Introduction

Textile industry is one of the most common and essential sectors in Tunisia. However, the treatment of textile effluents becomes a university because of their toxic impacts on waters, soils, flora, and fauna.

Materials and methods

The aim of this work was to evaluate the ability of Pseudomonas putida mt-2 to decolorize a textile wastewater and to compare the biologic decolorization process to the chemical one currently used by the textile industry.

Results

P. putida exhibited a high decolorizing capacity of the studied effluent, compared to the coagulation?Cflocculation method with decolorization percentage of 86% and 34.5%, respectively. Genotoxicity of the studied effluent, before and after decolorization by P. putida mt-2, was evaluated in vitro, using the SOS chromotest, and in vivo, in mouse bone marrow, by assessing the percentage of cells bearing different chromosome aberrations compared to not treated mice. In addition, textile effluent statistically significant influenced acetylcholinesterase and butyrylcholinesterase activities and lipid peroxidation (p?P. putida is a promising and improved alternative to treating industrial scale effluent compared to current chemical decolorization procedures used by the Tunisian textile industry.  相似文献

A new bioassay proposed in the patent P201300029 was applied to a pre-treated wastewater containing a mixture of commercial pesticides to simulate a recalcitrant industrial wastewater in order to determine its biodegradability. The test uses a mixture of standardized inoculum of the lyophilized bacteria Pseudomonas putida with the proper proportion of salts and minerals. The results highlight that biodegradation efficiency can be calculated using a gross parameter (chemical oxygen demand (COD)) which facilitates the biodegradability determination for routine water biodegradability analysis. The same trend was observed throughout the assay with the dehydrated and fresh inoculums, and only a difference of 5 % in biodegradation efficiency (E f) was observed. The obtained results showed that the P. putida biodegradability assay can be used as a commercial test with a lyophilized inoculum in order to monitor the ready biodegradability of an organic pollutant or a WWTP influent. Moreover, a combination of the BOD5/COD ratio and the P. putida biodegradability test is an attractive alternative in order to evaluate the biodegradability enhancement in water pre-treated with advanced oxidation processes (AOPs).  相似文献

The increasing usage and the persistence of polyester polyurethane (PU) generate significant sources of environmental pollution. The effective and environmental friendly bioremediation techniques for this refractory waste are in high demand. In this study, three novel PU degrading bacteria were isolated from farm soils and activated sludge. Based upon 16S ribosomal RNA gene sequence blast, their identities were determined. Particularly robust activity was observed in Pseudomonas putida; it spent 4 days to degrade 92 % of Impranil DLNTM for supporting its growth. The optimum temperature and pH for DLN removal by P. putida were 25 °C and 8.4, respectively. The degradation and transformation of DLN investigated by Fourier transformed infrared spectroscopy show the decrease in ester functional group and the emergence of amide group. The polyurethanolytic activities were both presented in the extracellular fraction and in the cytosol. Esterase activity was detected in the cell lysate. A 45-kDa protein bearing polyurethanolytic activity was also detected in the extracellular medium. This study presented high PU degrading activity of P. putida and demonstrated its responsible enzymes during the PU degradation process, which could be applied in the bioremediation and management of plastic wastes.  相似文献

Arsenic (As) removal through microbially driven biovolatilization can be explored as a potential method for As bioremediation. However, its effectiveness needs to be improved. Biostimulation with organic matter amendment and bioaugmentation with the inoculation of genetic engineered bacteria could be potential strategies for As removal and site remediation. Here, the experiments were conducted to evaluate the impacts of rice straw and biochar amendment, inoculation of genetic engineered Pseudomonas putida KT2440 (GE P. putida) with high As volatilization activity, on microbial mediated As volatilization and removal from three different arseniferous soils. In general, the addition of rice straw (5%) significantly enhanced As methylation and volatilization in comparison with corresponding non-amended soils. Biochar amendments and inoculation of the GE P. putida increased As methylation and volatilization, respectively, but less than that of rice straw addition. The effectiveness of As volatilizations are quite different in the various paddy soils. The combined amendments of rice straw and GE P. putida exhibited the highest As removal efficiency (483.2 μg/kg/year) in Dayu soil, with 1.2% volatilization of the total As annually. The highest water-soluble As concentration (0.73 mg/kg) in this soil could be responsible for highest As volatilization besides the rice straw and bacteria in this soil.  相似文献

