为了探讨甲醛暴露对小鼠骨髓组织的毒性影响,选用SPF级昆明雄性小鼠作为研究对象,用浓度为0.5,1.0,3.0mg/m3的气态甲醛对小鼠进行连续动态染毒72h,染毒后检测骨髓细胞中细胞周期变化、DPC(DNA-蛋白质交联)、DNA稳定性、骨髓细胞分化关键因子Nucleostemin和CYP1B1在各暴露浓度组中的表达情况.结果表明:甲醛暴露造成骨髓细胞的DNA损伤,其DNA稳定性和DPC效应均呈现良好的剂量-效应关系.骨髓细胞分化关键因子Nucleostemin表达量在各浓度染毒组中与空白组相比,1.0mg/m3浓度组有极显著性升高,而0.5和3.0mg/m3浓度组则是极显著性降低,与空白组相比CYP1B1基因在0.5,1.0,3.0mg/m3浓度组中表达,有极其显著性差异,并且1.0mg/m3浓度组上升较多.本次研究表明,甲醛暴露能影响小鼠骨髓组织细胞正常生长代谢.  相似文献

为研究吸入性甲醛的毒性能否进入动物骨髓组织,引起骨髓组织基因表达发生改变,选择小鼠某些原癌基因和抑癌基因为研究对象,以SPF级balb/c雄性小鼠为材料,采用动态吸入方式染毒2周(5+2模式),染毒浓度分别为0, 0.5, 3.0mg/m3,用半定量RT-PCR方法检测不同浓度甲醛对小鼠染毒后骨髓组织细胞中c-myc、MDM2和p53基因表达的变化.结果表明,在不同浓度的甲醛暴露条件下,与空白对照组相比,小鼠骨髓组织中的c-myc基因,MDM2基因和p53基因表达均发生改变,在3.0mg/m3浓度组与空白对照组存在显著差异(P<0.05), c-myc基因,MDM2基因呈现表达上调,p53基因则呈现表达下调.吸入性甲醛的毒性能进入动物骨髓组织,并能引起骨髓组织基因表达发生改变.  相似文献

为了探明骨炭粉对Cd污染土壤的修复效果及稳定性,以潮土与红壤Cd污染农田土壤为对象,基于土壤Cd有效态、作物降Cd率(DR,%)及降Cd失活率(IR,%)等指标,研究田间条件下施用3种浓度(0.25%、0.50%及1.0%)骨炭粉对Cd污染土壤的修复效果及120,360d后修复效果的稳定性.结果表明:骨炭粉处理均能有效促进潮土与红壤中作物生长,增加作物产量.施用骨炭粉能不同程度提高土壤中pH值、阳离子交换量(CEC)、全磷及有效磷含量;其中,0.5%和1.0%骨炭粉处理均显著(P<0.05)提高土壤pH值、土壤全磷及有效磷含量.与对照相比,不同骨炭粉处理120d后潮土和红壤中有效态Cd含量分别降低32.1%~48.4%、32.8%~56.2%,360d后土壤中有效态Cd仍然显著低于对照,降幅为26.9%~37.8%、25.8%~39.5%.对修复后三季作物Cd吸收测定结果表明,与对照相比,不同浓度(0.25%~1.0%)骨炭粉处理的第一季作物降Cd率(DR-Cd)分别达36.0%~60.4%、33.9%~58.2%,作物降Cd率随骨炭粉施用量增加而增加.基于降Cd失活率(IR-Cd)对骨炭粉修复效果的稳定性测定结果表明,潮土与红壤施用骨炭粉360d后,第三季作物降Cd失活率IR-Cd为21.2%~34.7%和6.2%~21.6%,表明在酸性红壤中修复效果的稳定性大于潮土.综上骨炭粉对Cd污染土壤的修复效果及其稳定性结果表明,骨炭粉在Cd污染农田土壤修复中具有较大应用价值.  相似文献

