Eelgrass (Zostera marina L.) has access to nutrient pools in both the water column and sediments. We investigated the potential for eelgrass to utilize nitrate nitrogen by measuring nitrate reductase (NR) activity with an in vivo tissue assay. Optimal incubation media contained 60 mM nitrate, 100 mM phosphate, and 0.5% 1-propanol at pH 7.0. Leaves had significantly higher NR activity than roots (350 vs 50 nmoles NO 2 produced g FW–1 h–1). The effects of growing depth (0.8 m MLW, 1.2 m, 3.0 m, 5.0 m) and location within the eelgrass meadow (patch edge vs middle) on NR activity were examined using plants collected from three locations in the Woods Hole area, Massachusetts, USA, in July 1987. Neither depth nor position within the meadow appear to affect NR activity. Nitrate enrichment experiments (200 M NO 3 for 6 d) were conducted in the laboratory to determine if NR activity could be induced. Certain plants from shallow depth (1.2 m) showed a significant response to enrichment, with NR activity increasing from >100 up to 950 nmoles NO 2 g FW–1 h–1 over 6 d. It appears that Z. marina growing in very shallow water (0.8 m) near a shoreline may be affected by ground water or surface run-off enrichments, since plants from this area exhibited rates up to 1 600 nmol NO 2 g FW–1 h–1. Water samples from this location consistently had slightly higher NO 3 concentrations (1.4 M) than all other collection sites (0.7 M). Thus, it is possible that chronic run-off or localized groundwater inputs can create sufficient NO 3 enrichment in the water column to induce nitrate reductase activity in Zostera leaves.  相似文献

The magnitude and physiological characteristics of biological nitrogen fixation have been studied in the oligotrophic waters of the North pacific gyre. The filamentous blue-green algae Trichodesmium spp. and Richelia intracellularis were the important nitrogen-fixing phytoplankton. Most of the nitrogen fixation occurs in the upper 40 m of the water column, with detectable fixation as deep as 90 m, which corresponds to about the 1 % light depth. There was no evidence of photoinhibition of nitrogen fixation, although CO2 reduction was depressed slightly at the highest light levels. The rate of nitrogen fixation in the water column varied throughout the day, being highest in mid-morning and in late afternoon. Relatively high fixation rates were also found during periods of darkness. Elevated oxygen concentrations had a marked inhibitory effect on rates of nitrogen fixation, a pO2 of 0.4 atm causing a 75% inhibition. Data from studies of nitrogen fixation and assimilation rates of 15N-labelled nitrate, ammonium, and urea indicate that nitrogen fixation furnished about 3% of the total daily fixed nitrogen requirement for phytoplankton growth. Studies with isolated colonies of Trichodesmium spp. indicated that 100% of their nitrogen requirement was met by nitrogen fixation. Chemical composition of the Trichodesmium colonies showed that the C:N ratio was 4.1 and that their phosphorus content relative to carbon or nitrogen was much lower than that of the total particulate material in the water column. Elevated ratios of carbon: adenosine triphosphate (ATP) also suggest that phosphorus deficiency may be limiting the growth of Trichodesmium. The magnitude of nitrogen fixation in the gyre is seasonally dependent, with high rates in late summer and autumn. At these times the water column is stratified, with phosphate and nitrate barely detectable in the upper 100 m. Our data suggest that during these months of stratification, biological fixation of nitrogen amounts to about 33 g-at N/m2/day.  相似文献

