Ozone degradation of a mixture containing methylparaben, ethylparaben, propylparaben, butylparaben and benzylparaben was carried out in aqueous solution. The degradation followed the pseudo-first-order kinetic model and occurs with two ozonation stages with the observed rate constants of second stage ozonation, k obs2, being higher than the observed rate constants in first stage, k obs1. The k obs1 of parabens was found to increase exponentially whilst k obs2 was found to maximize at 35°C. Both k obs1 and k obs2 were found to decrease exponentially with respect to the initial concentration of parabens. Both pH and ozone dose showed positive effects on the rate of degradation. It was also observed that an ozone dose of 0.67 g/h resulted in the removal of 99% of parabens in 12 min, and also the removal of 61 and 32% of chemical oxygen demand (COD) and total organic carbon (TOC), respectively, in 3 h of ozonation time for a 500 μM of solution of parabens.  相似文献

The persistence of bacteria in Anodonta cygnea largely depends on the hemolymph bactericidal activity against aquatic micro-organisms.

The aim of this study was to assess the in vitro bactericidal activity of whole hemolymph against multi-drug-resistant Escherichia coli and enterococci strains. Four mussels were injected with multi-resistant strains of E. coli in order to determine their in vivo bactericidal reactivity. In vitro experiments showed that the hemocytes viability decreased almost 70% in 4 h, the same happened in the control hemolymph. Enterococci tested in in vitro experiments were more susceptible than E. coli to whole hemolymph and hemocytes fractions. None of the enterococci bacteria was detected in the hemolymph fractions from 24 to 72 h, while E. coli was still detectable 72 h after inoculation both in the control and respective plasma fractions. The microbial inhibition on both hemolymph and cell pellets suggest that hemocytes may be the main responsibles for that process.

In vivo experiments showed that hemolymph inhibited the E. coli load injected and it was correlated with a high tendency for the increase in hemocytes counts, after 72 h. High adherence of E. coli and an intense pseudopods manifestation, mainly to the granulocytes, suggest a clear phagocytosis process.  相似文献

Fourteen benzophenone-type UV filters and four paraben preservatives were selected to examine their acute toxicities on Dugesia japonica. The 48-h LC50 values for planarians exposed to benzophenone-type UV filters can be ranked as oxybenzone?>?mexenone?>?5-chloro-2-hydroxybenzophenone?> 2,4-dihydroxybenzophenone?>?2-hydroxybenzophenone?>?dioxybenzone?>?benzophenone?>?2,2′,4,4′-tetrahydroxybenzophenone?>?4-hydroxybenzophenone?> 3-hydroxybenzophenone?>?4,4′-dihydroxybenzophenone?>?2,2′-dihydroxy-4,4′-dimethoxybenzophenone?>?2,3,4-trihydroxybenzophenone?>?sulisobenzone with a range from 0.9 to 145?mg?L?1 with a similar sequence for the 96?h LC50 values, ranging from 0.5 to 77?mg?L?1. The 48 and 96?LC50 values for planarians exposed to paraben preservatives can be ranked as butylparaben?>?propylparaben?>?ethylparaben?>?methylparaben. Among all the tested chemicals, oxybenzone was the most toxic and sulisobenzone the least toxic chemical to planarian at each exposure period. Most benzophenone-type UV filters are toxic to aquatic animals with 48?h LC50 values less than 10 mg?L?1, except for 2,2′-dihydroxy-4,4′-dimethoxybenzophenone, 2,3,4-trihydroxybenzophenone, and sulisobenzone. Because of their common occurrence in aquatic environment, more studies on aquatic toxicities of benzophenone-type UV filters and paraben preservativs are needed to provide important information to adequately assess their ecological risk.  相似文献

