山西高原天然油松群体过氧化物酶和多酚氧化酶分析   总被引:1,自引:0,他引:1

利用聚丙烯酰胺凝胶电泳技术,对山西高原5个油松(Pinus tabulaeformis)天然种群的过氧化物酶和多酚氧化酶进行了分析。从140份供试材料的带型计算出油松种群多态位点百分数为95%,等位基因平均数为1.95,期望杂合度为0.357,遗传分化系数为0.036。表明山西高原天然油松种群间产生一定程度的分化,在同工酶水平上呈现出遗传多态性,且大部分遗传变异存在于种群内。  相似文献

油松种子醇溶蛋白提取剂比较   总被引:1,自引:0,他引:1  
采用乙醇、尿素、2氯乙醇、2巯基乙醇等配成的不同浓度和成分的提取剂提取了山西太岳龙门口、陕西洛南、辽宁千山和河北雾灵山4个不同油松种群的种子醇溶蛋白;用酸性聚丙烯酰胺凝胶电泳(APAGE)对蛋白提取液进行分析.条带数目反映所提取醇溶蛋白的数量,谱带颜色深浅反映不同提取剂得到的某种醇溶蛋白的量.结果表明,不同提取剂提取的醇溶蛋白图谱存在明显差异.由1%2巯基乙醇和12%尿素组成的复合提取剂提取效果最佳,其次是由20%2氯乙醇、1%2巯基乙醇和12%尿素组成的复合提取剂.提取的醇溶蛋白的电泳图谱可以用于油松种群的遗传多样性分析和系统演化的研究.图1表2参11  相似文献

利用分子标记技术分析了山西5个天然油松种群140个个体的遗传特征,比较了不同种群的多态位点显性频率、遗传多样性指数、基因流和分化系数.筛选的5个ISSR引物和5个RAPD引物共扩增到位点68个,其中多态位点50个遗传参数的统计分析显示,多态位点的显性频率、遗传多样性指数、分化系数分别介于0.000 0~1.000 0、0.0200~0.500 0和0.003 7~0.615 62:间,表明油松种群遗传多样性的主要来源是等位基因频率的不同和部分位点较大程度的遗传分化.根据油松的种群遗传学特征,认为只有加强天然油松林的生境保护,使其不产生片断化,才能使油松的遗传资源得到有效保护.图2表4参16  相似文献

利用随机扩增多态性DNA(RAPD)方法对威海荣成地区鹿角菜(Silvetiasiliquosa)野生种群共30个个体的遗传多样性水平及遗传结构进行研究。结果表明,利用21条随机引物共检测到112个多态性位点,多态性位点百分率为93.75%,Nei基因多样性指数为0.3515±0.1352,Shannon多样性指数为0.5046±0.1126。通过聚类分析而划为一类的3个亚种群间的遗传分化指数为0.1349~0.2189,基因流为1.7793。研究结果表明威海荣成地区的鹿角菜种群遗传多样性较高,种群内存在较为明显的遗传分化,种群遗传资源不受遗传漂变的影响。针对当地鹿角菜遗传资源的现状,应加强现存种群的就地保护,恢复种群规模;进行取样养殖保护时则需避免因近交而造成种群资源退化。  相似文献

报道了青藏高原青稞B组醇溶蛋白的遗传多样性.在66份青藏高原青稞材料中,共有15种不同条带、29种不同条带组合,表明青藏高原青稞材料具有丰富的多态性.不同来源的群体材料间遗传多态性存在差异,西藏育成品种的多态性略低于四川农家品种,人工选择使其遗传多态性减弱.聚类分析将材料分成3组,材料聚类与材料来源地没有明显的相关性.图3表5参26  相似文献

为了解珠江流域重要野生经济鱼类赤眼鳟的遗传多样性和遗传分化现状,对柳江、西江、北江、郁江、南盘江和红水河6个自然种群共170尾个体的线粒体控制区(D-loop)序列进行测定与分析.结果显示,共获得长978 bp的序列,检测出106个单倍型,单倍型多样性平均为0.982 0,核苷酸多样性平均为0.013 53,表明珠江流域赤眼鳟具有较高的遗传多样性.遗传距离和遗传分化结果均显示珠江流域赤眼鳟未出现明显分化,各种群亲缘关系较近.基于D-loop序列构建的NJ树显示,106个单倍型分为两大支系(A支和B支).A支包含6个种群中的大部分单倍型,未形成明显单系群;B支由西江种群的两个特有单倍型组成.单倍型网络亲缘关系分析结果也表明其未出现明显分化.AMOVA分析结果表明,绝大部分变异出现在种群内(99.36%),而地理区间和地理区内种群间的变异很小.中性检验显示,珠江流域赤眼鳟历史上曾发生过种群扩张.综合上述研究结果,认为西江为珠江流域赤眼鳟的扩散中心.  相似文献

