The effect of various pesticides on the biofilm formation by the phytopathogenic bacterium Clavibacter michiganensis ssp. sepedonicus (Cms), the potato ring rot causative agent, was explored for the first time. Systemic herbicides: 2,4-D, diuron, glyphosate, clopyralid, fluorodifen, as well as the commercial preparations “Lazurite,” “Ridomil Gold,” and the mitochondria inhibiting pesticides analog, sodium monoiodoacetate, were studied. These pesticides' effect on the Cms biofilm formation was shown to be distinct and dependent on the agent under question. Cms biofilm formation was reduced when exposed to sodium monoiodoacetate, as well as “Lazurite” preparation, that could be due to the bactericidal effect of these agents. 2,4-D and “Ridomil Gold” preparation stimulated the biofilm formation. Systemic herbicides diuron, glyphosate, clopyralid, fluorodifen did not exert appreciable influence on the process of bacterial biofilm formation.  相似文献

In vitro antibacterial activity tests of seven biofungicides (Ekstrasol, Bisolbisan, Bisolbifit, Serenade, Sonata, Timorex, F-Stop) and two disinfectants (colloidal silver alone and in combination with hydrogen peroxide) against the Pseudomonas tolaasii strain (NS3B6) were carried out by the disc-diffusion, broth microdilution and broth macrodilution method. Biofungicides tested in this study did not exhibit any antimicrobial activity in neither one of the methods used. Disc diffusion method revealed high sensitivity of the tested P. tolaasii strain to Ecocute based on colloidal silver and hydrogen peroxide. Both microdilution and macrodilution methods identified the same MICs and MBCs of Ecocute (0.19 mg/L) for P. tolaasii strain. MICs and MBCs values of silver alone were much higher (10 mg/L) compared to silver in combination with hydrogen peroxide.  相似文献

Abstract

The toxicity of four essential oils extracted from Baccharis articulata, Baccharis ochracea, Baccharis psiadioides and Baccharis trimera was tested against the phytopathogen Alternaria alternata, which causes Alternaria stem canker on tomatoes. Diseases caused by Alternaria fungi are responsible for great economic losses in terms of production and are controlled by synthetic fungicides; however, essential oils offer an alternative, since they have been proven to be effective for controlling against various plant pathogens. In this way, the antifungal activity of Baccharis essential oils was tested using potato dextrose agar medium with concentrations ranging from 0.1 to 20.0?µL mL?1. Baccharis trimera and Baccharis ochracea essential oils presented 100% mycelial growth inhibition of A. alternata and were also able to control Alternaria stem canker disease under greenhouse conditions. Tomato plants treated with these essential oils exhibited area under the disease progress curve (AUDPC) values of 230.10 and 241.42, differing from the control condition, which showed an AUDPC value of 268.92. The essential oils of B. trimera and B. ochracea can be an alternative for controlling Alternaria stem canker disease of tomatoes and should be formulated as a potential fungicide against the A. alternata pathogen.  相似文献

The evaluation of the inhibition effect exerted by the essential oils of basil (Ocimum basilicum), sage (Salvia officinalis), thyme (Thymus vulgaris), lemon balm (Melissa officinalis) and goldenrod (Solidago virgaurea) on seeds germination and early seedling growth of velvetleaf (Abutilon theophrasti Medik.) weed was examined in a laboratory bioassay. The essential oils were obtained by hydrodistillation and characterized chemically by gas chromatography coupled with both mass spectrometry and flame ionization detector. The working solutions of essential oil emulsified with Tween 20 and dissolved in distilled water were prepared at three concentration levels (0.01%, 0.1% and 1%, vol/vol). The results obtained showed that increase of essential oil concentration leads to decrease of seed germination, shoot and radical length of velvetleaf. The obtained data revealed a highly significant effect (P?<?0.05) between control and 1% and 0.1% oil concentrations in all treatments. The essential oils of basil, thyme and lemon balm exhibited more powerful bio-herbicidal effect compared to sage and goldenrod essential oils on the germination and early seedling growth of velvetleaf weed.  相似文献

We evaluated the in vitro activity of citrus oils against Mycobacterium tuberculosis and other non-tuberculous Mycobacterium species. Citrus essential oils were tested against a variety of Mycobacterium species and strains using the BACTEC radiometric growth system. Cold pressed terpeneless Valencia oil (CPT) was further tested using the Wayne model of in vitro latency.

