The fatty-acid composition of lipids from ovulated eggs of wild and cultured turbot was investigated in order to estimate the nutritional requirements during embryonic and early larval development. Lipid comprised 13.8±0.5% (n=5) and 13.2±0.7% (n=7) of the egg dry weight in wild and cultured turbot, respectively. Polyunsaturated fatty acids (PUFA) of the (n-3) series accounted for 39% of total fatty acids in total lipid of both wild and cultured fish. The predominant (n-3) FUFA was docosahexaenoic acid (22:6 n-3), which also was the most abundant fatty acid in turbot eggs and comprised 24 and 23% of the total egg fatty acids in wild and cultured fish, respectively. Phospholipids, triacylglycerols and cholesterol-wax esters of turbot eggs all exhibited a specific fatty-acid profile distinctly different from that of total lipid. The general pattern of the fatty-acid distribution in lipids of eggs from wild and cultured turbot was similar, but the relative amount of 18:2(n-6) was considerably higher and 20:1(n-9) slightly higher in cultured fish. These differences were extended to all lipid classes and probably reflect the dietary intake of certain vegetable and marine fish oils. Calculations based on light microscopical studies showed that 55 to 60% of the total lipids in cultured turbot eggs are confined to the oil globule. The size of the oil globule remained constant during embryogenesis, and a reduction in size occurred first after hatching and mainly after yolk depletion. This implies that the total amount of lipids utilised during the embryonic development is considerably less than the total lipids present in ovulated turbot eggs. Comparison of the fatty-acid composition of total lipids from eggs and vitellogenin of wild turbot reveals that egg lipids contained a lower level of saturated and a higher level of monounsaturated fatty acids. Eggs also contained wax esters, which were not detected in vitellogenin, suggesting that vitellogenin is not the only source of lipids for turbot eggs.  相似文献

This paper reports on the firstPhaeocystis sp. (hereafterPhaeocystis) bloom in the Arabian Gulf. Total lipids from threePhaeocystis colonies sampled in November 1987 and in March and May 1988, comprised ca 11.0% of the dry biomass. Triacylglycerols (TG) and/or free fatty acids (FA) predominated in the lipids of all three samples. Polar lipids consisted of mixtures of phospholipids and glycolipids. Diacylglycerotrimethylhomoserines (DGTH) were present in all extracts, whilst conventional phospholipids only occurred in low concentrations. The predominant glycolipids were monogalactosyldiacylglycerols (MGDG) digalactosyldiacylglycerols (DGDG), sulfoquinovosyldiacylglycerols (SQDG) and steryl glycosides (SG). The predominant fatty acids in the total lipids of all three samples were palmitic (16:0) and oleic (18:1) acids. Total lipids of the November sample contained the smallest proportion of palmitic and the largest of polyunsaturated fatty acids (PUFA) — especially C16. The proportion of PUFA was considerable in individual polar lipids. C16 PUFA were more common in individual polar lipid classes of the November and March samples than that of May. The concentration of constituent C18 PUFA was relatively low. PUFA with chains longer than 18 C atoms only occurred in very low concentrations, and were confined to certain polar and nonpolar lipid classes of the November sample only.  相似文献

Many free-living copepods produce and store lipids prior to entering diapause (long-term dormancy). Heteropsyllus nunni Coull is the only marine harpacticoid copepod known to undergo any form of diapause. This study presents the first information on the types of lipids and fatty acids produced for long-term diapause in this benthic species. Sexually immature adults of H. nunni undergo diapause within a pliable self-made cyst. Prior to entering diapause (which lasts 3–4 months), they produce and store large amounts of orange lipid. The lipids apparently are utilized during diapause. Although some residual lipids remain, chiefly around the gonads, after the copepods emerge from their cysts, the lipid stores are visibly reduced. Typically, the copepods mate and produce eggs within 48 h after diapause is terminated. Light level and confocal laser scanning microscopy revealed that the