The crab Carcinus maenas (L.) was exposed to radioactively labelled cadmium dissolved in seawater at concentrations of 0.1, 1 and 10 ppm, the latter concentration being toxic to the crabs (50% mortality after 12.3 days). Net accumulation of cadmium from solution was proportional to the level and time period of cadmium exposure. Total absorbed cadmium levels reached 0.0043 and 0.0412 mg Cd g-1 dry weight after 40 days exposure to 0.1 and 1 ppm Cd, respectively, and 0.1115 mg Cd g-1 dry weight after 12.3 days average exposure to 10 ppm Cd. The highest tissue concentration was found in the midgut gland, reaching 0.786 mg Cd g-1 dry weight after 12.3 days average exposure to 10 ppm Cd. The midgut gland only contained about 10% of the total cadmium absorbed from solution, while the exoskeleton contained the bulk of obsorbed cadmium (59 to 80%) probably passively adsorbed onto the surface. When cadmium was absorbed by the crabs from a food source, the midgut gland contained 16.9% of the total absorbed cadmium whereas the exoskeleton now contained only 22.2%. Ten percent of the cadmium available in the food source (Artemia salina) was accumulated by the crabs. When placed in cadmium-free seawater, crabs that had accumulated cadmium from solution lost 69% of the absorbed cadmium in 10 days, mostly from the exoskeleton which lost 78% of its original absorbed cadmium concentration.  相似文献

The relative importance of accumulation of cadmium by Artemia salina (L.) directly from solution and from ingested food has been studied at 3 cadmium concentrations (0.1, 1 and 10 ppm) under controlled experimental conditions. At each cadmium concentration, A. salina were exposed to cadmium in 4 ways; in solution; in solution in the presence of latex food particles; in solution with cadmium-rich Dunaliella tertiolecta as a food source; and to cadmium-rich D. tertiolecta alone. Net accumulation of cadmium by A. salina continued throughout 5 days exposure under all 4 conditions. When the brine shrimp were removed to cadmium-free conditions, their accumulated cadmium concentrations declined and levelled off to a stabilised plateau after 10 days. From consideration of these stabilised levels it was shown that at least 30% of cadmium accumulation directly from solution occurs via uptake through the alimentary tract. The ratio of cadmium accumulated from solution to cadmium accumulated from food was found to be 1:4.9, 1:6.7 and 1:1.1 at 0.1, 1 and 10 ppm Cd exposure, respectively. At lower cadmium exposures uptake from food is the major route for cadmium accumulation, but at higher exposures the cadmium-saturated food source displaced cadmium-rich water from the gut and therefore actually inhibited cadmium accumulation. This study, therefore, concludes that the food chain will be the major source of cadmium as long as the previous trophic level has the ability to accumulate the metal to such an extent as to make it more available to the consumer than by direct uptake from seawater.  相似文献

Various forms of regression analysis are presented which show that there is a significant equilibrium relationship between total recoverable cadmium in seawater and its concentration in the mussel Mytilus edulis (P<0.001). The calculations show that the concentration of cadmium in seawater should not exceed 0.20 g l-1 if the mussel is not to reach a cadmium concentration of 2 mg kg-1 wet weight: a value frequently used as a food standard for human consumption. Further, when a cadmium value of 2 mg kg-1 wet weight is reached, the concentration factor by mussels for cadmium from seawater is 9 950.  相似文献

