Kratochvil et al. (Naturwissenschaften 95:209–215, 2008) reported recently that in the leopard gecko (Eublepharis macularius) of the family Eublepharidae with temperature-dependent sex determination (TSD), clutches in which eggs were incubated at the same temperature produce only same-sex siblings. Interpreting this result in light of studies of sex steroid hormone involvement in sex determination, they suggested that maternally derived yolk steroid hormones could constrain sex-determining mechanisms in TSD reptiles. We have worked extensively with this species and have routinely incubated clutches at constant temperatures. To test the consistency of high frequency same-sex clutches across different incubation temperatures, we examined our records of clutches at the University of Texas at Austin from 1992 to 2001. We observed that clutches in which eggs were incubated at the same incubation temperature produced mixed-sex clutches as well as same-sex clutches. Furthermore, cases in which eggs within a clutch were separated and incubated at different temperatures produced the expected number of mixed-sex clutches. These results suggest that maternal influences on sex determination are secondary relative to incubation temperature effects.  相似文献

The concentration of yolk steroids was suggested to influence offspring gender in oviparous animals subject to both temperature-dependent sex determination (TSD) and genotypic sex determination (GSD). However, the proposed mechanisms of steroid effects are thought to differ between TSD and GSD: a direct effect of oestrogens on gonad feminisation in TSD species vs a differential induction of male-producing or female-producing gametes in GSD species. Geckos offer an ideal opportunity for testing these suggested mechanisms. Closely related gecko species differ in their modes of sex determination. They lay clutches of two synchronously formed eggs; both eggs share equal steroid levels. If identical hormonal composition and environment during vitellogenesis, gravidity and incubation determine the sex of the progeny, siblings should share the same gender in both TSD and GSD geckos. We found strong support for this prediction in a TSD gecko species. Among clutches that were incubated at the temperature that produced both sexes, there were no clutches with siblings of the opposite sex. On the other hand, about half of the clutches yielded siblings of the opposite sex in four GSD species. These results suggest that sex-determining systems constrain the ability of the female to produce single-sex siblings and, hence, it seems that the GSD mechanism constrains the opportunities for sex ratio manipulation in geckos via yolk steroid manipulation.  相似文献

The resistance of the grain mite Acarus siro to unfavourable physical conditions was studied to determine how long some members of a population might survive so that development and increase could continue if conditions became favourable. Preliminary experiments showed that: (i) the eggs formed the most resistant stage, both to extreme temperatures and to desiccation; (ii) the most resistant eggs were those in an early stage of development, and that resistance declined with age; and (iii) environmental conditions prior to oviposition influenced resistance, resistance being greater in eggs from populations reared at near threshold conditions than in those reared at ecologically more favourable temperatures and humidities.Experiments with eggs exposed to lethal high and low temperatures and to both favourable and desiccating humidities show that above 30°C and below 0°C, relative humidity ceased to affect survival, the eggs being killed by heat or cold before the effects of desiccation became important. Eggs were fairly susceptible to high temperatures; none survived longer than 10 h at 35°C or 35 min at 40°C. They showed greater tolerance to low temperatures, surviving up to a maximum of 12–14 days at ?10°C and 24–26 days at ?5°C. At 0°C, the maximum survival period varied from 80 days at 20% RH to 180 days at 90% RH. For eggs exposed at temperatures within the developmental temperature range to relative humidities from 20 to 50%, survival appeared to be directly related to humidity and inversely to temperature, maximum survival periods ranging from 20 days (20% RH) to 45 days (50% RH) at 5°C, and from 1 day (20% RH) to 3 days (50% RH) at 30°C.  相似文献

