Abstract

Telomere length was found to be associated with omethoate exposure and polymorphisms in certain genes among occupational workers. However, whether the polymorphisms in telomere-binding protein genes influence telomere length remains unclear. To explore the correlation between telomere length and polymorphisms in telomere-binding protein genes, telomere length in peripheral blood leukocytes was determined by real-time quantitative polymerase chain reaction in 180 omethoate-exposed workers and 115 healthy controls. Polymorphisms in 10 pairs of alleles were detected using flight mass spectrometry or polymerase chain reaction–restriction fragment length polymorphism technique. The results showed that individuals with GG genotype in TRF1 rs3863242 had longer telomere lengths than those with AG?+?AA genotype in the control group (p?=?0.005). The multiple regression analysis suggested that both omethoate exposure (b?=?0.526, p?<?0.001) and TRF1 rs3863242 GG (b?=?0.220, p?=?0.002) were related to a longer telomere length. In conclusion, GG genotype in TRF1 rs3863242 is linked to prolongation of telomere length, and individuals with GG genotype are recommended to strengthen health protection in a Chinese occupational omethoate-exposed population.  相似文献

Polycyclic aromatic hydrocarbon (PAH) exposure and genetic susceptibility were conductive to genotoxic effects including gene damage, which can increase mutational probability. We aimed to explore the dose-effect associations of PAH exposure with damage of exons of epidermal growth factor receptor (EGFR) and breast cancer susceptibility gene 1 (BRCA1), as well as their associations whether modified by Flap endonuclease 1 (FEN1) genotype. Two hundred eighty-eight coke oven male workers were recruited, and we detected the concentration of 1-hydroxypyrene (1-OH-pyr) as PAH exposure biomarker in urine and examined base modification in exons of EGFR and BRCA1 respectively, and genotyped FEN1 rs174538 polymorphism in plasma. We found that the damage indexes of exon 19 and 21 of EGFR (EGFR-19 and EGFR-21) were both significantly associated with increased urinary 1-OH-pyr (both Ptrend < 0.001). The levels of urinary 1-OH-pyr were both significantly associated with increased EGFR-19 and EGFR-21 in both smokers and nonsmokers (both P < 0.001). Additionally, we observed that the urinary 1-OH-pyr concentrations were linearly associated with both EGFR-19 and EGFR-21 only in rs174538 GA+AA genotype carriers (both P < 0.001). Moreover, FEN1rs rs174538 showed modifying effects on the associations of urinary 1-OH-pyr with EGFR-19 and EGFR-21 (both Pinteraction < 0.05). Our findings revealed the linear dose-effect association between exon damage of EGFR and PAH exposure and highlight differences in genetic contributions to exon damage and have the potential to identify at-risk subpopulations who are susceptible to adverse health effects induced by PAH exposure.

In the present study, we have investigated the influence of polymorphism of GSTM1 and GSTT1 genes and confounding factors such as age, sex, exposure duration and consumption habits on cytogenetic biomarkers. Frequency of sister chromatid exchanges (SCEs), high frequency cell (HFC) and cytokinesis blocked micronuclei (CBMN) were evaluated in peripheral blood lymphocytes of 115 occupationally exposed road construction workers and 105 unexposed individuals. The distribution of null and positive genotypes of glutathione-S transferase gene was evaluated by multiplex PCR among control and exposed subjects. An increased frequency of CBMN (7.03 ± 2.08); SCE (6.95 ± 1.76) and HFC (6.28 ± 1.69) were found in exposed subjects when compared to referent (CBMN - 3.35 ± 1.10; SCE - 4.13 ± 1.30 and HFC - 3.98 ± 1.56). These results were found statistically significant at p < 0.05. When the effect of confounding factors on the frequency of studied biomarkers was evaluated, a strong positive interaction was found. The individuals having GSTM1 and GSTT1 null genotypes had higher frequency of CBMN, SCE and HFC. The association between GSTM1 and GSTT1 genotypes and studied biomarkers was found statistically significant at p < 0.05. Our findings suggest that individuals having null type of GST are more susceptible to cytogenetic damage by occupational exposure regardless of confounding factors. There is a significant effect of polymorphism of these genes on cytogenetic biomarkers which are considered as early effects of genotoxic carcinogens.  相似文献

