A cytogenetic analysis by several staining techniques was carried out on specimens of Seriola dumerili from the Tyrrhenian coast of Sicily. The karyotype was found to be composed of 48 chromosomes in all the specimens examined. All chromosomes are subtelo- and acrocentrics, except for one pair of small-sized submetacentric chromosomes. After Ag-staining, nucleolus organizer regions (NORs) can be observed on the short arms of a medium-sized subtelocentric chromosome pair, and after chromomycin A3-staining a bright fluorescent signal is evident at the same sites. The 18S rDNA mapping by fluorescence in situ hybridization confirms this unique NOR location. The constitutive heterochromatin is mainly restricted to centromeres and the fluorochrome-staining by chromomycin A3 and 4′,6-diamidino-2-phenylindole revealed a uniform base composition along the chromosomal arms; therefore stock/population-specific marker chromosomes cannot be identified.  A cytogenetic analysis was carried out on specimens of Epinephelus marginatus (Lowe, 1834) from three localities in the Mediterranean Sea, to deepen knowledge of the chromosome complement of the species and identify any possible population-specific cytogenetic markers. All specimens had a 2n = 48 acrocentric karyotype with two Ag-, chromomycin A3- and C-positive NORs (nucleolar organizer regions) in the subcentromeric region of the smallest chromosome pair. The constitutive heterochromatin was distributed centromerically. Except for NORs, neither eu- nor heterochromatin show fluorescence after fluorochrome staining, i.e. there is no localized increase of AT- or GC-rich DNA. In all populations, the (TTAGGG) n telomeric sequences are restricted to the telomeres, and the 18S and the 5S rDNA clusters are located on different chromosome pairs. In specimens from Sardinia, additional signals after C-banding, Ag-staining and FISH (fluorescence in situ hybridization) with 18S rDNA can be observed in the telomeric region of one or two large-sized chromosomes, classified as No. 2. This suggests that additional and variable NORs could be detected in the species in addition to those on the genus-specific, NOR-bearing Chromosome Pair 24. Chromosome preparations from gill tissue of the Australian flat oyster Ostrea angasi Sowerby were studied with conventional Giemsa staining, C-banding and silver staining techniques. A diploid complement of 2n = 20 was observed, consisting of five metacentric, three submetacentric and two subtelocentric pairs. Constitutive heterochromatin was distributed as large centromeric blocks in Chromosome Pairs 3, 6, 8, 9 and 10. The nucleolus organizer regions (NORs) were located terminally on long arms of Chromosome Pairs 9 and 10. This allowed the identification of homologous chromosomes in submetacentric and subtelocentric pairs. Intraspecific variability in NOR pattern as revealed by differences in the number of silver-stained NORs (Ag-NORs) per cell was found to be very common. Comparison of the patterns of karyotype, C-band and Ag-NORs between species of the larviparous oysters for which data have been published demonstrate that the chromosomal structure of the endemic Australian and New Zealand species O. angasi shows little similarity to the Southern Hemisphere oysters O. (Eostrea)puelchana Orbigny and Tiostrea chilensis (Philippi) and the Indo-West Pacific oyster O. denselamellosa, but very high resemblance to the European species O. edulis.  A cytogenetic analysis by several staining techniques was carried out on Oedalechilus labeo specimens from the west coast of Italy, in order to extend the karyological knowledge on Mediterranean mullets. The karyotype of the species is composed of 46 acrocentric and 2 subtelocentric chromosomes. Constitutive heterochromatin was found to be restricted to the centromeres. Fluorochrome staining revealed a uniform base composition along the chromosome arms. Nucleolus organizer regions (NORs) are located on the short arms of chromosome pair 9 and can be either homomorphic or quite heteromorphic in size. In some specimens chromomycin A3-staining and fluorescence in situ hybridization with 18S rDNA revealed a third additional and inactive NOR on the short arms of a medium-sized chromosome. This is the first report of NOR variability in Mugilidae, a family generally characterized by a quite conservative karyotype. Heterochromatin composition and NOR location differentiate O. labeo from a cytotaxonomical point of view from the Liza species studied, Liza being considered the genus from which the genus Oedalechilus derived. Chromosomes of Perna viridis were characterized by karyotype analysis, C-banding and nucleolus organizer regions (NORs). The diploid chromosome number was confirmed as 30 and the karyotype is composed of ten metacentric and five submetacentric chromosome pairs. Constitutive heterochromatin blocks were found on all chromosomal pairs except chromosomal pair 15, which showed uniform staining throughout the entire chromosomes. Silver staining revealed nucleolus organizer regions on telomeric region of four chromosomal pairs, viz. 1, 3, 7 and 11. This is the first comprehensive study undertaken on chromosomes of Perna viridis.  相似文献

