Microbial dechlorination of three PCB congeners in river sediment.   总被引:3,自引:0,他引:3

We investigated the potential for the anaerobic degradation of three PCB congeners (2,3,5,6-CB, 2,3,4,5-CB, and 2,3,4,5,6-CB) in sediments collected from five monitoring sites along the Keelung River in northern Taiwan. Optimal conditions for congener dechlorination were 30 degrees C and pH 7.0. Intermediate 2,3,4,5-CB products were identified as 2,3,5-CB, 2,4,5-CB, and 2,5-CB. Intermediate 2,3,4,5,6-CB products were identified as 2,3,5,6-CB, 2,3,6-CB, and 2,5-CB. For 2,3,5,6-CB, intermediate products were identified as 2,3,6-CB and 2,5-CB. Dechlorination rates for PCB congeners were observed as (fastest to slowest): 2, 3, 4-CB > 2, 3, 4, 5-CB > 2, 3, 4, 5, 6-CB > 2, 3, 5, 6-CB > 2, 2', 3, 3', 4, 4'-CB > 2, 2', 4, 4' 6, 6'-CB > 2, 2', 3, 4, 4', 5, 5'-CB > 2, 2', 3, 3', 4, 4', 5, 5'-CB. Rates decreased for mixtures of the eight congeners. Dechlorination rates for the three primary congeners under different reducing conditions occurred in the order of (fastest to slowest): methanogenic condition > sulfate-reducing condition > nitrate-reducing condition. Under methanogenic and sulfate-reducing conditions, dechlorination rates were enhanced by the addition of lactate, pyruvate, or acetate, but delayed by the addition of manganese oxide, or ferric chloride. Under nitrate-reducing condition, dechlorination rates were delayed by the addition of lactate, pyruvate, acetate, manganese oxide or ferric chloride. Treatment with such microbial inhibitors as bromoethanesulfonic acid (BESA) or molybdate revealed that methanogen and sulfate-reducing bacteria were involved in the dechlorination of these three PCB congeners.  相似文献

Polychlorinated biphenyls (PCBs) are toxic compounds ubiquitously distributed in ecosystems. Microbial attenuation of these contaminants is a potential means of remediation. Two promising microbial PCB remediation technologies, biostimulation and bioaugmentation, were investigated in different sediments. Biostimulation experiments in which electron donor was supplied (H2 via elemental iron, Fe(0)) resulted in only a marginal improvement in the dechlorination of amended 2,3,4,5-tetrachlorobiphenyl (2,3,4,5-CB), likely because of an inadequate population of indigenous H2-utilizing dechlorinators. Extensive dechlorination was observed, however, after bioaugmenting microcosms with a PCB-dechlorinating enrichment culture. Dechlorination of 2,3,4,5-CB began prior to the 20th day of incubation and proceeded to 2-chlorobiphenyl. This extensive dechlorination activity was maintained in both sediments over 70d at 10 and 25 degrees C. This research demonstrates that although past studies of biostimulation were promising, a great deal must be known about the PCB-dechlorinating organisms present before successful biostimulation is expected. Bioaugmentation, however, appears to be a promising PCB remediation strategy and should be further pursued.  相似文献

The effect of acclimating anaerobic granules from commercial bioreactors with different carbon/electron sources on their ability to reductively dechlorinate a tri-(2,3,4-CB) and heptachlorobiphenyl (2,2',3,3',4,5,6-CB) was studied. The anaerobic granules were first grown in upflow anaerobic sludge blanket (UASB) reactors fed with two different mixtures of carbon/electron sources, i.e., propionate/butyrate/methanol and formate/methanol. Differences in dechlorination patterns for 2,2',3,3',4,5,6-CB were observed in batch experiments inoculated with granules from these two sets of UASB reactors. Variation of the carbon/electron source, during the dechlorination process, had no effect on the dechlorination pathway, but the extents and rates of dechlorination were highest for ethanol and formate and lowest for pyruvate fed batches. Pre-acclimation of different anaerobic sludges to polychlorinated biphenyls (PCBs) shortened the lag period, but did not influence the PCB dechlorination pathway. This is the first time that similar acclimation conditions for several anaerobic microbial communities prior to inoculation were reported to yield similar substrate specificities for the reductive dechlorination of specific PCB congeners. This research demonstrates a successful strategy for the development of biocatalysts to serve as the inoculum of partially decontaminated sites in order to provide microorganisms with specificities complementary to those of naturally occurring dechlorinators.  相似文献