 The effects of cadmium exposure and dietary status on cadmium accumulation, fatty acid (FA) content and profiles were investigated in two colour forms of the shore crab Carcinus maenas. Groups of shore crabs were either starved or fed with blue mussels, Mytilus edulis, during a 40 d exposure period to 2 or 6 μM Cd2+ (as CdCl2). Starved green individuals accumulated more cadmium in haemolymph and hepatopancreas than did red crabs and green crabs fed during the experiments. In the red colour form, no difference in cadmium accumulation was observed between starved and fed individuals. In both colour forms, hepatopancreas contained more FA than gills and muscle. The FAs often present in the largest amounts in the tissues were 16:0, 16:1ω7, 18:1ω7, 18:1ω9, 20:4ω6, 20:5ω3 and 22:6ω3. However, saturated (SAFA) and mono-unsaturated fatty acids (MUFAs) were dominant in hepatopancreas, whereas poly-unsaturated fatty acids (PUFAs) were dominant in gills and muscles. At the beginning of the experiment, the total FA content in the hepatopancreas was 111.6 mg g−1 (dry weight) for red crabs and 78.4 mg g−1 for green shore crabs. During the experiment, however, the FA content decreased in red crabs. This decrease was more pronounced for starved individuals than for fed individuals. Also, the decrease in FA content was more pronounced in crabs exposed to 6 μM cadmium compared to crabs exposed to 2 μM or crabs not exposed to cadmium. No change in FA content was observed in green shore crabs, irrespective of diet and cadmium exposure. For both colour forms, no change in FA content was observed for gills and muscle. In red crabs, a decrease was observed for all FAs in the hepatopancreas. This decrease, however, was more pronounced for SAFAs and MUFAs than for PUFAs, indicating that the metabolism of FAs during starvation and cadmium exposure is selective. The experiments indicate that green colour forms of shore crabs are more tolerant of natural stress such as starvation and anthropogenic stress, e.g. cadmium exposure, than are red colour forms of shore crabs. We determined the temporal evolution of amylase, cellulase, laminarinase and protease in the digestive gland and crystalline style of cockles Cerastoderma edule held over 9 to 12 d in the presence and absence of food. Cockles were fed a constant diet of 1.5 mm3 l−1 of Tetraselmis suecica for 9 to 12 d and were then starved for 6 to 8 d in late summer (September 1992) and in winter (January 1993). Feeding increased the dry weight and total cellulase, laminarinase and protease activities of the digestive gland irrespective of season, whereas amylase activity remained unchanged. In winter (i.e. when cockles are metabolically weak) the response was faster and stronger, especially for protease. An additional experiment in September starved cockles for 20 d before resuming feeding. In agreement with the seasonal differences, the presence of food after prolonged starvation induced a rapid and marked increase in protease in the digestive gland of the cockles. In winter, the possible effects of the biochemical composition of food on their enzymatic response were tested by feeding two groups of cockles with the same ration of T. suecica but harvested at different growth phases. A compensatory induction of cellulases occurred in cockles fed on T. suecica with a lower carbohydrate content. In the crystalline style, the protein level and carbohydrase fell during the first day of feeding and increased during the first day of subsequent starvation. These results indicate that the release of enzymes from the style prevails over the incorporation of enzymes during the early stages of feeding, whereas the opposite occurs during starvation. Comparative studies on feeding, digestion, absorption and conversion have been made in the fish Megalops cyprinoides fed with mosquito fish, Gambusia affinis, and prawn, Metapenaeus monoceros, (Pandian, 1967). Feeding as well as digestion rates are faster in individuals fed with Gambusia than in those fed with prawn. The extent of the differences in food intake is more pronounced in smaller individuals than in larger ones. The prawn exoskeleton prolongs the duration of digestion and thus further reduces rate and amount of food intake. Changes in food quality do not affect absorption efficiency. Both, rate and efficiency of food conversion, are higher in M. cyprinoides fed with Gambusia than with prawn.  相似文献

室内采用离体叶片法研究了转sck单价基因抗虫水稻86AS1和转cry1Ac/sck双价基因抗虫水稻MSB不同生育期主茎顶叶对二化螟幼虫的杀虫效果.结果发现,不同生育期的MSB对二化螟初孵幼虫表现出了很强的毒杀效果,二化螟幼虫校正死亡率在扬花期前均达到90%以上;扬花期和灌浆期抗虫性有所下降,校正死亡率高于80%;成熟期校正死亡率在60%以上.在整个生育期,二化螟幼虫取食86AS1主茎顶叶的校正死亡率均未超过50%.MSB对二化螟幼虫的毒杀效果明显高于86AS1.采用透射电镜观察二化螟幼虫取食转cry1Ac/sck双价基因抗虫水稻和转sck单价基因抗虫水稻主茎顶叶后中肠组织的病理变化.结果发现,两种转基因水稻都能够引起二化螟幼虫中肠组织发生病理变化,但病变程度有明显差异.二化螟幼虫取食转cry1Ac/sck双价基因抗虫水稻3d,中肠组织受到严重破坏且病理变化明显;取食转sck单价基因抗虫水稻3d,幼虫中肠组织的病理变化却较缓慢.图2参10  相似文献

The distribution of cadmium within captive crabs (Cancer pagurus) exposed experimentally to cadmium-contaminated food and water is described and illustrated by triangular diagrams. Crabs from the Orkney Islands (Scotland) are known to contain relatively high levels of cadmium (up to 62 g g-1 wet wt) in the hepatopancreas. The distribution of cadmium between the hepatopancreas, gonad, gill, carapace, claw muscle, heart, and haemolymph, is described in crabs collected during 1978, and compared with similar data from crabs exposed to cadmium for ca. 300 d (September 1978 – June 1979) experimentally. It is concluded that the dominant uptake route of cadmium to Orkney crabs is through their diet.  相似文献

