The present study was performed to determine the toxic effects of endosulfan on the quantitative and qualitative aspects of acid and alkaline phosphatases (ACP and ALP) in Macrobrachium malcolmsonii. Intermoult juvenile prawns were exposed to 32.0 ng/l of endosulfan for a period of 21 days. Samples were taken from the hemolymph, hepatopancreas, gills and muscle of test prawns on the 21st day. The content of ACP and ALP in the hepatopancreas of test prawns were found to be higher in comparison to respective controls. The levels of these enzymes in the hemolymph, gills and muscle of test prawns were found to be lower than in the same tissues of controls. In non-denaturing PAGE, phosphatases appeared as white bands. The intensity of white bands in the hepatopancreas of test prawns were found to be higher when compared to controls. In the hemolymph, gills and muscle of test prawns, the intensity of white bands were found to be lower in comparison to controls. The results of the present study suggest that endosulfan affects the quantity and quality of ACP and ALP in the tissues of M. malcolmsonii.  相似文献

The present study was performed to investigate the toxic impact of carbaryl on biochemical metabolism in the hemolymph, brain, hepatopancreas, gills and muscle of intermoult juveniles of the economically important prawn, Macrobrachium malcolmsonii. The concentration of glutathione S-transferase (GST) was found to be higher in test prawns when compared with controls. This suggests that a mechanism of detoxification was in operation to neutralise carbaryl toxicity. However, the toxic effect of carbaryl was not fully neutralised, and hence, alterations were recorded in basic biochemical metabolism of test prawns. The concentration of acetylcholinestrase (AchE) was found to be lower in test prawns than that of controls. Carbaryl toxicity resulted in utilisation of major biochemical constituents, such as total carbohydrate, glycogen, protein and lipid to generate required energy as an attempt to withstand the toxic stress. Glycogenolysis resulted in elevation of total free sugar level in the hemolymph of test prawns. While proteolysis led to elevation of total free amino acid level in test prawns. The content of total lipid have also been found lower in test prawns than that of controls. This suggests that carbaryl toxicity resulted in severe energy crises in test prawns. In the present study, toxic effects of carbaryl impair basic metabolic functions and hence pose a threat to the life of M. malcolmsonii.  相似文献

This study determines the toxic effect of carbaryl (Sevin50% W.P) on the food utilization parameters in intermoult juveniles of the prawn, Macrobrachium malcolmsonii. The prawns (4.5-5.0 cm in length and 1.0-1.25 g wet wt.) were exposed to three sub-lethal concentrations of carbaryl (5.15, 7.73 and 15.47 microgl-1) for duration of 40 days. The toxic medium was renewed daily. The prawns were fed ad libitum with known energy quantity of boiled goat liver on daily basis. The overall wet weight gain was calculated. The energy lost through unconsumed food (15-60%), faeces (15-109%), ammonia excretion (9-27%) and moults (13-26%) of the prawns were calculated. The feeding rate, the rate and efficiency of absorption, the metabolic and food conversion rates and the gross and net food conversions efficiencies were found to be significantly declined (p<0.05) in test prawns when compared to that of the control. The energy lost through faeces, ammonia excretion and exuvia was found to be significantly elevated (p<0.05) in test prawns than that of the control. The effectof carbaryl on the bioenergetics parameters was severe in the highest sub-lethal concentration, less in the intermediate concentration and least in the lowest sub-lethal concentration. The results indicated that decrease in feeding, absorption, metabolism and food conversion are interdependent and toxicity of carbaryl diverting energy from production to maintenance pathways, which ultimately resulting in declined growth of M. malcolmsonii.  相似文献

