The cosmopolitan deposit-feeding polychaete Capitella is widely used as an indicator of organic pollution and plays an important role in waste recycling at aquaculture sites. Teflubenzuron is currently employed by salmon farms as an in-feed compound to control ectoparasite infestations. Its occurrence in sediments could pose a hazard to local sediment infauna. A bioassay to investigate the effects of exposing Capitella sp I and Capitella sp B to sediment spiked with environmentally relevant concentrations of teflubenzuron (8.4, 25 and 41.8 microg/g dry wt. sediment) is described. No mortality was recorded for Capitella sp I, but increasing teflubenzuron concentrations significantly reduced egestion rates. Capitella sp B exhibited mortality, although egestion was not affected. These results using Capitella spp. indicated that teflubenzuron could affect the rate of sediment processing by polychaetes close to fish farms treated with this compound. Further studies are recommended to assess the potential of contaminants in aquaculture wastes.  相似文献

The comparative experimental study of inorganic mercury (HgII), methylmercury (MeHg) and cadmium (Cd) bioaccumulation in the Asiatic clam Corbicula fluminea was based on a 14 days' exposure to the water column or sediment compartments, as initial contamination sources. For each contaminant and exposure source, a five-point concentration range was set up in order to quantify the relationships between the contamination pressure and bioaccumulation capacity, at the whole soft body level and in five organs: gills, mantle, visceral mass, kidney and foot. Hg and Cd bioaccumulation at the whole organism level was proportional to the metal concentrations in the water column or sediment. For similar exposure conditions, the average ratios between the metal concentrations in the bivalves - [MeHg]/[HgII] and [MeHg]/[Cd] - were close to 10 and 5 for the sediment source and 8 and 15 for the water column source. Metal distribution in the five organs revealed strong specificities, according to the different contamination modalities studied: kidney and gills were clearly associated with Cd exposure, mantle and foot with MeHg exposure and the visceral mass with inorganic Hg exposure.  相似文献

Fish, Noemacheilus barbatulus (stone loach), of different body weights were used to study rates of uptake and loss of cadmium during and after dietary exposure. Fish were kept singly in a continuous-flow system, and fed tubificid worms. The worms had a range of cadmium levels, but all levels were below that needed to cause acute lethal toxicity in the fish. Body weight affected both the maintenance ration and the amount of food consumed ad libitum, but the exponent for body weight (0.78+/-0.04), relating body weight to food consumption, was unaffected by either temperature or the size of feeding ration. The cadmium content of the worms did not affect the size of the maintenance ration. Metal burden in fish changed rapidly both during and after exposure. After exposure, the cadmium burden of starved fish usually declined more rapidly than in fed fish. A 58-fold increase in cadmium content in the food produced a 28% increase of body burden in the fish, and there was no evidence for biomagnification. Maintenance ration and ration ad libitum and rates of uptake and loss of cadmium increased with temperature within the range 8-18 degrees C, but exposure to cadmium at 16 degrees C yielded a higher asymptotic body burden than either 8 degrees C or 18 degrees C. Rate constants for loss of cadmium after exposure appear to be lower than for loss during exposure. Rates of uptake and loss of cadmium vary with metabolic rate. A maximum in the rate of oxygen consumption was measured at 16 degrees C, above which the rate dropped, presumably due to stress. The exponent for body weight was unaffected by activity or temperature. Body weight of fish appeared to affect both the rates of uptake and loss of cadmium, and feeding rations and respiration to the same extent: body weight exponents were not dissimilar.  相似文献

The effect of chronic exposure to a low concentration (0.5 microg l(-1)) of cadmium ions was investigated on escape behaviour of sea bass, Dicentrarchus labrax, using video analysis. Observations were also performed on the microanatomy of lateral system neuromasts. When fish were exposed for 4h per day over 8 days to the cadmium ions, most of both types of neuromasts observed remained intact. However, some of them presented damaged sensory maculae. Whereas before cadmium exposure, fish responded positively to nearly all the lateral system stimulations, after exposure they decreased by about 10% their positive responses to stimulations. From the 15th day after the beginning of cadmium exposure, neuromasts presented progressively less damage, cadmium accumulation in gills and scales decreased significantly and fish escape behaviour had recovered. This study presents a new concept in ecotoxicology: using behavioural change to reveal the effects of pollution levels, scarcely detectable by currently used techniques (physiological responses).  相似文献