The relative importance of 3 different sources for biological production of nitrite in seawater was studied. Decomposition of fecal pellets of the copepod Calanus helgolandicus (at a concentration of approximately 12 g-at N/l), in seawater medium, released small amounts of ammonia over a 6 week period. It nitrifying bacteria were added to the fecal pellets nitrite was barely detectable over the same period. Decomposition of phytoplankton (present at a concentration of about 8 g-at particulate plant N/l) with added heterotrophic bacteria, released moderate amounts of ammonia over a 12 week period. If the ammonia-oxidizing bacterium Nitrosocystis oceanus was added to the decomposing algae, nitrite was produced at a rate of 0.2 g-at N/l/week. Heterotrophic nitrification was not observed when 7 open-ocean bacteria were tested for their ability to oxidize ammonia. The diatom Skeletonema costatum, either non-starved or starved of nitrogen, produced nitrite when growing with 150 or 50 g-at NO 2 - -N/l at a light intensity of about 0.01 ly/min. When nitrate in the medium was exhausted, S. costatum assimilated nitrite. If starved of vitamin B12, both non-N-starved and N-starved cells of S. costatum produced nitrite in the medium with 150 g-at NO 3 - -N/l. Nitrate was not exhausted and cell densities reached 2x105/ml due to vitamin B12 deficiency. If light intensity was reduced to 0.003 ly/min under otherwise similar conditions, cells did not grow due to insufficient light, and nitrite was not produced. In the sea, it appears that, in certain micro-environments, decomposition of particulate matter releases ammonia with its subsequent oxidation to nitrite. The amounts of these nutrients and the rate at which they are produced are dependent upon the nature of the materials undergoing decomposition and the associated bacteria. In certain other areas of the sea, where phytoplankton standing stock is high and nitrate is non-limiting, excretion by these organisms is a major source of nitrite.  相似文献

Under conditions of natural irradiance, the development and decline of a flagellate-dominated phytoplankton population was followed in a coastal North Atlantic pond over a 3 d period in summer 1986. Irradiance negatively affected phytoplankton biomass estimated as chlorophyll a, which decreased during the day at photosynthetically available radiation (PAR) levels above 600 to 1000 mol m-2s-1; chlorophyll a increased at PAR values below this threshold. In addition, an inverse relationship was found between changes in chlorophyll a and changes in dissolved inorganic nitrogen, indicating synthesis of nitrogenous biomass mainly at night and degradation mainly during the day, with intense exchanges of material between the particulate and dissolved nitrogen fractions. The natural abundance of 13C in particulate matter increased initially, and then remained constant, and was controlled mainly by the ratio -carboxylases activity: ribulose biphosphate carboxylase activity. The hypothesis that the latter enzyme is broken down under high irradiance and is partly responsible for increases in external dissolved nitrogen was rejected.  相似文献

Nitrogen uptake by the kelp Ecklonia maxima Osbeck and phytoplankton was examined under different conditions of nutrient availability in a kelp bed off the Cape of Good Hope by measuring nutrient depletion in large plastic bags by the kelp and 15N uptake by phytoplankton. E. maxima took up nitrate and ammonia, but not urea, and showed only a weak preference for reduced nitrogen. Phytoplankton absorbed all three forms of nitrogen available, with a preference for ammonia and urea. Ambient nitrate concentration exhibited a marked and rapid decrease with northerly winds and an increase in response to offshore southerly winds. Nitrogen uptake by E. maxima was linearly related to ambient concentration and did not saturate even at nitrate concentrations >20g-at N l-1, resulting in a significantly higher tissue nitrogen content under upwelling conditions. Nitrate imported by upwelling was the chief source of nitrogen utilised within the kelp bed. Locally regenerated nitrogen (ammonia and urea) was calculated to contribute only ca 4% of total nitrogen uptake during upwelling and 30% during the relaxation or downwelling phase.  相似文献

We estimated primary productivity and distributions of carbon in the phytoplankton, micro-zooplankton, and suspended and dissolved matter in various areas of the World Ocean to increase our information about the organic carbon cycle in the surface layer of the sea. Primary productivity ranged from about 0.1 gC m–2 day–1 in the Gulf of Mexico to 9 gC m–2 day–1 in nutrient-rich water off Peru. Phytoplankton carbon ranged from less than 10 g/l in the former to 750 g/l in the latter and in nutrient-rich water off southwest Africa. Micro-zooplankton carbon usually was less than 50 g/l in all waters, and was dominated by ciliates, copepodids, and copepod nauplii in all areas. Concentrations of particulate carbon ranged from 12 g/l off the east coast of South America to 850 g/l off southwest Africa. Concentrations of dissolved organic carbon varied between 0.5 and 1.5 mg/l in all areas except off Peru, where maximum values of 4.5 mg/l were observed. Turnover rates of carbon by small standing crops of micro-flagellates (1 to 5 longest dimension) and dinoflagellates in nutrient-poor waters were lower than those by large standing crops of diatoms and micro-flagellates in nutrient-rich waters. Concentrations of phytoplankton usually accounted for 20 to 55% and micro-zooplankton for 2 to 30% of the particulate carbon in the surface layer of the sea. Concentrations of dissolved organic carbon were not related to concentrations of particulate carbon in most waters except off Peru, where they appear to be directly related.  相似文献