通过离体选择抗赖氨酸加苏氨酸玉米突变体,不仅获得了积累游离苏氨酸和赖氨酸等氨基酸的突变类型,也选择到了种子蛋白组份发生改变的高蛋氨酸突变体和高赖氨酸体细胞无性系变异体。高蛋氨酸种子中总蛋氨酸的含量比对照增高22.6%,这是由于醇溶蛋白Zein-2部分及其蛋氨酸含量增高所致。高赖氨酸变异体种子游离必需氨基酸增高显著(2-10倍),总赖氨酸比对照亲本增高28.1%,含量约占种子干重的0.40%,种子蛋白清蛋白、球蛋白和谷蛋白组份含量增高而醇溶蛋白减少。高赖氨酸和高蛋氨酸特性遗传稳定,植株育性正常。  相似文献

采用RAPD技术对中国对虾中国黄渤海沿岸种群(HB)和朝鲜半岛西海岸种群(PK)的遗传多样性进行检测。20个随机引物(OPD系列)在二个种群中都扩增出148条DNA片段,其中多态性片段数分别为54条和65条,多态性片段的比例分别为36.49%和43.92%,种群的平均杂合度分别为0.1981和0.2375。实验表明,中国对虾朝鲜半岛西海岸种群的遗传多样性要比中国黄渤海沿岸种群高,但二个种群都处于较  相似文献

综述了淡水腹足类微卫星位点的获取方法、微卫星重复序列的特点以及微卫星标记技术在其种群遗传学研究中的应用,着重分析了微卫星多态性在淡水腹足类种群遗传多样性、种群遗传结构、遗传分化、交配系统、种群关系研究中的应用.淡水腹足类普遍表现为杂合体缺失,种群遗传多样性降低.淡水腹足类近交和自体受精现象比较频繁以及地理隔离导致种群间遗传分化.利用微卫星标记研究遗传杂合度,结合群体近交系数和基因分化系数在一定程度上反映了淡水腹足类的交配系统.根据不同种群的遗传距离和相似性进行聚类分析在判别种群间的亲缘关系上起到了很大作用.针对淡水腹足类自身的特性,微卫星分子标记技术在淡水腹足类入侵生物学、分类阶元,寄生虫的分子流行病学等研究领域有广泛的应用前景.  相似文献

利用扩增片段长度多态性(amplified fragment length polymorphism,AFLP)分子标记技术,对金花茶(Camellia nitidissima) 4个天然种群的遗传多样性和遗传结构进行研究,为金花茶资源的有效保护和高效利用提供科学依据。结果表明,8对引物共扩增出1 619个DNA位点,其中1 473个位点具多态性,多态性位点百分率为90. 98%。金花茶物种水平基因多样性指数和Shannon信息指数为0. 146和0. 246,种群水平基因多样性指数和Shannon信息指数为0. 131和0. 215; 4个金花茶种群的多态位点百分率、基因多样性指数和Shannon信息指数变化趋势一致,其中多态位点百分率介于56. 83%~72. 11%之间,基因多样性指数介于0. 122~0. 138之间,Shannon信息指数介于0. 197~0. 228之间。金花茶种群间的遗传分化系数介于0. 139~0. 289之间,遗传变异主要存在于种群内。分子变异分析(AMOVA)结果表明,金花茶种群内遗传变异为81. 52%,种群间遗传变异为18. 48%,遗传变异主要存在于种群内。非加权组平均法(UPGMA)聚类结果表明,种群间遗传变异与其地理距离无明显相关性。应加强现有金花茶种群的就地保护,促进种群自然更新,同时加强迁地保护。  相似文献

采用随机扩增多态DNA技术研究了5个茸鹿品种(品系)75个个体的遗传变异关系。在所使用的41个引物中,有40个引物扩增出多态谱带,共检测到395条扩增片段,其中259条(65.6%)出现变异,用Shaknnon指数计算了5个品种(品系)的遗传变异及其遗传变异在群体内和群体间的分布。利用Nei氏片段共享度计算了75个个体间的遗传距离,用UPGMA聚类法构建了75个个体的系统发育树状图,反映5个茸鹿品  相似文献