The genetic structure of Mycedium elephantotus (Pallas, 1766) populations from three regions around Taiwan was examined using allozyme electrophoresis. Eight loci were scored from seven enzyme systems. Seven loci were polymorphic under the 95% criterion. The high ratio of observed to expected genotypic diversities at the collection sites (G O:G E=0.8 to 1.0) indicate that M. elephantotus propagates predominantly by sexual reproduction. Allele frequencies of M. elephantotus differed significantly among regions (D=0.024 to 0.256, F ST=0.032 to 0.218, p < 0.001), while populations among collection sites within each region were homogeneous (D=0.000 to 0.015, F ST=0.010 to 0.022, p > 0.05). Genetic differentiation between populations from southern Taiwan and the Penghu Islands is greater than that between southern and northern Taiwan populations, although the former two regions are much closer geographically. The UPGMA dendrogram based on Nei's unbiased genetic distance showed a clear subdivision of populations into two groupings, northern Taiwan/Penghu Islands and southern Taiwan. A higher level of gene flow was found between M. elephantotus populations in northern Taiwan and the Penghu Islands (N m=7.56) than that between populations in southern Taiwan and other regions (N m=0.90 to 1.72). The pattern of genetic subdivision among regions is consistent with the pattern of ocean currents, indicating that genetic differentiation is likely driven by surface circulation vicariance. A karyotypic analysis was carried out in turbot (Scophthalmus maximus) using conventional banding techniques, fluorochromes and restriction endonucleasebanding. The standard karyotype of turbot is 2n=44 chromosomes, with 48 chromosome arms. Nucleolar organizer regions, which were localized in the short arm of Chromosome Pair No. 3, showed C-and chromomycin A3-positive staining. C-bands were mainly located at the centromeres, but were also present in some interstitial and telomeric locations. No differences were revealed among constitutive heterochromatin bands after treatment of fixed chromosomes with fluorochrome staining and restriction endonucleases. Digestion with DdeI restriction enzyme produced a pattern of interstitial banding which suggested some degree of differentiation along the chromosome arms in turbot. Neither variation in chromosome number of arm number nor polymorphism in nucleolar organizer regions was revealed in the turbot analyzed.  相似文献

The aim of this study was to determine whether the composition of the demersal fish fauna in coastal marine waters in temperate Australia changes markedly with increasing water depth and distance from the shore and whether the composition of the fish fauna in water depths of 5 to 35 m undergoes cyclic, seasonal changes. Samples of demersal fishes were therefore collected by trawling over the predominantly sandy substrate at nine sites located in water depths of 5 to 15 m or 20 to 35 m and within 20 km of the shore in four regions along ∼200 km on the lower west coast of Australia. The sampling regime involved trawling for fishes at each site at night in seven consecutive seasons between the summer of 1990/1991 and winter of 1992. A total of 72 435 fishes, representing 77 families, 143 genera and 172 species was caught. The compositions of the fish faunas in offshore waters with depths of 5 to 35 m were shown to differ markedly from those previously recorded for nearshore marine waters in the same regions. However, as some species, such as Sillago burrus, S. vittata, S. bassensis and Rhabdosargus sarba, increase in size, they move out from their nursery areas in nearshore waters into deeper and more offshore waters, where spawning occurs. Ordination showed that, in each of the four regions, the composition of the fish fauna in depths of 5 to 15 m differs from that in depths of 20 to 35 m. This difference is attributable to the fact that some species, such as  S. burrus, S. vittata and Upeneichthys lineatus, are far more abundant in depths of 5 to 15 m, whereas other species, such as S. robusta, U. stotti and