Freshwater fish Prochilodus lineatus were exposed to sublethal concentrations of cypermethrin (0.3 and 0.6 microg/l) for 2, 5 and 8 days. It was observed that with the increase of exposure time total erythrocyte (RBC), hemoglobin (Hb), hematocrit (Ht) and mean corpuscular hemoglobin concentration (MCHC) values decreased but mean corpuscular volume (MCV) and mean corpuscular hemoglobin (MCH) values increased. These reports indicate that hematological parameters, may be useful as a diagnostic test for cypermethrin exposure in aquatic organisms.  相似文献

Adults Swiss mice were administered 5% solution of textile industry wastewater orally for 25 days and haematological parameters like RBC, WBC, Hb, and PCV were studied. Red cell indices like MCV, MCH and MCHC were calculated. Results indicate significant reduction in RBC, Hb and PCV levels. It is inferred that toxic effluents cause metabolic alteration in erythrocytes and reduce their Hb carrying capacity.  相似文献

The effect of heavy metals and mixture were studied on some haematological and biochemical parameters in fish, Cyprinus carpio at sublethal level over the period of 30 days. Heavy metal significantly decreased total RBC count, haemogoblin, heamatocrit (Hct) (except copper after 10 days exposure). The WBC count was increased significantly in all the treated fish. The MCH, MCHC, MVC were increased depending upon the exposure period; declined PVC was noticed at 1% level of significance after 30 days on all the treated fish. Serum level of chloride and calcium content were increased where as serum glucose, cholesterol and total serum protein were decreased significantly. The alteration in haematological and biochemical parameters shows more toxic in mixture after 30 days, showed more than 'addition action' of the individual toxicant.  相似文献

Excessive fluoride consumption leads to accelerated red blood cell death and anaemia. Whether that increases the haematological alteration in subjects with haematological disorders (iron deficiency, thalassaemia, and G-6-PD deficiency) is still unclear. The fluoride in serum and urine and haematological parameters of students at Mae Tuen School (fluoride endemic area) were analysed and compared to those of students at Baan Yang Poa and Baan Mai Schools (control areas). Iron deficiency, thalassaemia, and G-6-PD deficiency were also diagnosed in these students. The students at Mae Tuen School had significantly (P < 0.001) higher levels of mean fluoride in the serum and urine than those in control areas. In both control and fluoride endemic areas, students with haematological disorders had significantly lower levels of Hb, Hct, MCV, MCH, and MCHC than those without haematological disorders. Moreover, the lowest levels of Hb, MCH, and MCHC were observed in the students with haematological disorders who live in the fluoride endemic area. Thus, the excessive fluoride consumption increased haematological alteration in subjects with iron deficiency, thalassaemia, and G-6-PD deficiency and that may increase the risk of anaemia in these subjects.  相似文献

Electrophoresis release test (ERT) was established by our lab to observe the re-released hemoglobin (Hb) from red blood cells (RBCs) and whole blood. In this study, ERT was performed to study the effects of different plasma components including plasma, serum, albumin, globulin, fibrinogen, glucose, amino acid, vitamin, insulin, hormone, and inorganic ions on re-released Hb from RBC and whole blood samples during ERT. The results showed that plasma, serum, albumin, globulin, compound amino acid, essential amino acid, vitamin C, insulin, hormone, NaCl, KCl, CaCl2, and NaHCO3? decreased re-released RBC Hb; while glucose, vitamin B1, vitamin B2, and vitamin B12 elevated re-released RBC Hb. The differing effects of various plasma components on re-released Hb of RBC may play a significant role in blood conservation.  相似文献

