The injection of bacteria in the subsurface has been identified as a potential method for in situ cleanup of contaminated aquifers. For high bacterial loadings, the presence of previously deposited bacteria can result in decreased deposition rates--a phenomenon known as blocking. Miscible displacement experiments were performed on short sand columns (approximately 5 cm) to determine how bacterial deposition on positively charged metal-oxyhydroxide-coated sands is affected by the presence of previously deposited bacteria. Approximately 8 pore volumes of a radiolabeled bacterial suspension at a concentration of approximately 1 x 10(9) cells ml-1 were introduced into the columns followed by a 2-pore-volume flush of cell-free buffer. It was found that the presence of Al- and Fe-coated sand increased both deposition rates and maximum fractional surface coverage of bacteria on the sediment surfaces. The effect of grain size on maximum bacterial retention capacity, however, was not significant. Decreasing ionic strength from 10(-1) to 10(-2) M KCl resulted in noticeable decreases in sticking efficiency (alpha) and maximum surface coverage (thetamax) for clean silica sand--results consistent with DLVO theory. In columns containing positively charged Al- and Fe-coated sands, however, changes in alpha and thetamax due to decreasing ionic strength were minimal. These findings demonstrate the importance of geochemical controls on the maximum bacterial retention capacity of sands.  相似文献

Filtration of Bacillus subtilis spores and the F-RNA phage MS2 (MS2) on a field scale in a coarse alluvial gravel aquifer was evaluated from the authors' previously published data. An advection-dispersion model that is coupled with first-order attachment kinetics was used in this study to interpret microbial concentration vs. time breakthrough curves (BTC) at sampling wells. Based on attachment rates (katt) that were determined by applying the model to the breakthrough data, filter factors (f) were calculated and compared with f values estimated from the slopes of log (cmax/co) vs. distance plots. These two independent approaches resulted in nearly identical filter factors, suggesting that both approaches are useful in determining reductions in microbial concentrations over transport distance. Applying the graphic approach to analyse spatial data, we have also estimated the f values for different aquifers using information provided by some other published field studies. The results show that values of f, in units of log (cmax/co) m(-1), are consistently in the order of 10(-2) for clean coarse gravel aquifers, 10(-3) for contaminated coarse gravel aquifers, and generally 10(-1) for sandy fine gravel aquifers and river and coastal sand aquifers. For each aquifer category, the f values for bacteriophages and bacteria are in the same order-of-magnitude. The f values estimated in this study indicate that for every one-log reduction in microbial concentration in groundwater, it requires a few tens of meters of travel in clean coarse gravel aquifers, but a few hundreds of meters in contaminated coarse gravel aquifers. In contrast, a one-log reduction generally only requires a few meters of travel in sandy fine gravel aquifers and sand aquifers. Considering the highest concentration in human effluent is in the order of 10(4) pfu/l for enteroviruses and 10(6) cfu/100 ml for faecal coliform bacteria, a 7-log reduction in microbial concentration would comply with the drinking water standards for the downgradient wells under natural gradient conditions. Based on the results of this study, a 7-log reduction would require 125-280 m travel in clean coarse gravel aquifers, 1.7-3.9 km travel in contaminated coarse gravel aquifers, 33-61 m travel in clean sandy fine gravel aquifers, 33-129 m travel in contaminated sandy fine gravel aquifers, and 37-44 m travel in contaminated river and coastal sand aquifers. These recommended setback distances are for a worst-case scenario, assuming direct discharge of raw effluent into the saturated zone of an aquifer. Filtration theory was applied to calculate collision efficiency (alpha) from model-derived attachment rates (katt), and the results are compared with those reported in the literature. The calculated alpha values vary by two orders-of-magnitude, depending on whether collision efficiency is estimated from the effective particle size (d10) or the mean particle size (d50). Collision efficiency values for MS-2 are similar to those previously reported in the literature (e.g. ) [DeBorde, D.C., Woessner, W.W., Kiley, QT., Ball, P., 1999. Rapid transport of viruses in a floodplain aquifer. Water Res. 33 (10), 2229-2238]. However, the collision efficiency values calculated for Bacillus subtilis spores were unrealistic, suggesting that filtration theory is not appropriate for theoretically estimating filtration capacity for poorly sorted coarse gravel aquifer media. This is not surprising, as filtration theory was developed for uniform sand filters and does not consider particle size distribution. Thus, we do not recommend the use of filtration theory to estimate the filter factor or setback distances. Either of the methods applied in this work (BTC or concentration vs. distance analyses), which takes into account aquifer heterogeneities and site-specific conditions, appear to be most useful in determining filter factors and setback distances.  相似文献

