Improved methodology for ATP determination in marine environments   总被引:1,自引:0,他引:1  
A simple technique is described for de-salting and concentrating adenosine triphosphate (ATP) extracted from seawater or marine sediment samples prior to assays with the standard luciferin-luciferase procedure. The technique involves chromatography of H2SO4 extracts on columns of activated carbon. The efficiency of ATP recovery from marine sediments using this pre-treatment was superior to that attained with either boiling Tris extraction or with H2SO4 extraction without subsequent purification. All ions which interfere with the luciferase reaction or precipitate ATP upon neutralization of acid extracts are removed with this procedure, thus eliminating the 50- to 100-fold dilutions required with other acid-extraction procedures. In addition, the purified ATP extracts may be concentrated up to 100-fold, thus greatly improving the sensitivity of ATP measurement in samples containing very low biomass. This procedure has been applied to ATP determination in marine sediments in the upwelling area off northwest Africa. The average ATP concentrations in the upper 6 cm of sediment from 1000 and 200 m were 195 and 545 ng per ml wet sediment, respectively.  相似文献

A method for measuring ATP, ADP and AMP levels in environmental samples was devised, and applied to seawater and bacterial cell extracts. This procedure is specifically designed for measuring the extremely low concentrations of total adenine nucleotides ([AT]=[ATP]+[ADP]+[AMP]) that are apt to occur in most natural ecosystems (i.e., 10 ng AT ml-1 of sample extract). Although the current assay methodology can be used with purified firefly luciferase reagents, it has been suitably modified to accept crude luciferase preparations as well. ATP, ADP and AMP levels have been measured, and the corresponding energy charge (EC) ratios determined for seawater samples collected off the Southern California coast. The EC ratios ranged from 0.50 to 0.89, with peak values corresponding to the subsurface maxima in ATP and chlorophyll a concentrations, and the minimum values corresponding to the deepest water sampled (1500 m). The measurement of adenylate energy charge ratios in environmental samples can be a useful indicator of mean community metabolic activity and potential for cell growth.  相似文献

Latitudinally separate populations of the sea anemone Metridium senile (L.) are very similar genetically by electrophoretic criteria, yet respond differently to temperature. Anemones from southern and northern California (USA) have different oxygen consumption patterns in response to acclimatory and acute changes in temperature. Northern anemones show a pronounced increase in Q10 at temperatures just above the normal environmental range, but southern anemones do not. The two populations also differed in the extent of metabolic compensation to temperature following several weeks of acclimation. This acclimation regime resulted in changes in the activities of several enzymes of intermediary metabolism, yet the extent and direction of these changes did not display a consistent trend with regard to acclimation temperature or population. The biochemical concomitants of acclimatory and acute temperature effects were studied further by measuring the concentrations of adenylates (ATP, ADP, and AMP) in anemones from the two populations exposed to different temperature regimes. During cold acclimation for several weeks, total adenylate concentrations (AT) increased in both the southern and northern populations, possibly due to metabolic rate compensation, since AT is positively correlated with tissue metabolic rate in many species. Moreover, the extremely low weight-specific oxygen consumption rates of M. senile are probably related to its very low AT values. Acute temperature decreases had no effect on adenylate concentrations and adenylate energy charge (AEC); in contrast, acute temperature increases led to large changes in adenylate concentrations. The effects of starvation on adenylate concentrations are pronounced, and the effect is temperature-dependent. In starved individuals held at 20°C, AEC values fell to 50% of normal values after 8 d, while those held at 10°C maintained normal AEC values.  相似文献

To aid the recovery of a species, understanding the extent to which populations are connected is useful for targeting conservation efforts. Pacific hake within waters of Puget Sound, Washington State, USA, and Georgia Strait, British Columbia, Canada are listed as a species of concern under the U.S. Endangered Species Act due to dramatic declines in the Puget Sound population. To assess the role of dispersal in the recovery of Pacific hake, we sought to quantify patterns of connectivity between populations in Puget Sound and Georgia Strait. Using natural chemical markers from otoliths of fish sampled from these two populations, we linked natal signatures of fish to signatures of individuals from known spawning grounds. Results indicated that 82 % of individuals collected from Puget Sound (n = 78) were estimated to have originated there, while 40 and 92 % of the individuals collected from two cohorts within Georgia Strait (n = 9 and 24, respectively) had originated from Puget Sound. A trend of “population abandonment” of fish from Puget Sound suggests that recovery of this Pacific hake population will depend on local management practices.  相似文献

