Michaelis-Menten uptake kinetics were observed at all light intensities. With constant illumination, the Vmax and K1 in nitrate uptake over the natural light intensity range of 0 to 2000 E were 0.343 g-at NO3–N(g)-1 at protein-N h-1 and 26 E, respectively. Nitrate uptake was inhibited at higher light intensities. The Ks for nitrate uptake did not vary as a function of light intensity remaining relatively constant at 0.62 g-at NO3–N 1-1. With intermittent illumination, the Vmzx for light intensity in nitrate uptake over a light intensity range of 0 to 5000 E was 0.341 g-at NO3–N(g)-1-at protein-N h-1. No inhibition of nitrate uptake was observed at higher than natural light intensities. Chaetoceros curvisetus will probably never experience light inhibition of nitrate uptake under natural conditions.  相似文献

Batch cultures of the marine unicellular centric diatom Chaetoceros calcitrans (Paulsen) Takano were maintained by serial subculturing every 4 d into nutrient-enriched natural sea-water medium supplemented with 350, 950 and 1 400 g-at Si l-1. The diatom cultures removed initial silica concentrations of 350 and 950 g-at l-1 from the medium within 2 and 3 d, respectively. About 30 g-at l-1 of the highest initial concentration remained in the medium after 4 d. The mean final cell density with an enrichment of 350 g-at Si l-1 was 3.43±0.26×104 cells l-1 (median cell volume = 77.5±5.0 m3); with 950 g-at Si l-1, 8.55±0.55×104 cells l-1 (50.0±4.5 m3); and with 1 400 g-at Si l-1, 9.72±0.48×104 cells l-1 (37.3±5.0 m3). There was no significant difference in the final total organic weight of cells produced, which was in the range of 170 to 190 mg per 250 ml culture. This consisted of proportionately more lipid and carbohydrate and less protein from the treatment with 350 g-at Si l-1 than from the 1 400 g-at Si l-1 treatment.  相似文献

The uptake of cadmium by the shoots ofZostera marina L. (eelgrass) was examined in the laboratory. Experiments were carried out in experimental chambers which allowed the separation of the leaves from the root-rhizome portions of intact shoots. Cadmium uptake by the root-rhizome portions over 24 h was directly related to substrate cadmium concentration (1.0, 5.0 and 10.0 g Cd ml-1) and varied from 6.5 to 30.0 g Cd g-1. Cadmium uptake by the root-rhizomes and the leaves in a substrate concentration of 1 g Cd ml-1 was also related to exposure times (24, 48 and 72 h). Maximum uptake by the root-rhizomes and the leaves was observed after 72 h at a substrate concentration of 1 g Cd ml-1 and was equivalent to 48 and 94 g Cd g-1, respectively. Translocation of cadmium from the leaves to the root-rhizomes was observed after 24 h, and at the end of 72 h was equivalent to 27% of the total leaf uptake. No cadmium movement from the root-rhizomes to the leaves was detected.  相似文献

A study of phosphate assimilation by coastal marine plankton revealed that both phytoplankton and microheterotrophs incorporated radioactive phosphorus (33P). Size fractionation of the particulate matter (using 1 m pore diameter Nucleopore® membrane filters), antibiotic treatment (using garamycin), and independent estimaties of photoautotrophic (14CO2 uptake) and heterotrophic (3H-glucose uptake) activities were employed to separate phyto- and bacterioplankton phosphate uptake. Results indicated that phytoplankton 33P-uptake was best estimated by the fraction of particulate matter retained on the 1 m membrane filters. Usually, less than 10% of the phytoplankton (based on chlorophyll a measurements) passed the 1 m pore-diameter filters, whereas about 90% of the heterotrophic activity passed. At least 50% of the 33P-uptake was associated with the <1 m fraction. It may be possible to resolve the phytoplankton and bacterial contributions to 33P-uptake by comparing the percent of total 33P-uptake with the percent of total 3H-glucose uptake associated with the >1 m fraction.  相似文献