The present study was to evaluate heavy metal toxicity stress symptoms in fish blood during long-term exposure of sublethal concentration of copper sulphate (CuSO4). The effects of copper on various heamatological parameters were evaluated exposing Indian fresh water fish, Channa punctatus to a sub-lethal concentration of copper sulphate (0.36 mg l(-1)) for different periods. Exposure of fish to copper showed a significant decrease in the haemoglobin (Hb) content from 10.73 to 6.60%, red blood cells (RBC) from 2.86 to 1.84 x 10(6)/mm3 and packed cell volume (PCV) from 31.00 to 23.33% at the end of 45th day as compared to control. Whereas the white blood cells (WBC) increased from 60.00 to 92.48 x10(3)/mm3, clotting time (CT) from 27.66 to 43.00 second, erythrocyte sedimentation rate from 5.0 to 13.66 mm/hr and mean corpuscular volume from 108.11 to 126.85 cmu, significantly, with increase in exposure periods. Although mean corpuscular haemoglobin concentration (MCHC) and mean corpuscular haemoglobin (MCH) values showed significant increase during 15 and 30 days exposures, both the values were found significantly increased at the end of the experimental period. The differential leucocytes count (DLC) showed a significant increase in the populations of lymphocytes and eosinophils, whereas neutrophils, monocytes and basophiles were found decreased after exposure to copper.  相似文献

The acute toxicity of three formula grade pesticides namely, triazophos (an organophosphate, OP), deltamethrin (a pyrethroid) and combined pesticide (triazophos?+?deltamethrin) was determined in earthworm, Eudrilus eugeniae. They were exposed to different concentrations of these pesticides for 48 h by paper contact toxicity method. The LC50 values for triazophos, deltamethrin and of combination were determined as 0.076, 0.031 and 0.065?μg/cm2, respectively. To study the sublethal effect of these pesticides, E. eugeniae were exposed to 5% and 10% of LC50 of pesticides for 48 h. Morphological alterations such as coiling, clitellar swelling, mucus release and bleeding followed by body segmentation were observed in exposed earthworm. Acetylcholinesterase (AChE) activity assayed in different regions of body segment exhibited a significant (p?<?0.05) decrease in its activity particularly in the pre-clitellar region as compared to other regions. The altered behavioural responses in pesticides exposed earthworms would have been due to decline in AChE activity of the nervous system.

The effects of exposure of tilapia (Oreochromis mossambicus) to sublethal concentrations of lead on hematological and immunological parameters were investigated. The fish were semistatically exposed to low (L) (0.5 mg L?1), medium (M) (2.5 mg L?1), and high (H) (5 mg L?1) lead concentrations for 14 days. At the end of the trial, red blood cell (RBC) count, hematocrit (Hct) ratio and hemoglobin levels showed significant decreases in M and H groups, while RBC was significantly increased in L group compared to control group. Lysozyme activity showed a decrease in all groups on day 14 compared to control. A decrease in myeloperoxidase activity was seen in M and H groups on day 7, and in all treatment groups on day 14. In conclusion, it was found that exposure of O. mossambicus to lead concentrations affected hematological and immunogical parameters adversely.  相似文献

Fish are often exposed to highly contaminated water, especially in areas where the dilution rate of waste water is low. Environmental pollutants or other stress may affect one or more of the immunological functions in fish. Effective control of toxic substances is essential for the success of fish culture, which requires fish health monitoring using biomarkers. This study evaluated the effects of heavy metal copper (Cu) at environmentally relevant concentrations on the hematopoiesis in Labeo rohita. L. rohita fingerlings were exposed to different Cu concentrations for 6, 24, 48, or 72 hr. Following exposure, percentage blast cells was found to be increased after 6 and decreased after 24, 48, and 72 hr both at sublethal and LC50 concentrations, whereas percentage mature erythrocytes decreased after 6 and increased after 24, 48, and 72 hr. Erythropoietic and leukopoietic efficiencies were also affected upon exposure. Erythropoietic and leucopoietic efficiencies increased markedly after 6, 24, and 48 hr but decreased after 72 hr in sublethal-treated fish. Erythropoietic efficiency decreased significantly after 6, 24 and 48 hr but increased after 72 hr in LC50-treated fish. Leukopoietic efficiency decreased significantly after 6 hr exposure and increased after 24 and 48 hr but decreased after 72 hr. Flow cytometric studies of head kidney cell cycle phase distribution revealed cell death and DNA synthesis. The percentage cell death increased in fish exposed to sublethal concentrations and rose further at LC50 during the earlier hours of exposure. Synthesis of DNA was increased at sublethal concentrations but was significantly reduced at LC50 dose.  相似文献