Waterpipe (aka hookah) tobacco smokers are exposed to toxicants that can lead to oxidative DNA and RNA damage, a precursor to chronic disease formation. This study assessed toxicant exposure and biomarkers of DNA [8-oxo-7, 8-dihydro-2′-deoxyguanosine (8-oxodG)] and RNA [8-oxo-7,8-dihydroguanosine (8-oxoGuo)] oxidative damage during smoking of flavored and non-flavored waterpipe tobacco. Thirty waterpipe smokers completed two counterbalanced 2-h lab waterpipe smoking sessions (flavored vs. non-flavored waterpipe tobacco). Urinary concentrations of 8-oxodG and 8-oxoGuo and expired carbon monoxide (eCO) were measured before and after the smoking sessions. A significant increase in the urinary concentrations of 8-oxodG (from 2.12 ± 0.83 to 2.35 ± 0.91 ng/mg creatinine, p = 0.024) and 8-oxoGuo (from 2.96 ± 0.84 to 3.45 ± 0.76 ng/mg creatinine, p = 0.003) were observed after smoking the non-flavored and flavored waterpipe tobacco, respectively. Our results also showed that the mean ± SD of eCO increased significantly after smoking the flavored (from 1.3 ± 1.1 to 20.3 ± 23.6 ppm, p < 0.001) and non-flavored waterpipe tobacco (from 1.8 ± 1.2 to 24.5 ± 26.1 ppm, p < 0.001). There were no significant differences in the means of 8-oxodG (p = 0.576), 8-oxoGuo (p = 0.108), and eCO (p = 0.170) between the flavored and non-flavored tobacco sessions. Smoking non-flavored and flavored waterpipe tobacco leads to oxidative stress and toxicant exposure. Our findings add to the existing evidence about the adverse effects of waterpipe tobacco smoking (WTS) and the need for strong policies to inform and protect young people from the risks of WTS.

Chronic exposure to heavy metals has long been recognized as being capable of increasing head and neck cancer (HNC) incidence, such as laryngeal (LC) and nasopharyngeal (NPC), among exposed human populations. The aim of the present study was to evaluate the concentrations of arsenic (As) and cadmium (Cd) in the blood of 145 patients (LC and NPC) and 351 controls in order to establish a potential relationship between these factors and the occurrence of LC and NPC. Mean blood levels of As and Cd in patients (5.67 and 3.51 μg/L, respectively) were significantly higher than those of controls (1.57 and 0.74 μg/L, respectively). The blood levels of As and Cd were mostly significantly higher than those of controls (p?<?0.05) after controlling the other risk factors of HNC including tobacco smoking and chewing, and alcohol drinking. Cd levels in blood increase significantly with the number of occupational exposure years for patients (p?<?0.05). However, seafood was not found to be contributing as an exposure source. Among these risk factors, smoking (>30 pack years) and occupational exposure (>20 years) presented the most significant association with HNC (OR?=?10.22 and 10.38, respectively, p?<?0.001). Cd level in blood sample of cases that are occupationally exposed/tobacco users (smokers and chewers) were higher than that of non-occupationally exposed/nontobacco users (p?<?0.001). Microbial remediation, especially the application of probiotics, has recently gained popularity in improving water quality and maintaining aquatic animal health. The efficacy and mechanism of mixed Bacillus for improvement of water quality and its effects on aquatic microbial community structure remain unknown. To elucidate these issues, we applied two groups of mixed Bacillus (Bacillus megaterium and Bacillus subtilis (A0?+?BS) and Bacillus megaterium and Bacillus coagulans (A0?+?BC)) to the aquaculture system of Crucian carp. Our results showed that the improvement effect of mixed Bacillus A0?+?BS on water quality was better than that of A0?+?BC, and the NH4+-N, NO2?-N, NO3?-N, and total phosphorus (TP) concentrations were reduced by 46.3%, 76.3%, 35.6%, and 80.3%, respectively. In addition, both groups of mixed Bacillus increased the diversity of the bacterial community and decreased the diversity of the fungal community. Microbial community analysis showed that mixed Bacillus A0?+?BS increased the relative abundance of bacteria related with nitrogen and phosphorus removal, such as Proteobacteria, Actinobacteria, Comamonas, and Stenotrophomonas, but decreased the relative abundance of pathogenic bacteria (Acinetobacter and Pseudomonas) and fungi (Epicoccum and Fusarium). Redundancy analysis showed that NH4+-N, NO2?-N, and TP were the primary environmental factors affecting the microbial community in aquaculture water. PICRUST analysis indicated that all functional pathways in the A0?+?BS group were richer than those in other groups. These results indicated that mixed Bacillus A0?+?BS addition produced good results in reducing nitrogenous and phosphorus compounds and shaped a favorable microbial community structure to further improve water quality.

