Genetic differentiation between North Sea and Baltic Sea Hediste diversicolor (O.F. Müller, 1776) (Polychaeta: Nereididae) populations was studied by allozyme electrophoresis on starch gel. Thirteen loci were analyzed in eight populations. The level of genetic variation was very low (mean H o = 0.000 to 0.015). Differentiation between H. diversicolor populations is quite high (F ST = 0.892) and reflected by three enzyme loci (MDH-I*, MDH-II*, IDH-I*). The reduced gene flow (N m<1) may be explained by the limited dispersal capacity of the species. Regardless of whether found in the North Sea or Baltic Sea, there appear to be two different genetic types which are parapatric or sympatric in some places. The two types hybridize at three localities, but no signs of hybridization have been found at one (Tallinn). Received: 26 June 1997 / Accepted: 10 September 1997  相似文献

The genetic structure of Mycedium elephantotus (Pallas, 1766) populations from three regions around Taiwan was examined using allozyme electrophoresis. Eight loci were scored from seven enzyme systems. Seven loci were polymorphic under the 95% criterion. The high ratio of observed to expected genotypic diversities at the collection sites (G O:G E=0.8 to 1.0) indicate that M. elephantotus propagates predominantly by sexual reproduction. Allele frequencies of M. elephantotus differed significantly among regions (D=0.024 to 0.256, F ST=0.032 to 0.218, p < 0.001), while populations among collection sites within each region were homogeneous (D=0.000 to 0.015, F ST=0.010 to 0.022, p > 0.05). Genetic differentiation between populations from southern Taiwan and the Penghu Islands is greater than that between southern and northern Taiwan populations, although the former two regions are much closer geographically. The UPGMA dendrogram based on Nei's unbiased genetic distance showed a clear subdivision of populations into two groupings, northern Taiwan/Penghu Islands and southern Taiwan. A higher level of gene flow was found between M. elephantotus populations in northern Taiwan and the Penghu Islands (N m=7.56) than that between populations in southern Taiwan and other regions (N m=0.90 to 1.72). The pattern of genetic subdivision among regions is consistent with the pattern of ocean currents, indicating that genetic differentiation is likely driven by surface circulation vicariance. Samples of the scleractinian coral Pocillopora damicornis were collected from six sites located around four islands in the Ryukyu Archipelago, southern Japan, and subjected to allozyme electrophoresis. Seven polymorphic loci were examined for their allelic patterns. The ratio of observed to expected genotypic diversity (0.30 < G o :G e  < 0.64), the ratio of the observed number of genotypes to the number of individuals (0.47 < N g :N i  < 0.75), and deviations from Hardy–Weinberg equilibrium indicated that asexual reproduction plays a major role in the maintenance of established populations. However, populations were not completely dominated by a single or a few clones, and most clones were represented by only a few individual samples. The high frequency of typhoons in the region suggests that, in P. damicornis, fragmentation caused through occasional exposure to powerful waves is a major mode of asexual reproduction, but asexual production of planulae may also be contributing to the maintenance of populations. A significant genetic differentiation (F ST) was found between the six populations examined (0.027 < F ST < 0.092, average F ST = 0.056). The moderate gene flow is discussed according to characteristics of the larval stage of the species, and to circulation patterns in the region.  In south-eastern Australia, the prosobranch gastropods Morula marginalba (Blainville), Cominella lineolata (Lamarck) and Bedeva hanleyi (Angas) have similar fine-scale distributions, but appear to possess very different dispersal capabilities due to contrasting modes of larval development. M.marginalba produce planktonic larvae, whereas C. lineolata and B. hanleyi undergo direct development in benthic egg capsules and emerge as crawling juveniles. To test for possible effects of contrasting life histories on levels of genetic variation within and among populations, a survey was conducted of allozyme variation at six polymorphic loci in 8 to 9 local populations of each species. Collections of snails were made between June 1992 and November 1993. Sampling ranges spanned between 162 and 180 km of coast. Regardless of larval type, proportions of single-locus genotypes in each collection were consistent with the recruitment of offspring which had been generated through random mating. However, genotypic diversity was lower in those species that undergo direct development. Loci surveyed in C. lineolata and B. hanleyi were polymorphic (i.e. frequency of most common allele <95%) in fewer populations than those examined for M.