Lepidotrigla modesta, occur predominantly in depths of 20 to 35 m. However, the samples collected from the single site that was inshore but in deeper water demonstrate that the composition of the fish fauna is influenced by distance from shore as well as by water depth. The compositions of the fish faunas differed with latitude, largely due to the fact that some subtropical species, such as Polyspina piosae, S. burrus and  S. robusta, did not extend down into the more southern regions. Ordination also showed that the composition of the fish faunas at all but one of the nine sites underwent pronounced and consistent cyclic, seasonal changes. This seasonal cyclicity at the different sites was attributable to sequential patterns of immigrations and emigrations by a number of fish species during the course of the year. These seasonal migrations involved, inter alia (1) movements of certain species from their nursery areas into these deeper waters, e.g.  S. bassensis and Scobinichthys granulatus; (2) migrations into and off the sandy areas of the inner continental shelf, e.g. Arnoglossus muelleri; (3) migrations to spawning areas, e.g. Sillago robusta; and (4) movements into areas where detached macrophytes accumulate in winter, e.g. Cnidoglanis macrocephalus and Apogon rueppellii.  The European fanworm Sabella spallanzanii (Gmelin, 1791) was recently introduced to Port Phillip Bay and is now a conspicuous component of most benthic communities. Reproduction of the worm was investigated in a population at Queenscliff over a 2 yr period (October 1995 to October 1997) using gonadal histology. The worms are dioecious (sex ratio 1:1, n=250), and attained sexual maturity at ∼50 mm body length. Reproductive periodicity followed a distinct annual cycle, and spawning proceeded through an extended autumn/winter period. Spawning was broadly synchronous between sexes, and coincided with falling seawater temperatures and shorter day-lengths. The females were highly fecund, and >50 000 eggs were probably shed from large females (>300 mm body length) during the annual spawning period. Breeding cycles of S. spallanzanii in Port Phillip Bay are ∼6 mo out of phase with endemic populations located at similar latitudes in the northern hemisphere. The spread of S. spallanzanii within Port Phillip Bay has been monitored by divers on an annual basis since 1994. The most recent dive survey (1998) indicates that S. spallanzanii has extended its range through out the entire 2000 km2 embayment, and has invaded most subtidal habitats. Quantitative estimates of S. spallanzanii abundances were highest on pier pylons (12.5 individuals m−2, 0.5 to 7 m depths). On sediments, estimates were highest at shallow sites (0.3 m−2, 7 m depth), but numbers declined significantly with depth (0.1 m−2, 17 to 22 m depth). Mean worm lengths and biomass were, by contrast, significantly higher at intermediate depths (12 to 17 m) than in shallower (7 m) or deeper (22 m) locations. S. spallanzanii demonstrates a clear preference for growth in sheltered, nutrient-enriched waters, so it may not spread from Port Phillip Bay into the adjacent oceanic waters of Bass Strait; however, in view of S. spallanzanii's current high abundance, fecundity and extended spawning periodicity, there is a high risk of future range expansions, mediated by shipping, into other temperate-water ports. The genetic structure of 12 reef populations of the soft coral Sinularia flexibilis (Octocorallia, Alcyoniidae) was studied along the Great Barrier Reef (GBR) at a maximum separation of 1,300 km to investigate the relative importance of sexual and asexual reproduction, genetic differentiation and gene flow among these populations. S. flexibilis is a widely distributed Indo-Pacific species and a gamete broadcaster that can form large aggregations of colonies on near-shore reefs of the GBR. Up to 60 individuals per reef were collected at a minimum sampling scale of 5 m at two sites per reef, from December 1998 to February 2000. Electrophoretic analyses of nine polymorphic allozymes indicated that genotypic frequencies in most populations and loci did not differ significantly from those expected from Hardy–Weinberg predictions. Analysis of multi-locus genotypes indicated a high number of unique genotypes (N go) relative to the number of individuals sampled (N) in each reef population (range of 0.69–0.95). The maximum number of individuals likely to have been produced sexually (N*) was similar to the number of individuals sampled (i.e. N*:N ˜ 1), suggesting that even repeated genotypes may have been produced sexually. These results demonstrated a dominant role of sexual