Exposure of M. tuberculosis and M. bovis BCG to 0.025 % cold pressed terpeneless Valencia orange oil (CPT) resulted in a 3-log decrease in viable counts versus corresponding controls. Inhibition of various clinical isolates of the M. avium complex and M. abscessus ranged from 2.5 to 5.2-logs. Some species/strains were completely inhibited in the presence of CPT including one isolate each of the following: the M. avium complex, M. chelonae and M. avium subsp. paratuberculosis. CPT also inhibited the growth of BCG more than 99 % in an in vitro model of latency which mimics anaerobic dormancy thought to occur in vivo. The activity of CPT against drug-resistant strains of the M. avium complex and M. abscessus suggest that the mechanism of action for CPT is different than that of currently available drugs. Inhibition of latently adapted bacilli offers promise for treatment of latent infections of MTB. These results suggest that the antimycobacterial properties of CPT warrant further study to elucidate the specific mechanism of action and clarify the spectrum of activity.  相似文献

This study aimed to isolate and characterize bacteria able to use sulfentrazone in the commercial formulation as their sole carbon source. The isolation of the potential sulfentrazone-degrading bacteria was made from soil samples with a recent history of herbicide application and from isolates identified through rDNA sequencing. Subsequently, we assessed the growth of the isolates and their sulfentrazone degradation ability using high-performance liquid chromatography. Twenty-six potential sulfentrazone-degrading bacterial isolates were obtained in pure culture. Through analysis of the rDNA sequences, the predominance of bacterial species of the genus Pseudomonas was found. The isolates presented a differentiated ability of sulfentrazone degradation. The presence of herbicide in the culture medium reduced the log phase of four isolates. Pseudomonas putida, Pseudomonas lutea, Pseudomonas plecoglossicida and three isolates of Pseudomonas sp. showed higher sulfentrazone degradation capacity, which varied from 4 to 15%. This is the first report of the Pseudomonas genre capable of sulfentrazone degradation. The isolates obtained present potential use in bioremediation programs for soil contaminated with sulfentrazone.  相似文献

In the present study, the antimicrobial and antiradical activities of 15 essential oils were investigated. The antimicrobial activities were determined by using agar disc diffusion and broth microdilution methods against Clostridium genus and antioxidant properties of essential oils by testing their scavenging effect on DPPH radicals activities. We determined the antibacterial activity of Clostridium butyricum, Clostridium hystoliticum, Clostridium intestinale, Clostridium perfringens and Clostridium ramosum. We obtained the original commercial essential oils samples of Lavandula angustifolia, Carum carvi, Pinus montana, Mentha piperita, Foeniculum vulgare Mill., Pinus sylvestris, Satureia montana, Origanum vulgare L. (2 samples), Pimpinella anisum, Rosmarinus officinalis L., Salvia officinalis L., Abies alba Mill., Chamomilla recutita L. Rausch and Thymus vulgaris L. produced in Slovakia (Calendula a.s., Nova Lubovna, Slovakia). The results of the disk diffusion method showed very high essential oils activity against all tested strains of microorganisms. The best antimicrobial activity against C. butyricum was found at Pimpinella anisum, against C. hystoliticum was found at Pinus sylvestris, against C. intestinale was found at Satureia hortensis L., against C. perfringens was found at Origanum vulgare L. and against C. ramosum was found at Pinus sylvestris. The results of broth microdilution assay showed that none of the essential oils was active against C. hystoliticum. The best antimicrobial activity against C. butyricum was found at Abies alba Mill., against C. intestinale was found at Abies alba Mill., against C. perfringens was found at Satureia montana and against C. ramosum was found at Abius alba and Carum carvi. Antioxidant DPPH radical scavenging activity was determined at several solutions of oil samples (50 μL.mL?1–0.39 μL.mL?1) and the best scavenging effect for the highest concentration (50 μL.mL?1) was observed. The antioxidant properties were different in particular plant species. The highest% of inhibition after 30 min. of reaction was observed at Origanum vulgare (93%), Satureia montana (90.66%) and Lavandula augustifolia (90.22%).  相似文献