␣marginalba (P <0.001) and, where polymorphisms occurred, also possessed significantly fewer alleles (P <0.001). Consequently, average levels of expected heterozygosity were greater in populations of M. marginalba than in those of either of the other species (P <0.001). Genetic variation among populations, expressed as the standardised variance in allele frequencies (F ST ), was inversely related to expected larval dispersal capability. The nine collections of M. marginalba showed little overall differentiation (F ST  = 0.017; P <0.001), reflecting the ability of planktonic larvae to interconnect local populations, and so limit divergence due to drift and natural selection. In contrast, there were high levels of allelic heterogeneity among the nine collections of C. lineolata (F ST  = 0.523; P <0.001) and eight collections of B. hanleyi (F ST  = 0.140; P <0.001). These data imply that for species which undergo direct development, local populations are effectively closed and evolve largely independent of one another.  Although oysters are commercially very important in Brazil, there is still much dispute about the number of Crassostrea species occurring on the Brazilian coast. The dispute is centered around C. brasiliana, considered by some authors to be a junior synonym of C. rhizophorae. In this paper we compared, by allozyme electrophoresis, sympatric and allopatric populations of the two putative species. Of the 17 loci analysed, five were diagnostic for the two species in sympatry (gene identity = 0.46 to 0.47), clearly demonstrating that they are distinct biological species. Heterozygosity (h) levels were high for both species (h = 0.24 to 0.28), and no heterozygote deficiencies were observed in any population (local inbreeding, F IS  = 0.141; P > 0.70). Levels of population structure in C. rhizophorae along 1300 km of coast were very low (population inbreeding, F ST  = 0.026; P > 0.15), indicating that the planktonic, planktotrophic larvae of these species are capable of long-range dispersal.  Genetic variation in 15 Holothuria (Microthele) nobilis (Selenka, 1867) populations on the Great Barrier Reef was studied at seven polymorphic allozyme loci. Although populations were separated by distances up to 1300 km, there were no apparent restrictions to gene flow (F ST values were not significantly different from 0) and the maximum Nei's unbiased genetic distance was 0.003. Populations were in Hardy–Weinberg equilibrium at all loci. The estimated maximum sexual input [this is the ratio of the number of sexually produced individuals (N*) to the sample size (N i )] and the minimum sexual input [this is the ratio of the number of genotypes (N go ) over the sample size (N i )] were used as estimators for the amount of asexual reproduction. Both parameters suggested that H. nobilis reproduces solely by sexual means (N*:N i : = 1; N go :N i  = 0.74 to 1). The allozyme data indicated high gene flow between populations, but the possibility that allozyme frequencies may not be at equilibrium means that it was not possible to distinguish whether the patterns reflect present-day dispersal or dispersal that occurred in the past. The genetic structure of 12 reef populations of the soft coral Sinularia flexibilis (Octocorallia, Alcyoniidae) was studied along the Great Barrier Reef (GBR) at a maximum separation of 1,300 km to investigate the relative importance of sexual and asexual reproduction, genetic differentiation and gene flow among these populations. S. flexibilis is a widely distributed Indo-Pacific species and a gamete broadcaster that can form large aggregations of colonies on near-shore reefs of the GBR. Up to 60 individuals per reef were collected at a minimum sampling scale of 5 m at two sites per reef, from December 1998 to February 2000. Electrophoretic analyses of nine polymorphic allozymes indicated