Human perfluorooctanesulfonate (PFOS) body burdens are attributable to both direct PFOS and indirect PFOS precursor (PreFOS) exposure. The relative importance of these two pathways has been estimated, but the relative temporal trajectory of exposure to PFOS and PreFOS has not been examined. Here, two hypothesized biomarkers of PreFOS exposure, PFOS isomer profiles (quantified as percent branched PFOS, %br-PFOS) and chiral 1m-PFOS enantiomer fractions (1m-PFOS EF) were analyzed in archived human serum samples of individual American adults (1974–2010) and pooled samples of Swedish primiparous women (1996–2010). After correcting for potential confounders, significant correlations between %br-PFOS and 1m-PFOS EFs were observed in American samples and in Swedish samples for the 1996–2000 period, supporting the hypothesis that both %br-PFOS and 1m-PFOS EF are biomarkers of PreFOS exposure. Significant trends of increasing %br-PFOS, from 2000 to 2010, and increasingly non-racemic 1m-PFOS EFs, from 1996 to 2000, were detected in Swedish samples. No statistically significant trend for %br-PFOS or 1m-PFOS EF was observed in American samples, but American males had significantly higher %br-PFOS and significantly lower 1m-PFOS EF (i.e. more non-racemic) than females, and a similar significant difference was shown in the older age group, relative to the younger age group. These temporal trends in %br-PFOS and 1m-PFOS EF are not easily explained and the results highlight uncertainties about how humans are exposed to PFOS.  相似文献

Perfluoroalkyl substances (PFASs) have been well studied in human daily intake for assessment of potential health risks. However, little is known about the isomeric compositions of PFASs in daily intake and their impacts on isomeric profiles in humans. In this study, we investigated the occurrence of PFASs with isomeric analysis in various human exposure matrices including foodstuffs, tap water and indoor dust. Perfluorooctanesulfonate (PFOS) and/or perfluorooctanoate (PFOA) were predominant in these exposure matrices collected in Tianjin, China. In fish and meat, linear (n-) PFOA was enriched with a percentage of 92.2% and 99.6%, respectively. Although n-PFOS was higher in fish (84.8%) than in technical PFOS (ca. 70%), it was much lower in meat (63.1%) and vegetables (58.5%). Dietary intake contributed > 99% of the estimated daily intake (EDI) for the general population. The isomeric profiles of PFOA and PFOS in human serum were predicted based on the EDI and a one-compartment, first-order pharmacokinetic model. The isomeric percentage of n-PFOA in the EDI (98.6%) was similar to that in human serum (predicted: 98.2%, previously measured: 99.7%) of Tianjin residents. The results suggest direct PFOA intake plays an important role in its isomeric compositions in humans. For PFOS, the predicted n-PFOS (69.3%) was much higher than the previously measured values (59.2%) in human serum. This implies that other factors, such as indirect exposure to PFOS precursors and multiple excretion pathways, may contribute to the lower percentage of n-PFOS in humans than of technical PFOS.  相似文献

