Some experiments on phosphate assimilation by coastal marine plankton   总被引:2,自引:0,他引:2  
A study of phosphate assimilation by coastal marine plankton revealed that both phytoplankton and microheterotrophs incorporated radioactive phosphorus (33P). Size fractionation of the particulate matter (using 1 m pore diameter Nucleopore® membrane filters), antibiotic treatment (using garamycin), and independent estimaties of photoautotrophic (14CO2 uptake) and heterotrophic (3H-glucose uptake) activities were employed to separate phyto- and bacterioplankton phosphate uptake. Results indicated that phytoplankton 33P-uptake was best estimated by the fraction of particulate matter retained on the 1 m membrane filters. Usually, less than 10% of the phytoplankton (based on chlorophyll a measurements) passed the 1 m pore-diameter filters, whereas about 90% of the heterotrophic activity passed. At least 50% of the 33P-uptake was associated with the <1 m fraction. It may be possible to resolve the phytoplankton and bacterial contributions to 33P-uptake by comparing the percent of total 33P-uptake with the percent of total 3H-glucose uptake associated with the >1 m fraction.  相似文献

In order to determine whether phytoplankton growth rates were normal or depressed, total plant carbon (g l–1) and in situ production rates (g C l–1 d–1) were measured for phytoplankton assemblages at Weathership Station P (50°N; 145°W) and at 53°N; 145°W in the subarctic Pacific in May and August 1984. Plant carbon, estimated from cell volumes determined using epifluorescence microscopy, was distributed as follow: 28% in the <2 m fraction, 38% in the 2 to 5 m size fraction, and the remainder in size classes >5 m. Carbon-specific growth rates (k), as doublings d–1, were calculated for the phytoplankton assemblages as a whole at each sampling depth down to 100 m for three days in May and for four days in August. The populations in the upper part of the euphotic zone showed average doubling rates of 1 d–1 and thus appeared to be growing at rates normally expected for the prevailing conditions of light and temperature. The low chlorophyll concentrations (0.3 to 0.4 mg chl a m–3) characteristically found in this oceanic region do not seem to be due to very slow growth of algal populations.Contribution No. 1695 of the School of Oceanography, University of Washington, Seattle, Washington 98195, USA  相似文献

Michaelis-Menten uptake kinetics were observed at all light intensities. With constant illumination, the Vmax and K1 in nitrate uptake over the natural light intensity range of 0 to 2000 E were 0.343 g-at NO3–N(g)-1 at protein-N h-1 and 26 E, respectively. Nitrate uptake was inhibited at higher light intensities. The Ks for nitrate uptake did not vary as a function of light intensity remaining relatively constant at 0.62 g-at NO3–N 1-1. With intermittent illumination, the Vmzx for light intensity in nitrate uptake over a light intensity range of 0 to 5000 E was 0.341 g-at NO3–N(g)-1-at protein-N h-1. No inhibition of nitrate uptake was observed at higher than natural light intensities. Chaetoceros curvisetus will probably never experience light inhibition of nitrate uptake under natural conditions.  相似文献

Juveniles of the prawnPenaeus chinensis (3.96 ±0.18 cm, 0.36±0.06 g) reared in Taiwan in 1989 were exposed to different concentrations of ammonia and nitrite, by a static renewal method in 33 seawater at pH 7.94 and at 26 °C. The 24, 48, 96 and 120 h LC50 (median lethal concentration) of ammonia were 3.29, 2.10, 1.53 and 1.44 mg l–1 for NH3-N (un-ionized ammonia as nitrogen) and 79.97, 51.14, 37.00 and 35.09 mg l–1 for ammonia-N (un-ionized plus ionized ammonia as nitrogen). The 24, 96, 120, 144 and 192 h LC50 of nitrite-N were 339, 37.71, 29.18, 26.98 and 22.95 mg l–1. The LC50 decreased with increasing exposure time. During the first 96 h,P. chinesis juveniles were more susceptible to ammonia than nitrite. However, prawns were less tolerant to nitrite than ammonia when exposed for more than 96 h. The threshold was found at 120 and 192 h for ammonia and nitrite, respectively, on the toxicity curves. Incipient LC50 was 1.44 mg l–1 for NH3-N, 35.09 mg l–1 for ammonia-N and 22.95 mg l–1 for nitrite-N. The safe value forP. chinensis juveniles was 0.14, 3.51 and 2.30 mg l–1, respectively.  相似文献