We studied the effects of heavy metal exposure on host plant choice and performance of the grass miner Chromatomyia milii (Diptera, Agromyzidae). Cadmium decreased plant growth in a dose-dependent way. C. milii preferred the control to the cadmium-exposed plants for feeding and oviposition. Moreover, preference for the control plants increased with increasing cadmium exposure of the alternative choice. Adult and offspring performance decreased with increasing plant cadmium exposure. This suggests that, at least under our laboratory conditions, host choice of C. milii is adaptive under pollution stress. Foliar cadmium concentration increased and the soluble sugar concentration decreased with increasing cadmium exposure. Regression analysis showed that both latter components might be responsible for the decrease in performance of C. milii on cadmium-exposed plants. The protein and amino acid concentration of the leaves, the amount of structural defenses, and water concentration were not affected by the cadmium treatment.  相似文献

Exposure to specific metallic compounds can cause severe deleterious modifications in organisms. Fishes are particularly prone to toxic effects from exposure to metallic compounds via their environment. Species that inhabit estuaries or freshwater environments can be chronically affected by persistent exposure to a large number of metallic compounds, particularly those released by industrial activities. In this study, we exposed yellow eels (European eel, Anguilla anguilla) for 28 days to environmentally relevant concentrations of four specific metals; lead (300, 600, and 1,200 μg/l), copper (40, 120, and 360 μg/l), zinc (30, 60, and 120 μg/l) and cadmium (50, 150, and 450 μg/l). The selected endpoints to assess the toxicological effects were neurotransmission (cholinesterasic activity in nervous tissue), antioxidant defense, and phase II metabolism (glutathione-S-transferase [GST] activity, in both gills and liver tissues), and peroxidative damage. The results showed an overall lack of effects on acetylcholinesterase for all tested metals. Lead, copper, and cadmium exposure caused a significant, dose-dependent, increase in GST activity in gill tissue. However, liver GST only significantly increased following zinc exposure. No statistically significant effects were observed for the thiobarbituric acid reactive substances assay, indicating the absence of peroxidative damage. These findings suggest that, despite the occurrence of an oxidative-based response after exposure to lead, copper, and cadmium, this had no consequence in terms of peroxidative membrane damage; furthermore, cholinergic neurotoxicity caused by lead, copper, and cadmium did not occur. Male Sprague-Dawley rats were fed on different levels of cadmium mixed with purified diets containing iron or no iron for 8 weeks. The body weight gain, tissue weights, hemoglobin, hematocrit, liver and brain ATPase were measured at 2, 4 and 8 weeks after feeding. The hemoglobin and hematocrit values were the same in all rats fed on cadmium. The rats fed on iron-deficient diets mixed with cadmium showed a significant decrease in body weight gain. However, the rats receiving only the 100 ppm of cadmium in iron-sufficient diet showed a significant decrease in body weight gain. There were no significant changes in the weights of thymus, spleen, kidney, heart, brain and testes. However, the liver weights were decreased in the highest treatment of cadmium but the liver weight/body ratios were uneffected. Na+-K+ activated ATPase activity in brains of rats fed on cadmium were significantly decreased at 2, 4 and 8 weeks of treatment. The decrease was more pronounced in rats fed on iron-deficient diets. Oligomycin-sensitive (Mitochondrial) Mg2+ ATPase activity was also significantly decreased in liver and brain tissues of rats fed on cadmium. Oligomycin-insensitive Mg2+ ATPase activity, however, was not altered in any tissues tested. It appears that cadmium may be interfering with energy (ATP) production and utilization processes in rat brain and liver tissues.  相似文献

Phytoextraction has shown great potential as an alternative technique for the remediation of metal contaminated soils. The objective of this study was to investigate cadmium (Cd) phytoextraction ability of high biomass producing weeds in comparison to indicator plant species. The pot study conducted with 10 to 200 mg Cd kg(-1) soil indicated that Ipomoea carnea was more effective in removing Cd from soil than Brassica juncea. Among the five species, B. juncea accumulated maximum Cd, but I. carnea followed by Dhatura innoxia and Phragmytes karka were the most suitable species for phytoextraction of cadmium from soil, if the whole plant or above ground biomass is harvested. In the relatively short time of this experiment, I. carnea produced more than 5 times more biomass in comparison to B. juncea. There were significant differences (p < 0.05) between the shoot length and shoot mass of control and treated plants.  相似文献