A unique data set from lead risk assessments performed on 67 public housing developments from across the United States was made available for analyzes. The data set includes results of lead analysis from 5906 dust wipes and from 1222 soil samples. A total of 487 dwelling units in these developments, as well as associated common areas, were sampled, all by the same team of inspectors. The number of dwelling units within a development that were sampled reflected the guidelines then in force, the 1990 Interim HUD Guidelines, rather than those specified in the 1995 Guidelines. Median dust lead loadings for floors, 151gm–2 (14gft–2), and window sills, 936gm–2 (87gft–2), were much less than former HUD limits of 1076gm–2 (100gft–2) and 5380gm–2 (500gft–2), respectively and are only about one-third of the recently established limits of 431gm–2 (40gft–2) and 2690gm–2 (250gft–2). In contrast, the median lead loading for window troughs, 8560gm–2 (795gft–2), was almost identical to the HUD clearance limit of 8610gm–2 (800gft–2). There was a strong positive correlation between floor and window trough lead loading values for samples from the same dwelling units and those from common areas of the housing developments. Door threshold samples, which may reflect conditions exterior to the dwelling unit, were collected from 53 dwelling units. Median lead loading levels of these samples were more than ten times higher than those in floor samples from the same dwelling units, were about the same as window sill samples and about one-half of levels in window trough samples. Composite sample results, simulated by averaging results from four samples within a dwelling unit, revealed that in order to have the same rate of excedence of standards, the composite standards would have to be reduced, for example, from the single sample value of 1076gm–2 (100gft–2) to 527gm–2 (49gft–2) for floor samples and from the single sample value of 8610gm–2 (800gft–2) to 5160gm–2 (479gft–2) for window troughs. For this public housing data set, the portion of the units in developments containing more than 225 units which exceeded the established limit for window samples was the same when using either the full data set or a random one-half of the data set. This suggests that, for this data set, the number of dwelling units sampled was excessive . Thus, the required increase in the number of dwelling units to be sampled specified in the 1995 Guidelines for developments with more than 225 dwelling units, may not have been necessary if this data set is representative of public housing developments in the United States.  相似文献

Analyses of individual content of carbon (C), nitrogen (N), and hydrogen (H) were carried out for all larval stages of Pagurus bernhardus and Carcinus maenas, and for newly metamorphosed crabs. Maximum range in total larval development is 12.8 to 165.8 g C, 3.2 to 35.1 g N, and 1.9 to 24.9 g H in P. bernhardus and 3.1 to 43.2 g C, 0.7 to 10.1 g N, and 0.4 to 6.3 g H in C. maenas. From these data energy equivalents were calculated. Maximum range in total larval life is 0.43 to 6.38 J ind. -1 in P. bernhardus and 0.1 to 1.49 J ind. -1 in C. maenas. There is a 32.4% mean loss of energy in P. bernhardus megalopa development; this seems to describe the normal developmental pattern in this stage. Biomass was determined as fresh and dry weight respectively. The shortterm (10–22 d) effect of Zn, Hg, Cu, Cd, Pb, and Ni on the length growth of Mytilus edulis is studied. Significant reductions of growth rate was found at 0.3 g Hgl-1, 3 g Cul-1, 10 g Znl-1, and 10 g Cdl-1 added to the local sea water, while concentrations of up to 200 gl-1 of Pb and Ni had no effect on the growth. With exposure to Cu and Zn, there was a linear reduction in growth rate with increasing metal concentration up to about 6 g Cul-1 and 100 g Znl-1. Above these levels, growth stopped with Cu, while with Zn it was stabilized at about 20% of control growth. When Hg and Cd were added, a curvilinear relationship between growth and metal concentration is indicated. With Hg, growth rate is nearly zero above 3–4 g Hgl-1, while the growth rate was 50% of control after 10 d of exposure to 100 g Cdl-1. At 2 g Cdl-1 there was a significant stimulation of length increase. Observed EC50-values for growth were 0.3–0.4 g Hgl-1, 3–4 g Cul-1, 60 g Znl-1, and 100 g Cdl-1.  相似文献