Cadmium (Cd) is a toxic-heavy metal that induces a wide range of behavioural, biochemical and physiological effects in aquatic organisms. Oxidative damage has been proposed as a possible mechanism involved in cadmium toxicity. The current study was carried out to evaluate the antioxidant activity of Spirulina as feed additive (1?mg/L) against the toxicity of cadmium (Cd) 0.5?mg/L in freshwater mussel Unio ravoisieri. At the end of the exposed period of 4 days, digestive gland antioxidant status Superoxide dismutase, Catalase, Glutathione-S-transferase and damage markers such as Malondialdehyde and Protein carbonyl were determined. Associations between biomarkers were assessed by a multivariate analysis technique, principal component analysis (PCA). The results of this study revealed that digestive gland antioxidant status showed a significant decrease when mussels were exposed to Cd. The difference between the cadmium uptake via food and seawater in Mytilus edulis has been studied. This was done by labelling algae with Cd-109 and seawater with Cd-115m. Mussels were fed on six different quantities of Isochrysis galbana. Cadmium uptake via algae was more efficient at low food levels, while accumulation from seawater was linearly correlated with food quantities. Cadmium from food contributed only little to the body burden (0.2–0.5%). Half-lives for the elimination of cadmium ranged from 96–190 d and increased with decreasing availability of algae, presumably due to slowed down metabolism. Differences in elimination patterns suggest a release of both isotopes from different storage depots. A computer model shows that the food pathway can only play a significant role if algae are highly contaminated. It also demonstrates the paradox that in long-term studies the highest contribution of food-derived cadmium to the body burden must be expected near maintenance food concentrations.  相似文献

In the carnivorous isopod Eurydice pulchra Leach the mid-gut caeca have a pH of 5.6 and produce a strong acid proteinase, a lipase and a carbohydrase. Electron microscope studies of the digestive caeca of starved and fed animals show that secretion is merocrine and may originate from a complex involving the basal cell membrane and associated mitochondria, producing lysosomes. Absorbed materials, in fed animals, appear to take the form of neutral lipid bodies, glycogen, and proteinaceous crystals, the latter forming within the golgi complex. Digestion of both lipid and protein inclusions was observed to occur intra-cellularly by fusion with free lysosomes. Secretion and absorption may occur in the same cell, and the scheme for the sequence of epithelial cell stages is proposed.  相似文献

The effects of sublethal concentrations of cadmium (Cd) on the morphology of the digestive gland of the periwinkle Littorina littorea have been investigated using a planimetric procedure. Five planimetric parameters were recorded (mean epithelial thickness: MET; mean diverticular radius: MDR; mean luminal radius: MLR; MLR/MET and MET/MDR) and tested by diverse statistical methods. The results indicate that MET is depressed after long exposures at high Cd concentrations whilst MLR increases. Variations in MDR have been attributed to different conditions in the tissues surrounding digestive tubules, and are not-significant in relation to the treatment. However, certain increases might be significant due to the presence of parasites or hemocyte proliferation. The ratios MLR/MET and MET/MDR are highly sensitive, and can be expressed as statistical functions of metal concentration and exposure time. Combination of these five planimetric parameters is reliably indicative of the structural changes occurring in the digestive gland of molluscs under conditions of environmental stress.  相似文献

Flux of the heavy metal cadmium through the euphausiid Meganyctiphanes norvegica was examined. Radiotracer experiments showed that cadmium can be accumulated either directly from water or through the food chain. When comparing equilibrium cadmium concentration factors based on stable element measurements with those obtained from radiotracer experiments, it is evident that exchange between cadmium in the water and that in euphausiid tissue is a relatively slow process, indicating that, in the long term, ingestion of cadmium will probably be the more important route for the accumulation of this metal. Approximately 10% of cadmium ingested by euphausiids was incorporated into internal tissues when the food source was radioactive Artemia. After 1 month cadmium, accumulated directly from water, was found to be most concentrated in the viscera with lesser amounts in eyes, exoskeleton and muscle, respectively. Use of a simple model, based on the assumption that cadmium taken in by the organism must equal cadmium released plus that accumulated in tissue, allowed assessment of the relative importance of various metabolic parameters in controlling the cadmium flux through euphausiids. Fecal pellets, due to their relatively high rate of production and high cadmium content, accounted for 84% of the total cadmium flux through M. norvegica. Comparisons of stable cadmium concentrations in natural euphausiid food and the organism's resultant fecal pellets indicate that the cadmium concentration in ingested material was increased nearly 5-fold during its passage through the euphausiid. From comparisons of all routes by which cadmium can be released from M. norvegica to the water column, it is concluded that fecal pellet deposition represents the principal mechanism effecting the downward vertical transport of cadmium by this species.  相似文献