While sampling intertidally in Puget Sound, Washington, USA, for juvenile Dungeness crab (Cancer magister) in 1984, we found evidence of two distinct cohorts of the same year-class based on sizes of first-instar juveniles (J1) and the spatial/temporal patterns of settlement. In 1988, three distinct cohorts were observed to settle in Puget Sound and its approaches. Settlement of one cohort occurred during May in the Strait of Juan de Fuca and in those areas of Puget Sound closest to the Strait. J1 individuals of this cohort were large (x=7.4 mm carapace width, CW) and comparable in both size and timing of settlement to populations along the Washington coast (e.g. Grays Harbor and Willapa Bay). Initial settlement density of the May cohort was as high as 215 crabs/m2 in intertidal eelgrass beds along the Strait of Juan de Fuca and decreased to <2 crabs/m2 within Puget Sound and the Strait of Georgia. A second cohort apparently originated in Hood Canal (a deep inland fjord), its size upon settlement in June was significantly smaller (J1 x= 5.3 mm CW) than the May cohort, and it was limited to Hood Canal and areas of Puget Sound close to the mouth of Hood Canal. A third cohort, which settled in late July and August, was the smallest of the three cohorts (J1 x= 4.8 mm CW), and was widely distributed around Puget Sound from Seattle in the south to the USA/Canadian border in the north. We hypothesize that most juvenile recruitment in Hood Canal and Puget Sound originates from parental stocks endemic to their respective basins (Hood Canal and Puget Sound cohorts), but that, on occasion, oceanographic conditions allow substantial influx of Pacific Ocean Dungeness crab larvae (oceanic cohort) through the Strait of Juan de Fuca into Puget Sound. Tracking of spatial/temporal settlement patterns and comparison of J1 sizes proved useful for estimating the probable sources and dispersion of Dungeness crab larvae. Differences in size and time of settlement between various larval cohorts of C. magister may prove useful as biomarkers for tracing circulation patterns within and between inland waters of Washington and the Pacific Ocean. Benthic, free-living marine nematodes from two stations, one subtidal, one intertidal, in the North Inlet Estuary, South Carolina, USA, have been characterized by measurement of adenosine triphosphate (ATP), carbon, caloric content, and ash-free dry weight. Two methods of extracting the organisms from the sediment were used. Resulting carbon to ATP ratios and population density data from ongoing North Inlet meiofauna studies show that the nematodes account for 0.11 to 6.33 g ATP per g dry weight of sediment, varying seasonally. The percentage of nematode ATP/g sediment is higher in the winter and lowest in the summer, and accounts for 68% to over 90% of the total sediment ATP. The findings of this study indicate that meiofauna, particularly nematodes, can no longer be overlooked as a separate entity in attempts to partition benthic systems.Contribution No. 173 from the Belle W. Adenosine triphosphate (ATP) measurements are used to determine vertical and seasonal distributions of microorganisms and meiofauna in sediments from a 14 m-deep mud bottom in central Long Island Sound on 12 sampling dates from April 1975 to October 1976. Below the topmost 1 cm of sediment, ATP measurements can be useful in estimating and comparing standing stocks of microorganisms and meiofauna. In the top 1 cm, however, large quantities of newly settled bivalves (Yoldia limatula, Nucula annulata, and Mulinia lateralis) and juvenile polychaetes (Owenia fusiformis) in summer and fall months account for total ATP concentrations. The ATP content of individual meiofauna ranges from 1.97 ng individual copepod nauplius-1 to 190.7±60 ng individual M. lateralis -1. In general, the total ATP content of individual polychaetes and bivalves is much higher than that of individuals of other groups. However, on a mg ATP per g wet or dry tissue basis, the ATP content of micro- and meiofaunal taxa are not significantly different. In addition to providing a means for comparing micro- and meiofaunal standing stocks, ATP measurements permit examination of the relative contribution of different meiofauna to the total living biomass of meiofauna in sediments. Total sediment ATP concentrations are greatest in the top 1 cm at all seasons, and decrease with increasing depth in the sediment. Annual concentrations in the topmost centimeter average 4.22 g g dry sediment-1 and range seasonally from 1.09 to 7.64 g g dry sediment-1. At a depth of 10 cm, values average 0.16 and range from 0.019 to 0.35 g g dry sediment-1. High ATP concentrations in surface sediment reflect high concentrations of microorganisms and meiofauna at the sediment-water interface. The top 2 cm of sediment contain 71% of all meiofauna, with 41% occurring in the topmost cm. In general, densities are lowest in the winter and highest during the spring and summer, averaging 490 individuals 10 cm-2, and varying from 87 to 1366 individuals 10 cm-2. Because of wide variation in recruitment patterns of the benthos in Long Island Sound, the extremes of the range in meiofaunal densities can be observed in the same month in two different years. In order to monitor and compare standing stocks of organisms less than 1 mm in size in sediments, the ATP assay can save hours of processing time compared with alternate methods such as direct counts.  相似文献