This study was carried out during two consecutive seasons, 2020 and 2021, on 12-year-old mango (Mangifera indica L.). cv. Ewaise grown in region Idku, El Beheira Governorate, Egypt. The trees were planted at 5?×?4 m apart and grafted on “Sokary” root stock to study the influence of zeolite and biochar on growth, yield, and fruit quality of “Ewaise” mango cultivar irrigated by agricultural drainage water. The trees were treated by the following treatments: zeolite or biochar solely at 1, 2, and 3 kg for tree and their different combinations such as 1 kg zeolite?+?1 kg biochar; 1 kg zeolite?+?2 kg biochar; 1 kg zeolite?+?3 kg biochar; 2 kg zeolite?+?1 kg biochar; 2 kg zeolite?+?2 kg biochar; 2 kg zeolite?+?3 kg biochar; 3 kg zeolite?+?1 kg biochar; 3 kg zeolite?+?2 kg biochar; and 3 kg zeolite?+?3 kg biochar as well as control zero soil application. The obtained results showed that the soil application of zeolite or biochar gave a positive effect on improving the soil characteristics which reflects on the tree trunk thickness, shoot length and thickness, number of inflorescences, yield in kg per tree, and fruit quality. The greatest positive effect on the previous mentioned parameters was obtained by the combined application of the soil application of 2 kg zeolite?+?3 kg biochar; 2 kg zeolite?+?2 kg biochar; 3 kg zeolite?+?2 kg biochar; and 3 kg zeolite?+?3 kg biochar over the rest-applied treatments or control in the two seasons.

The paper reports the kinetics and adsorption isotherm modeling for imidacloprid (IMIDA) and azoxystrobin (AZOXY) in rice straw (RS)/corn cob (CC) and peat (P)/compost (C) based biomixtures. The pseudo-first-order (PFO), pseudo-second-order (PSO), Elovich and intraparticle diffusion models were used to describe the kinetics. The adsorption data were subjected to the Langmuir and the Freundlich isotherms. Results (r2Adj values) suggested that the modified Elovich model was the best suited to explain the kinetics of IMIDA sorption while different models explained AZOXY sorption kinetics in different biomixtures (PFO in RS?+?C and RS?+?P; PSO in CC?+?P and Elovich in CC?+?C). Biomixtures varied in their capacity to adsorb both pesticides and the adsorption coefficient (Kd) values were 116.8–369.24 (AZOXY) and 24.2–293.4 (IMIDA). The Freundlich isotherm better explained the sorption of both pesticides. Comparison analysis of linear and nonlinear method for estimating the Freundlich adsorption constants was made. In general, r2Adj values were higher for the nonlinear fit (AZOXY?=?0.938–0.982; IMIDA?=?0.91–0.970) than the linear fit (AZOXY?=?0.886–0.993; IMIDA?=?0.870–0.974) suggesting that the nonlinear Freundlich equation better explained the sorption. The rice straw-based biomixtures performed better in adsorbing both the pesticides and can be used in bio-purification systems.  相似文献