that genotypic frequencies in most populations and loci did not differ significantly from those expected from Hardy–Weinberg predictions. Analysis of multi-locus genotypes indicated a high number of unique genotypes (N go) relative to the number of individuals sampled (N) in each reef population (range of 0.69–0.95). The maximum number of individuals likely to have been produced sexually (N*) was similar to the number of individuals sampled (i.e. N*:N ˜ 1), suggesting that even repeated genotypes may have been produced sexually. These results demonstrated a dominant role of sexual reproduction in these populations at the scale sampled. Significant genetic differentiation between some populations indicated that gene flow is restricted between some reefs (F ST=0.026, 95% CI= 0.011 − 0.045) and even between sites within reefs (F ST=0.041, 95% CI=0.027 − 0.055). Nevertheless, there was no relationship between geographic separation and genetic differentiation. Analyses comparing groups of populations showed no significant differentiation on a north-south gradient in the GBR. The pattern in the number of significant differences in gene frequencies in pairwise population comparisons, however, suggested that gene flow may be more restricted among inner-shelf reef populations near to the coast than among mid/outer-shelf populations further from the coast. Starch-gel electrophoresis of allozymes was used to differentiate the two red mullet species (Mullus barbatus L. and M. surmuletus L.) in the Mediterranean Sea and, further, to investigate the genetic stock structure of M. barbatus in the eastern Mediterranean area. Twenty putative enzyme-coding loci were examined in eight M. barbatus samples caught in the Aegean and Ionian Seas (Greece) and in the Gulf of Lion (France), and two M. surmuletus samples caught in the Aegean and Gulf of Lion. A high degree of genetic polymorphism was found in both species. Species-specific electrophoretic patterns were found in PGI* and PGM*. Estimates of variance of allele frequencies among samples (F ST) and 2 analyses both revealed significant differences (P < 0.05) among the M. barbatus samples. Most of the genetic variation was among samples regardless of region. The mean value of Nei's genetic distance between the two species was 0.329. Genetic distance among M. barbatus samples was low (maximum Nei's D = 0.012), with the sample from Platania differing most from other M. barbatus samples. This is probably be due to founder effects existing at this area. These results suggest that allozyme analysis may provide important information on the genetic structure of the red mullet to ensure sustainable management of this species. Zooplankton samples from the North Arabian Sea Environment and Ecosystem Research (NASEER) cruises were analyzed to determine the basic taxonomic composition, biomass (standing stock) and the total and copepod numeric abundance; these characteristics are discussed with reference to the different monsoon periods. Cruises carried out during March 1993 and May 1994, categorized as pre-southwest monsoon periods, and a cruise in December 1994, categorized as a northeast monsoon period, are discussed in detail. The biomass of January 1992 versus August 1992 and August 1992 versus March 1993 differed significantly (F = 6.44, P≤ 0.05). Ranges of highest and lowest biomass from each cruise are also given. Distinct “high” and “low” production areas of statistically significant difference (F = 12.67, P≤ 0.05) were observed. The “high” and “low” production areas were mobile and followed the surface wind circulation patterns (wind reversal pattern) during the northeast and southwest monsoon periods. Overall zooplankton showed a patchy distribution. The overall zooplankton abundance and total copepod counts differed significantly between the Cruises 3 versus 4 and 4 versus 5 (F = 15.67, P≤ 0.05 and F = 34.39, P≤ 0.05, respectively). There was no significant difference (P≥ 0.05) in biomass, between eutrophic and oligotrophic stations, suggesting no difference between near shore and offshore waters. Thirty-eight taxonomic groups were identified from the samples, with copepods being the most dominant group, followed by chaetognaths and siphonophores. Copepods constitute an average of 52.50 to 74.93% of the total zooplankton count and reach maxima of 92.14% of the total zooplankton count at