Aluminium accumulation by the freshwater snail Lymnaea stagnalis is correlated with behavioural depression which is ameliorated by addition of orthosilicic acid. We hypothesised that Si is relocated to the digestive gland in response to Al, leading to the formation of non-toxic hydroxyaluminosilicates (HAS). Exposure to 500 microg l(-1) Al for 30 days was associated with an initial period of behavioural depression, followed by apparent tolerance and subsequent depression, suggesting saturation of the cellular detoxification pathway during prolonged exposure. Exogenous Si (7.77 mg l(-1)) completely ameliorated all behavioural effects of Al but did not prevent its accumulation. In the presence of added Al, significantly more of this Si was accumulated by the tissues, compared to controls and snails exposed to Si alone. In snails exposed to Al plus Si, Al and Si concentrations were significantly correlated, with a ratio around 3:1 Al:Si, consistent with the presence of the non-toxic HAS protoimogolite.  相似文献

This study examined the accumulation of aluminium (Al), mostly as the insoluble (Al(OH)(3)) species, by the freshwater crustacean Asellus aquaticus at neutral pH. Animals were exposed to a range of Al concentrations (5-356 microg l(-1)) in three experiments. The first two were of 30 and 50 days duration, respectively, followed by transfer of the A. aquaticus to water containing no Al for 20 days. The third used live and dead animals in order to investigate the contribution made by surface adsorption of Al to the total accumulated. Significant accumulation of Al in the whole tissues occurred by day 10 in all animals in the 30- and 50- day experiment. Peak concentrations of Al were measured in animals between days 10 and 20 with high concentration factors ranging from 1.4 x 10(4) to 5.5 x 10(3). By day 30, accumulated Al had fallen but was still significantly greater than the control in the 50- day exposure experiment. This 30- day increase followed by decreased accumulation of Al was repeated over the remaining exposure period (i.e. 30-50 days) although rates of uptake and loss and peak tissue levels of Al were higher. Proportionality between environmental (water) and tissue concentrations of Al occurred at day 20 but not at day 45. Significantly more Al was accumulated by dead animals than live animals at all Al exposure concentrations. These results suggest that Al is available to the crustacean at neutral pH and that the cuticle may provide an important site of uptake.  相似文献

In this study the great ramshorn snail (Planorbarius corneus), one of the most abundant gastropod of Turkish limnic systems, was investigated to determine the histopathological effects of endosulfan on the digestive gland, foot and mantle under laboratory conditions. Samples were collected from small artificial pools in Karaot at Gelendost-Isparta (southwest of Turkey), where agricultural activities are widespread. The snails were exposed to two sublethal concentrations of endosulfan (0.4 and 0.8 mg/l) for periods of 10, 20 and 30 days. Fifteen snails were kept in 2.5 l glass jars containing dechlorinated tap water and exposed under semi-static test (daily exchange of test water). All the testing was carried out on adult specimens, and snails were maintained on a photothermal period with 16 light hours at 22 +/- 2 degrees C. The histopathological examinations revealed the following changes: amoebocytes infiltration, dilatation in hemolymphatic spaces between the tubules, degeneration of cells, abnormal lumen, necrosis of cells and atrophy in the connective tissue of digestive gland; desquamation of the epithelium cells, changes in the number of mucocytes and protein gland cells, lipid vacuolus and atrophy of the columnar muscle fibers of the foot and mantle tissues. Pycnotic state of cells was also seen in the mantle tissues. Endosulfan caused significant histopathological alterations in the digestive gland, foot and mantle tissues of the snail, irrespective of concentrations of the pesticide and its exposure periods. The results are discussed, particularly in comparison to those of other aquatic organisms.  相似文献