A synergistic action of silver (Ag) and groundnut husk carbon was shown, which markedly enhanced inactivation of Escherichia coli. The purpose of this study was to assess the feasibility of using a silver-impregnated groundnut husk carbon adsorbent (AdSG) for drinking water disinfection and evaluate % disinfection of E. coli. The results showed that contact of bacterial cells with AdSG resulted in a transfer of Ag ion to the cell, as evidenced by bactericidal activity. It is also possible that Ag produced generation of reactive oxygen species in the cell that led to bactericidal activity of the adsorbent. Data indicated that AdSG was effective as a bactericidal agent and may prove to be economically beneficial in drinking water disinfection.  相似文献

随着抗生素抗性污染日益严重,快速评估环境中典型病原菌与条件性致病菌的抗生素抗性水平,对掌握区域环境抗生素抗性污染状况、揭示抗性污染传播规律至关重要。通过以最低抑菌浓度浸入抗生素改进MI、VJ培养基,并结合滤膜法,建立了针对近岸海洋环境中指示性病原微生物大肠杆菌(Escherichia coli,E.coli)与金黄色葡萄球菌(Staphylococcus aureus,S.aureus)的抗生素抗性监测方法。水体和沉积物样品抗生素抗性水平评估实验结果显示,该方法具有较好重现性(水体和沉积物中E.coli和S.aureus抗生素抗性水平的相对标准偏差分别为11%、8%)与准确度(水体和沉积物中E.coli和S.aureus的平均回收率分别为83.5%、68.4%;相对于CLSI药敏试验的偏离度为±0.1)。且与CLSI药敏实验相比,该方法过程简便、耗时短(36 h/84 h),能最大限度节约经济和人员成本提高抗性评价效率。应用该方法评估辽河口与莱州湾环境中2种病原微生物磺胺类抗生素抗性水平,结果显示辽河口水体中E.coli和S.aureus磺胺二甲嘧啶的平均抗性率分别为27.0%、28.4%,沉积物中分别为35.5%、34.6%;莱州湾水体中E.coli和S.aureus磺胺二甲嘧啶的平均抗性率分别为26.0%、14.5%,沉积物中分别为12.0%、32.9%。该方法适用于河口、近岸海洋及入海排污口水体与沉积物样品中E.coli与S.aureus的快速分析及抗生素抗性水平评估。  相似文献

为了揭示海陆衔接区环境中抗生素与抗性细菌分布特征及其内在相关性,以莱州湾及其主要入海河流为研究区域,利用HPLC-MS/MS分析样品中15种磺胺类抗生素(SAs)和6种喹诺酮类抗生素(QNs)的浓度,并通过改良的Method 1604(US EPA)评估海水与沉积物中2种典型水传病原微生物大肠杆菌(E.coli)与金黄色葡萄球菌(S.aureus)的抗生素抗性水平,进而探讨该区域水体中抗性菌株的分布特点以及微生物抗性率与相应抗生素浓度的相关性。结果显示,莱州湾水体与沉积物中普遍存在磺胺与喹诺酮类抗生素残留及抗性污染问题。两大类抗生素在水体中平均残留浓度分别为3.89 ng·L~(-1)(SAs)和234.68ng·L~(-1)(QNs),在沉积物中分别为0.91 ng·g~(-1)(SAs)和49.37 ng·g~(-1)(QNs),且分布特征基本呈现自河流向海洋逐渐递减的趋势,说明河流输入是莱州湾抗生素污染的主要来源。在水体中,具有磺胺类抗性的E.coli和S.aureus平均检出量分别达到2 018和4 683 CFU·L~(-1),抗性率范围分别在0%~37.3%和10.6%~45.8%之间;而2种喹诺酮类抗性病原微生物的平均检出量则相对较低,分别为1 315 CFU·L~(-1)(E.coli)和1 461 CFU·L~(-1)(S.aureus),抗性率分别为0%~50.0%和0%~20.8%;此外,相比于E.coli,S.aureus为沉积物中的主要抗性病原微生物,磺胺与喹诺酮类抗性S.aureus检出率均高于80%,平均检出量分别为24CFU·g~(-1)和18 CFU·g~(-1)。相关性分析表明,莱州湾近岸海域水体中磺胺类抗生素浓度与磺胺类抗性微生物总量之间具有良好的线性关系,然而其与微生物抗性率之间并未表现出相似的规律,说明近岸海洋环境中抗生素的残留量不是影响抗性菌株丰度的唯一因素。  相似文献

