Biopurification systems (BPS) have been introduced to minimise the risk for point source contamination of natural water resources by pesticides. Their depuration efficiency relies mostly on the high biodegradation of their packing substrate (biomixture). Despite that, little is known regarding the interactions between biomixture microflora and pesticides, especially fungicides which are expected to have a higher impact on the microbial community. This study reports the dissipation of the fungicides azoxystrobin (AZX), fludioxonil (FL) and penconazole (PC), commonly used in vineyards, in a biomixture composed of pruning residues and straw used in vineyard BPS. The impact of fungicides on the microbial community was also studied via microbial biomass carbon, basal respiration and phospholipid fatty acid analysis. AZX dissipated faster (t 1/2?=?30.1 days) than PC (t 1/2?=?99.0 days) and FL (t 1/2?=?115.5 days). Fungicides differently affected the microbial community. PC showed the highest adverse effect on both the size and the activity of the biomixture microflora. A significant change in the structure of the microbial community was noted for PC and FL, and it was attributed to a rapid inhibition of the fungal fraction while bacteria showed a delayed response which was attributed to indirect effects by the late proliferation of fungi. All effects observed were transitory and a full recovery of microbial indices was observed 60 days post-application. Overall, no clear link between pesticide persistence and microbial responses was observed stressing the complex nature of interactions between pesticides in microflora in BPS.  相似文献

Abstract

This study reports degradation of azoxystrobin (AZOXY) and imidacloprid (IMIDA) in the rice straw (RS)/corn cob (CC) and peat (P)/compost (C)-based biomixtures. The effect of biomixture preconditioning (10?days incubation prior to pesticide application), pesticide concentration and moisture content was evaluated. Results suggested that conditioning of biomixture greatly affected IMIDA degradation where half-life (t1/2) was reduced by 5–9 times. This was attributed to higher microbial biomass carbon content and dehydrogenase activity in the conditioned biomixtures. Pesticide application in the conditioned biomixture did not show any negative impact on soil microbial parameters. Both pesticides degraded at faster rate in the rice straw-based biomixtures than in the corn cob-based biomixtures. Degradation slowed down with increase in initial concentration of pesticides in biomixture and 1.6–3.0 (AZOXY) and 2.4–3.6 (IMIDA) times increase in t1/2 values was observed. The moisture content of biomixture showed positive effect on degradation which increased when moisture content was increased from 60 to 80% water holding capacity. The effect was significant for IMIDA degradation in the corn cob-based biomixtures and AZOXY degradation in the peat biomixtures. The rice straw-based biomixtures were better in degrading AZOXY and IMIDA and can be used in biopurification systems.  相似文献

The biomixture is a principal element controlling the degradation efficacy of the biobed. The maturity of the biomixture used in the biobed affects its overall performance of the biobed, but this is not well studied yet. The aim of this research was to evaluate the effect of using a typical composition of Swedish biomixture at different maturity stages on the degradation of chlorpyrifos. Tests were made using biomixture at three maturity stages: 0 d (BC0), 15 d (BC15) and 30 d (BC30); chlorpyrifos was added to the biobeds at final concentration of 200, 320 and 480 mg kg−1. Chlorpyrifos degradation in the biomixture was monitored over time. Formation of TCP (3,5,6-trichloro-2-pyrinidol) was also quantified, and hydrolytic and phenoloxidase activities measured. The biomixture efficiently degraded chlorpyrifos (degradation efficiency >50%) in all the evaluated maturity stages. However, chlorpyrifos degradation decreased with increasing concentrations of the pesticide. TCP formation occurred in all biomixtures, but a major accumulation was observed in BC30. Significant differences were found in both phenoloxidase and hydrolytic activities in the three maturity stages of biomixture evaluated. Also, these two biological activities were affected by the increase in pesticide concentration. In conclusion, our results demonstrated that chlorpyrifos can be degraded efficiently in all the evaluated maturity stages.  相似文献