Contributions of direct and indirect (via precursors) pathways of human exposure to perfluorooctane sulfonic acid (PFOS) isomers and perfluoroalkyl carboxylic acids (PFCAs) are estimated using a Scenario-Based Risk Assessment (SceBRA) modelling approach. Monitoring data published since 2008 (including samples from 2007) are used. The estimated daily exposures (resulting from both direct and precursor intake) for the general adult population are highest for PFOS and perfluorooctanoic acid (PFOA), followed by perfluorohexanoic acid (PFHxA) and perfluorodecanoic acid (PFDA), while lower daily exposures are estimated for perfluorobutanoic acid (PFBA) and perfluorododecanoic acid (PFDoDA). The precursor contributions to the individual perfluoroalkyl acid (PFAA) daily exposures are estimated to be 11–33% for PFOS, 0.1–2.5% for PFBA, 3.7–34% for PFHxA, 13–64% for PFOA, 5.2–66% for PFDA, and 0.7–25% for PFDoDA (ranges represent estimated precursor contributions in a low- and high-exposure scenario). For PFOS, direct intake via diet is the major exposure pathway regardless of exposure scenario. For PFCAs, the dominant exposure pathway is dependent on perfluoroalkyl chain length and exposure scenario. Modelled PFOS and PFOA concentrations in human serum using the estimated intakes from an intermediate-exposure scenario are in agreement with measured concentrations in different populations. The isomer pattern of PFOS resulting from total intakes (direct and via precursors) is estimated to be enriched with linear PFOS (84%) relative to technical PFOS (70% linear). This finding appears to be contradictory to the observed enrichment of branched PFOS isomers in recent human serum monitoring studies and suggests that either external exposure is not fully understood (e.g. there are unknown precursors, missing or poorly quantified exposure pathways) and/or that there is an incomplete understanding of the isomer-specific human pharmacokinetic processes of PFOS, its precursors and intermediates.  相似文献

Tetrabromobisphenol-A (TBBP-A), hexabromocyclododecane (HBCD) and its degradation products were determined in 34 human milk samples from Birmingham, UK. TBBP-A was detected in 36% of samples (average=0.06 ng g(-1) lw), with HBCDs detected in all samples (average ΣHBCDs=5.95 ng g(-1) lw). α-HBCD comprised 62-95% ΣHBCDs while β- and γ-HBCD constituted 2-18% and 3-33% respectively. Enantioselective enrichment of (-)-α-HBCD (average enantiomer fraction=0.29) was observed indicating potential enantioselectivity associated with HBCD absorption, metabolism and/or excretion. The degradation products pentabromocyclododecenes (average=0.04 ng g(-1) lw; n=9) and tetrabromocyclododecadienes (average=0.15 ng g(-1) lw; n=25) were detected for the first time in human tissues. Average exposures of a nursing infant to ΣHBCDs and TBBP-A (35 and 1 ng kg(-1) bw day(-1) respectively) via breast milk exceeded upper-bound dietary intakes of both UK adults and toddlers. Using a simple pharmacokinetic model, intakes of UK adults via inhalation, diet and dust ingestion were converted to predicted body burdens. Predictions compared well with those observed for HBCDs but observed body burdens of TBBP-A exceeded predictions. This may indicate the human half-life of TBBP-A is greater than observed previously, that intakes may be underestimated, or that concentrations reported here reflect recent elevated episodic exposure.  相似文献

Exposure to aqueous film forming foam (AFFF) was evaluated in 149 firefighters working at AFFF training facilities in Australia by analysis of PFOS and related compounds in serum. A questionnaire was designed to capture information about basic demographic factors, lifestyle factors and potential occupational exposure (such as work history and self-reported skin contact with foam). The results showed that a number of factors were associated with PFAA serum concentrations. Blood donation was found to be linked to low PFAA levels, and the concentrations of PFOS and PFHxS were found to be positively associated with years of jobs with AFFF contact. The highest levels of PFOS and PFHxS were one order of magnitude higher compared to the general population in Australia and Canada. Study participants who had worked ten years or less had levels of PFOS that were similar to or only slightly above those of the general population. This coincides with the phase out of 3M AFFF from all training facilities in 2003, and suggests that the exposures to PFOS and PFHxS in AFFF have declined in recent years. Self-reporting of skin contact and frequency of contact were used as an index of exposure. Using this index, there was no relationship between PFOS levels and skin exposure. This index of exposure is limited as it relies on self-report and it only considers skin exposure to AFFF, and does not capture other routes of potential exposure. Possible associations between serum PFAA concentrations and five biochemical outcomes were assessed. The outcomes were serum cholesterol, triglycerides, high-density lipoproteins, low density lipoproteins, and uric acid. No statistical associations between any of these endpoints and serum PFAA concentrations were observed.  相似文献