The vertical distribution, diel gut pigment content and oxygen consumption of Calanus euxinus were studied in April and September 1995 in the Black Sea. Gut pigment content of C. euxinus females was associated with diel vertical migration of the individuals, and it varied with depth and time. Highest gut pigment content was observed during the nighttime, when females were in the chlorophyll a (chl a) rich surface waters, but significant feeding also occurred in the deep layer. Gut pigment content throughout the water column varied from 0.8 to 22.0 ng pigment female–1 in April and from 0.2 to 21 ng pigment female–1 in September 1995. From the diel vertical migration pattern, it was estimated that female C. euxinus spend 7.5 h day–1 in April and 10.5 h day–1 in September in the chl a rich surface waters. Daily consumption by female C. euxinus in chl a rich surface waters was estimated by taking into account the feeding duration and gut pigment concentrations. Daily carbon rations of female C. euxinus, derived from herbivorous feeding in the euphotic zone, ranged from 6% to 11% of their body carbon weight in April and from 15% to 35% in September. Oxygen consumption rates of female and copepodite stage V (CV) C. euxinus were measured at different temperatures and at different oxygen concentrations. Oxygen consumption rates at oxygen-saturated concentration ranged from an average of 0.67 g O2 mg–1 dry weight (DW) h–1 at 5°C to 2.1 g O2 mg–1 DW h–1 at 23°C for females, and ranged from 0.48 g O2 mg–1 DW h–1 at 5°C to 1.5 g O2 mg–1 DW h–1 at 23°C for CVs. The rate of oxygen consumption at 16°C varied from 0.62 g O2 mg–1 DW h–1 at 0.65 mg O2 l–1 to 1.57 g O2 mg–1 DW h–1 at 4.35 mg O2 l–1 for CVs, and from 0.74 g O2 mg–1 DW h–1 at 0.57 mg O2 l–1 to 2.24 g O2 mg–1 DW h–1 at 4.37 mg O2 l–1 for females. From the oxygen consumption rates, daily requirements for the routine metabolism of females were estimated, and our results indicate that the herbivorous daily ration was sufficient to meet the routine metabolic requirements of female C. euxinus in April and September in the Black Sea.Communicated by O. The life-history of the crown-of thorns starfish (Acanthaster planci) includes a planktotrophic larva that is capable of feeding on particulate food. It has been proposed, however, that particulate food (e.g. microalgae) is scarce in tropical water columns relative to the nutritional requirements of the larvae of A. planci, and that periodic shortages of food play an important role in the biology of this species. It has also been proposed that non-particulate sources of nutrition (e.g. dissolved organic matter, DOM) may fuel part of the nutritional requirements of the larval development of A. planci as well. The present study addresses the ability of A. planci larvae to take up several DOM species and compares rates of DOM uptake to the energy requirements of the larvae. Substrates transported in this study have been previously reported to be transported by larval asteroids from temperate and antarctic waters. Transport rates (per larval A. planci) increased steadily during larval development and some substrates had among the highest mass-specific transport rates ever reported for invertebrate larvae. Maximum transport rates (J max in) for alanine increased from 15.5 pmol larva–1 h–1 (13.2 pmol g–1 h–1) for gastrulas (J max in=38.7 pmol larva–1 h–1 or 47.4 pmol g–1 h–1) to 35.0 pmol larva–1 h–1 (13.1 pmol g–1 h–1) for early brachiolaria (J max in just prior to settlement=350.0 pmol larva–1 h–1 or 161.1 pmol g–1 h–1) at 1 M substrate concentrations. The instantaneous metabolic demand for substrates by gastrula, bipinnaria and brachiolaria stage larvae could be completely satisfied by alanine concentrations of 11, 1.6 and 0.8 M, respectively. Similar rates were measured in this study for the essential amino