The distribution and structure of nematode populations in 4 sedimentary environments (muds, muddy sands, fine sands and medium-coarse sands) in Long Island Sound were studied. Mean population densities were highest in muds and muddy sands. Cluster analysis suggested the presence of two basic faunistic units; a mud unit characterized by high species dominance, low species diversity and low species endemism, and a sand unit characterized by low species dominance, high species diversity and high species endemism. Species diversity in all habitats was a direct function of both species richness and equitability. Limited niche separation among deposit feeders, usually the dominant nematode trophic type in muddy sediments, is proposed as the cause for the high species dominance so often characteristic of shallow marine muds. The study afforded the opportunity to examine the quantitative and qualitative aspects of population structure in relation to environmental impact (as indicated by large differences in the heavy metal and organic carbon concentrations within each of the 4 sedimentary regimes). Within each sediment type no differences in population densities, species composition or species diversity of nematodes existed between heavily impacted and apparently non-impacted sediments, or between Long Island Sound and similar coastal regions. These findings cast doubt on (1) the use of heavy metal and organic carbon concentrations as indicators of environmental stress for marine nematodes; (2) the use of diversity indices alone as indicators of environmental deterioration; and (3) the usefulness of field monitoring studies alone for the assessment of pollution impact on marine nematodes.Contribution no. 100 from the Institute of Marine and Atmospheric Sciences, City College of New YorkCommunicated by M.R. The standing stock of benthic macroalgae, sediment-associated microalgae and eelgrass (Zostera marina L.) was sampled in conjunction with irradiance and water properties from June 1982 through March 1984 to examine the relationship between the dynamics of benthic primary producers and environmental factors in central Puget Sound, Washington, USA. Sediment-associated microalgal standing stock (measured by chlorophylla) peaked in April and August. The seaweed assemblage, dominated by bladed green algae (e.g.Ulva fenestrata) and eelgrass exhibited maximum standing stocks in August. Although water temperature correlated best with changes in standing stock of all vegetation types, solar irradiance appeared to trigger the onset of biomass buildup and autumn die-back by the plants. Seasonal variations in dissolved oxygen reflected the buildup and loss of plant standing-stock. Nutrient concentrations, with the exception of ammonia, exhibited seasonal trends. Most nutrients were in greatest concentration in winter and reached minimum concentration in late spring-summer. Regeneration of nutrients in autumn followed shortly after the autumn dieback of the benthic vegetation. We concluded that irradiance was the primary controlling factor in the system. Nutrient limitation (primarily nitrate) may control standing-stock accumulations from the period May–October when light is not limiting. The bioluminescent fish Porichthys notatus (plainfin midshipman), has a discontinuous distribution along the Pacific coast of North America. The fish is present from Cape Mendocino southward to Baja California, Mexico, absent off the coast of Oregon, USA, and abundant, northward, in Puget Sound, Washington. Interestingly, the population in Puget Sound lacks the substrate (luciferin) necessary for the luminescence reaction and, despite possessing an otherwise fully functional photophore system, is nonluminescent. The California population of P. notatus is uniformly luminescent south of Monterey Bay, but 15% of the speciments tested from San Francisco Bay and the Gulf of the Farallons have been reported to be nonluminescent. Explanations for nonluminescent midshipman in both Puget Sound and the San Francisco Bay area have focussed on a dietary requirement for luciferin. To gain further insight into reasons for nonluminescence in the San Francisco Bay region, the distribution of bioluminescence in P. notatus was studied from Monterey Bay to Cape Mendocino during 1985. A complex pattern of bioluminescence was found, in which nonluminescent individuals reflected neither a local anomaly in the San Francisco Bay region nor a simple gradient of decreasing luminescence towards the northern end of the range of the California population. Instead, a distinct size-dependent component in luminescence capability of the fish was observed. The ability of microbial populations to mediate the anaerobic transformation of four aromatic compounds (aniline, benzoic acid, pyridine, and quinoline) in sediments of the Tsengwen River was examined. Along the river, from a freshwater (0.0° salinity) to an oceanic (37.0° salinity) environment, five sampling stations were chosen to collect the sediment. Sediment slurries were incubated in an anaerobic mineral salts medium that was amended with multiple electron acceptors, including manganese (IV) and ferric (III) oxides, and the concentrations of the aromatic substrates were followed over a 3- to 4-month period. Most sediment samples showed a complete loss of benzoic acid and quinoline (0.12–0.21 mM) within approximately three months. Pyridine was transformed after a lag period of 53 days in the sediment slurries from the freshwater environment which had been amended with both metal oxide (either Fe (OH)3 or MnO2) and inhibitor (either BESA or molybdate). Pyridine was not transformed in other sediment slurries. No significant metabolism of aniline was apparent in any of the sediments.  相似文献