Daily dietary intake of perfluorinated chemicals (PFCs) in relation to serum levels was assessed by determination of nine PFCs including perfluorooctane sulfonate (PFOS) and perfluorooctanoic acid (PFOA) in matched daily diet duplicates and serum samples. Diet and serum were collected in year 2004 from 20 women in Osaka and Miyagi, Japan. Only PFOS and PFOA were detected in the diet samples and no significant difference between cities was seen. After adjusted by water content, diet concentration of PFOA was significantly higher in Osaka. The median daily intake calculated using the measured diet concentrations was 1.47 ng PFOS/kg b.w. and 1.28 ng PFOA/kg b.w. for Osaka, and 1.08 ng PFOS/kg b.w. and 0.72 ng PFOA/kg b.w. for Miyagi. A significant difference between cities was seen for the serum concentrations with median of 31 ng/mL PFOS and PFOA in Osaka, compared to 14 ng/mL PFOS and 4.6 ng/mL PFOA in Miyagi. Carboxylates such as perfluorononanoic acid (PFNA) and perfluoroundecanoic acid (PFUnDA) were also detected in serum at median levels 6.9 ng/mL and 3.2 ng/mL (Osaka), and 2.8 ng/mL and 5.1 ng/mL (Miyagi). Based on one-compartment model under steady state, dietary intake of PFOS and PFOA accounted for only 22.4% and 23.7% of serum levels in Osaka females, and in contrast 92.5% and 110.6% in Miyagi females, respectively.  相似文献

The widespread presence of perfluorooctanesulfonate (PFOS), perfluorooctanoate (PFOA), and perfluorohexanesulfonate (PFHxS) in human general populations and their slow elimination profiles have led to renewed interest in understanding the potential human neonatal exposures of perfluoroalkyls (PFAs) from consumption of human milk. The objective of this study was to evaluate the concentrations of PFOS, PFHxS, and PFOA in pooled human milk samples obtained in Sweden between 1972 and 2008 (a period representing the most significant period of PFA production) and to see whether the time trend of these analytes parallels that indicated in human serum. Chemical analysis of PFOS, PFHxS, and PFOA was performed on pooled Swedish human milk samples from 1972 to 2008 after methodological refinements. The 20 samples which formed the 2007 pool were also analyzed individually to evaluate sample variations. Analyses were performed by HPLC-MS/MS. Due to the complexities of the human milk matrix and the requirement to accurately quantitate low pg/mL concentrations, meticulous attention must be paid to background contamination if accurate results are to be obtained. PFOS was the predominant analyte present in the pools and all three analytes showed statistically significant increasing trends from 1972 to 2000, with concentrations reaching a plateau in the 1990s. PFOA and PFOS showed statistically significant decreasing trends during 2001-2008. At the end of the study, in 2008, the measured concentrations of PFOS, PFHxS, and PFOA in pooled human milk were 75 pg/mL, 14 pg/mL, and 74 pg/mL, respectively. The temporal concentration trends of PFOS, PFHxS, and PFOA observed in human milk are parallel to those reported in the general population serum concentrations.  相似文献

Per- and polyfluoroalkyl substances (PFASs) are a class of compounds with unique chemical properties that have been shown useful in a wide variety of applications because they provide materials with reduced surface tension and exceptional non-stick properties. PFASs are commonly found in impregnation materials, coatings of papers and textiles, fire-fighting foams, pesticides, and cleaning agents. The potential for human exposure to PFASs is high because of their widespread distribution. The aim of this study was to investigate levels of PFASs in men and women from Sweden and to assess the influence of gender and parity among women. Levels of 13 PFASs were determined in plasma samples collected during 2001–2004 from 1016 (507 women) 70 year-old participants from the population-based Prospective Study of the Vasculature in Uppsala Seniors (PIVUS). The PFASs studied were nine perfluorinated carboxylic acids (PFCAs), four perfluorinated sulfonic acids (PFSAs) and perfluorooctane sulfonamide (PFOSA). In addition, structural isomers of perfluorooctane sulfonic acid (PFOS) were determined in a subset of 398 individuals. The detection rates were high and the majority of the studied compounds were detected in more than 75% of the participants. Levels of the selected analytes were found to be similar to other studies of non-occupationally exposed populations. Gender differences were observed in levels of PFHpA which was higher in men, while PFHxS was higher in women. Parity among women was shown to have a minor effect on PFAS concentrations and we found primi- and multiparous women to have slightly lower levels of PFUnDA when compared to nulliparous women.  相似文献