The genetic variation in populations of Anatolian black pine (Pinus nigra Arn. subsp. pallasiana (L.) Holmboe.), one of the species covering large areas in Turkey, was investigated. Open pollinated seeds were collected from 13 populations in a natural distribution range. Six characters of seeds (length, width, ratio of length to width, weight/1000 seeds) and seedling characters (cotyledon number and hypocotyls height) and two enzyme systems viz. leucine aminopeptidase (LAP) and glutamate oxaloacetate transaminase, (GOT) were investigated. Significant differences were detected among the populations for the morphological characters. In addition, isozyme patterns of two enzyme systems revealed that LAP has two loci (one with 2 alleles and the other with 3), while GOT has three loci (two with 3 alleles and the third one with 2 alleles). Polymorphic loci were 74% on the average. The mean number of alleles per loci was 1.94 and expected heterozygosity was 19%. The mean total genetic diversity was calculated as 0.203; the mean gene diversity within populations was determined as 0.188, and the average between subpopulations diversity was 0.016. The relative magnitude of genetic differentiation among subpopulations was measured as 0.074 indicating that only 7.4% of the total genetic diversity was there between populations. 为了阐明加拿大一枝黄花成功入侵的机制,利用简单序列重复区间标记(ISSR)方法对加拿大一枝黄花和本地一枝黄花的遗传多样性进行比较研究。从100条引物中筛选出12条引物用于PCR扩增,利用POPGEN32软件对2种一枝黄花进行遗传多样性分析。结果表明,加拿大一枝黄花在物种水平上的多态位点百分率为95.19%,Nei’s基因多样性指数为0.308 5,Shannon’s信息指数为0.415 8;本地一枝黄花在物种水平上的多态位点百分率(89.80%)、Nei’s基因多样性指数(0.249 1)和Shannon’s信息指数(0.383 4)都比加拿大一枝黄花小。加拿大一枝黄花和本地一枝黄花居群间遗传分化系数分别为0.118 2和0.131 3,居群内变异分别为0.881 8和0.868 7,表明2个物种居群间的遗传分化不明显,遗传一致度高,且主要的遗传变异存在于居群内。入侵植物加拿大一枝黄花具有较高遗传多样性,且高于本地一枝黄花,这可能是加拿大一枝黄花成功入侵的原因之一。  相似文献

从1800m到3400m五个海拔连续取样,用RAPD分子标记研究了卧龙自然保护区中国沙棘种群的遗传结构和遗传变异.用11条寡核苷酸引物,扩增得到151个重复性好的位点,其中143个多态位点,多态率达94.7%.在5个沙棘种群中,总遗传多样性值(HT)为0.289,B种群内的遗传多样性值为0.315,这完全符合沙棘这种多年生、远交的木本植物高遗传变异的特性.5个种群内遗传多样性随海拔升高呈低—高—低变异趋势,在2200m海拔处的B种群遗传多样性达最大值0.315,3400m海拔处的E种群则表现最小,仅0.098.5个种群间的遗传分化值GST=0.406,也即是说有40.6%的遗传变异存在于种群间,59.4%存在种群内.1800m海拔处的A种群与其他种群的明显分离是造成种群间遗传分化大的原因.UPGMA聚类图和PCoA散点图分别进一步确证了5个种群间关系和所有个体间的关系.最后,经过Mantel检测,遗传距离与海拔表现了明显的相关性(r=0.646,P=0.011).  相似文献

