Freshwater edible catfish Clarias batrachus (Linn.) was treated with sublethal concentration of two different groups of pesticides- carbaryl, a carbamate and phorate, an organophosphate for 24 ,72 ,120 and 168 h. The disorders of lipid metabolism were observed in serum. Cholesterol levels in the serum decreased significantly throughout the exposure period with both the pesticides. These results indicate one significant manifestation of the toxic response from the fish under the stress of pesticide exposure.  相似文献

To better understand sublethal effects of harmful algal blooms (HABs) on fish, mummichog, Fundulus heteroclitus (L.), were exposed in the laboratory to varying, environmentally relevant densities of Pfiesteria shumwayae (Glasgow et Burkholder, CCMP 2089, dinoflagellate) and Chaetoceros concavicornis (Mangin, CCMP 169, diatom). Two experiments were conducted during the spring of 2003 and 2004 to quantitatively examine the effects of acute (2 h) P. shumwayae and C. concavicornis algal exposure on mummichog brain activity using c-Fos expression as a marker of altered neuronal activity. Brains from HAB-exposed fish were removed, sectioned, and stained using immunocytochemistry prior to quantifying neuronal c-Fos expression. Fish exposed to P. shumwayae and C. concavicornis showed increased c-Fos expression compared to unexposed control fish. A significant dose-response relationship was observed, with increased labeling in brains of fish exposed to higher cell densities for both HAB species tested (P ≤ 0.01). Increased labeling was found in the telencephalon, optic lobes, midbrain, and portions of the medulla. The greatest increases in expression were observed in the telencephalon of P. shumwayae-exposed fish, and in the telencephalon and optic lobes of C. concavicornis-exposed fish (P ≤ 0.01). These increases in c-Fos expression are consistent with other physical and chemical stress exposures observed in fish. Neuronal stress, evidenced by c-Fos expression, demonstrates a sublethal effect of exposure and changes in brain activity in fish exposed to HAB species.  相似文献

Fish are often exposed to highly contaminated water, especially in areas where the dilution rate of waste water is low. Environmental pollutants or other stress may affect one or more of the immunological functions in fish. Effective control of toxic substances is essential for the success of fish culture, which requires fish health monitoring using biomarkers. This study evaluated the effects of heavy metal copper (Cu) at environmentally relevant concentrations on the hematopoiesis in Labeo rohita. L. rohita fingerlings were exposed to different Cu concentrations for 6, 24, 48, or 72 hr. Following exposure, percentage blast cells was found to be increased after 6 and decreased after 24, 48, and 72 hr both at sublethal and LC50 concentrations, whereas percentage mature erythrocytes decreased after 6 and increased after 24, 48, and 72 hr. Erythropoietic and leukopoietic efficiencies were also affected upon exposure. Erythropoietic and leucopoietic efficiencies increased markedly after 6, 24, and 48 hr but decreased after 72 hr in sublethal-treated fish. Erythropoietic efficiency decreased significantly after 6, 24 and 48 hr but increased after 72 hr in LC50-treated fish. Leukopoietic efficiency decreased significantly after 6 hr exposure and increased after 24 and 48 hr but decreased after 72 hr. Flow cytometric studies of head kidney cell cycle phase distribution revealed cell death and DNA synthesis. The percentage cell death increased in fish exposed to sublethal concentrations and rose further at LC50 during the earlier hours of exposure. Synthesis of DNA was increased at sublethal concentrations but was significantly reduced at LC50 dose.  相似文献

The knowledge of fecundity of fish from a specific aquatic body is extremely important in the successful management and exploitation of its fishery. In the present investigation the fecundity of the freshwater fish, Notopterus notopterus was studied in fish collected from a natural aquatic body (Sirnoor nala) near Gulbarga and also in fish exposed to some heavy metal contamination (HgCl2, CdCl2 and their combination) at sublethal concentration for 15 days in the laboratory. The mathematical relationship between fecundity and total length, body weight, ovary length and ovary weight were determined in both unexposed and exposed fish. The fish, N. notopterus has bigger oocytes and are few in number. Studies in the fish exposed to heavy metals indicate that significant reduction in these parameters after exposure to heavy metals at sublethal concentration was noticed. The fecundity has straight line relationship with total length, body weight, ovary length and ovary weight in control fish which did not alter after heavy metal exposure. This study provides the viability of species in only specific environment.  相似文献