lipid stores are distributed throughout the body in numerous oil droplets and not as a single oil sac, as seen in many marine calanoid copepods prior to overwintering (winter diapause). Transmission electron microscopy showed lipid spheres within the gut epithelium and large droplets of lipids stored extracellularly. Confocal laser scanning microscopy of copepods in pre-diapause, during diapause (encysted), post-diapause (recently excysted), and in reproductive condition, revealed that lipid stores are reduced following diapause, but are not totally absent. Analysis of lipid classes showed that H. nunni store predominantly wax esters/sterol esters (83% of total lipids) during diapause. The predominant lipid is most likely wax esters, as sterol esters typically are found only in small amounts in copepods. Fatty acid (FA) profiles of the copepods in diapause showed 16:0 to be most abundant followed by 16:1n-7 and 18:0; other FA occurred at concentrations <10% of total FA. Three polyunsaturated fatty acids (PUFA), 20:5n-3, 18:2n-6 and 20:4n-6, were found at concentrations <2% of total FA. These PUFA are "essential fatty acids" in H. nunni, obtained through dietary sources. The lipid classes and fatty acids present in H. nunni during diapause are compared to those of other copepods, some in a state of diapause and others not. It appears that lipid class and FA profiles are indicative of genetic makeup, type of diet or amount of food consumed prior to dormancy. The Caribbean reef-building corals Porites porites (Pallas) and Montastrea annularis (Ellis and Solander) and the Red Sea corals Pocillopora verrucosa (Ellis and Solander), Stylophora pistillata (Esper) and Goniastrea retiformis (Lamark) were analysed for total lipid, wax ester and triglyceride content, and fatty acid composition. M. annularis contained about 32% of dry weight as total lipid, whereas much lower values of between 11 and 17% were recorded for the other four species. It is concluded that there is greater variation in coral total lipid than hitherto thought. The total lipid contained a substantial proportion of wax ester (22 to 49%) and triglyceride (18 to 37%). The storage lipids (wax esters and triglycerides) accounted for between 6 and 20% of the dry weight and between 46 and 73% of the total lipid. Variation in lipid content between species could not be attributed to geographical location, but the low values for total lipid in Red Sea corals may in part be due to environmental factors as these samples were collected in winter. All corals analysed contained high levels of saturated fatty acids, the most abundant fatty acids being 16:0, 18:0 and 18:1n-9. Marked differences were observed in polyunsaturated fatty acid (PUFA) content between species, with comparatively low levels of 10 and 11% of fatty acids being recorded in M. annularis and G. retiformis, respectively. The values for the other species ranged between 21 and 37%. Fatty acid composition may vary according to the proportions of fatty acids obtained from diet, algal photosynthesis and synthesis by the animal tissues.  相似文献

The lipid/fatty acid composition of marine fish eggs and larvae is linked with buoyancy regulation, but our understanding of such processes is largely restricted to species with pelagic eggs. In this study, we examined developmental changes in the lipid/fatty acids of eggs and embryos of Pacific cod (Gadus macrocephalus), a species that spawns demersal eggs along coastal shelf edges, but as larvae must make a rapid transition to the upper reaches of the water column. Adult Pacific cod were collected in the Gulf of Alaska during the spawning season and eggs of two females were artificially fertilized with sperm from three males for each female. The eggs were subsequently reared in the laboratory to determine (1) how lipids/fatty acids were catabolized during egg and larval development, and (2) whether lipid/fatty acid catabolism had measurable effects on egg/embryo density. Eggs incubated at 4°C began hatching after 3-weeks and continued to hatch over a 10-day period, during which there was a distinct shift in lipid classes (phospholipids (PL), triacyglycerols (TAG), and sterols (ST)) and essential fatty acids (EFAs: 22:6n-3 (DHA), 20:5n-3 (EPA), and 20:4n-6 (AA)). In the egg stage, total lipid content