为研究纳米银和银离子对土壤微生物的影响,采用土壤培养方式,对不同剂量纳米银(10、50、100 mg·kg~(-1))和银离子(1、5、10 mg·kg~(-1))暴露下黄褐土、砖红壤中可培养微生物数量及土壤酶活性(脲酶、荧光素二乙酸酯水解酶、蔗糖酶、过氧化氢酶)进行研究,并采用纯培养方法对纳米银和银离子暴露下的大肠杆菌(Escherichia coli)、金黄色葡萄球菌(Staphylococcus aureus)凋亡情况进行检测,对纳米银释放的银离子毒性进行评估。结果表明,随着纳米银剂量的增加,土壤可培养微生物数量显著减少,脲酶和过氧化氢酶活性降低,蔗糖酶、荧光素二乙酸酯水解酶(FDA酶)活性没有显著变化;银离子处理中微生物数量明显减少,但土壤酶活性被激活。10 mg·L~(-1)纳米银暴露1 h后大肠杆菌、金黄色葡萄球菌凋亡率、死亡率增高;随着培养时间的延长,纳米银缓慢释放银离子,并促进大肠杆菌的凋亡。综上分析,纳米银能够抑制土壤可培养微生物生长和酶活性,其中脲酶、过氧化氢酶对纳米银较为敏感,蔗糖酶、FDA酶受纳米银的影响较小;纳米银的毒性一方面是其本身的特异抗菌性,也有部分来自缓慢释放的银离子。  相似文献

Bacterial systems have not had success predicting metal carcinogenicity. Hypotheses explaining this failure are examined. Using a broad genetic endpoint, λ prophage induction, under sub‐toxic growing conditions, genotoxicity is seen for compounds of chromium, manganese, lead, molybdenum and tungsten. Copper, manganese, arsenic and molybdenum compounds enhanced UV mutagenesis in E. coli WP2.

The toxicity of metal compounds to cultured mammalian cells correlates well with rat oral LD50 values. Whereas insolubility can present problems in bacterial studies, concentrations of metal compounds toxic to mammalian cells can be determined even in the presence of precipitate, and sometimes [Pb(NO3)2, BaCl2 and BeCl2] occurs only in its presence. PbS and MnS, which are insoluble, are much more toxic than the more soluble compounds Pb(NO3)2 and MnCl2. These results demonstrate the importance of cellular phagocytosis of insoluble metal compounds as a factor in studying the toxicity and genotoxicity of metal compounds.  相似文献

Aqueous and flavonoid-enriched extract as well as essential oil (EO) obtained from leaves of Pistacia lentiscus were assessed for antibacterial and antimutagenic activities. Antibacterial activity of different extracts and EO were evaluated against six bacterial strains. A marked inhibitory effect was observed against Salmonella typhimurium, whereas lower activity was observed against Staphylococcus aureus, Pseudomonas aeruginosa, and Salmonella enteritidis. EO showed significant inhibitory effects against Salmonella typhimurium, Salmonella enteritidis and Staphylococcus aureus. The antimutagenic activity of the different extracts against Aflatoxin B1 (AFB1) and sodium azide was demonstrated with the Salmonella typhymurium assay. The number of revertants per plate decreased significantly when the plant extracts were added to the assay system using Salmonella typhimurium TA100, TA98 and TA1535.  相似文献