The uptake rates of the three nitrogen compounds ammonium, nitrate, and urea were measured in the oligotrophic North Central Pacific Gyre in August–September 1985. The measurements were performed by using 15N-labelled substrates and incubating for short-time periods (3 to 4 h) under simulated in situ conditions. Ambient concentrations of the nitrogenous nutrients were generally below 0.10 mol l-1. The average total daily nitrogen uptake rate, integrated over the euphotic zone, was 12.5 mmol N m-2 d-1. Diel studies in the upper water mass resulted in a calculated phytoplankton growth rate of 1.3 d-1. Ammonium was the dominating nutrient, accounting for on the average 54% of the total nitrogen uptake, while urea uptake represented 32% and nitrate 14%. Ammonium uptake rates at a coastal station off the Hawaiian Islands were very close to the rates found at the oceanic station. Organisms <3 m dominated the nitrogen assimilation, being responsible for about 75% of the ammonium uptake. The nitrogen uptake rates in this study seem to be higher than those found by earlier investigations in the area, but correlated well with other productivity measurements performed during the same cruise.  相似文献

Seasonal variations in tissue nitrogen (ethanol soluble nitrate and ninhydrin positive substances, as well as total nitrogen) of different thallus parts of Pleurophycus gardneri Setchell and Saunders were monitored simultaneously with ambient seawater nitrate from 1982 until 1984 in Bamfield, Vancouver Island, British Columbia, Canada. A trend of low, nearly zero levels in ambient nitrate typical for the area in late spring and early summer normally contrasts with average nitrate concentrations of 10 mol NO3 - l-1 in late fall and winter. Total nitrogen content was greater in the perennial thallus parts, stipe and holdfast than in the annual blade and peaked in fall and early winter. The longitudinal thallus distribution of nitrate revealed a distinct and significant concentration of nitrate in the haptera reaching at maximum 8% nitrate-N of the internal total nitrogen. Internal nitrate concentration ranged from 20 to 5 000 times the ambient nitrate concentration in the midrib, and from 40 to 3 100 times in the wing, while the range was greatest with 400 to 14 000 times in the haptera. P. gardneri contained at most about 7 mol NO3 - g fresh wt-1 in the blade, which corresponds to about 6% of total tissue nitrogen. Ninhydrin positive substances comprised the major portion of the soluble N pool in P. gardneri and showed a pronounced seasonality. Concentrations of ninhydrin positive substances ranged from 20 to 800 g N g fresh wt-1 in the midrib and in the wing. In the stipe, ninhydrin positive substances varied from 180 to 2 200 g N g fresh wt-1, and from 250 to 1 200 g N g fresh wt-1 in the haptera. Evidence is given that (1) the perennial parts, stipe and haptera of P. gardneri contain the majority of nitrogen products independent of season and ontogenetic stage; (2) ninhydrin positive substances are the most abundant internal nitrogen constituents; (3) the low N values in the blade in summer suggest a nitrogen limited growth; and (4) nitrate may not be the predominant external nitrogen source.  相似文献