We aimed to estimate the levels and time trends of perfluorinated alkyl acids (PFAAs) in serum of 1533 Danish pregnant nulliparous women between 2008 and 2013. The selection criterion of only including nulliparous women was chosen to avoid confounding from parity. The serum samples were analyzed for sixteen PFAAs using solid phase extraction and liquid chromatography tandem mass spectrometry (LC-MS/MS). We investigated the time trends for seven PFAAs, which were detected in more than 50% of the samples: perfluorohexane sulfonate (PFHxS), perfluoroheptane sulfonate (PFHpS), perfluorooctane sulfonate (PFOS), perfluorooctanoic acid (PFOA), perfluorononanoic acid (PFNA), perfluorodecanoic acid (PFDA), and perfluoroundecanoic acid (PFUnA). We found that the serum levels of all seven PFAAs decreased during the period from 2008 to 2013; on average PFHxS decreased with 7.0% per year, PFHpS with 14.8%, PFOS with 9.3%, PFOA with 9.1%, PFNA with 6.2%, PFDA with 6.3%, and PFUnA with 7.1% per year. Adjustment for maternal age, body mass index (BMI), educational level and gestational age at blood sampling did not change the time trends much. To our knowledge, we are the first to report decreasing trends of PFNA, PFDA and PFUnA since year 2000, thereby indicating that the phase-out of these compounds are beginning to show an effect on human serum levels.  相似文献

Perfluorooctane sulfonate (PFOS) and perfluorooctanoic acid (PFOA) blood levels are commonly used as biomarkers of human environmental exposure to these compounds. Many biomonitoring studies indicate 100% detection for PFOS and PFOA thus justifying a concern of possible risk for the most exposed individuals.This study addresses the predictive value of hazard quotients (HQs) calculated on the basis of serum PFOS and PFOA in male and female populations of reproductive age in Greenland, Poland and Ukraine.Overall, 2026 results of PFOS and PFOA serum concentrations (589 males, 1437 females) were obtained from the INUENDO database. HQs were calculated from the actual biomonitoring results and literature-based animal data linking toxicological outcomes and critical PFOS/PFOA serum levels. HQs for serum PFOS were calculated based on Points of Departure (PoD) at 13 μg mL 1 (cynomolgus monkeys, 183 days, changes in THS and T3) and for PFOA at 7.1 μg mL 1 serum (male rats, 90 days, hepatocellular necrosis, increased liver weight). Uncertainty factors were applied to reflect interspecies differences and human variability. Serum HQs were expressed as a ratio relative to the point of departure for each PFOS and PFOA. Only in the three cases of males in Greenland were there serum PFOS levels showing HQ values exceeding 1, so indicating that such serum levels may be of concern. The mean serum concentration of PFOS was significantly higher in male than in female populations. Despite significant differences between HQ profiles for PFOS and PFOA in donors from Greenland, Poland and Ukraine, the concentrations of these perfluoroalkylated compounds do not indicate a cause for concern, except for the three aforementioned cases from Greenland.This study demonstrates that the HQ approach can help to interpret human biomonitoring data and thus serve as a valuable tool in further risk assessment priority settings and may also be used as a basis for taking decisions in risk management.  相似文献

