Canada's National Agri-Environmental Standards Initiative sought to develop an environmental benchmark for low-level waterborne pathogen occurrence in agricultural watersheds. A field study collected 902 water samples from 27 sites in four intensive agricultural watersheds across Canada from 2005 to 2007. Four of the sites were selected as reference sites away from livestock and human fecal pollution sources in each watershed. Water samples were analyzed for Campylobacter spp., Salmonella spp., Escherichia coli O157:H7, Cryptosporidium spp., Giardia spp., and the water quality indicator E. coli. The annual mean number of pathogen species was higher at agricultural sites (1.54 ± 0.07 species per water sample) than at reference sites (0.75 ± 0.14 species per water sample). The annual mean concentration of E. coli was also higher at agricultural sites (491 ± 96 colony-forming units [cfu] 100 mL(-1)) than at reference sites (53 ± 18 cfu 100 mL(-1)). The feasibility of adopting existing E. coli water quality guideline values as an environmental benchmark was assessed, but waterborne pathogens were detected at agricultural sites in 80% of water samples with low E. coli concentrations (<100 cfu 100 mL(-1)). Instead, an approach was developed based on using the natural background occurrence of pathogens at reference sites in agricultural watersheds to derive provisional environmental benchmarks for pathogens at agricultural sites. The environmental benchmarks that were derived were found to represent E. coli values lower than geometric mean values typically found in recreational water quality guidelines. Additional research is needed to investigate environmental benchmarks for waterborne pathogens within the context of the "One World, One Health" perspective for protecting human, domestic animal, and wildlife health.  相似文献

The moisture and manure contents of soils at cattle feedlot surfaces vary spatiotemporally and likely are important factors in the persistence of Escherichia coli O157 in these soils. The impacts of water content (0.11-1.50 g H2O g(-1) dry feedlot surface material [FSM]) and manure level (5, 25, and 75% dry manure in dry FSM) on E. coli O157:H7 in feedlot soils were evaluated. Generally, E. coli O157:H7 numbers either persisted or increased at all but the lowest moisture levels examined. Manure content modulated the effect of water on E. coli growth; for example, at water content of 0.43 g H2O g(-1) dry FSM and 25% manure, E. coli O157:H7 increased by 2 log10 colony forming units (CFU) g(-1) dry FSM in 3 d, while at 0.43 g H2O g(-1) dry FSM and 75% manure, populations remained stable over 14 d. Escherichia coli and coliform populations responded similarly. In a second study, the impacts of cycling moisture levels and different drying rates on naturally occurring E. coli O157 in feedlot soils were examined. Low initial levels of E. coli O157 were reduced to below enumerable levels by 21 d, but indigenous E. coli populations persisted at >2.50 log10 CFU g(-1) dry FSM up to 133 d. We conclude that E. coli O157 can persist and may even grow in feedlot soils, over a wide range of water and manure contents. Further investigations are needed to determine if these variables can be manipulated to reduce this pathogen in cattle and the feedlot environment.  相似文献

In regions where animal agriculture is prominent, such as southern Alberta, higher rates of gastrointestinal illness have been reported when compared with nonagricultural regions. This difference in the rate of illness is thought to be a result of increased zoonotic pathogen exposure through environmental sources such as water. In this study, temporal and spatial factors associated with bacterial pathogen contamination of the Oldman River, which transverses this region, were analyzed using classification and regression tree analysis. Significantly higher levels of fecal indicators; more frequent isolations of Campylobacter spp., Escherichia coli O157:H7, and Salmonella enterica spp.; and higher rates of detection of pig-specific Bacteroides markers occurred at downstream sites than at upstream sites, suggesting additive stream inputs. Fecal indicator densities were also significantly higher when any one of these three bacterial pathogens was present and where there were higher total animal manure units; however, occasionally pathogens were present when fecal indicator levels were low or undetectable. Overall, Salmonella spp., Campylobacter spp., and E. coli O157:H7 presence was associated with season, animal manure units, and total rainfall on the day of sampling and 3 d in advance of sampling. Several of the environmental variables analyzed in this study appear to influence pathogen prevalence and therefore may be useful in predicting water quality and safety and in the improvement of watershed management practices in this and other agricultural regions.  相似文献

