The effect of exposure to sodium arsenite during development was tested on adult fluctuating asymmetry (FA) in sternopleural bristle number, bristle number, body size and survivorship in Drosophila melanogaster. Three genetic strains of flies were used, CT-106, PVM and Oregon-R, and arsenite concentrations ranged from relatively mild, sub-lethal doses, to concentrations with pronounced negative effects on survivorship. At arsenite concentrations of 1.0 and 0.125 mM, mortality was on average 38% greater than in controls, and surviving flies emerged significantly smaller and had fewer bristles than controls. Neither the effect of arsenite or the genotype x environment interaction on asymmetry were significant. However, given the high mortality, any increase in FA could have been masked by the outcome of developmental selection against developmentally unstable phenotypes. We tested for this effect by contrasting FA values between (1) flies reared at the highest concentrations used previously, (2) flies reared at sub-lethal dosage, and (3) controls. Positional fluctuating asymmetry (PFA), which is expected to be a sensitive indicator of underlying developmental stability, was significantly reduced among flies reared at the highest concentration, and at which flies suffered significant mortality. Moreover, the slope of the regression relating mean PFA to emergence per bottle was significantly positive. These data support the hypothesis that developmental selection occurred in this experiment, and that the expected positive relationship between asymmetry and stress may be altered when the stressor eliminates individuals from the population. In contrast, FA of flies reared at sub-lethal dosage did not differ from that in controls, a result that fails to support the hypothesis that arsenite disrupts developmental stability. Our results call for caution in FA-based biomonitoring, especially of potentially lethal forms of stress, because in the presence of developmental selection, and under the common assumption that FA should increase under stress, erroneous conclusions may be drawn about the health and well being of a population. It is suggested that FA-based biomonitoring efforts integrate the use of FA with other bioindicators, and experimentally validate any expected FA-stress relationship before attempting to infer the presence of environmental stress.  相似文献

In this study, the interaction between temperature and heavy metal stress was investigated in two wheat varieties (Triticum aestivum L. cv. Gerek-79 and Bolal-2973). Effects of different concentrations of lead and cadmium (0, 50, 100, 250, 500 mg l(-1)) simultaneously applied at various temperatures (8/4, 25/18, 35/26 degrees C day/night) to the seedlings were detected by measuring changes in the plant length, percentage of dry weight, chlorophyll (a, b, a/b), total soluble phenolics, and free proline. At the high cadmium concentrations, significant changes occurred in plant length and in the amounts of dry weight, chlorophyll, total soluble phenolics, and free proline. Significant differences were not observed in the parameters examined for the lead treatments. These data indicated that heavy metal toxicity increases in parallel with temperature. In addition, lead did not have any toxic effects on plants in sand-perlite mixture at 500 mg l(-1), whereas cadmium showed toxic effect even at such low concentrations as 50 mg l(-1).  相似文献

Many ecotoxicological studies have addressed the effects of contaminant exposure at various levels of biological organization. However, little information exists on the effects of toxicants on wildlife populations. Here we examined exposure of populations of the greater white-toothed shrew Crocidura russula (Soricomorpha, Soricidae) occupying two protected Mediterranean sites (a polluted area, the Ebro Delta, and a control site, Garraf Massif). Bioaccumulation of selected elements (Pb, Hg, Cd, Zn, Cu, Fe, Mn, Cr, Mo, Sr, Ba, and B), a body condition index (BCI) and fluctuating asymmetry (FA) were used to assess the chronic exposure to environmental pollution. BCI was correlated neither to metal concentrations nor to FA, suggesting that this fitness measure only reflects environmental disturbances at a local level. However, shrews from the polluted area showed higher concentrations of metals and metalloids (Pb, Hg, B, and Sr) and greater shape FA than specimens from the reference area. A correlation between FA was found for both first and second principal component vectors suggesting that developmental instability increases as a result of exposure to multiple pollutants. Our results corroborate the suitability of C. russula as a bioindicator of environmental quality and show that FA is an appropriate index to examine impact of developmental stressors in populations inhabiting disturbed areas.  相似文献

