Oogenesis in the marine mussel Mytilus edulis: an ultrastructural study   总被引:1,自引:0,他引:1

Ultrastructural changes occurring during the course of development in oocytes of Mytilus edulis are described for mussels collected at monthly intervals over a period of one year (September 1981 to October 1982) from a site in Cornwall, England. During early stages of oogenesis the oocyte is surrounded by a small number of follicle cells but, as development proceeds, the follicle cells are restricted to the stalk region which attaches the oocyte to the acinar wall. Contact between the follicle cells and the developing oocyte is maintained by means of desmosomelike gap junctions. Organelles and inclusion bodies present in the ooplasm during oogenesis include rough endoplasmic reticulum (RER), Golgi bodies, mitochondria, free ribosomes, Balbiani's vitelline body, annulate lamellae and yolk and cortical granules. The RER, in particular, varies considerably throughout the course of development. Evidence for uptake of exogenous macromolecules into oocytes by pinocytosis is presented; it occurs in the basal region of previtellogenic oocytes prior to the formation of the vittelline coat. Lipid-yolk granules invariably have mitochondria in close association and, during the winter months, develop in close proximity to small, apparently glycogen-rich vesicles possibly suggesting that conversion of glycogen to lipid takes place in developing oocytes. Oocyte degeneration was commonly observed and involves initial breakdown of the plasma membrane followed by rupture of the vitelline coat. The oocyte contents once released into the acinar lumen are resorbed by the epithelial cells of the gonoducts, which are prevalent throughout the mantle of ripe individuals.  相似文献

Histological examinations of the gonads of Promachocrinus kerguelensis Carpenter, 1888 collected in November 1984 from McMurdo Sound, Antarctica, revealed ripe ovaries containing three distinct size classes of oocytes: 20 to 90, 91 to 150 and 151 to 180 m diameter as well as ova. Testes were ripe, and filled with mature sperms. There was marked inter-pinnule and inter-individual synchrony of gametogenesis and spawning is probably synchronous, at least within the sub-population sampled. The number of eggs per pinnule is allometrically related to body size. Body weight and arm length are positively correlated with total egg number. Annual fecundity was estimated to be 29 000 eggs (full-grown oocytes and ova) for a crinoid weighing 53.6 g wet weight with an arm length of 16.3 cm. The eggs float. Female genital pinnules contain more lipid and energy than male pinnules. Reproductive output and effort (23.4 to 65.2 kJ and 26 to 38%, respectively) increase with body size and are similar to values reported for other echinoderms with lecithotrophic larvae. The greatest overall energetic contribution is represented by the arms and genital pinnules. Spawning in P. kerguelensis probably occurs in November and December within McMurdo Sound. If development is slow, as in all other antarctic echinoderms studied to date, then settlement most likely occurs 2 to 3 mo later, after the short summer period of high phytoplankton productivity. High fecundity, a pelagic mode of reproduction, and the high probability of a slow rate of development may contribute to the wide geographic distribution of this common antarctic crinoid.  相似文献

InMytilus edulis L., gametogenesis takes place in the mantle at the expense of the connective storage tissue. There are two main types of storage cells: vesicular (VC) cells storing large amounts of glycogen and adipogranular (ADG) cells containing large numbers of protein granules, lipid droplets and lesser amounts of glycogen. One of the ways in which stored reserves can be mobilized for gamete formation is by controlled autophagy, in which the cellular constituents are degraded by lysosomes. Mussels were collected from the Menai Strait, North Wales, and monthly measurements made, over two years (1984–1986), of the activities of lysosomal acid hydrolases (acid phosphatase,-N-acetyl-D-glucosaminidase and-glucuronidase) and Cathepsins B and L in the mantle tissue, isolated ADG cells, low-density cells and, during spawning, in the mature oocytes of female mussels. The lysosomal proteinases, Cathepsins B, L and H, were further characterised by activation with thiol compounds and inhibition with thiol blockers and by leupeptin. Because of the low activity in the mantle tissue ofM. edulis, Cathepsin H was not assayed on a seasonal basis. There was a general increase in lysosomal enzyme activity during the winter, which can be related to increased autolysis in the storage cells and to the process of maturation in the developing oocytes. The activity of Cathepsin B was highest in the ADG and low-density cells, implying an important role in proteolysis within the ADG cells. By contrast, Cathepsin L displayed the highest activity in the mature oocytes, suggesting a major function of Cathepsin L in the development and maturation of the oocytes. Two different-glucosidase activities were measured in the monthly assays, one with a pH optimum of 4.5 (acid) and the other at pH 7.5 (neutral). Highest activities of the acid-glycosidase were found in the low-density cells, but there were no significant seasonal changes in the mantle tissue as a whole. Activities of the neutral-glucosidase were low in the ADG cells and mature oocytes, but showed high activities in the mantle tissue, with marked seasonal changes that corresponded to the mobilization of glycogen reserves in the VC cells.  相似文献