Human serum samples (n=61) were collected in Attika, Greece between June and October 2007 and analyzed for polybrominated diphenyl ethers (PBDEs), hexabromocyclododecane (HBCD), polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs). Thirty samples were collected from computer clerks of a large computer company in Athens working full-time with computers, and thirty-one from a control population in the Attika region with no computer use. Σ(5)PBDE concentrations (sum of tri- to hexa-BDEs) in all samples (n=61) ranged from 0.68 to 13.3 ng g(-1) lipid, with a median of 1.07 ng g(-1) lipid. These concentrations are on the lower end of those reported from other countries, probably reflecting lower usage of PBDE-containing products or lower exposures to these chemicals. Individual and Σ(5)PBDE concentrations did not statistically differ between the two groups of computer clerks and non-computer users (p>0.05), with the exception of BDE 153 (p=0.033). The predominant congener was BDE 153, followed by BDEs 47, 100, 99, 183, 154 and 28. HBCD was also detected in 70% of the samples. BDE 209 was detected in 8 out 61 samples (13%), with concentrations ranging from 1.18 to 19.1 ng g(-1) lipid, and a median of 2.94 ng g(-1) lipid. No age dependency was found for PBDEs. Σ(11)PCB (sum of PCBs 74, 99, 118, 138, 146, 153, 156, 170, 180, 183 and 187,) in all samples (n=61) ranged from 36 to 402 ng g(-1) lipid, with a median of 110 ng g(-1) lipid. pp'-DDE concentrations ranged from 53.8 to 1649 ng g(-1) lipid, with a median of 268 ng g(-1) lipid. This is the first study to report levels of PBDEs in a possibly occupationally exposed subset of the Greek population.  相似文献

Perfluorinated compounds (PFCs) are worldwide present in the environment and the general population. Animal studies have shown developmental toxicity of these compounds. To investigate the PFCs exposure of neonates from mother during gestation and lactation, we analyzed twelve PFCs in matched maternal serum, cord serum and breast milk samples collected from 50 pairs of women and their newborns between June and July 2009 in Jinhu, China. Eight PFCs were detected in serum samples, and five of them were also detectable in breast milk. A significant intercorrelation between PFCs concentrations in matched maternal serum, cord serum and breast milk was observed (p<0.01, r=0.435-0.911). The median partition ratio was from 0.39:1 (PFDA) to 1.74:1 (PFTrDA) for seven PFCs through the placenta, and was from 0.02:1 (PFOS) to 0.09:1 (PFOA) for five PFCs through the lactation. A high transport efficiency of PFOA both through placental barrier and lactation was observed. The postnatal exposure of PFCs through lactation was higher compared to prenatal exposure, especially for PFOA.  相似文献

Perfluorooctane sulfonate (PFOS) found extensive use for over 60 years up until its restriction in the early 2000s, culminating in its listing under the Stockholm Convention on Persistent Organic Pollutants (POPs) in 2009. Efforts to minimise human body burdens are hindered by uncertainty over their precise origins. While diet appears the principal source for the majority of western populations (with other pathways like dust ingestion, drinking water and inhalation also important contributors); the role played by exposure to PFOS-precursor compounds followed by in vivo metabolism to PFOS as the ultimate highly stable end-product is unclear. Such PFOS-precursor compounds include perfluorooctane sulfonamide derivates, e.g., perfluorooctane sulfonamides (FOSAs) and sulfonamidoethanols (FOSEs). Understanding the indirect contribution of such precursors to human body burdens of PFOS is important as a significant contribution from this pathway would render the margin of safety between the current exposure limits and estimates of external exposure to PFOS alone, narrower than hitherto appreciated. Estimates derived from mathematical modelling studies, put the contribution of so-called “precursor exposure” at between 10% and 40% of total PFOS body burdens. However, there are substantial uncertainties associated with such approaches. This paper reviews current understanding of human exposure to PFOS, with particular reference to recent research highlighting the potential of environmental forensics approaches based on the relative abundance and chiral signatures of branched chain PFOS isomers to provide definitive insights into the role played by “precursor exposure”.  相似文献