The possible effect of Alexandrium spp. containing paralytic shellfish poisoning (PSP) toxins on the hemocytes of oysters was tested experimentally. In one trial, eastern oysters, Crassostrea virginica Gmelin, were exposed to bloom concentrations of the sympatric dinoflagellate, Alexandrium fundyense Balech, alone and in a mixture with a non-toxic diatom, Thalassiosira weissflogii (Grun) Fryxell et Hasle. Subsequently, another experiment exposed Pacific oysters, Crassostrea gigas Thunberg, to a mixed suspension of the sympatric, toxic species Alexandrium catenella (Whedon et Kofoid) Balech, with T. weissflogii. Measurements of numbers of oyster hemocytes, percentages of different cell types, and functions (phagocytosis, reactive oxygen species (ROS) production, and mortality) were made using flow-cytometry. During and after exposure, almost no significant effects of Alexandrium spp. upon hemocyte numbers, morphology, or functions were detected, despite observations of adductor-muscle paralysis in C. virginica and measured toxin accumulation in C. gigas. The only significant correlation found was between toxin accumulation at one temperature and higher numbers of circulating live and dead hemocytes in C. gigas. The PSP toxins are known to interfere specifically with sodium-channel function; therefore, the finding that the toxins had no effect on measured hemocyte functions suggests that sodium-channel physiology is not important in these hemocyte functions. Finally, because oysters were exposed to the living algae, not purified toxins, there was no evidence of bioactive compounds other than PSP toxins affecting hemocytes in the two species of Alexandrium studied.  相似文献

Freshwater fish Prochilodus lineatus were exposed to sublethal concentrations of cypermethrin (0.3 and 0.6 microg/l) for 2, 5 and 8 days. It was observed that with the increase of exposure time total erythrocyte (RBC), hemoglobin (Hb), hematocrit (Ht) and mean corpuscular hemoglobin concentration (MCHC) values decreased but mean corpuscular volume (MCV) and mean corpuscular hemoglobin (MCH) values increased. These reports indicate that hematological parameters, may be useful as a diagnostic test for cypermethrin exposure in aquatic organisms.  相似文献

Fish are often exposed to highly contaminated water, especially in areas where the dilution rate of waste water is low. Environmental pollutants or other stress may affect one or more of the immunological functions in fish. Effective control of toxic substances is essential for the success of fish culture, which requires fish health monitoring using biomarkers. This study evaluated the effects of heavy metal copper (Cu) at environmentally relevant concentrations on the hematopoiesis in Labeo rohita. L. rohita fingerlings were exposed to different Cu concentrations for 6, 24, 48, or 72 hr. Following exposure, percentage blast cells was found to be increased after 6 and decreased after 24, 48, and 72 hr both at sublethal and LC50 concentrations, whereas percentage mature erythrocytes decreased after 6 and increased after 24, 48, and 72 hr. Erythropoietic and leukopoietic efficiencies were also affected upon exposure. Erythropoietic and leucopoietic efficiencies increased markedly after 6, 24, and 48 hr but decreased after 72 hr in sublethal-treated fish. Erythropoietic efficiency decreased significantly after 6, 24 and 48 hr but increased after 72 hr in LC50-treated fish. Leukopoietic efficiency decreased significantly after 6 hr exposure and increased after 24 and 48 hr but decreased after 72 hr. Flow cytometric studies of head kidney cell cycle phase distribution revealed cell death and DNA synthesis. The percentage cell death increased in fish exposed to sublethal concentrations and rose further at LC50 during the earlier hours of exposure. Synthesis of DNA was increased at sublethal concentrations but was significantly reduced at LC50 dose.  相似文献

The known negative effects of shellfish toxin-producing dinoflagellates on feeding, burrowing and survival of some bivalve mollusks has prompted questions concerning whether they might also impair the internal defense system of affected bivalves and make them more susceptible to disease agents. The primary components of the cellular defense system are hemocytes. Many toxic dinoflagellates are too large to be ingested whole by hemocytes and would most likely be exposed to intracellular toxins only after the algae are consumed, broken down, and the water-soluble toxins, released. Therefore, we conducted a series of experiments in which hemocytes from two suspension-feeding bivalves—the Manila clam, Ruditapes philippinarum, and the softshell clam, Mya arenaria—were exposed in vitro to filtered extracts of one highly toxic paralytic shellfish toxin (PST)-producing and one nonPST-producing strain of Alexandrium tamarense (isolates PR18b, 76 ± 6 STXeq cell−1 and CCMP115, with undetectable PST, respectively). We measured adherence and phagocytosis, two hemocyte attributes known to be inhibited by bacterial pathogens and other stressors. We found no measurable effect of a cell-free extract from a highly concentrated suspension of the PST-producing strain on hemocytes of either bivalve species. Instead, extract from the nonPST-producing strain had a consistent negative effect on both clams, resulting in significantly lower adherence and phagocytosis compared to strain PR18b and filtered seawater controls. The bioactive compound produced by strain CCMP115, which has yet to be characterized, may be similar to the PST-independent allelopathic compounds described for Alexandrium spp., which act on other plankters. These compounds and those produced by other harmful algae are known to cause immobilization, cellular deformation and lysis of co-occurring target organisms. Thus, nonPST producing Alexandrium spp., which do not cause paralysis and burrowing incapacitation of clams, may still produce a compound(s) that has negative effects not only on hemocytes, but on other molluscan cell types and their functions, as well.  相似文献