The present investigation was undertaken to assess the effects of sublethal concentrations of zinc (0.1 and 0.2 mgl(-1)) on the nutritional value of fish Channa punctatus after exposure for 135 days. The parameters studied were, levels of total proteins, glycogen, total lipids, cholesterol and vitamins (A and D) in the muscle. Total proteins, glycogen, total lipids and vitamin D show highly significant decline from day 90 to 135, while cholesterol and vitamin A, show no significant changes upto 105 days. This study signifies a drastic reduction in the nutritive value of fish, which is compounded by the fact that under natural conditions fish are exposed to these levels of zinc for prolonged periods.  相似文献

The unrestricted use of agrochemicals in crop fields lowers the overall prospects of survival and yield of wetland fish species like Anabas testudineus. In the present study, the impact of different doses of SUMIDON-40, an organophosphate (OP) pesticide on the hematopoiesis of Anabas testudineus has been examined. The small lymphoid hemoblast and consequently the polychromatophilic erythroblasts decreased significantly at both sublethal and LC50 dose-treated fish. The percentage of mature erythrocyte also fell significantly in the pesticide-treated groups. The erythropoietic efficiency was increased in fish exposed to sublethal dose but fell in fish exposed to LC50 dose. Among leucocytes, the percentage of neutrophils rose in Anabas testudineus in both treatment groups and percentage of macrophage decreased significantly only in LC50 dose-treated group. The percentage of lymphocyte increased significantly in sublethal dose-treated groups. The overall leukopoietic efficiency, however, was elevated significantly in both treatment groups. These facts clearly indicated that pesticides affected the overall process of hematopoiesis in this fish species. Flow cytometric analysis of cell cycle in pronephric kidney cells confirmed changes in percentage of cell death and DNA synthesis following pesticide exposure. Data indicate that habitat deterioration from agrochemicals impedes the hematopoiesis in this species resulting in reduced tolerance in their usually hypoxic habitat environment.  相似文献

Various heavy metals released into the rivers and lakes affect aquatic fauna including Clarias batrachus. Elevated levels of chromium (Cr) in aquatic organisms, especially fish, represent an ecological and human concern. Amino acid levels were estimated in five tissues (gills, liver, kidneys, muscle, and brain) of C. batrachus after 28 days of exposure to Cr. This study showed that Cr elevated levels of free amino acids in organs of fish C. batrachus, indicating a compensatory mechanism to counter sublethal metallic stress. Data suggest that the measurement of amino acids may serve as a biomarker for Cr toxicity in fish.  相似文献

Background

Available literature and regulatory studies show that the severity of effects of beta-cyfluthrin (a synthetic pyrethroid) on fish is influenced by the magnitude and duration of exposure. To investigate how the exposure pattern to beta-cyfluthrin (constant vs peak) may influence the response of the fish, we used a mechanistic effect model to predict the survival and growth of the rainbow trout over its early life stages (i.e. egg, alevin and swim-up fry). We parameterized a toxicokinetic–toxicodynamic (TKTD) module in combination with a dynamic energy budget model enabling us to describe uptake and elimination, as well as to predict the threshold concentration for survival and sublethal effects (feeding behaviour and growth). This effect model was calibrated using data from an early life stage experiment where trout was exposed to a constant concentration of cyfluthrin. The model was validated by comparing model predictions to independent data from a pulsed-exposure study with early life stages of rainbow trout.

Results

The co-occurrence of effects on behaviour and growth raised the possibility that these were interrelated, i.e. impairment of feeding behaviour may have led to reduced food intake and slower growth. We, therefore, included ‘effect on feeding’ as mode of action in the TKTD module. At higher concentrations, the constant exposure led to death. The model was able to adequately capture this effect pattern in the calibration. The model was able to adequately predict the response of fish eggs, alevins and swim-up fry, from both the qualitative (response pattern) and quantitative points of view.

Conclusions

Since the model was successfully validated, it can be used to predict survival and growth of early life stages under various realistic time-variable exposure profiles (e.g. profiles from FOCUS surface water modelling) of beta-cyfluthrin.