The role of algal concentration in the transfer of organic contaminants in a food chain has been studied using the ubiquitous model polycyclic aromatic hydrocarbon benzo[a]pyrene (BaP) as the contaminant, Isochrysis galbana as the phytoplankton food source, and the common mussel (Mytilus edulis) as the primary consumer. The effect of algal concentration on BaP uptake by M. edulis was determined by feeding M. edulis daily with I. galbana which had previously been kept in the presence of BaP for 24 h. Four combinations of concentrations of algae and BaP were used to give final exposure concentrations of 30,000 or 150,000 algal cells ml(-1) in combination with either 2 or 50 microg BaP l(-1). BaP concentrations were determined fluorometrically in rest tissues (excluding digestive glands) and digestive gland microsomal fractions of M. edulis after 1, 7 and 15 days exposure, and also in isolated algae. Potentially toxic effects of BaP on M. edulis were examined in terms of blood cell lysosomal membrane damage (neutral red dye retention assay) and induction of digestive gland microsomal mixed-function oxygenase (MFO) parameters [BaP hydroxylase (BPH) and NADPH-cytochrome c (P450) reductase activities]. BaP bioaccumulation in rest tissues (and to a lesser extent in digestive gland microsomes) of M. edulis increased with both increasing BaP and algal exposure concentrations, and over time, producing maximal bioconcentration factors in rest tissues after 15 days exposure to 150,000 algal cells ml(-1) and 50 microg BaP l(-1) of 250,000. The five-fold higher concentration of algae increased BaP bioaccumulation by a factor of approximately 2 for 50 microg BaP l(-1) at day 15. Blood cell neutral red dye retention time decreased linearly with increasing log(10) tissue BaP body burden, indicating an increased biological impact on M. edulis with increasing BaP exposure possibly due to a direct effect of BaP on blood cell lysosomal membrane integrity. An increase was seen in NADPH-cytochrome c reductase activity, and indicated in BPH activity, with 1 but not 7 or 15 days exposure to BaP, indicating a transient response of the digestive gland microsomal MFO system to BaP exposure.  相似文献

The distributions of copper, zinc, iron, and cadmium among the tissues of Dorippe granulata were determined. The highest copper concentrations were found in the haemolymph (c. 53 microg ml(-1)) while the highest iron concentrations occurred in the gills (c. 720 microg g(-1) dry weight) and the highest zinc concentrations in the exoskeleton (c. 200 microg g(-1) dry weight). By comparison, concentrations of the non-essential metal, cadmium, were low in all tissues (mean = 10 microg g(-1) dry weight). The highest value was recorded from the midgut gland of a female crab (18.5 microg Cd g(-1) dry weight). Concentrations of copper, zinc, and iron were positively correlated with tissue-hydration levels. Such a relationship was not found for cadmium. The findings are discussed with regard to trace-metal levels found in temperate and tropical brachyurans from clean and polluted localities.  相似文献

Cd, Cu and Zn were analysed in the organs and tissues of the three scallop species from the Bay of Biscay, the variegated Chlamys varia L., the queen scallop Aequipecten opercularis L. and the common scallop Pecten maximus L. for interspecific comparisons. In P. maximus the greatest concentrations of Cd and Cu were found in the digestive gland, whereas the two other species showed similar levels of Cd between digestive gland and kidneys and higher renal Cu concentrations. However, the digestive gland of all Pectinidae species contained from 75% to 93% and 52% to 74% of the total body burdens of Cd and Cu, respectively. Whatever the species, kidneys displayed the highest Zn concentrations, which therefore contained from 53% to 97% of the total body burden of this metal. Also using reported results, ratios between the concentrations in the digestive gland and that in the kidneys discriminated two groups of Pectinidae: (1) the Pecten group (P. maximus, P. jacobeus and Adamussium colbecki) with a Cd ratio > or = 4, a Cu ratio > or = 1 and a Zn ratio > 20x10(-3); (2) the Chlamys group with a Cd ratio < or = 1, a Cu ratio < or = 1 and a Zn ratio < or = 6x10(-3). However, no differences in the detoxification processes in the digestive gland were found between groups in this study. 72-80% of the total Cd was found to be soluble, probably bound to metalloproteins such as metallothioneins, while Cu and Zn were mainly found associated to the insoluble fraction of the digestive gland cells whatever the species. Queen scallops from the Faroe Islands were also considered to examine variations of the metal concentrations due to the geographical origin. In this northern area, queen scallops displayed Cd concentrations 2, 4 and 6 times higher in kidneys, muscle and digestive gland, respectively, compared to those from the Bay of Biscay. Consequently, the Cd concentrations in the whole soft parts of the queen scallops were more than 6 times higher in the Faroe Islands than in the Bay of Biscay. In contrast to Cd, both Cu and Zn concentrations in the whole soft parts were 2 times lower in the Faroe Islands, suggesting lower bioavailability of these essential elements.  相似文献