The planktonic marine copepodCalanus pacificus exhibits an enhanced feeding rate, or hunger response, when exposed to food following short periods of starvation. In a scries of laboratory experiments with copepods collected from the main basin of Puget Sound, Washington, during 1982 and 1984, we measured maximum ingestion rate, assimilation efficiency, and digestive enzyme activity to determine the time scales over which the feeding behavior ofC. pacificus responds to increases in food. These laboratory results were then compared to field studies of diel fluctuations in digestive enzymes and gut fluorescence ofC. pacificus in Dabod Bay, a fjord of Puget Sound, during September, 1980, and the closely relatedC. marshallae off the Washington coast, in August, 1981. Laboratory experiments demonstrated that the hunger response ofC. pacificus lasts approximately 6 h before ingestion rate returns to a steady state level of about one-half maximum. On the order of 12h of starvation were required to induce the maximum ingestion rate of the hunger response. Digestive enzyme activities did not change over these time scales. Assimilation efficiency peaked within a few hours of the onset of feeding, with low initial rates possibly related to the period of starvation prior to feeding. These results were consistent with diel patterns observed in the field. The abundance and species composition of diatoms were investigated along the PN line from the Okinawa Islands to the inner continental shelf in the East China Sea in the early spring of 1996. Viable diatom resting stages in sediments on the shelf were also enumerated by the extinction dilution method (most probable number method). Clear differentiation in the water masses was observed, with less saline, cold water (shelf water) on the shelf region, and warm, saline water (Kuroshio water) prevalent off the shelf and on the shelf edge. In the Kuroshio water, the abundance of diatoms was generally low but species diversity of diatoms was relatively high. In contrast, the spring bloom of diatoms was clearly observed in the shelf water where the water column was weakly stratified. The bloom was dominated by Chaetoceros debilis, contributing occasionally >80% of the diatom community. Resting stages of this species were also the most abundant taxon in the sediments on the shelf. Resuspension of the sediment during winter mixing of the water column should have enabled the resting stages to germinate at the surface. Subsequent vegetative growth after germination led to the formation of an early spring bloom of C. debilis when the water column was stratified and light availability had increased. Intermittent resuspension of sediment on the shelf, driven by strong winds and tidal currents, probably provides opportunities for diatoms with a resting stage to exploit favorable conditions for their germination and subsequent vegetative growth. It is further suggested that complex hydrographic conditions in the East China Sea result in a dynamic bloom with contributions by both autochthonous and allochthonous species.Communicated by T. Morphological characters and 14 enzyme loci were examined for 1 040 sea stars, currently recognized as forms ofLeptasterias hexactis, from Lynn Canal, Alaska, and Puget Sound, Washington, USA, between March 1988 and April 1989. Three morphologically and two genetically distinctLeptasterias forms were identified. The most common form found at both localities wasL. epichlora (Brandt)sensu Verrill.L. hexactis (Stimpson)sensu Verrill co-occurred withL. epichlora at all study sites and apparently hybridizes extensively withL. epichlora in the Puget Sound region, but rarely, if at all, in Alaska. The presumptive product of this hybridization morphologically resembledL. aequalis (Stimpson)sensu Fisher, and was conspicuously absent from Alaskan samples. Considerable genetic distance existed betweenL. epichlora andL. hexactis (Nei'sD=0.19±0.01) and moderate genetic differentiation occurred between populations of each species from Alaska and Washington (Weir and Cockerham'sF RT =0.29±0.04 forL. epichlora and 0.21±0.15 forL. hexactis). A significant (p<0.05) deficiency in the proportion of heterozygous individuals was found compared to Hardy-Weinberg expectations (Wright's fixation index,F ID=0.12±0.04 and 0.31±0.08 forL. epichlora andL. hexactis, respectively). However, mean observed heterozygosity for each species (0.09±0.03, 0.14±0.04 and 0.14±0.04 forL. epichlora, L. hexactis andL. aequalis, respectively) fell within the range of reported values for other asteroid species (ca. 0.04 to 0.37). The results of this study indicate that considerable genetic integrity is maintained betweenL. epichlora andL. hexactis, which warrants their recognition as distinct species despite their apparent hybridization in the Puget Sound region.  相似文献