Polychlorobiphenyl (PCB) biodegradation was followed for 1 year in microcosms containing marine sediments collected from Mar Piccolo (Taranto, Italy) chronically contaminated by this class of hazardous compounds. The microcosms were performed under strictly anaerobic conditions with or without the addition of Dehalococcoides mccartyi, the main microorganism known to degrade PCBs through the anaerobic reductive dechlorination process. Thirty PCB congeners were monitored during the experiments revealing that the biodegradation occurred in all microcosms with a decrease in hepta-, hexa-, and penta-chlorobiphenyls (CBs) and a parallel increase in low chlorinated PCBs (tri-CBs and tetra-CBs). The concentrations of the most representative congeners detected in the original sediment, such as 245-245-CB and 2345-245-CB, and of the mixture 2356-34-CB+234-245-CB, decreased by 32.5, 23.8, and 46.7 %, respectively, after only 70 days of anaerobic incubation without any bioaugmentation treatment. Additionally, the structure and population dynamics of the microbial key players involved in the biodegradative process and of the entire mixed microbial community were accurately defined by Catalyzed Reporter Deposition Fluorescence In Situ Hybridization (CARD-FISH) in both the original sediment and during the operation of the microcosm. The reductive dehalogenase genes of D. mccartyi, specifically involved in PCB dechlorination, were also quantified using real-time PCR (qPCR). Our results demonstrated that the autochthonous microbial community living in the marine sediment, including D. mccartyi (6.32E+06 16S rRNA gene copy numbers g?1 sediment), was able to efficiently sustain the biodegradation of PCBs when controlled anaerobic conditions were imposed.  相似文献

The microbial dechlorination of seven kinds of polychlorinated biphenyls (PCBs) by anaerobic microorganisms from river sediment was investigated. Dechlorination rates were found to be affected by the chlorine level of PCB congeners; dechlorination rates decreased as chlorine levels increased. Dechlorination rates were fastest under methanogenic conditions and slowest under nitrate-reducing conditions. The addition of individual electron donors (acetate, pyruvate, and lactate) enhanced the dechlorination of PCB congeners under methanogenic and sulfate-reducing conditions but delayed the dechlorination of PCB congeners under nitrate-reducing conditions. PCB congener dechlorination also was delayed by the addition of various polycyclic aromatic hydrocarbons (PAHs) under three reducing conditions and by surfactants, such as brij30, triton SN70, and triton N101. The results suggest that methanogen, sulfate-reducing bacteria, and nitrate-reducing bacteria all are involved in the dechlorination of PCB congeners.  相似文献

Known concentrations of phenanthrene, pyrene, anthracene, fluorene and acenapthene were added to soil samples to investigate the anaerobic degradation potential of polycyclic aromatic hydrocarbon (PAH). Consortia-treated river sediments taken from known sites of long-term pollution were added as inoculum. Mixtures of soil, consortia, and PAH (individually or combined) were amended with nutrients and batch incubated. High-to-low degradation rates for both soil types were phenanthrene > pyrene > anthracene > fluorene > acenaphthene. Degradation rates were faster in Taida soil than in Guishan soil. Faster individual PAH degradation rates were also observed in cultures containing a mixture of PAH substrates compared to the presence of a single substrate. Optimal incubation conditions were noted as pH 8.0 and 30 degrees C. Degradation was enhanced for PAH by the addition of acetate, lactate, or pyruvate. The addition of municipal sewage or oil refinery sludge to the soil samples stimulated PAH degradation. Biodegradation was also measured under three anaerobic conditions; results show the high-to-low order of biodegradation rates to be sulfate-reducing conditions > methanogenic conditions > nitrate-reducing conditions. The results show that sulfate-reducing bacteria, methanogen, and eubacteria are involved in the PAH degradation; sulfate-reducing bacteria constitute a major component of the PAH-adapted consortia.  相似文献