Laboratory experiments were conducted to examine the impact of cadmium (Cd) on survival of Diplostomum sp. cercariae. Freshly emerged cercariae were exposed to Cd solutions at concentrations ranging between 0.2 and 200 microg/l. The mortality patterns and mean survival times (MSTs) of the exposed parasites were determined by the Kaplan-Meier estimator and were compared (Log-Rank test) to corresponding patterns and estimators of the unexposed controls. Cd concentrations >20 microg/l caused statistically significant changes in the mortality pattern of Diplostomum sp. cercariae and reduced MST by at least five hours compared to the control group, where MST was 52 h. The results show that longevity of the free-living Diplostomum sp. cercariae can be reduced by direct exposure to heavy metals. Cd contamination of aquatic habitats thus potentially can have an impact on parasite populations and communities.  相似文献

Based on a three compartment microcosm-water column, natural sediment,Hexagenia rigida nymphs-an experimental study was set up to compare cadmium (Cd) and methylmercury (MeHg) bioaccumulation by a burrowing mayfly species, after exposure via the water column or the sediment as initial contamination sources. Results from a wide concentration range for each exposure condition revealed very marked differences between the two metals: MeHg was readily accumulated from the two contamination sources, leading to important metal concentrations in the nymphs after the 2 weeks’ exposure; Cd bioaccumulation, on the other hand, was negligible when the metal was added to the water compartment, even though significant transfers were observed from the sediment source. The average Cd concentrations in the nymphs were proportional to the sediment contamination levels. Turbidity measurements in the water column, reflecting the bioturbation activity of the nymphs, revealed that the effect of Cd was significant, but only when the metal was initially added to the sediment. The results are discussed according to the uptake routes and the structural and functional properties of the biological barriers involved (gills and gut).  相似文献

The levels of copper, lead, chromium, zinc, cadmium, arsenic and silver were determined in periphyton specimens obtained with a diatometer collector. Stations selected were along three important bayous of the Calcasieu River system. Distributions of some metals in the organisms were similar to those found in sediment from the same locations, while other metals appeared to be similar to water concentrations. Concentration ratios of periphyton over sediment greatly exceeded one for the metals chromium, zinc, cadmium, arsenic and silver. The concentrations of heavy metals in the periphyton appeared to yield more information about pollutants than either water or sediment samples collected at the periphyton stations.  相似文献

The beet armyworm (Spodoptera exigua) were fed on artificial food contaminated with zinc (200 mg kg(-1) dry mass) or cadmium (66 mg kg(-1) dry mass) for 15 generations. In 15th generation, O2 output and CO2 production of pupae were measured. Exposure to cadmium did not cause any effects whilst exposure to zinc led to a significant increase in the respiration rate of pupae. Several field and laboratory assays were employed below an urban storm sewer outfall to define the relationship between stormwater runoff and contaminant effects. Specifically, two bioassays that measure feeding rate as a toxicological endpoint were employed in the field and in the laboratory, along with bioassays measuring survival and growth of test organisms. In 7 to 10 d in situ exposures, amphipod leaf disc processing, growth and survival were monitored. Different exposure scenarios were investigated by varying the mesh size (74 microns or 250 microns mesh) and method of deployment (water column, sediment surface, or containing sediment) of in situ exposure chambers. Hyalella azteca, Daphnia magna, and Pimephales promelas survival were monitored in 48 h in situ exposures. Feeding inhibition was investigated via enzyme inhibition of H. azteca and D. magna and via leaf disc processing measurements of the detritivore H. azteca. Additionally, we investigated the extent of phototoxicity at this site via field exposures in sun and shade and laboratory exposures with and without UV light. The measurement of detritivore leaf disc processing, and thus its usefulness as an endpoint, was hindered by individual variability in the amount of leaf consumed and by leaf weight gain during the summer field exposures. For D. magna, enzyme inhibition measured in a laboratory exposure did not reveal the toxicity observed in field exposures. For H. azteca, enzyme inhibition measured in the laboratory indicated toxicity similar to that observed in short term chronic in situ exposures. Enzyme inhibition also did not detect differences in toxicity due to variations in flow conditions. There were no statistically significant effects of any exposure on P. promelas survival or H. azteca growth, and there were no statistically significant effects due to mesh size or sun exposure. Survival of H. azteca was the most sensitive and the least variable endpoint. Effects on survival were noted in the same treatments over short-term chronic exposures in the laboratory and in situ. Significant differences in survival were noted due to the method of deployment under low flow conditions. In situ chambers containing sediment resulted in greater mortality in the 10 d low flow in situ experiments. Under high flow conditions, significant reductions in survival and leaf disc processing were noted under all methods of deployment at the two impacted sites over a 7 d exposure. Also under high flow conditions, significantly greater mortality of H. azteca was reported at the downstream field site when sediment was included in the chamber at deployment. These results suggest that significant toxicity at this site is due to accumulation of contaminants in the sediment and the mobilization of these contaminants during a storm event. In situ exposures detected toxicity not observed in laboratory exposures. These results suggest that a combination of laboratory and field bioassays is most useful in defining field effects.  相似文献