Postlarval megalopae of the Dungeness crabCancer magister inhabiting offshore coastal waters along the west toast of North America are larger and settle earlier than do those occupying the inland waters of the Puget Sound basin (Washington, USA, and British Columbia, Canada). The Puget Sound habitat is characterized by low initial temperatures that steadily increase during the course of zoeal development, while offshore temperatures are more moderate and stable. Larvae were raised in the laboratory from hatching to megalopa in three temperature treatments: (1) constant 10 °C (CO), (2) a regime that temporally simulated temperatures found off the central California coast during the larval season (CA); and (3) a regime simulating temperatures found within the Puget Sound basin (PS). Zoeal duration was 44% longer in the PS treatment than in the other two, at least partially accounting for observed differentes between outer-toast and inland-water settlement times. Although differentes were measured in megalopal weights and carapace lengths among treatments, results do not explain differences in megalopal size observed between outer-toast and inland-water individuals. Survival to megalopa was highest in the PS treatment, with the difference due to significantly lower mortality than in CO and CA treatments during the terminal zoeal stage. Daily instantaneous mortality rates were lower in the PS than in CO or CA treatments, indicating Chat extended larval duration will not necessarily result in reduced settlement success.  相似文献

Sediment was collected from three locations along a pollution gradient in Narragansett Bay and transplanted to controlled mesocosms. Total hydrocarbons and eleven individual polycyclic aromatic hydrocarbons (PAHs) were measured in these sediments over a period of 394 days. Total hydrocarbon concentrations increased in the “relatively uncontaminated”; (Rhode Island Sound) sediment that was held in the mesocosms, but did not change in the two other sediments. The concentrations of four PAHs: naphthalene, 2‐methyl naphthalene, 1‐methyl naphthalene and biphenyl, decreased in the “contaminated”; (Providence River) sediment during the experiment and the calculated half‐lives for these compounds were 287, 353, 321 and 333 days, respectively.  相似文献

Relative rates of the anaerobic decomposition of organic matter in the upper 10 cm of sediment from two stations in central Long Island Sound, USA, were compared. Sediment samples from discrete depth intervas were incubated anoxically and changes in SO 4 = , NH 4 + , bacterial numbers, extractable adenosine triphosphate (ATP), organic matter, and organic carbon were measured as a function of time and temperature. At both stations (15 and 34 m water-depths, respectively), the calculated rates of SO 4 = reduction and NH 4 + production decreased exponentially (approximately) with depth below the sediment-water interface. Over the same depth interval, ATP concentrations dropped by a factor of 6 to 7 and bacterial numbers were lower by a factor of 2 to 3. These decreases in SO 4 = reduction, NH 4 + production, bacterial numbers, and ATP, reflect a change in the physiological state of microbial populations with depth in the sediment and are consistent with the conclusions that the quantity of easily utilizable organic matter changes rapidly below the sediment surface and that food limitation controls the basic depth distribution of microbial activity. The average rates of SO 4 = reduction, 29 to 39 mM year-1 (22°C), in the top 10 cm are similar at both stations studied here, as well as at an additional station from a previous study. In contrast, average NH 4 + production differs by a factor of 2 at the two stations, reflecting differences in the C:N ratio of the organic matter supplied to the sediment surface and differences in particle reworking by macrofauna at each site. The apparent activation energy of SO 4 = reduction was 19±1 kcal mole-1 and that of NH 4 + production, 18±3 kcal mole-1. The overall quantity of carbon required to support the calculated average SO 4 = reduction rate in the top 10 cm is 23 g C m-2 year-1 and represents 36% of all the carbon available to the benthos annually and 11% of the net primary production in the water column. Directly measured fluxes of NH 4 + from sediments to overlying water at both stations agree well with those predicted from production rates obtained by the incubation techniques.  相似文献