The reproductive cycles of two populations of the Recent articulate brachiopodTerebratulina retusa (Linnaeus) are described, based on samples taken between October 1985 and October 1986. Stereological analysis of the gonads revealed a single synchronised spawning event between late November and the end of January in the Firth of Lorn. In a second population, in Loch Fyne, spawning took place repeatedly throughout the spring and summer, with greatest spawning activity occurring in the late autumn.T. retusa from Loch Fyne were approximately three times more fecund than those from the Firth of Lorn. Productivity in the Clyde Sea area is approximately five times higher than in Firth of Lorn and higher fecundities and spawning frequencies of the Loch Fyne population are attributed, in part, to greater food availability. The initiation of gametogenesis appears to be mediated by the mobilisation of reserves stored in the outer mantle epithelium during the winter. Primary productivity increases dramatically in the spring and gametogenesis shows a corresponding increase.  相似文献

The euryalinid snake star Astrobrachion constrictum (Farquhar), a normally deep-water species, exists within SCUBA diving range in New Zealand flords, and thus presents a unique opportunity for study. A population of A. constrictum in Doubtful Sound was investigated from February 1990 to July 1994 to determine the reproductive cycle. Unlike most ophiuroids, the gonads of A. constrictum are positioned along the aboral coelom in the proximal portion of the arms. The proximal 2 to 3 cm of the arms of individuals were thus removed on an approximately monthly basis for histological examination of the gonads. Image analysis was used to determine oocyte size-frequency distributions. A gonad index was calculated for each individual based on the ratio of cross-sectional area of the gonads to cross-sectional area of the arm. Maturity indices were calculated based on stage of development of the gonads of each individual. A. constrictum has a period of enhanced breeding activity in April–June, after which both the gonad and maturity indices decline. The increase in gametogenic activity during these months was correlated with rising sea temperature within the fiord. Although the females showed an annual pattern of gonad development, oogenesis was continuous throughout the year, with oocytes at different stages of development present in the gonads. Males also showed continuous spermatogenesis and, although the same annual pattern of gonad development was evident as seen in the females, it was less marked. The presence of females with large oocytes and ripe males throughout the study indicates that breeding in the Doubtful Sound population is prolonged, and possibly continuous, as found in many deep-sea ophiuroids.  相似文献

The present study investigated the sexual pattern of the yellow-tailed dascyllus, Dascyllus flavicaudus (Randall and Allen), through histological examination of the process of gonadal differentiation and maturation and through male-removal experiments in the laboratory. The study was conducted in Moorea, French Polynesia (17°32S; 149°50W), from 31 December 1994 to 11 March 1995. For gonadal histology, two populations were sampled, one consisting of large aggregations (n=54) and the other consisting of small- to medium-sized groups around isolated corals (n=55). An additional small sample (n=21) was also collected from a population that consisted of small groups around isolated corals. After an initially undifferentiated state, gonads of D. flavicaudus developed an ovarian lumen, followed by oocytes in the primary growth stage. From this ovarian state or from more developed ovaries with cortical-alveolus stage oocytes, some gonads developed into testes through degeneration of oocytes and development of spermatogenic tissue. In all three populations, spermatogenic tissue developed only in gonads that contained pre-vitellogenic oocytes (termed as mixed stage 1–2 gonads). The two main populations did not differ in their expression of sex change despite differences in their social organization. In both populations, size of individuals with mixed stage 1–2 gonads overlapped mainly with the size range of immature females, which suggests that functional female-to-male sex change was rare. The hypothesis that D. flavicaudus is primarily gonochoristic is further supported by removal experiments in laboratory groups, in which removal of a dominant male(s) failed to induce sex change in any of the high-ranking females. Yet, in all three field populations, some fish with mixed stage 1–2 gonads were found within the size ranges of mature females, which indicates possible occurrences of functional sex change. These conflicting results indicate that it may be premature to draw any definitive conclusions about the sexual pattern of D. flavicaudus.Communicated by T. Ikeda, Hakodate  相似文献