The potential value of the marine prosobranch Nerita saxtilis as an efficient biological monitor to heavy metal pollution in the Red Sea was investigated. Storage ability of lead and cadmium was compared in shell, headfoot and digestive gland of the marine snail N. saxtilis collected from Al-Hamrauin area at El-kuseir (lead, 300.35 +/- 28.53 microg/l, 1,716 +/- 16.14. cadmium 20.01 +/- 1.8 microg/l, 161.72 +/- 21.4 mean +/- S.D. for water and sediment, respectively) relative to that of inhabiting marine water and sediment employing atomic absorption spectrometry to determine the organ with highest capability of heavy metal accumulation. The influence of metal storage on light microscopic structure of that organ was investigated. Also, the ultrastructure localization of storage sites in the same organ was determined employing transmission electron microscopy. The digestive gland was shown to accumulate both metals at conccntrations that are several orders of magnitude higher than those in the surrounding marine water. The bioaccumulation capability of lead and cadmium was ranked in the following order; digestive gland > headfoot > shell for lead and digestive gland > shell > headfoot for cadmium. In spite of its evident highest metal storage capability, no histopathological changes were observed in the digestive gland of that marine prosobranch. Enlarged electron dense vesicles and many granules were observed in ultrathin sections in digestive cells of these snails and are suggested to be the sites of storage of detoxified metals. The results of that finding indicate the possibility of using the marine prosobranch N. saxtilis as biomonitor for heavy metal contaminants in the Red Sea.  相似文献

This study assesses the potential of the scallop Chlamys varia as a biomonitor of metal contamination in the field. Cd, Cu, and Zn concentrations were determined in the soft tissues and organs of individuals from the French Atlantic coast sampled over a 1 year period and covering a wide range of size. All metals were selectively distributed among the different body compartments considered, and their concentrations were influenced by the size of the specimens or the sampling-season. The present work shows the importance of considering the body compartment, the sampling period and the size in studies aiming at using this scallop as a biomonitor species. Among tissues, the digestive gland and kidneys exhibited the highest metal concentrations whatever the season or the size. The digestive gland contained 65 and 48% of the whole Cd and Cu body burdens, respectively, and kidneys accounted for 85% of the Zn load. Those tissues are therefore particularly recommended for use in biomonitoring programs.  相似文献

The biological availability of metals in municipal wastewater effluents is strongly influenced by the physical and chemical conditions of both the effluent and the receiving water. Aquatic organisms are exposed to both dissolved and particulate (food ingestion) forms of these metals. In the present study, the distribution of metals in specific tissues was used to distinguish between exposure routes (i.e. dissolved vs. particulate phase) and to examine metal bioavailability in mussels exposed to municipal effluents. Caged Elliptio complanata mussels were deployed at sites located between 1.5 km upstream and 12 km downstream of a major effluent outfall in the St. Lawrence River. Metals in surface water samples were fractionated by filtration techniques to determine their dissolved, truly-dissolved (<10 kDa), total-particulate and acid-reactive-particulate forms. At the end of the exposure period (90 days), pooled mussel soft tissues (digestive gland, gills, gonad, foot and mantle) were analyzed for several metals. The results showed that gills and digestive gland were generally the most important target tissues for metal bioaccumulation, while gill/digestive gland metal ratios suggest that both exposure routes should be considered for mussels exposed to municipal effluents. We also found that Ag and Cd in the dispersion plume nearest the outfall, in contrast to other metals such as Cu and Zn, are more closely associated with colloids and were generally less bioavailable than at the reference site in the St. Lawrence River.  相似文献