The textile industry is a favor to the Tunisian economy by offering several job positions. However, it’s not environmentally friendly. In fact, textile industries discharge high volumes of wastewater which contain several toxic pollutants such as dyes, fixator, and whiteness. In our study, Pseudomonas peli, isolated and characterized from Oued Hamdoun (center of Tunisia), was found able to decolorize textile effluent about 81 % after 24 h shaking incubation. On the other hand, the in vitro antiproliferative effects of the untreated and treated effluent was evaluated by their potential cytotoxic activity using the MTT colorimetric method against three human cancer cell lines (A549, lung cell carcinoma; HT29, colon adenocarcinoma; and MCF7, breast adenocarcinoma). Results showed that intact textile effluent and its content azo dyes didn’t inhibit the proliferation of all tested cell lines. However, the cytotoxic effect was remarkable when we tested effluent obtained after treatment by P. peli in a dose-dependent manner. This activity was attributed to the presence, in our treated effluent, of some azo products of dyes which are responsible for inhibition of human cell lines proliferation. Thus, the use of this strain for testing on the industrial scale seems impossible and disadvantageous.  相似文献

In this study we evaluated genotoxicity and cytotoxicity of native samples of wastewaters (15 samples), surface waters (28 samples) and potable waters (8 samples) with the SOS/umuC assay with Salmonella typhimurium TA1535/pSK1002 and MTT assay with human hepatoma HepG2 cells. The genotoxicity of selected samples was confirmed with the comet assay with HepG2 cells. In the SOS/umuC assay 13 out of the 51 samples were genotoxic: two effluent samples from chemical industry; one sample of wastewater treatment plant effluent; two hospital wastewater samples; three river water samples and four lake water samples. Six samples were cytotoxic for HepG2 cells: both effluent samples of chemical industry, two wastewater treatment plant effluent samples, and two river water samples, however, only the chemical industry effluent samples were genotoxic and cytotoxic, indicating that different contaminants are responsible for genotoxic and toxic effects. Comparing genotoxicity of river and lake water samples with the chemical analytical data of the presence of the residues of pharmaceutical and personal care products (non-steroidal anti-inflammatory drugs, UV filters and disinfectants) in these samples, indicated that the presence of UV filters might be linked to the genotoxicity of these samples. The results showed that the application of the bacterial SOS/umuC assay and mammalian cell assays (MTT and comet assay) with HepG2 cells was suitably sensitive combination of assays to monitor genotoxicity and cytotoxicity of native samples of wastewaters and surface waters. With this study we also confirmed that the toxicity/genotoxicity bioassays should be an integral tool in the evaluation of toxicity of complex wastewaters before the release into environment, as well as for the monitoring of surface water quality, providing data useful in risk assessment.  相似文献

Organic pollutants present in the soil of a microcosm containing pulp and paper mill black liquor were extracted with hexane/acetone (1:1 v/v) to study the biodegradation and detoxification potential of a Bacillus sp. gas chromatography-mass spectroscopic (GC-MS) analysis performed after biodegradation showed formation of simpler compounds like p-hydroxyhydrocinnamic acid (retention time [RT] 19.3 min), homovanillic acid methyl ester (RT 21.6 min) and 3,5-dimethoxy-p-coumaric alcohol (RT 24.7 min). The methyltetrazolium (MTT) assay for cytotoxicity, 7-ethoxyresorufin-O-deethylase (EROD) assay for dioxin-like behavior and alkaline comet assay for genotoxicity were carried out in the human hepatocarcinoma cell line HuH-7 before and after bacterial treatment. Bioremediation for 15 days reduced toxicity, as shown by a 139-fold increase in black liquor’s LC50 value, a 343-fold reduction in benzo(a)pyrene equivalent value and a 5-fold reduction in olive tail moment. The EROD assay positively correlated with both the MTT and comet assays in post biodegradation toxicity evaluation.  相似文献