The Trivers–Willard hypothesis predicts sex biases in parental investment according to parental condition. In addition, parents may need to sex bias their investment if there is an asymmetry between the sexes in offspring fitness under different conditions. For studying maternal differential investment, egg resources are ideal subjects because they are self contained and allocated unequivocally by the female. Recent studies show that yolk androgens can be beneficial to offspring, so here we test for sex-biased investment with maternal investment of yolk testosterone (T) in zebra finch (Taeniopygia guttata) eggs. From the Trivers–Willard hypothesis, we predicted females to invest more in male eggs in optimum circumstances (e.g. good-condition mother, early-laid egg), and more in female eggs under suboptimal conditions (e.g. poor-condition mother, late-laid egg). This latter prediction is also because in this species there is a female nestling disadvantage in poor conditions and we expected mothers to help compensate for this in female eggs. Indeed, we found more yolk T in female than male eggs. Moreover, in accordance with our predictions, yolk T in male eggs increased with maternal quality relative to female eggs, and decreased with laying order relative to female eggs. This supports our predictions for the different needs and value of male and female offspring in zebra finches. Our results support the idea that females may use yolk androgens as a tool to adaptively manipulate the inequalities between different nestlings.  相似文献

A bacterial strain, ZY3, growing on sex steroid hormones as the sole source of carbon and energy was isolated from the sewage treatment plant of a prophylactic steroids factory. ZY3 degrades the 3-methoxy-17β-hydroxy-1,3,5(10),8(9)-δ-4-estren (MHE). This strain was preliminarily identified as Raoultella sp. ZY3 according to its morphology and its 16S rRNA gene sequence. During the experimental period (72 h), the optimum temperature, pH and 3-MHE concentration for the degradation of hydride by the strain ZY3 were 35°C, 10 and 10 mg/L, respectively. The degradation rate of the sex steroid hormones increased to 87% and 85% after the addition of maltose and peptone, respectively. The aim of this preliminary work is to reach to a cleaner production of intact shell-table eggs free from Salmonella enteritidis (SE) contamination, by application of pasteurization and dry heat. Three highly virulent strains of SE, previously reported to the World Animal Health Organization, were used in this study. The three strains acquired plasmid profiles similar to SE isolates reported in severe food illness of humans. The plasmid profiles of the three SE strains were: strain 1 (14.1 kb), strain 2 (14.1 and ≈50.0 kb), and strain 3 (1.8, 14.1, and ≈50.0 kb).Each of the three SE strains grown up to 10 h (stationary phase) and adjusted to the same light transmission of 12% at a wavelength of 540 nm, was homogenized with yolk of eggs to form three individual inocula. Strain 1 inoculum to eggs resulted in initial SE count/ml of egg content equivalent to 1.80×1019 colony forming unit (cfu); however, strain 2 inoculum resulted in 2.3×1019 cfu, and strain 3 inoculum resulted in 2.20×1019 cfu (P>0.05). Treatment of inoculated intact shell eggs in a hot water bath at 57°C for 25 min, followed by application of hot air at 55°C for another 57 min resulted in the highest reduction in SE count/ml of the egg content for strain 1 (average 33.33 cfu), followed by strain 2 (average of 0.73×104 cfu), and strain 3 (average of 1.60×104 cfu). All counts after treatment were significantly less than the initial count before treatment (P<0.05).Decreasing the density of the SE in the three inocula by about 13 logs to be 1.80×106 cfu/ml of egg content for strain 1, 2.34×106 for strain 2, and 2.20×106 for strain 3 (P>0.05) and applying the same pasteurization and dry heat treatment on inoculated eggs resulted in complete absence of viability in the three strains, averaging zero cfu/ml of egg content.The pasteurization and dry heat treatment of intact shell-table eggs results in significant reduction of the high initial count of SE strains, regardless of their plasmid profile or density of contamination. The production of table eggs completely clean of SE could be obtained by the same treatment when the initial contamination is low (around 106 cfu/ml of egg content).  相似文献

In egg-laying animals, mothers can influence the development of their offspring via the suite of biochemicals they incorporate into the nourishing yolk (e.g. lipids, hormones). However, the long-lasting fitness consequences of this early nutritional environment have often proved elusive. Here, we show that the colorful carotenoid pigments that female zebra finches (Taeniopygia guttata) deposit into egg yolks influence embryonic and nestling survival, the sex ratio of fledged offspring, and the eventual ornamental coloration displayed by their offspring as adults. Mothers experimentally supplemented with dietary carotenoids prior to egg-laying incorporated more carotenoids into eggs, which, due to the antioxidant activity of carotenoids, rendered their embryos less susceptible to free-radical attack during development. These eggs were subsequently more likely to hatch, fledge offspring, produce more sons than daughters, and produce sons who exhibited more brightly colored carotenoid-based beak pigmentation. Provisioned mothers also acquired more colorful beaks, which directly predicted levels of carotenoids found in eggs, thus indicating that these pigments may function not only as physiological ‘damage-protectants’ in adults and offspring but also as morphological signals of maternal reproductive capabilities.  相似文献