We studied the contribution of each of the components of a compost-based biomixture (BX), commonly used in Europe, on pesticide degradation. The impact of other key parameters including pesticide dose, temperature and repeated applications on the degradation of eight pesticides, applied as a mixture, in a BX and a peat-based biomixture (OBX) was compared and contrasted to their degradation in soil. Incubation studies showed that straw was essential in maintaining a high pesticide degradation capacity of the biomixture, whereas compost, when mixed with soil, retarded pesticide degradation. The highest rates of degradation were shown in the biomixture composed of soil/compost/straw suggesting that all three components are essential for maximum biobed performance. Increasing doses prolonged the persistence of most pesticides with biomixtures showing a higher tolerance to high pesticide dose levels compared to soil. Increasing the incubation temperature from 15 °C to 25 °C resulted in lower t(1/2) values, with biomixtures performing better than soil at the lower temperature. Repeated applications led to a decrease in the degradation rates of most pesticides in all the substrates, with the exception of iprodione and metalaxyl. Overall, our results stress the ability of biomixtures to perform better than soil under unfavorable conditions and extreme pesticide dose levels.  相似文献

The use of biopurification systems can mitigate the effects of pesticide contamination on farms. The primary aim of this study was to evaluate the effect of pesticide dissipation on microbial communities in a pilot biopurification system. The pesticide dissipation of atrazine, chlorpyrifos and iprodione (35 mg kg?1 active ingredient [a.i.]) and biological activity were determined for 40 days. The microbial communities (bacteria, actinomycetes and fungi) were analyzed using denaturing gradient gel electrophoresis (DGGE). In general, pesticide dissipation was the highest by day 5 and reached 95%. The pesticides did not affect biological activity during the experiment. The structure of the actinomycete and bacterial communities in the rhizosphere was more stable during the evaluation than that in the communities in the control without pesticides. The rhizosphere fungal communities, detected using DGGE, showed small and transitory shifts with time. To conclude, rhizosphere microbial communities were not affected during pesticide dissipation in a pilot biopurification system.  相似文献

A field experiment was performed with the aim to assess the amount of penconazole losses during field application and the spatial variability of penconazole concentration in a vineyard soil, under two different management techniques (tilled and grass covered). The field dissipation of penconazole under the two techniques was also followed for 114 days, highlighting the effects of spatial variability of the initial concentration and of the grass covering in terms of the different soil metabolic activity. Data found show that a high percentage of penconazole, from 42.5 % to 67.43 %, can reach the soil during the treatments, despite the fact that penconazole is applied to the foliage. The high values of the coefficient of variation for data within rows show considerable variability in all applications, ranging from 30 to 65 in the first application, from 35 to 79 in the second and from 36 to 75 in the third. Since the applications of penconazole occurred under almost same climatic conditions in terms of wind speed and direction, the high variability of concentration of penconazole found within mid-rows was attributed to the uneven slope of the vineyard causing an irregular speed of the sprayer equipment. However, least significant differences of the mean values did not end in a significant difference of penconazole concentration among mid-rows for all applications, indicating that the variability between rows does not contribute to the overall variability. The calculated half-life values for penconazole in tilled soil were 62.4 days for tilled and 33.0 for grassed soil, highlighting the tendency of penconazole to faster dissipate in grassed than in tilled soil. Grassed soil was characterised by a higher metabolic activity in terms of microbial biomass carbon content, basal respiration and total hydrolytic activity, than tilled soil. A significant correlation between penconazole dissipation and total hydrolytic activity, was found. The faster dissipation of penconazole in grassed soil was attributed to the higher amount of the total hydrolytic activity which, in grassed soil, was 1.5 fold-higher than in tilled soil.  相似文献