A consistent diploid number of 2n = 36 was determined for the sea urchinParacentrotus lividus from the Gulf of Palermo by analysis of mitotic chromosomes of both early developing embryos and male gonads. The haploid numbern = 18 was determined by counts of spermatocyte bivalents at diakinesis. A heteromorphic chromosome sex mechanism of the XY type is likely present in this species. This is indicated by the occurrence of a chromosomal pair, pair No. 2, which is heteromorphic in both morphology and size in about 50% of the mitotic figures (metaphases and anaphases) of einbryos. In addition, heteromorphism of the same pair of chromosomes occurred during spermatogonial metaphases in the five male specimens investigated here. The detection of a low chromosome number (2n = 36) compared to other echinoids (2n = 42 to 44), a heteromorphic chromosome sex system and the involvement of three chromosome pairs in nucleolar organization (NORs) provide evidence of the specialization of theP. lividus karyotype.  相似文献

Monthly samples of gonads of 3 common species of intertidal trochids, Monodonta lineata (da Costa), Gibbula umbilicalis (da Costa) and G. cineraria (L.) were fixed for examination by light microscopy. Numbers and proportions of immature and mature oocytes in sections of ovaries were determined. In M. lineata and G. umbilicalis, the annual reproductive cycles were very similar, the testis and ovary of both species being empty brown structures from September to January/February, when gametogenesis began and the gonads began to swell and turn green in both sexes. As ripening continued, the ovary remained green in G. umbilicalis, but in M. lineata turned greygreen by the end of May. The testis of both species changed, to yellowish in G. umbilicalis and creamy-pink in M. lineata. Spawning occurred throughout summer and early autumn, and the gonads reverted to their former empty state. These changes were matched by the increase and subsequent decrease in proportion of ripe oocytes in the gonad. Some ripe oocytes are not shed during the spawning season, but no evidence of cytolysis of such oocytes was found. In G. cineraria, in contrast, the ovary remained green and swollen and the testis remained creamy-white and packed with sperm throughout the year. The proportion of ripe to unripe oocytes in the ovary fluctuated for the entire sampling period, and did not indicate the time of spawning. In the testis of all three species, spermatogonia were rarely seen, and spermatid stages were uncommon.  相似文献

Potential fecundity in Loligo vulgaris reynaudii was estimated to be about 17 000 eggs, calculated as the total number of discernible oocytes in the ovary and oviduct. Squid were observed to spawn up to 8140 eggs over a 36 h period in captivity. First estimations of actual fecundity are therefore between 8000 and 17 000 eggs. Factors complicating a more accurate estimation of actual fecundity in this species include difficulties with aquarium maintenance, their behaviour of spawning over a protracted period and in multiple sites, and atretic oocytes observed in both developing and mature ovaries. Detailed morphological and histological analysis of gonads collected at regular intervals over a complete spawning season will allow a more precise calculation of actual fecundity. The lipid content and fatty acid composition of the small common wrasse Symphodus ocellatus was analysed within two populations located in very different biotopes on the French shore: a Caulerpa taxifolia meadow located in Cap Martin and a Posidonia oceanica bed located in Villefranche. Polar lipids represented 82–90% of the total lipid in adult female livers, gonads and in eggs. Cholesterol in gonads and eggs and triacylglycerols in livers were the dominant neutral lipids. Differences in lipid classes were found in livers between the two populations, but not in their gonads and eggs. Moreover, a quantitative difference in total lipid content was evident. Young fish living on the C. taxifolia meadow in winter had 21% less lipid than those living on the P. oceanica bed. The populations of S. ocellatus showed differences between triacylglycerol versus phospholipid fatty acid compositions and between gonad versus liver fatty acid compositions. Moreover, a significant difference was found between both populations in their liver triacylglycerol fatty acid compositions, suggesting a difference in diet. Gut content analysis supported this hypothesis as it showed important differences in the ingested preys between the two populations in January (planktonic vs. benthic, frequency of isopods) and in June (frequency of gastropods). Thus, we found that the fish populations from Villefranche and Cap Martin showed significant differences in body lipids due to different nutritional habits.Communicated by S.A. An account is given about the development of the gametes of the holopelagic coronate scyphomedusa Periphylla periphylla (Péron and Lesueur 1809). The gonads of the species are complex and differ from those thus far described in Scyphozoa in having this characteristic composition of trophocytes, follicle cells, gamete-releasing pores, mucus cells, and resorption cells. Our results differ from those of previous coronate studies with respect to the contact of the oocytes with the gonad tissue throughout the duration of development. Among the Medusozoa, follicle cells have thus far been considered as an apomorphy for the Staurozoa, but their presence in the Scyphozoa casts some doubts on this assumption. From morphological structure and examination of the gonads and gametes, it can be deduced that P. periphylla has true organs and that gamete release is continuous. These results supplement our knowledge of the reproductive biology of this mesopelagic species.  相似文献