This investigation was undertaken to evaluate the chemopreventive effect of hydroalcoholic extract of Indigofera aspalathoides (HAIA) against 20-methylcholanthrene (20-MC)-induced carcinogenesis in mice. Tumor was induced by single subcutaneous administration of 20-MC (200 µg per mouse) in Swiss albino mice. After 24 h of 20-MC administration, HAIA was administered at the doses of 250 and 500 mg kg?1 body weight orally for 90 consecutive days. Mice of all groups were observed for 15 weeks to record tumor incidence (fibrosarcoma) and survival time. After 15 weeks the mice were sacrificed for the estimation of hematological profiles like hemoglobin (Hb), white blood cell (WBC), red blood cell (RBC), and liver biochemical parameters, namely lipid peroxidation, reduced glutathione (GSH), glutathione-S-transferase (GST), superoxide dismutase (SOD), and catalase (CAT). HAIA treatment markedly reduced tumor incidence and prolonged the life span of sarcoma-bearing mice as compared to 20-MC control mice. Hematological profiles were significantly (p < 0.001) restored to normal levels in HAIA-treated mice as compared to 20-MC control mice. HAIA treatment significantly (p < 0.001) modulated the aforesaid liver biochemical parameters as compared to 20-MC control. The results concluded that I. aspalathoides demonstrated a remarkable chemopreventive effect in chemical-induced carcinogenesis in mouse. The potential chemopreventive action may be due to its antioxidant and detoxifying properties.  相似文献

This study investigated the relative toxicity of water-based cuttings (WBC) and synthetic oil-based cuttings (SOBC) to the marine species, Metamysidopsis insularis. Results obtained indicate that SOBC (LC50 1.2 (0.85–1.6)%) was more toxic to M. insularis than WBC (LC50 9.9 (8.3–11.8)%), with similar metal contents in both types of cuttings. The elevated levels of metals found in the cuttings when compared to surficial sediments may be due to both drilling fluids, as well as the rock strata from which the cuttings were obtained. Furthermore, total petroleum hydrocarbon (TPH) analyses demonstrated significantly higher concentrations of TPH present in SOBC (14,680?±?1250?mg?kg?1) compared to WBC (860?±?115?mg?kg?1). This may also be due to the increased depth and hence oil bearing rock formations in the selected sampling area, along with the associated synthetic oil-based drilling fluid. These findings therefore supply evidence that drill cuttings after treatment prior to discharge are potentially toxic to marine organisms.  相似文献

Deltamethrin, a pyrethroid insecticide widely used in many countries, causes significant adverse effects in aquatic ecosystems. The concentration of deltamethrin in water reservoirs and the run off from agricultural areas (in water) in many countries range up to 24.0?µg?L?1 which is higher than the recommendation of the European Union standard. Hence, in this study the effects of deltamethrin were investigated, i.e. its toxic impacts on the freshwater catfish Heteropneustes fossilis in terms of serum calcium and prolactin cells (located in the rostral pars distalis region of the pituitary). The fish were subjected to deltamethrin for a short-term experiment (96?h; 1.5?µg?L?1 e.0.8 of 96?h LC50) and a long-term experiment (28 days; 0.37?µg?L?1 e.0.2 of 96?h LC50). After short-term deltamethrin exposure, serum calcium levels decrease. No histological change in prolactin cells is noticed throughout the short-term experiment. Fish exposed to deltamethrin for 7 days also exhibit a decrease in serum calcium level. This decrease persisted until the end of the experiment (28 days). Prolactin cells of fish treated for 14 days with deltamethrin exhibit increased nuclear volume and degranulation, increasing progressively from the 21st day onwards. After 28 days, a few degenerated cells are discerned. The results of this study show that deltamethrin is moderately toxic for the freshwater fish H. fossilis by producing adverse effects on serum calcium and prolactin cells. Hence, it should be used with caution in areas near fish-inhabited waters.  相似文献