The rational for the study was to review the literature on the toxicity and corresponding mechanisms associated with lead (Pb), mercury (Hg), cadmium (Cd), and arsenic (As), individually and as mixtures, in the environment. Heavy metals are ubiquitous and generally persist in the environment, enabling them to biomagnify in the food chain. Living systems most often interact with a cocktail of heavy metals in the environment. Heavy metal exposure to biological systems may lead to oxidation stress which may induce DNA damage, protein modification, lipid peroxidation, and others. In this review, the major mechanism associated with toxicities of individual metals was the generation of reactive oxygen species (ROS). Additionally, toxicities were expressed through depletion of glutathione and bonding to sulfhydryl groups of proteins. Interestingly, a metal like Pb becomes toxic to organisms through the depletion of antioxidants while Cd indirectly generates ROS by its ability to replace iron and copper. ROS generated through exposure to arsenic were associated with many modes of action, and heavy metal mixtures were found to have varied effects on organisms. Many models based on concentration addition (CA) and independent action (IA) have been introduced to help predict toxicities and mechanisms associated with metal mixtures. An integrated model which combines CA and IA was further proposed for evaluating toxicities of non-interactive mixtures. In cases where there are molecular interactions, the toxicogenomic approach was used to predict toxicities. The high-throughput toxicogenomics combines studies in genetics, genome-scale expression, cell and tissue expression, metabolite profiling, and bioinformatics.  相似文献

The study aimed to monitor heavy metal (chromium, Cr; cadmium, Cd; nickel, Ni; copper, Cu; lead, Pb; iron, Fe; manganese, Mn; and zinc, Zn) footprints in biological matrices (urine, whole blood, saliva, and hair), as well as in indoor industrial dust samples, and their toxic effects on oxidative stress and health risks in exposed workers. Overall, blood, urine, and saliva samples exhibited significantly higher concentrations of toxic metals in exposed workers (Cr; blood 16.30 μg/L, urine 58.15 μg/L, saliva 5.28 μg/L) than the control samples (Cr; blood 5.48 μg/L, urine 4.47 μg/L, saliva 2.46 μg/L). Indoor industrial dust samples also reported to have elevated heavy metal concentrations, as an example, Cr quantified with concentration of 299 mg/kg of dust, i.e., more than twice the level of Cr in household dust (136 mg/kg). Superoxide dismutase (SOD) level presented significant positive correlation (p?≤?0.01) with Cr, Zn, and Cd (Cr?>?Zn?>?Cd) which is an indication of heavy metal’s associated raised oxidative stress in exposed workers. Elevated average daily intake (ADI) of heavy metals resulted in cumulative hazard quotient (HQ) range of 2.97–18.88 in workers of different surgical units; this is an alarming situation of health risk implications. Principal component analysis-multiple linear regression (PCA-MLR)-based pie charts represent that polishing and cutting sections exhibited highest metal inputs to the biological and environmental matrices than other sources. Heavy metal concentrations in biological matrices and dust samples showed a significant positive correlation between Cr in dust, urine, and saliva samples. Current study will help to generate comprehensive base line data of heavy metal status in biomatrices and dust from scientifically ignored industrial sector. Our findings can play vital role for health departments and industrial environmental management system (EMS) authorities in policy making and implementation.  相似文献

Heavy metal pollution is a severe environmental problem. Remediation of contaminated soils can be accomplished using environmental materials that are low cost and environmentally friendly. We evaluated the individual and combination effects of humic acid (HA), super absorbent polymer (SAP), zeolite (ZE), and fly ash composites (FC) on immobilization of lead (Pb) and cadmium (Cd) in contaminated soils. We also investigated long-term practical approaches for remediation of heavy metal pollution in soil. The biochemical and morphological properties of maize (Zea mays L.) were selected as biomarkers to assess the effects of environmental materials on heavy metal immobilization. The results showed that addition of test materials to soil effectively reduced heavy metal accumulation in maize foliage, improving chlorophyll levels, plant growth, and antioxidant enzyme activity. The test materials reduced heavy metal injury to maize throughout the growth period. A synergistic effect from combinations of different materials on immobilization of Pb and Cd was determined based on the reduction of morphological and biochemical injuries to maize. The combination of zeolite and humic acid was especially effective. Treatment with a combination of HA?+?SAP?+?ZE?+?FC was superior for remediation of soils contaminated with high levels of Pb and Cd.  相似文献

Background, aim, and scope

The purpose of this study was to monitor and present the heavy metal concentrations in the blood of residents of areas near municipal waste incinerators (MWIs), who are more prone to environmental pollution. We also sought to compare and analyze the residents?? perception of environmental pollution as one of the factors affecting heavy metal concentrations in the blood using a survey about the perceived damage caused by the facilities. Since heavy metal levels in the blood can be affected not only by local environmental pollution but also by personal and occupational factors, heavy metal levels in the blood need to be verified and consistently monitored.