The genotoxic effects of oxidative metabolites of trichloroethylene (TCE), namely chloral hydrate, trichloroacetic acid (TCA), dichloroacetic acid (DCA), and trichloroethanol (TCEOH) were examined in human peripheral blood lymphocytes. In this context, lymphocytes were exposed in vitro to 25, 50, and 100 μg/ml concentrations of these metabolites separately for a period of 48 h and examined for micronucleus (MN) induction through flow cytometer. At 50 μg/ml TCE metabolites, TCA (6.33?±?0.56 %), DCA (5.06?±?0.55), and TCEOH (4.70?±?1.73) induced highly significant (p?<?0.001) frequency of MN in comparison to control (1.03?±?0.40) suggestive of their genotoxic potential. However, exposure of 100 μg/ml of all the metabolites consistently declined the frequencies of MN which in some cases was equable to that of observed at 25 μg/ml. Further, cytotoxicity and cell cycle disturbances were also measured to find out the association of these endpoints with the MN induction. DNA content analysis revealed 3–4-fold elevation of S-phase at all the concentrations tested. Particularly, at 100 μg/ml, treatment elevation of S-phase was significantly (p?<?0.0001) higher as compared to the control. Present findings together with earlier reports indicate that TCE induces genotoxicity through its metabolites. Interaction of these metabolites with DNA, as evident by elevated S-phase, seems to be the major cause of MN induction. However, involvement of spindle disruption cannot be ruled out. This comparative study also suggests that after TCE exposure, the metabolic efficiency of human to generate oxidative metabolites determines the extent of genotoxicity.  相似文献

The genotoxicity of industrial wastewaters from Jajmau (Kanpur), was carried out by Ames Salmonella/microsome test, DNA repair-defective mutants, and Allium cepa anaphase–telophase test. Test samples showed maximum response with TA98 strain with and without metabolic activation. Amberlite resins concentrated wastewater samples were found to be more mutagenic as compared to those of liquid–liquid extracts (hexane and dichloromethane extracts). The damage in the DNA repair defective mutants in the presence of Amberlite resins concentrated water samples were found to be higher to that of liquid–liquid-extracted water samples at the dose level of 20 μl/ml culture. Among all the mutants, polA exhibited maximum decline with test samples. Mitotic index (MI) of root tip meristematic cells of A. cepa treated with 5, 10, 25, 50, and 100 % (v/v) wastewaters were significantly lower than the control. Complementary to the lower levels of MI, the wastewaters showed higher chromosomal aberration levels in all cases investigated.  相似文献

A mixture of urban and hospital effluents (50 % v/v) was evaluated for ecotoxicity with an advanced bioassay battery. Mixed effluents were tested before any treatment, after biological treatment alone, and after biological treatment followed by a tertiary ozonation (15 mg O3/L). Laying a high value on the continuance of organisms’ fitness, essential to preserve a healthy receiving ecosystem, the main objective of this study was to combine normalized bioassays with newly developed in vivo and in vitro tests in order to assess alteration of embryo development, growth and reproduction, as well as genotoxic effects in aquatic organisms exposed to complex wastewater effluents. Comparison of the bioassays sensitivity was considered. Contrary to the lack of toxicity observed with normalized ecotoxicity tests, endpoints measured on zebrafish embryos such as developmental abnormalities and genotoxicity demonstrated a residual toxicity in wastewater both after a biological treatment followed or not by a tertiary O3 treatment. However, the ozonation step allowed to alleviate the residual endocrine disrupting potential measure in the biologically treated effluent. This study shows that normalized bioassays are not sensitive enough for the ecotoxicological evaluation of wastewaters and that there is a great need for the development of suitable sensitive bioassays in order to characterize properly the possible residual toxicity of treated effluents.  相似文献