A post-harvest experiment was conducted further to our previous greenhouse pot study on upland kangkong (Ipomoea aquatica Forsk.) and Alfred stonecrop (Sedum alfredii Hance) intercropping system in Cd-contaminated soil inoculated with arbuscular mycorrhizal (AM) fungi. Previously, four treatments were established in the intercropping experiment, including monoculture of kangkong (control), intercropping with stonecrop (IS), and IS plus inoculation with Glomus caledonium (IS?+?Gc) or Glomus versiforme (IS?+?Gv). Both kangkong and stonecrop plants were harvested after growing for 8 weeks. Then, the tested soils were reclaimed for growing post-harvest kangkong for 6 weeks. In the post-harvest experiment, there were no significant differences between the IS and control treatments, except for a significantly decreased (p?<?0.05) soil available P concentration with IS treatment. Compared with IS, both IS?+?Gc and IS?+?Gv significantly decreased (p?<?0.05) soil DTPA-extractable (phytoavailable) Cd concentrations, but not total Cd, by elevating soil pH, causing significantly lower (p?<?0.05) Cd concentrations in both the root and shoot of kangkong. In addition, both Gc and Gv significantly increased (p?<?0.05) soil acid phosphatase activities and available P concentrations and hence resulted in significantly higher (p?<?0.05) plant P acquisitions. However, only Gv significantly increased (p?<?0.05) kangkong yield, while Gc only significantly elevated (p?<?0.05) the shoot P concentration. It suggested that AM fungi have played key roles in Cd stabilization and P mobilization in the intercropping system, and such positive responses seemed to be sustainable and valuable in post-harvest soils.  相似文献

To assess the effect of coumaphos [O-(3-chloro-4-methyl-2-oxo-2H-1-benzopyran-7-yl) O,O-diethyl phosphorothioate] exposure on physiological responses during bovine production, acetylcolinesterase (AChE) and butyrylcholinesterase (BuChE) activities were measured in whole blood, erythrocytes, and plasma of healthy male steers (Bos Taurus x Bos indicus) sprayed with coumaphos at a non-lethal dose of 1 mg kg? 1 body weight per day once every 14 (in vivo group) or 21 days (southern and central groups). Coumaphos topically administered at 1 mg/kg body weight per day to cattle under normal management practices in tropical areas produced a significant inhibition in erythrocyte (RBC) AChE and BuAChE activities when compared to baseline levels. RBC-AChE activity for the in vivo group decreased 71.3% (P < 0.05) and BuChE activity 59.1% (P < 0.05); RBC-AChE activity decreased 55.1% (P < 0.05) (southern group) and 43.4% (P < 0.05) (central group). Compared to the control specimens, steers from in vivo, southern, and central groups after 150 days of exposure had lower (P < 0.05) leukocyte count, absolute lymphocyte, erythrocyte, and platelet counts. Decreases in RBC-AChE activities correlated with decreased lymphocyte (r = 1.000, p = 0.01), erythrocyte (r = 1.000, p = 0.003), and platelet counts (r = 0.841, p = 0.036). Significantly increased BUN levels (P < 0.05) correlated with the decrease in RBC-AChE activities (r = ? 0.997, p = 0.047) and with the decrease in absolute red blood cell (r = ? 0.883, p = 0.020) and lymphocyte (r = ? 0.825, p = 0.043) counts; increased (P < 0.05) total plasma protein levels correlated with the decrease in RBC-AChE activities (r = ?0.998, p = 0.043), absolute red blood cell (r = ? 0.998, p = 0.040), lymphocyte (r = ? 0.893, p = 0.017), and platelet (r = -0.855, p = 0.030) counts. The physiological responses correlated with the erythrocyte acetylcholinesterase inhibition could be considered as early indicators or warning responses of bovine exposures to organophosphorus pesticides (OPs).  相似文献