The sublethal toxicity of sodium arsenite on protein metabolism was investigated in teleost fish, Tilalpia mossambica at the end of 24, 48, 72 and 96 h of exposure. Total protein content, free amino acid content and activities of the enzymes aspartate amino transferase (AAT) and alanine amino transferase (ALAT) in liver, gill, brain and muscle exhibited significant (P<0.05) alterations throughout the investigation in relation to that of control. It is suggested that the fish is able to respond to the stressful situations by gearing up the metabolic activity as revealed by the elevated protein, amino acid content and the activities of AAT and ALAT.  相似文献

Freshwater fish Prochilodus lineatus were exposed to sublethal concentrations of cypermethrin (0.3 and 0.6 microg/l) for 2, 5 and 8 days. It was observed that with the increase of exposure time total erythrocyte (RBC), hemoglobin (Hb), hematocrit (Ht) and mean corpuscular hemoglobin concentration (MCHC) values decreased but mean corpuscular volume (MCV) and mean corpuscular hemoglobin (MCH) values increased. These reports indicate that hematological parameters, may be useful as a diagnostic test for cypermethrin exposure in aquatic organisms.  相似文献

The biochemical composition of juvenile blue crabs, Callinectes sapidus, exposed to sublethal concentrations of the water-soluble fraction of South Louisiana crude oil (0 to 2 504 ppb) for 21 d were examined. Although growth took place in all crabs, tissue content varied inversely with exposure concentration while percentage tissue water varied directly with exposure concentration. Total protein, lipid and RNA content of crabs exposed to crude oil were significantly less than that of control crabs by Day 21. DNA content was not significantly different from that of control crabs, suggesting that the difference in tissue content was due to differences in cell volume and not cell number. There were no consistent differences in the concentration of the major biochemical components, indicating that the relative contribution of each of the components remained stable during the period of sublethal stress. Ratios of RNA:DNA and protein:DNA decreased in exposed crabs and were positively correlated with scope for growth and observed growth. The ratios may be useful as indirect indicators of physiological condition. Analysis of lipid classes indicated that structural lipids in stressed crabs were less affected than were lipids used for energy storage. The changes in biochemical composition suggest that the pattern of energy utilization was altered in crabs exposed to crude oil. Growth in size without comparable growth in tissue resulted in decreased tissue content. Available energy was used for growth, with little being stored in lipid reserves.  相似文献

The objectives of this study were to evaluate the effects of sublethal cadmium concentrations on the levels of cadmium, metallothionein (MT) and histological changes in gills of East Java strain tilapia (Oreochromis niloticus) at different salinity levels. The levels of cadmium in control gills were not significantly different at 0, 5 and 10 practical salinity unit (PSU). The cadmium concentrations in gills of cadmium-exposed fish were significantly higher at 0 PSU than at 5, 10 and 15 PSU. The MT concentrations in control gills were not significantly different at 0, 5, and 10 PSU. The MT concentrations of cadmium-exposed fish were significantly higher than those in respective control groups at 0, 5 and 10 PSU. Significant gill damage occurred in fish exposed to cadmium at lower salinity. The epithelial lifting was noted at gills of fish exposed to 2.5 mg/L of cadmium at 0 PSU, and telangiectasis was observed at gills exposed to 5 mg/L of cadmium at 0 PSU. The level of gill damage decreased with increasing salinity of media. The increased MT and histological changes in gills of our findings could be a protective response of animals to toxic effect of cadmium.  相似文献