Bioassays were conducted to determine the effects of temperature and salinity on the acute toxicity of mercury, copper, cadmium, zinc, nickel and lead to juvenile banana prawns (Penaeus merguiensis de Man). Tests were conducted at all combinations of 35, 30 and 20°C with 36 and 20° salinity over 96 h. The general rank order of metal toxicity was Hg > (Cu, Cd, Zn) > Ni > Pb. The toxicity of all metals increased with increased temperature. This was most noticeable in the high salinity treatments, particularly for copper and zinc. Salinity appeared to influence the toxicity of all metals tested although significant differences were only found for copper and lead at 20°C. At this temperature prawns were markedly more susceptible to both metals in low salinity sea water. The data are compared with lethal concentrations found for other crustaceans and are discussed in relation to existing water quality criteria.  相似文献

Considering that amphibians are good sentinels of environmental conditions, Rhinella arenarum embryos were used to investigate the effects of sublethal concentrations of the organophosphorus insecticide azinphos-methyl, focusing on its anticholinesterasic or pro-oxidant actions and its possible connection with the appearance of morphological alterations. Early amphibian embryos exposed to azinphos-methyl displayed a protective response through glutathione S-transferase induction, along with superoxide dismutase inhibition. At intermediate embryonic stages, embryos exposed to azinphos-methyl displayed superoxide dismutase inhibition and morphological alterations, although cholinesterase activity was not altered, suggesting that molecular targets other than cholinesterase were involved in the development of morphological alterations. At the end of embryonic development, decreases in reduced glutathione and cholinesterase inhibition were observed, along with a significant increase in the number of malformed embryos. The connection between biochemical alterations and the appearance of malformations was not evident in R. arenarum embryos. However, increased glutathione S-transferase and decreased superoxide dismutase activities could be considered as early markers of exposure to azinphos-methyl. The results obtained demonstrate that sublethal concentrations of azinphos-methyl are a serious threat to toad embryos in their natural habitats because biochemical and morphological alterations could impair their ability to deal with environmental stresses.  相似文献

Sublethal effects of mercury, cadmium and copper on the diatom Skeletonema costatum (Grev.) Cleve, grown in batch and bacteria-free culture are studied. Division rate, maximum yield growth, mean cell volume, particulate carbon and nitrogen, and 14C-bicarbonate uptake are used as toxic impairment criteria. Division rate is the first-affected and most sensitive parameter, but algal responses vary according to the metal. Hg produces an acute decrease in division rate, followed by a temporary recovery of growth capacity within the first 48 h after metal addition. Cd, on the other hand, increases division rate, followed by an obvious decrease. Cu reduces division rate slowly or quickly, depending on the metal concentration. Cell synthesis capacity (culture biovolume, particulate carbon and nitrogen, carbon assimilation) is less affected than division rate, especially with Hg. The C:N cell ratio is unchanged at sublethal concentrations, even when production is reduced. The mean cell volume is slightly affected: the variations are not greater than those of the control during its growth phases. Markedly teratological forms are never observed. In the authors' opinion, these results confirm that many parameters and growth kinetic aspects must be considered to fully appreciate the effects of sublethal concentrations of heavy metals. It would also be of advantage to develop a better methodology for such research, applying, for example, techniques already employed in enzymology.  相似文献