Abstract

The potential for dechlorinating 2,4‐dichlorophenoxyacetic acid (2,4‐D) and 2,4,5‐trichlorophenoxyacetic acid (2,4,5‐T) in soil with a consortium showing stable dechlorinating activity was investigated. The effects of adding electron donors and/or acceptors under three anaerobic reducing conditions was compared. Results show that both 2,4‐D and 2,4,5‐T dechlorination rates were enhanced in methanogenic conditions, delayed in sulfate‐reducing conditions, and inhibited in denitrifying conditions. Also under the same three conditions dechlorination was be enhanced by the addition of lactate, pyruvate, and acetate, delayed by the addition of manganese oxide and vitamin B12, and inhibited by the addition of ferric chloride. Response to treatment with such microbial inhibitors as bromoethane sulfonic acid (BESA), vancomycin, and molybdate suggests that the major bacteria involved in 2,4‐D and 2,4,5‐T dechlonnation is methanogen followed joined by sulfate‐reducing bacteria and eubacteria.  相似文献

In order to develop more robust insight into the natural attenuation of polychlorinated biphenyls (PCBs), the chlorine isotopic composition of residual 2,3,4,5-tetrachlorobiphenyl (2,3,4,5-CB) was monitored as it underwent microbial reductive dechlorination to 2,3,5-trichlorobiphenyl (2,3,5-CB) in laboratory cultures. Reverse-phase high performance liquid chromatography (HPLC) was employed to isolate the former compound from the experimental matrix for delta37Cl measurement. No detectable isotopic fractionation was observed over the 90 day incubation with sterile control, standard, and inoculated samples all exhibiting delta37Cl values with a range of approximately 0.5 per thousand. These results show that this type of biological activity can be discriminated from other transformations by the absence of a measurable isotope effect during microbial reductive dechlorination. The utility of HPLC isolation for compound-specific delta37Cl analyses of environmentally relevant species is also demonstrated.  相似文献

Anaerobic reductive dechlorination of polychlorinated biphenyls (PCBs) in heat treated, bromoethanesulfonate (BES) treated, and untreated slurries of PCB-contaminated upper Hudson River sediment was investigated to better understand the microorganisms that mediate this process. Extensive meta-and para-dechlorination of Aroclor 1242 and 2,3,4-trichlorobiphenyl (2,3,4-CB) occurred in both the untreated and heat treated slurries. Heat treatment of the slurries eliminated methanogenesis, enhanced 2,3,4-CB dechlorination, and caused extensive meta- and para-dechlorination of Aroclor 1242 earlier than in untreated slurries. BES treatment (1 mM) of the slurries caused a 90% reduction in methanogenesis and inhibited metadechlorination of PCB congeners containing 2,3- and 2,5-dichlorophenyl groups. The results suggest that there are at least two distinct microbial PCB reductive dechlorination activities in PCB-contaminated upper Hudson River sediment, a meta-dechlorination activity and a para-dechlorination activity on Aroclor 1242. Both of these microbial activities are apparently not methanogenic and are resistant to or activated by heat treatment. In addition, the meta- but not the para-dechlorinating activity is inhibited by BES treatment.  相似文献