The activities of phosphatases and transaminases were studied in muscle and liver of the fresh water fish, Oreochromis mossambicus on exposure to different sublethal concentrations (0.25, 0.5, 0.75 and 1 mgl(-1)) of cartap hydrochloride (insecticidal derivative from marine polycheate) for 96 h. In the present study, an attempt has been made to quantify the fenvalerate accumulated in different tissues (gill, muscle and liver) and observe changes involved in the levels of sodium, potassium and calcium ions and Na+–K+, Mg2+ and Ca2+ adenosine triphosphatase (ATPase) activities in the freshwater fish, Cirrhinus mrigala on short-term and long-term exposure to the median lethal and sublethal concentration of fenvalerate. Residue analysis using gas–liquid chromatography (GLC) technique revealed that fenvalerate accumulated in highest quantity in gill followed by liver and muscle under median lethal concentration (6?µg?L?1). Whereas in sublethal concentration (0.6?µg?L?1), muscle accumulated highest quantity followed by gill and liver, which might be due to the fact that fenvalerate is highly lyphophilic. The ion concentration and ATPase activity were found effected in fish exposed to lethal and sublethal concentrations of fenvalerate. Concentration of Na+, K+ and Ca2+ ions decreased in gill, muscle and liver on being exposed to median lethal concentration to a significant level. Whereas the changes were not highly pronounced at sub lethal level indicating low concentration of fenvalerate and its non-toxic effect at chronic exposure. Na+–K+, Mg2+ and Ca2+ ATPases activity were also found decreased in correspondence to the ionic change under median lethal and sub lethal concentrations in target tissues. This might have lead to behavioural changes and create wide-spread disturbance in the normal physiology, ultimately causing the death of the fish. The results suggest that in biomonitoring programmes, ions and associated ATPases can be a good diagnostic tool for fenvalerate toxicity.  相似文献

Experiments were conducted on the effects of high, sublethal temperature on activity, social behavior, and feeding in the adult tautog Tautoga onitis held under controlled laboratory conditions. Norms of these behaviors were established during acclimation to temperatures at which these fish are commonly found. During acclimation, the fish were active and feeding throughout the day. A dominance hierarchy, based primarily on size, was established for each group, with subordinate fish being displaced from all areas of the aquarium. Under elevated temperature, changes in established norms of these behaviors were observed, with activity, feeding, aggression, and the spacing between fish showing a decrease. Following return to pre-rise temperatures, although aggression remained reduced, activity, interfish distance, and feeding approached levels established during acclimation. 针对浙江某地苯酚泄漏事件,结合实地监测和实验室研究,探索苯酚的水体污染化学行为对水体生态系统的影响和急性毒性效应.本研究选取水生生态系统中明亮发光杆菌T3、优势藻四尾栅藻Scenedesmus quadricauda以及模式鱼斑马鱼Brachydanio rerio为研究对象,探索水体中生产者、消费者及分解者等整个生态系统对不同苯酚浓度胁迫下的生态响应.结果表明,水生生物对苯酚污染浓度的生态响应差异显著,虽然苯酚对发光细菌、四尾栅藻及鱼体都具有一定的毒性影响,但四尾栅藻抑制率与苯酚浓度之间并没有良好的线性关系.发光细菌的相对发光率与苯酚浓度的关系表明,0.2—25 mg.L-1苯酚浓度对水体会产生低毒作用,而30—100 mg.L-1苯酚浓度对水体会产生中毒作用.苯酚对四尾栅藻的影响主要表现为光合作用的暗反应破坏,高浓度苯酚处理组中四尾栅藻会将残余苯酚浓度作为自身碳源而短暂地促进其生长.斑马鱼在苯酚浓度高于15 mg.L-1时,会发生体重下降、死亡率上升、呼吸急促、游动迅速以及导致水质变坏等现象,并且在100 mg.L-1苯酚浓度处理下,鱼体内富集大量苯酚,通过食物链进而对人类健康产生影响.  相似文献

Sublethal effects of chromium trioxide on mosquito fish, Gambusia affinis, were carried out for 28 days on day 4, 8, 12, 16, 20, 24, and 28. The sublethal effects of chromium (LC10, 77.62?mg?L?1) on antioxidant enzymes, thiobarbituric acid reactive substance (TBARS), locomotor behavior, and gill morphology were studied. The antioxidant enzymes like superoxide dismutase (SOD) and catalase were induced throughout the exposure span of 28 days. Percent induction was also increase with the days of exposure. Lipid peroxidation product, malondialdehyde was enhanced in the viscera tissue of chromium-exposed fish. Perturbation of locomotor behavior like decrease in distance travelled (m?min?1) and in swimming speed (cm?s?1) was observed in exposed fish. Chromium toxicity caused alterations in gill morphology like hypertrophy and hyperplasia in secondary lamellae followed by detached epithelium with severe necrosis. It can be concluded from the results that Cr intoxication in Gambusia fish, caused induction in antioxidant enzymes and lipid peroxidation, plus changes in gill morphology and locomotor behavior.  相似文献