the outset of the southwest monsoon (March 1993) and 91.39% at the outset of the active northeast monsoon (December 1994). Allozyme variation was used to investigate the genetic structure of the coral reef fishes Stegastes nigricans, Epinephelus merra and Acanthurus triostegus around New Caledonia. Each species was sampled from each of three sites in the ≃1000 km circumference of the lagoon of New Caledonia. Allelic variation was recorded for each species at 14, 13 and 17 loci, respectively, and heterozygosity diversity (H s) was 0.082, 0.065 and 0.116, respectively. Analysis of genetic differentiation between sites produced inconsistent results between species, with spatial heterogeneity in two species (S. nigricans, F st  = 0.038; A. triostegus, F st = 0.049) and homogeneity in one species (E. merra, F st = 0.000). Hydrological and climatic data from the lagoon suggest that the eastern and western sides of the lagoon are isolated, since they lie in water masses of different origin. This may explain the genetic differentiation and restricted gene flow found at a local scale for S. nigricans and A. triostegus. Homogeneity in populations of E. merra is discussed in relation to its low genetic diversity and its reproductive behaviour. Received: 23 April 1997 / Accepted: 25 September 1997  相似文献

An allozyme survey, using starch-gel electrophoresis, was carried out on eight populations of the Antarctic nemertean worm Parborlasia corrugatus (McIntosh, 1876) collected from locations around the South Orkney Islands, Antarctica. These populations were separated by distances in the order of tens of kilometres. Genetic variation was estimated over 22 enzyme loci for all populations examined, giving an observed heterozygosity of 0.142. This was much lower than the expected heterozygosity (H e  = 0.201), and it was found that there was a significant deficiency of het‐erozygotes across four enzyme loci ( p ≤ 0.01). A more detailed examination of this deficiency of heterozygotes was undertaken for the six populations and six variable enzyme loci for which the most complete data sets existed. A significant deficiency of heterozygotes was found at the enzyme locus Odh-1 for four of the six populations examined ( p ≤ 0.01). Mean F is (0.240) indicated a significant ( p ≤ 0.01) within-population component of the heterozygote deficiency estimated for the six populations sampled, and this was mainly due to the␣Ap-1, Odh-1 and Pgm-1 loci. The mean F st value (0.036) was also significant ( p ≤ 0.01), indicating a degree of genetic differentiation between populations. The observed levels of genetic differentiation between populations of P. corrugatus and the significant heterozygote deficiencies were unexpected, because this species has been reported to have a long-lived planktotrophic larva. It is hypothesised that recruitment of P. corrugatus in the South Orkney Islands originates from genetically distinct populations located in the Weddell Sea and to the west of the Antarctic Peninsula. Shifts in the relative position of the Weddell Sea Front, Weddell–Scotia Confluence and Scotia Front, relative to the South Orkney Islands, provide a mechanism for variation in the origin of recruits over time. Parma microlepis (Günther) were collected from Malabar, an urban location close to the centre of Sydney, Australia, and from Jervis Bay, a reference location 170 km south of the city centre. At each location, fish were collected from two sites separated by 100 to 200 m. The ultrastructure of normal liver tissue is described based on 20 female fish collected from Jervis Bay, where fish are known to be exposed to low levels of organochlorine contaminants. Alterations in the endoplasmic reticulum, mitochondria, lysosomes and nuclei of hepatocytes were identified and quantified in the liver tissue of fish from this location and compared to alterations in 20 female fish collected from Malabar, where fish are exposed to higher concentrations of organochlorine pesticides such as DDT compounds. There were significant differences in the percentage of hepatocytes with swollen mitochondria (F = 124.025, df = 2, 2, P = 0.008) and atypical nuclei (F = 22.198, df = 2, 2, P = 0.043) between sites (100 to 200 m apart), but there were no clear differences between the percentage of structural alterations in the hepatocytes of P. microlepis from Jervis Bay and