Orthophosphate uptake by a natural estuarine phytoplankton population was estimated using two methods: (1) 32P uptake experiments in which filters of different pore sizes were used to separate plankton size-fractions; (2) 33P autoradiography of phytoplankton cells. Results of the first method showed that plankton cells larger than 5 m were responsible for 2% of the total orthophosphate uptake rate. 98% of the total uptake rate occurred in plankton composed mostly of bacteria, which passed the 5 m screen and were retained by the 0.45 m pore-size filter. There was no orthophosphate absorption by particulates in a biologically inhibited control containing iodoacetic acid. Orthophosphate uptake rates of individual phytoplankton species were obtained using 33P autoradiography. The sum of these individual rates was very close to the estimated rate of uptake by particulates larger than 5 m in the 32P labelling experiment. Generally, smaller cells were found to have a faster uptake rate per m3 biomass than larger cells. Although the nannoplankton constituted only about 21% of the total algal biomass, the rate of phosphate uptake by the nannoplankton was 75% of the total phytoplankton uptake rate. Results of the plankton autoradiography showed that the phosphate uptake rate per unit biomass is a power function of the surface: volume ratio of a cell; the relationship is expressed by the equation Y=2x10-11 X 1.7, where Y is gP m-3 h-1 and X is the surface: volume ratio. These results lend support to the hypothesis that smaller cells have a competitive advantage by having faster nutrient uptake rates.  相似文献

The coralline alga Phymatolithon calcareum was dredged from 13 m in the Kattegatt, Baltic Sea, in December, 1980, and its rate of calcification was measured by 45Ca++-uptake methods. Light-saturated calcification rates at 5°C ranged from 15.8 g CaCO3 g-1 dry wt h-1 for the basal parts of the plants to 38.7 g CaCO3 g-1 dry wt h-1 for the tips. These age gradients were not apparent when calcification rates were expressed on the basis of surface area. Experiments with salinity (10, 20, 30) and temperature (0°, 5°, 10°, 20°C) indicated that optimum conditions for calcification were at 30 S and at temperatures above 10°C. Salinity had a greater influence on calcification rate than did temperature, and there was a positive relationship between salinity and calcification rate at all temperatures. In 6 mo old cultures, salinity was again the important factor, with all plants remaining healthy at 30 except those at the highest temperature (20°C). These trends, and the low calcification rates at 10S (4.6 g CaCO3 g-1 dry wt h-1 at 5°C to 8.6 g CaCO3g-1 dry wt h-1 at 20°C) suggest that low salinity may be the explanation for the general absence of P. calcareum from the brackish waters of the Baltic Sea. Short-term experiments in which salinity was kept constant while Ca++ concentration was altered, and experiments in which salinity was varied and Ca++ concentration kept constant, suggest that it is the calcium ion concentration and not salinity per se which affects calcification rates.  相似文献

Monthly variation in photosynthesis, dark respiration, chlorophyll a content and carbon: nitrogen (C:N) ratios in different lamina sections of adult plants of Ascoseira mirabilis Skottsberg from King George Island, Antarctica, was investigated between September 1993 and February 1994. Light saturated net photosynthesis (P max) showed maximum values in September (12 to 25 mol O2 g-1 fr wt h-1), and decreased towards the summer to values ranging between 2.0 and 5.0 mol O2 g-1. In the distal section, however, a second optimum occurred in December (25 mol O2 g-1 fr wt h-1). Dark respiration rates were also highest in October and November and decreased strongly in December to February (6.0 and 1.0 mol O2 g-1 fr wt h-1, respectively). Gross photosynthesis exhibited high values between September and December. Concomitant with the seasonal decrease of photosynthetic efficiency () from mean values of 1.2 mol O2 g-1 fr wt h-1 (mol photons cm-2 s-1)-1 in September to 0.3 mol O2 g-1 fr wt h-1 (mol photons cm-2 s-1)-1 in January, the initial light saturating point (I k) gradually increased from 19 to 60 mol photons m-2 s-1. Likewise C:N ratios were low in spring (12 to 13) and increased in summer (20). In general, the photosynthetic parameters P max, gross photosynthesis, and Chl a concentrations were significantly higher in the distal section of the thallus. In contrast, C:N ratios were lower in the distal section of the lamina. The results show that photosynthesis obviously strongly supports growth of the alga in late winter to spring, as it does in some morphologically related brown algae from temperate and polar regions. Mussels, Mytilus edulis planulatus, were collected from Port Phillip Bay, Victoria, Australia, in February 1984, to test the assumption that they are integrators of cadmium pollution. Groups of mussels were subjected to the same average dose of cadmium (21 g l-1), administered according to different dosing regimes over 4 wk (Regimes 1, 2, 3, 5); other groups received twice the average dose (42 g l-1) in half the time (Regime 4). During each regime, the mussels were exposed to the different cadmium concentrations for one week at each concentration. Nominally, the regimes were: (1) 36 g (Wk 1) to 26 g (Wk 2) to 16 g (Wk 3) to 6 g (Wk 4) Cd l-1; (2) 6 to 16 to 26 to 36 g Cd l-1; (3) 21 to 21 to 21 to 21 g Cd l-1; (4) 42 to 42 g Cd l-1; (5) 42 to 42 g Cd l-1 to background (<0.5 g) to background. Differences in cadmium accumulation by mussels from Regimes 1 and 4 were not statistically significant, nor were differences in accumulation between mussels from Regimes 2 and 3. However, mussels from Regimes 1 and 4 had accumulated significantly more cadmium than had mussels from Regimes 2 and 3. Accumulation by mussels from Regime 5 was not significantly different from that by mussels from any of the other regimes. These results suggest that, at least for cadmium, the assumption that mussels are integrators of pollution should be treated with caution. They also have implications with regard to the quantitative biological monitoring of pollution. For example, even in a carefully controlled monitoring program, using mussels of standard size and condition, significant differences in cadmium content between mussels need not indicate exposure to different levels of contamination. Rather, these differences could reflect differences in the regime by which the contamination was received.  相似文献