There are fears that global warming will lead to degradation of peatlands, higher emissions of greenhouse gases from peat, and accelerated warming. Anaerobic decomposition of organic soils produces methane (CH4), a potent greenhouse gas. Two peat bogs differing in mean annual temperature, Velke Darko (VD, Czech Republic, 7.2 °C), and Stor Åmyran (SA, Sweden, 4.0 °C), were selected for a comparative study of how organic soils in different climatic zones will respond to warmer and drier conditions. Twenty peat cores from each bog were incubated in growth chambers. Under present-day summer conditions, VD produced 14 times more CH4 than SA. Two different warming scenarios were used. Peat-core replicates were kept at temperatures of 11 versus 16 °C, and 11 versus 22 °C. From 11 to 16 °C, the CH4 production slightly decreased at SA, and slightly increased at VD. From 11 to 22 °C, the CH4 production increased 9 times at SA, but slightly decreased at VD. After an 8-month incubation, peat cores under drying conditions (water table at ?14 cm) were compared to samples with original water table (?2 cm). Drying conditions led to a steeper reduction in CH4 production at VD, compared to SA. The CH4 production decreased more than 100 times at VD. Then, the combined effect of simultaneous warming and drying at 11 and 22 °C was studied. We did not find any significant effect of interactions between increasing temperature and decreasing water table level. Overall, the warmer site VD responded more strongly to the simulated climate change than the colder site SA.  相似文献

Cinnamaldehyde (CNMA), an organic compound that gives cinnamon its flavor and odor, was investigated for its virucidal activity on norovirus surrogates, murine norovirus (MNV) and feline calicivirus (FCV), and hepatitis A virus (HAV). Initially, different concentrations of CNMA (0.1, 0.5 and 1 %) were individually mixed with each virus at titers of ca. 6–7 log10 TCID50/ml and incubated 2 h at 4 and 37 °C. CNMA was effective in reducing the titers of norovirus surrogates in a dose-dependent manner after 2 h at 37 °C, while HAV titers were reduced by 1 log10 after treatment with 1 % of CNMA. When incubation time was extended, HAV titers were reduced by 3.4 and 2.7 log10 after overnight incubation at 37 °C with 1 and 0.5 % of CNMA, respectively. Moreover, this paper analyzed, for the first time, the antiviral activity of adding an active electrospun interlayer based on zein and CNMA to a polyhydroxybutyrate packaging material (PHB) in a multilayer form. Biodegradable multilayer systems prepared with 2.60 mg/cm2 (~9.7 %) of CNMA completely inactivated FCV according to ISO 22196:2011, while MNV titers were reduced by 2.75 log10. When the developed multilayer films were evaluated after one month of preparation or at 25 °C, the antiviral activity was reduced as compared to freshly prepared multilayer films evaluated at 37 °C. The results show the excellent potential of this system for food contact applications as well as for active packaging technologies in order to maintain or extend food quality and safety.  相似文献

Unlike natural selection, phenotypic plasticity allows organisms to respond quickly to changing environmental conditions. However, plasticity may not always be adaptive. In insects, body size and other morphological measurements have been shown to decrease as temperature increases. This relationship may lead to a physiological conflict in ants, where larger body size and longer legs often confer better thermal resistance. Here, we tested the effect of developmental temperature (20, 24, 28 or 32 °C) on adult thermal resistance in the thermophilic ant species Aphaenogaster senilis. We found that no larval development occurred at 20 °C. However, at higher temperatures, developmental speed increased as expected and smaller adults were produced. In thermal resistance tests, we found that ants reared at 28 and 32 °C had half-lethal temperatures that were 2 °C higher than those of ants reared at 24 °C. Thus, although ants reared at higher temperatures were smaller in size, they were nonetheless more thermoresistant. These results show that A. senilis can exploit phenotypic plasticity to quickly adjust its thermal resistance to local conditions and that this process is independent of morphological adaptations. This mechanism may be particularly relevant given current rapid climate warming.  相似文献