Propylene and butylene are highly reactive volatile organic compounds (HRVOCs) in terms of ground-level ozone formation. This study examined the effectiveness of biofiltration in removing propylene and butylene as separate compounds. Specific objectives were (1) to measure maximum removal efficiencies for propylene and butylene and the corresponding microbial acclimation times, which will be useful in the design of future biofilters for removal of these compounds; (2) to compare removal efficiencies of propylene and butylene for different ratios of compost/hard wood-chip media; and (3) to identify the microorganisms responsible for propylene and butylene degradation. Two laboratory-scale polyvinyl chloride biofilter columns were filled with 28 in. of biofilter media (compost/wood-chip mixtures of 80:20 and 50:50 ratios). Close to 100% removal efficiency was obtained for propylene for inlet concentrations ranging from 2.9 x 10(4) to 6.3 x 10(4) parts per million (ppm) (232-602 g/m3-hr) and for butylene for inlet concentrations ranging from 91 to 643 ppm (1.7-13.6 g/m3-hr). The microbial acclimation period to attain 100% removal efficiency was 12-13 weeks for both compounds. The lack of similar microbial species in the fresh and used media likely accounts for the long acclimation time required. Both ratios of compost/wood chips (80:20 and 50:50) gave similar results. During the testing, media pH increased slightly from 7.1 to 7.5-7.7. None of the species in the used media that treated butylene were the same as those in the used media that treated propylene, indicating that different microbes are adept at degrading the two compounds.  相似文献

GOAL, SCOPE AND BACKGROUND: Pentachlorophenol (PCP) is the second highest volume pesticide used in the United States. It is a mutagenic compound whose exposure poses significant health effects, One of the most desirable, environmentally friendly treatment methods is bioremediation. For soil-based contamination, the effectiveness of bioremediation will also be affected by the presence of an active indigenous population, sorption of the contaminant onto the soil, and environmental parameters. METHODS: Two pure strains and their mixed culture were used to evaluate PCP biodegradation in two different field soils, Columbia (CO) and New Mexico (NM). Biostimulation of the indigenous microbes was evaluated by adding nutrients. The efficiency of adding bacteria strains (bioaugmentation) for degrading PCP was determined with Arthrobacter sp., Flavobacterium sp. and a 50:50 mixture of the two bacteria strains. RESULTS: In CO soil, only 24%, 12% and 25% of the initial PCP concentration were degraded by Flavobacterium sp., Arthrobacter sp. and mixed culture, respectively. Arthrobacter sp. was used in NM soil with two initial concentrations and achieved degradation efficiencies of 57% and 61% for 361 and 95 mg kg- concentrations, respectively. Discussion. Analysis via statistical methods showed that the bacteria had different efficiencies on PCP degradation in each soil. 2 CONCLUSIONS: All bacteria catalyzed a higher PCP degradation when present in NM soil. Second, Flavobacterium sp. degraded more PCP than Arthrobacter sp. in CO soil. The mixed culture achieved the highest degradation efficiency regardless of the initial concentration or soil origin. RECOMMENDATIONS AND PERSPECTIVES: The effect of the soil properties, such as the soil organic matter (SOM) on PCP biodegradation should be investigated. Future work can also investigate the effect of aging time on biodegradation.  相似文献