Adult Elasmopus rapax, collected from the eastern coast of Venezuela in 1990, were exposed to seawater containing various CdCl2 concentrations ranging from 0.25 to 5.5 mol l-1. The 48-h and 96-h LC50 values obtained were 4.0 and 1.6 mol Cd l-1, respectively. In amphipods exposed to 1 mol Cd l-1 for up to 240 h, the apparent rate of cadmium uptake was higher in dead animals (most of which had molted during the preceding 24 to 48 h) than in those which survived throughout the treatments without molting. Thus, whole-body cadmium content reached 1.74 mol g-1 dry weight (dw) in the former and only 0.85 mol g-1 dw in the latter; the higher body Cd-load may have caused the increased mortality observed in molters. On exposure to cadmium levels above 0.5 mol l-1 the oxygen consumption rate of non-molters decreased from 2.2 to about 1.5 ml O2 g-1 dw h-1 over the first 24 h, remaining unchanged thereafter. The results place E. rapax among the most sensitive marine organisms yet studied concerning cadmium toxicity, and emphasize the usefulness of the Amphipoda as bioindicators and research tools for bioassays.  相似文献

Captan did not affect the survival of Dungeness crab (Cancer magister Dana) zoea exposed to 30 g l-1 during a chronic toxicity test lasting 69 days, but larvae were quickly killed (mean survival time = 9 days) in the same test by exposure to 450 g l-1 of the fungicide. Delay of molting occurred, however, for later stages at 30 g l-1. Survival of juvenile crabs was not reduced by exposure to captan for 36 days at 510 g l-1 or, in a second test, for 80 days at 290 g l-1. No deaths of adults exposed for 75 days to 340 g l-1 of captan were observed. Captan appeared to accelerate hatching of eggs at all concentrations tested from 100 to 10,000 g l-1. The development from prezoeae during a 24-h period was not inhibited by the fungicide, but at 3,300 and 10,00 g l-1, the two highest concentrations tested, developing zoeae exhibited a morphological deformity and were largely inactive. Under the prevailing conditions in the toxicity tests, the half-life of captan was estimated to be from 23 to 54 h. Because of the relatively low toxicity of captan to crab stages and its high rate of degradation in sewater, it is suggested that the agricultural application of captan near marine waters is not likely to affect natural crab populations or crabs in laboratory culture. Macrofauna living on subtidal rocks reefs in southern California excrete ammonium, a potentially important nutrient for benthic algae. Ammonium excretion rates of eleven macroinvertebrate and five fish taxa were determined from a total of 324 in situ incubations conducted between October 1984 and August 1985 at 14 to 17 m depths off Santa Catalina Island, California. Total ammonium excretion ranged from over 100 mol h-1 by the kelp bass Paralabrax clathratus to less than 0.1 mol h-1 by the gastropod Conus californicus. Weight-specific ammonium excretion generally ranged from 0.5 to 4 mol g-1 h-1 in invertebrates and from 3 to 7 mol g-1 h-1 in fishes. Intraspecific excretion rates varied substantially. Coefficient of variation of excretion rates were higher than reported for laboratory studies and multiple regression indicated that 50 to 90% of the variation in ammonium excretion rates of five species studied in detail could not be explained by the combined variation in dry weight, water temperature, time of day, and incubation dates. The excretion data, along with estimates of population densities and size-frequency distributions, indicate that benthic macrofauna