Manure amendment in agricultural practice can have a large effect on herbicide dissipation because the period of manure plowing is close to the period of herbicide application. In addition, manure amendment is among the frequently encountered options in ameliorating pesticide pollution. In this research, the dissipation of the herbicide pendimethalin was examined after amendment with two common green manures, Lupinus luteus (L) or Cosmos bipinnatus (C), for 110 days in pH 5.2 and 7.7 soils (Sankengtzu [Sk] and Erhlin [Eh] soil, respectively). The microbial activity and ecology changes were examined by using Biolog EcoPlate and denaturing gradient gel electrophoresis (DGGE). In Sk soil, the half-lives of pendimethalin with L, C, and blank treatment were 49.0, 54.9, and 62.2 days, respectively, whereas that in Eh soil they were 46.3, 52.6, and 34.8 days, respectively. Pendimethalin dissipated quickly in more neutral soil (Eh soil), but the addition of manure can only increase the dissipation rate in acidic soil (Sk soil), indicating that the amendment of manures exerted different effect in pendimethalin dissipation rates in different pH soils. The application of pendimethalin and/or manure altered the microbial community activity after 24 h of incubation. After 110 days, the microbial community activities in green manure–amended soil were more similar to that with blank than pendimethalin treatment in both types of soils. In comparison with treatment C, microbial communities were more similar between treatment L and blank, indicating the superior effect over pendimethalin on microbial communities when applying Lupinus luteus. The research showed that the application of herbicide pendimethalin changed soil microbial community, and the amendment of manures exerted different effect in pendimethalin dissipation rates in different pH soils. It is assumed that the change in dissipation rates was originated from the microbial community change after different manure amendment.  相似文献

采用高效菌强化膜生物反应器对溴氨酸废水进行处理,考察了生物强化前后系统对溴氨酸的降解能力及其内部微生物生理状态变化.并采用核糖体基因间隔序列分析技术(RISA)解析了高效菌投加前后污泥系统的群落变化.实验表明,投菌后在进水负荷增加的条件下,上清液和膜出水的溴氨酸脱色率分别约为50%和65%,相应的COD去除率分别约为25%和50%,和投菌前基本保持一致;TTC-脱氢酶活性和胞外聚合物(EPS)浓度略有波动,但是运行一段时间后即恢复到投菌前水平.群落分析表明高效菌可以在系统中稳定存在,并且不对原菌群结构造成较大影响.  相似文献

Acetochlor is a widely used herbicide in maize fields; however, the ecological risk of its residue in the soil–plant system remains unknown. We investigated the dissipation dynamics of field dose acetochlor and clarified its impact on microbial biomass and community structure both in the rhizosphere and bulk soil over 1 month after its application. Soil microbial parameters such as quantities of culturable bacteria and fungi represented by colony-forming units, soil microbial biomass carbon (SMBC), and phospholipid fatty acids (PLFAs) were determined across different sampling times. The results showed that the dissipation half-lives of acetochlor were, respectively, 2.8 and 3.4 days in the rhizosphere and bulk soil, and 0.02–0.07 μg/g residual acetochlor could be detected in the soil 40 days after its application. Compared to the bulk soil, microbial communities in the rhizosphere soil were inclined to be affected by the application of acetochlor: SMBC content and bacterial growth were most likely to be increased; however, fungal growth was prone to be inhibited. The principal component analysis of PLFAs, as well as the comparisons of fungi/bacteria and cy17:0/C16:1ω9c ratios between different treatments over sampling time, revealed that the soil microbial community composition was significantly affected by acetochlor at its early application stage (at day 15); thereafter, the effects of acetochlor were attenuated or even could not be detected. Our results suggested that residual acetochlor did not confer a long-term impairment on viable bacterial groups in the rhizosphere and bulk soil.  相似文献