对裂变产物~(170)Tm的体内蓄积特性和诱发细胞突变效应的监测结果表明,当机体摄入~(170)Tm后,在短时间内即能迅速进入并滞留于骨组织中,其滞留量占体内各器官组织中的首位呈现高度选择性亲骨特征,在机体摄入~(170)Tm后的不同阶段,骨组织中的累积吸收剂量随着观察时间的延长而增升,其诱发骨髓细胞的染色体畸变率亦相应增高。由~(170)Tm内污染所诱发的染色体结构异常为染色单体型畸变,其中主要为染色单体断裂。随着骨组织中累积吸收剂量的增升,可观察到在一个细胞中同时有两个畸变发生,也诱发个别的染色体易位。  相似文献

为了探究甲醛(Formaldehyde,FA)对造血组织的毒性,分析甲醛的毒性是否与中间代谢产物——甲醇(Methanol,MeOH)的毒性一致,以72只昆明小鼠为受试动物,随机分为6组〔①对照组,仅灌服生理盐水;②FA40组,单独灌服甲醛(FA),w(FA)为40 mg/kg;③ MeOH40组,单独灌服甲醇(MeOH),w(MeOH)为40 mg/kg;④FA10+MeOH30组,同时灌服甲醛和甲醇,w(FA)为10 mg/kg、w(MeOH)为30 mg/kg;⑤FA20+MeOH20组,同时灌服甲醛和甲醇,w(FA)为20 mg/kg、w(MeOH)为20 mg/kg;⑥FA30+MeOH10组〕,同时灌服甲醛和甲醇,w(FA)为30 mg/kg、w(MeOH)为10 mg/kg,连续灌胃7 d,检测小鼠的肝脏氧化损伤程度、血液和骨髓内甲醛含量以及甲醛脱氢酶的相对表达量,比较甲醇和甲醛毒性的差异性.结果表明:与对照组相比,各染毒组小鼠肝脏内ROS(reactive oxygen,活性氧)含量极显著增加(P<0.01),FA30+MeOH10组的ROS含量显著上升(P<0.05);肝脏内MDA(malondialdehyde,丙二醛)含量极显著下降(P<0.01),GSH(glutathione,谷胱甘肽)含量极显著升高(P<0.01);在甲醛和甲醇联合染毒组中,随着甲醛含量的增加和甲醇含量的减少,氧化损伤程度呈上升趋势.AHMT(4-amino-3-hydrazine-5-mercapto-1,2,4-triazole)法测定结果显示,各染毒组小鼠血液和骨髓中甲醛含量与对照组相比均无显著差异;RT-PCR法测定结果显示,各染毒组小鼠中甲醛脱氢酶的相对表达量较对照组显著增加;FA40组与MeOH40组甲醛脱氢酶的相对表达量差异显著(P<0.01);与FA30+MeOH10组相比,FA10+MeOH30组中甲醛脱氢酶的相对表达量极显著增加(P<0.01),甲醛和甲醇联合染毒组中甲醛脱氢酶的相对表达量随着甲醛含量的增加和甲醇含量的减少呈上升趋势.研究显示,甲醛和甲醇的毒性不一致,甲醛对造血组织的毒性可能存在其他方式.   相似文献

小鼠吸入过滤或未过滤的柴油机排出物,染毒8h/d,分别染毒2、5、8d后,发现小鼠骨髓细胞微核率明显升高。吸入过滤后排出物的小鼠骨髓细胞微核出现率低于未过滤组。柴油机排出物对小鼠染色体有一定的损伤作用,过滤后的柴油机排出颗粒物中,致染色体损伤的物质减少,不同吸入时间(2、5、8d)对小鼬骨髓细胞微核出现率,没有显著差异。  相似文献