Rainwater concentrations of either ammonium or nitrate were sufficient to stimulate chlorophyll a (chl a) production in bioassay experiments using Gulf Stream surface water collected off North Carolina during the summer of 1991. Previous studies primarily examined inshore waters and did not address the impact of rainwater ammonium. An increase in chl a occurred within 1 d of the addition of synthetic rainwater (2 or 5% rainwater, 98 or 95% seawater) containing up to 10 M ammonium; this increase was followed by a decrease in chl a the following day. A similar response to nitrate addition (5% addition of 20 M nitrate rain) was observed. In separate experiments, natural rainwater having nitrate and ammonium concentrations less than those in the experimental synthetic rain yielded a greater chl a response than synthetic rain when added at similar dilutions (0.5 to 5.0% rain). The maximum dissolved inorganic nitrogen concentration in the enriched seawater in these bioassays was 1.8 M; prior to enrichment the maximum was < 0.4 M. Bioassay experiments begun 2 d after a major storm event (sustained NE winds with gusts to 13 m s-1 and ca. 390 mol m-2 inorganic nitrogen deposition from rain) showed a chl a increase in response to addition of natural rainwater, but not to synthetic rainwater with similar dissolved inorganic nitrogen concentration. These results suggest that phytoplankton stimulants, in addition to nitrate and ammonium, exist in natural rain but not in the synthetic rain used in these experiments.  相似文献

Productivity was studied in two diatom species, Chaetoceros armatum T. West and Asterionella socialis Lewin and Norris, which form persistent dense blooms in the surf zone along the Pacific coast of Washington and Oregon, USA. Past observations have shown that surf-diatom standing stock usually declines in summer along with concentrations of nitrate and ammonium. Using the 14C method, photosynthetic rates in natural surf samples were measured monthly for one year (October 1981 through September 1982) at a study site on the Washington coast. Also measured were temperature, salinity, dissolved nutrients, particulate carbon and nitrogen (used as estimates of phytoplankton C and N), and chlorophyll a. Assimilation numbers (P max) were higher in summer (5 to 8 g C g-1 chl a h-1) than in winter (3 to 4gC). Specific carbon incorporation rates (µmax) showed no obvious seasonality, mostly falling within the range of 0.09 to 0.13 g C g-1 C(POC) h-1. The discrepancy between the seasonal trends for chlorophyll-specific and carbon-specific rates reflects a change in the carbon-to-chlorophyll ratio. Because of seasonal differences in daylength and light intensity, daily specific growth rates () are thought to be higher in summer than in winter. Neither ammonium enrichment assays nor particulate carbon-to-nitrogen ratios provided convincing evidence for nitrogen limitation during summer, and the observed changes in diatom abundance cannot be explained on this basis. Both the high diatom concentrations and their seasonal variations probably are due mainly to factors other than growth rates; two factors considered important are diatom flotation and seasonal changes in wind-driven water transport. Seston composition [particulate organic carbon (POC), particulate nitrogen (PN), phyto- and microzooplankton numbers and biomass] was investigated in the bottom waters of Great Lameshur Bay, St. John, Virgin Islands (USA), in October, 1970 during Tektite II Mission 17–50. Mean values of 67.4 g POC · I-1 and 7.2 g PN · I-1 were determined. A mean phytoplankton carbon content of 42.2 g · I-1 and zooplankton carbon content of 5.5 g · I-1 were calculated from counts. The phytoplankton consisted mainly of dinoflagellates 71.2% phytoplankton carbon. Copepods were the dominant zooplankters (61.8% zooplankton carbon), followed by larvaceans (30.9% zooplankton carbon). Organic carbon content of counted zooplankton faecal pellets ranged between 0.4 and 1.6 g · I-1, and amounted probably to about 15% of the total zooplankton carbon value. Plankton and detritus components as possible food for coral-reef animals are discussed. The ratio carbon: nitrogen of suspended particles is compared to that of sedimented matter.  相似文献

The uptake of nitrate and ammonium was investigated experimentally during early spring 1989 in the Greenland Sea, with particular attention placed on the roles of irradiance, nitrogen concentrations and nitrateammonium interactions. The phytoplankton assemblage was dominated by the colonial prymnesiophyte Phaeocystis pouchetii. Nitrate concentrations ranged from undetectable at the end of the cruise to greater than 10 M, and ammonium levels ranged from less than 0.1 to 1.9M. The uptake of both nitrate and ammonium as a function of irradiance was found to be a saturation response. Photoinhibition occurred and was found to be greater for ammonium uptake. Ammonium uptake also saturated at irradiance levels five times lower than those needed to saturate nitrate uptake. Nitrate and ammonium uptake as a function of nitrogen concentration also was characterized by a saturation response, with the estimated half-saturation constant (K s) value for nitrate uptake being 0.29 M. Elevated ammonium concentrations inhibited nitrate uptake, and the response appeared to be one of exponential decrease with increasing concentrations of ammonium. The most important factor in the Greenland Sea influencing ammonium uptake during the spring was irradiace, while both irradiance and ammonium concentrations played major roles in regulating nitrate uptake and new production.  相似文献