release a total of 25 to 30 mol NH 4 + m-2 h-1 both day and night. The species that generally make the largest contributions are a gobiid fish (Lythrypnus dalli), followed by three gastropods (Astraea undosa, Tegula eiseni, and T. aureotincta) and a sea urchin (Centrostephanus coronatus). The amount of ammonium excreted by these macrofauna on rocky reefs is insignificant compared to our previously published data on the nighttime excretion of blacksmith (Chromis punctipinnis), a pomacentrid fish that feeds in the water column during the day and shelters on the reef at night. Including blacksmiths, we estimate that the amount released by rocky-reef macrofauna at night is >280 mol m-2 h-1, a rate that is similar to that for many other marine communities. Aquaria containing clean sand received a continuous supply of seawater from Santa Rosa Sound, Florida, USA, mixed with known concentrations (7, 76 and 622 g l-1) of pentachlorophenol (PCP). After 9 weeks, nematodes accounted for 87% of the total meiofauna. Nematode biomass and densities were greatest in aquaria exposed to 76 g PCP l-1 and were least in aquaria exposed to 622 g PCP l-1. Epistrate feeders were abundant in control aquaria and aquaria exposed to 7 and 76 g PCP l-1, but not in aquaria exposed to 622 g PCP l-1. Selective deposit feeders were not abundant in the control aquaria and aquaria exposed to 7 g PCP l-1, but comprised 19% of the nematodes in aquaria exposed to 76 g PCP l-1 and 61% in aquaria exposed to 622 g PCP l-1.  相似文献

In order to assess the intake of lead and cadmium by consumers of home grown vegetables in urban areas, replicated experimental plots of uniform size, comprising summer and winter crops, were established in 94 gardens and allotments in nine towns and cities in England.The geometric mean lead and cadmium concentrations for the soils (n = 94) were 217 g g–1 (ranging from 27 to 1,676 g g–1) and 0.53 g g–1 (<0.2–5.9 g g–1), respectively. Compared with agricultural soils, the garden and allotment soils contained elevated levels of lead but not cadmium.Lead concentrations in the vegetables ranged from <0.25 g g–1 to 16.7 g g–1 dry weight and cadmium concentrations ranged from <0.025 g g–1 to 10.4 g g–1 dry weight. Lead concentrations were higher than reported background levels, although <1% exceeded the statutory limit for saleable food in the UK (1 g g–1 fresh weight). Experiments were conducted to develop a sensitive sublethal toxicity test protocol to determine the toxicity of municipal wastewater effluents to larvae of the red abalone Haliotis rufescens. In multiple tests, fertilized abalone embryos were exposed for 48 h to dilutions of a reference toxicant, zinc sulfate, and to dilutions of primary-and secondary-treated effluents. The resulting veliger larvae were examined microscopically for larval shell abnormalities. In a longer flowthrough experiment, abalone were exposed for the entire larval phase, from the two-cell stage through metamorphosis, to compare zinc effects on metamorphosis with zinc effects on short-term larval shell development. Dissolved oxygen, pH, salinity and temperature were measured daily in test solutions, and zinc concentrations were verified by chemical analysis. No observed effect concentrations (NOECs) for zinc were 39±2.1 g l-1 in three 48 h exposures, and 19 g l-1 for the 9 d exposure through metamorphosis. Median effect concentrations (EC50s) were 68±6.9 g l-1 in 48 h tests and 50 g l-1 in the 9 d test. Abalone larvae were affected at lower concentrations of primary than of secondary effluent.  相似文献