Objective and Background

PCB (polychlorinated biphenyle) are typical man-made environmental pollutants that cause a broad spectrum of effects in vertebrates. Although a lot has been studied about the toxic effects of xenobiotic substances on aquatic organisms, their subacute effects are not yet well known. Fish occupies a central position in freshwater food webs and therefore, carp was chosen to be our test organism. The aim of our study was the investigation of subacute effects of Aroclor 1254 (22μg l?1) on respiration, swimming activity, and biotransformation, allowing discussion of changed interaction between the available energy resources.

Methods

The respiration experiments under controlled laboratory conditions were run for 29 days (5 d before, 16 d with and 8 d after chemical exposure) with exposed carp compared to the non-exposed fish. Furthermore, the BehavioQuant system quantitatively monitors the positions of each individual fish before (8 d) and during (21 d) PCB exposure and swimming activity (number of horizontal turnings s?1) of animals was calculated. Thereafter, liver samples were taken from animals for analysis of the phase 2 enzyme activity (glutathione-S-transferase). In the respiration experiments were 6 exposure, 6 controll, and 3 solubilizer controll groups (12 animals each group); in the behavioral experiments were 9 exposure, 6 controll- und 3 solubilizer controll groups (6 animals each group), and in the enzyme activity measurements were 9 exposure, 6 controll and 3 solubilizer controll groups (6 animals each group) examined.

Results and Discussion

A clearly increased (33%) oxygen consumption due to the PCB-exposure is found to be reversible; after the exposure period the respiration of carp shows a level comparable to the oxygen consumption during non-exposure time. By evaluating quantitative behavioral parameters of animals, it becomes evident that the exposure to PCB also causes a reversible change in their swimming behavior. Chemical stress leads to a decrease of mean daily swimming activity combined with an increase of the mean number of turnings during the night. Thereafter, the swimming activity shifts to the circadian swimming behavior under control conditions. Furthermore, our measurement of soluble and microsomal glutathione-S-transferase activity of fish liver shows a significant elevation after exposure period.

Conclusion

Our results prove that it was feasible to detect sublethal effects of PCB-mixture on all parameters under these conditions successfully. Our findings imply that fish are able to cope with the chemicals and we purse the hypothesis that the higher physiological energy demand caused by increased activity of enzymes, which are involved in biotransformation of foregin substances, may be related to a higher respiration of fish stressed by xenobiotics.

Recommendation and Outlook

Further experiments should investigate the effects of PCB on ecophysiological parameters of carp fed with exposed natural food allowing discussion of ecological consequences.  相似文献

The present study was to evaluate heavy metal toxicity stress symptoms in fish blood during long-term exposure of sublethal concentration of copper sulphate (CuSO4). The effects of copper on various heamatological parameters were evaluated exposing Indian fresh water fish, Channa punctatus to a sub-lethal concentration of copper sulphate (0.36 mg l(-1)) for different periods. Exposure of fish to copper showed a significant decrease in the haemoglobin (Hb) content from 10.73 to 6.60%, red blood cells (RBC) from 2.86 to 1.84 x 10(6)/mm3 and packed cell volume (PCV) from 31.00 to 23.33% at the end of 45th day as compared to control. Whereas the white blood cells (WBC) increased from 60.00 to 92.48 x10(3)/mm3, clotting time (CT) from 27.66 to 43.00 second, erythrocyte sedimentation rate from 5.0 to 13.66 mm/hr and mean corpuscular volume from 108.11 to 126.85 cmu, significantly, with increase in exposure periods. Although mean corpuscular haemoglobin concentration (MCHC) and mean corpuscular haemoglobin (MCH) values showed significant increase during 15 and 30 days exposures, both the values were found significantly increased at the end of the experimental period. The differential leucocytes count (DLC) showed a significant increase in the populations of lymphocytes and eosinophils, whereas neutrophils, monocytes and basophiles were found decreased after exposure to copper.  相似文献