Nano-titania is widely used in the food industry due to its efficient antimicrobial activity. However, the mechanism of microbial toxicity of nano-titania is poorly known. Here, nano-TiO2 has been fabricated by microwave-irradiation chemistry, a new method, and then tested for antimicrobial activity. Mutagenicity of nano-TiO2 was evaluated using Salmonella typhimurium histidine-auxotrophic strains. The reactive oxygen generation test was performed using 2,7-dichlorofluorescein diacetate dye. To test membrane permeabilization, E. coli cultures were grown in nutrient broth at an optical density of 0.3–0.5 at 610 nm, harvested by centrifugation at 11,000g for 10 min, washed and resuspended in 0.5 % NaCl solution. We also analyzed superoxide formation and membrane integrity, and we used scanning electron microscopy. Results show that nano-TiO2 has a minimum inhibitory concentration of 15 µg/mL, and a minimum bactericidal concentration of 20 µg/mL for E. coli. The bacterial inner wall was ruptured, and cytoplasmic content was released after 5 min of treatment in a dose-dependent manner. Notably, superoxide formation was not observed, which establishes the fact that reactive oxygen generation and alteration of membrane integrity, as well as permeability, is the major mechanism of antimicrobial activity of nano-TiO2.  相似文献

Electronic waste (e-waste) poses a major public health threat for developing countries’ populations. The hazards of e-waste are exacerbated by crude recycling methods. In this study, the presence of metals in e-waste samples obtained from Lagos, Nigeria, was assessed using atomic absorption spectrometry. The effect of e-waste on Escherichia coli (E. coli) PQ-37 genomic integrity was evaluated using the SOS chromotest. The means of metal concentrations in the evaluated samples were 16 (cadmium), 7.3 (nickel), 11 (chromium), 20 (lead), 3100 (iron), 90 (zinc), and 2000 (copper) μg_L?1. Damage to E. coli deoxyribonucleic acid (DNA) increased proportionally to the metal concentrations. Significant amounts of DNA damaging agents from inadequately processed e-waste are present in the studied environment, which will have implications for adverse effects on public and ecological health. Existing policies against dumping of toxic materials in susceptible communities should be enforced.  相似文献

A polyaniline Zr(IV) selenotungstophosphate nanocomposite was prepared via sol-gel method and characterized by thermogravimetric analysis, scanning electron microscopy, energy dispersive X-ray spectroscopy, and transmission electron microscopy. The combined sorptional–photocatalytic activity of the nanocomposite for degradation of methylene blue and malachite green was investigated and was found to be more efficient than separate adsorption in the dark followed by photocatalysis. The dyes were degraded in 3 h by 96% and 89% by the combined process, as compared to 86% and 72% by the two-step process in 5 h. The nanocomposite material showed antimicrobial activity against Staphylococcus aureus and Escherichia coli.  相似文献

Summary. The scope of this work was to examine whether leaf constitutive secondary metabolites play a role in determining bacterial colonization of the phyllosphere. To this aim, we surveyed nineteen native or cultivated plant species that share a common bacterial pool in a North Mediterranean area, and estimated the size of total and ice nucleation active (INA) bacterial populations on their leaves. Large differences in the colonization of their phyllosphere were found; the population size of epiphytic bacteria ranged from 7.5 × 102 to 1 × 106 CFU/g fresh weight, in eucalypt and celery, respectively. Species native in Mediterranean-type climate areas, particularly those belonging to the group of aromatic plants, are characterized by scarce presence of INA bacteria. The antibacterial activity of essential oils, surface phenolics and leaf tissue extracts was also estimated against the INA strains P. syringae and E. herbicola, isolated from two of these plant species. E. herbicola proved more sensitive than P. syringae. Of the species examined, oregano [Origanum vulgare L. subsp. hirtum (Link.) Ietswaart], an aromatic plant, had the highest antimicrobial activity, whereas six species showed no activity at all. Further experiments were performed with oregano and bean (Phaseolus vulgaris L.) that represent two extremes in their secondary metabolite content. Both plants were inoculated with P. syringae. By the end of incubation, the bacterial population on bean plants was about 100 times higher than that on oregano leaves. Scanning electron micrographs showed that bacterial growth on oregano leaves was confined to sites away from glandular hairs. Results from the bacterial colonization survey together with those from the toxicity tests showed that all species rich in antibacterial secondary metabolites harbored low leaf bacterial populations. These results provide substantial evidence that leaf secondary metabolites function as constitutive defense chemicals against microbial invasions. However, the fact that species with non- or moderately active leaf secondary metabolites are not always highly colonized suggests mediation of other unknown factors, the contribution of which requires further investigation.  相似文献