reproduction in these populations at the scale sampled. Significant genetic differentiation between some populations indicated that gene flow is restricted between some reefs (F ST=0.026, 95% CI= 0.011 − 0.045) and even between sites within reefs (F ST=0.041, 95% CI=0.027 − 0.055). Nevertheless, there was no relationship between geographic separation and genetic differentiation. Analyses comparing groups of populations showed no significant differentiation on a north-south gradient in the GBR. The pattern in the number of significant differences in gene frequencies in pairwise population comparisons, however, suggested that gene flow may be more restricted among inner-shelf reef populations near to the coast than among mid/outer-shelf populations further from the coast. Samples of the scleractinian coral Pocillopora damicornis were collected from six sites located around four islands in the Ryukyu Archipelago, southern Japan, and subjected to allozyme electrophoresis. Seven polymorphic loci were examined for their allelic patterns. The ratio of observed to expected genotypic diversity (0.30 < G o :G e  < 0.64), the ratio of the observed number of genotypes to the number of individuals (0.47 < N g :N i  < 0.75), and deviations from Hardy–Weinberg equilibrium indicated that asexual reproduction plays a major role in the maintenance of established populations. However, populations were not completely dominated by a single or a few clones, and most clones were represented by only a few individual samples. The high frequency of typhoons in the region suggests that, in P. damicornis, fragmentation caused through occasional exposure to powerful waves is a major mode of asexual reproduction, but asexual production of planulae may also be contributing to the maintenance of populations. A significant genetic differentiation (F ST) was found between the six populations examined (0.027 < F ST < 0.092, average F ST = 0.056). The moderate gene flow is discussed according to characteristics of the larval stage of the species, and to circulation patterns in the region. We studied the correlation between male-male aggressive behavior of the spider mite, Schizotetranychus miscanthi and winter coldness, a factor hypothesized to correlate with the degree of relatedness of the interacting males through the frequent occurrence of mother-son mating in the spring. Thirty-one samples from various populations throughout Japan were added to previous data. A cluster analysis revealed two clinal trends for male-male aggression in S. miscanthi in regions of light or no snowfall. The two clines may represent a dimorphism within the species, each having a similar clinal trend within Japan. Further support for the hypothesis that male overwintering survival affects male-male aggressiveness came from a comparison between regions of heavy snowfall and those of light or no snowfall. In regions with heavy snow insulation, overwintering survival may not be as high as in other areas, and there was no correlation between male aggressiveness and the minimum winter temperature. The abundance, biomass and distribution of Solenicola setigera, a colonial heterotrophic protist found only with the centric chain-forming diatom Leptocylindusmediterraneus, are reported for four major ocean basins. The distribution is cosmopolitan, and abundances and biomass are usually low (<500 colonies l−1); however, in the summer of 1993, we observed a major biomass component (range = 5 to 31 μg C l−1) in the surface waters of the North Atlantic attributable to S. setigera. These colonies of S. setigera were exceptionally large, and unusual in possessing high abundances of Synechococcus sp., a normally solitary cyanobacterium, embedded in the matrix covering the cells. We have examined the variability and potential adaptive significance of the wavelengths of light produced by gelatinous zooplankton. Bioluminescence spectra were measured from 100 species of planktonic cnidarians and ctenophores collected between 1 and 3500 m depth. Species averages of maximal wavelengths for all groups ranged from 440 to 506 nm. Ctenophores (41 species) had characteristically longer wavelengths than medusae (34 species), and the wavelengths from siphonophores (25 species) had a bimodal distribution across species. Four species each produced two different wavelengths of light, and in the siphonophore Abylopsistetragona these differences were associated with specific body regions. Light from deep-dwelling species had significantly shorter wavelengths than light from shallow species in both ctenophores (p = 0.010) and medusae (p = 0.009). Although light production in these