The loss of Pb from snail soft tissues may depend on the excretion of Ca, and involve the mobilization of shell Ca. Most sub-adults in three populations of Cantareus aspersus (syn. Cornu aspersum Müller) either failed to add, or lost, shell mass on a diet with 250 μg g−1 Pb. Their failure to mineralize shell extensions occurred irrespective of food consumed, time or dietary Mg. Budgets of metals for 36 individuals in each of two populations showed that Ca loss would account for the reduction in shell mass. Lead concentrations were higher in the reduced shells but this may be a consequence of their smaller mass, rather than its cause. In both populations shell reduction correlated with the total mass of Pb assimilated. Any shell growth may have been dependent on the initial Ca reserve in each snail. Differential movement of Mg, Pb and Ca occurred between the shell and soft tissues.  相似文献

The transfer and the toxic effects of Cd were studied in an experimental food chain involving the snail Helix aspersa as prey organism and one of its natural predators, the carabid beetle Chrysocarabus splendens. Juvenile snails were fed plant-based food enriched with 0, 10, 50 and 100 microg g(-1) of Cd, then were offered as prey to beetle larvae from egg hatching to pupation stage. Cd concentrations in snail tissues increased with increasing Cd concentration in food and with duration of exposure. Bioaccumulation factors ranged from 1.87 to 3.39, showing that H. aspersa snails, even in their early life stages, belong to macroconcentrator species for Cd. No significant reduction of snail consumption by beetles was found in exposed groups. Cd concentrations in beetle larvae remained very low (lower than 1 microg g(-1) for all groups), demonstrating a very effective regulation capacity in beetle larvae. However, Cd concentrations in highest exposed groups were higher than those found in control groups. Cd contents in adult beetles were lower than in larvae, showing a loss of Cd during metamorphosis. Despite the low Cd concentrations found in beetles, their exposure to Cd contaminated snails led to 31% of mortality, which occurred only during pupation and for the highest exposure level. No clear sublethal effects were found. These results showed that snails inhabiting heavily polluted areas may represent a risk of secondary poisoning for predatory invertebrates and provided quantitative data on the transfer of Cd between two compartments of a terrestrial food chain.  相似文献

Anuran tadpoles are found in a variety of habitats, many of which are acidified or have high ambient concentrations of metals from anthropogenic sources. A few studies that have been conducted on metals in tadpoles demonstrate that they can contain high concentrations of some metals but have not demonstrated clear relationships between ambient conditions and metal concentrations. This study examines the influence of soil, water treatment, amphibian species, and body portion analyzed on metal concentration in tadpoles. In northern cricket frogs, gray treefrogs, and green frogs, concentrations of Al and Fe exceeded 10 000 microg.g(-1) and Mg and Mn exceeded 1000 microg g(-1). Body concentrations of Ba, Be, Fe, Mg, Mn, Ni, Pb, and Sr increased with soil concentrations. Acidification reduced body concentrations of Be and Sr, and pH correlated with Be, Mg, and Sr. Gray treefrogs had significantly lower concentrations of most metals compared to northern cricket frogs, possibly because of differences in microhabitats and soil ingestion. More than half of most metals was sequestered in the gut coil of green frog tadpoles, probably mixed with soil. Depending on bioavailablity, many of the metals in gut coils and whole bodies of these tadpoles could be potentially toxic to predators.  相似文献

Purpose

The differential ability of apple snail tissues, endosymbionts, and eggs to bioaccumulate several metals (Sb, As, Ba, Br, Zn, Cr, Fe, Hg, Se, and U) was investigated.

Methods

Metal concentrations were determined by neutron activation analysis in several tissues, endosymbionts, and eggs from mature apple snails cultured in either drinking water or reconstituted water (prepared with American Society for Testing and Materials type I water).

Results

The highest bioconcentration factors (BCFs) in the midgut gland were found for Ba, Zn, Se, As, U, Br, and Hg (in decreasing order), while the highest in the kidney were for Ba, Br, and Hg. The foot showed the highest BCFs for Ba, Hg, Br, and Se (in decreasing order). Calcified tissues (uterus, shell) and eggs showed low BCFs, except for Ba. Both C corpuscles and gland tissue showed statistically higher BCFs than K corpuscles for Ba, Fe, U, Br, and Sb. The concentration of most of the studied elements was significantly lower in tissues and endosymbionts obtained from snails cultured in reconstituted water instead of drinking water. Snails cultured in reconstituted water and then exposed or not to Hg, As, and U (at the maximum contaminant level allowed by the US Environmental Protection Agency) also resulted in high levels accumulated in midgut gland, endosymbionts and kidney.