acid leucine, with rates increasing from 11.0 pmol larva–1 h–1 (or 9.4 pmol g–1 h–1) for gastrulas (J max in=110.5 pmol larva–1 h–1 or 94.4 pmol g–1 h–1) to 34.0 pmol larva–1 h–1 (or 13.0 pmol g–1 h–1) for late brachiolaria (J max in=288.9 pmol larva–1 h–1 or 110.3 pmol g–1 h–1) at 1 M substrate concentrations. The essential amino acid histidine was transported at lower rates (1.6 pmol g–1 h–1 at 1 M for late brachiolaria). Calculation of the energy contribution of the transported species revealed that larvae of A. planci can potentially satisfy 0.6, 18.7, 29.9 and 3.3% of their total energy requirements (instantaneous energy demand plus energy added to larvae as biomass) during embryonic and larval development from external concentrations of 1 M of glucose, alanine, leucine and histidine, respectively. These data demonstrate that a relatively minor component of the DOM pool in seawater (dissolved free amino acids, DFAA) can potentially provide significant amounts of energy for the growth and development of A. planci during larval development.  相似文献

The photosynthesis–irradiance response of Ecklonia radiata (C. Agardh) J. Agardh, a common kelp in the temperate southern hemisphere, was investigated in situ throughout the year and across a depth profile at West Island, South Australia. Temperature and irradiance environment altered throughout the year, varying at 3 m between 14–20°C and 279–705 mol photons m–2 s–1. Photosynthetic capacity (Pm) varied throughout the year between 177–278 mol O2 g–1 dry wt h–1 at 3 m and 133–348 mol O2 g–1 dry wt h–1 at 10 m. The irradiance required for sub-saturation of photosynthesis (Ek) varied between 97–152 and 81–142 mol photons m–2 s–1 for 3 m and 10 m respectively, and the respiration rate varied between 15–36 and 13–20 mol O2 g–1 dry wt h–1 for 3 m and 10 m. A clear seasonal change in photokinetic parameters was detected and provided strong evidence for a seasonal acclimation response. During winter an increase in the efficiency of light utilisation at low irradiance () was accompanied by a decrease in both Ek and that required for photosynthetic compensation. Pm also increased during the winter and autumn months and respiratory requirements decreased. These changes enable E. radiata to display an optimal photosynthetic performance throughout the year despite significant changes in the surrounding environment.Communicated by P.W. Eelgrass (Zostera marina L.) has access to nutrient pools in both the water column and sediments. We investigated the potential for eelgrass to utilize nitrate nitrogen by measuring nitrate reductase (NR) activity with an in vivo tissue assay. Optimal incubation media contained 60 mM nitrate, 100 mM phosphate, and 0.5% 1-propanol at pH 7.0. Leaves had significantly higher NR activity than roots (350 vs 50 nmoles NO 2 produced g FW–1 h–1). The effects of growing depth (0.8 m MLW, 1.2 m, 3.0 m, 5.0 m) and location within the eelgrass meadow (patch edge vs middle) on NR activity were examined using plants collected from three locations in the Woods Hole area, Massachusetts, USA, in July 1987. Neither depth nor position within the meadow appear to affect NR activity. Nitrate enrichment experiments (200 M NO 3 for 6 d) were conducted in the laboratory to determine if NR activity could be induced. Certain plants from shallow depth (1.2 m) showed a significant response to enrichment, with NR activity increasing from >100 up to 950 nmoles NO 2 g FW–1 h–1 over 6 d. It appears that Z. marina growing in very shallow water (0.8 m) near a shoreline may be affected by ground water or surface run-off enrichments, since plants from this area exhibited rates up to 1 600 nmol NO 2 g FW–1 h–1. Water samples from this location consistently had slightly higher NO 3 concentrations (1.4 M) than all other collection sites (0.7 M). Thus, it is possible that chronic run-off or localized groundwater inputs can create sufficient NO 3 enrichment in the water column to induce nitrate reductase activity in Zostera leaves.  相似文献