organisms was limited to the blue-green wavelengths, it appears that within this range, colors are well-adapted to the particular environment which the species inhabit. This paper reports the results of cytogenetic analyses carried out on Porichthys plectrodon using conventional Giemsa staining, C-banding and silver staining techniques. A diploid chromosome count of 2n=44 was observed, consisting of 8 metacentric, 10 submetacentric, 6 subtelocentric and 20 acrocentric chromosomes. Differences in length made it possible to identify homologous chromosomes within the metacentric group. Constitutive heterochromatin was distributed as large pericentromeric blocks in pairs 1 and 2, while the rest of the chromosomes were marked in centromeric regions, some more conspicuously than others. Ammonium concentrations of ∼1 M are commonly cited as being the threshold for inhibition of NO3 uptake, but the applicability of this threshold to phytoplankton from different taxonomic classes has rarely been examined. Additionally, little is known about the influence of environmental variables (e.g. growth temperature) on the interaction between ambient NH4 + and NO3 uptake. Four species of estuarine phytoplankton, two diatom [Chaetoceros sp., and Thalassiosira weissflogii (Grunow) Fryxell et Hasle] and two dinoflagellate [Prorocentrum minimum (Pavillard) Schiller, and Gyrodinium uncatenum Hulburt], were grown on NO3 at several different temperatures (4, 10, 15, or 20 °C), and the impact of NH4 + additions on NO3 uptake/assimilation (non-TCA-extracted) and assimilation (TCA-extracted) was assessed. For all species at all temperatures, NO3 uptake/assimilation and assimilation rates decreased in a roughly exponential manner with increasing NH4 + concentrations but were not completely inhibited even at elevated NH4 + concentrations of 200 μM. Estimated half-inhibition concentrations (K i) were significantly greater in the diatom species (mean ± SE; 2.70 ± 0.67 μM) than in the dinoflagellate species (1.26 ± 0.55 μM). Half-inhibition constants were positively related to temperature-limited relative growth rate although not significantly. The observed inhibition of NO3 uptake and assimilation, as a percentage of NO3 uptake in the absence of NH4 +, averaged about 80% and ranged from 49 to 100%. For all species, a significant (P < 0.001) positive correlation was found between percent inhibition of NO3 assimilation and temperature-limited relative growth rate. Two experiments on Chesapeake Bay phytoplankton during an April 1998 diatom bloom showed that in short-term (∼1 h) temperature manipulation experiments, percent inhibition of NO3 uptake/assimilation was also positively related (P = 0.05) to experimental temperature. The observed relationships between temperature-limited relative growth rate and percent inhibition of NO3 assimilation rates for the species tested suggest that at the enzyme level, the inhibitory mechanism of NO3 assimilation is similar among species, but at the whole cell level may be regulated by species-specific differences in the accumulation of internal metabolites. These findings add not only to our understanding of species-specific variability and the role of growth temperature, but also provide additional data with which to evaluate current models of NH4 + and NO3 interactions. Using a biopsy dart system, samples of skin tissue were collected from southern right whales (Eubalaena australis) in 1995 on two wintering grounds, southwest Australia (n = 20) and the Auckland Islands of New Zealand (n = 20); and on offshore feeding grounds at Latitudes 40 to 43°, south of Western Australia (n = 5). A variable section of the mitochondrial DNA control-region (289 nucleotides) was amplified and sequenced from these 45 individuals (21 males, 20 females and 4 of unknown sex), distinguishing a total of seven unique sequences (i.e. mtDNA haplotypes). Two haplotypes were found on both wintering grounds (including a common type representing 45% of each sample), and five types were unique to only one wintering ground. An analysis of variance adapted for molecular information revealed significant genetic differentiation between the two wintering grounds (p = 0.017). The feeding-ground sample was too small for statistical comparison with the wintering grounds, but included two haplotypes found only in the Auckland Islands as well as the common haplotype found on both wintering grounds. The nucleotide diversity and differentiation of mtDNA among the right whales was similar to that among humpback whales (Megaptera novaeangliae) from the same regions (Baker et al. 1998), but haplotype diversity was significantly