The influence of zoonotic pathogens in animal manure on human health and well-being as a direct or indirect cause of human enteric illness is examined. Available international data are considered, but the study is focused on the developing situation in western Canada, where it is certain there will be further rapid growth in livestock numbers, particularly hogs. Major pathogens considered are Escherichia coli O157:H7, Salmonella, Campylobacter, Yersinia, Cryptosporidium, and Giardia. Canada is now the leading exporter of pork internationally, but recent increases in production contrast with constant domestic levels of pork consumption and declining levels of foodborne illness caused by pork. Effects of increased levels of manure production are not quantifiable in terms of effects on human health. The presence of major pathogens in manure and movement to human food sources and water are considered on the basis of available data. Survival of the organisms in soil, manure, and water indicate significant variability in resistance to environmental challenge that are characteristic of the organisms themselves. Generally, pathogens survived longer in environmental samples at cool temperatures but differences were seen in liquid and solid manure. Based on actual data plus some data extrapolated from cattle manure environments, holding manure at 25 degrees C for 90 d will render it free from the pathogens considered above.  相似文献

Southern Alberta, which has a cold climate dominated by strong chinook winds, has the highest density of feedlot cattle in Canada. However, the quantity and quality of runoff from beef cattle (Bos taurus) feedlots in this unique region has not been investigated. Our objectives were to compare runoff quantity (1998-2002) with catch-basin design criteria; determine concentrations of selected inorganic chemical parameters (1998-2000) in runoff in relation to water quality guidelines and the potential implications of irrigating adjacent crop-land; and determine if total heterotrophs, total coliforms, and Escherichia coli (1998-2000) persisted in the catch-basin water and soil. Runoff (< 0.1 to 42.5 mm) for a 24-h duration that included maximum peak discharge was less than the recommended design criteria of 90 mm based on runoff from 24 h of rainfall with a 30-yr return period. We found that curve numbers between 52 and 96 (mode of 90) were required to match the USDA Natural Resources Conservation Service predicted runoff and actual runoff volumes. Total P posed the greatest threat to water quality guidelines, and K posed the greatest threat for exceeding crop fertilizer requirements if catch-basin effluent was used as irrigation water. Water in the catch basin had continually high populations of E. coli throughout the study, with values ranging between log10 2 and log10 8 100 mL(-1). In contrast, soil in the catch basin generally had low populations of E. coli that were < log10 2 g(-1) wet wt., but at times higher populations between log10 2 and log10 6 g(-1) wet wt. were also found.  相似文献

Swine (Sus scofa domestica) lagoon effluent is a valuable resource. In the U.S. Mid-South it is applied from April to September to fertilize grass hay in spray-irrigated fields. Lagoon levels of nutrients and bacteria, and soil levels of nutrients have been documented, but little was known of effluent bacterial levels in soil. The present study examined levels of selected effluent bacteria and nutrients in soils inside and outside spray fields after >15 yr of effluent irrigation. Samples were collected February to March 2009 from contiguous soils spanning adjacent irrigated and nonirrigated areas. Separate soil cores for bacterial and nutrient tests were collected in pairs <10 cm apart. Five cores each were collected at 15-m intervals and combined, respectively, to comprise inside and outside samples from each of 20 soils (four each from five farms/spray fields). Analyses of data combined across all soils showed higher pH and Mehlich-3-extracrable (M3-) P, Mg, K, Na, Cu, and Zn inside than outside spray fields, while total N, total C, M3-Ca, and M3-Mn did not differ. Bacterial levels were higher inside than outside spray fields for heterotrophic plate counts, thermotolerant coliforms, Staphylococcus spp., and Clostridium perfringens, but levels of Escherichia coli and Enterococcus spp. were not different. Cultural presence/absence tests for three pathogens (Listeria spp., Campylobacter spp., and Salmonella spp.) detected only Listeria spp., which did not differ inside (23% positive samples) and outside (28% positive). Molecular tests detected all three pathogens at low levels that were not different inside and outside. We found no evidence of cumulative buildup of Campylobacter spp., Listeria spp., or Salmonela s. in spray field soils.  相似文献