The transport of bacteriophage PRD1, a model virus, was studied in columns containing sediment mixtures of quartz sand with goethite-coated sand and using various solutions consisting of monovalent and divalent salts and humic acid (HA). Without HA and in the absence of sand, the inactivation rate of PRD1 was found to be as low as 0.014 day(-1) (at 5+/-3 degrees C), but in the presence of HA it was much lower (0.0009 day(-1)), indicating that HA helps PRD1 to survive. When the fraction of goethite in the sediment was increased, the removal of PRD1 also increased. However, in the presence of HA, C/C0 values of PRD1 increased by as much as 5 log units, thereby almost completely eliminating the effect of addition of goethite. The sticking efficiency was not linearly dependent on the amount of goethite added to the quartz sand; this is apparently due to surface charge heterogeneity of PRD1. Our results imply that, in the presence of dissolved organic matter (DOM), viruses can be transported for long distances thanks to two effects: attachment is poor because DOM has occupied favourable sites for attachment and inactivation of virus may have decreased. This conclusion justifies making conservative assumptions about the attachment of viruses when calculating protection zones for groundwater wells.  相似文献

Flaring of waste gases is a common practice in the processing of hydrocarbon (HC) materials. It is assumed that flaring achieves complete combustion with relatively innocuous byproducts such as CO2 and H2O. However, flaring is rarely successful in the attainment of complete combustion, because entrainment of air into the region of combusting gases restricts flame sizes to less than optimum values. The resulting flames are too small to dissipate the amount of heat associated with 100% combustion efficiency. Equations were employed to estimate flame lengths, areas, and volumes as functions of flare stack exit velocity, stoichiometric mixing ratio, and wind speed. Heats released as part of the combustion process were then estimated from a knowledge of the flame dimensions together with an assumed flame temperature of 1200 K. Combustion efficiencies were subsequently obtained by taking the ratio of estimated actual heat release values to those associated with 100% complete combustion. Results of the calculations showed that combustion efficiencies decreased rapidly as wind speed increased from 1 to 6 m/sec. As wind speeds increased beyond 6 m/sec, combustion efficiencies tended to level off at values between 10 and 15%. Propane and ethane tend to burn more efficiently than do methane or hydrogen sulfide because of their lower stoichiometric mixing ratios. Results of theoretical predictions were compared to nine values of local combustion efficiencies obtained as part of an observational study into flaring activity conducted by the Alberta Research Council (ARC). All values were obtained during wind speed conditions of less than 4 m/sec. There was generally good agreement between predicted and observed values. The mean and standard deviation of observed combustion efficiencies were 68 +/- 7%. Comparable predicted values were 69 +/- 7%.  相似文献

Bioleaching of metals can be achieved in sewage sludge using Thiobacillus ferrooxidans, which obtains its energy requirements from the oxidation of added ferrous iron. The purpose of this study was to verify the presence of indigenous T. ferroxidans and to evaluate their adaptive capacity and leaching potential. Nineteen sludges (primary, secondary, aerobically and anaerobically digested, oxidation pond) were tested and all of them contained indigenous iron-oxidizing bacteria. The acclimation of these organisms by successive transfers allowed a rise of sludge redox potential over 450 mV and a decrease of sludge pH between 3.8 and 2.2 over a 10-day incubation period. The metal solubilization efficiencies were Cd: 55-98%, Cr: 0-32%, Cu: 39-94%, Mn: 71-98%, Ni: 37-98%, Pb: 0-31% and Zn: 66-98%, were reached with these indigenous strains. The results obtained show that the metal bioleaching may be easily realized by direct acclimation of sludge microflora.  相似文献

Fugitive dust emission from limestone extraction areas is a significant pollution source. The cracking operation in limestone extraction areas easily causes high total suspended particulate (TSP) concentrations in the atmosphere, occasionally exceeding the 1-hr national emission standard of Taiwan (500 microg/m3). The concentration and size distribution were measured at different distances (0.05-15 km) in the extraction areas. The highest hourly concentrations of TSP, PM10 (suspended particulate matter [PM] smaller than 10 microm), and PM2.5 (suspended PM smaller than 2.5 microm) are 1111, 825, and 236 microg/m3, respectively, during the cracking process. Measurement results obtained from the Micro-Orifice Uniform Deposit Impactor indicated that the mass median aerodynamic diameter is approximately 0.7 microm, with the geometric standard deviation exceeding 7. In addition, the emission factors are 0.143 and 0.211 kg/t for both vertical well and stair extraction operations, respectively. Experimental results demonstrate that the corresponding TSP control efficiencies for spraying water, planting grass, setting short walls, paving gravel roads, and establishing vertical well transportation are approximately 55, 50, 44, 22, and 30%, respectively. Furthermore, the PM10 control efficiencies are approximately 45, 41, 54, 35, and 30%, respectively, whereas the PM2.5 control efficiencies are roughly 23, 31, 15, 11, and 10%, individually.  相似文献