The synthetic growth-promoting hormones trenbolone and melengestrol acetate have been detected in the environment near beef cattle feedlots and are reportedly transported via wind-borne particulate matter. Therefore, movement of synthetic hormones from beef cattle feedlots to water bodies via particulate matter is possible. Our objective was to evaluate potential effects of 17α-trenbolone (17α-TB), melengestrol acetate (MGA), and combinations of both on growth, development, and survival of Xenopus laevis larvae. On post-hatch day 2 (stage 33/34), X. laevis larvae were exposed to three nominal concentrations of 17α-TB (10, 100, and 500 ng/L), MGA (1, 10, and 100 ng/L), a combination of both (1/10, 10/100, and 100/500 ng/L MGA/17α-TB), frog embryo teratogenesis assay-Xenopus medium, or a solvent control. Significant increases in all X. laevis growth metrics were observed among larvae in the 1 ng/L MGA?+?10 ng/L 17α-TB and 10 ng/L MGA?+?100 ng/L 17α-TB treatments. Stage of development was increased among larvae in the 1 ng/L MGA?+?10 ng/L 17α-TB treatment group and significantly decreased among those in the 500 ng/L 17α-TB treatment. Total body mass and snout–vent length of X. laevis larvae were significantly reduced in the 100 ng/L MGA and 100 ng/L MGA?+?500 ng/L 17α-TB treatment groups. Larvae exposed to 500 ng/L 17α-TB had decreased total body mass, snout–vent length, and total length. In general, growth measurements decreased with increasing concentration of MGA, 17α-TB, or a combination of both. Survival among all treatments was not significantly different from controls. Amphibians exposed to MGA and 17α-TB in the environment may experience alterations in growth and development.  相似文献

We evaluated 41 rural workers occupationally exposed to pesticides and 32 subjects as a control group, using the micronucleus (MN) and the comet assay. For the comet assay, we evaluated the peripheral blood, and for the MN, we sampled cells from the oral epithelium. Damage to DNA was measured by tail length, % DNA in tail (% tail), olive tail moment (OTM), and tail moment (TM). The exposed group presented an 8× increase in MN frequency, when compared to the control group (p <0.05). When we contrasted the MN frequencies between the individuals that use and do not use personal protective equipment, we found a mean of 7.5 MN (57 % variance) and 12.1 MN (130 % variance), respectively. The binucleated cells were 0.04 and 0.005, in the exposed and control groups, respectively, indicating 8× increase in the number of binucleated cells, when comparing the groups (p <0.05). In the comet assay, we demonstrated statistically significant differences in three parameters (% DNA, OTM, and TM) indicating that the rural workers presented high levels of genomic damages. Our results indicate that occupational exposure to pesticides could cause genome damage in somatic cells, representing a potential health risk to Brazilian rural workers that deal constantly with agrochemicals without adequate personal protection equipment.  相似文献

The study of lead exposure among workers in Selangor and the Federal Territory was carried out based on the delta-aminolevulinic acid (ALA) level in urine. Occupations which are expected to have higher lead exposure were chosen in this research. The ALA level in the workers' urine was linked to a few variables which may contribute to the lead level in the body. The result of this study showed that the ALA level of the urine of university students (0.352 +/- 0.038 mg/100 ml) < clerical staff (0.560 +/- 0.043 mg/100 ml) < traffic police (0.612 +/- 0.064 mg/100 ml) < vehicle workshop workers (0.673 +/- 0.099 mg/100 ml) < petrol kiosk workers (0.717 +/- 0.069 mg/100 ml) < bus drivers/conductors (0.850 +/- 0.055 mg/100 ml) which was similar to workers in the printing industry (0.852 +/- 0.110 mg/100 ml). The ALA levels in the urine of the exposed workers were significantly different from the control group (university students). However, results obtained from clerical staff revealed that they were also in the exposed group category. Analysis of variance showed that the exposed groups are in a population which is different from the control population. Correlation tests suggest that there is no significant connection between the ALA level in the urine and the variables tested. Furthermore, Duncan's Multiple Range Test showed no significant differences between the smoking/non smoking group, alcoholic/non-alcoholic group, race and sex (p > 0.05).  相似文献