Catastrophic die-offs can have important consequences for vertebrate population growth and biodiversity, but catastrophic risks are not commonly incorporated into endangered-species recovery planning. Natural (e.g., landslides, floods) and anthropogenic (e.g., toxic leaks and spills) catastrophes pose a challenge for evolutionarily significant units (ESUs) of Pacific salmon listed under the Endangered Species Act and teetering at precariously low population levels. To spread risks among Puget Sound chinook salmon populations, recovery strategies for ESU-wide viability recommend at least two viable populations of historical life-history types in each of five geographic regions. We explored the likelihood of Puget Sound chinook salmon ESU persistence by examining spatial patterns of catastrophic risk and testing ESU viability recommendations for 22 populations of the threatened Puget Sound chinook salmon ESU. We combined geospatial information about catastrophic risks and chinook salmon distribution in Puget Sound watersheds to categorize relative catastrophic risks for each population. We then analyzed similarities in risk scores among regions and compared risk distributions among strategies: (1) population groups selected using the ESU viability recommendations of having populations spread out geographically and including historical life-history diversity, and (2) population groups selected at random. Risks from individual catastrophes varied among populations, but overall risk from catastrophes was similar within geographic regions. Recovery strategies that called for two viable populations in each of five geographic regions had lower risk than random strategies; strategies that included life-history diversity had even lower risks. Geographically distributed populations have varying catastrophic-risks profiles, thus identifying and reinforcing the spatial and life-history diversity critical for populations to respond to environmental change or needed to rescue severely depleted or extirpated populations. Recovery planning can promote viability of Pacific salmon ESUs across the landscape by incorporating catastrophic risk assessments.  相似文献

This paper describes a quantitative investigation of relationships between the growth of phytoplankton, and climatic and hydrodynamci conditions in temperate fjords with marked tides, as exemplified by Puget Sound, Washington (USA). Algal growth in the open waters of the central basin of the Sound is dominated by a number of intense blooms beginning in late April or May and recurring throughout the summer. Rarely, and only briefly, does nitrate become exhausted. The phytoplankton production rate in the central basin of Puget Sound is about 465 g C m-2 year-1. During the springs of 1966 and 1967, oceanographic measurements were carried out at a mid-channel station with sufficient frequency to allow investigation of physical and biological processes with time scales of the order of a day. The principal investigative tool is a numerical model in which the hydrodynamical conditions are represented by an approximate analysis of the gravitational convection mode of circulation. Algal concentration is represented as a continous function of space and time in the model which ascribes changes in phytoplankton density to variations in photosynthetic and respiratory activity, algal sinking, grazing by herbivores, and to mixing and advection. Computations adequately reproduce the principal features of phytoplankton concentrations observed during 75 days and 35 days in the springs of 1966 and 1967, respectively. Numerical experiments assess the relative importance of various processes which govern the level of primary production in Puget Sound. It is concluded that phytoplankton growth is limited by a combination of factors, including vertical advection and turbulence, modulation of underwater light intensity by self-shading and inorganic particulates, sinking of algal cells, and occasional rapid horizontal advection of the population from the area by sustained winds. The high primary productivity of the Sound is due to intensive upward transport of nitrate by the estuarine mechanism. These results should be generally applicable to other temperate fjords because of the largely conventional choice of the biological functions.  相似文献

Tidal currents, seston, and sediments separately influence growth of the hard clam, Mercenaria mercenaria, but it is uncertain how these factors may interact. A 3×3 factorial field experiment, carried out in Great Sound, a coastal lagoon in Southern New Jersey, USA, between May and September 1986, determined the relative effects of three sediment types and three site-specific seston/tidal current regimes on the individual growth of M. mercenaria. Analysis of variance of the change in shell length after 15 wk (differences in initial and final lengths) demonstrated a significant difference (P=0.0064) in growth among sites, but no significant differences (P=0.1331) for growth in different sediments, although trends were evident. Effects of sites were independent of sediment type (P=0.2621). Shell growth rates differed by 10.7% between the slowest and fastest sites, but only differed by 5.7% between sediment types, with fastest growth in sand and slowest in mud. Tidal current speeds and four measures of seston (chlorophyll a, particulate inorganic and organic matter, PIM and POM, and energy content) were measured >20 times in near-bottom waters at each site. Horizontal fluxes of POM exhibited higher correlation coefficients with growth rates, than did seston concentrations or current speeds alone. We attribute significant site differences to differences in horizontal seston fluxes fluxes among sites.