The sexuality, reproductive mode, and timing of reproduction of Pocillopora verrucosa from the Republic of Maldives, Indian Ocean, were assessed using serial histological sections. These showed that P. verrucosa is an annual simultaneous hermaphrodite, with gonads arranged in two opposing arcs of alternating testes and ovaries, six gonads in each arc. No planulae were observed in any dissection or histological analysis carried out, therefore making brooding unlikely. Broadcast spawning is inferred from the disappearance of mature gametes from samples collected between late March and April 1991. Mean oocyte size at spawning was 53.5 m and mean potential fecundity was 7300 ooctyes cm-2yr-1. The reproductive pattern of P. verrucosa in the Maldives is compared to that of the same species in different locations, the short breeding season in March to April occurring earlier in the Maldives than in Red Sea populations. The maximum mean oocyte diameter found in Maldivian specimens was much smaller than elsewhere. The year-to-year variation in numbers of oocytes/polyp within single colonies, and the variation between colonies within the population was significant (p<0.01). Therefore, it is possible that fecundity is not a good variable to use when monitoring stress on coral reefs unless larger numbers of estimates can be routinely made. Some colonies contained immature oocytes at spawning that were not released and continued to grow to approximately twice the size of the spawned oocytes that were presumed to be mature. These unspawned oocytes were oosorbing, and were characterised by the presence of zooxanthellae and large numbers of vacuoles in their cytoplasm. This gradual expansion and oosorption of unspawned oocytes has not been documented previously for corals.  相似文献

The majority of published accounts on scleractinian coral reproduction are from the tropical Pacific and Caribbean, with very little information known about Red Sea species. This report examines variation in reproductive mode in 24 species of hermatypic corals (belonging to seven families) in the Gulf of Eilat, Red Sea. Eighteen species are hermaphroditic broadcasters, two are hermaphroditic brooders and three are gonochoric broadcasters. In the Pocilloporidae, the gonads project into the body cavity, while in the other six families the gonads reside inside the mesenteries. The number of gonads per polyp in broadcasting species follows family or genus lines. Fecundity (eggs per polyp) increases with polyp size. Brooding species usually exhibit one or two gonads per polyp and each gonad contains only one to three oocytes. Oocyte size varies widely and does not relate to mode of reproduction. The largest oocytes (diameter = 450 μm) occur in the brooding coral Alveopora daedalea and in broadcasting species of the genus Acropora (diameter = 420 μm). Gonad morphology and gonochorism versus hermaphroditism appear to be constrained phylogenetically at the family or genus level. Lastly, this report compares the data presented for Red Sea scleractinian species with the data available on scleractinian corals from other geographical regions. Reproduction of two populations of Ophionereis schayeri (Müller and Troschel) in New South Wales was investigated from November 1991 through January 1993. The reproductive cycle was documented by histological examination of the gonads and by image analysis to determine the oocyte size-frequency distributions. Both populations of O. schayeri were mature in December and January and had a major summer spawning period between January and February. Thereafter, the condition of the gonads exhibited interindividual variability, with asynchronous low-intensity gamete release through August. O. schayeri has the potential to spawn for up to 8 mo of the year. This breeding pattern with synchronous spawning in summer and asynchronous gamete release through winter was similar in both populations. During autumn and winter, the gonads contained developing gametes and advanced gametes ready for spawning. The eggs spawned by O. schayeri during these seasons started their growth during the previous spring vitellogenic period, and continued to grow through summer. Upon reaching full size, they were stored for spawning outside the main breeding period. Spring is marked by increased spermatogenic and vitellogenic activity followed by maturation and spawning. Throughout their development, the oocytes of O. schayeri are surrounded by a follicle consisting of periodic acid-Schiff-positive (PAS+) haemal fluid and PAS+ yolk by the oocytes. In the testes, the haemal sinus projects into the centre of the spermatocyte columns, thus appearing to play a nutritive role in support of both oogenesis and spermatogenesis. Late vitellogenic oocytes of O. schayeri are firmly anchored to the germinal epithelium by an attachment complex consisting of specialised attachment cells and basophilic strands that radiate between the oolemma and the nucleus. Spawning is associated with rupture of the follicles, which remain as prominent, empty U-shaped profiles in the ovaries. O. schayeri produces copious numbers of 200 m-diam oocytes, suggesting that this species is a broadcast spawner and that it has a modified ophiopluteus or vitellaria larva.  相似文献