Blueberry and blueberry extracts are known for their health benefits and antimicrobial properties. Natural therapeutic or preventive options to decrease the incidences of foodborne viral illnesses are becoming popular and being researched. This study aimed to determine the antiviral effects of blueberry juice (BJ) and blueberry proanthocyanidins (BB-PAC, B-type PAC structurally different from A-type PAC found in cranberries) against the infectivity of hepatitis A virus (HAV) and human norovirus surrogates (feline calicivirus (FCV-F9) and murine norovirus (MNV-1)) at 37 °C over 24 h using standard plaque assays. Viruses at ~5 log PFU/ml were mixed with equal volumes of BJ (pH 2.8), neutralized BJ (pH 7.0), BB-PAC (1, 2, 4, and 10 mg/ml), malic acid (pH 3.0), or phosphate-buffered saline (pH 7.2) and incubated over 24 h at 37 °C. Each experiment was carried out in duplicate and replicated thrice. FCV-F9 titers were found to be reduced to undetectable levels with 1 and 2 mg/ml BB-PAC after 5 min, with 0.5 mg/ml BB-PAC after 1-h, and with BJ after 3-h. MNV-1 titers were reduced to undetectable levels after 3 h with 1, 2, and 5 mg/ml BB-PAC and after 6 h with BJ. HAV titers were reduced to undetectable levels after 30 min with 2 and 5 mg/ml BB-PAC, after 3 h with 1 mg/ml BB-PAC, and by ~2 log PFU/ml with BJ after 24-h. BB-PAC shows preventive potential against infection by the tested enteric viruses in a dose- and time-dependent manner, although further in vitro studies in model food systems and in vivo studies using animal models are warranted.  相似文献

In biofilm fermentation, from the very early moments, surfactin was produced along with the biofilm development in the lipopeptide antibiotic production medium by using Bacillus subtilis. However, almost no iturin A was produced in its first 24 hours of cultivation and the production of iturin A began much later. Volumes of the nutrient medium and available surface area of the biofilm reactors were found to be important with the relative production of these two antibiotics. Production of iturin A was increased from 12 mg to about 50 mg per reactor when the culture size was increased from 5 mL to 20 mL, as the depth of the medium was increased. The production level was saturated thereafter with larger volumes. On the other hand, surfactin production was remained similar, which was about 10 mg per reactor, from all the 5 mL to 80 mL of biofilm culture. Optimized temperature for iturin A and surfactin production was observed at 25 and 37°C, respectively. In the biofilm fermentation, production of surfactin was increased when the incubation temperature was increased within the temperature range of 25 to 37°C, on the other hand, iturin A production was gradually decreased with the increase of the incubation temperature.  相似文献

Aqueous Hibiscus sabdariffa extracts possess antimicrobial properties with limited information available on their antiviral effects. Aichi virus (AiV) is an emerging foodborne pathogen that causes gastroenteritis. Vaccines are currently unavailable to prevent their disease transmission. The objective of this study was to determine the antiviral effects of aqueous H. sabdariffa extracts against AiV. AiV at ~5 log PFU/ml was incubated with undiluted (200 mg/ml), 1:1 (100 mg/ml) or 1:5 (40 mg/ml) diluted aqueous hibiscus extract (pH 3.6), phosphate-buffered saline (pH 7.2 as control), or malic acid (pH 3.0, acid control) at 37 °C over 24 h. Treatments were stopped by serially diluting in cell-culture media containing fetal bovine serum and titers were determined using plaque assays on confluent Vero cells. Each treatment was replicated thrice and assayed in duplicate. AiV did not show any significant reduction with 1:1 (100 mg/ml) or 1:5 (40 mg/ml) diluted aqueous hibiscus extracts or malic acid after 0.5, 1, or 2 h at 37 °C. However, AiV titers were reduced to non-detectable levels after 24 h with all the three tested concentrations, while malic acid showed only 0.93 log PFU/ml reduction after 24 h. AiV was reduced by 0.5 and 0.9 log PFU/ml with undiluted extracts (200 mg/ml) after 2 and 6 h, respectively. AiV treated with 1:1 (100 mg/ml) and 1:5 (40 mg/ml) diluted extracts showed a minimal ~0.3 log PFU/ml reduction after 6 h. These extracts show promise to reduce AiV titers mainly through alteration of virus structure, though higher concentrations may have improved effects.  相似文献