The kinetic response of ammonium- or silicate-limited and ammonium- or silicatestarved populations of Chaetoceros debilis, Skeletonema costatum, and Thalassiosira gravida was determined by a single addition of the limiting nutrient to a steady-state culture and subsequent monitoring of the nutrient disappearance of the limiting and non-limiting nutrients at frequent time intervals. The kinetic response of nonlimited (nutrient) populations of these three species was also determined. Three distinct modes of the uptake of the limiting nutrient were observed for ammonium-or silicate-limited populations of these three species, surge uptake (V s ), internally (cellular) controlled uptake (V i ), and externally (ambient limiting nutrient concentration) controlled uptake (V e ). Non-limited populations did not exhibit the three distinct segments of uptake, V s , V i and V e . Estimates of the maximal uptake rate (V max) and the Michaelis constant (K s ) were obtained from nutrient-limited populations during the V e segment of the uptake curve. Pooled values of V e for the three ammonium-limited populations yielded V max and K s estimates of 0.16 h-1 and 0.5 g-at NH4–N l-1. Kinetic data derived from the V e segment of the uptake curve for silicate-limited populations yielded different values of V max and K s for each of the three species. In a number of parameters that were measured, T. gravida was clearly different from C. debilis and S. costatum and its recovery from nutrient starvation was the slowest. Recovery of all species from silicate limitation or starvation was slower than from ammonium limitation or starvation. Ammonium-starved populations maintained a maximal uptake rate at a substrate concentration an order of magnitude lower (0.1 g-at NH4–N l-1) than that observed for NH4-limited populations (1.0 g-at NH4–N l-1). Adaptation to the severity of the nutrient limitation occurred as changes in the magnitude of cellular characteristics, such as short-term uptake potential (V s ) and affinity for the substrate (K s ). Acute toxicity of ammonia was determined for cultured larval, postlarval, and wild adult lobsters (Homarus americanus) in 1988. Ammonia tolerance was found to increase with ontogenetic development. Based on 96-h LC50 values of 58 mg l–1 NH4 + + NH3 l–1 seawater (0.72 mg NH3 l–1) for Stage I larvae, 87 mg NH4 + + NH3 l–1 (1.7 mg NH3 l–1) for Stage II larvae, 125 mg NH4 + + NH3 l–1 (2.13 mg NH3) for Stage III larvae, 144 mg NH4 + + NH3 l–1 (2.36 mg NH3 l–1) for Stage IV postlarvae, 377 mg NH4 + + NH3 l–1 (5.12 mg NH3 l–1) for adult lobsters at 5°C and 219 mg NH4 + + NH3 l–1 (3.25 mg NH3 l–1) for adult lobsters at 20°C, recommendations for safe levels of total ammonia and un-ionized ammonia were calculated using an application factor of 0.1. Effects of ammonia on osmoregulatory capacity were studied on postlarvae and adults. Ability of postlarvae and adults to hyper-regulate in low-salinity media decreased after exposure to ammonia. In postlarval lobsters, osmoregulatory capacity was significantly affected in ammonia concentrations exceeding 32 mg l–1. Osmoregulatory capacity in adult lobsters (5 and 20°C) was affected at 150 mg l–1. In postlarval lobsters, a minimum exposure time of 12 h was required to impair osmoregulatory capacity. The decrease in hemolymph osmotic pressure was caused by lower hemolymph sodium concentrations. The presence of ammonia in the external medium could markedly affect the Na+/NH4 + transport mechanism by permanently, temporarily, or partially impairing the transport sites for sodium.  相似文献