It has been confirmed that metallothioneins play an important role in the accumulation of cadmium (Cd) in the digestive gland cells of mussels (Mytilus galloprovincialis Lam.). The content of Cd in the tissue of mussels exposed for 9 d to the metal (estimated dosage of 180 g Cd mussel-1 d-1) was 66.2 ppm. This value is about the same as the metal content found in the digestive gland of Cd-exposed mussels kept in clean water for a recovery period of 28 d. At the end of the recovery period, however, the Cd bound to thionein had increased by approximately 250%. Our data demonstrate that the stability of lysosomes, a biological parameter adopted as a cellular stress index, is extremely low in mussels exposed to Cd for 9 d, but returns to control values in the digestive gland cells of mussels allowed to recover for 28 d in uncontaminated sea water. At this point most of the Cd present in the cytosol is bound to thionein. These data demonstrate the importance of metallothionein induction in the reduction of the cytotoxic effects exerted by high levels of Cd accumulation. The results of tests designed to clarify the reasons for the long biological half-life of Cd demonstrated that, in the digestive gland of mussels, the lysosomes are not able to eliminate Cd either bound to insoluble thionein polymers or to lipid peroxidation products such as lysosomal lipofuscin, both of which are apparently involved in the elimination of copper. The absence of these two mechanisms of metal sequestration and elimination via excretion of residual bodies (tertiary lysosomes) is in agreement with the persistence of cadmium in the digestive gland of mussels. Finally, the results also demonstrate that simultaneous exposure of mussels to Cd and phenanthrene, an established lysosomal membrane destabilizer, did not significantly alter the accumulation of Cd or the kinetics of the metal in mussels.  相似文献

Trace-metal distribution in tissues of the shrimp Pleoticus muelleri Bate from the Patagonian region, Argentina, was related to sex, size and physiological condition. Concentrations of cadmium, copper, manganese and zinc were determined in the digestive gland, male reproductive system and muscle of adult specimens. Significant (p<0.01) sex-dependent differences in mean metal concentrations (g g-1 wet wt) were found in the following tissues of males and females, respectively: digestive gland, Cu, 82.9 and 30.8; Zn, 32.5 and 44.9; reproductive system, Zn, 12.8 and 38.6; Mn, 1.7 and 3.1; Cd, 0.29 and 0.58. Metal levels of muscle showed no significant differences between sexes. Cadmium was not detected in muscle, suggesting that its incorporation into this tissue was strictly regulated. Metal concentrations in the male reproductive system were in general weakly correlated (0.001<p<0.05) with either body size or the concentration of metals in the digestive gland. The developing ovary incorporated substantial amounts of zinc, while cadmium and copper levels decreased at proportional rates (p<0.001). The patterns of the variations in the concentrations of these metals in the ovary strongly suggested that a regulatory mechanism operated throughout oogenesis. Manganese was not involved in this presumptive mechanism. The results of this study are discussed in terms of concentrations and in absolute amounts for standardized individuals.  相似文献

The performance of an artificial practical diet, kappacarrageenan microbound diet (C-MBD) was assessed on Penaeus monodon larvae at the SEAFDEC Broodstock and Maturation Experimental Laboratory in March 1986. Shrimps were reared from zoea1 to post-larvae1 using five dietary treatments: (a) natural food — Chaetoceros calicitrans and Artemia salina (b) C-MBD; (c) combination of natural food and C-MBD; (d) commercial diet (microencapsulated, MED); (e) combination of natural food and commercial diet. Results showed slow development with larvae fed the commercial diet. Feeding with C-MBD in combination with natural food resulted in the highest % survival among treatments (69.6), but this was not significantly different (P>0.05) from those obtained with larvae fed natural food alone, C-MBD alone or their combination. While mean values for survival of larvae fed the commercial diet, either alone or in combination, was significantly lower (p<0.05) than all other treatments, their mean growth indices were comparable with larvae fed C-MBD alone or in combination. The low levels of protein, lipid and essentially fatty acids (which are considered important nutrients during larval development) contained in the commercial diet may well justify the results on metamorphosis, survival and growth of the larvae fed this diet. The good performance of C-MBD in this experiment suggests that this kind of diet can be used as partial or total replacement to the traditional algal food.  相似文献