抗生素的滥用造成的环境安全问题已不可忽视,关于抗生素联合毒性效应研究较多,但联合突变效应研究较少。因此,本文以大肠杆菌为受试生物,研究了2种磺胺类抗生素(磺胺氯哒嗪(SCP)、磺胺二甲嘧啶(SMZ))和3种四环素类抗生素(二甲胺四环素(MH)、盐酸四环素(TH)和盐酸强力霉素(DH))单一及联合暴露时对大肠杆菌的突变效应。结果表明:在单一暴露下,磺胺类抗生素会促进大肠杆菌的突变效应,四环素类抗生素没有明显的促进作用;联合暴露下,磺胺类抗生素对大肠杆菌突变效应为相加,磺胺和四环素类抗生素对大肠杆菌突变效应为拮抗。本研究初步探索了抗生素对大肠杆菌的联合致突变风险,为今后环境中抗生素混合暴露的生态风险评价和抗生素污染控制标准制定提供一种理论支撑。  相似文献

The influence of added organic nutrients on the bactericidal capacity of natural and synthetic sea water was investigated, with special reference to levels below the normal concentration of orgnnic matter in natural sea water. Resting cells of Escherichia coli, Staphylococcus aureus and Serratia marinorubra were used as test organisms. The inactivation of test bacteria in synthetic and filter-sterilized natural sea water could be increased by addition of ZoBell's 2216E-broth (containing peptone and yeast extract), peptone or glucose. This effect of the nutrients depended on chemical nature and concentration as well as on specific properties of the test strains. In regard to peptone, less than 10-4 mg/ml must be added to induce bacterial kill exceeding that observed with unsupplemented controls. With raw sea water, no fundamentally different results were obtained; however, the influence of marine bacteria was clearly discernible. All findings support a previously offered hypothesis of nutrient-dependent antibacterial activity of sea water which is discussed in detail.  相似文献

抗生素的滥用导致细菌耐药问题日益严重,人类迫切需要开发出新的抗菌药物以减少细菌耐药问题。基于纳米银制备而成的纳米银复合材料在兼顾纳米银抗菌性能的同时不仅能够克服单一纳米银释放速度快、不稳定等缺点,还能缓解细菌耐药的问题,因此被认为是一类具有广泛应用前景的新型抗菌剂。已有研究表明,单一纳米银与某些抗生素的联合使用可以达到协同抗菌效果,但目前尚缺乏对纳米银复合材料与抗生素的联合抗菌性能及机制的研究。本文首先制备出3种不同结构的纳米银复合材料,包括二氧化硅-聚多巴胺-纳米银复合材料(SiO_2-PD-AgNPs)、纳米银@二氧化硅复合材料(AgNPs@SiO_2)和纳米银@二氧化硅-聚多巴胺-纳米银复合材料(AgNPs@SiO_2-PD-AgNPs)。随后测定了纳米银复合材料对大肠杆菌(Escherichia coli, E. coli)和枯草芽孢杆菌(Bacillus subtilis, B. subtilis)的单一毒性效应。结果显示,AgNPs@SiO_2-PD-AgNPs复合材料对2种菌的单一毒性均大于其余2种纳米银复合材料。因此,笔者以AgNPs@SiO_2-PD-AgNPs作为代表,测定了纳米银复合材料与硫酸卡那霉素(kanamycin sulfate, KS)/盐酸土霉素(oxytetracycline hydrochloride, OH)的二元联合抗菌性能,发现AgNPs@SiO_2-PD-AgNPs与KS联合可以对E. coli产生协同效应。协同效应产生的主要原因可能是:AgNPs@SiO_2-PD-AgNPs释放出的纳米银会和KS发生键合反应生成KS-纳米银复合物,导致纳米银释放出大量的Ag+增加了细胞膜的通透性,从而使得进入细菌内的Ag~+和KS比单独作用时进入胞内的抗菌剂增多,产生更强的抗菌性能,从而表现出协同抗菌效应。本研究基于新型纳米银复合材料与抗生素的联合抗菌性能实验探究了纳米银复合材料与特定抗生素联合用药的最佳组合和相关机制,为今后开发新型抗菌材料提供了新思路并为相关联合用药提供参考。  相似文献