steadily decreased during the first 60% of development, but just prior to hatch we observed an unexpected 2–3-fold lipid increase (~6–9 μg individual−1) and a significant drop in egg density. The increase in lipids was largely driven by PL, with evidence of long-chained fatty acid synthesis. Late-hatching larvae had progressively decreasing lipid and fatty acid reserves, suggesting a shift from lipogenesis to lipid catabolism with continued larval development. Egg density measures suggest that lipid/fatty acid composition is linked to buoyancy regulation as larvae shift from a demersal to a pelagic existence following hatch. The biochemical pathway by which Pacific cod are apparently able to synthesize EFAs is unknown, therefore representing a remarkable finding meriting further investigation.  相似文献

To discern species-specific patterns in omnivory indices in copepods from the North Water Polynya, lipid classes and fatty acids were determined in Calanus hyperboreus Krøyer copepodite stage 5 (CV), C. glacialis Jaschnov (CV) and Metridia longa Lubbock (females) sampled below the mixed layer during autumn 1999. Generally, M. longa contained higher proportions of triacylglycerols, polar lipids and 18:1(n-9) than the other species. M. longa also had lower relative amounts of wax esters (WE), polyunsaturated fatty acids (PUFA), 20:1(n-9), 22:1(n-11), and lower absolute (g lipid copepod-1) and relative (% lipid) lipid levels. Unsaturation coefficients (UC; ratio of polyunsaturated to total WE) were usually lowest in M. longa. These differences probably relate to a lesser dependence on phytoplankton in M. longa, and hence more ingestion of PUFA-poor prey. Moreover, levels of the carnivory index 18:1(n-9)/18:1(n-7) were highest in M. longa. Our data support the widespread contention that M. longa is more omnivorous than C. hyperboreus and C. glacialis. Proportions of bacterial fatty acids [odd-numbered and/or branched (OBFA); 18:1(n-7)] and PUFA/SFA (SFA: saturated fatty acids) ratios were largely unrelated to feeding strategy. Relationships between relative and absolute amounts of 16:1(n-7) and 18:1(n-7) in copepods suggest that the latter fatty acid is formed in vivo by chain elongation of the former. However, elevated levels of 18:1(n-7) and OBFA in M. longa at stations dominated by the microbial loop imply that these indices can be used to track ingestion of PUFA-poor protozoans when diatoms are scarce.Communicated by J.P. The total lipid and wax ester content as well as the fatty acid and alcohol composition of all copepodid stages and adults of Calanus finmarchicus s.l. were investigated at different locations in the North Sea in 1983 and 1984. Total lipid and the wax ester proportion increased exponentially until Copepodid V. The females were sometimes lower in lipids than the Stage V. The wax ester proportion reached about 90% of total lipids in males and Copepodid V and up to 40% in Copepodid I. The major fatty acids were 16:0, 20:5, and 22:6 and the major fatty alcohols were 16:0, 20:1 and 22:1. At one station the 18:4 acid became one of the dominant acids, because of a Phaeocystis sp. bloom, indicating that the fatty acids of the diet are incorporated mostly unchanged into the lipids of the copepods. The other main fatty acids 20:1 and 22:1 are probably synthesized de novo, serving as precursors for the principal alcohols 20:1 and 22:1. Their levels decreased in the younger stages due to increases in 16:0 alcohol. The fatty alcohol-forming enzyme seems to be specific for saturated and monounsaturated acids, which may be synthesized de novo or derived from diet.  相似文献

The variation in the concentration and fatty acid composition of lipid classes during the molting cycle of the prawn Penaeus japonicus was investigated. The lipid concentration of the whole body reached a maximum at mid-premolt (Stage D2) and then decreased to low level at late premolt (Stage D3–4). The accumulation of lipids during the premolt period seemed to be attributable to the increase of both polar and neutral lipids. The increase of neutral lipids at Stage D2 was derived from not only triglycerides but also free sterols and free fatty acids. Regarding the fatty acid composition of every lipid class, a marked variation occurred mainly at the intermolt (Stage C). In this stage, the polar lipids were rich in monoenoic acids such as 18:1 and poor in polyenoic acids such as 20:53 and 22:63. The triglycerides were rich in polyenoic acids at Stage C, but poor in monoenoic acids such as 16:1 and 18:1. The steryl esters contained large amounts of saturated acids such as 16:0 and 18:0 throughout the molting cycle, however the level of polyenoic acids increased at Stage C.  相似文献