Uptake of 109Cd in freshly caught sea-skaters Halobates nereis and H. flaviventris was measured under the influence of abiotic parameters as salinity and stable Cd concentration. After 6 d of accumulation a concentration factor of 8 over the seawater level was reached with no indication for an equilibration. The calculated biological half-life of 109Cd amounted to 9.3 d. Cadmium uptake from food (wingless Drosophila sp. flies reared on cadmium enriched agar) can be considerable. A large pond-skater Limnogonus fossarum, measured for comparative purposes, accumulated cadmium, but at a much lower rate. The results are discussed with respect to observed natural cadmium concentrations in open ocean sea-skater populations.  相似文献

Food selectivity was examined in amictic female rotifers, Brachionus plicatilis, fed Chlamydomonas sp. and Olisthodiscus sp. Filtering and ingestion rates of the rotifers in a single food suspension were higher with Chlamydomonas sp. than with Olisthodiscus sp. In every mixture of two food species, the apparent filtering rate on Chlamydomonas sp. was much higher than that on Olisthodiscus sp. Total filtering rate and apparent filtering rate in food suspensions containing Chlamydomonas sp. decreased with increasing cell concentration of Chlamydomonas sp. Total and apparent filtering rates were calculated from the decrease in total food concentration as well as from decreases in concentrations of each of the two food algae, respectively. B. plicatilis ingested Olisthodiscus sp. at an extremely low constant rate in all mixtures. The degree of food selectivity of the rotifers fed Chlamydomonas sp. (i.e., selective filtration) from the mixture of two food algae decreased with increasing cell concentration of Chlamydomonas sp. Filtering and ingestion rates of rotifers fed senescent Chlamydomonas sp. were relatively lower than those fed Chlamydomonas sp. in the exponential phase. This indicates that Brachionus plicatilis displays selectivity in regard to condition of cells as well as type of food.  相似文献

Melipona panamica foragers can deposit a scent beacon that influences the orientation of foragers near a food source. In misdirection experiments, newcomers (bees from the same colony as trained foragers) consistently preferred the feeder at which trained foragers had fed (training feeder) over an identical feeder at which bees had never fed (control feeder) even when the training feeder was placed at a site where experienced foragers had never foraged. Through similar misdirection experiments, the effective radius of the scent beacon was determined to be greater than 6 and less than 12 m. Foragers may deposit this beacon during a sequence of departure behaviors performed at the feeder. Prior to leaving the feeder with a load of sugar solution, bees tended to perform the following sequence of behaviors: (1) spinning, (2) grooming, (3) abdomen dragging, (4) excreting anal droplets, and (5) producing sounds, although not all behaviors were performed prior to each departure or at all sucrose concentrations (0.5–2.5 m). As sucrose concentration increased, the number of newcomers significantly increased, and the number of experienced foragers producing sounds and spinning on the feeder increased. The exact source of the scent beacon remains a mystery. However, three important sources have been excluded. When choosing between identical paired feeders, foragers were not attracted to the feeders with (1) anal droplets, (2) extracts of sucrose solution at which foragers had fed, or (3) mandibular gland extracts. They were indifferent to the first two preparations and exhibited only typical alarm behavior towards the mandibular gland (MG) extract: they oriented towards the feeder with MG extract but consistently landed on the feeder with no MG extract. Other authors have suggested that Melipona foragers deposit anal droplets to attract recruits, however the frequency of anal droplet production and the mass of anal droplets produced by M. panamica foragers are negatively correlated with sucrose concentration. Thus the scent beacon is evidently not deposited with anal droplets, infused into the feeder solution, or produced by mandibular glands. Two heavy metal-binding proteins occur naturally in the midgut glands of Carcinus maenas (L.) collected from the Firth of Clyde, Scotland. These proteins, of approximately 27,000 and 11,500 MW (molecular weight) have previously been described as Cd-binding proteins after their induction by high concentrations of cadmium in the laboratory. The 27,000 MW heavy metal-binding protein is bound to about 0.10 g-at of Cd, 0.70 g-at of Zn and 0.31 g-at of Cu per mole of protein; 7.7, 7.9 and 1.1%, respectively, of the soluble Cd, Zn and Cu in the midgut gland are associated with this 27,000 MW protein (6.7, 1.6 and 0.9% of the total midgut gland Cd, Zn and Cu). The 11,500 MW protein is bound to about 0.04 g-at of Cd, 0.37 g-at of Zn and 1.54 g-at of Cu per mole of protein; 29.3, 31.3 and 41.7%, respectively, of the soluble Cd, Zn and Cu in the midgut gland are associated with this protein (25.7, 6.3 and 34.4% of the total midgut gland Cd, Zn and Cu).