Reproduction of the commercial sea urchinParacentrotus lividus (Lamarck) from contrasting habitats on the west coast of Ireland was examined from May 1986 through August 1988. Urchins were collected intertidally from an exposed rocky shore and subtidally from a protected bay. Monthly measurements of the gonad index and histological examination of the gonads demonstrated thatP. lividus has an annual reproductive cycle. Although the two populations exhibited similar reproductive patterns over three breeding seasons, there were inter-annual and inter-population differences in the amplitude of gonadal growth and in the time when spawning started. The subtidal urchins had significantly larger gonads and exhibited a longer period of reproductive maturity compared with the intertidal urchins. This difference was also evident in the histology of the ovaries at the beginning of breeding, when most of the subtidal females contained mature ovaries, whereas most of the intertidal females contained premature ovaries. An inter-annual difference in the onset of spawning was observed, with the start of gamete release differing by as much as four weeks between years. It appears that inter-annual differences in sea temperature influence the temporal variation in spawning byP. lividus and that increasing temperature may serve as an exogenous cue for gamete release. The inter-annual variability in the onset of spawning suggests that photoperiod does not cue gamete release. Gonadal growth, on the other hand, occurs during the coldest months of the year and during the period of shortest days, suggesting that temperature and photoperiod may both influence gonadal growth during the winter. Oogenesis and spermatogenesis ofP. lividus were examined histologically. During the post-spawning recovery and growth stages, the gonads gained weight through growth of the nutritive phagocytes and the accumulation of periodic acid Schiff (PAS) + droplets by these cells. The PAS + material appears to play a nutritive role in gametogenesis. For the females, the frequencies of six ovarian maturity stages was assessed at approximately monthly intervals. Small oocytes were present throughout the year and clusters of early oocytes were most apparent during the spent and recovery stages. With the onset of vitellogenesis and subsequent accumulation of ova, the nutritive phagocytes and their PAS + droplets became depleted. The ovarian condition at the onset of breeding was variable, due to differences in the number of vitellogenic oocytes, differences in the number of ova in storage, and differences in the amount of PAS + material. In general, the nutritive phagocyte tissue is reduced by the onset of spawning and is exhausted by the termination of breeding. A similar series of events occurs during spermatogenesis. The relevance of this study for the use ofP. lividus as a brood-stock organism for mariculture is discussed.  相似文献

Heavy metal concentrations (Cd, Co, Cr, Cu, Fe, Mn, Ni, Pb and Zn) were determined in various tissues (hepatopancreas, branchial hearts, salivary gland, gills, genital tract, mantle, arms and skin) of Octopus vulgaris collected from three different contaminated sites in front of Alexandria (Egypt) during 2000. All collected tissues displayed high enrichment factors when compared to ambient levels. Heavy metal concentrations in most tissues displayed significant differences among sites, sizes and sex. This study suggests that hepatopancreas, and to a lesser extent branchial hearts, are better indicators of chronic Cu, Fe, Zn and Cd contamination than edible tissues. The enrichment factor (EF) for heavy metals in the hepatopancreas and in edible tissues allowed discriminating our samples into three main groups; (1) EF?>?55 (Cu), (2) EF ranging from 15 to 7.5 (Fe, Cd and Zn) and (3) EF?相似文献