We have carried out first-trimester prenatal diagnosis of hypophosphatasia in 1 6 pregnancies with a 1 in 4 risk of this condition. The liver/bone/kidney isoenzyme of alkaline phosphatase was measured in chorionic villus samples using a specific monoclonal antibody and an enzymatic amplification system. Fifteen of the 16 pregnancies were correctly predicted, while one has been lost to follow up. We suggest that this assay system is likely to be superior to DNA-base methods for the first-trimester prenatal diagnosis of hypophosphatasia.  相似文献

以淡水养殖鱼塘中的鱼类为研究对象,采用碱化乙酸乙酯超声提取样品中的氯霉素(CAP),用C18 SPE固相萃取净化,液相色谱-串联质谱(HPLC-MS/MS)法进行分析,研究了同一鱼塘4种鱼类鱼肉样品及福寿鱼不同器官或组织对氯霉素的蓄积能力,初步评价其安全性.结果表明:同一淡水养殖鱼塘中4种鱼肉样品w(CAP)分别是福寿鱼(0.40 μg/kg)>青鱼(0.22 μg/kg)>鲢鱼(0.15 μg/kg)>鲮鱼(0.13 μg/kg),其中福寿鱼w(CAP)超过欧盟和美国FAD规定的检出限(0.3 μg/kg),而青鱼、鲢鱼和鲮鱼符合该检测标准,可安全食用;福寿鱼不同器官或组织的w(CAP)从高到低依次是鱼鳃1.07 μg/kg,鱼头0.79 μg/kg,鱼肠0.67 μg/kg,鱼鳞0.52 μg/kg,鱼肉0.40 μg/kg,鱼骨0.13 μg/kg,只有鱼骨中的w(CAP)符合欧盟和美国FAD规定的安全标准.   相似文献

湖北省鄂西自治州恩施市为我国第一个高硒区,硒源于石煤.州内还有低硒环境并曾有克山病流行,将硒均值为100mg/kg的石煤颗粒施入低硒土壤后,作物硒及人畜内环境硒水平有明显的剂量效应关系.根据足硒区粮食、饲草及人发硒的推荐值,每公顷施石煤15×10~3-18×10~3kg为宜.此外,讨论了这一措施得以增产的可能因素和该措施可能维持有效作用的年限.  相似文献

 Metal implants are the preferred materials to generate articular prostheses, plates, or bone pegs in orthopedic surgery. Although titanium and titanium alloys show a relatively good biocompatibility, clinical experience revealed that coating of the metallic implant surface may increase the biocompatibility. In a search for optimum bone implant surfaces, we determined polarity and contact angle parameters of a variety of polymers and substances and correlated the findings in a biocompatibility assay using an in vitro bone cell model. We report that an optimum adherence of SAOS-2 cells to such surfaces and a good vitality for polymers are characterized by water-based contact angles of 80° and 20° for advancing and receding probes, respectively. 农田土壤Cd污染是我国最为突出的环境问题之一,开展新型钝化修复Cd污染土壤材料的研究和技术开发对保障农产品安全及保护人体健康具有重要意义.以北京某蔬菜生产基地设施大棚Cd污染土壤为试验对象,温室条件下采用盆栽试验,研究牛骨炭与伊/蒙黏土复合组配的改良剂对Cd污染土壤的钝化效果,探讨不同配比的组配改良剂对土壤Cd有效态含量以及小白菜Cd吸收量的影响.结果表明:在污染土壤中添加1%、2%和5%的组配改良剂,可明显降低土壤有效态Cd含量及小白菜对Cd的吸收,土壤有效态Cd含量降幅最大值达42.3%,小白菜地上部分Cd含量降幅最大值达22.7%,组配改良剂中骨炭成分比例越高,钝化效果越好,尤其以添加5%组配改良剂(添加组配改良剂中牛骨炭含量为50%)对Cd的稳定化效果最佳,且不同组配改良剂处理均不会对小白菜生长产生不良影响,表明牛骨炭与伊/蒙黏土组配的改良剂在修复Cd污染土壤上具有较好的应用潜力.研究显示,牛骨炭与伊/蒙黏土组配的改良剂符合绿色修复技术的发展要求,可作为钝化修复农田重金属污染土壤的环保新材料.   相似文献