Seasonal lipid dynamics of the copepod Pseudocalanus acuspes were studied in the Bornholm Basin (Central Baltic Sea) on a monthly basis from March 2002 until March 2003 and were interpreted in light of life cycle strategies and diet selection. The individual total lipid content of females ranged from 0.9 to 1.8 μg, with relative wax ester content reaching a significant maximum in May (44% of total lipids) and minimum (17% of total lipids) in April and November. Significant changes in size, lipid content, lipid classes and fatty acid composition of structural as well as storage lipids suggested five characteristic seasonal phases that were induced by different feeding histories and environmental conditions. Storage lipids were characterized by 18:1(n−9) as major component, which ranged between 44% of total fatty acids in June and 23% in February. The strong coherence between 18:1(n−9) in the seston lipids and the occurrence of ciliates emphasized the importance of ciliates in the diet of P. acuspes. As indicated by changes in the amounts of fatty acid markers, other food sources varied over the year, suggesting an opportunistic feeding behavior. The spring period was characterized by an increase in typical diatom and dinoflagellate markers, whereas other sources, potentially cyanobacteria, became more important during summer. The life cycle strategy is discussed with respect to extant adaptations to high latitudinal habitats.  相似文献

Fatty acids were extracted from the surface sediments (10 cm) of three sampling sites of Rufiji estuary to infer their sources and biogeochemical pathways. The fatty acids ranging from C8 to C24 were distinguished from this study, and were broadly classified into saturated (SFAs), monounsaturated (MUFAs) and polyunsaturated fatty acids (PUFAs). SFAs were found to be the major fraction at station 1 and 3 where as at station 2, MUFAs dominated. A total of 19 fatty acids with a total concentration of 64.91 μg/g dry weight were characterised at station 1. C16:0 was the most abundant fatty acid contributing 21.94% of total fatty acids (TFAs). C22:2 was the second most abundant, which accounted for 9.46% of TFAs. Fatty acids ranging from C12 to C24 were identified at station 2. C20 fatty acid was the most abundant fatty acid contributing 21.94%, followed by C16:0. At station 3, fatty acids ranging from C8 to C24 were obtained. The PUFA C20:5n-3 was the most abundant fatty acid contributing 21.65%, followed by C24:0 (15.00% of TFAs). The ratio of lower molecular weight (LMW) to higher molecular weight (HMW) biomarkers was used as an indicators to distinguish higher plants organic matter from algae-derived fatty acids.  相似文献

Developing eggs and larvae of laboratory-reared gilthead sea bream (Sparus aurata) maintained in filtered seawater (40 ppt) at 18°C, were measured for oxygen uptake, ammonia excretion, contents of free amino acids (FAA), protein, fatty acids (FA) accumulated ammonia, and volumes of yolk-sac and oil globule. Absorption of the yolk coincided with the consumption of FAA and was complete ca. 100 h post-fertilisation. Amino acids from protein were mobilised for energy in the last part of the yolk-sac stage. Absorption of the oil globule occurred primarily after hatching following yolk absorption, and correlated with catabolism of the FA neutral lipids. Overall, FAA appear to be a significant energy substrate during the egg stage (60 to 70%) while FA from neutral lipids derived from the oil globule are the main metabolic fuel after hatching (80 to 90%).  相似文献