Extremely high organic load and the toxic nature of olive mill wastewater (OMW) prevent their direct discharge into domestic wastewater treatment systems. In addition to the various treatment schemes designed for such wastewater, controlled land spreading of untreated OMW has been suggested as an alternative mean of disposal. A field study was conducted between October 2004 and September 2005 to assess possible effects of OMW on soil microbial activity and potential phytotoxicity. The experiment was carried out in an organic orchard located on a Vertisol-type soil (Jezre'el Valley, Israel) and included two application levels of OMW (36 and 72m(3)ha(-1)). Total microbial counts, and to less extent the hydrolytic activity and soil respiration were increased following the high OMW application level. A bench-scale lab experiment showed that the rate of OMW mineralization was mainly dependent on the general status of soil activity and was not related to previous acclimatization of the soil microflora to OMW. Soil phytotoxicity (% germination and root elongation) was assessed in soil extracts of samples collected before and after each OMW application, using germinating cress (Lepidium sativum L.) seeds. We found direct short-term effect of OMW application on soil phytotoxicity. However, the soil was partly or completely recovered between successive applications. No further phytotoxicity was observed in treated soils as compared with control soil, 3 months after OMW application. Such short-term phytotoxicity was not in correlation with measured EC and total polyphenols in the soil extracts. Overall, the results of this study further support a safe controlled OMW spreading on lands that are not associated with sensitive aquifers.  相似文献

This investigation was undertaken to examine the dissipation rate of pyriproxyfen as well as the change in the soil bacterial community. Residues of pyriproxyfen were measured using high performance liquid chromatography (HPLC) and the changes in bacterial community were determined by comparing the 16S rDNA bands on patterns by denaturing gradient gel electrophoresis (DGGE). The dissipation of pyriproxyfen was affected by both the concentration applied and incubation temperature. Lower concentrations (1 mg Kg(-1)) and higher incubation temperatures (30 and 40°C) showed more rapid dissipation rates. The population of microbial community decreased rapidly after incubation with 10 mg Kg(-1) of pyriproxyfen for 91 days, indicating the toxicity of pyriproxyfen toward bacterial communities in a closed soil ecosystem. Lower concentrations of pyriproxyfen showed less toxicity toward the microbial community. From cluster analysis, the structure of the bacterial community showed roughly a 60 % similarity throughout the experiment period in the control experiment, indicating the stability within soil microbiota without chemical agitation. However, the similarity was lower than 50 % both in the one and 10 mg Kg(-1) of insecticide pyriproxyfen spiked experiment, indicating the soil bacterial community changed after the insecticide pyriproxyfen was applied.  相似文献

The aim of this study was to evaluate the soil microbial characteristics in historically heavy-metal polluted soil, which was also affected by organic co-contaminants, 2,4-dichlorophenol or pentachlorophenol, which often occur due to the conventional use of pesticides. It was observed that the normalized microbial biomass (microbial biomass per unit soil organic C) of the contaminated soil was very low, less than 1% in both non-planted and ryegrass planted soil, and showed a decreasing trend with the treatment of organic co-contaminants. The microbial biomass and substrate-induced respiration (SIR) in the ryegrass planted soil were much larger, as compared with the non-planted soil with or without organic pollutants. The different resistant bacterial community and its physiological diversity in the rhizosphere further suggested that the effect of vegetation on microbial activity was not just a general increase in the mass or activity of pre-existing microorganisms, but rather acted selectively on microbial growth so that the relative abundance of different microbial groups in soil was changed. In sum, high concentrations of organic co-contaminants, especially pentachlorophenol (PCP), could strengthen the deterioration of microbial ecology. The adverse effect of heavy metal-organic pollutants on the soil microbial biomass and activity might be the reason for the slow degradation of PCP that has high chlorinated and high toxicity. Vegetation might be the efficient way to assist in improving and restoring the utilization of agricultural ecosystems. The beneficial microbial effect of vegetation could cause the rapid dissipation of 2,4-dichlorophenol (2,4-DCP) that has less chlorinated and less toxicity in the planted soils.  相似文献