以鱼骨粉(FBM)为原料,利用NaOH处理制备高性能改性鱼骨粉材料(FBMT),通过比表面积仪(BET)、X射线衍射仪(XRD)、傅里叶红外光谱(FTIR)、扫描电镜-光电子能谱(SEM-EDS)、透射电镜(TEM)等手段对吸附前后材料形貌、孔结构、功能基团进行表征分析.结果表明,与FBM相比,FBMT出现一定数目中孔和大孔,具备了高孔容和平均孔径,分别增加68.57%和24.69%.FBMT材料表面-OH、C=O和PO43-等官能团含量增加,显示出一定的缓冲性能.FBM和FBMT对Cd(Ⅱ)的吸附经历快速的分配和慢速的吸附两个阶段,符合准二级动力学模型,相关系数R2分别达到0.8635及0.9480.Langmuir模型较好地拟合FBM和FBMT对Cd(Ⅱ)的等温吸附过程,对Cd(Ⅱ)饱和吸附量由改性前的9.28mg/g增至改性后的22.47mg/g,对应的吸附强度KF值也随之增加.分离因子0 < RL < 1表明Cd(Ⅱ)的去除过程为有利吸附.功能材料与Cd(Ⅱ)的作用机理主要包括阳离子专性吸附、静电吸附、离子交换、表面络合及沉淀作用.以上研究为鱼骨粉修复Cd污染提供一种经济有效的方法.  相似文献

2267 singleton fetuses who had one ultrasound examination between 15 and 21 weeks' gestation were prospectively evaluated for echogenic small bowel. Thirty-two cases of echogenic small bowel were detected—a prevalence of 1·4 per cent. Echogenic fetal small bowel was divided into two grades: grade 1, where the small bowel was more echogenic than the liver; and grade 2, where the small bowel had the echogenicity of bone. In contrast to 19/23 fetuses with grade 1 small bowel echogenicity, only 2/9 fetuses with grade 2 echogenic bowel had a normal pregnancy outcome (Fisher's exact test; P⩽0·01). Complications associated with second-trimester echogenic small bowel included in utero cytomegalovirus infection, second-trimester growth restriction, intrauterine fetal demise, and chromosomal abnormalities. Second-trimester fetal echogenic small bowel is associated with an increased risk of an adverse outcome. The prevalence of perinatal and neonatal complications is significantly greater when small bowel echogenicity approaches that of bone.  相似文献

对乌鲁木齐市具有代表性的民用住宅室内氡浓度进行了连续一年的监测调查,调查结果显示室内氡浓度平均值55.4 Bq/m3,低于国标的“200 Bq/m3”,并据此估算出其所致居民的年均有效剂量为1.400 mSv,以及所致居民支气管上皮组织、肺、性腺、骨髓及骨表面细胞等关键组织的年吸收剂量分别为1886.49,378.96,2.80,3.20,3.20μGy/a,大于UNSCEAR2000年报告的世界平均值1.008 mSv,结合室内氡的主要来源,提出了对建筑材料进行放射性监测、控制装修、工程竣工后开展室内环境监测、加强室内通风以及开展室内氡污染治理等措施控制室内氡浓度水平。  相似文献