The role of algal concentration in the transfer of organic contaminants in a food chain has been studied using the ubiquitous model polycyclic aromatic hydrocarbon benzo[a]pyrene (BaP) as the contaminant, Isochrysis galbana as the phytoplankton food source, and the common mussel (Mytilus edulis) as the primary consumer. The effect of algal concentration on BaP uptake by M. edulis was determined by feeding M. edulis daily with I. galbana which had previously been kept in the presence of BaP for 24 h. Four combinations of concentrations of algae and BaP were used to give final exposure concentrations of 30,000 or 150,000 algal cells ml(-1) in combination with either 2 or 50 microg BaP l(-1). BaP concentrations were determined fluorometrically in rest tissues (excluding digestive glands) and digestive gland microsomal fractions of M. edulis after 1, 7 and 15 days exposure, and also in isolated algae. Potentially toxic effects of BaP on M. edulis were examined in terms of blood cell lysosomal membrane damage (neutral red dye retention assay) and induction of digestive gland microsomal mixed-function oxygenase (MFO) parameters [BaP hydroxylase (BPH) and NADPH-cytochrome c (P450) reductase activities]. BaP bioaccumulation in rest tissues (and to a lesser extent in digestive gland microsomes) of M. edulis increased with both increasing BaP and algal exposure concentrations, and over time, producing maximal bioconcentration factors in rest tissues after 15 days exposure to 150,000 algal cells ml(-1) and 50 microg BaP l(-1) of 250,000. The five-fold higher concentration of algae increased BaP bioaccumulation by a factor of approximately 2 for 50 microg BaP l(-1) at day 15. Blood cell neutral red dye retention time decreased linearly with increasing log(10) tissue BaP body burden, indicating an increased biological impact on M. edulis with increasing BaP exposure possibly due to a direct effect of BaP on blood cell lysosomal membrane integrity. An increase was seen in NADPH-cytochrome c reductase activity, and indicated in BPH activity, with 1 but not 7 or 15 days exposure to BaP, indicating a transient response of the digestive gland microsomal MFO system to BaP exposure.  相似文献

The objective of the present study is to examine the growth and energetic performance of juvenile turbot after exposure to contaminated sediment and during the subsequent recovery period with or without food limitation. We designed a two-step experiment by first exposing juvenile turbot to harbour sediment for 26 days and then transferring them to clean sea water with different frequencies of feeding for 35 days. Without food limitation, fish previously exposed to contaminated sediment compensated for weight, length and lipid reserve losses; we did not record any differences in size, Fulton’s K condition index and triacylglycerol/sterol (TAG/ST) ratio after the 35-day depuration period compared to the reference fish. This result could be related to the compensatory growth mechanism observed in a wide range of fish species following a period of growth depression. With food limitation during the 35-day depuration period, recovery growth was not sufficient to restore length and weight values similar to the reference fish. Moreover, turbot previously exposed to contaminated sediment and subsequently fed twice or once a week exhibited extremely low TAG/ST ratios, but the reference fish submitted to the same restrictive feeding conditions did not. This study indicates that juvenile fish affected by chemical pollution can improve their biological performance if pollution events are followed by a period of abundant food. However, if pollution events occur during periods of food scarcity, e.g. in winter, storage of energy reserves will be compromised.  相似文献