Mediterranean coastal ecosystems are known to be highly subject to natural and anthropic environmental stress. In this study, we examine the effects of anthracene as a common pollutant on the total microbial communities from a Pinus halepensis litter of a typical Mediterranean coastal site (Les Calanques, Marseille). The main objective was to identify the microbial factors leading the resilience of this ecosystem. Two questions were addressed: (i) how lignin-degrading enzymes (Laccase, Lignin-peroxidase and Mn-peroxidase) are affected by the presence of this molecule, (ii) whether the indigenous consortia are involved in its degradation in mesocosms under favorable incubation conditions (25 °C, 60% WHC) and after different time intervals (1 and 3 month(s)). We found a strong increase in laccase production in the presence of anthracene after 3 months, together with anthracene degradation (28% ± 5). Moreover 9,10-anthraquinone is detected as the product of anthracene oxidation after 3 months. However neither lignin-peroxidase activity nor Mn-peroxidase activity is detected. Laccase proteins directly extracted from litter were sequenced via Nano-LC-MS/MS and reveal twelve different peptide sequences induced by the presence of anthracene in the mesocoms. Our study confirms the major detoxification role of this enzymatic system and highlights the high degradation potential of fungal species inhabiting P. halepensis litter, a factor in the resilience of Mediterranean ecosystems.  相似文献

A single tertiary isomer which is believed to be one of the major branched isomers of the isomeric nonylphenol was synthesized for use in investigations on its metabolism and estrogenicity in aquatic organisms. The physico-chemical properties of the isomer were determined to enable the prediction of its behaviour in aquatic environments. From laboratory investigations on its dissipation and distribution in lake water, which are reported in this paper, it was found that it had a half-life of dissipation of 38.1 days and 20.1 days in an open lake water and in an open lake water/ sediment system, respectively, and to be rapidly partitioned in to sediment giving a high concentration factor of 1.76 after 28 days with an initial dose concentration of 2.52 ppm. The main dissipation route was found to occur through volatilization and co-distillation. The isomer was, however, found to be resistant to biodegradation in both the lake water and sediment, showing only a slight 9% loss (after 56 days) and 4.2% loss (after 28 days), of the 14C-residues in lake water and lake water/sediment systems, respectively, by microbial activity. Transformation to other more polar metabolites possibly by hydroxylation was also found to be minimal in both lake water and sediment samples after 14 days by HPLC analysis. After 7 days, only 2.25 and 7.4% transformation to a more polar metabolite was detected in lake water and sediment samples, respectively.  相似文献

Effects of trenbolone (TBOH), a hormone used in cattle production, on the structure and function of microbial communities in a fresh water sediment from a lake in Southern Germany were studied in a microcosm experiment. The microbial community structure and the total gene pool of the sediment, assessed by 16S rRNA/rDNA and RAPD fingerprint analysis, respectively, were not significantly affected by TBOH. In contrast, the N-acetyl-glucosaminidase activity was almost 50% lower in TBOH treated samples (P<0.05). Also, the substrate utilization potential, measured using the BIOLOG system, was reduced after TBOH treatment. Interestingly, this potential did not recover at the end of the experiment, i.e. 19 days after the addition of the chemical. Repeated application of TBOH did not lead to an additional reduction in the substrate utilization potential. Overall results indicate that microbial community function was more sensitive to TBOH treatment than the community structure and the total gene pool.  相似文献

采用富集-分离-筛选的方法,从土壤中筛选得到3株具有较高絮凝活性的菌种,将其两两混合培养后,发现其对高岭土悬液的絮凝率均有较明显的提高,并且具有更好的热稳定性。实验结果表明,其最佳絮凝条件为:在pH=9时,微生物絮凝剂的最佳投量为2.0 mL,以2.0 mL 2.0%的CaCl2为助凝剂,对100 mL浓度为1 g/L的高岭土悬液的絮凝率可以达到90%以上;在与无机絮凝剂(10%PAC)和有机絮凝剂(0.1%PAM)的对比中,表现出了更高的絮凝活性和更好的絮凝效率。  相似文献