Neurotoxic effect of BHC, the organochlorine pesticide in Heteropneustes fossilis has been studied exposing at the dose concentrations of 1 ppm, 5 ppm and 10 ppm in lab aquarium for 96 hours over a period of one year. The results showed the behavioural abnormalities in different exposure concentrations such as dysfunction of endocrine gland, excretion of mucus, dispigmentation, sign of restlessness, erratic swimming with rapid jurkey movement, spiralling and convolution showing severe effect in central nervous system. Therefore an attempt has been made for monitoring of BHC residues viz. alpha, beta, gamma isomers in the brain tissue exposed to different sublethal concentrations using Gas liquid chromatography. The mean values of isomers were found to be 1.587 microg/gm for 1 ppm, 2.993 microg/gm for 5 ppm and 3.78 microg/gm for 10 ppm test group. Severe behavioural abnormalities were recorded at high dose concentration of pesticides with higher accumulation of pesticide residues in brain tissue.  相似文献

This study was undertaken to document the impact of heavy metal pollution on the Libyan jird, Meriones libycus and to contribute to an environmental impact statement for the rapidly growing City of Riyadh. All metal concentrations in surface soil of a polluted site (within Riyadh City) were higherthan those from a reference site (outside the city).Although Pb declined versus earlier reports on Riyadh soil, Cd (0.97 microg g(-1)) and Hg (0.28 microg g(-1)) were above some of the most stringent quality guidelines (0.07-0.62 microg g(-1) for Cd and 0.14-0.18 microg g(-1) for Hg). Metal distribution in M. libycus proved site-related and organ-specific, recognizing a higher affinity of most tested metals towards the kidneys, liver and brain than the lung and heart. The comparatively lower site-specific accumulation of Pb in soft tissues was attributed primarily to its major hypothetical accumulation in bones, whereas, the transition rate of Hg from the liver was suggested to be lower to the brain than to the kidneys. Although a non hazardous status was assumed for Cu (11.27-13.16 microg g(-1)) and Hg (up to 0.207 microg g(-1)) in tissues of M. libycus, a potential risk was imposed by mean tissue concentrations of Cd (up to 3.29 microg g(01)), Ni (up to 1.48 microg g(-1)) and Pb (up to 1.94 microg g(-1)). On the grounds of the significantly higher metal levels in polluted soft tissues versus reference subjects, Libyan jirds possess high exposure potential and can be useful biomonitors of environmental metal contamination.  相似文献

The waste water showed high values of total solid (TS), hardness and chloride with slightly alkaline pH along with high concentrations of Cr (2.03 mg l(-1)), Ni (1.59 mg l(-1)) and Zn (0.46 mg l(-1)). The concentration of Cu (0.21 mg l(-1)) and Zn in industrial waste water was low than Ni and Cr. The diluted (25 and 50%), undiluted (100%) waste water was used to irrigate the lettuce plants grown in alluvial soils. Plants accumulated heavy metals in their shoot (Ni, 13.65; Cr, 19.73; Zn, 21.6 and Cu 14.76 microg g(-1) dry weight) and root (Ni, 41.4; Cr, 31.6; Zn, 30.2 and Cu 15.85 microg g(-1) dry weight) in high concentrations after irrigation with undiluted industrial waste water. Maximum accumulation of heavy metals was found in the root than the shoot (13.65-21.60 microg g(-1) dry weight). Dry matter yield and biomolecules (Chlorophyll a, b and sugar contents) was found to increase with increase in concentration of waste water up to 50%, which declined at the exposure of undiluted waste water. Catalase activity was found to increase with increase in waste water concentrations up to 100%, while carotenoids content increased in plants only up to the 50% waste water irrigation. Use of industrial waste water in such form, on agricultural lands is not found suitable without proper treatment. It could be injurious to plants growth and may be a potential threat to food web.  相似文献