This paper analyzes potential impacts of climate change on biomass carbon (C) density and water-use (actual evapotranspiration, AET) of savannah woodlands in Sudan. Climate change scenarios were developed from five General Circulation Models (GCMs; CGCM2, CSIRO2, ECHam4, HadCM3 and PCM) under two IPCC (Intergovernmental Panel on Climate Change) emission scenarios (A1FI and B1). Baseline (1961-90) climate and climate change scenarios for 2080s for eight map sheet grids (1° latitude x 1.5° longitude) were constructed. Compared to baseline values, mean annual precipitation (MAP) showed both increases (+112 to +221 mm) and decreases (?13 to ?188 mm) but mean annual temperature (MAT) only showed increases (+1.2 to +8.3 °C). Baseline biomass C densities showed an exponential relationship with MAP (y?=?6.798 e 0.0054x, R2?=?0.70). Depending on climate change MAP, biomass C densities increased (+14 to +241 g C m?2) or decreased (?1 to ?148 g?C m?2). However, because of uncertainty in biomass C density estimates, the changes were only significant (P <0.05) for some of the climate change scenarios and for grids with MAP >260 mm. Under A1FI emission scenarios, only HadCM3 did not have a significant effect while under B1 emission scenarios, only CGCM2 and ECHam4 had a significant effect on biomass C density. AET also showed both increases (+100 to +145 mm for vertisols and +82 to +197 mm for arenosols) and decreases (?12 to ?178 mm for vertisols and ?12 to ?132 mm for arenosols). The largest relative changes in AET (up to 31 %) were associated with grids receiving the lowest rainfall. Thus, even if MAP increases across the study region, the increase will have little impact on biomass levels in the driest areas of the region, emphasizing the need for improved management and use of savannah woodlands.  相似文献

Human Adenovirus 41 (Ad41) is an important human enteric pathogen and widely prevalent in the environment. The aim of this study was to assess the survival of Ad41 based on genome stability and infectivity in different types of manure and three types of biosolids. For viral survival studies, Ad41 was added to pelletized poultry litter (PL), alum-treated poultry litter (AL), raw poultry litter (RPL), liquid dairy manure (DM), swine manure (SM), and three types of biosolids 1, 2, 3. All samples were stored at 20 or 4°C and analyzed every 10 days for up to 60 days. Quantification PCR (qPCR) standard curves were generated for PL, AL, biosolids 1, and DM to measure the number of viral genomic copies remaining in the samples. To study the infectivity, all contaminated manure/biosolids samples were added to mammalian cell culture and viral mRNA was detected using one-step RT–PCR. Overall, Ad41 viral genomes were stable at both 20 and 4°C and there was no significant loss of viral DNA after 60 days in PL, AL, biosolids type 1, and DM. However, infectivity was lost almost immediately in high pH biosolids type 2 and 3, and infectivity decreased quickly in DM, with estimated T90 of 4.3 and 8.7 days at 20 and 4°C, respectively. Ad41 had ~1.9 log loss of infectivity after added in SM and biosolids type 1 at day 0, and estimated T90 was 12.5 and 28.6 days for biosolids type 1, and 19.1 and 51.0 days for SM at 20 and 4°C, respectively. Ad41 maintained infectivity in all three poultry litter, and after 60 days incubation, there were significantly more infectious virus in PL, AL, and RPL than biosolids 1, SM, and DM at 20°C.  相似文献

The effect of incubation temperature and wet-dry cycle on the availabilities of Cd, Pb and Zn was studied. Three soils with pH ranging from 3.8 to 7.3, organic carbon (OC) from 0.7% to 2.4%, and clay from 12.3% to 35.6% were selected. Soils were spiked with reagent grade Cd(NO3)2, Pb(NO3)2, and Zn(NO3)2 at concentrations of 30 mg Cd/kg soil, 300 mg Zn/kg soil and 2000 mg Pb/kg soil. The soils were incubated at 35, 60, 105℃, respectively and went through four wet-dry cycles. Metal availability in soils was estimated by soil extraction with 0.1 mol/L Ca(NO3)2. According to this study, the effect of the spiking temperature on the metal availabilities was different among the metals, soils and wet-dry cycles. Mostly, 35 ~C was the first recommended spiking temperature for Cd and Pb while no spiking temperature was obviously better than others for Zn. Three wet-dry cycles was recommended regardless of the type of metals and incubation temperature.  相似文献