A novel liquid-solid circulating fluidized bed bioreactor (LSCFB) configured with anoxic and aerobic columns and lava rock as the biofilm carrier was used to treat synthetic municipal wastewater. Four different empty bed contact times (EBCTs) of 0.82, 0.65, 0.55, and 0.44 h were examined to optimize nutrient removal capability of the system. The LSCFB demonstrated tertiary effluent quality organic and nitrogen removal efficiencies. Effluent characteristics of the LSCFB were soluble biological oxygen demand (SBOD)10 mg l(-1) and total nitrogen (TN)<10 mg l(-1) at organic loading rate (OLR) of 5.3 kg m(-3)d(-1) and nitrogen loading rate of 0.54 kg Nm(-3)d(-1). Remarkably low yields of 0.14, 0.17, 0.19, and 0.21 g VSS g(-1)COD were observed at OLR of 2.6, 3.2, 4.1 and 5.3 kg COD m(-3)d(-1), where increment of biomass growth and detachment rate were also experienced with increasing OLR. However the system demonstrated only 30% phosphorus removal, and mass balances along the anoxic and aerobic columns showed biological phosphorus removal in the system. Organic mass balance showed that approximately 40% of the influent COD was utilized in the anoxic column and the remaining COD was oxidized in the aerobic column. The system is very efficient in nitrification-denitrification, with more than 90% nitrification of ammonium and overall nitrogen removal in the LSCFB was 70+/-11% even at an EBCT of 0.44 h.  相似文献

In this paper, we used the continuous time random walk (CTRW) framework to characterize the transport process in 1250-cm long one-dimensional homogenous and heterogeneous soil columns at the experiments conducted by Huang et al. [Huang, K., Toride, N., van Genuchten, M.Th., 1995. Experimental investigation of solute transport in large, homogeneous and heterogeneous, saturated soil columns. Trans. Porous Media. 18, 283-302]. The transport process was also simulated by using the advection-dispersion equation (ADE) and the spatial fractional advection-dispersion equation (FADE) for comparison. In the homogeneous soil column, the non-Fickian behavior is found at the distances less than 1000cm with beta values larger than 1.60, but less than 2, and Fickian form transport is obtained at distances larger than 1000cm with beta values larger than 2. In the heterogeneous soil column, we found the most anomalous behavior at distances from 200cm to 700cm with beta values ranging from 0.894 to 0.958, and non-Fickian transport process is observed at distances larger than 800cm with beta values in the range between 1 and 1.3. More significant non-Fickian behavior is found for transport in the heterogeneous soil column than that in the homogeneous soil column. The CTRW fits to the breakthrough curves (BTCs) have lower values of root mean square error (RMSE) and higher values of determination coefficient (r(2)), with respect to the fits of ADE and FADE. The CTRW model also is better captures the full evolution of BTCs, and especially their tails.  相似文献

In the northeast of Italy, high volatile organic carbon (VOC) emissions originate from small-medium companies producing furniture. In these conditions it is difficult to propose a single, efficient, and economic system to reduce pollution. Among the various choices, the biofiltration method could be a good solution, because microbial populations possess multiple VOC degradation potentials used to oxidize these compounds to CO2. Starting from the air emissions of a typical industrial wood-painting plant, a series of experiments studied in vitro microbial degradation of each individual VOC. Isolated strains were then added to a laboratory-scale biofiltration apparatus filled with an organic matrix, and the different VOC behavior demonstrated the potential of single and/or synergic microbial removal actions. When a single substrate was fed, the removal efficiency of a Pseudomonas aeruginosa inoculated reactor was 1.1, 1.17, and 0.33 g m(-3) hr(-1), respectively, for xylene, toluene, and ethoxy propyl acetate. A VOC mixture composed of butyl acetate, ethyl acetate, diacetin alcohol, ethoxy propanol acetate, methyl ethyl ketone, methyl isobutyl ketone, toluene, and xylene was then fed into a 2-m(3) reactor treating 100 m3 hr(-1) of contaminated air. The reactor was filled with the same mixture of organic matrix, enriched with all of the isolated strains together. During reactor study, different VOC loading rates were used, and the behavior was evaluated continuously. After a short acclimation period, the removal efficiency was > 65% at VOC load of 150-200 g m(-3) hr(-1). Quantification of removal efficiencies and VOC speciation confirmed the relationship among removal efficiencies, compound biodegradability, and the dynamic transport of each mixture component within the organic matrix. Samples of the fixed bed were withdrawn at different intervals and the heterogeneous microbial community evaluated for both total and differential compound counts.  相似文献