Conclusions

Our findings suggest that the midgut gland (and the symbionts contained therein), the kidney, and the foot of Pomacea canaliculata may be useful bioindicators of Hg, As and U pollution in freshwater bodies and that the unrestricted use of ampullariid snails as human and animal food must be considered with caution..  相似文献

Marine mussels (Mytilus edulis) were exposed to seawater spiked with tritiated water (HTO) at a dose rate of 122 and 79 μGy h−1 for 7 and 14 days, respectively, and tritiated glycine (T-Gly) at a dose rate of 4.9 μGy h−1 over 7 days. This was followed by depuration in clean seawater for 21 days. Tissues (foot, gills, digestive gland, mantle, adductor muscle and byssus) and DNA extracts from tissues were analysed for their tritium activity concentrations. All tissues demonstrated bio-accumulation of tritium from HTO and T-Gly. Tritium from T-Gly showed increased incorporation into DNA compared to HTO. About 90% of the initial activity from HTO was depurated within one day, whereas T-Gly was depurated relatively slowly, indicating that tritium may be bound with different affinities in tissues. Both forms of tritium caused a significant induction of micronuclei in the haemocytes of mussels. Our findings identify significant differential impacts on Mytilus edulis of the two chemical forms of tritium and emphasise the need for a separate classification and control of releases of tritiated compounds, to adequately protect the marine ecosystem.  相似文献

Mercury levels were determined in the tissues and organs (lung, liver, kidney, skin, muscle, bone) of five toothed-whales stranded along the Corsican coast between November 1993 and February 1996. The species taken into consideration were the bottlenose dolphin Tursiops truncatus, the common dolphin Delphinus delphis, the striped dolphin Stenella coeruleoalba, the pilot whale Globicephala melas and the Risso's dolphin Grampus griseus. The variation in mercury levels between the different tissues and organs (lung, liver, kidney, skin, muscle, bone) of the cetacean species are discussed as regards storage, biotransformation and elimination. In all cases, the liver appears to be the preferential organ for mercury accumulation (with concentrations as high as 4250 microg Hg/g dw and 3298 microg Hg/g in the livers of Tursiops truncatus and Grampus griseus, respectively). The kidney and lung are the next organs in terms of mercury uptake followed by the muscle, bone and skin. The stomach contents of Grampus griseus and D. delphis were determined and consisted of cephalopods for Grampus griseus, and of sardines Sardina pilchardus and mackerels Trachurus sp. for D. delphis. Cephalopods had higher mercury concentrations (25.4 microg Hg/g dw) than fish (ca 1 microg Hg/g). These contents represent only one meal and mercury levels found in livers may integrate mercury uptake having occurred during the whole life span of animals.  相似文献

In this study, the bioavailability and accumulation of mercury from external environmental exposure to mixed, cured, milled, sieved and proportioned dental amalgam was examined in the common goldfish, Carassius auratus. Fish were exposed to dental amalgam (particle size range from < 0.10 to 3.15 mm) in order to represent the particle size and distribution of that found within the typical dental office wastewater discharge stream. Experimental amalgam water loadings were 0 g/l, 0.5 g/l and 1 g/l in glass aquaria at 15 degrees C for 28 days. Fish tissues were sampled at 5 min and 28 days of exposure, and the liver, brain, muscle and whole body analyzed for total mercury using cold vapor atomic fluorescence spectroscopy. Mercury was found in several tissues examined and generally increased with exposure to higher amounts of dental amalgam. The highest levels were found in the whole body (17.68 +/- 5.73 microg/g) followed by the liver (0.80 +/- 0.16 microg/g) and muscle (0.47 +/- 0.16 microg/g). The lowest concentrations were seen in the brain (0.28 +/- 0.19 microg/g). Compared to controls, concentrations in the whole body, muscle and liver in fish exposed for 28 days to the highest concentration of amalgam were 200-, 233-, and 40-fold higher, respectively. This study shows that mercury from an environmental exposure to representative samples of dental amalgam typically found within the dental wastewater discharge stream is bioavailable to fish and may accumulate in internal tissues.  相似文献