The impact of supplementing lipid emulsions rich in eicosapentaenoic acid (EmEPA), docosahexaenoic acid (EmDHA) or saturated fatty acids (EmCOCO) to a standard algal diet [3:1 mixture of Isochrysis galbana (T-iso) and Chaetoceros neogracile, St-diet] on Argopecten purpuratus broodstock was evaluated. Broodstock fecundity was compared as well as the egg quality in terms of lipid content, fatty acid composition and lipid class distribution. Fecundity was defined as the number of eggs released in the spawning process, since spawning was virtually complete. Results indicated that the total lipid content of the eggs of A. purpuratus was diet independent. A greater energy reserve was spent on a larger number of oocytes and not on bigger sized oocytes with a higher lipid content. The lipids supplied through the emulsions were at least partially allocated to the eggs, demonstrating that the fatty acid composition of the eggs could be manipulated, especially the neutral lipid fraction. Levels of EPA changed more rapidly than DHA levels, supporting the observation that they fulfilled an energetic and structural role, respectively. The St-diet supplemented with 50%EmCOCO resulted in a significantly higher fecundity compared to the algal diet supplemented with 25%EmEPA+25%EmDHA and the non-supplemented algal diet. It would seem that saturated fatty acids (SAFA) were more easily or preferentially incorporated in the female gonads of A. purpuratus. The relative content of SAFA and 18:2( n-6) in these eggs rose significantly. The relative content of the highly unsaturated fatty acids, EPA and DHA, on the other hand was substantially lower in the neutral lipid fraction, but hardly affected in the polar lipid fraction. It appeared that the maintenance of an adequate DHA/EPA ratio (approximately 1.2) was more important than the absolute levels of the two fatty acids, as long as a threshold value was reached.  相似文献

A 6-week feeding trial was conducted with 44-d-old European sea bass (Dicentrarchus labrax L.) in order to examine the effect of various dietary phospholipid (PL) sources on the incorporation of n-3 highly unsaturated fatty acids (HUFA) in tissue lipids. From weaning onwards the fish received diets prepared by coating different lipid fractions (7.5% diet) on an extruded basal diet (92.5% diet). The two PL-free control diets contained 0.5 and 2% of an emulsifier blend, respectively. Seven other diets contained 2% PL, differing by their purity and origin (vegetable or animal). All diets were rendered isolipidic by the addition of hydrogenated coconut oil. Feeding the PL-supplemented diets, except the diet containing hydrolyzed soybean PL (lyso PL), resulted in a higher survival and a 10 to 30% better growth as compared to the PL-free diets. No difference according to the PL origin was observed. The sea bass final lipid content increased with increasing body weight. Also the lipid class composition of the fish was clearly correlated with the final weight gain. Total neutral lipid increased from 51% of total lipid (initial fish) to 76% for fish fed the PL-free diets, and up to 88% for fish fed the sunflower PL. Weaning the fish on the experimental diets induced important changes in their fatty acid profiles characterized by a decrease in 18:3n-3, 20:5n-3 and 20:4n-6 and an increase in saturated fatty acids and 22:6n-3 (DHA). According to the fatty acid composition of both total and polar lipid, the weaned fish could be divided into three groups reflecting the dietary fatty acids: a group fed the vegetable PL, a group fed the animal PL and a PL-deprived group. An effect of dietary PL on the incorporation of dietary n-3 HUFA, more particularly DHA, was noticed. For a similar supply of DHA through the neutral lipids in the diet, fish fed PL-supplemented diets (except for the lyso PL diet) had 10 to 25% higher DHA levels in total and polar lipid than PL-deprived fish. This PL effect was already clear at the end of the weaning and was not related to the presence of n-3 HUFA in the PL source, as suspected in a previous study when feeding egg yolk PL. A better incorporation of DHA was not obtained by replacing the PL by an emulsifier or by lyso PL with higher emulsifying properties. Aquaculture studies have revealed that polyunsaturated fatty acids are critical for maintaining substantial growth, survival and reproductive rates, and high food conversion efficiencies for a wide variety of marine and freshwater organisms. The aim of this study was to investigate the gross biochemical and fatty acid composition of both neutral and polar lipid compartments of the razor clam Solen marginatus throughout embryonic and larval development. High