Crop losses due to bacterial pathogens are a major global concern. Most of the available pesticides for these pathogens suffer from various drawbacks such as complicated synthesis, high cost, high toxicity, pesticide resistance and environmental hazards. To overcome these drawbacks, the present study was undertaken to find a potent bactericide. Therefore, a series of compounds comprising bioactive furyl and oxazolone rings was synthesized under microwave irradiation and screened for in vitro antibacterial activity. The reactions were completed in fewer than 2 minutes with minimal use of solvents and resulted in high yields. These compounds were screened for antibacterial activity against plant pathogens, Xanthomonas oryzae, Ralstonia solanacearum and nitrifying bacteria, Nitrosomonas species under laboratory conditions. Five compounds were active as antibacterial agents against Xanthomonas oryzae and Ralstonia solanacearum. However, all compounds were effective against the Nitrosomonas species and the best one was 2-furyl-4-(3-methoxy-4-hydroxybenzylidene)-5(4H)-oxazolone. The study revealed the fast and environmentally friendly synthesis of bioactive title compounds, which also hold promise to be used as prototypes for the discovery of potent analogues.  相似文献

The aim of the present study was to assess the antifungal and anti-toxinogenic activity of 15 essential oils (EOs) against three fungi of the genus Aspergillus (A. parasiticus KMi-227-LR, A. parasiticus KMi-220-LR and A. flavus KMi-202-LR). The minimum inhibitory doses (MIDs) of the tested essential oils and their antifungal activity were determined using the micro-atmosphere method. The original commercial essential oil samples of Jasminum officinale L., Thymus vulgaris L., Syzygium aromaticum (L.) Merrill &; Perry, Rosmarinus officinalis L., Ocimum basilicum L., Eucalyptus globulus Labill., Salvia officinalis L., Citrus limon (L.) Burm, Origanum vulgare L., Lavandula angustifolia Mill., Carum carvi L., Citrus sinensis (L.) Osbeck., Zingiber officinalis Rosc., Mentha piperita L. and Cinnamomum zeylanicum Nees. (C. verum J.S.Presl.) were produced in Slovakia (Calendula a.s., Nová ?ubovňa, Slovakia). All essential oils exhibited activity against all tested strains of fungi. After 14 days of incubation, A. flavus (KMi-202-LR) showed the highest susceptibility with a growth inhibition percentage (GIP) of 18.70% to C. limon and 5.92% to C. sinensis, while A. parasiticus (KMi-220-LR) exhibited a GIP of 20.56% to J. officinale. The minimum inhibitory doses (MIDs) of EOs with the most significant activity were recorded. The best antifungal activity, using the micro-atmosphere method was found in S. aromaticum with an MID of 62.5 μL L?1 air, T. vulgaris (MID of 62.5 μL L?1 air) and O. vulgare (MID of 31.5 μL L?1 air) against all tested strains. Mycotoxin production of the tested strains was evaluated by the thin layer chromatography (TLC) method. Mycotoxin production of AFB1 and AFG1 was inhibited following all treatments with C. carvi, R. officinale and S. officinale, Eucalyptus globulus L. and O. basilicum L. Essential oils exhibited a potential inhibition activity against toxic fungi, although, these affected only the production of AFB1.  相似文献

Two essential oils (EO), thymol and carvacrol, were used in six ratio (100:00, 80:20, 60:40, 40:60, 20:80 and 00:100) combinations of both EO and in a dose of 0.2?g L?1 in bovine ruminal culture medium, 24-h cultures, to evaluate effects on total gas production (TGP), methane production, in vitro dry matter digestibility (IVDMD) and in vitro culture population dynamics of methanogenic and total bacteria. Total DNA extracted from ruminal microorganisms was subjected to denaturing gradient gel electrophoresis (DGGE)-polymerase chain reaction (PCR) to examine effects on bacterial populations. The effect of EO on TGP and IVDMD were assessed by comparison to untreated control cultures. In general, methane production by the microbial populations appeared to be higher with treatments containing the highest concentration of thymol than with treatments containing more carvacrol resulting in a tendency for greater methane-inhibiting activity achieved as the thymol concentration in the thymol:carvacrol mixtures decreased linearly. The population of total bacteria with a 74.5% Dice similarity coefficient for comparison of DGGE band patterns indicating shifts in bacterial constituents as EO ratios changed. No effects on TGP, IVDMD while only slight shifts in the methanogenic populations were seen with an overall 91.5% Dice similarity coefficient.  相似文献