Fresh and healthy specimens of Oreochromis niloticus (mean weight 70.5± 1.23 g and mean length, 12.3± 1.5cm) and Clarias gariepinus (mean weight, 41.8±2.3 g and mean length 13.78 ± 1.2 cm) were exposed to heavy metal lead, ranging in concentration from 10 to 18 mg L‐1 for O. niloticus and 20 to 28 mg L‐1 for C. gariepinus respectively. The 96 h LC50 for both the species was computed as 12.45 mg L‐1 (O. niloticus) and 22.65 mg L‐1 (C. gariepinus). The cichlid fish was exposed to sublethal concentration of lead (2, 4 and 6 mgL‐1) and catfish (4, 8 and 12 mgL‐1). The erythrocytes count, haemoglobin concentration, haematocrit and the value of serum glucose increased significantly (P < 0.01), whereas leucocytes count, serum protein and serum triglyceride after intoxication declined in both the species with the same pattern but in different quantity. After 96 h of time a significant depletion of muscle and liver glycogen was noted, being greater in muscle (16.7% in O. niloticus) and in liver (15.9% in C. gariepinus). The response of fish to lead was dose dependent.  相似文献

Capoeta damascina has a large distribution in the Middle East and is one of the most abundant fishes in inland waters of Iran. Histopathological symptoms of the blood, gill, liver and kidney of C. damascina were investigated over 1, 5 and 9-day exposure to 0 (control), 0.5, 1 and 1.5?mg?L?1 diazinon. The data were analysed using analysis of variance and non-parametric multivariate analysis of variance. Diazinon concentration was more important than the exposure time on changes of the tissues. Diazinon decreased red blood cell (RBC), white blood cell (WBC), haematocrit (Hct) and haemoglobin (Hb). The symptoms of the gill were mainly desquamation, epithelial hyperplasia, hyperplasia at the base of secondary lamella, epithelial lifting and curling. The prevalent symptoms of the liver were degeneration of nuclei, alternation in size and shape of hepatocytes, focal necrosis and pyknosis. Degeneration of Bowman's capsule, necrosis in renal tubule and haematopoietic tissue were the most common symptoms found in the kidney. In conclusion, the blood, gill, liver and kidney of C. damascina are sensitive enough to respond to low concentrations of diazinon over a short period and this species can thus be a bioindicator of diazinon.  相似文献

The toxicity of fenitrothion was determined in larvae (nauplii, Zoeae 1 to 3, Mysis 1 to 3), postlarvae (PL stages) and juvenile shrimp (Penaeus japonicus Bate), in two media, seawater (SW) and diluted seawater (DSW) (1100 and 550 mosM kg−1, ≃ 37 and 19‰ S). The effects of fenitrothion on the osmoregulatory capacities (OC) of juveniles were recorded. A gill and epipodite histopathological study was also conducted. For larvae in seawater, 24 and 48 h LC50s ranged from 32.9 μg l−1 (Zoeae 2) to 10.7 μg l−1 (Mysis 3), and from 3.9 μg l−1 (Zoeae 3) to 2.0 μg l−1 (Mysis 3), respectively; 48 and 96 h  LC50s in postlarvae (PL) at the same salinity ranged from 1.8 μg l−1 (PL1) to 0.6 μg l−1 (PL5), and from 0.3 μg l−1 (PL7) to 0.4 μg l−1 (PL15). In juveniles, 96 h LC50s were 0.8 μg l−1 in seawater and 1.5 μg l−1 in diluted seawater. From hatching to juvenile stages, the overall trend was a rapid decrease (from nauplii to PL5–PL7) followed by a slight increase (from PL7 to PL15 and juveniles) in the shrimp's ability to tolerate the insecticide. In juveniles kept in seawater and in diluted seawater, fenitrothion decreased the osmoregulatory capacity (OC = difference between the hemolymph osmotic pressure and the osmotic pressure of the medium) at both lethal and sublethal concentrations. This effect was time- and dose-dependent. In SW, the decrease in hypo-OC was ˜ 25% at sublethal concentrations and ˜ 35% at the 96 h LC50. In DSW, the decrease in hyper-OC was ˜ 10 to 15% at sublethal concentrations. In SW, shrimp were able to recover their OC in less than 48 h when transferred to water free of pesticide. In DSW, recovery at 48 h was only possible after exposure to the lowest tested sublethal concentration. Haemocytic congestions (thrombosis) of the gills, lamellae necrosis and other alterations of gills and epipodites (breakage of the cuticle, reduction of the hemolymph lacunae) were noted in juveniles exposed to lethal and sublethal concentrations of fenitrothion. Hematological responses of broiler chicken exposed to simulated crude petroleum emissions at varying distances over a period of 16 h daily were evaluated for 56 days in a poultry house. The burning of crude petroleum was simulated with a specially designed burner. The measured distances were 4, 8, or 12 m from the flame point. Control birds were located in a separate poultry building without the flame treatment. Gaseous emissions from the burning crude oil were monitored. Proprietory broiler starter and finisher diets were fed ad libitum. Blood samples were taken at the fourth and eighth weeks for hematological assay from each treatment. The hematological parameters assayed were hemoglobin (Hb), packed cell volume (PCV), white blood cells (WBC), neutrophils, and lymphocytes. Results indicated that the treatment did not significantly affect the Hb, PCV, and WBC constituents. Neutrophils significantly decreased while lymphocytes increased. The age of birds significantly increased Hb, PCV, and WBC levels in the blood.  相似文献