The hydrothermal vent crab Bythograea thermydron is exposed to high environmental concentrations of sulfide and low levels of oxygen for extended periods of time. It has previously been shown that hydrogen sulfide is oxidized to the relatively non-toxic thiosulfate (S2O 3 2– ), which accumulates in the hemolymph. Hemolymph thiosulfate levels in freshly captured crabs vary significantly among crabs from different hydrothermal vent sites as well as between crabs from different microhabitats within the same site. Hemolymph thiosulfate concentrations were not significantly different between crabs captured at the same site 6 mo apart. Hemolymph thiosulfate concentrations ranged from 66 mol 1–1 in a crab captured at a site with relatively low sulfide venting, to 3206 mol 1–1 in an individual that was netted from an active smoker vent with much higher sulfide exposure. The differences in hemolymph thiosulfate between sites and the stability of hemolymph thiosulfate in crabs captured at the same site at different times suggest that sulfide exposure is significantly different between sites and that this exposure may not vary significantly over the course of a few months. B. thermydron experimentally exposed to sulfide had high levels of thiosulfate in their hemolymph and increased abilities to regulate oxygen consumption in conditions of low oxygen. This enhancement of regulatory abilities suggests that the previously demonstrated increased hemocyaninoxygen (Hc–O2) affinity due to elevated thiosulfate may be adaptive in vivo. Average oxygen-consumption rates were much higher in crabs experimentally exposed to sulfide than in unexposed crabs. Crabs injected with isosmotic thiosulfate did not have increased oxygen-consumption rates as did the sulfide-exposed individuals, but did show a similar reduction in P c (the critical partial pressure of oxygen at which crabs can no longer regulate oxygen consumption). This suggests that it is the sulfide exposure and/or detoxification rather than the elimination of thiosulfate that causes the increase in metabolic rate. Thiosulfate diffuses into dead crabs and into live crabs exposed to 15 mmol S2O 3 2- l–1, indicating substantial permeability, and yet live crabs are able to eliminate thiosulfate when incubated in sea water containing 1.5 mmol S2O 3 2- l–1, suggesting a process that has an active component.  相似文献

The pattern of growth (biomass accumulation) in Ecklonia radiata throughout the year and across a depth profile was investigated using the traditional hole-punch method, and the information presented in context with concurrently measured in situ net productivity rates. The rate of net daily productivity showed a lack of consistent seasonal variability, remaining constant throughout the year at two of the four depths measured (3 m and 12 m), and becoming higher during winter at another (5 m). Throughout the year, rates of net daily productivity differed significantly across the depth profile. Net daily productivity rates averaged 0.017 g C g–1 dwt day–1 and 0.005 g C g–1 dwt day–1 at a depth of 3 m (1,394 mol O2 g–1 dwt day–1) and 10 m (382 mol O2 g–1 dwt day–1) respectively. In contrast, the biomass accumulation rate of E. radiata was highly seasonal, with low rates of growth occurring in autumn (0.002 g dwt g–1 dwt day–1 at both 3 and 10 m) and summer (0.007 and 0.004 g dwt g–1 dwt day–1 at 3 and 10 m respectively) and higher rates in spring (0.016 and 0.007 g dwt g–1 dwt day–1 at 3 and 10 m respectively) and winter (0.015 and 0.008 g dwt g–1 dwt day–1 at 3 and 10 m respectively). The proportion of assimilated carbon used for biomass accumulation varied throughout the year, between 5% and 41% at 3 m and between 28% and 128% at 10 m. Beryllium and aluminium contents in uncontaminated soils from six countries are reported. The means and ranges of beryllium in the surface soils were as follows: 1.43(0.20–5.50)g g–1 in Thailand (n=28), 0.7 (0.31–1.03) g g–1 in Indonesia (n=12), 0.99(0.82–1.32) g g–1 in New Zealand (n=3), 0.58(0.08-1.68)g g–1 in Brazil (n=16), 3.52(2.49–4.97)g g–1 in the former Yugoslavia (n=10), and 1.56(1.01–2.73) g g–1 in the former USSR (n=8). The mean and range of beryllium contents of the surface soils in Japan (1.17(0.27–1.95)g g–1 n=27) are situated within the values of the soils from these countries except for the Yugoslav soils derived from limestones. The mean of the mean beryllium contents of the surface soils in all these countries is 1.42 g g–1 which will be used as a tentative average content of beryllium in uncontaminated surface soils, except for the soils derived from parent materials high in beryllium content. The beryllium contents of the subsoils were higher than those of the surface soils in New Zealand and Yugoslavia as is the case with Japan. The correlation coefficient between the contents of beryllium and aluminium in all the soil samples (n=113) including surface soils and subsoils was 0.505 (p < 0.001).  相似文献