Contamination by bacteria is a leading cause of impairment in U.S. waters, particularly in areas of livestock agriculture. We evaluated the effectiveness of several practices in reducing Escherichia coli levels in runoff from fields receiving liquid dairy (Bos taurus) manure. Runoff trials were conducted on replicated hay and silage corn (Zea mays L.) plots using simulated rainfall. Levels of E. coli in runoff were approximately 10(4) to 10(6) organisms per 100 mL, representing a significant pollution potential. Practices tested were: manure storage, delay between manure application and rainfall, manure incorporation by tillage, and increased hayland vegetation height. Storage of manure for 30 d or more consistently and dramatically lowered E. coli counts in our experiments, with longer storage providing greater reductions. Manure E. coli declined by > 99% after approximately 90 d of storage. On average, levels of E. coli in runoff were 97% lower from plots receiving 30-d-old and > 99% lower from plots receiving 90-d-old manure than from plots where fresh manure was applied. Runoff from hayland and cornland plots where manure was applied 3 d before rainfall contained approximately 50% fewer E. coli than did runoff from plots that received manure 1 d before rainfall. Hayland vegetation height alone did not significantly affect E. coli levels in runoff, but interactions with rainfall delay and manure age were observed. Manure incorporation alone did not significantly affect E. coli levels in cornland plot runoff, but incorporation could reduce bacteria export by reducing field runoff and interaction with rainfall delay was observed. Extended storage that avoids additions of fresh manure, combined with application several days before runoff, incorporation on tilled land, and higher vegetation on hayland at application could substantially reduce microorganism loading from agricultural land.  相似文献

Current methods for tracking pathogens across farmland and into surrounding waterways via runoff are limited and typically have been developed using artificially created landscapes. No studies have investigated how Giardia in farm runoff moves across the landscape, despite high prevalence rates in dairy cattle (Bos taurus) worldwide. Here, we report the development of a field-based tracking method specific for Giardia movement in runoff and use this technique to compare the pathogen reduction capability of recently planted vegetation strips with bare soil strips cleared of vegetation. Such scenarios represent typical events in schemes to plant vegetation barriers aimed at reducing waterway contamination. A significant treatment effect was identified, with 26% fewer Giardia detected in runoff collected from the planted strip (P = 0.006). These results highlight the immediate benefit of pathogen removal to be gained from vegetation planting. The successful discrimination of treatment effects by this new technique will enable the assessment of different vegetation types on runoff reduction and the effects of plant development over time.  相似文献

The influence of aerobic and anaerobic conditions on the survival of the enteropathogens Escherichia coli O157:H7 and Salmonella serovar Typhimurium was investigated in microcosms with broth, cattle manure or slurry. These substrates were inoculated with a green fluorescent protein transformed strain of the enteropathogens at 10(7) cells g(-1) dry weight. Survival data was fitted to the Weibull model. The survival curves in aerobic conditions generally showed a concave curvature, while the curvature was convex in anaerobic conditions. The estimated survival times showed that E. coli O157:H7 survived significantly longer under anaerobic than under aerobic conditions. Survival ranged from approximately. 2 weeks for aerobic manure and slurry to more than six months for anaerobic manure at 16 °C. On average, in 56.3% of the samplings, the number of recovered E. coli O157:H7 cells by anaerobic incubation of Petri plates was significantly (p < 0.05) higher in comparison with aerobic incubation. Survival of Salmonella serovar Typhimurium was not different between aerobic and anaerobic storage of LB broth or manure as well as between aerobic and anaerobic incubation of Petri dishes. The importance of changes in microbial community and chemical composition of manure and slurry was distinguished for the survival of E. coli O157:H7 in different oxygen conditions.  相似文献