Methods

Residents who live within 300?m of MWIs in Seoul are acknowledged to be under indirect influence according to the Waste Disposal Act. A survey was given to 841 residents living within 300?m of a MWI from 2006 to 2009. The concentrations of heavy metals (lead, cadmium, and mercury) in the blood were measured in the 841 surveyed residents and in 105 residents in reference areas. Additionally, the perception of the damage caused by municipal waste incinerators was investigated using scores from 1 to 5 on a Likert scale.

Results

The measurements of the heavy metal concentrations in the blood showed that the mean concentrations of lead, cadmium, and mercury were 43.1, 1.7, and 1.3?ug/L, respectively. The blood levels of lead and cadmium were slightly higher in the group of the subjects who had resided the longest near the municipal waste incinerators. When compared with the domestic investigation by the Ministry of Environment, the concentrations of lead and cadmium were a little higher, while that of mercury was a little lower. Overall, there was no significant difference in the distribution of heavy metal levels in the blood among age groups. Background, aim, and scope  

Heavy metals such as lead are well known to cause harmful health effects. Especially children are particularly susceptible to increased levels of lead in their blood. It is also a fact that lead concentration is increasing in the environment due to increased anthropogenic activity. The risk of heavy metal contamination is pronounced in the environment adjacent to large industrial complexes. In a combined case study, the environmental pollution by heavy metals was related to children’s health in the vicinity of an industrial area located 4 km south-east from Bucharest about 2 km east from the nearest town—Pantelimon. This site includes companies processing different, nonferrous solid wastes for recovery of heavy metals and producing different nonferrous alloys and lead batteries. Genotypic and environmental variation in Cr, Cd and Pb concentrations of rice grains and the interaction between these metals were investigated by using 138 rice genotypes grown in three contaminated soils. There were significant genotypic differences in the three heavy metal concentrations of rice grains, with the absolute difference among 138 genotypes in grain Cr, Cd and Pb concentrations being 24.5-, 9.1- and 23.8-folds, respectively, under the slightly contaminated soil (containing 4.61mgkg(-1) Cr, 1.09mgkg(-1) Cd and Pb 28.28mgkg(-1), respectively). A highly significant interaction occurred between genotype and environment (soil type) in the heavy metal concentrations of rice grains. Cr concentration in rice grains was not correlated with Cd and Pb concentration. However, there was a significant correlation between Cd and Pb in slightly and highly contaminated soils. The results suggest the possibility to develop the rice cultivars with low Cd and Pb concentrations in grain.  相似文献

The plant metabolic response to heavy metal stress is largely unknown. The present investigation was undertaken to examine the influence of different concentrations of potassium dichromate on the Zea mays L. plantlets. A clear effect of chromium on maize plantlets growth and seed germination was observed strating from 100-300 ppm up to 1500 ppm. In this concentration range, chromium uptake was dependent on the concentration in the medium. Metallothioneins, involved in heavy metal binding, were measured by capillary electrophoresis (CE), and showed a dose-response induction. Protein profile analyzed by two-dimensional gel electrophoresis showed differential expression of several proteins. Identification of spots of upregulated proteins was performed by MALDI mass spectrometry. Results showed that proteins induced by heavy metal exposure are principally involved in oxidative stress tolerance or in other stress pathways. Induction of proteins implicated in sugar metabolism was also observed. Identification of factors involved in plant response may lead to a better understanding of the mechanisms involved in cell protection and tolerance. This information could be used to improve agricultural production and environmental quality.  相似文献

Natural fulvic acids (FA) from the Rio Santiago, an affluent of the Rio de la Plata estuary, were isolated by adsorption on XAD-2 at pH 2, and were then characterized by IR and fluorescence spectra, acidity and complexing capacity to Cu(II). The effect of the interaction between FA and Cu(II) on the acute toxicity of this heavy metal was assayed with the rotifer Brachionus calyciflorus in standardized in-vitro tests. A CuSO(4).5H(2)O concentration of 0.080 ppm produced 89.2% mortality in the absence of FA. At FA/Cu ratios >/= 1 no mortality was observed. Based on the values of the FA-Cu binding parameters and acute toxicity inhibition, it was possible to predict the effect of dissolved organic matter on the toxicity of copper in this surface freshwater body.  相似文献