reduced, perhaps as a result of more intensive hunting during the last century and continued illegal hunting during this century. Phenoloxidase (PO) from the colonial ascidian Botryllus schlosseri was purified using two different chromatographic strategies. A three-step purification was developed in order to maintain enzyme activity, whereas an easier purification procedure was adopted to obtain enough PO for the production of specific polyclonal antibodies. The enzyme showed optimal pH and temperature values of 7.0 to 7.5 and 35 °C, respectively, and a K m value of 4.62 ± 0.76 mM was estimated using l-DOPA as substrate. A molecular weight of 160 kDa was determined after SDS-PAGE under non-reducing conditions. The addition of the reducing agent β-mercaptoethanol caused the disappearance of the 160 kDa band and the appearance of a new band at 80 kDa, suggesting that active PO is a dimer and the two subunits are linked by disulphide bridges. Oxygen and pH microelectrodes were used to investigate the microenvironment of the planktonic foraminifer Orbulina universa and its dinoflagellate endosymbionts. A diffusive boundary layer surrounds the foraminiferal shell and limits the O2 and proton transport from the shell to the ambient seawater and vice versa. Due to symbiont photosynthesis, high O2 concentrations of up to 206% air saturation and a pH of up to 8.8, i.e. 0.5 pH units above ambient seawater, were measured at the shell surface of the foraminifer at saturating irradiances. The respiration of the host–symbiont system in darkness decreased the O2 concentration at the shell surface to <70% of the oxygen content in the surrounding air-saturated water. The pH at the shell surface dropped to 7.9 in darkness. We measured a mean gross photosynthetic rate of 8.5 ± 4.0 nmol O2 h−1 foraminifer−1. The net photosynthesis averaged 5.3 ± 2.7 nmol O2 h−1. In the light, the calculated respiration rates reached 3.9 ± 1.9 nmol O2 h−1, whereas the dark respiration rates were significantly lower (1.7 ± 0.7 nmol O2 h−1). Experimental light–dark cycles demonstrated a very dynamic response of the symbionts to changing light conditions. Gross photosynthesis versus scalar irradiance curves (P vs E o curves) showed light saturation irradiances (E k) of 75 and 137 μmol photons m−2 s−1 in two O. universa specimens, respectively. No inhibition of photosynthesis was observed at irradiance levels up to 700 μmol photons m−2 s−1. The light compensation point of the symbiotic association was 50 μmol photons m−2 s−1. Radial profile measurements of scalar irradiance (E o) inside the foraminifera showed a slight increase at the shell surface up to 105% of the incident irradiance (E d). The growth rates of two fish species, the winter flounder Pseudopleuronectes americanus (Walbaum) (19.3 to 42.6 mm total length, TL) and the tautog Tautogaonitis (Linnaeus) (23.9 to 55.9 mm TL), were used to evaluate habitat quality under and around municipal piers in the Hudson River estuary, USA. Growth rates were measured in a series of 10 d field caging-experiments conducted at two large piers in the summers of 1996 and 1997. Cages (0.64 m2) were deployed along␣transects that stretched from underneath the piers to beyond them, encompassing the pier edge (the transitional zone between the pier interior and the outside). Growth in weight (G w ) was determined at five locations along the transect, 40 m beneath the pier, 20 m beneath the pier, at the pier edge, 20 m beyond the pier edge, and 40 m beyond. Under piers, mean growth rates of winter flounder and tautogs were negative (xˉG W  = −0.02 d−1), and rates were comparable to laboratory-starved control fishes (xˉG W  = −0.02 d−1). In contrast, mean growth rates at pier edges and in open waters beyond piers were generally positive (xˉG W ranged from −0.001 to +0.05 d−1), with growth at pier edges often being more variable and less rapid than at open-water sites. Analyses of stomach contents upon retrieval of caged fishes revealed that dry weights of food were generally higher among fishes caged at open-water stations ( range = 0.02 to 0.72 mg dry wt) than at pier-edge ( range = 0.01 to 0.54 mg) or under-pier ( range = 0.03 to 0.11 mg) stations, although it was apparent that benthic prey were available at all stations on the transect. Our results indicate poor feeding conditions among fishes caged under piers, and suboptimal foraging among fishes caged at pier edges. Inadequate growth rates can lead to higher rates of mortality, and, based on these and other earlier experiments, we conclude that under-pier environments are poor-quality habitats for some species of juvenile fishes.
