Particulate extracts from six different environmental emission sources were assayed for genotoxic activity in mouse BALB/c 3T3 clone A31-1 cells. All compounds were tested simultaneously for both transforming and mutagenic (induction of ouabain-resistance) potential with and without exogenous metabolic activation in the form of a 9000 × g postmitochondrial hepatic supernatant fraction from Aroclor-1254 induced Fischer 344 rats. Dichloromethane particulate extracts from the exhaust of two light duty diesel engines (Oldsmobile and Nissan), one heavy duty diesel engine (Caterpillar) and one late model gasoline engine (Mustang II) were assayed in an identical manner to particulate extracts from the emissions of a roofing tar pot and a coke oven. No clear dose-dependent responses were observed, but several of the samples showed significant transforming and mutagenic activity. A qualitative ranking system showed the activity of these particulate extracts for either mutagenesis or transformation was: coke oven = Mustang II gasoline engine > Nissan diesel engine > roofing tar. Particulate extracts from the Oldsmobile diesel engine and the Caterpillar diesel engine showed essentially no activity.  相似文献

Measurement of personal exposure to ambient level particulate concentrations is often extremely difficult because of a lack of personal exposure monitors capable of collecting measurable quantities within a meaningful sampling period. A new personal exposure monitor for two fractions of inhalable particulates (i.e., the 3–15 μm aerodynamic diameter and the < 3 μm or respirable fraction) has been developed and characterized. This monitor is capable of collecting a sample of each fraction that is quantifiable with ambient concentrations of inhalable/respirable particulates as low as 25 μg/m3 in a 24-h sampling period. Wind tunnel tests have been made on the particulate personal exposure monitor to determine sampling efficiency as a function of relative wind speed and orientation with respect to the sampler.  相似文献

The concentrations of chromium, iron, copper, nickel, lead, vanadium, and zinc in fish were determined to assess the impact of petroleum-refining activities and the 1991 Gulf War. Twenty-eight species of fish from the Kuwait coast were collected 50 m apart. Sample solutions prepared were analyzed using the atomic absorption spectrophotometry technique. Results show that fish samples contain 6.2 to 6.8 μg/g Cr, 4.2 to 96.2 μg/g Cu, 18.8 to 126.4 μg/g Fe, 0.5 to 20.4 μg/g Ni, 0.2 to 14.6 μg/g Pb, 0.02 to 15.6 μg/g V, and 7.6 to 81.3 μg/g Zn. Copper, nickel, and zinc in Station I, covering the Kuwait City area, were often greatly in excess of those present in Station II, covering the Al-Ahmadi area, while chromium, iron, lead, and vanadium were greater in Station II. Significant correlations were observed between some trace metals in fishes, indicating their common association.  相似文献

The concentrations of environmental tobacco smoke (ETS) constituents including benzene were measured in the living rooms of 10 nonsmoking households and 20 households with at least one smoker situated in the city and suburbs of Munich. In the city, the median benzene levels during the evening, when all household members were at home, were 8.1 and 10.4 μg/m3 in nonsmoking and smoking homes, respectively. The corresponding levels of 3.5 and 4.6 μg/m3 were considerably lower in the suburbs. Median time-integrated 1-week benzene concentrations in the city were 10.6 μg/m3 in nonsmoking homes and 13.1 μg/m3 in smoking homes. In the suburbs, the corresponding values were 3.2 and 5.6 μg/m3. While the benzene concentrations in nonsmoking homes located in the city were significantly higher (p < 0.05) than in suburban nonsmoking households, no difference was found between smoking and nonsmoking households located either in the city or in the suburbs. Individual exposures to benzene and to specific markers for tobacco smoke of all household members (82 nonsmokers and 32 smokers) were determined by questionnaire, personal monitoring, and biomonitoring. Within the city, the benzene exposure determined by personal samplers was 11.8 μg/m3 for nonsmokers living in nonsmoking homes and 13.3 μg/m3 for nonsmokers in smoking homes. The corresponding values for nonsmokers living in the suburbs were 5.9 and 6.9 μg/m3, respectively. Neither difference was statistically significant. Nonsmokers living in nonsmoking households in the city had significantly higher exposure to benzene compared to their counterparts living in the suburbs (personal samplers: 11.8 vs 5.9 μg/m3, p < 0.001; benzene in exhalate: 2.4 vs. 1.1 μg/m3, p < 0.05; trans,trans-muconic acid excretion in urine: 92 vs. 54 μg/g creatinine, p < 0.05). Nonsmokers from all households with smokers were significantly more exposed to benzene than nonsmokers living in the nonsmoking households (personal samplers: 13.2 vs. 7.0 μg/m3, p < 0.05; benzene in exhalate: 2.6 vs. 1.8 μg/m3, p < 0.01; trans,trans-muconic acid excretion in urine: 73 vs. 62 μg/g creatinine), but the contribution of ETS to the total benzene exposure was relatively low compared to that from other sources. Analysis of variance showed that at most 15% of the benzene exposure of nonsmokers living in smoking homes was attributable to ETS. For nonsmokers living in nonsmoking households benzene exposure from ETS was insignificant.  相似文献