Vegetative treatment areas (VTAs) are commonly being used as an alternative method of agricultural process wastewater treatment. However, it is also apparent that to completely prevent discharge of pollutants to the surrounding environment, settling of particulates and bound constituents from overland flow through VTAs is not sufficient. For effective remediation of dissolved agricultural pollutants, VTAs must infiltrate incoming wastewater. A simple water balance model for predicting VTA soil saturation and surface discharge in landscapes characterized by sloping terrain and a shallow restrictive layer is presented and discussed. The model accounts for the cumulative effect of successive rainfall events and wastewater input on soil moisture status and depth to water table. Nash–Sutcliffe efficiencies ranged from 0.65 to 0.81 for modeled and observed water table elevations after calibration of saturated hydraulic conductivity. Precipitation data from relatively low, average, and high annual rainfall years were used with soil, site, and contributing area data from an example VTA for simulations and comparisons. Model sensitivity to VTA width and contributing area (i.e. barnyard, feedlot, silage bunker, etc.) curve number was also investigated. Results of this analysis indicate that VTAs should be located on steeper slopes with deeper, more-permeable soils, which effectively lowers the shallow water table. In sloping landscapes (>2%), this model provides practitioners an easy-to-use VTA design and/or risk assessment tool that is more hydrological process-based than current methods.  相似文献

Drinking water regulations in the United States and elsewhere are based on the occurrence of fecal indicator bacteria. Though not meeting all the criteria of an ideal indicator, nonpathogenic strains of Escherichia coli (E. coli) are used worldwide as an indicator of potential fecal contamination for drinking water and for distribution systems. This is, in part, because real illnesses are related to human pathogens, such as E. coli O157:H7, whose presence may be predicted better by E. coli than by total coliform bacteria. Our objective was to estimate the number of E. coli O157:H7 illnesses attributable to drinking water exposures in the United States and the feasible relationships between positive occurrences of the indicator bacteria E. coli and E. coli O157:H7 in drinking water. Results of the modeling indicate that in undisinfected drinking water systems, the ratio of bacterial indicator E. coli positives to E. coli O157:H7 organisms is estimated to be between 6:1 and 90:1 with few model parameters accounting for the vast majority of the uncertainty. These results provide context for considering the potential public health implications of a positive E. coli result from routine monitoring of undisinfected drinking water.  相似文献

Silage bunker runoff can be a very polluting substance and is increasingly being treated by vegetative treatment areas (VTAs), but little information exists regarding nutrient removal performance of systems receiving this wastewater. Nutrient transport through the shallow subsurface of three VTAs (i.e. one VTA at Farm WNY and two VTAs at Farm CNY) in glaciated soils containing a restrictive layer (i.e., fragipan) was assessed using a mass balance approach. At Farm WNY, the mass removal of ammonium was 63%, nitrate was 0%, and soluble reactive phosphorus (SRP) was 39%. At Farm CNY, the mass removal of ammonium was 79% in the West VTA, but nitrate and SRP increased by 200% and 533%, respectively. Mass removal of ammonium was 67% in the East VTA at Farm CNY; nitrate removal was 86% and SRP removal was 88%. The East VTA received a much higher nutrient loading, which was attributed to a malfunctioning low-flow collection apparatus within the settling basin. Results demonstrate that nutrient reduction mechanisms other than vegetative uptake can be significant within VTAs. Even though increases in nitrate mass were observed, concentrations in 1.65m deep wells indicated that groundwater impairment from leaching of nitrate was not likely. These results offer one of the first evaluations of VTAs treating silage bunker runoff, and highlight the importance of capturing concentrated low flows in VTA systems.  相似文献

ABSTRACT. Samples for water-quality analyses were collected from beneath eighty commercial cattle feedlots in the Texas High Plains. Twenty-two feedlots were drilled and/or cored to establish vertical gradients of dissolved solids. Sample and gamma logs, size analyses and vertical permeability of cores were determined from samples beneath these lots. Relationships of groundwater saturated thickness, depth to watertable, and age of lots to specificion distribution were evaluated. The study includes lots ranging in age from 35 years to new installations. Runoff collection-systems on lots include playas, man-made ponds, and dammed and undammed stream channels. Infiltration of feedlot liquid waste to the watertable below feedyards is insignificant in most localities in the Texas High Plains. Infiltration of “collected” feedlot runoff and subsequent concentration of dissolved ions in groundwater in the High Plains are dependent upon, among other things, (1) surface and subsurface geology, (2) depth to water, (3) thickness of the groundwater zone, and to (4) differences in lateral and vertical permeabilities of the Ogallala Formation, the major aquifer. Certainly, no regional subsurface pollution problem exists today nor is one foreseen from cattle feedlot runoff in the Texas High Plains.  相似文献