Titanium dioxide (TiO(2)) photocatalysts have attracted great attention as a material for photocatalytic sterilization in the food and environmental industry. This research aimed to design a new photobioreactor and its application to sterilize selected food borne pathogenic bacteria, Salmonella choleraesuis subsp., Vibrio parahaemolyticus, and Listeria monocytogenes. The photocatalytic reaction was carried out with various TiO(2) concentrations and Ultraviolet (UV) illumination time. A feasible synergistic effect was found that the bactericidal effect of TiO(2) on all bacterial suspension after UV light irradiation was much higher than that of without TiO(2). As the concentration of TiO(2) increased to 1.0 mg/ml, bactericidal effect increased. However, the bactericidal effect was rapidly abbreviated at TiO(2) concentration higher than 1.25 mg/ml to all selected bacteria. UV illumination time affected drastically the viability of all bacteria with different death rate. Similar trends were obtained from S. choleraesuis subsp. and V. parahaemolyticus that their complete killing was achieved after 3 h of illumination. However, L. monocytogenes was more resistant and its death ratio was about 87% at that time.  相似文献

The molluscicidal activity of essential oils from two endemic (Juniperusbrevifolia; Laurus azorica) and three introduced (Hedychium gardnerianum; Pittosporum undulatum; Psidium cattleianum) Azorean plants against the snail Radix peregra was studied under laboratory conditions. Essential oils from leaves of H. gardnerianum, L. azorica and J.brevifolia presented promising molluscicidal activity on both adults and juveniles stages of R. peregra. The molluscicidal activity of these essential oils was found to be both time and concentration dependent. Lethal concentrations (LC50) varied between 15.4 (L. azorica) and 44.6 ppm (H. gardnerianum) for juveniles and from 45.3 (H. gardnerianum) to 54.6 ppm (J. brevifolia) for R. peregra adults. Ovicidal effect, calculated as percentage of egg hatching, at 100 ppm concentration, was observed in essential oils from P. undulatum flowers (4.2% of hatching) and leaves of H. gardnerianum (4.9%), L. azorica (7.4%) and J. brevifolia (17.7%). The present study is the first attempt to assess the molluscicidal potential of some Azorean plants essential oils against a Lymnaeidae snail. In fact, the H. gardnerianum, L. azorica and J. brevifolia can offer natural alternative tools for the control of R. peregra population, but more research is needed in order to determine the mode of action of these oils and determine the side effects on the ecosystem where this freshwater snail occurs.  相似文献

The essential oils from leaves of Schinus molle var. areira, Aloysia citriodora, Origanum vulgare and Thymus vulgaris have showed potential as phytoinsecticides against the green stink bug, Nezara viridula. In this work were evaluated their toxicological and behavioral effects on the parasitoid Trissolcus basalis, a biological control agent of this pest insect. Essential oils were obtained via hydrodestillation from fresh leaves. Insecticide activity in T. basalis females was evaluated in direct contact and fumigation bioassays. Behavioral effects were evaluated in olfactometer bioassays. To evaluate the residual toxicity, females of the parasitoids were exposed to oil residues; in these insects, the sublethal effects were evaluated (potential parasitism and survivorship of immature stages). The essential oils from O. vulgare and T. vulgaris proved to be highly selective when used as fumigant and did not change parasitoid behavior. After one week, the residues of these oils were harmless and did not show sublethal effects against T. basalis. According with these results, essential oils have potential applications for the integrated management of N. viridula.  相似文献

Background, aim, and scope

Solid surfaces in contact with water have been found to be biofouled due to the attachment of various organisms. For better understanding of the biofilm formation, the important initial stage of bacterial attachment was investigated with Pseudomonas aeruginosa PAO1 as a model microorganism. Effects of the biosurfactant rhamnolipids and the shear conditions were particularly examined.  相似文献

Methionine is the first limiting amino acid in poultry feed. Currently, methionine supplement is synthesized from an expensive chemical process requiring hazardous chemicals. Therefore, the objectives of this study were isolation of methionine producing bacteria from environmental samples and quantification of methionine production in these isolated bacteria. MCGC medium was selected as the isolation medium for methionine-producing bacteria by using Corynebacterium glutamicum ATCC13032 and Escherichia coli ATCC23798 as the positive and negative controls, respectively. Thirty-nine bacterial strains were obtained from environmental samples. Only strains A121, A122, A151 and A181 were able to tolerate up to 0.1% (w/v) of ethionine or norleucine. These isolated strains were identified by sequencing small subunit rRNA genes. The results revealed that bacterial strains A121, A122, A151and A181 were Klebsiella species, Acinetobacter baumannii, A. baumannii and Pseudomonas aeruginosa, respectively. When methionine production in strains A121 and A181 was quantitated, strains A121 and A181 generated methionine up to 31.1 and 124.6 μg/ml, respectively.  相似文献