Mercury-resistant bacteria belonging to the genera Bacillus, Escherichia, Klebsiella, Micrococcus, Pseudomonas, Salmonella, Sarcina, Shigella, Staphylococcus and Streptococcus were isolated from gills and guts of fresh water fish collected from wetland fisheries around Calcutta, India, contaminated with mercury compounds. The total number of bacteria, as well as Hg-resistant bacteria, were always higher in guts than gills. Bottom-dwelling fish contained higher number of bacteria, including Hg-resistant bacteria, than surface and middle water dwelling fish. They belonged either to narrow-spectrum or to broad-spectrum Hg-resistant groups and they also possessed other heavy metal and antibiotic resistant properties. In the presence of toxic levels of HgCl(2), phenylmercuric acetate (PMA) and methylmercuric chloride (MMC), the lag in growth of the bacterial strains gradually increased with increasing concentration of Hg-compounds. Narrow-spectrum Hg-resistant bacterial strains volatilized only HgCl(2) from the liquid medium in the range of 64-89%, whereas the broad-spectrum group exhibited a high level of HgCl(2) (80-94%), PMA (72-84%) and MMC (64-80%) volatilizing capacity with inducible mercuric reductase and organomercurial lyase enzyme activities in their cell-free extracts. Cell-free extracts prepared from narrow-spectrum Hg-resistant bacterial strains induced by HgCl(2) exhibited Hg(+2)-dependent NADPH oxidation, indicating the presence of only mercuric reductase enzyme.  相似文献

进行了微波辅助工艺处理天然气净化废水的应用研究,考察了废水色度、COD、SS、含油量和细菌的去除效果以及对废水腐蚀率的降低作用,并进行了同等条件下的常温对照实验。研究结果表明:微波絮凝工艺中絮凝剂PAC和PAM的用量较常温下减少了100 mg/L和5 mg/L,絮凝沉降时间仅为常温絮凝的1/4,最佳微波辐照时间下废水的COD去除率比常温条件下提高4.6%。微波工艺处理后,难降解天然气净化废水的含油量去除率达到了94%、废水中硫酸盐还原菌死亡率达97%以上,废水腐蚀率降为原来的1/7,废水可生化性得到了很大提高,可直接进行好氧生化处理。实验表明,微波辅助工艺对难降解天然气净化废水具有很好的处理应用效果。  相似文献

The main objective of this study was to evaluate the combined effects of grain size and pore water velocity on the transport in water saturated porous media of three waterborne fecal indicator organisms (Escherichia coli, MS2, and ΦX174) in laboratory-scale columns packed with clean quartz sand. Three different grain sizes and three pore water velocities were examined and the attachment behavior of Escherichia coli, MS2, and ΦX174 onto quartz sand was evaluated. The mass recoveries of the biocolloids examined were shown to be highest for Escherichia coli and lowest for MS2. However, no obvious relationships between mass recoveries and water velocity or grain size could be established from the experimental results. The observed mean dispersivity values for each sand grain size were smaller for bacteria than coliphages, but higher for MS2 than ΦX174. The single collector removal and collision efficiencies were quantified using the classical colloid filtration theory. Furthermore, theoretical collision efficiencies were estimated only for E. coli by the Interaction-Force-Boundary-Layer, and Maxwell approximations. Better agreement between the experimental and Maxwell theoretical collision efficiencies were observed.  相似文献