Human serum samples (n=142) from Iassy county (Eastern Romania) collected in 2005 were analyzed for p,p'-DDT and its metabolites, hexachlorobenzene, hexachlorocyclohexane (HCH) isomers, chlordane and metabolites and 14 polychlorinated biphenyl (PCB) congeners. In all samples, p,p'-DDE (principal metabolite of p,p'-DDT) and beta-HCH (most persistent HCH isomer) were the most abundant organochlorine pesticides (OCPs) with median concentrations of 1975 and 923 ng/g lipid weight (lw), respectively. Hexachlorobenzene and p,p'-DDT were also detected in all samples, but at a lower median concentration of 30 and 340 ng/g lw, respectively. The large variation of the ratio p,p'-DDT/p,p'-DDE (range between 0.02 and 0.80) suggests various degrees of exposure to DDT, including recent exposure, at least to some subgroups of the population. Chlordane and its metabolites were in most cases close to the limit of quantification suggesting a very low use of chlordane formulations in Eastern Romania. Unexpectedly, PCBs were present in all samples at sometimes considerable levels (up to 4970 ng/g lw for sum of 14 PCB congeners) indicating a higher exposure of the Romanian population than previously reported. The PCB profile consisted of persistent congeners such as 138, 153, 170 and 180 which contributed for approximately 75% to the sum PCBs. Concentrations of most pollutants correlated significantly with age (r>0.86, p<0.01). Except for p,p'-DDT and gamma-HCH, the mean levels of OCPs in females were statistically higher than in males, while, except for octa-CBs, no gender differences were found for PCBs. Levels of p,p'-DDE, penta-CBs, hexa-CBs and hepta-CBs were significantly higher in individuals with a rural main residence. In a pooled serum sample, 6 polybrominated diphenyl ether (PBDE) congeners were measured at a level of 1.04 ng/g lw. Compared to results available from Central and Eastern European countries, human serum samples from Romania contained higher levels of contamination. This emphasizes that an extensive and rigorous program for the monitoring of OCPs and PCBs in Romanian population is highly needed in the light of possible adverse health effects acknowledged for these pollutants.  相似文献

Releases of aqueous film-forming foams (AFFFs) from airport firefighting activities have been identified as important local point sources of per- and polyfluoroalkyl substances (PFASs) in nearby waterways. PFASs can be taken up by fish, and in turn by the humans that consume them. Despite the global extent of AFFF emissions, few studies exist on related impacts on humans. We aimed to investigate the associations between the consumption of fish from AFFF-affected waters and serum PFAS concentrations in humans using a combination of statistical tools, empirical data, and toxicokinetic modeling. Participants of the SAMINOR 2 Clinical Study were the basis for this study sample, which comprised 74 persons. Fifty-nine participants who reported consuming fish from AFFF-affected waters and 15 nonconsumers completed a questionnaire and gave serum samples. Participants were classified based on their consumption of trout and char: high (n = 16), moderate (n = 16), low (n = 27), and nonconsumers (n = 15); and serum samples were tested for the presence of 15 PFASs. Perfluorooctane sulfonic acid (PFOS) was found in all participants, with the highest concentrations detected in the high consumption group (geometric means, 28 ng/mL) compared to the low consumption group and nonconsumers (10 and 11 ng/mL, respectively). In an analysis of variance contrast model, a significant, positive increasing trend was seen for fish consumption and PFOS, perfluorohexane sulfonic acid (PFHxS), and perfluorononanoic acid (PFNA). Toxicokinetic modeling allowed us to predict the median increases in serum concentrations of PFOS, PFHxS, and PFNA among high consumers within a factor of 2.2. The combination of statistical evaluation and toxicokinetic modeling clearly demonstrated a positive relationship between consumption of fish from AFFF-affected waters and serum PFAS concentrations. Further studies on dietary exposure to other PFASs present in AFFF and its consequences on human health are warranted.  相似文献