Considering its richness in organic and inorganic mineral nutrients, the recycling of sewage sludge (SS) is highly considered as a soil supplement in agriculture. However, the fate of hazardous heavy metal accumulation in the crops cultivated in SS amended soils is always a source of concern. Since nanoparticles are widely recognized to reduce heavy metal uptake by crop plants; thus, the present experiment deals with okra (Abelmoschus esculentus L. Moench) cultivation under the combined application of SS and TiO2-nanoparticles (NPs). Triplicated pot experiments were conducted using different doses of SS and TiO2-NPs such as 0 g/kg SS (control), 50 g/kg SS, 50 g/kg SS?+?TiO2, 100 g/kg SS, and 100 g/kg SS?+?TiO2, respectively. The findings of this study indicated that among the doses of treatment combinations investigated, 100 g/kg SS?+?TiO2 showed a significant (p?<?0.05) increase in the okra plant yield (287.87?±?4.06 g/plant) and other biochemical parameters such as fruit length (13.97?±?0.54 cm), plant height (75.05?±?3.18 cm), superoxide dismutase (SOD: 110.68?±?3.11 μ/mg), catalase (CAT: 81.32?±?3.52 μ/mg), and chlorophyll content (3.12?±?0.05 mg/g fwt.). Also, the maximum contents of six heavy metals in the soil and cultivated okra plant tissues (fruit, stem, and root regions) followed the order of Fe?>?Mn?>?Cu?>?Zn?>?Cr?>?Cd using the same treatment. Bioaccumulation and health risk assessment indicated that foliar application of TiO2-NPs significantly reduced the fate of heavy metal accumulation under higher doses of SS application. Therefore, the findings of this study suggested that the combined use of SS and TiO2-NPs may be useful in ameliorating the negative consequences of heavy metal accumulation in cultivated okra crops.

Dental amalgam in fillings exposes workers to mercury. The exposure to mercury was investigated among 1871 dental health care workers. The aim of the study was to evaluate the risk of mercury exposure among dental compared to nondental health care workers and to determine other risk factors for mercury exposure. Respondents answered questionnaires to obtain demographic, personal, professional, and workplace information and were examined for their own amalgam fillings. Chronic mercury exposure was assessed through urinary mercury levels. In total, 1409 dental and 462 nondental health care workers participated in the study. Median urine mercury levels for dental and nondental health care workers were 2.75 μg/L (interquartile range [IQR] = 3.0175) and 2.66 μg/L (IQR = 3.04) respectively. For mercury exposure, there were no significant risk factor found among the workers involved within the dental care. The Mann–Whitney test showed that urine mercury levels were significantly different between respondents who eat seafood more than 5 times per week compared to those who eat it less frequently or not at all (p = 0.003). The urinary mercury levels indicated significant difference between dental workers in their practice using squeeze cloths (Mann–Whitney test, p = 0.03). Multiple logistic regression showed that only the usage of cosmetic products that might contain mercury was found to be significantly associated with the urinary mercury levels (odds ratio [OR] = 15.237; CI: 3.612–64.276). Therefore, mean urinary mercury levels of health care workers were low. Exposure to dental amalgam is not associated with high mercury exposure. However, usage of cosmetic products containing mercury and high seafood consumption may lead to the increase of exposure to mercury.

Implications: Exposure to the high levels of mercury from dental amalgam can lead to serious health effects among the dental health care workers. Nationwide chronic mercury exposure among dental personnel was assessed through urinary mercury levels. Findings suggest low urinary mercury levels of these health care workers. Exposure to dental amalgam is not associated with high mercury exposure. However, the usage of cosmetic products containing mercury and high seafood consumption may lead to the increase of exposure to mercury.  相似文献

Polychlorinated biphenyls (PCBs) contaminate 19% of US Superfund sites and represent a serious risk to human and environmental health. One promising strategy to remediate PCB-contaminated sediments utilizes organohalide-respiring bacteria (OHRB) that dechlorinate PCBs.

However, functional genes that act as biomarkers for PCB dechlorination processes (i.e., reductive dehalogenase genes) are poorly understood. Here, we developed anaerobic sediment microcosms that harbor an OHRB community dominated by the genus Dehalococcoides. During the 430-day microcosm incubation, Dehalococcoides 16S rRNA sequences increased two orders of magnitude to 107 copies/g of sediment, and at the same time, PCB118 decreased by as much as 70%. In addition, the OHRB community dechlorinated a range of penta- and tetra-chlorinated PCB congeners including PCBs 66, 70?+?74?+?76, 95, 90?+?101, and PCB110 without exogenous electron donor. We quantified candidate reductive dehalogenase (RDase) genes over a 430-day incubation period and found rd14, a reductive dehalogenase that belongs to Dehalococcoides mccartyi strain CG5, was enriched to 107 copies/g of sediment. At the same time, pcbA5 was enriched to only 105 copies/g of sediment. A survey for additional RDase genes revealed sequences similar to strain CG5’s rd4 and rd8. In addition to demonstrating the PCB dechlorination potential of native microbial communities in contaminated freshwater sediments, our results suggest candidate functional genes with previously unexplored potential could serve as biomarkers of PCB dechlorination processes.