levels of stored reserves in S. marginatus eggs allow a short larval development, lasting only 8 days. The energy required for embryogenesis was obtained from stored proteins. During larval development from D-shaped veliger until settlement, protein, lipid, and carbohydrate reserves were indistinctly stored for metamorphosis. Although total lipids increased, fatty acids in both neutral and polar lipids decreased during embryonic development. The depots allow a short larval development in which settlement is reached with lower amounts of stored neutral and polar lipids than the contents found in the oocytes. Non-methylene-interrupted dienoic fatty acid levels were similar to those of some polyunsaturated fatty acids, with increasing percentages at the onset of metamorphosis. This study indicates that S. marginatus exhibits a different pattern in the use of gross biochemical and fatty acid reserves during larval development compared to other razor clam and bivalve species, mainly due to the large size of its eggs and the short larval development stage reported in this species.  相似文献

Summary Pseudocrenilabrus multicolor is an iteroparous maternal mouthbrooding cichlid fish living in small and shallow waters in East Africa. It is subjected to large seasonal variations of population density. Laboratory experiments show that with increasing density and time after spawning the number of offspring present in the mouth decreased. By experimentally varying density and the numbers of fertilized and unfertilized eggs in the mouth at the end of spawning the following causes of brood size reduction were identified: (1) At high densities some of the spawned eggs are stolen by conspecifics before they are taken into the mouth. Of the eggs taken up a considerable proportion is unfertilized. (2) Unfertilized eggs are selectively swallowed from day 2 to day 5 after spawning. (3) Some of the fertilized eggs are also swallowed during mouthbrooding. (4) If the number of eggs in the mouth falls below a critical value of approximately 20% of the number of eggs spawned, they are all swallowed. By contrast, predation by conspecifics is insignificant as long as the brood is in the mother's mouth. The causes and functions of the mother's filial cannibalism are discussed.  相似文献

The lipid composition of tropical marine reef fishes is poorly known, despite their use as food by local human populations and recent interest in health-related benefits of fish lipids. We examined the composition of lipids from epaxial muscle, liver, and two storage sites [mesenteries surrounding the gut (intraperitoneal fat, IPFs) and retroperitoneal fat bodies (FBs) posterior to the peritoneal cavity] in three species of surgeonfishes from Ishigaki Island, Japan: Naso lituratus (Bloch and Schneider, 1801), Acanthurus lineatus (Linnaeus, 1758), and A. bariene (Lesson, 1830). Triacylglycerols dominated all samples of neutral lipid and constituted ≥ 99% of FBs and IPFs. Polar lipids generally contained large fractions of phosphatidylethanolamine and phosphatidylcholine. Quantified fatty acids ranged in length from C14 to C24. C16 fatty acids prevailed (>35% of neutral fatty acids, >23% of polar fatty acids), although C18 (>16 and >14%, respectively) and C20 acids (>8 and >19%, respectively) were also common. Saturated fatty acids, dominated by palmitic acid (16:0), comprised 38.7 to 50.7% of acids from neutral lipids and 30.8 to 41.1% from polar lipids. The most common monounsaturated acids were 18:1n9 and 20:1n9. Polyunsaturated acids were prevalent in polar lipids (especially 20:4n6, 20:5n3, 22:2n3, 22:5n3, 22:5n6 and 22:6n3). Common polyunsaturated acids of neutral lipids were 18:2n6, 18:4n3, several n-3 and n-6 C20 acids, 22:2n3 and 22:5n3. IPF and FB were almost identical across species, and lipids of fat bodies (IPFs, FBs) were more similar to those of muscle than those of liver for all three species. The FBs appear to constitute an accessory storage site, which overcomes constraints on lipid storage imposed by a small, inflexible abdominal cavity that contains both viscera and consistently voluminous gut contents. Fatty acid signatures indicate that largely overlooked epiphytic or epilithic diatoms contribute significantly to lipid acquisition. The combination of large quantities of both saturated