We investigated changes in population and taxonomic distribution of cultivable bacteria and diazotrophs with butachlor application in rice paddy soils. Population changes were measured by the traditional plate-count method, and taxonomic distribution was studied by 16S rDNA sequencing, then maximum parsimony phylogenic analysis with bootstrapping (1,000 replications). The bacterial population was higher after 39 than 7 days of rice cultivation, which indicated the augmentation of soil microbes by rice root exudates. The application of butachlor increased the diazotrophic population in both upper (0–3 cm) and lower (3–15 cm) layers of soils. Especially at day 39, the population of diazotrophs was 1.8 and 1.6 times that of the control in upper and lower layer soils, respectively. We found several bacterial strains only with butachlor application; examples are strains closest to Bacillus arsenicus, B. marisflavi, B. luciferensis, B. pumilus, and Pseudomonas alvei. Among diazotrophs, three strains closely related to Streptomyces sp. or Rhrizobium sp. were found only with butachlor application. The population of cultivable bacteria and the species composition were both changed with butachlor application, which explains in part the contribution of butachlor to augmenting soil nitrogen-fixing ability.  相似文献

Essential oils, predominantly comprised of a group of aromatic chemicals, have attracted increasing attention as they are introduced into indoor environments through various forms of consumer products via different venues. Our study aimed to characterize the profiles and concentrations of emitted volatile organic compounds (VOCs) when evaporating essential oils indoors. Three popular essential oils in the market, lavender, eucalyptus, and tea tree, based on a nation-wide questionnaire survey, were tested. Specific aromatic compounds of interest were sampled during evaporating the essential oils, and analyzed by GC-MS. Indoor carbon monoxide (CO), carbon dioxide (CO2), total volatile organic compounds (TVOCs), and particulate matters (PM10) were measured by real-time, continuous monitors, and duplicate samples for airborne fungi and bacteria were collected in different periods of the evaporation. Indoor CO (average concentration 1.48 vs. 0.47 ppm at test vs. background), CO2 (543.21 vs. 435.47 ppm), and TVOCs (0.74 vs. 0.48 ppm) levels have increased significantly after evaporating essential oils, but not the PM10 (2.45 vs. 2.42 ppm). The anti-microbial activity on airborne microbes, an effect claimed by the use of many essential oils, could only be found at the first 30–60 min after the evaporation began as the highest levels of volatile components in these essential oils appeared to emit into the air, especially in the case of tea tree oil. High emissions of linalool (0.092–0.787 mg m−3), eucalyptol (0.007–0.856 mg m−3), d-limonene (0.004–0.153 mg m−3), ρ-cymene (0.019–0.141 mg m−3), and terpinene-4-ol-1 (0.029–0.978 mg m−3), all from the family of terpenes, were observed, and warranted for further examination for their health implications, especially for their potential contribution to the increasing indoor levels of secondary pollutants such as formaldehyde and secondary organic aerosols (SOAs) in the presence of ozone.  相似文献

Carbonyls emitted from essential oils can affect the air quality when they are used in indoors, especially under poor ventilation conditions. Lavender, lemon, rose, rosemary, and tea tree oils were selected as typical and popular essential oils to investigate in terms of composition, thermal characteristics and fifteen carbonyl constituents. Based on thermogravimetric (TG) analysis, the activation energy was 7.6–8.3 kcal mol?1, the reaction order was in the range of 0.6–0.7 and the frequency factor was 360–2838 min?1. Formaldehyde, acetaldehyde, acetone, and propionaldehyde were the dominant carbonyl compounds, and their concentrations were 0.034–0.170 ppm. The emission factors of carbonyl compounds were 2.10–3.70 mg g?1, and acetone, propionaldehyde, acetaldehyde, and formaldehyde accounted for a high portion of the emission factor of carbonyl compounds in essential oil exhaust. Some unhealthy carbonyl species such as formaldehyde and valeraldehyde, were measured at low-temperature during the vaporization of essential oils, indicating a potential effect on indoor air quality and human health.  相似文献