A study was conducted to estimate the retention efficiency of vegetative buffers for Escherichia coli deposited on grasslands in cattle fecal deposits and subject to natural rainfall-runoff conditions. The study was conducted on annual grasslands in California's northern Sierra Nevada foothills, a region with a distinct wet-dry season Mediterranean climate. We used 48, 2.0- by 3.0-m runoff plots to examine the efficacy of 0.1-, 1.1-, and 2.1-m buffers at three land slopes (5, 20, and 35%) and four dry vegetation matter levels (225, 560, 900, and 4500 kg/ha) across 27 rainfall-runoff events during two rainfall seasons. Buffer width treatments were implemented by placement of cattle fecal material containing known loads of E. coli 0.1, 1.1, or 2.1 m upslope of the plot runoff collector. Mean total runoff to total rainfall ratio per plot ranged from 0.014:1 to 0.019:1 and reflected the high infiltration capacity of these soils. Approximately 94.8 to 99.995% of total E. coli load applied to each plot appears to be either retained in the fecal pat and/or attenuated within 0.1 m downslope of the fecal pat, irrespective of the presence of a wider vegetated buffer. Relative to a 0.1-m buffer, we found 0.3 to 3.1 log10 reduction in E. coli discharge per additional meter of vegetative buffer across the range of residual dry vegetation matter levels, land slope, and rainfall and runoff conditions experienced during this project. Buffer efficiency was significantly reduced as runoff increased. These results support the assertion that grassland buffers are an effective method for reducing animal agricultural inputs of waterborne E. coli into surface waters.  相似文献

E. coli O157:H7 is a pathogen that can be present in sewage contaminated waters. This organism poses a health risk for humans who come in contact with these waters via drinking, swimming, or shellfish consumption. A risk assessment model is needed to evaluate or quantify this risk. One possibility is the use of a computer model to simulate the fate and transport of E. coli O157:H7 downstream from a discharge point [e.g., a separate sanitary sewer overflow (SSO)]. However, this computer model would require input data regarding characteristics of this organism, which have not been previously available. One necessary input parameter is the rate at which die off of this organism occurs in a stream or river environment. Several studies were conducted to evaluate the die‐off rate of E. coli O157:H7 in an SSO impacted stream. Indicator bacteria (total coliforms, E. coli, and enterococci) were evaluated simultaneously. The results suggest that E. coli O157:H7 is not persistent — decay rates are high relative to the indicator bacteria. However, the decay plots suggest a biphasic response: initial decay is rapid, followed by an attenuated, slower decay. Hence traditional simulation methods using a single, first‐order decay rate may be inaccurate.  相似文献

Nutrients, soluble salts, and pathogenic bacteria in feedlot-pen manure have the potential to cause pollution of the environment. A three-year study (1998-2000) was conducted at a beef cattle (Bos taurus) feedlot in southern Alberta, Canada to determine the effect of bedding material [barley (Hordeum vulgare L.) straw versus wood chips] and season on the chemical and bacterial properties of pen-floor manure. Manure was sampled for chemical content (N, P, soluble salts, electrical conductivity, and pH) and populations of four groups of bacteria (Escherichia coli, total coliforms, and total aerobic heterotrophs at 27 and 39 degrees C). More chemical parameters of manure were significantly (P < or = 0.05) affected by season (SO4, Na, Mg, K, Ca, sodium adsorption ratio [SAR], total C, NO3-N, NH4-N, total P, and available P) than by bedding (K, pH, total C, C to N ratio, NH4-N, and available P). Bedding had no significant (P > 0.05) effect on the four bacterial groups whereas season affected all four groups. Numbers of E. coli and total coliforms (TC) were significantly higher by 1.72 to 2.02 log10 units in the summer than the other three seasons, which was consistent with a strong positive correlation of E. coli and TC with air temperature. The low ratio of bedding to manure in the pens was probably the major cause of the lack of significant bedding effects. Bedding material and seasonal timing of cleaning feedlot pens and land application of manure may be a potential tool to manage nutrients, soluble salts, and pathogens in manure.  相似文献