This study investigates the mercury (Hg) contaminations in soil and foodstuffs along the artisanal gold mining areas, Gilgit-Baltistan Province, Pakistan. For this purpose, soils were analyzed for Hg concentrations and evaluated for the enrichment/contamination using enrichment factor or contamination factors (CF). The CF values ranged from 18.9 to 153 showed multifold higher levels of Hg contamination as compared to background or reference site. Foodstuffs including vegetables, seeds or grains and fish muscles showed Hg accumulation. Results revealed that Hg concentrations in foodstuffs were higher than the critical human health value set by European Union. The Hg in foodstuffs was consumed and, therefore, evaluated for the risk assessment indices using the daily intake (DI) and health risk index (HRI) for the exposed human population both children and adults. Results of this study revealed that cumulative HRI values through foodstuffs consumption were <1 (within safe limit), but if the current practices continued, then the Hg contamination could pose potential threat to exposed population in near future.  相似文献

The coastal saline soils, Kharlands, have great potential for their use in aquaculture. This study has been taken up to understand the limnology of the ponds in Kharland area for assessing their prawn culture potential. This study was carried out during September, 1999 to August, 2001. Each Kharland pond has an area of 0.045 hectare. During the study, depth of pond water was 47.7 to 120.0 cm, temperature varied from 25.7 to 34.5 degrees C; transparency from nil to 65.0 cm; specific conductivity from 1.78 to 94.5 microS.cm(-1); total dissolved solids from 0.89 to 27.55 ppt; pH 5.42 to 8.25; dissolved oxygen 1.6 to 8 mg.l(-1); free carbon dioxide 10.00 to 44.00 mg.l(-1); total alkalinity 5.00 to 142.00 mg.l(-1); salinity 0.45 to 39.55 ppt; total hardness 245.00 to 5945.00; calcium 56.05 to 1827.6; magnesium 110.74 to 4507.75 mg.l(-1); dissolved organic matter 1.45 to 9.68 mg.l(-1); ammonia 1.00-8.00 microg.l(-1); nitrite nil to 20.00 micro l(-1) and nitrate 7.5 to 17.5 microg.l(-1). These Kharland ponds are unique in physio-chemical characteristics during their seasonal cycle. From July to October, these ponds have nearly freshwater while from November to May pond water becomes saline. Thus, there is a great possibility of taking up monoculture of both the freshwater and brackish water prawns as well as polyculture of prawns and fishes in the Kharland ponds.  相似文献

The marine amphipodAllorchestes compressa Dana, fed on the seagrassHeterozostera tasmanica, was exposed to sublethal concentrations of Cd, Cr, Cu and Zn for 4 wk in flowing sea water, and the concentrations producing the minimum detectable decreases (the minimum effect concentrations, MECs) in average weight, survival and biomass (average weight × survival proportion) were estimated by interpolation from regression models. Survival and biomass were more sensitive than average weight as indicators of sublethal effects. The lowest values of MEC for Cd, Cr, Cu and Zn were 11, >250, 3.7 and 99µg 1–1, respectively. For Cu, this value fell below the minimum risk concentration (MRC) calculated from acute toxicity tests (LC50) and application factors (AF); for Cd, the MEC was similar to the MRC; for Cr and Zn, the MECs were well above the MRCs. The metal concentrations in the amphipods at the MECs were 46, >46, 364 and 139µg g–1 dry wt for Cd, Cr, Cu and Zn, respectively. Accumulation of the nutrient metals (Cr, Cu and Zn) showed some evidence of metabolic regulation, but the non-nutrient Cd was accumulated without regulation until the amphipods died. In general, those metals that were more highly accumulated by the amphipods were the more toxic.  相似文献

We studied the demographic responses of Ceriodaphnia dubia exposed to copper at 3 different levels (0.1, 0.2 and 0.4 mg l(-1)) and at 4 exposure periods (3, 6, 12 and 24 hr) in addition to controls (without the heavy metal). The tested levels of Cu concentration and the chosen periods of exposure affected some demographic variables (average lifespan, gross and net reproductive rates and generation time). Depending on the heavy metal concentration and the period of exposure, the average lifespan of C. dubia varied from 18 to 24 day Net reproductive rates (NRR) ranged from 50 to 80 offspring per female per lifespan, while the rate of population increase (r) varied from 0.30 to 0.34 per day Though gross reproductive rates (especially at low and intermediate copper levels and shorter duration of exposure) showed significantly higher values than controls, both NRR and r were either unaffected or decreased. It is therefore necessary to regulate the release of untreated industrial effluents containing copper into freshwater ecosystems. Our study thus highlights the importance of including, not only toxicant concentrations but also exposure time, in ecotoxicological evaluations.  相似文献