Malabar. Associations between liver morphology and the organochlorines aldrin, dieldrin, DDE and chlordane were examined using a Pearson correlation matrix. Significant correlations were detected between the percentage of hepatocytes with dilated endoplasmic reticulum and the concentrations of the pesticide aldrin (r = 0.600, df = 11, r crit(α = 0.05) = 0.553). Significant associations were also detected between the percentage of hepatocytes with disorganised endoplasmic reticulum and the concentrations of dieldrin and DDE residues in fish (r = 0.576, r = 0.567, respectively, df = 13, r crit (α = 0.05) = 0.514). However, there was little evidence that ultrastructural alterations in fish responded to increasing concentrations of these pesticides in a consistent dose-response manner. The genetic relationships between morphologically indistinguishable marine and brackish populations of Syllis gracilis Grube, 1840 (Polychaeta: Syllidae) were studied by means of allozyme electrophoresis. Samples of S. gracilis from marine coastal and brackish-water habitats were examined for variation at 13 presumptive loci. In addition, a sample of the closely related species S. prolifera (Krohn, 1852) was analysed. Five loci were multiallelic in at least one population of S. gracilis and eight loci in S. prolifera. Low to moderate levels of within-population genetic variability were found, with average expected heterozygosity values ranging from H = 0.068 (±0.043 SE) to 0.187 (±0.069 SE) in the populations of S. gracilis; higher values were found in S. prolifera (H = 0.325 ± 0.076). The presence of various private alleles indicated a marked genetic divergence among populations of S. gracilis, with Nei's genetic distances ranging from D = 0.000 to 0.833 and a highly significant F ST value. Furthermore, evidence for strong genetic heterogeneity between two sympatric marine populations was found. UPGMA cluster analysis and multidimensional scaling pointed out a clear genetic divergence between brackish and marine populations. At least two genetically divergent entities occurred in marine and brackish habitats. This could be due to local adaptation of individuals coming from marine populations to brackish habitats, but more presumably to the occurrence of a species complex within S. gracilis. Genetic divergence among ten populations of small- and large-egg forms of the brackish-water polychaete Hediste japonica complex was investigated on 14 isozyme loci by electrophoretic analysis. The two forms were distinguishable by complete allele substitutions at five loci, resulting in high genetic differentiation (Nei's D: 0.533 to 0.662). No genetic evidence of hybridization between the two forms was detected in sympatric populations in three rivers. These results indicate that the two forms are reproductively isolated, clearly showing that the two forms are distinct species. The genetic differentiation among populations was higher in the large-egg form (D: 0.005 to 0.111, G ST: 0.435) than that in the small-egg form (D: 0.000 to 0.001, G ST: 0.020). This genetic difference between the two forms seems to be attributable to a difference in their life histories. The average expected heterozygosity was low in populations of both the large-egg form (0.005 to 0.068) and the small-egg form (0.014 to 0.038) in comparison with other marine invertebrates. The growth rates of two fish species, the winter flounder Pseudopleuronectes americanus (Walbaum) (19.3 to 42.6 mm total length, TL) and the tautog Tautogaonitis (Linnaeus) (23.9 to 55.9 mm TL), were used to evaluate habitat quality under and around municipal piers in the Hudson River estuary, USA. Growth rates were measured in a series of 10 d field caging-experiments conducted at two large piers in the summers of 1996 and 1997. Cages (0.64 m2) were deployed along␣transects that stretched from underneath the piers to beyond them, encompassing the pier edge (the transitional zone between the pier interior and the outside). Growth in weight (G w ) was determined at five locations along the transect, 40 m beneath the pier, 20 m beneath the pier, at the pier edge, 20 m beyond the pier edge, and 40 m beyond. Under piers, mean growth rates of winter flounder and tautogs were negative (xˉG W  = −0.02 d−1), and rates were comparable to laboratory-starved control fishes (xˉG W  = −0.02 d−1). In contrast, mean growth rates