Although the long incubation time of biochemical oxygen demand (BOD7) measurements has been addressed by the use of microbial biosensors, the resulting sensor-BOD values gained from the measurements with specific industrial wastewaters still underestimates the BOD value of such samples. This research aims to provide fast and more accurate BOD measurements in the dairy wastewater samples. Unlike municipal wastewater, wastewater from the dairy industry contains many substrates that are not easily accessible to a majority of microorganisms. Therefore, a bacterial culture, Microbacterium phyllosphaerae, isolated from dairy wastewater was used to construct a semi-specific microbial biosensor. A universal microbial biosensor based on Pseudomonas fluorescens, which has a wide substrate spectrum but is nonspecific to dairy wastewater, was used as a comparison. BOD biosensors were calibrated with OECD synthetic wastewater, and experiments with different synthetic and actual wastewater samples were carried out. Results show that the semi-specific M. phyllosphaerae-based microbial biosensor is more sensitive towards wastewaters that contain milk derivates and butter whey than the P. fluorescens-based biosensor. Although the M. phyllosphaerae biosensor underestimates the BOD7 value of actual dairy wastewaters by 25–32 %, this bacterial culture is more suitable for BOD monitoring in dairy wastewater than P. fluorescens, which underestimated the same samples by 46–61 %.  相似文献

Background

Di-(2-ethylhexyl) phthalate (DEHP) is a common plasticizer used in industrial and diverse consumer products. Animal studies indicate DEHP caused developmental, reproductive, and hepatic toxicities. However, human studies of the potential effects of DEHP are limited.

Methods

The exposed site with a history of over 20 years of waste plastic recycling was located in Hunan Province, China. The reference site without known DEHP pollution source was about 50 km far away from the exposed site. In this study, 181 workers working in plastic waste recycling and 160 gender?Cage matched farmers were recruited. DEHP concentrations in water and cultivated soil samples, serum thyroid-stimulating hormone, malondialdehyde (MDA), superoxide dismutase (SOD), urinary 8-hydroxy-2??-deoxyguanosine (8-OHdG), and micronuclei frequency in human capillary blood lymphocytes were analyzed.

Results

Mean levels of DEHP were greater in environment at the recycling site than at reference site (industry wastewater for the exposed: 42.43 ??g/l; well water: 14.20 vs. 0.79 ??g/l, pond water: 135.68 vs. 0.37 ??g/l, cultivated soil: 13.07 vs. 0.81 mg/kg, p?p?p?Conclusions The occupational DEHP exposure might contribute to oxidative deoxyribonucleic acid damage in the male workers.  相似文献

It is the first report in which a novel psychrotrophic Pseudomonas putida SKG-1 strain was evaluated for simultaneous bioremediation of pentachlorophenol and Cr6+ under various cultural and nutritional conditions. Pentachlorophenol (PCP) dechlorination products, bacterial structure, and functional groups were characterized by gas chromatography and mass spectrometry (GC–MS), scanning electron microscope and energy dispersive X-ray spectroscopy (SEM–EDS), and Fourier-transform infrared (FTIR) techniques. The strain was extremely tolerant to excessively higher individual concentration of PCP (1,400 mg l?1) and Cr6+ (4,300 mg l?1). Increasing concentration of PCP and Cr6+ exerted inhibitory effect on bacterial growth and toxicants’ removal. The strain exhibited growth, and concomitantly remediated both the pollutants simultaneously over a broad pH (7.0–9.0) and temperature (28–32 °C) range; maximum growth, PCP dechlorination (87.5 %), and Cr6+ removal (80.0 %) occurred at optimum pH 8.0 and 30 °C (from initial PCP 100 mg l?1 and Cr6+ 500 mg l?1) under shaking (150 rpm) within 72 h incubation. Optimization of agitation (125 rpm) and aeration (0.4 vvm) in bioreactor further enhanced PCP dechlorination by ~10 % and Cr6+ removal by 2 %. A direct correlation existed between growth and bioremediation of both the toxicants. Among other heavy metals, mercury exerted maximum and cobalt minimum inhibitory effect on PCP dechlorination and Cr6+ removal. Chromate reductase activity was mainly associated with the supernatant and cytosolic fraction of bacterial cells. GC–MS analysis revealed the formation of tetrachloro-p-hydroquinone, 2,4,6-trichlorophenol, and 2,6-dichlorophenol as PCP dechlorination products. FTIR spectrometry indicated likely involvement of carbonyl and amide groups in Cr3+ adsorption, and SEM–EDS showed the presence of chromium on P. putida surface. Thus, our promising isolate can be ecofriendly employed for biotreatment of various industrial wastes contaminated with high PCP and Cr6+ concentrations.  相似文献