A simple model predicting bathing water concentrations of Escherichia coli from livestock in the Irvine catchment in SW Scotland has been adapted for intestinal enterococci (IE). This has been used to predict risk of bather illness by extrapolation of published data on bather IE exposure vs incidence of gastro-enteritis. Simulated reduction in the risk of illness by reduced faecal loading was multiplied by a willingness to pay for risk reduction to estimate the annual benefits of mitigation. Health benefits of reducing loading by 75% at Irvine Beach were estimated by a willingness to pay method to be about pound 276k pa. Estimated annualised costs of diffuse pollution mitigation measures across the catchment were higher (> pound 1m), and it is very unlikely that 75% mitigation is achievable with current stocking rates. Further work should explore the influence of uncertainty of model parameters, and use emerging epidemiological information on specific zoonotic pathogens such as E. coli O157 and Cryptosporidium. Other components of the value of clean water should also be included to obtain a complete estimate of the cost:benefit of mitigation.  相似文献

Due to the difficulties in testing for specific pathogens, water samples are tested for the presence of nonpathogenic indicator organisms to determine whether a water supply has been contaminated by fecal material. An implicit assumption in this approach is that where pathogenic microorganisms are present fecal indicator organisms are present as well; yet surprisingly few studies have been conducted that directly compare the transport of indicator organisms with pathogenic organisms in ground water environments. In this study we compared the cell properties and transport of Escherichia coli, a commonly used indicator organism, and Campylobacter jejuni, an important enteropathogen commonly found in agricultural wastes, through saturated porous media. Differences in cell properties were determined by measuring cell geometry, hydrophobicity, and electrophoretic mobility. Transport differences were determined by conducting miscible displacement experiments in laboratory columns. Under the experimental conditions tested, C. jejuni was much more negatively charged and more hydrophobic than E. coli. In addition, C. jejuni cells were slightly longer, narrower, and less spherical than E. coli. The variations in cell properties, primarily surface charge, resulted in significant differences in transport between these two microorganisms, with the transport of C. jejuni exceeding that of E. coli when conditions favored low attachment rates, thus calling into question the usefulness of using E. coli as an indicator organism for this important pathogen.  相似文献

Erosion and runoff from pastures may lead to degradation of surface water. A 2-yr grazing study was conducted to quantify the effects of grazing management on sediment, phosphorus (P), and pathogen loading of streams in cool-season grass pastures. Six adjoining 12.1-ha pastures bisected by a stream in central Iowa were divided into three treatments: continuous stocking with unrestricted stream access (CSU), continuous stocking with restricted stream access (CSR), and rotational stocking (RS). Rainfall simulations on stream banks resulted in greater ( < 0.10) proportions of applied precipitation and amounts of sediment and P transported in runoff from bare sites than from vegetated sites across grazing treatments. Similar differences were observed comparing vegetated sites in CSU and RS pastures with vegetated sites in CSR pastures. Bovine enterovirus was shed by an average of 24.3% of cows during the study period and was collected in the runoff of 8.3 and 16.7% of runoff simulations on bare sites in CSU pastures in June and October of 2008, respectively, and from 8.3% of runoff simulations on vegetated sites in CSU pastures in April 2009. Fecal pathogens (bovine coronavirus [BCV], bovine rotavirus group A, and O157:H7) shed or detected in runoff were almost nonexistent; only BCV was detected in feces of one cow in August of 2008. Erosion of cut-banks was the greatest contributor of sediment and P loading to the stream; contributions from surface runoff and grazing animals were considerably less and were minimized by grazing management practices that reduced congregation of cattle by pasture streams.  相似文献

ABSTRACT: Measured stream discharge plus calculated ground water discharge (total measured runoff) were compared with runoff calculated by the unit-runoff method for the two largest watersheds of Mirror Lake for 1981–1983. Runoff calculated by the unit-runoff method, using Hubbard Brook watershed 3 as the index watershed, was greater than the total measured runoff into Mirror Lake during periods of high flow and slightly less than the total measured runoff into Mirror Lake during periods of low flow. Annual calculated unit runoff was 17 to 37 percent greater than total measured runoff. Differences in monthly runoff are far greater, ranging from 0 to greater than 100 percent. For high flows the calculated unit runoff is about 2 times greater than total measured runoff. For low flows the northwest basin of Mirror Lake has the greatest ground water contribution compared to the other two basins. In contrast, Hubbard Brook watershed 3 has the least ground water contribution.  相似文献