以鲫鱼(Carassius auratus)作为实验对象,经过40 d Pb2+、Cd2+不同浓度的暴露后,运用SDS-PAGE和Western Blotting方法检测鱼肝脏组织内应激蛋白HSP70的诱导表达情况.结果表明,在实验浓度下,与对照组相比,Pb2+、Cd2+对鱼肝脏内HSP70有显著的诱导(P<0.05),但在Cd2+浓度为0.2 mg/L有停止表达HSP70的现象,可能是由于浓度过高造成了组织病理损伤而破坏了诱导表达机制.实验还发现,在实验浓度低于国家渔业用水标准时,HSP70仍然表现为明显诱导(P<0.05),充分说明运用分子生物学指标要比传统的环境检测指标敏感,具有对污染物早期预警的作用.  相似文献

Among heavy metals, lead (Pb) is a non-essential metal having a higher toxicity and without any crucial known biological functions. Being widespread, non-biodegradable and persistent in every sphere of soil, air and water, Pb is responsible for severe health and environmental issues, which need appropriate remediation measures. However, microbes inhabiting Pb-contaminated area are found to have evolved distinctive mechanisms to successfully thrive in the Pb-contaminated environment without exhibiting any negative effects on their growth and metabolism. The defensive strategies used by bacteria to ameliorate the toxic effects of lead comprise biosorption, efflux, production of metal chelators like siderophores and metallothioneins and synthesis of exopolysaccharides, extracellular sequestration and intracellular bioaccumulation. Lead remediation technologies by employing microbes may appear as potential advantageous alternatives to the conventional physical and chemical means due to specificity, suitability for applying in situ condition and feasibility to upgrade by genetic engineering. Developing strategies by designing transgenic bacterial strain having specific metal binding properties and metal chelating proteins or higher metal adsorption ability and using bacterial activity such as incorporating plant growth-promoting rhizobacteria for improved Pb resistance, exopolysaccharide and siderophores and metallothionein-mediated immobilization may prove highly effective for formulating bioremediation vis-a-vis phytoremediation strategies.

The aim of this research was to assess the degree of heavy metal (cadmium, lead and mercury) contamination of wild boars from central Croatia and thereby conduct further tests to connect the contamination to oxidative stress occurrence. The highest cadmium concentration was noticed in the kidneys of older boars, the lead concentration was approximately the same in the liver and kidneys, but it was much higher than the concentration in the muscles, while the highest mercury concentration was measured in the kidneys of all wild boars. The correlation between the malondialdehyde (MDA) and the heavy metal concentrations was higher in tissue samples taken from younger animals (1 to 3 years of age) than in the tissue samples taken from older animals (5 to 6 years of age). In the kidneys of all wild boars there was a statistically significant connection between the MDA and cadmium concentration; in the kidneys of younger wild boars there was a statistically significant connection between the MDA and lead concentration. Further research on this topic should focus on MDA potential to find its use, not only as a contamination biomarker in the area of ecotoxicology, but also in the evaluation of the hygienic acceptability of animal products.  相似文献

Plants need to survive with changing environmental conditions, be it different accessibility to water or nutrients, or attack by insects or pathogens. Few of these changes, especially heavy metal stress, can become more stressful and needed strong countermeasures to ensure survival of plants. Priming, a pre-sowing hydration treatment, involves pre-exposure of plants to an eliciting component which enhance the plant’s tolerance to later stress events. By considering the role of micronutrients in aiding plants to cope up under adverse conditions, this review addresses various aspects of micronutrient seed priming in attenuating heavy metal stress. Priming using micronutrients is an adaptive strategy that boosts the defensive capacity of the plant by accumulating several active or inactive signaling proteins, which hold considerable importance in signal amplification against the triggered stimulus. Priming induced ‘defence memory’ persists in both present generation and its progeny. Therefore, it is considered a promising approach by seed technologist for commercial seed lots to enhance the vigour in terms of seed germination potential, productivity and strengthening resistance response against metalloid stress. The present review provides an overview regarding the potency of priming with micronutrient to ameliorate harmful effects of heavy metal stress, possible mechanism how attenuation is accomplished, role of priming in enhancing crop productivity and inducing defence memory against the metalloid stress stimulus.