The present work aims at evaluating the ability of the River Tiber natural microbial community to degrade naproxen in water samples collected downstream from a wastewater treatment plant. For this purpose, different water microcosms were set up (microbiologically active vs sterile ones) and treated with naproxen (100 μg/L) alone or in the co-presence of gemfibrozil in order to evaluate if the co-presence of the latter had an influence on naproxen degradation. The experiment was performed in the autumn and was compared with the same experimental set performed in spring of the same year to highlight if seasonal differences in the river water influenced the naproxen degradation. Pharmaceutical concentrations and microbial analysis (total cell number, viability, and microbial community composition) were performed at different times in the degradation experiments. The overall results show that the natural microbial community in the river water had a key role in the naproxen degradation. In fact, although there was a transient negative effect on the natural microbial community in all the experiments (3 h after adding the pharmaceutical), the latter was able to degrade naproxen within about 40 days. On the contrary, no decrease in the pharmaceutical concentration was observed in the sterile river water. Moreover, the co-presence of the two drugs lengthened the naproxen lag phase. As regards the natural microbial community composition detected by Fluorescence in situ Hybridization, Alpha and Gamma-Proteobacteria increased when the pharmaceutical halved, suggesting their role in the degradation. This study shows that with the concentration studied, naproxen was degraded by the natural microbial populations collected from a river chronically contaminated by this pharmaceutical.  相似文献

Utilization of agrowaste materials for the production of activated carbon, as an excellent adsorbent with large surface area, is well established industrially, for dephenolation of wastewater. In the present work, dried pods of Prosopis cineraria—a novel and low-cost agrowaste material—were used to prepare activated carbons by zinc chloride activation. Batch adsorption experiments were carried out to study the effects of various physicochemical parameters such as initial phenol concentration, adsorbent dose, initial solution pH, and temperature. Pseudo-first-order second-order and diffusion kinetic models were used to identify the possible mechanisms of such adsorption process. The Langmuir and Freundlich equations were used to analyze the adsorption equilibrium. Maximum removal efficiency of 86 % was obtained with 25 mg?L?1 of initial phenol concentration. The favorable pH for maximum phenol adsorption was 4.0. Freundlich equation represented the adsorption equilibrium data more ideally than the Langmuir. The maximum adsorption capacity obtained was 78.32 mg?g?1 at a temperature of 30 °C and 25 mg?L?1 initial phenol concentration. The adsorption was spontaneous and endothermic. The pseudo-second-order model, an indication of chemisorption mechanism, fitted the experimental data better than the pseudo-first-order Lagergren model. Regeneration of spent activated carbon was carried out using Pseudomonas putida MTCC 2252 as the phenol-degrading microorganism. Maximum regeneration up to 57.5 % was recorded, when loaded phenol concentration was 25 mg?L?1. The data obtained in this study would be useful in designing and fabricating an efficient treatment plant for phenol-rich effluents.  相似文献

Introduction

The effect of oceanic CO2 sequestration was examined exposing a deep-sea bacterium identified as Vibrio alginolyticus (9NA) to elevated levels of carbon dioxide and monitoring its growth at 2,750 psi (1,846 m depth).

Findings

The wild-type strain of 9NA could not grow in acidified marine broth below a pH of 5. The pH of marine broth did not drop below this level until at least 20.8 mM of CO2 was injected into the medium. 9NA did not grow at this CO2 concentration or higher concentrations (31.2 and 41.6 mM) for at least 72 h. Carbon dioxide at 10.4 mM also inhibited growth, but the bacterium was able to recover and grow. Exposure to CO2 caused the cell to undergo a morphological change and form a dimple-like structure. The membrane was also damaged but with no protein leakage.  相似文献