The impact of pesticides, namely thiobencarb (TBC), molinate (MOL) and chlorpyrifos (CPF), on soil microbial processes was studied in two Australian soils. Substrate induced respiration (SIR), substrate induced nitrification (SIN) and phosphatases and chitinase enzymatic activities were assessed during a 30-day microcosm study. The pesticides were applied to soils at recommended rates either alone, or as binary mixtures with TBC. Soil samples were sampled at 5, 15 and 30 days after pesticide treatments. Substrate induced respiration was only transiently affected by pesticides in both soils. In contrast, the process of indigenous nitrification was affected by the presence of pesticides in both soils, especially when the pesticides were applied as binary mixtures. Substrate induced nitrification increased with pesticides in the Griffith soil (except with MOL+TBC after 5 days) whereas SIN values were non-significantly different to the control on the Coleambally soil. The binary mixtures of pesticides with TBC resulted in a decrease in SIN in both soils, but the effects disappeared within 30 days. The enzymatic activities were not consistently affected by pesticides, and varied with the soil and pesticides studied. This study showed that, when applied at recommended application rates, TBC, MOL, and CPF (individually or as binary mixtures), had little or only transitory effects on the functional endpoints studied. However, further investigations are needed to assess the effect on microbial densities and community structure despite the low disturbance to the functions noted in this work.  相似文献

Effect of concentration, moisture and soil type on dissipation of flufenacet from soil has been studied under laboratory condition. The treated soil samples (1 and 10 microg/g levels) were incubated at 25+/-1 degrees C. The effect of moisture was studied by maintaining the treated soil samples (10 microg/g level) at field capacity and submerged condition. In general, flufenacet persisted for 60-90 days at lower and beyond 90 days at high rate. The dissipation of flufenacet from soil followed first order kinetics with half-life (DT50) values ranging from 10 to 31 days. The dissipation of flufenacet was faster at low rate than high rate of application. The slow dissipation at high rate could be attributed to inhibition of microbial activity at high rate. There was little overall difference in rate of dissipation in Ranchi and Nagpur soil maintained at field capacity and submerged condition moisture regimes. In Delhi soil net dissipation was faster under field capacity moisture than submerged condition. Soil types greatly influenced the dissipation of flufenacet. Dissipation was fastest in Delhi soil (DT50 10.1-22.3 days) followed by Ranchi soil (DT50 10.5-24.1 days) and least in Nagpur soil (DT50 29.2-31.0 days). The difference in dissipation could be attributed to the magnitude of adsorption and desorption of flufenacet in these soils.  相似文献

To investigate the effects of moist olive husks (MOH-residues) on soil respiration, microbial biomass, and enzymatic (o-diphenoloxidase, beta-glucosidase, dehydrogenase and alkaline phosphatase) activities, a silty clay soil was incubated with 0 (control), 8 x 10(3) (D), 16 x 10(3) (2D) and 80 x 10(3) (10D) kg ha-1 of MOH-residues on a dry weight basis. Soil respiration and microbial biomass data indicated that the addition of MOH-residues strongly increased microbial activity proportionally to the amounts added. Data of qCO2 suggested that the respiration to biomass ratio of the microbial population was strongly modified by MOH-residues additions during the first 90 days of incubation. The qCO2 data suggested a low efficiency in energy yields from C oxidation during the first 2 months of soil incubation. qFDA seemed to be relatively unaffected for treatments D and 2D as compared to the control, but was significantly lowered by the application of 10D, showing the lowest hydrolytic activity of microbial biomass in this treatment up to 360 days of incubation. o-Diphenoloxidase activity was delayed, and this delay was extended with the addition of larger quantities of MOH-residues. Alkaline phosphatase, beta-glucosidase and dehydrogenase activities were in line with the findings on microbial biomass changes and activities. The biological and biochemical data suggest that the addition of a large quantity of MOH-residues (80 x 10(3) kg ha-1) strongly modifies the soil characteristics affecting the r- and K-strategist populations, and that these changes last for at least the 360 days of incubation. The data also suggest that application rates exceeding 16 x 10(3) kg ha-1 are not recommended until the agro-chemical and -physical functions of the soil are further studied.  相似文献