To understand the dechlorination ability of chlorobenzenes (CBs) and polychlorinated biphenyls (PCBs) by untamed microorganisms under anaerobic condition and to correlate gas chromatographic properties with the occurrence of reductive dechlorination, introduction of CBs and PCBs in the culture medium inoculated with microorganisms from sludge and sediment, respectively, were performed. Three kinds of culture media preparing from sludge, river water and a synthetic medium were used in the experiments. HCB was degraded to 1,3,5-trichlorobenzene (1,3,5-TCB) and 1,3-dichlorobenzene (1,3-DCB) in both sludge medium and synthetic medium with inoculated microorganisms. Three PCB congeners including 2,3,4-, 3,4,5- and 2,3,4,5-CBp (chlorinated biphenyl) were not found to be dechlorinated in the river water medium with inoculation culture but to be dechlorinated in the synthetic medium. MNDO methodology was used to compute theoretical dechlorination reaction heats and GC-ECD techniques were used to estimate chromatographic data of CB and PCB congeners. Both CB and PCB congeners showed that dechlorination by untamed microorganisms under anaerobic mixed cultures were more likely to occur when larger amounts of energy were released and greater deltaln RRT value between the parent congener and the daughter product was observed. Deltaln RRT provided a more precise information on the singularity of PCBs ortho-dechlorination in an aspect of thermodynamic favorable rule.  相似文献

Twelve sewage sludges from rural, urban and industrial waste water treatment works in north west England were analysed for polychlorinated biphenyls by capillary GC-ECD. The total polychlorinated biphenyl content ranged between 106 to 712 μg/kg, with a mean of 292 μg/kg. Di- and penta-chlorinated congeners made the greatest contribution to the ΣPCB concentrations at most sites. The 6 IUPAC congeners were found in the following order of abundance, 101>180>153>138>52>28. Industrial inputs would appear to influence levels of ΣPCB in the sludge.

If these sludges were ploughed into arable land at typical rates only a slight elevation in soil ΣPCB levels would result. Alternatively, if the same quantity of sludge were applied to the surface of pasture grassland, significant elevations in the potential transfer of PCBs to grazing livestock may result.  相似文献

Dechlorination of polychlorinated biphenyls (PCBs) by nanoscale zerovalent iron (NZVI) is often strongly hindered by increased pH because large amounts of H+ ions were consumed during the surface reaction. The main objective of this work was to evaluate the effect of pH control in acid on the dechlorination processes of PCBs and to compare the dechlorination efficiency between 2,4,4′-trichlorobiphenyl (2,4,4′-CB) and the extracted PCBs from the field PCBs-contaminated soil in this system.  相似文献

用自制的污泥活性炭处理亚甲基蓝与酸性品红组成的染料废水,研究了pH、吸附时间、温度等因素对复合组分染料废水脱色率的影响,测试分析了污泥活性炭在处理亚甲基蓝与酸性品红复合组分染料废水过程中的重金属浸出毒性。结果表明:与处理单一组分染料废水相比较,处理复合染料废水时pH的影响较为复杂,2种染料在污泥活性炭上存在竞争吸附,但是污泥活性炭对复合组分染料的脱色效果较好。污泥活性炭对复合染料的吸附过程符合Langmuir型吸附。在处理染料废水的过程中,污泥活性炭中的重金属镉、锌及铬会浸出,重金属镉、锌的浸出浓度符合国家标准,但铬的浸出浓度已接近国家标准上限。  相似文献

The reductive dechlorination and behaviour of 14C-hexachlorobenzene (HCB) was investigated in an arable soil. The activity of the native anaerobic microbial communities could be induced by saturating the soil with water. Under these conditions high rates of dechlorination were observed. After 20 weeks of incubation only 1% of the applied 14C-HCB could be detected in the fraction of extractable residues. Additional organic substances, like wheat straw and lucerne straw, however considerably delayed and reduced the dechlorination process in the soil. The decline of HCB was not only caused by dechlorination but also by the formation of non-extractable residues, whereby their amounts varied with time depending on the experimental conditions. Several dechlorination products were detected, indicating the following main HCB transformation pathway: HCB → PCB → 1,2,3,5-TeCB → 1,3,5-TCB → 1,3-DCB, with 1,3,5-TCB as main intermediate dechlorination product. The other TeCB-, TCB- and DCB-isomers were also detected in low amounts, showing the presence of more than one dechlorination pathway. Since the methane production rates were lowest when the dechlorination rates were highest, it can be assumed that methanogenic bacteria were not involved in the dechlorination process of HCB. The established 14C-mass balances show, that with increasing dechlorination and incubation times, the 14C-recoveries decreased.  相似文献