and n-3 and other polyunsaturated fatty acids in surgeonfishes, in contrast to low saturates and high polyunsaturated acids in lipids of commercially important cool-water fishes, suggests that a study of dietary effects of fish lipids on human inhabitants of the tropics may be instructive insofar as human health and nutrition are concerned. We tested the usefulness of the fatty acid signature-method in investigating the diet of seabirds in conjunction with the conventional technique of stomach-content analysis. We compared the fatty acid composition of subcutaneous white adipose tissue (SWAT) of king penguin chicks (Aptenodytes patagonicus) during fattening periods to that of total lipids from their food. In both spring and autumn, the fatty acid composition of chick SWAT was identical to that of the dietary lipids. Because the diet of adult king penguins feeding for self-maintenance (i.e. not for their chicks) was essentially unknown, we subsequently analysed their SWAT fatty acid patterns after premolting and prebreeding foraging trips (during which they build up large energy reserves). The fatty acid composition of SWAT from adults was identical to that of chick adipose tissue and food. King penguin diet and SWAT were characterized by high levels of very long-chain mono-unsaturated fatty acids (20 to 24 carbon atoms, 16 to 23% by mass) and (n-3) poly-unsaturated fatty acids (19 to 27%); these consisted mainly of 20:1n-9 (5 to 8%) and 22:1n-11 (5 to 8%), and 22:6n-3 (10 to 13%) and 20:5n-3 (3 to 9%), respectively. Prey items identified from chick stomach contents indicated that the bulk of the food was oceanic myctophid fishes, mainly Electrona carlbergi, Krefftichthys anderssoni and Protomyctophum tenisoni. The fatty acid composition of four other species of myctophid fishes was similar to that of penguin diet and SWAT, but markedly different from that measured for a squid species and that reported for crustaceans. These findings indicate that adult king penguins prey on myctophid fish not only to feed their chicks but also for their own nutrition. The fatty acid signature-technique is therefore a reliable method to gain information on the food and feeding ecology of seabirds when more conventional techniques are of limited value. Such information is important to the understanding of trophic relationships between key species of the ecosystems, and also to provide insight into the nature of avian adaptations to the marine environment. The distribution of n-3 highly unsaturated fatty acids (HUFA) over the major neutral and polar lipid classes was determined for two predominant types of live food used in the larviculture of marine fish and shrimp, i.e. freshly hatched and HUFA-enriched Artemia, and compared with data reported in the literature for wild copepods, representing the natural diet of these larvae. Lipid class composition and their content of n-3 HUFA, particularly eicosapentaenoic acid (EPA, 20:5n-3) and docosahexaenoic acid (DHA, 22:6n-3), were assessed in freshly hatched, HUFA-enriched and subsequently starved Artemia franciscana. The n-3 HUFA enrichment was based on feeding Artemia a lipid emulsion in which either fatty acid ethyl esters (EE, diluted with olive oil) or triacylglycerol (TAG) provided a level of 30% n-3 HUFA. Enrichment of Artemia with either type of the lipid emulsions resulted in an increase of total lipid content from 20.0 to 28.2–28.7% of dry matter mainly due to the accumulation of neutral lipid, primarily TAG (from 82 to 158 mg g−1 dry wt in freshly hatched and 24-h enriched Artemia). Enriched brine shrimp utilized up to 27–30% of their TAG content during 72 h of starvation at 12 °C. The absolute tissue concentrations of polar lipids remained constant at 71 to 79 mg g−1 dry wt throughout the enrichment and subsequent starvation. The level of n-3 HUFA increased drastically during enrichment from 6.3% of total fatty acids (8.2 mg g−1 dry wt) in freshly hatched nauplii to between 20.4 and 21.8% (40.4 to 43.2 mg g−1 dry wt) in 24-h enriched Artemia and was not significantly affected by the source of n-3 HUFA. During starvation, 18:0, 20:4n-6 and 20:5n-3 were retained, whereas 18:4n-3, 22:5n-3 and 22:6n-3 were specifically catabolized. The major polar lipids, phosphatidylethanolamine (PE) and phosphatidylcholine (PC), of