The Ames bacterial mutagenicity test system was used to evaluate parameters which may affect the mutagenic activity of diesel particulate extracts. The optimal extraction conditions, extractability of mutagens by simulated biological fluids and the effect of collection method were investigated. The role of solvent was examined by extracting diesel particles with methanol, acetone, cyclohexane, ethyl acetate, n-hexane, dichloromethane, benzene and a benzene-ethanol mixture. Of these, the dichloromethane extract exhibited the highest activity in the Ames test, although methanol yielded the largest extractable mass. Diesel particles were also extracted by dimethyl sulfoxide (DMSO) and four other simulated biological fluids for 48 h at 25, 37, and 45°C to study the effects of temperature. The mutagenic activity of the DMSO extract began to decline at temperatures higher than 37°C after 8 h of incubation. Fetal calf serum was the only simulated biological fluid which eluted mutagenic activity from the particles. No activity was detected in the 0.5% bovine serum albumin, simulated lung surfactant and saline extracts. Diesel particles collected by electrostatic precipitation (ESP) and filtration were studied. The mutagenic activities of both extracts were comparable when expressed as revertants per mg of particle. After the extracts were separated into nine fractions by a solvent partitioning scheme, the majority of the activity was found in the neutral-nonpolar II, neutral polar, strong acid and weak acid fractions. The acid salt fraction from the ESP sample was inactive. These results demonstrate that differences in the extraction conditions can result in differences in the mutagenic activity of diesel particulate extract. Since the mutagens in the extracts are not readily extractable by simulated biological fluids, the question of bioavailability of mutagens in diesel particles must be considered in the final assessment of their potential effects in biological systems and organisms.  相似文献

A new automated version of the piezoelectric microbalance measures the mass concentration of airborne particles smaller than a preselected aerodynamic cutoff diameter. It is designed for near-real-time, unattended, round-the-clock measurements of nearly any aerosol environment inhabited by humans. The instrument uses an electrostatic precipitator to deposit particles with greater than 95% efficiency onto a piezoelectric quartz crystal sensor which is able to detect a deposit of 0.005 μg. The precipitator and sensor are nearly identical to those in the portable instrument reported previously. Measurements comparing within ± 15% with gravimetrically measured filter samples are documented for a wide variety of aerosols in the 50 μg/m3 to 5.5 mg/m3 range. The instrument measures particles from 10 μm down to 0.01 μm in diameter, including submicron combustion smokes and metallic fumes. The piezoelectric microbalance technique senses the mass concentration of the aerosol, rather than light scattering properties as is done by photometers and nephelometers. The piezobalance, with 1 L/min sample flow, is more sensitive than any other mass-sensing instrument, making it especially suited for low concentration indoor measurements, even below 50 μg/m3. An automatic piezobalance recently measured respirable aerosol mass concentrations in several offices. Each measurement was the average concentration during a 30-min period. The 24-h/day measurements continued for several days. Especially interesting is the diurnal pattern, both for offices with and without smokers. The effect of a single nearby smoker was clearly illustrated when the smoker was absent one day in the middle of a week. Normal daytime peak concentrations in that office reached 80–110 μg/m3 with a smoker present, but only 50–60 μg/m3 when the smoker was absent. Curious smokers who briefly stopped byt o see the instrument caused single half-hour averages to triple, to values as high as 294 μg/m3 in one office.  相似文献