Fluorinated organic compounds (FOCs) are a group of chemicals widely used as surfactants, lubricants, polymers, and fire-fighting foams. Recent studies have shown the ubiquitous distribution of FOCs in the environment, wildlife, and humans. We here report the results of a pilot study conducted to provide preliminary data on the levels of 13 FOCs in the blood of 48 residents in Catalonia, Spain, in relation to gender and age (25+/-5 and 55+/-5 years). The highest mean concentration was obtained for perfluorooctane sulfonate (PFOS, 7.64 ng/ml), followed by perfluorohexane sulfonate (PFHxS, 3.56 ng/ml) and perfluorooctanoic acid (PFOA, 1.80 ng/ml). Four other FOCs showed mean levels between 0.30 and 0.44 ng/ml, whereas those of the remaining 6 compounds were below the detection limit. Regarding gender, the blood levels of PFHxS and PFOA were significantly higher (p<0.05) in men than in women, while differences according to age were only noted for PFHxS (p<0.05) and perfluorooctane sulfonamide (PFOSA) (p<0.001), for which the levels were higher in the younger (25+/-5 years) group of subjects. A significant correlation between PFOS levels and those of the remaining detected FOCs (except PFDA) was found. In general terms, the current FOC concentrations were lower than those found in recent studies concerning levels of these chemicals in human blood and serum of subjects from different countries.  相似文献

We aimed to characterize levels of polyfluorinated compounds (PFCs) in indoor dust from offices, homes, and vehicles; to investigate factors that may affect PFC levels in dust; and to examine the associations between PFCs in dust and office workers' serum. Dust samples were collected in 2009 from offices, homes, and vehicles of 31 individuals in Boston, MA and analyzed for nineteen PFCs, including perfluorooctanoate (PFOA), perfluorooctane sulfonate (PFOS), fluorotelomer alcohols (FTOHs), and sulfonamidoethanols (FOSEs). Serum was collected from each participant and analyzed for eight PFCs including PFOA and PFOS. Perfluorononanoate, PFOA, perfluoroheptanoate, perfluorohexanoate, PFOS and 8:2 FTOH had detection frequencies > 50% in dust from all three microenvironments. The highest geometric mean concentration in office dust was for 8:2 FTOH (309 ng/g), while PFOS was highest in homes (26.9 ng/g) and vehicles (15.8 ng/g). Overall, offices had the highest PFC concentrations, particularly for longer-chain carboxylic acids and FTOHs. Perfluorobutyrate was prevalent in homes and vehicles, but not offices. PFOA serum concentrations were not associated with PFC dust levels after adjusting for PFC concentrations in office air. Dust concentrations of most PFCs are higher in offices than in homes and vehicles. However, indoor dust may not be a significant source of exposure to PFCs for office workers. This finding suggests that our previously published observation of an association between FTOH concentrations in office air and PFOA concentrations in office workers was not due to confounding by PFCs in dust.  相似文献