Dissolved total carbohydrate (TCHO), polysaccharide (PCHO), monosaccharide (MCHO) and organic carbon (DOC) were determined at 3-h intervals over 5 diel cycles in the mixed layer of the northwestern Caribbean Sea while following a drogued buoy. These data have been compared to populations of phototrophic (PNAN) and heterotrophic (HNAN) nanoplankton (2–20 m diameter) and heterotrophic bacteria (HBAC) (0.2–2.0 m diameter) estimated by epifluorescence counts, as well as to CO2, phosphate, chlorophyll a and phaeopigment data determined simultaneously. Two different types of apparent diel dissolved carbohydrate (CHO) patterns were found. On 3 d when no sustained net CO2 uptake was evident, TCHO and PCHO generally declined during the afternoon and early evening while MCHO tended to increase. On two other days when apparent sustained CO2 uptake occurred during the day, there were large evening TCHO and PCHO peaks with constant or declining MCHO levels. These accumulations probably resulted from the release of recently produced PCHO from phototrophs. As was found earlier in the Sargasso Sea, PNAN populations were inversely related to PCHO concentrations. The sample to sample fluctuations of PNAN also were inversely related to the apparent rates of change of TCHO and PCHO, possibly due to an inverse relation between the rates of PNAN cell division and CHO excretion. Fluctuations in HBAC populations were inversely correlated with PCHO dynamics and directly related to MCHO variations, possibly due to extracellular hydrolysis of PCHO to MCHO during periods of rapid bacterial growth as well as to net heterotrophic PCHO uptake. A direct relationship between HNAN and TCHO fluctuations suggests the importance of HNAN excretion in the release of dissolved organics. The combined PNAN and HBAC fluctuations accounted for a more significant fraction of the variance in the apparent rates of change of PCHO than did any single population parameter indicating that intimate interactions between the microbial plankton groups are important in the in-situ regulation of CHO dynamics. Total system net TCHO release and uptake rates for 5 d averaged 56 and 53 g C l-1 d-1 respectively, assuming that the observed fluctuations resulted from temporal planktonic processes in homogeneous water masses. While the data contain indications that this was the case, this assumption is not definitive.  相似文献

The red algaGelidium sesquipedale (Clem.) Born. et Thur. has been cultured in chemostats to assess the effects of light quality and photon-fluence rate (PFR) on growth, photosynthesis and biochemical composition. Plants under blue and red light (BL and RL) showed higher growth rates than under white light (WL) of the same PFR (40 mol m–2 s–1). The light-saturated rate of photosynthesis was higher for algae grown under BL and RL than for algae grown under WL. When algae were transferred to WL of moderate PFR (100 mol m–2 s–1), the light-saturated rate of photosynthesis decreased, being higher in previously RL-grown algae than in previously BL- and WL-grown algae. The initial slope of photosynthesis-irradiance (PI) curves () was affected by PFR but not by light quality. Pigment content was little affected by light quality. Light-quality treatments also affected the biochemical composition of the alga; previous exposure to various light treatments activate or repress several metabolical pathways that are fully expressed in the subsequent phase of WL of moderate PFR. Thus, phycobiliproteins and soluble proteins increased for previously BL- and RL-grown algae, whereas insoluble carbohydrate concentration was reduced, indicating a change of the C-partitioning between carbon compounds and organic nitrogen compounds. Inorganic nitrogen metabolism