The air quality in a newly built preschool was investigated in a longitudinal study. Typical air contaminants emanating from building materials were determined, their variation over time (0–18 months) was measured, and the influence of the ventilation system (81%–91% recirculation of return air) on contaminant concentrations was studied. Volatile organic compounds were sampled by adsorption on porous polymer, analysed by a GC/FID system, and identified by MS. A spatial build-up in concentration (ppb or μg/m3 levels) is evident for all the organic compounds, as well as for CO2, from the outdoor air, through the ventilation system, and through the rooms to the exhaust air. The longitudinal comparison over time shows that all the organic compounds decline in concentration mainly within the first 6 months of occupancy: 1-butanol 4–14 times, toluene and pentanal + hexanal 2–4 times, while formaldehyde remained at a constant low level of 90 ppb (110 μg/m3). It is difficult to believe that the problems of poor air quality in 100 preschools in Stockholm are caused by the organic compounds alone unless interactions occur. A preschool building needs to be gassed off during the first 6 months after its construction with no recirculation of return air allowed (outdoor air rate approx 4–5 ach). During at least 1–2 additional years, it is desired that the recirculation rate of return air is restricted, perhaps to 50%.  相似文献

Leaves of six plant species from 31 different locations in Ibadan, Nigeria, were analysed for their lead content. The lead concentrations found ranged between 12 and 32 μg/g, 50 m or more away from the roadside, between 47 and 115 μg/g in the residential low traffic density areas, 5 m from the roadside, and between 165 and 312 μg/g in the commercial high traffic density roadsides, 5 m away from the roadside.  相似文献

PM2.5 is the breathable fraction of the particulate matter and some adverse health effects, such as respiratory functionality, cardiological diseases and cancer, can be in some measure attributable to this risk factor exposure. Some of the most carcinogen compounds transported by PM2.5 are nitro-compounds. In this study, a strengthened in vitro bioassay — able to predict the mutagenic/carcinogenic activity of the environmental mixtures — was conducted on PM2.5 organic extracts to define the nitro-compounds burden. PM2.5 air pollution was daily monitored, during 2006, in three cities located in the Northern part of Italy (Torino, Pavia and Verona) and the mutagenic properties of the PM2.5 organic extracts were assessed with the Ames test. The bacterial used in this study were three Salmonella typhimurium strains: TA98, nitroreductase-less mutant TA98NR and YG1021 carrying a nitroreductase-producing plasmid. The annual PM2.5 mean level measured in Torino was 46.5 (± 31.6) μg/m3, in Pavia 34.8 (± 25.1) μg/m3, and in Verona 37.3 (± 27.8) μg/m3, while the mutagenicity expressed as TA98 net reverants/m3 was 28.0 (± 22.1), 28.3 (± 24.9), and 34.2 (± 30.9) respectively. Monthly pool bioassays, conducted with the three different strains, showed a greater mutagenic response of the YG1021 in each city. The relationship among the mutagenic answers for YG1021:TA98:TA98NR was about 6:3:1 (p < 0.001). Over nitroreductase activity enhanced the response of 2.2, 2.0 and 1.7 times for Torino, Pavia, and Verona (ANOVA Torino p < 0.05) respectively. Without nitroreductase activity the genotoxicity was limited. These biological findings are able to describe a relevant role played by the nitro compounds in the mutagenic properties of the urban PM2.5 in the Padana plain; moreover the bacterial nitroreductase plays a predominant role in DNA interaction primarily for Torino PM2.5 extracts.  相似文献

Oxidants of significance to human health include ozone, nitrogen dioxide, and peroxyacetylnitrate. All of these compounds are involved in complex photochemical reactions which makes quantification and prediction of their individual health effects difficult. Ozone causes trauma to lung tissues and interferes with enzyme systems in the lungs and other tissues causing a broad range of symptoms. Measurable health impacts can occur at concentrations as low as 390 μg/m3. Acute effects of ozone exposure are reversible at normal urban concentrations (80–120 μg/m3). A special problem of concern, however, is increased susceptibility to infectious diseases contracted through the lungs. Nitrogen dioxide also causes trauma to lung tissues and interferes with enzyme systems. Measurable impacts can occur at concentrations as low as 100 μg/m3, but recovery is rapid and it is not known whether repeated exposures at this level have cumulative effects or predispose the lungs to permanent damage. Chronic exposure of laboratory animals to higher nitrogen dioxide levels can cause emphysema-like conditions and reduction in resistance to respiratory infection. Epidemiological studies of children in houses with gas stoves confirm the finding of reduced resistance to respiratory infection. The U.S. EPA estimates that health effects may occur in young children exposed to concentrations in excess of 280 to 560 μg/m3 one-hour average. These concentrations occur routinely in houses having gas stoves. Peroxyacetylnitrate is a powerful eye irritant in photochemical smog. Other health effects are similar to those of ozone, but less important because of the relatively low concentrations of this pollutant compared to other oxidants.  相似文献