Absorption of fluorine compounds produces chronic toxicity and the hematological changes appear earlier in blood than other organs. A study was undertaken to determine the status of hematological changes in the individual residing in fluoride-contaminated area. Two hundred and fifty nine and 233 subjects from the contaminated area and an unaffected area, respectively, were selected for hematological investigations. Total erythrocytes (red blood corpuscles (RBC)), total leucocytes (white blood corpuscles (WBC)), and hemoglobin (Hb) concentrations were determined in these populations. Results revealed that Hb values were significantly lower among all age groups in the exposed population compared to control subjects. There was a quantitative rise in WBC counts in study subjects compared to controls. The RBC counts showed reduction among exposed population.  相似文献

In this study, the effects of environmental hypercapnia on hemato-immunological parameters and the activities of respiratory enzymes such as carbonic anhydrase (CA) and Na+, K+-ATPase were investigated in rainbow trout (Oncorhynchus mykiss) tissues (gill, liver and kidney). Batches of 12 fish were exposed to 4.5 mg L?1 (control) and 14 mg L?1 CO2. No mortalities occurred during the 14 days of the experimental period. Red blood cell (RBC), hemoglobin (Hb), and hematocrit (Ht) levels, and innate immune parameters such as nitro blue tetrazolium (NBT), lysozyme, and myeloperoxidase activities, and the melano-macrophage frequency were negatively affected by elevated CO2 levels. Patterns of change in CA activity differed among the gill, liver, and kidney. Compared with the activities of CA in the control group, the CA enzyme was significantly stimulated at day 7 in the gill tissue, whereas it was stimulated at day 14 of the experiment in the liver tissue of fish exposed to 14 mg L?1 CO2 (P < 0.05). In contrast to the pattern of CA enzyme activities, the Na+, K+-ATPase enzymes were stimulated significantly in the liver after day 7 but inhibited in the kidney and gill (P < 0.05). These results suggest that a subchronic exposure to hypercapnia of rainbow trout tissues may lead to adaptive changes in the respiratory enzymes and negatively affects hemato-immunological parameters.  相似文献

The unrestricted use of agrochemicals in crop fields lowers the overall prospects of survival and yield of wetland fish species like Anabas testudineus. In the present study, the impact of different doses of SUMIDON-40, an organophosphate (OP) pesticide on the hematopoiesis of Anabas testudineus has been examined. The small lymphoid hemoblast and consequently the polychromatophilic erythroblasts decreased significantly at both sublethal and LC50 dose-treated fish. The percentage of mature erythrocyte also fell significantly in the pesticide-treated groups. The erythropoietic efficiency was increased in fish exposed to sublethal dose but fell in fish exposed to LC50 dose. Among leucocytes, the percentage of neutrophils rose in Anabas testudineus in both treatment groups and percentage of macrophage decreased significantly only in LC50 dose-treated group. The percentage of lymphocyte increased significantly in sublethal dose-treated groups. The overall leukopoietic efficiency, however, was elevated significantly in both treatment groups. These facts clearly indicated that pesticides affected the overall process of hematopoiesis in this fish species. Flow cytometric analysis of cell cycle in pronephric kidney cells confirmed changes in percentage of cell death and DNA synthesis following pesticide exposure. Data indicate that habitat deterioration from agrochemicals impedes the hematopoiesis in this species resulting in reduced tolerance in their usually hypoxic habitat environment.  相似文献