This paper reports data on 28 allozyme loci in wild and artificially reared sea bass (Dicentrarchus labrax) samples, originating from either coastal lagoon or marine sites in the Mediterranean Sea. F ST analysis (θ estimator) indicated strong genetic structuring among populations; around 34% of the overall genetic variation is due to interpopulation variation. Pairwise θ estimates showed that, on average, the degree of genetic structuring was much higher between marine populations than between samples from lagoons. Six polymorphic loci showed differences in allele frequencies between marine and lagoon samples. Multivariate analyses of individual allozymic profiles and of allele frequencies suggested that different arrays of genotypes prevail in lagoons compared to marine samples, particularly at those loci that, on the basis of previous acclimation experiments, had been implicated in adaptation to freshwater. On the other hand, variation at “neutral” allozyme loci reflects to a greater extent the geographic location of populations. Allozyme differentiation was also studied in a D. labrax population from the Portuguese coast. Average genetic distance between this population and the Mediterranean populations was quite high (Nei's D = 0.236) and calls into question the taxonomic status of the Portuguese population. Finally, genetic relationships between D. labrax and D. punctatus were evaluated. Average Nei's D was 0.648, revealing high genetic differentiation between the two species, even for two sympatric populations of these species in Egypt; thus gene flow was not indicated between species. Mangrove forests, with their ecological significance and economic benefits, are vital inter-tidal wetland ecosystems. Lumnitzera littorea (Combreataceae) is a non-viviparous mangrove distributed in tropical Asia and North Australia. Due to natural and human impacts, populations of this species have been isolated, fragmented, and highly disturbed. In China, L. littorea is an endangered species, restricted to small regions of Hainan Island. The genetic composition of five populations of this species from the Indo-West Pacific (South China, Malay Peninsula, Sri Lanka, North Australia) was assessed using inter simple sequence repeat (ISSR) makers. At the species level, expected mean heterozygosity (He) was 0.240 with 75.6% of loci polymorphic (P). However, genetic variation was much lower at the population level (P = 37.1%, He = 0.118). A high coefficient of gene differentiation (Gst = 0.515) and low level of gene flow (Nm = 0.470) indicated significant genetic differentiation among populations. AMOVA also indicated that more than half the total variation (58.4%) was partitioned among populations. The high degree of differentiation observed among populations emphasizes the need for appropriate conservation measures that incorporate additional populations into protected areas, and achieve the restoration of separate, degraded populations.  相似文献

 We describe three highly polymorphic microsatellite loci which have been isolated from the giant tiger prawn Penaeus monodon. The number of alleles present among 312 samples at the loci Pmo9, Pmo25 and Pmo27 were 84, 34 and 35, respectively, with heterozygosities all >90%. Analyses of the distribution of length variation at three microsatellite loci among five Australian P. monodon populations revealed strong differentiation between populations from the west and those from the northern and eastern coasts. Tests for population differentiation (F st) values and an analogous measure for microsatellite loci (R st) all demonstrated that Western Australian P. monodon are a separate genetic stock which exhibits reduced genetic variation relative to the other populations. Reduced variability is consistent with a recent population bottleneck or colonization by a small founding population from the east when sea links between Indonesia, New Guinea and Australia were re-established following the last ice age. Variation in the endospermal protein patterns of seeds amongst and between inland and coastal populations of the dune building grassLeymus arenarius was examined in Iceland. Seeds were collected from six coastal populations and five inland populations in Iceland. Endospermal proteins (prolamins) of seeds were extracted with Tris-buffered 2-propanol (50%) and β-mercaptoethanol (0.5% v/v). We used 8% and 12% sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) to separate the prolamin protein bands, which were stained with Coomassie Brilliant Blue R. Coastal populations were differentiated from inland populations on the basis of high frequency of high molar weight (HMW) (110–150 kg mol−1) prolamins. Coastal populations had significantly higher proportions of their seed with more than two HMW prolamins, than those of inland populations. Subtle differences were found among the HMW prolamins of coastal and inland populations indicating limited intrapopulation variation. The dynamic environment of sand dunes probably influences the genetic composition of these populations. The results suggest that seeds of the inland and coastal populations should be treated separately in sand dune reclamation and restoration works  相似文献

Planktonic populations of the calanoid copepod Labidocera aestiva show significant biochemical genetic heterogeneity along the Atlantic coast of the USA. In summer, 1981, copepods were collected by surface tows at Beaufort Inlet, North Carolina; Fort Pierce Inlet, Florida; and Vineyard Sound, Massachusetts. Genetic variation within each population and genetic differentiation among the three populations were studied by micro-acrylamide gel electrophoresis of six loci encoding four enzymes. All six enzyme loci were polymorphic when all populations were considered together, but the North Carolina population was monomorphic at two loci. High genetic variability was indicated by the following: (1) the number of alleles per locus averaged over all loci was 2.57±0.26 SD; (2) the average proportion of loci for which the frequency of the most common allele was not greater than 0.95 was 0.78±0.10; (3) the frequency of heterozygous individuals was 0.25±0.07. Genetic differentiation among population samples in the three regions was demonstrated in several ways: allele frequencies at one aminopeptidase-I locus, Lap-1, differed significantly among samples of the three populations, and there were unique alleles of high frequency at this locus in two population samples. Values of the statistic of genetic distance (D) averaged 0.20±0.08 for pairwise comparisons between all samples. Compared to expected heterozygosity if individuals across the entire range sampled mated at random, there were highly significant heterozygote deficiencies at five of the six loci. Genetic differentiation of populations of L. aestiva may result from (1) differential selection on populations in the three regions, or (2) restricted gene flow between the populations.