Two microbial extracellular enzyme activities (MEEA) were studied in HUMEX Lake Skjervatjern: acid phosphatase (APHA) and leucine aminopeptidase (LeuAMPA). Both enzyme activities varied in the vertical and horizontal scale in both lake sites. APHA varied in the acidfied Basin A between 945–1706 nmol L−1 h−1 and LeuAMPA between 3.7–25 nmol L−1 h−1. Both MEEA reached maxima in 0.5 m depth. In the control site (Basin B), APHA was lower by a factor of two, and varied between 156–669 nmol L−1 h−1. LeuAMPA reached similar values as in Basin A and varied between 7.8–34.8 nmol L−1 h−1. Maxima of APHA were found in the upper layer (0–2 m), while LeuAMPA had only one distinct maxima at 2–2.5 m depth. The number of bacteria (AFDC) varied between 4.4–8.8 106 cells mL−1 and was not significantly different in either side, but both had maxima in the thermocline. Highest specific LeuAMPA activities were found in the thermocline (3.2–4.5 fmol L−1 h−1 cell−1) in both sides and varied between 0.4–4.5 fmol L−1 h−1 cell−1 in both water columns. The main contributor (60–70%) to LeuAMPA was found in the microplankton fraction, retained on Nuclepore filters with pore sizes between 2.0-0.2 μm. APHA was retained less even on a filter with pore size smaller than 0.2 μm. About 50–70% of APHA passed through 0.2 μm-0.1 μm Nuclepore filters and could be found in the dissolved organic matter (DOM) fraction. APHA and bacteria counts (AFDC) showed a distinct gradient from the littoral zone to the pelagial in the surface water samples (0.2 m depth). APHA and LeuAMPA are regarded as important regulators for nutrient availability to microplankton. However, all data from vertical as important regulators for nutrient availability to microplankton. However, all data from vertical and horizontal samples showed that Lake Skjervatjern is a strongly gradient-dominated aquatic ecosystem. Watershed-littoral effects are more pronounced in the shallow, acidified Basin A than in the control side, Basin B.  相似文献

The present field study addressed the need for a procedure that provides a defined unit of measurement of perceived annoyance from environmental odors, calibrating the estimates for individual scaling behavior and context effects. In including 25 subjects, the purpose was to demonstrate the applicability of the master-scale procedure with magnitude estimation to perform such a calibration of odor-annoyance estimates for target stimuli such as road-traffic combustion exhausts (13 000 vehicles/d; averaging 47 μg/m3 over the day/night with peaks exceeding 100 μg/m3 of nitrogen dioxide; NO2). For comparison, calibrated estimates were also obtained for a backyard expected to be considerably less polluted (comparable with 18 μg/m3 of NO2) and for blank stimuli presented indoors. The data transformation for the calibration procedure with which annoyance is expressed in either master-scale units or pyridine equivalents requires estimates of a reference stimulus for which seven concentrations of pyridine were used. The results provide an illustration of master scaling of odor annoyance, and imply that use of a modulus (standard stimulus with a predefined annoyance magnitude), in contrast to master scaling, is not sufficient for calibration for individual scaling behavior and context effects.  相似文献

Two studies evaluating the levels and sources of nitrogen dioxide in approximately 90 employee homes in the Richmond area with continuous sampling during the weeks of August 5, 1980, and February 9, 1981, were performed using samplers in the living room, bedroom, kitchen, and outdoors. Additional data were collected concerning appliance usage, heating/cooling plant, ventilation and cigarette smoking. Results were analyzed using BMDP routines. The largest contributor to NO2 concentration was found to be gas-fired kitchen appliances. The mean kitchen level for homes with gas appliances during the winter study was 188 μg/m3. Excluding participants with gas kitchens, incremental influence due to cigarette smoking was detected. The 7-day, 3-room average level of NO2 in the homes of nonsmokers and smokers without gas-fired appliances was 12 and 15 μg/m3, respectively, in the summer. The corresponding winter values were 19 and 22 μg/m3. Furthermore, the individual levels of NO2 in the homes of smokers were generally below both the adjacent outdoor level and the National Ambient Air Quality Standard limit for annual exposure.  相似文献