Concern about the effects of environmental contaminants on immune function in both humans and wildlife is growing and practically nothing is known about this impact on terrestrial invertebrates, even though they are known to easily accumulate pollutants. We studied the effect of industrial heavy metal contamination on immune defense of a free-living wood ant (Formica aquilonia). To find out whether ants show an adapted immune function in a polluted environment, we compared encapsulation responses between local and translocated colonies. Local colonies showed higher heavy metal levels than the translocated ones but the encapsulation response was similar between the two groups, indicating that the immune system of local ants has not adapted to high contamination level. The encapsulation response was elevated in moderate whereas suppressed in high heavy metal levels suggesting higher risk for infections in heavily polluted areas.  相似文献

In industrialized countries river floodplains can be strongly polluted with heavy metals. Published studies on effects of heavy metal pollution on soil invertebrates in floodplains, however, are inconclusive. This is unexpected since studies in other less dynamic environments reported clear effects at even lower levels of pollution. Flooding induces extra variation in invertebrate biomass and abundance which may reduce the probability to detect heavy metal effects. In this paper we combine reported data from studies on river floodplains in The Netherlands and Belgium and statistically analyze the effect of heavy metals on species composition, biomass, density and individual weight of earthworms. Interaction effects of heavy metal stress and flooding are also considered. The results suggest clear effects of zinc and copper on all variables and interaction of heavy metals and flooding for individual weight.  相似文献

Phytoextraction has been proposed as an alternative remediation technology for soils polluted with heavy metals or radionuclides, but is generally conceived as too slow working. Enhancing the accumulation of trace pollutants in harvestable plant tissues is a prerequisite for the technology to be practically applicable. The chelating aminopolycarboxylic acid, ethylene diamine tetraacetate (EDTA), has been found to enhance shoot accumulation of heavy metals. However, the use of EDTA in phytoextraction may not be suitable due to its high environmental persistence, which may lead to groundwater contamination. This paper aims to assess whether ethylene diamine disuccinate (EDDS), a biodegradable chelator, can be used for enhanced phytoextraction purposes. A laboratory experiment was conducted to examine mobilisation of Cd, Cu, Cr, Ni, Pb and Zn into the soil solution upon application of EDTA or EDDS. The longevity of the induced mobilisation was monitored for a period of 40 days after application. Estimated effect half lives ranged between 3.8 and 7.5 days for EDDS, depending on the applied dose. The minimum observed effect half life of EDTA was 36 days, while for the highest applied dose no decrease was observed throughout the 40 day period of the mobilisation experiment. Performance of EDTA and EDDS for phytoextraction was evaluated by application to Helianthus annuus. Two other potential chelators, known for their biodegradability in comparison to EDTA, were tested in the plant experiment: nitrilo acetic acid (NTA) and citric acid. Uptake of heavy metals was higher in EDDS-treated pots than in EDTA-treated pots. The effects were still considered insufficiently high to consider efficient remediation. This may be partly due to the choice of timing for application of the soil amendment. Fixing the time of application at an earlier point before harvest may yield better results. NTA and citric acid induced no significant effects on heavy metal uptake.  相似文献

Measurements were made regarding the adsorption of lead, cadmium, copper, zinc and barium to freshwater surface coatings (biofilms and associated minerals), which were collected in Nanhu Lake in Jilin Province, PR China, in order to investigate the variability of adsorption capacities of these heavy metals mentioned in the above surface coatings. The adsorption of lead and other heavy metals to the biofilms was observed to decrease in the following order: copper, lead, zinc, cadmium, and barium. Generally, the values of Gamma(max) (the maximum adsorption, micromol/m(2)) increased with the standard electrode potential of metal elements used and were recorded as 166.7, 40.0, 29.4, 10.8, and 1.8 for copper, lead, zinc, cadmium and barium, respectively. The values of 1/Gamma(max) increased linearly with the decrease in values of the standard electrode potential of metal elements with a significant correlation (n=5, p=0.01) and increased linearly with the increase in values of covalent radius of metal elements with a significant correlation (n=5, p=0.05). This indicates that standard electrode potential and covalent radius were two of the principal characteristics of metals employed, causing the variation of lead and other heavy metal adsorption to the surface coatings.  相似文献