was also affected by light: under WL of moderate PFR, NO3 was totally depleted from sea water, and maximal values of NO3 uptake were recorded. In addition, neither NO2 nor NH4 + was released. However, when algae were transferred to a low PFR, there was a drastic reduction of NO3 uptake under WL, which only partially recovered over time. It was accompanied by the release of NO2 , but not NH4 +, to the culture medium. Under BL and RL, however, there was a transient enhancement of NO3 uptake that was followed by a net release of NO2 and NH4 . Growth rates were not correlated with PFR. This could be due to the the dynamics of internal carbon mobilization and accumulation in the algae. When algae were exposed to a moderate PFR of WL, carbon requirements for growth were satisfied by photosynthesis. Thus, there was a net accumulation of carbon in the tissue. In contrast, when algae were exposed to low PFRs of either WL, BL or RL, observed growth rates could not be maintained by photosynthesis and carbon was mobilized.  相似文献

The activity of Na–K-ATPase was measured in crude homogenates prepared from various organs (leg muscle, pincer muscle, heart, testes, digestive gland, hypodermis, gills 1–9) of shore crabs, Carcinus maenas L., acclimated to salinities ranging between 10 and 50 S (in steps of 10 S). In all salinities tested, Na–K-ATPase activity was highest in posterior gills 7–9 (10–12 mol Pi mg protein-1 h-1), followed by anterior gills 1–6 (ca. 2.5 mol Pi mg protein-1 h-1) and the other organs (in most cases far below 2mol Pi mg protein-1 h-1). In gills only, Na–K-ATPase activity was salinity-dependent, with the highest values in the lowest salinities and vice versa. In gills 7–9, Na–K-ATPase activity was increased more than threefold following a reduction in salinity from 50 to 10 S. Na–K-ATPase activity, expressed as percentage of total ATPase activity, amounted to 60–80% in gills, about 60% in hypodermis and 20–40% in the other organs. Ouabain, a specific inhibitor of Na–K-ATPase activity, reduced serum osmolalities in crabs kept at 9–10 S only when injected into the hemolymph (1 and 5 · 10-5 M), but had no effect when dissolved in ambient water (10-4 M). The results obtained underline that crustacean gills are the main organs for ionic regulation, and confirm the hypothesis of the central role of the Na–K-ATPase in active Na uptake as the basic mechanism of hyperregulation in dilute media. Reduction of serum osmolalities following injection of ouabain into the hemolymph confirms previous reports on localization of the sodium pump in the basolateral parts of epithelial cells.  相似文献

Adult Elasmopus rapax, collected from the eastern coast of Venezuela in 1990, were exposed to seawater containing various CdCl2 concentrations ranging from 0.25 to 5.5 mol l-1. The 48-h and 96-h LC50 values obtained were 4.0 and 1.6 mol Cd l-1, respectively. In amphipods exposed to 1 mol Cd l-1 for up to 240 h, the apparent rate of cadmium uptake was higher in dead animals (most of which had molted during the preceding 24 to 48 h) than in those which survived throughout the treatments without molting. Thus, whole-body cadmium content reached 1.74 mol g-1 dry weight (dw) in the former and only 0.85 mol g-1 dw in the latter; the higher body Cd-load may have caused the increased mortality observed in molters. On exposure to cadmium levels above 0.5 mol l-1 the oxygen consumption rate of non-molters decreased from 2.2 to about 1.5 ml O2 g-1 dw h-1 over the first 24 h, remaining unchanged thereafter. The results place E. rapax among the most sensitive marine organisms yet studied concerning cadmium toxicity, and emphasize the usefulness of the Amphipoda as bioindicators and research tools for bioassays.  相似文献

