membered ring in the bornane skeleton. Such persistence is probably the result of lower metabolization of toxaphene residues in soils, water and sediments leading to a similar pattern in the atmosphere following volatilization.  相似文献

In soils of the Moscow region, we examined PCB concentrations in bulk samples and aggregate fractions. Topsoils under grassland and forest at five locations along a southeast-bound transect from Moscow and at a northeastern background location (grassland only) were analysed. We collected aggregates >1 cm and fractionated them into interior and exterior portions and also analysed the remaining soil without the aggregates >1 cm. The concentrations of the sum of 17 PCBs (sigma17PCBs) in 35 bulk soil samples ranged from 3.1 to 42 microg kg(-1). This was 48-61% of the sigma33PCBs determined in 23 selected samples. The congeners 138(+158), 101 and 52 were most abundant. All PCB concentrations and the degree of chlorination declined with increasing distance from Moscow. The PCBs were accumulated in the aggregate exterior (on average 146% of the sigma17PCBs in bulk soil). We conclude that the ecotoxicological risk of PCBs in soils may not be properly assessed with the conventional bulk soil analysis.  相似文献

A laboratory experiment was carried out to measure volatilisation fluxes of polychlorinated biphenyls (PCBs) from sewage sludge-amended soils. The most commonly practised methods of applying sludge to agricultural land in the UK, namely, surface application, ploughing in to soil and subsurface injection, were simulated inside glass experimental chambers using an anaerobically digested sludge and a sandy loam soil. Humidified air was blown over the surface of the soil/sludge in the chambers for a period of 32 days, in order to sample a sufficient air volume to detect the volatilising PCBs. The resulting PCB volatilisation fluxes from the different sludge application methods were quantified and compared. Volatilisation fluxes of individual congeners were generally highest for the surface sludge (1-cm depth) application and slightly lower for the plough layer (5-cm depth) application. Fluxes from the subsurface layer of sludge (5-cm depth) were only quantified for the lightest congeners near to the end of the experimental run-time. Results from a multiple regression analysis showed that volatilisation fluxes of PCBs from the surface application are highly dependent on both the sludge concentration and the log of the octanol-air partition coefficient (K(OA)). A well-known soil volatilisation model, developed by Jury et al., was adapted and used to predict fluxes for the different sludge application methods during the experiment. The model predicted volatilisation fluxes that were reasonably comparable to measured fluxes for some congeners, but for others predicted fluxes that were more than an order of magnitude lower than measured fluxes. The model predicted similar loss kinetics to those observed in the experiment. Possible reasons for the dissimilarity between measured and predicted fluxes include inaccuracies in model input parameters and the fact that the models were not developed for predicting fluxes from sludge-amended soils.  相似文献

A novel and simple method based on polymer monolith microextraction (PMME) coupled to gas chromatography with electron-capture detection (GC-ECD) was developed for the determination of six polychlorinated biphenyls (PCBs) residues in water samples. The proposed method used poly-(methyl methacrylate-co-ethylene dimethacrylate) (MMA-co-EDMA) monolith as extraction media. Several factors affecting experiments such as sample flow rate, sample volume, the type of eluent, eluent volume, eluent flow rate, effect of salt addition and carry over effect were investigated and optimized systematically. The limits of detection (LODs) for six PCBs were 0.028-0.043 ng mL−1 in water samples. The intra-day and inter-day precisions (R.S.D.) were less than 9.2% and 9.6%, respectively. The proposed method was successfully applied to the determination of six PCBs in tap water, lake water and industrial waste water and the trueness has been evaluated by recovery experiments. The obtained relative recoveries were in the range of 63.3-105.6%.  相似文献

Organochlorine pesticides and polychlorinated biphenyls (PCBs) were analysed in extracts of human adipose tissue. The samples consisted of abdominal, mammary, and perirenal fat tissue of 27 Finnish adult males and females. Lipids were separated from the low-molecular compounds by preparative gel permeation chromatography (GPC) and the extracts were further cleaned-up using Florisil chromatography. The compounds were analysed in the extracts by gas chromatography (GC) using electron capture detection and by GC-mass spectrometry in the selected ion monitoring mode. Of the 23 analysed pesticide residues only seven could be detected in the extracts. All the extracts contained the DDT metabolite 4,4'-DDE, hexachlorobenzene, and PCBs. Other compounds found in the extracts were 4,4'-DDT, 4,4'-DDD, pentachlorobenzene, and beta- and gamma-hexachlorocyclohexane (HCH). The antiandrogenic 4,4'-DDE was the most abundant pesticide with concentration levels ranging from 3.5 to 3229 ng/g lipids (ppb). The mean concentration of DDE was 567 ppb, of pesticides in all 1008 ppb and of PCBs 504 ppb. Several of the identified compounds have been shown to exhibit endocrine disrupting effects. Statistical analysis showed a positive age correlation of DDE in females and hexachlorobenzene in males. No statistically significant differences were found for concentrations of individual compounds between sexes.  相似文献