The attenuation of methane and seven volatile organic compounds (VOCs) was investigated in a dynamic methane and oxygen counter gradient system simulating a landfill soil cover. The VOCs investigated were: Tetrachloromethane (TeCM), trichloromethane (TCM), dichloromethane (DCM), trichloroethylene (TCE), vinyl chloride (VC), benzene, and toluene. Soil was sampled at Skellingsted landfill, Denmark. The soil columns showed a high capacity for methane oxidation, with oxidation rates up to 184 g/m2/d corresponding to a 77% reduction of inlet methane. Maximal methane oxidation occurred at 15-20 cm depth, in the upper part of the column where there were overlapping gradients of methane and oxygen. All the chlorinated hydrocarbons were degraded in the active soil columns with removal efficiencies higher than 57%. Soil gas concentration profiles indicated that the removal of the fully chlorinated compound TeCM was because of anaerobic degradation, whereas the degradation of lower chlorinated compounds like VC and DCM was located in the upper oxic part of the column. Benzene and toluene were also removed in the active column. This study demonstrates the complexity of landfill soil cover systems and shows that both anaerobic and aerobic bacteria may play an important role in reducing the emission of trace components into the atmosphere.  相似文献

A microbiological study conducted as a complement to kinetic studies of biological denitrification as a process for treating high-sodium-nitrite wastewaters generated from ship-boiler-tube cleaning is described. The number, genera, and denitrifying capabilities of the organisms inhabiting anoxic suspended-growth reactors used in the kinetic studies were evaluated for four experimental phases. The results regarding the enumeration of bacteria supported the findings of the kinetic studies as follows: (i) the better nitrite-removal efficiencies observed in the nitrification/denitrification system as compared with direct denitrification were confirmed by the presence of larger populations of organisms capable of completely reducing nitrate or nitrite; (ii) the presence of metals in concentrations associated with boiler-tube wastewater did not affect removal performance in the nitrification/denitrification systems, nor did it affect the density of complete denitrifiers; (iii) increasing sludge ages resulted in increasing nitrite-removal efficiencies as well as populations of complete denitrifiers; and (iv) a decrease in nitrate-removal efficiencies when the actual wastewater was introduced to a system that had been acclimated to the synthetic wastewater coincided with a reduction in the number of complete denitrifiers. Regarding the types of organisms found in this study, denitrifying strains of Alcaligenes and Pseudomonas were always present in the anoxic reactors along with other denitrifying and non-denitrifying bacteria of the same genera, or other genera such as Acinetobacter and Flavobacterium. However, members of the genus Alcaligenes were the only complete denitrifiers found in the anoxic reactors, and hence they are likely to play a key role in the denitrification process.  相似文献

针对露天矿破碎站粉尘污染严重、治理技术中存在的缺陷,提出了超声波雾化喷雾降尘技术。以平朔安家岭煤矿为例,根据破碎站区域粉尘污染情况,应用数学模型对破碎站粉尘逸散规律进行解算,将模拟结果与现场实测数据对比,确定对卸载坑采用150~300μm雾滴粒径和对破碎机各产尘点采用1~10μm雾滴粒径的机体梁上外喷雾方式,通过对采取治理方案后破碎站粉尘浓度逸散的数值模拟和现场实际应用后的测试数据,显示全尘和呼吸性粉尘降尘效率分别达到94%和82%。结果表明,超声波喷雾降尘技术能有效降低破碎站粉尘浓度,保障了作业人员工作环境和安全。  相似文献

Bench-scale packed zeolite columns were set up and operated to investigate the continuous removal of ammonium ions from compost leachate. The effects of hydraulic retention time (HRT), and particle size of the zeolite on the ammonia adsorption capacity were studied. For both the coarse particle and the powdered zeolite columns, higher ammonia removal efficiencies were achieved with longer HRT (i.e., lower influent flow rate) tests. At the same HRT, ammonia removal efficiencies from tests with powdered zeolite were generally 20% higher than tests with the coarse particle zeolite. A HRT of 6 hours was found appropriate for efficient ammonia removal, and an operating capacity of 1.31 mg N/g zeolite was obtained. Over 98% of the ammonia input from the influent was consistently removed for over 5 bed volumes (BV) of compost leachate flowing through the zeolite column. Zeolite proved to have a great potential as a medium for ammonia removal in treating composting leachate.  相似文献