Extracts of 19 of 24 species of sponge collected from Queensland. Australia, inhibited the growth of test microorganisms in in-vitro assays. A similar result has been obtained by earlier workers for extracts of species of sponge obtained from temperate waters. Some of the extracts, including extracts of Thorecta vasiforis, Arenochalina mirabilis and Acanthella kleutha, showed activity against bacterial strains that was comparable with that exhibited by penicillin G and streptomycin against these strains. Gram-positive bacteria were expecially sensitive to many extracts. Little activity was exhibited by any extract against four species of fungi tested. Some of the extracts were markedly toxic to one or more of the test organisms (a fish, a crustacean and a hydroid) used, but no clear pattern linking toxicity to these organisms with antimicrobial properties of the extracts emerged. In general, there bas a negative correlation between antimicrobial activity and surface-fouling, raising the possibility of using freedom from surface-fouling as an indicator of antimicrobial activity. Four of five encrusting species from the undersides of coral boulders showed antimicrobial activity. This study confirmed the value of using methanol-toluene extracts in field-based screening programmes, but emphasised the need to use additional extracting media such as methylene chloride in order to augment the number of antimicrobial compounds detected. There are indications that antimicrobial activity may be widespread in the order Dictyoceratida, in the order Verongida and in the order Axinellida, but sporadic in other orders of Porifera.  相似文献

Flow cytometry (FCM) has been widely used in multi-parametric assessment of cells in various research fields, especially in environmental sciences. This study detected the metabolic activity of Escherichia coli and Staphylococcus aureus by using an FCM method based on 5-cyano-2,3-ditolyltetrazolium chloride (CTC); the accuracy of this method was enhanced by adding SYTO 9 and 10%R2A broth. The disinfection effects of chlorine, chloramine, and UV were subsequently evaluated by FCM methods. Chlorine demonstrated stronger and faster destructive effects on cytomembrane than chloramine, and nucleic acids decomposed afterwards. The metabolic activity of the bacteria persisted after the cytomembranewas damaged as detected using CTC. Low-pressure (LP) UV or medium-pressure (MP) UV treatments exerted no significant effects on membrane permeability. The metabolic activity of the bacteria decreased with increasing UV dosage, and MP-UV was a stronger inhibitor of metabolic activity than LP-UV. Furthermore, the membrane of Gram-positive S. aureus was more resistant to chlorine/chloramine than that of Gram-negative E. coli. In addition, S. aureus showed higher resistance to UV irradiation than E. coli.

Summary. Many plant species contain essential oils with allelochemical properties, yet the extent to which these same chemicals can be autotoxic is unclear. In this study, we tested the toxicity of several essential oil components to three species that produce them—Pastinaca sativa and Petroselinum crispum (Apiaceae), and Citrus jambhiri (Rutaceae). The effects of exogenous application of small amounts of essential oil components to the surface of foliage, followed by a pinprick to allow entry into the leaf, were monitored by chlorophyll fluorescence imaging. Rapid and spatially extensive declines in photosynthetic capacity were detected within 200 s. The most toxic constituents were monoterpenes. Two sesquiterpenes, caryophyllene and farnesene, and the phenylpropanoid myristicin, by comparison, were not toxic. Autotoxicity of endogenous essential oil was investigated by slicing through containment structures (glands or tubes); significant toxicity, manifested by reduced photosynthetic activity, was observed in all three species but was most pronounced in P. sativa and P. crispum, both of which possess oil tubes.  相似文献