Specimens of the oceanic decapod Systellaspis debilis were collected from six sites in the East Atlantic Ocean between 1970 and 1984, and were analysed for Mn, Fe, Cu, Zn and Cd. The data confirm that there are small but significant differences in mean metal concentrations from some sites which showed no obvious pattern in relation to geographic location of the samples. As a result, ranges of site means are quoted as baseline levels for each metal (g g-1 dry wt): 2.3 to 2.9 g Mn g-1, 31.2 to 77.8 g Fe g-1, 25.9 to 83.4 g Cu g-1, 41.9 to 92.9 gZn g-1, 11.1 to 31.8 g Cd g-1. The concentration of cadmium in S. debilis from all sites was raised relative to cadmium concentrations reported for coastal decapods, perhaps as a result of dietary enrichment. Metal accumulation may provide useful information for understanding the complex feeding behaviour of many oceanic animals.  相似文献

Respiration rates of Thais haemastoma and Callinectes sapidus were determined as a function of salinity with a flow-through respirometer at 20°C. Respiration rates were measured at 10, 20 and 30 S for acclimated animals. The effects of 10-5-10, 20-10-20, 30-10-30 and 10-30-10 S semidiurnal cycles (12 h) of fluctuating salinity on the rate of respiration of the oyster drill were studied. During each cycle, salinity was changed from the acclimation salinity over a 4 h interval, held at that salinity for 2 h, returned to the acclimation salinity over 4 h and held at that salinity for 2 h. The effects of diurnal (24.8 h) salinity cycles on respiration in the oyster drill and blue crab were also studied. Salinity was changed from the acclimation salinity over a 10.4 h interval, held at that salinity for 2 h, then returned to the acclimation salinity over 10.4 h and held at that salinity for 2 h. The respiration rate of 30 S acclimated oyster drills (679 l O2 g dry weight–1 h–1) was significantly higher than for individuals acclimated to 10 S (534 l O2 g dry weight–1 h–1). Blue crab respiration was 170 l O2 g dry weight–1 h–1 at 30 S, and was significantly higher at 10 and 20 S than at 30 S. With the exception of the 20-10-20 S semidiurnal cycle, the respiration rate of oyster drills declined as salinity fluctuated in either direction from the acclimation salinity and increased as ambient salinity returned to the acclimation salinity. Semidiurnal cycles (12 h) of fluctuating salinity produced greater changes in the respiration rate of snails than analogous diurnal cycles (24.8 h). A 10-30-10 S pattern of fluctuation caused a greater percentage reduction in the steady state respiration rate of oyster drills than the 30-10-30 S pattern. The respiration rate of blue crabs varied inversely with fluctuating salinity. Relatively minor changes occurred in blue crab respiration rate with fluctuating salinity. Blue crab respiration rate characteristically dropped during the initial phase of declining salinity at a rate directly proportional to the rate of salinity decrease, perhaps representing a metabolic adjustment period by the blue crabs. The respiratory response of T. haemastoma to salinity is consistent with its incomplete volume regulation, while the response of C. sapidus is compatible with its ability to regulate extracellular fluid osmotic and ionic composition.  相似文献

The object of this study was to evaluate the acute toxicity of cadmium in different post-embryonic stages of the penaeid shrimp Penaeus japonicus (Bate, 1888) and to determine the effect of sublethal cadmium on the osmoregulatory capacity used as an indicator of physiological condition. Tolerance to cadmium increases with the developmental stage. The least tolerant stages are the nauplii (48 h LC50: 124 g Cd l-1) and the zoeae (96 h LC50: 10 to 30 g Cd l-1). The most tolerant stages are the postlarvae (96 h LC50: 200 to 3500 Cd l-1) and juveniles (96 h LC50: 5500 g Cd l-1). In juvenile shrimp, 2000 g Cd l-1 significantly reduce hypo- and hyper-OC. The effect of cadmium on hypo- and hyper-osmoregulatory capacity illustrates a dose- and time-dependent response. Surviving shrimp recover their hypo-osmoregulatory capacity after 6 d of readaptation in cadmium-free seawater.  相似文献