The levels of 1,1,1-trichloro-bis-2,2'-(4-chlorophenyl) ethane (p,p'-DDT) and its metabolites, hexachlorobenzene (HCB), hexachlorocyclohexane isomers (HCHs), chlordanes (trans- and cis-chlordane, oxychlordane and trans-nonachlor), 11 polychlorinated biphenyl (PCB) congeners, and 7 polybrominated diphenyl ether (PBDE) congeners were measured in 37 individual human milk samples from Kahramanmara? region, Turkey. Organochlorine pesticides were the major contaminants in the milk samples. p,p'-DDE and p,p'-DDT were detected in all samples, while beta-HCH had a detection frequency of 97%. The sum of the DDTs in human milk samples varied between 0.52 and 315.8 ng/g wet weight (ww) with a mean ratio between p,p'-DDE and p,p'-DDT equal to 31.1. p,p'-DDD could be measured only in six samples. beta-HCH was the most prevalent HCH isomer with a mean value of 2.08 ng/g ww. The mean concentration of gamma-HCH was 0.38 ng/g ww, while alpha-HCH was not detected in any sample. HCB is found in 95% of the milk samples with a mean concentration of 0.30 ng/g ww. The mean value for the sum of chlordanes was 0.39 ng/g ww, with oxychlordane and trans-nonachlor being the principal contributors. PCBs could be measured only in 8 out of 37 samples and their concentration ranged between <0.15 and 1.92 ng/g ww for the sum of PCBs. PCB profiles were dominated by congeners 153, 180 and 138. PBDEs were detected only in 3 out of 37 samples, with the highest value being 0.014 ng/g ww (0.40 ng/g lipid weight) and BDE 47 was the dominant congener. Although the number of samples is relatively low and they are not representative for the whole Turkish population, the results of the present study are important to provide additional data on the concentrations of persistent organochlorinated pollutants in Turkey and show as first the PBDE levels in Turkish population.  相似文献

Current understanding of the thyroid disruptive properties of perfluoroalkyl substances (PFASs), particularly in aging populations, is limited. The objectives of this study were to (i) assess associations between thyroid function, as measured by serum thyrotropin (thyroid stimulating hormone, TSH), free thyroxine (fT4), total thyroxine (T4), and total triiodothyronine (T3), and serum perfluorooctane sulfonate (PFOS) and perfluorooctanoic acid (PFOA) in an aging population and (ii) determine if other persistent organic pollutants with thyroid disruptive properties including polychlorinated biphenyls (PCBs) and polybrominated diphenyl ethers (PBDEs) modify such associations. We conducted a cross-sectional study of 87 men and women 55 to 74 years of age, without clinically-diagnosed thyroid disease, who resided in upper Hudson River communities in New York. Geometric means (standard deviations) of serum PFOS and PFOA were 31.6 (1.7) ng/mL and 9.17 (1.72) ng/mL, respectively. Multivariable linear regression analyses indicated that one interquartile range difference in PFOS corresponded to 4% and 9% increases in fT4 and T4 respectively. We detected statistical interactions between PFOA and age for effects on fT4 and T4; joint increases in PFOA and age were associated with increases in fT4 and T4, of 3% and 7%, respectively. We also detected statistical interactions between PFOS and total PCBs for the effect on T3 and between PFOA and total PBDEs for the effect on TSH. Our results suggest that PFASs are associated with subtle alterations in thyroid hormone levels in this population, and that these associations are likely to vary by age, and levels of PCBs and PBDEs.  相似文献

Perfluorooctane sulfonate (PFOS) is the stable end product of the degradation of various perfluorinated compounds and is the predominant compound found in the environment and biota. PFOS is a widespread environmental contaminant that is found in a great diversity of wildlife species with more elevated tissue concentrations in animals from populated and industrialized areas. In this study we determined the PFOS accumulation in blood and livers of a small songbird, the great tit (Parus major), in the vicinity of a large fluorochemical plant in Antwerp, Belgium. PFOS concentrations ranged from 553 ng/g to 11359 ng/g in liver and ranged from 24 to 1625 ng/ml in blood, which are among the highest ever reported in free-living animals, and exceeded in almost all birds the hepatic benchmark concentrations for the protection of avian species [Beach SA, Newsted JL, Coady K, Giesy JP. Ecotoxicological evaluation of perfluorooctanesulfonate (PFOS). Rev Environ Contam Toxicol 2006;186:133-174]. Although PFOS concentrations in liver and blood decreased significantly within approximately 5.5 km of the plant, differences were smaller than previously described for wood mice (Apodemus sylvaticus) and nestling great tits. PFOS concentrations in liver and blood were higher in young birds (one-year old). No significant sex differences were found. A highly significant correlation between liver and blood concentrations indicates the usefulness of blood as a non-destructive matrix for biomonitoring purposes.  相似文献