Evidence has shown that polymorphisms of various genes known to be involved in estrogen biosynthesis and function are associated with estrogen-dependent diseases (EDDs). These genes include CYP17A1, estrogen receptor 1 (ESR1), and 2 (ESR2). Phthalates are considered estrogenic endocrine disruptors, and recent research has suggested that they may act as a risk factor for EDDs. However, extremely few studies have assessed the effects of gene–environment interaction on these diseases. We recruited 44 patients with endometriosis or adenomyosis, 36 patients with leiomyoma, and 69 healthy controls from a medical center in Taiwan between 2005 and 2007. Urine samples were collected and analyzed for seven phthalate metabolites using liquid chromatography tandem mass spectrometry. Peripheral lymphocytes were used for DNA extraction to determine the genotype of CYP17A1, ESR1, and ESR2. Compared to controls, patients with leiomyoma had significantly higher levels of total urinary mono-ethylhexyl phthalate (ΣMEHP) (52.1 vs. 29.6 μg/g creatinine, p?=?0.040), mono-n-butyl phthalate (MnBP) (75.4 vs. 51.3 μg/g creatinine, p?=?0.019), and monoethyl phthalate (MEP) (103.7 vs. 59.3 μg/g creatinine, p?=?0.031). In contrast, patients with endometriosis or adenomyosis showed a marginally increased level of urinary MEHP only. Subjects who were homozygous for both the ESR1 C allele (rs2234693) and CYP17A1 C allele (rs743572) showed a significantly increased risk for leiomyoma (OR?=?19.8; 95 % CI, 1.70; 231.5; p?=?0.017) relative to subjects with other genotypes of ESR1 and CYP17A1. These results were obtained after adjusting for age, cigarette smoking, MEHP level, GSTM1 genotype and other covariates. Our results suggested that both CYP17A1 and ESR1 polymorphisms may modulate the effects of phthalate exposure on the development of leiomyoma.  相似文献

Abstract

This study paper reports on two cases of poisoning with the organophosphorus insecticides, fenthion and omethoate. The two victims were admitted in the Intensive Care Unit (ICU) a few hours after ingestion of the two insecticides. They received appropriate treatment for organophosphorous poisoning (gastric lavage, activated charcoal, atropine and pralidoxime) and supportive care. Both patients survived. Organophosphate blood levels were determined on admission (fenthion 2.9 (ig/ml, omethoate 1.6 Hg/ml) and during the hospitalisation and proved to be considerably high. Slow elimination rate of the poison already distributed in the body was indicated for both pesticides. The patient with omethoate poisoning remained clinically well (Glasgow Coma Scale : 15) and was discharged three days later. The patient with fenthion poisoning, who had also ingested 30 mg of bromazepam and 720 mg of oxetoron, developed cholinergic crisis six hours after admission and was intubated for 24 days, with concomitant complications.  相似文献

The combination of zero-valent iron (ZVI) with anaerobic sludge for enhancing reductive transformation and dechlorination of p-chloronitrobenzene (p-ClNB) was investigated in this study. p-ClNB was quickly reduced into p-chloroaniline (p-ClAn) and subsequently dechlorinated into aniline in the complex system, and the strengthening factor for pseudo-first-order transformation rate constant of p-ClNB (Q, k ZVI?+?sludge/(k sludge?+?k ZVI)) was above 3. The Q values for the different ZVI types with anaerobic sludge were as following: Reduced ZVI (RZVI)?>?Industrial ZVI?>?Nanoscale ZVI (NZVI). Thereinto, the aggregation of NZVI occurred, and its reaction activity declined. Furthermore, the increase of ZVI dosage promoted the p-ClNB transformation, but the p-ClAn dechlorination rate and Q value were not improved. With the anaerobic biomass increasing, the dechlorination rate of p-ClAn was significantly enhanced, and the Q value had positive relation with the mass ratio of anaerobic sludge to RZVI.  相似文献