freshly hatched Artemia showed very low levels of DHA (<0.1% of total fatty acids) and carried about 45% of the total EPA present. Enrichment with either of the emulsions resulted in an increase of the neutral lipid fraction which concentrated >64% of the EPA and >91% of the total DHA present. This is in sharp contrast with the high levels of n-3 HUFA, in particular DHA, in the polar lipid fraction reported for wild copepods. The contrasting distribution of DHA in the neutral and polar lipid fractions of enriched brine shrimp compared to the natural diet may influence the efficacy of this essential fatty acid for marine fish larvae in aquaculture systems. The present work is a comprehensive study of reproduction and embryonic development of Armases cinereum. Ovigerous A. cinereum (Bosc, 1802) females from Sebastian Inlet, Florida (9.88–19.4 mm CW) lay 2,000–12,000 eggs per brood, depending on their CW (mm): fecundity = 24.662 CW1.9432. A. cinereum displayed significant brood loss through development (ca. 500 eggs per brood) independently from their CW (no senescence). However, since smaller females lay fewer eggs than larger ones, the percentage of eggs lost during embryonic development is greater in smaller females. The number of eggs carried on a later stage of development (potential fertility = 5.5593 CW2.4417) is a more accurate estimate of the reproductive output and subsequent recruitment. Egg volume increased during development (64%, 0.025–0.041 mm3 or 0.36–0.43 mm of diameter, N = 270) and was strongly correlated with egg water content increase (19.21%, r = 0.89). Lipids, particularly fatty acids, seem to be the major energy source for embryonic development, decreasing 56.31 and 37.08% (respectively) during embryonic development; both are negatively correlated with egg volume (r = −0.90). The utilization of fatty acids through the different developmental stages of A. cinereum is presented. The most consumed fatty acids are the monounsatured (43.33 μg mg−1 dw), followed by the saturated (29.91 μg mg−1 dw) and polyunsaturated (24.03 μg mg−1). Palmitic (16:0) and linoleic (18:2n-6) acids are preferentially consumed (19.5 and 17.9 μg mg−1 dw, respectively). The high proportion of essential polyunsaturated fatty acids of C18 and C20 reflects the consumption of primary producers such as mangrove leaves. EPA/DHA ratio (2.85–3.84) and low DHA content indicated that this species appears in a medium-low level of the trophic chain. The low ratio of 18:1n-7/18:1n-9 and high percentage of 18:1n-9 (marker of carnivory) may be a sign of the consumption of juvenile invertebrates. The high percentage of odd-numbered FA indicated the occurrence of detritivores/scavenger behaviours. The fatty acid composition of the eggs reflects adult feeding ecology (omnivorous) and habitat.  相似文献

This paper presents quantitative data for the changes in the contents of total lipids, lipid classes and their associated fatty acids, together with the changes in caloric contents of developing eggs and yolk-sac larvae of Atlantic cod (Gadus morhua L.). During development between Day 0 to 28 post fertilisation, 32% of catabolic metabolism was fuelled by lipids. On a mass-specific basis, polar lipids (mainly phosphatidyl choline) contributed 60%, and neutral lipids (mainly triacyl-glycerol) contributed 40% to this catabolic component, with each class supplying similar amounts of fatty acids as fuel. The fatty acids supplied by phosphatidyl choline were catabolised non-selectively (i.e. in proportion to their presence in the egg), with about half of them being polyunsaturated. However, of the fatty acids esterified in triacylglycerol, the larvae showed an apparent oxidation preference for monoenes over polyunsaturates or saturates. Routine rates of oxygen consumption and ammonia production were related to the caloric contents of the eggs and larvae in order to derive an enthalpy balance equation (of the form P=C+R+E) for an Atlantic cod larva during its period of endogenous nutrition. For the interval of Day 0 to 25 post fertilisation (the period of yolk dependence), integration of the physiological and caloric data revealed that Atlantic cod larvae conserved 53% of yolk enthalpy (C) for growth (P), 42% was dissipated due to metabolism (R) while only 5% was lost via excretion (E).  相似文献