at pier edges and in open waters beyond piers were generally positive (xˉG W ranged from −0.001 to +0.05 d−1), with growth at pier edges often being more variable and less rapid than at open-water sites. Analyses of stomach contents upon retrieval of caged fishes revealed that dry weights of food were generally higher among fishes caged at open-water stations ( range = 0.02 to 0.72 mg dry wt) than at pier-edge ( range = 0.01 to 0.54 mg) or under-pier ( range = 0.03 to 0.11 mg) stations, although it was apparent that benthic prey were available at all stations on the transect. Our results indicate poor feeding conditions among fishes caged under piers, and suboptimal foraging among fishes caged at pier edges. Inadequate growth rates can lead to higher rates of mortality, and, based on these and other earlier experiments, we conclude that under-pier environments are poor-quality habitats for some species of juvenile fishes. Harp seals (Pagophilus groenlandicus Erxleben, 1777) comprise three populations based upon whelping areas in the Greenland Sea, White Sea, and Northwest Atlantic. The last comprises two subpopulations, one whelping in the Gulf of St. Lawrence (“Gulf ”) and one on the pack ice of the southern Labrador/northern Newfoundland coastal shelf (“Front”). A total of 40 female seals from the four whelping areas were collected during the 1990 and 1992 whelping seasons. DNA sequence variation was examined in a 307 bp region of the mitochondrial cytochrome b gene. Eleven variable nucleotide positions defined 13 genotypes: a significant fraction of the genotypic variance (F ST=0.12, or 0.09 as measured by Weir's coancestry coefficient θ) is attributable to differentiation between Northwest and Northeast Atlantic populations. There was no significant differentiation between the two whelping areas in the Northwest Atlantic, or between the Greenland Sea and White Sea. These findings suggest significant reproductive isolation exists between trans-Atlantic breeding populations. Received: 18 January 1999 / Accepted: 22 February 2000  相似文献

We examined the population structure of the black tiger prawn, Penaeus monodon Fabricius, 1798, in the Indo-West Pacific by analyzing the geographic distribution of elongation factor 1-alpha intron sequences from specimens collected during the winter and spring of 1997. Both the molecular phylogeny of alleles and F-statistics indicated very strong differentiation between populations from the western Indian Ocean and western Pacific. This pattern is concordant with other recent studies of marine species in this region, implying that the Indo-Australian Archipelago represents a biogeographic break between populations in the Indo-West Pacific. F ST-values among populations in the western Indian Ocean also indicate structure within this region, whereas no structure was found among western Pacific populations. Nucleotide diversity was significantly lower in the western Indian Ocean populations than in the western Pacific, implying that the populations have regional differences in demographic history. The genetic relationships among South-East Asian populations of the scad mackerels Decapterus macarellus, D. macrosoma and D. russelli (Pisces: Carangidae) were investigated. In 1995 and 1996, 216 fish were sampled in seven localities spanning the seas of Indonesia and were examined for restriction-site polymorphisms using ten restriction enzymes for the mitochondrial (mt) DNA control region, amplified by the polymerase chain-reaction. The inferred phylogeny of haplotypes led to the recognition of three distinct mitochondrial lineages or phylads consistent with the distinctions of current taxonomy. All 15 mtDNA haplotypes found in D. macarellus and all 9 haplotypes found in D. macrosoma were arranged as star-like clusters, suggesting recent evolutionary history. In contrast, the phylad formed by 6 haplotypes in D. russelli from the Sulawesi Sea exhibited diffuse topology, suggesting that ancestral lineages of this species have been retained to the present. Average nucleotide-divergence estimates between haplotypes of different phylads were between 0.042 and 0.135, suggesting ancient separation, in consistency with published allozyme data. High levels of haplotype diversity, but no geographical heterogeneity, was detected within D. macarellus from the Molucca Sea and the Banda Sea. Populations