The effect of ambient ammonium concentration on the nitrate uptake rate of marine phytoplankton was investigated. These studies consisted of laboratory experiments using unialgal species and field experiments using natural phytoplankton communities. In laboratory experiments, ammonium suppressed the uptake rates of nitrate and nitrite. Approximately 30 min were required for ammonium to exhibit its fully inhibitory effect on nitrate uptake. At high ammonium concentration (>3 g-at/l), a residual nitrate uptake rate of approximately 0.006 h-1 was observed. When the ambient ammonium concentration was reduced to a value less than 1 g-at/l, the suppressed nitrate uptake rate subsequently attained a value comparable to that observed before the addition of ammonium. A range of 25 to 60% reduction in the nitrate uptake rate of natural phytoplankton communities was observed at ambient ammonium concentrations of 1.0 g-at/l. A mechanism is proposed for the suppression of nitrate uptake rate by ammonium through feedback control of the nitrate permease system and/or the nitrate reductase enzyme system. Influence of calcium on sodium fluxes was investigated in the brackish-water crab Carcinus mediterraneus Csrn., after activation of sodium regulatory mechanisms, during longterm acclimation in diluted (15.9 S) sea water. The 22Na outflux constants measured in whole crabs are noticeably lower (0.188 to 0.374h-1) in diluted sea water enriched by calcium (5.8 to 10.4 mM Ca2+/l), than in ordinary diluted sea water (0.545 h-1). The sodium-outflux constants in hemolymph, gills and muscle show the same trend of slower exchange of 22Na in calcium-enriched sea water. In ordinary sea water, the total sodium-outflux rate from the hemolymph amounts to 46.31 M Na/g/h, while in calcium-enriched sea water (8.23 mM Ca2+/l) it is inhibited, amounting to 13.86 M Na/g/h. Sodium and potassium concentrations of intracellular muscles in diluted sea water enriched with calcium and control diluted sea water are similar. The outflux of intracellular sodium from the muscle amounts to 2.84 M Na/g/h in crabs acclimated to diluted sea water.  相似文献

The uptake, storage and excretion of cadmium by the common mussel Mytilus edulis (L.) has been studied at sub-lethal concentrations using the radioactive isotope 115mCd as a marker. After an initial lag period, the uptake at low concentrations in sea water is linear with time and directly proportional to the sea water concentration, with a maximum concentration factor of 165 at 0.7 g Cd/ml sea water. A decrease occurs at higher concentrations indicating saturation of the available binding capacity. Prior complexation of the cadmium with either EDTA, humic and alginic acids or pectin doubles both the rate of accumulation and the final tissue concentrations (order: kidneyviscera>gillsmantle>muscle) and eliminates the lag period, suggesting that ionic cadmium must first be complexed before uptake can occur. A mechanism for this effect, which may involve thionein, is described. The rate of excretion of cadmium is 18 times slower than that of uptake, with the major route via the kidney but not via the byssal threads as with particulate iron. The need to detoxify and store cadmium by an immobilization mechanism is a consequence of this slower rate of elimination.  相似文献

Grazing rates of larger (Calanus finmarchicus) and smaller (Acartia clausii Pseudocalanus elongatus etc.) copepods on naturally occurring phytoplankton populations were measured during a declining spring phytoplankton bloom. During the initial period, dominated by Chaetoceros spp. diatoms, constant ingestion rates were observed in Calanus finmarchicus at suspended particulate concentrations above 300 g carbon l-1. Average daily intake during this time amounted to 35 to 40% of body carbon and reached a maximum of 50%. The feeding response of the smaller copepods was not so well defined, although a maximum daily intake of 56% body carbon was recorded. In both groups, feeding thresholds were at particulate concentrations around 50 g C l-1. The feeding response of C. finmarchicus was correlated with both a change in their own population and in the food cell type. Linear regressions describing the concentration-dependent feeding response were: ingestion rate (IR)=1.16 total particulate volume (TPV)-36.15 during the initial part of the period compared with IR=0.41 TPV-12.18 for the latter period. C. finmarchicus filtered out slightly larger (x 1.2 diameter) particles than the small copepods and, in both groups, some filtering adjustment was made to accomodate to modal changes in the phytoplankton population from 20–30 m to 10 m diameter cells. Particle production during feeding was frequently evident in the smallest size ranges of particles and the ratio of particle production to ingestion rate was greater at low feeding rates.  相似文献