Photodynamic inactivation (PDI) is extensively used to inactivate different type of pathogens through the use of photosensitizers (PS). Curcumin has been identified as an excellent natural photosensitizer with some potential applications in the food industry. The aim of this study was to assess the antiviral activity of photoactivated curcumin on norovirus surrogates, feline calicivirus (FCV), and murine norovirus (MNV). Initially, different concentrations of curcumin (13.5–1358 µM) were individually mixed with each virus at titers of ca. 6–7 log TCID50/ml and photoactivated by LED blue light with light dose of 3?J/cm2. Results showed that photoactivated curcumin at 50 µg/mL reduced FCV titers by almost 5 log after incubation at 37 °C for 30 min. Lower antiviral activity (0.73 log TCID50/mL reduction) was reported for MNV. At room temperature, curcumin at 5 µg/mL reduced FCV titers by 1.75 log TCID50/mL. These results represent a step forward in improving food safety using photoactivated curcumin as an alternative natural additive to reduce viral contamination.  相似文献

Genogroup II.4 norovirus (NoV) remains the predominant NoV strain in food- and water-borne outbreaks. Capsid integrity as well as viral RNA persistence were determined for GII.4 NoV by real-time RT-PCR after 1?C14 freeze/thaw (F/T) cycles (?80?°C/+22?°C) or after ?80?°C storage for up to 120?days. In both cases, capsid integrity and viral RNA titers remained stable. RNase was exogenously added after 1?C14 F/T cycles, but did not alter the amount of genomic NoV RNA detected, indicating that capsids remained intact. Presumptive NoV infectivity was evaluated in functional studies by a porcine gastric mucin binding assay. Viruses frozen and thawed up to 14× bound similarly to porcine mucin, suggesting no reduction in virus infectivity. Overall, this study shows that a) NoV particles retain their integrity for at least 14 F/T cycles, b) long-term (120?day) frozen storage does not decrease NoV RNA titers, and c) capsid binding to receptor-like glycoprotein moieties remains unaltered after 14 F/T cycles. This work indicates that freezing and thawing of foods or beverages would not be a practical processing intervention to reduce NoV contamination. Likewise, repeated freezing and thawing, as might be encountered during winter months, is not expected to inactivate NoV in the environment. Results do show that laboratory samples destined for molecular biological analyses or for use as positive controls may be repeatedly frozen and thawed without any anticipated reduction in NoV RNA titers. This study documents the cryostability of NoV capsids and RNA to freezing and thawing and to the possible retention of virus infectivity.  相似文献

Proteolytic enzyme isolated from Aspergillus tamarii prepared by solid state fermentation was studied for dehairing of goat skins in the tannery. The stability of the enzyme for dehairing was optimized at different environmental parameters. Unhairing of goat skins could be obtained at pH 9–11 and temperatures 30–37°C with enzyme concentration of 1% w/v and incubation periods of 18–24 h. The physical properties of the experimental leathers in comparison with the control sets gave better results with respect to tensile strength and elongation at break.  相似文献

The kinetics of free βhCG concentrations were measured in 30 maternal whole blood samples from second-trimester pregnancies during 72 h incubation at 3, 20, and 30°C. Dissociation of intact hCG (ihCG) was undetectable at 3°C and produced a more than 20 per cent increase of free βhCG at 20°C and a more than 100 per cent increase at 30°C. hCG dissociation at 30°C was not reduced by a protease inhibitor (sodium iodoacetate) and also occurred in purified hCG dissolved in a protease-free incubation medium. These results were reproduced under conditions of sample transport by post at different environmental temperatures. In conclusion, reliable free βhCG assessment requires that the specimen be kept cool from vene puncture until assay or completely other transport strategies have to be considered. Evaluation of free βhCG as an effective marker in prenatal Down syndrome screening must be reconsidered from this aspect.  相似文献