Juveniles of the dendrobranchiate decapod Penaeus indicus take up radiolabelled zinc from solution at all exposure concentrations studied from 5.6 to 100 g l–1, with an uptake rate constant of 0.045 l g–1 day–1 at 15 salinity and 25°C. Over the first 10 days of zinc exposure, the new zinc taken up is added to the existing zinc content of the prawn with no significant excretion; over this period the rate of accumulation of radiolabelled zinc is a measure of the absolute zinc uptake rate from solution. Over the next 10 days of zinc exposure to 10 g Zn l–1, however, zinc is excreted at about half the rate of uptake resulting in a raised body concentration of zinc. Moulting had no significant effect on the accumulation of zinc. Newly accumulated zinc is distributed to all organs with the highest proportions of body content being found in the exoskeleton, followed by the muscle, the hepatopancreas and the antennal organs. Radiolabelled zinc is subsequently lost from all organs. Unlike caridean shrimps or prawns (pleocyemate decapods), therefore, penaeids (dendrobranchiate decapods) do not show regulation of zinc body concentrations to a constant level over a range of dissolved zinc bioavailabilities by matching zinc excretion to zinc uptake. Nevertheless, unlike amphipod crustaceans, P. indicus does excrete some of the zinc newly accumulated from solution after a time delay. Unlike their caridean counterparts, penaeid prawns inhabiting anthropogenically contaminated coastal waters with raised zinc bioavailabilities can be expected to contain raised body concentrations of zinc.Communicated by J.P. The control mechanisms within the pelagic microbial food web of the oligotrophic Gulf of Aqaba and the northern Red Sea were investigated in the spring of 1999. Nutrient conditions and potential grazer impact were manipulated in a series of dilution experiments. Ambient nutrient concentrations and autotrophic biomass were very low (0.23–1.21 µmol NO3 l–1, 0.06–0.98 µmol NH4 l–1, 1.08–1.17 µmol Si l–1, 0.08–0.12 µmol P l–1, 0.15–0.36 µg chlorophyll a l–1). The planktonic community was characterized by low abundances [3.0–5.5×105 heterotrophic bacteria ml–1, 0.58–7.2×103 ultraphytoplankton <8 µm ml–1 (small eukaryotic photoautotrophs and Prochlorococcus sp., excluding Synechococcus sp.), 0.45–4.4×104 Synechococcus sp. ml–1, 0.32–1.2×103 heterotrophic nanoflagellates ml–1, 1.3–3.8×103 phytoplankton >8 µm l–1, 0.93–5.4×102 microzooplankton l–1] and dominated by small forms (0.2–8 µm). Dinoflagellates and oligotrichous ciliates were the most common groups in initial samples among the phytoplankton >8 µm and microzooplankton, respectively. Results show that bottom-up and top-down control mechanisms operated simultaneously. Small organisms were vulnerable to grazing, with maximum grazing rates of 1.1 day–1 on heterotrophic bacteria and 1.3 day–1 on ultraphytoplankton. In contrast, algae >8 µm showed stronger signs of nutrient limitation, especially when the final assemblages were dominated by diatoms. Synechococcus sp. were not grazed and only showed moderate to no response to nutrient additions. The high spatial and temporal variation of our results indicates that the composition of the planktonic community determines the prevailing control mechanisms. It further implies that, at this transitional time of the year (onset of summer stratification), the populations fluctuate about an equilibrium between growth and grazing.Communicated by O. Photosynthetic parameters for netplankton (>22 m) and nanoplankton (<22 m) varied over similar ranges but exhibited different seasonal and geographic patterns of variation. Nanoplankton a was relatively constant (0.06 mg C [mg Chl · h]-1 [E m-2 s-1]-1), but P m B (mg C [mg Chl · d]-1) was an exponential function of temperature independent of nutrient concentration and vertical stability in the euphotic zone. The temperature function gives a P m B of 24 at 25°C for nanoplankton growing in an estuarine environment characterized by high nutrient concentrations and a shallow, stratified euphotic zone. Variations in netplankton a and P m B were less predictable and were not correlated with temperature, nutrients or vertical stability. Chain forming diatoms with small cells were able to achieve high (0.10 to 0.15) and P m B (20 to 24) that were 3 to 5 times higher than large-celled diatoms and dinoflagellates were able to achieve.  相似文献