Soil can be a storage place and source of pollutants for interfacial environments. This study looked at a site contaminated with wood preservatives as a source of mutagens, defined routes and extent of the dispersion of these contaminants by particle remobilization and atmospheric deposition, considering an evaluation of risk to human health by quantifying mutagenic risk. Soil sampling sites were chosen at gradually increasing distances (150, 500 and 1700 m) from SI (industrial area pool) and indoor dust (pool in an area at risk at 385 m and at 1700 m). Mutagenesis was evaluated in the Salmonella/microsome assay, TA98, TA97a and TA100 strains with and without S9 mix, YGs strains 1041, 1042 and 1024 for nitrocompounds. Acid extracts were analyzed to define the effects of metals and organics for polycyclic aromatic hydrocarbons (PAHs) and nitroderivates, besides concentrations of these compounds and pentachlorophenol (PCP). Risk to human health was obtained from the relation between the quantified potential of mutagenic risk and estimated soil ingestion for children according to USEPA. Metal concentrations showed a gradient of responses with As, Cr and Cu (total metal) or Cr and Cu (fraction available) higher for SI. However, mutagenic effects of the mixtures did not show this grading. Site SR1700, without a response, was characterized as a reference. In organic extracts, the mutagenesis responses showed the mobility of these compounds from the source. In the surrounding area, a smaller pattern similar to SI was observed at SR150, and at the other sites elevated values of direct mutagenesis at SR500 and diminished effects at SR1700. Tests with YG strains indicated that nitrated compounds have a significant effect on the direct mutagenesis found, except SR500. The investigation of indoor dust in the surrounding area enabled confirmation of the particle resuspension route and atmospheric deposition, showing responses in mutagenicity biomarkers, PAH concentrations and PCP dosage similar to SI. The range of values obtained, considering the soil masses needed to induce mutagenicity was 0.02 to 0.33 g, indicating a high risk associated with human populations exposed, since these values found surpass the standard estimate of 200 mg/day of rate of soil ingestion for children according to USEPA. The study showed that it is essential to evaluate the extent of contamination from the soil to delimit remedial measures and avoid damage to the ecological balance and to human health.  相似文献

Diesel exhaust particles were used to compare methods and techniques used in the preparation of particulate matter for microbial mutagenesis testing. Investigated in this study were extraction, concentration, and solvent exchange methodologies as they affected recovery of mutagenic material from diesel samples using a Salmonella typhimurium plate incorporation assay. Solvent removal methods applicable for use in determining the mass concentration of extracts were also evaluated. Results indicated that particle samples Soxhlet extracted with dichloromethane yielded higher levels of mutagenic activity than did comparative samples utilizing sonication. No difference was seen between rotary evaporation or Kuderna-Danish macro concentration of extracts to volumes > 50 mL. In comparison of micro concentration techniques to volumes < 10 mL, vortex evaporation was found to be more efficient than a modified micro Kuderna-Danish method in recovery of mass and mutagenicity. Solvent exchanged samples were found to yield higher recoveries of mutagenic activity than samples taken to dryness and then reconstituted in the bioassay solvent. A dry mass weighing procedure utilizing desiccation was found to be more acceptable than either the use of an infrared heat lamp or nitrogen blowdown for solvent removal.  相似文献

Mutagenic activity in diesel emission particulate extracts was detected by the Salmonella typhimurium/microsome assay. Direct-acting mutagens as well as promutagens requiring metabolic activation were detected. The extracts were fractionated into acidic, basic, and neutral fractions, and the neutral fraction was chromatographed into seven subfractions. Differences in the mutagenic potency of these fractions and subfractions were determined by the Salmonella assay. Fractions containing as yet unidentified compounds, but not polynuclear aromatic hydrocarbons, were found to make a major contribution to mutagenic activity of the extracts.  相似文献

A standardized questionnaire was used in a two-year follow-up study to test the effectiveness of non-regulatory recommendations to improve indoor air quality of 103 ice arenas in Finland. In addition, the performance of a state-of-the-art emission control technology (ECT) on propane-fueled resurfacers was evaluated by measuring the one-week average nitrogen dioxide (NO2) concentration in a small sample of arenas. The number of retrofitted ECT on propane-fueled resurfacers increased from 6 to 37 (8% to 37% of ice arenas) and the number of electric resurfacers from 7 to 9 (both 9% of arenas) in 1994–1996. At the same time, the prevalence of inadequate ventilation increased among the most susceptible small arenas (volume <30 000 m3) from 11 (31%) to 19 (38%). Combustion-powered resurfacers (88%) and inadequate ventilation (24%) were prevalent also among the 17 new arenas built in 1994–1996. ECT resurfacers significantly decreased the mean indoor NO2 concentration of eleven arenas from 650 μg/m3 to 147 μg/m3. Thus, retrofitting resurfacers with ECT seems to be a feasible mitigation option to improve indoor air quality in ice arenas, but the ultimate solution is an electric ice resurfacer. Non-regulatory recommendations seem to be partially effective in abatement against the air quality problems, but additional regulatory measures are needed for full compliance in all arenas.  相似文献