Experiments were conducted to assess the effects of azadirachtin (AZ) on survival capacity, fertilization success, and swimming speed in larval stages of the cane toad, Bufo marinus . LC50 values (96-hr) for various larval stages exposed to 0.50 mg/l AZ were as follows: 5.35 (stage 12), 5.77 (stage 16), 787 (stage 20), 13.43 (stage 24), and 21.54 (stage 30). Tadpoles did not exhibit any apparent disorientation or production of mucous in response to AZ concentrations ranging from 0.3 to 0.5 mg/l. Mortality was significantly higher at 0.50 mg/l AZ than at lower concentrations. At 0.50 mg/l, mortality rate decreased significantly from 36% for stage 12 tadpoles, to 24% for stage 20 tadpoles. Mortality for early stages (stage 12) dropped significantly, from 36% at 0.50 mg/l, to 21% at 0.40 mg/l AZ There were no significant differences in percent mortality for any developmental stage at an AZ concentration of 0.3 mg/l, or for controls (no AZ). Exposure of gametes to 0.30 mg/l AZ did not reduce fertilization success as compared to controls (89.8 vs. 83.7%). At 0.40 mg/l, percent fertilization decreased significantly (68.8%). This effect was even more pronounced at 0.50 mg/l. No significant difference was found in swimming speeds between controls and subjects exposed to 0.30 mg/l AZ. However, at a concentration of 0.40 mg/l, tadpoles required a significantly longer period of time to swim a distance of 140 cm as compared to controls. The potential impact of AZ on amphibian populations is discussed.  相似文献

The short term human exposure studies conducted on populations exposed to high concentrations of inorganic arsenic in soil have been inconsistent in demonstrating a relationship between environmental concentrations and exposure measures. In Australia there are many areas with very high arsenic concentrations in residential soil most typically associated with gold mining activities in rural areas. This study aimed to investigate the relationship between environmental arsenic and urinary inorganic arsenic concentrations in a population living in a gold mining area (soil arsenic concentrations between 9 and 9900 mg kg(-1)), and a control population with low arsenic levels in soil (between 1 and 80 mg kg(-1)). Risk factors for increased urinary arsenic concentrations were also explored. There was a weak but significant relationship between soil arsenic concentrations and inorganic urinary arsenic concentration with a Spearman correlation coefficient of 0.39. When participants with greater than 100 mg kg(-1) arsenic in residential soil were selected, the coefficient increased to 0.64. In this study, the acute toxicity of butyl benzyl phthalate (BBP) to freshwater cladoceran Moina macrocopa was tested, and its chronic effects on survival and reproduction of two successive generations of the cladoceran were studied using life-table demographic method. The results showed that the 48-hr LC50 of BBP for M. macrocopa was 3.69 mg l(-1). Compared to the blank controls, BBP at 125, 500, 1000 and 2000 microg l(-1) significantly shortened the life expectancy at birth, BBP at 125-2000 microg l(-1) decreased the net reproductive rate, and BBP at 500 and 1000 microg I(-1) shortened the generation time but increased the intrinsic rate of population increase of the parental M. macrocopa. BBP at 62.5,125, 500,1000 and 2000 microg l(-1) increased the intrinsic rate of population increase of the F1 generation. A significant dose-effect relationship existed between BBP concentration and life expectancy at birth, net reproductive rate as well as intrinsic rate of population increase of the parental M. macrocopa. The parental M. macrocopa were more sensitive in survival, development and reproduction to BBP than the F1 generation, but the reverse was also true in the population growth. Extending chronic toxicity tests to the second generation of M. macrocopa increased the cost-effectiveness of the assays.  相似文献