of D. macrosoma exhibited both significant differences between adjacent regions (Sunda Strait and Java Sea), and broadscale genetic homogeneity from the South China Sea to the Sulawesi Sea via the Java Sea and Makassar Strait. The geographic isolation of the D. macrosoma population sampled in the Sunda Strait suggests that this region constitute a sharp transition zone between the Indian Ocean and the Sunda Shelf. Near-monomorphism of haplotypes and low nucleotide diversity (d X) were observed in the samples of D. macrosoma from the continental shelf (haplotype-diversity estimates, h, = 0.00 to 0.25 ± 0.08 and d X = 0.000 to 0.002). This was in contrast to the comparatively high haplotype and nucleotide diversities observed in other pelagic fish species including D. macarellus (h = 0.82 ± 0.05, d X = 0.012 to 0.015) and D. russelli (h = 0.63 ± 0.12, d X = 0.016), and in the oceanic D. macrosoma population sampled in the Sunda Strait (h = 0.67 ± 0.31, d X = 0.005). We hypothesise that this may be the consequence of recent and perhaps repeated bottleneck events that have affected the D. macrosoma population sampled on the continental shelf. Results of isozyme electrophoresis were used to explore the genetic relationships between several Mediterranean morphs of Cerithium (Gastropoda: Prosobranchia), for which taxonomy is currently uncertain because of high intraspecific variability and low interspecific differentiation. The large species, classically known as C. vulgatum Bruguière, 1789 was identified at four sites (two in the French Mediterranean and two in southern Spain). Two different larval types were found in the French sites, but poecilogony could not be demonstrated. Individuals collected from harbours were not genetically distinct from open-sea populations of classic C. vulgatum. However, a population in the Embiez lagoon (French Mediterranean) which morphologically resembles C. vulgatum did display distinct genetic traits, supporting its status as a separate species. Of the small Cerithium species usually known as C. rupestre, two sympatric species (C.rupestre” Risso, 1826 and C. lividulum Risso, 1826) were distinguished. Genotype frequencies within the analysed populations revealed much heterozygote deficiency. F ST values (fixation index measuring the effects of population subdivision) suggest a higher genetic differentiation for C. lividulum populations than for C. vulgatum populations. We assume that a high larval dispersal capability (via planktotrophy) allows a high gene flow between populations of C. vulgatum. Received: 24 November 1998 / Accepted: 24 September 1999  相似文献

Photosynthetic rates of eight seagrass species from Zanzibar were limited by the inorganic carbon composition of natural seawater (2.1 mM, mostly in the form of HCO3 ), and they exhibited more than three time higher rates at inorganic carbon saturation (>6 mM). The intertidal species that grew most shallowly, Halophila ovalis, Halodule wrightii and Cymodocea rotundata, showed the highest affinity for inorganic carbon (K 1/2 = ca. 2.5 mM), followed by the subtidal species (K 1/2 > 5 mM). Photosynthesis of H. wrightii, C. rotundata, Cymodocea serrulata and Enhalus acoroides was >50% inhibited by acetazolamide, a membrane-impermeable inhibitor of carbonic anhydrase, indicating that extracellular HCO3 dehydration is an important part of their inorganic carbon uptake. Photosynthetic rates of H. wrightii, Thalassia hemprichii, Thalassodendron ciliatum, C. serrulata and E. acoroides were strongly reduced by changing the seawater pH from 8.2 to 8.6 in a closed system. In H. ovalis, C. rotundata and Syringodiumisoetifolium, photosynthesis at pH 8.6 was maintained at a higher level than could be caused by the ca. 30% CO2 concentration which remained in the closed experimental systems at that pH, pointing toward HCO3 uptake in those species. It is suggested that the ability of H. ovalis and C. rotundata to grow in the high, frequently air-exposed, intertidal zone may be related to a capability to take up HCO3 directly, since this is a more efficient way of HCO3 utilisation than extracellular HCO3 dehydration under such conditions. The inability of all species to attain maximal photosynthetic rates under natural conditions of inorganic carbon supports the notion that seagrasses may respond favourably to any future increases in marine CO2 levels.