As part of a program aimed at developing a field process for cleanup of PCB contaminated soils using photochemistry in basic 2-propanol, additional details of the dechlorination pathway are presented. The mechanism involves a chain reaction with both homolytic photochemical C-Cl bond fission and electron transfer steps producing PCB anion radicals. Kinetics of dechlorination of various congeners show patterns of relative rates associated with the basic 2-propanol medium that are not found in other media because both electron transfer and photochemical homolysis steps determine overall rates of dechlorination and govern the pathways and relative concentrations of intermediates. The electron transfer steps display opposite structure-reactivity correlations to the photo-homolysis, C-Cl bond fission steps. Oxygen quenching is shown to differentially affect both types of steps. In contrast to the suggestion that inter system crossing can be highly efficient with reaction originating from a PCB triplet, oxygen quenching data suggest that a significant minimum of the quantum yield is non-quenchable, presumably because of a reaction path from the PCB singlet. This may help to explain why exclusion of air is not entirely necessary in practice.  相似文献

采用城市污水厂污泥作为嗜酸微生物菌株来源,通过添加一定量的单质硫,使其中的嗜酸硫杆菌群大量增殖,并使污泥pH大幅降低.取得的培养物可用于废旧干电池中重金属沥滤等的处理.由于培养物对重金属沥滤效率和污泥的酸化速率密切相关,为此进行了不同的污泥种类、加硫量、污泥浓度和曝气强度对污泥酸化速率影响的实验.实验表明,初沉泥、二沉泥和混合浓缩污泥都能迅速利用硫产酸;加硫量(以100 mL污泥计)在0.5、1.0、2.0、4.0 g时,快速酸化的趋势相同.0.5 g的加硫量显示出略微慢的酸化速率,最后达到的最低pH在1.2左右,其他三个都降至1.0以下.污泥质量分数在0.5%、1.0%、2.0%、3.0%、4.0%时,也具有相同的酸化趋势,0.5%质量分数的污泥下降速率最快,这与较低浓度下污泥对pH的缓冲能力较小有关;曝气强度在0.45 L/min和0.3 L/min差别较小,5 d内能迅速降低pH,0.2 L/min的酸化速率较慢,足够长的时间(12 d)也能将pH降至2.5左右,0.1 L/min的曝气强度的酸化速率最慢,不能达到预期的酸化效果.  相似文献

Polychlorinated biphenyls (PCBs) are a class of persistent organic pollutants that are distributed worldwide. Although industrial PCB production has stopped, legacy contamination can be traced to several different commercial mixtures (e.g., Aroclors in the USA). Despite their persistence, PCBs are subject to naturally occurring biodegradation processes, although the microbes and enzymes involved are poorly understood. The biodegradation potential of PCB-contaminated sediments in a wastewater lagoon located in Virginia (USA) was studied. Total PCB concentrations in sediments ranged from 6.34 to 12,700 mg/kg. PCB congener profiles in sediment sample were similar to Aroclor 1248; however, PCB congener profiles at several locations showed evidence of dechlorination. The sediment microbial community structure varied among samples but was dominated by Proteobacteria and Firmicutes. The relative abundance of putative dechlorinating Chloroflexi (including Dehalococcoides sp.) was 0.01–0.19% among the sediment samples, with Dehalococcoides sp. representing 0.6–14.8% of this group. Other possible PCB dechlorinators present included the Clostridia and the Geobacteraceae. A PCR survey for potential PCB reductive dehalogenase genes (RDases) yielded 11 sequences related to RDase genes in PCB-respiring Dehalococcoides mccartyi strain CG5 and PCB-dechlorinating D. mccartyi strain CBDB1. This is the first study to retrieve potential PCB RDase genes from unenriched PCB-contaminated sediments.  相似文献