The present study used an eco-ditch system that employed Eichhornia crassipes, Bacillus subtilis, and Bellamya aeruginosa (E–B–B) during the summer and fall (high temperature) seasons and a second eco-ditch system that employed Elodea nuttallii, a compound microbial preparation called “EM bacteria”, and Hypophthalmichthys molitrix (E–E–H) during the winter and spring (low temperature) seasons successively to purify the discharged wastewater produced by Chinese soft-shelled turtle greenhouse cultivation. The wastewater was sampled, and the dynamic changes in the major nutrient pollutant indicators over several months were analysed. After the E–B–B and E–E–H eco-ditch purification systems were operated for nearly 140 days each, the following results were observed: the total nitrogen (TN) removal rates in the wastewater were 75 % and 69 %, respectively; the total phosphorus (TP) removal rates were 82 % and 86 %, respectively; the NH4 +-N removal rates were 91 % and 75 %, respectively; the chemical oxygen demand (CODcr) decreased 54 % and 44 %, respectively; the dissolved oxygen (DO) contents increased nearly 3 to 4 times; and the wastewater was maintained at neutral or alkaline pH values. The wastewater physical traits gradually changed from being yellow, brown, and muddy to being pale yellow, slightly turbid, and odourless. Both eco-ditch systems were observed to have a relatively favourable effect on the purification of Chinese soft-shelled turtle aquaculture wastewater. The continuous use of both eco-ditch systems could result in a year-round purification effect on Chinese soft-shelled turtle greenhouse aquaculture wastewater; therefore, this method has good prospects for promotion and application.  相似文献

The anoxic–oxic (A/O) process has been extensively applied for simultaneous removal of organic contaminants and nitrogen in wastewater treatment. However, very little is known about its ability to remove toxic materials. Municipal wastewater contains various kinds of pollutants, some of which have recalcitrant genotoxicity and may cause potential threat to environment, and even can lead to extinction of many species. In this study, we have selected three municipal wastewater treatment plants (WWTPs) employing anoxic–oxic (A/O) process to evaluate their ability to remove acute toxicity and genotoxicity of wastewater. Mortality rate of zebrafish (Danio rerio) was used to evaluate acute toxicity, while micronucleus (MN) and comet assays were used to detect genotoxicity. Results showed that in this process the acute toxicity was completely removed as the treatment proceeded along with decrease in chemical oxygen demand (COD) (<50 mg L?1) in the effluent. However, in these treatment processes the genotoxicity was not significantly reduced, but an increase in genotoxicity was observed. Both MN and comet assays showed similar results. The eliminated effluent may pose genotoxic threaten although its COD level has met the Chinese Sewage Discharge Standard. This study suggests that further treatment of the wastewater is required after the A/O process to remove the genotoxicity and minimize the ecotoxicological risk.  相似文献

The wastewater discharged from leather industries lack biodegradability due to the presence of xenobiotic compounds. The primary clarification and aerobic treatment in Bacillus sp. immobilized Chemo Autotrophic Activated Carbon Oxidation (CAACO) reactor removed considerable amount of pollution parameters. The residual untreated organics in the wastewater was further treated in algal batch reactor inoculated with Synechocystis sp. Sodium nitrate, K2HPO4, MgSO4.7H2O, NH4Cl, CaCl2·2H2O, FeCl3 (anhydrous), and thiamine hydrochloride, rice husk based activated carbon (RHAC), immobilization of Bacillus sp. in mesoporous activated carbon, sand filter of dimensions diameter, 6 cm and height, 30 cm; and the CAACO reactor of dimensions diameter, 5.5 cm and height, 30 cm with total volume 720 ml, and working volume of 356 ml. In the present investigation, the CAACO treated tannery wastewater was applied to Synechocystis sp. inoculated algal batch reactor of hydraulic residence time 24 h. The BOD5, COD, and TOC of treated wastewater from algal batch reactor were 20?±?7, 167?±?29, and 78?±?16 mg/l respectively. The integrated CAACO system and Algal batch reactor was operated for 30 days and they accomplished a cumulative removal of BOD5,COD, TOC, VFA and sulphide as 98 %, 95 %, 93 %, 86 %, and 100 %, respectively. The biokinetic constants for the growth of algae in the batch reactor were specific growth rate, 0.095(day?1) and yield coefficient, 3.15 mg of algal biomass/mg of COD destructed. The degradation of xenobiotic compounds in the algal batch reactor was confirmed through HPLC and FT-IR techniques. The integrated CAACO–Algal reactor system established a credible reduction in pollution parameters in the tannery wastewater. The removal mechanism is mainly due to co-metabolism between algae and bacterial species and the organics were completely metabolized rather than by adsorption.  相似文献