We suggest that gonad development in the mantle tissue of the bivalve molluscMytilus galloprovincialis Lmk. is an example of epithelial/mesenchymal interactions (i.e. soma/germline interactions) and morphogenesis in the adult state. According to this concept, the aim of the present study was to use biochemical and immunochemical methods for identifying and characterizing the mantle cell polypeptide markers whose expression is seasonally and morphogenetically regulated. We showed for the first time thatM. galloprovincialis mantle, of both males and females, contains polypeptides (with an apparent mol. wt of 45 to 53 kDa) specific for connective tissue (mantle connective tissue polypeptides; MCTPs). Electrophoretic, immunoblotting and immunofluorescent experiments demonstrated that MCTPs are primarily localized in the adipogranular (ADG) cells, and their expression in the mantle is seasonally regulated. There is a positive correlation between MCTP expression and connective tissue volume in the mantle. MCTPs are overexpressed during the rest period, when the mantle consists of connective tissue mainly, whereas mature gonads contain only trace amounts of MCTPs. Moreover, there is a temporal correlation between the onset and decrease of MCTP expression and the appearance and disappearance of the ADG cells in the mantle. MCTP localization in the mantle tissue should not be associated with the ADG cells only, because positive immunofluorescence was also detected in follicle membranes (but not in germ cells) and superficial mantle epithelium. Using immunoblotting and immunofluorescence, MCTPs were found in the connective tissue of the mantle, posterior adductor muscle and visceral mass, but not gills, foot or hepatopancreas. Possible mechanisms by which MCTPs could participate in the annual processes of mantle gonad/connective tissue development and involution are discussed.  相似文献

Samples of the deep-sea spinulosan asteroid Hymenaster membranaceus Wyville Thomson were collected in a timeseries of 19 bottom trawls spanning the period April 1978 to October 1981 from a 2200 m-deep station in the northern Rockall Trough. The reproductive biology of this species was studied from histological sections of the gonad, and compared with that of H. gennaeus H. L. Clark which was collected in the same hauls. At first sexual development, oogonia develop in nests surrounded by small accessory cells. Previtellogenic oocytes remain in the periphery but, at maturity, oocytes ranging up to 1 100 m fill the ovary. A variety of accessory cells pack the lumen and may be nutritive or degenerative. It appears that a small number of oocytes are spawned intermittently, but there is no evidence of overproduction and break-down of superfluous oocytes. A few large oocytes become senescent and undergo internal break-down, releasing periodic acid Schiff-positive material into the lumen. Size-frequencies of oocytes indicate that eggs may be spawned as a continuous slow release, and there is no evidence of reproductive synchrony between or within samples. On reaching maturity, males appear always to be ready to release spermatozoa. Spawning is probably stimulated by egg release during chance encounters with mature females. There is no evidence for brooding, and from the large size and yolky nature of the egg direct lecithotrophic development at or near the seabed is inferred. A limited histological study of H. gennaeus indicates that egg production is very similar, but the two species differ in the nature of the accessory cells and amorphous material filling the lumen.  相似文献

Laboratory studies on the reproductive cycle of the planktonic copepod Temora stylifera, collected from the Gulf of Naples between October 1987 and March 1988, showed that females oscillated between a light and dark gonadal condition. Histological preparations indicated that the dark condition corresponded to a ripe stage of oogenesis as compared to the light or unripe maturation state of primary oocytes. The number of eggs released within 24 h was strongly dependent on the reproductive status of the female at the time of incubation. Light unfed females rarely reverted to a dark or ripe condition and egg deposition in such cases was almost always zero. Dark females produced eggs within 24 h under all experimental food conditions, reverting from dark to light soon after egg deposition. Successively, the length of the interclutch period was strongly dependent on the type of food present. A high percentage of infertile eggs was almost always present. We suggest that short-term studies, not taking into consideration the condition of gonads at the time of incubation, will underestimate potential egg production rates. Females maintained for longer than three or four days produced mainly infertile eggs. After remating, females reinitiated the production of fertile eggs but egg viabilities never reached 100% hatching success, indicating poor efficiency in the fertilization of mature oocytes. We hypothesize a possible cause and effect relationship between egg viabilities and seasonal fluctuations in population densities.Died tragically at sea during a cruise  相似文献