The objective of this study was to conduct a comprehensive comparison of the effect on acetylcholinesterase (AChE) from various tissues of Mytilus trossulus caused by in vitro exposure to several pesticides and metals, because results available in the literature are inconsistent, difficult to compare, and sometimes contradict each other. For this purpose, fraction S10 extracted from gills, digestive gland, mantle and muscles, and the whole soft tissue of the mussel was exposed to several pesticides (dichlorvos, chlorpyrifos, fenitrothion, carbofuran and carbaryl) and metals (Cu, Zn, Cd, Hg, Pb) at a wide range of concentrations. AChE was inhibited in 50% or more in all the tissues exposed to dichlorvos, Cu, Hg and the mixture of Cu+Cd, and in some tissues exposed to carbaryl and carbofuran. The IC50 was calculated where possible. No inhibition was found in the case of chlorpyrifos, Cd, Pb, and Zn.  相似文献

A collaborative study was performed on scallops (Chlamys farreri) exposed to 0.5, 3 and 10 microg/L benzo[a]pyrene (B[a]P) for 20 days. The levels of aryl hydrocarbon hydroxylase (AHH) activity, B[a]P accumulation and DNA strand break were assayed in the gill and digestive gland. Results showed that AHH activity and B[a]P accumulation were significantly related to B[a]P dose. AHH activity was induced and then became stable gradually. B[a]P accumulation increased first and showed an incoming plateau. DNA strand break levels in the 0.5 and 3 microg/L B[a]P groups remained high and significantly different from control values until day 6, followed by a reduction in the gill and no recovery in the digestive gland. The 10 microg/L B[a]P groups remained significantly lower than control until the end. These results suggested that the application of comprehensive indices may give information on the actual exposure of organisms to pollutants, and also information on toxic effects.  相似文献

Uptake of waterborne Cd, Co, Mn and Zn was determined in laboratory experiments using radiotracer techniques (109Cd, 57Co, 54Mn and 65Zn). Labelled Zn was mainly accumulated in the digestive gland (65%) and Co in kidneys (81%); Cd and Mn were similarly distributed in digestive gland and gills. In a complementary field study, Ag, As, Cd, Co, Cr, Cu, Fe, Mn, Ni, and Zn were analysed in scallops collected at two stations showing different contamination levels. Digestive gland and kidneys displayed the highest concentrations. Ag, As, Cd, and Fe differed in soft tissues from the two stations, suggesting that Comptopallium radula could be a valuable local biomonitor species for these elements. Low Mn and Zn concentrations found in kidneys suggest that their content in calcium-phosphate concretions differs from the other pectinids. Preliminary risk considerations suggest that As would be the only element potentially leading to exposure of concern for seafood consumers.  相似文献

Tissue distributions and seasonal dynamics of the hepatotoxic microcystins-LR and -RR in a freshwater snail (Bellamya aeruginosa) were studied monthly in a large shallow, eutrophic lake of the subtropical China during June-November, 2003. Microcystins (MCs) were quantitatively determined by High-Performance Liquid Chromatography (HPLC) with a qualitative analysis by a Finnigan LC-MS system. On the average of the study period, hepatopancreas was the highest in MC contents (mean 4.14 and range 1.06-7.42 microg g(-1)DW), followed by digestive tracts (mean 1.69 and range 0.8-4.54 microg g(-1)DW) and gonad (mean 0.715 and range 0-2.62 microg g(-1)DW), whereas foot was the least (mean 0.01 and range 0-0.06 microg g(-1)DW). There was a positive correlation in MC contents between digestive tracts and hepatopancreas. A constantly higher MC content in hepatopancreas than in digestive tracts indicates a substantial bioaccumulation of MCs in the hepatopancreas of the snail. The average ratio of MC-LR/MC-RR showed a steady increase from digestive tracts (0.44) to hepatopancreas (0.63) and to gonad (0.96), suggesting that MC-LR was more resistant to degradation in the snail. Since most MCs were present in the hepatopancreas, digestive tracts and gonad with only a very small amount in the edible foot, the risk to human health may not be significant if these toxic parts are removed prior to snail consumption. However, the possible transference of toxins along food chains should not be a negligible concern.  相似文献