For the first time, zooplankton and environmental factors have been studied in winter in the deep stratified lakes Siverskoe, Borodaevskoe, and Vydogoshch. The zooplankton consists of 18–22 species of rotifers, copepods, and cladocerans, including 15–18 specifically winter, cryophilic and eurythermic forms. The populations of zooplankters reproduce actively: males and egg-bearing females (in copepods), females with embryos and eggs in brood pouches (in cladocerans), and females with attached eggs (in rotifers) are present. In lakes Siverskoe and Borodaevskoe (depth 20–24 m), where oxygen deficiency is observed only in the bottom water layer, the zooplankton populates the greater part of the water column and forms a large aggregation in the oxycline and near the bottom. The abundance of bacteria in these zones is high (up to 4–6 × 106 cells/ml). In Lake Vydogoshch (depth 16 m), oxygen deficiency extends to the middle layers, and all zooplankton is distributed above the oxycline. The biomass of winter zooplankton in Lake Siverskoe in 1983 averaged 1.26 g/m3, with a maximum of 4 g/m3; in 1993, 1.4 and 8.04 g/m2; the respective values in Lake Borodaevskoe were 0.98 and 3.1 in 1983 and 0.14 and 1.6 in 1993; in Lake Vydogoshch, 0.15 and 0.24 in 1983 and 1.25 g/m3 in 1993. The development of winter zooplankton is limited by the size of the zone of oxygen deficiency and depends on the amount of food (bacterioplankton). A chronic exposure study was initiated to determine the effects of diesel exhaust on the health of experimental animals. For this purpose, test atmospheres of clean air (control) or freshly diluted diesel exhaust at concentrations of 250, 750, and 1500 μg/m3 were supplied to four 12.6 m3 inhalation chambers which housed rats and guinea pigs. Diesel aerosol size and concentration, as well as chamber temperature and relative humidity, were continually monitored and controlled to maintain the exposure dose levels and an environment of 22±2°C and 50%±20% relative humidity. The concentrations of CO and NOx were found to be 5.8±1.0 mg/m3 and 7.9±1.0 mg/m3 above ambient in the chamber containing 1500 μg/m3 of particulate. Animals were supplied from the chambers, on a random basis, for both intramural and extramural studies throughout the exposure period. The experiment ran uninterrupted for over twelve months with mean diesel particle mass concentrations within 2% of the target values.  相似文献

Biomonitoring studies have increased as a consequence of risks and effects to human health on exposure to environmental contaminants, mainly air pollutants. Genetic biomarkers are useful tools for the early assessment of exposure to occupational and environmental pollution. The objective of the present study was to investigate genotoxic effects on people residing and/or working downwind from an oil refinery in southern Brazil and the mutagenic activity of airborne particulate matter (PM10). Samples of peripheral blood and buccal mucosa cells were evaluated using the single-cell gel electrophoresis assay (comet assay) and the micronucleus (MN) assay, respectively. PM10 samples were collected in the target site and the organic matter extraced with dichloromethane was assessed for mutagenic activity in the Salmonella/microsome assay. The exposed group (n = 37) was compared to a reference group (n = 37) of subjects living in an urban area with limited traffic and industrial influence, located far from the main industrial areas. All PM10 organic extracts showed mutagenic positive responses and the effect decreased in the presence of S9 mix indicating that the predominant compounds present were direct-acting mutagens. The responses of YGs strains are consistent with aromatic amines and nitroarenes being present in the PM10 extracts. The group in the area under the influence of the oil refinery (exposed group) showed significantly higher DNA damage in lymphocytes than the reference group. The MN frequencies in buccal mucosa were very low for both groups and no difference between groups was observed. No association was found between age and tobacco smoking habit and level of DNA damages measured by the comet assay. The results indicate that the comet assay was a sensitive tool to detect DNA damage in subjects under the influence of an oil refinery, with marked genotoxic activity in the atmospheric environment.  相似文献