The goal of this study was to contribute to understanding of the mechanisms behind sensitivity differences between early and late instar larvae of Chironomus riparius and to address the influence of the differences in standard testing approaches on the toxicity evaluation. A 10-day contact sediment toxicity test was carried out to assess sensitivity to cadmium exposure in relation to different age and laboratory culture line origin of test organisms. Chironomid larvae of early (OECD 218 method) and late instar (US-EPA600/R-99/064 method) differed substantially in sensitivity of traditional endpoints (OECD: LOEC 50 and 10 μg Cd/g dry weight (dw); US-EPA: LOEC?>?1000 and 100 μg Cd/g dw for survival and growth, respectively). Bioaccumulated cadmium and metallothioneins (MTs) concentrations were analyzed to investigate the role of MTs in reduced sensitivity to cadmium in late instar larvae. Metallothioneins were induced after treatment to greater Cd concentrations, but their levels in relation to cadmium body burdens did not fully explain low sensitivity of late instars to cadmium, which indicates some other effective way of detoxification in late instars. This study brings new information related to the role of MTs in age-dependent toxicant sensitivity and discusses the implications of divergence in data generated by chironomid sediment toxicity tests by standardized methods using different instars.  相似文献

Heavy metals damage the structure, chemistry, and function of cells, including enzyme systems inside them. Variation in the profile of biochemical biomarkers in prevalent species should be used for assessing environmental contamination. The present study pays attention to the phosphatases present in the midgut of gypsy moth fourth instar caterpillars, which had been exposed to short- and long-term cadmium intake at 10 and 30 μg Cd/g dry food. Chronic cadmium ingestion significantly inhibited the activity of all examined phosphatases, while only the activity of lysosomal phosphatase was acutely decreased. Total acid phosphatase activity recovered from both long-term cadmium treatments within 3 days. The low index of phenotypic plasticity was connected to high variability of plasticity. Dependence of phosphatase isoforms on genotype and duration of cadmium treatment was determined. We concluded that, with further investigations, profiling of total acid phosphatase activity, as well as the lysosomal fraction can be used as a biomarker for acute sublethal metal toxicity.  相似文献

In this study, laboratory experiments were carried out in order to come to a better understanding of the fate of polycyclic aromatic hydrocarbons (PAHs) in the marine environment and especially on their bioaccumulation, biotransformation and genotoxic effects in fish. Juveniles of turbot (Scophthalmus maximus) were exposed to PAHs through different routes via (1) a mixture of dissolved PAHs, (2) a PAH-polluted sediment and (3) an oil fuel elutriate. Fish were exposed 4 days followed by a 6-day depuration period. In each experiment, PAH concentrations in the seawater of the tanks were analysed regularly by gas chromatography coupled with mass spectrometry. Muscle and liver samples were also analysed for parent PAH levels and PAH bioconcentration factors were calculated. Biotransformation was evaluated by measuring the levels of PAH metabolites in fish bile. Genotoxicity was assessed by the alkaline comet assay. Regardless of exposure route, the parent PAH concentrations in the liver and muscle showed a peak level 1 day after the beginning of the exposure, followed by a decrease up to the background level towards the end of the experiment, except for the exposure to dissolved PAHs for which levels were relatively low throughout the study. As a consequence, no bioaccumulation was observed in fish tissues at the end of the experiment. In contrast, regardless of exposure routes, a rapid production of biliary metabolites was observed throughout the whole exposure experiment. This was especially true for 1-hydroxypyrene, the major metabolite of pyrene. After 6 days of recovery in clean water, a significant decrease in the total metabolite concentrations occurred in bile. Fish exposed through either route displayed a significant increase in DNA strand breaks after 4 days of exposure, and significant correlations were observed between the level of biliary PAH metabolites and the level of DNA lesions in fish erythrocytes. Overall results indicate that exposure to either a mixture of dissolved PAHs, a PAH-contaminated sediment or a dispersed oil fuel elutriate leads to biotransformation and increase in DNA damage in fish. The quantification of PAH metabolites in bile and DNA damage in erythrocytes appear to be suitable for environmental monitoring of marine pollution either in the case of accidental oil spills or sediment contamination.  相似文献

A greenhouse trial investigated the uptake of cadmium and zinc by the bird-cherry oat aphid (Rhopalosiphum padi) feeding on wheat grown on sewage sludge amended soil. The trial was conducted at application rates of 0, 5, 7.5, 10, 15 and 20 tonnes ha(-1) dry solids. Concentrations of Cd and Zn were within current UK limits for potentially toxic elements in soils amended with sewage sludge. Cd and Zn in wheat plants were significantly greater than controls. Batches of aphids feeding on the wheat also showed a significant increase in the uptake of Cd and Zn. This study demonstrates a potential route of exposure to Cd and Zn for the predators of cereal aphids.  相似文献