Laboratory water flumes are artificial troughs of moving water widely used in hydraulic studies of fluvial systems to investigate real-world problems at smaller, more manageable scales. Water flumes have also been used to understand bone transportation sorting and bone orientation found in the fossil record using actual bones. To date, these studies have not involved scaled bones. A 1/12 scale model of a 21.8-m long skeleton of Apatosaurus, a long-necked sauropod dinosaur from the Late Jurassic, was used to explore three problems at Dinosaur National Monument (USA) that cannot be explained by tradition bone flume studies: (1) why there is an abrupt bend in articulated vertebrae, (2) why articulated dorsals are inverted relative to the pelvis, and (3) how bone jams form. The flume experiments established that (1) bed friction with the wing-like transverse processes of vertebrae resists the force of the water flow, whereas those vertebrae lacking the processes are free to pivot in the flow; (2) elevation of the dorsal vertebrae by the transverse processes subjects the vertebrae to the energy of the flow stream, which causes the vertebrae to flip. Computation fluid dynamics (CFD) software shows this flip was due to differential pressure on the upstream and downstream sides. (3) The formation and growth of bone clusters or jams (analogous to log jams in rivers) occur as transported bones pile against an initial obstruction and jammed bones themselves become obstacles. These preliminary studies show that scale models can provide valuable insights into certain taphonomic problems that cannot be obtained by traditional bone flume studies.

Objectives The aim of this study was to evaluate the role of nasal bone assessment in first-trimester screening for Down syndrome (DS) in the Korean population. Methods From July 2004 to March 2006, we prospectively evaluated the fetal nasal bones at 11–14 weeks' gestation in the Korean population. Results A successful evaluation was possible in 6490 of 6787 fetuses (95.6%). Absent, hypoechoic, and short nasal bones were seen in 4 (26.7%), 4 (26.7%), and 1 (6.7%) of 15 fetuses with DS, respectively, whereas in 5 (0.1%), 11 (0.2%), and 246 (3.8%) of 6456 normal fetuses. The incidence of absent and hypoechoic nasal bone showed significant differences between normal fetuses and fetuses with DS (P < 0.0005, both). Screening for DS using an absent or hypoechoic nasal bone resulted in a sensitivity of 53.3%, a specificity of 99.8%, a positive likelihood ratio of 215.2, and a negative likelihood ratio of 0.5. Conclusion Our study showed that nasal bone abnormality at 11–14 weeks of gestation had a high association with DS in the Korean population. This suggests that nasal bone assessment can be used to supplement the current first-trimester screening for DS in the Korean population. Copyright © 2007 John Wiley & Sons, Ltd.  相似文献

During the Mesozoic, one of the most significant evolutionary processes was the secondary adaptation of tetrapods to life in water. Several non-related lineages invaded from the terrestrial realms and from the oceans of the entire world. Among these lineages, ichthyosaurs were particularly successful. Advance parvipelvian ichthyosaurs were the first tetrapods to evolve a fish-shaped body profile. The deep skeletal modifications of their bodies, as well as their biology, depict advance ichthyosaurs as the paradigm of secondary adaptation of reptiles to marine life. Functional inferences point to them as off-shore cruising forms, similar to a living tuna, and some of them were capable of deep diving. Bone histology of some genera such as Temnodontosaurus, Stenopterygius, Ichthyosaurus, and Caypullisaurus, characterized by overall cancellous bone, is consistent with the idea of a fish-shaped ichthyosaurs as fast and far cruisers. Here, we provide histological examination of the ribs of the Middle Jurassic parvipelvian Mollesaurus. Contrasting with the bone histology of other parvipelvian, Mollesaurus ribs are characterized by a compact and thick cortex. Our data indicate that the rib cage was heavy and suggest that not all advanced ichthyosaurs were fast cruisers. The compact and dense ribs in these parvipelvian show that advance ichthyosaurs were ecologically more diverse than previously thought and that the lightening of the skeleton reversed, as also occurred in the evolution of cetacean, at least once along the evolutionary history of ichthyosaurs.  相似文献

采用现场监测,色-质联用分析以及小鼠骨髓微核试验方法,评价了兰州市交通要道颗粒物的污染状况,分析了颗粒提取物(PE)的有机组成及致突变活性。结果表明,兰州市总悬浮颗粒物(TSP)污染严重。颗粒提取物中含有近100种有机化合物,其中包括致癌性多环芳烃。微核试验显示PE具有致突变活性,对人群健康的影响不容忽视。  相似文献