Changes in the phosphorus components of the particulate matter in seawater were studied in the eutrophicated waters of Mikawa Bay, Japan, during summer 1981. The contents of particulate phosphorus and hot-water extractable intracellular phosphorus displayed remarkable changes associated with phytoplankton blooms caused by wind-induced or upwelling-associated nutrient enrichment from the bottom water layers. Nanoplankton <10 m accounted for much of the particulate phosphorus (70 to 79% in June and July, and 44 to 78% in August and September); the contribution of large-sized phytoplankton >25 m varied from 9 to 49%, the peak values being attained under red-tide conditions. The capacity for phosphorus storage in cells was low in nanoplankton cells, high in large phytoplankton species. Differences in rates of phosphorus storage and growth between nanoplankton and large phytoplankton accounted for fluctuations in particulate phosphorus which were closely associated with fluctuations in phytoplankton blooms in Mikaw Bay.  相似文献

The distribution and composition of suspended particulate matter in the sea is very complex and not well understood. In this study, 3 different approaches were used to estimate the quantity and quality of suspended particulate matter in 34 samples from the euphotic zone of 9 stations in the Gulf of California. The results from electronic, microscopic and chemical analyses showed that most parameters measured were significantly correlated, e.g. the total particulate volume from particles of 2 to 150 diameter, as obtained from the Coulter Counter, proved to be significantly related to such parameters as seston, particulate nitrogen, particulate carbon, phytoplankton carbon and chlorophylla. It can be concluded from this study that the Coulter Counter can be a very useful instrument to determine, with little effort, the size, distribution, and volume of particulate suspended matter in the sea. These data can then be used to calculate some important biological parameters which are necessary to establish meaningful models of phytoplankton production. A study of phosphate assimilation by coastal marine plankton revealed that both phytoplankton and microheterotrophs incorporated radioactive phosphorus (33P). Size fractionation of the particulate matter (using 1 m pore diameter Nucleopore® membrane filters), antibiotic treatment (using garamycin), and independent estimaties of photoautotrophic (14CO2 uptake) and heterotrophic (3H-glucose uptake) activities were employed to separate phyto- and bacterioplankton phosphate uptake. Results indicated that phytoplankton 33P-uptake was best estimated by the fraction of particulate matter retained on the 1 m membrane filters. Usually, less than 10% of the phytoplankton (based on chlorophyll a measurements) passed the 1 m pore-diameter filters, whereas about 90% of the heterotrophic activity passed. At least 50% of the 33P-uptake was associated with the <1 m fraction. It may be possible to resolve the phytoplankton and bacterial contributions to 33P-uptake by comparing the percent of total 33P-uptake with the percent of total 3H-glucose uptake associated with the >1 m fraction.  相似文献

To reveal the distribution characteristics of phytoplankton and the main influence factors under different conditions in the urban rivers, the investigations were conducted during autumn and winter 2014 in Changzhou City, East China. 178 taxa of phytoplankton belonging to 28 functional assemblages were identified. In autumn, the phytoplankton community compositions have high similarity for enhanced hydrological connectivity. The chlorophytes and diatoms (prevailing functional groups C, F, J, P), together with euglenoids (W1), showed high proportions of biomass in the main rivers and connected rivers. It was related to the well mixed eutrophic conditions. The phytoplankton community exhibited spatiotemporal heterogeneity in winter. Affected by the low water level and temperature, the free-living phytoflagellates (X2) replaced groups F and J in the main rivers. Phytoplankton productivity was the highest in the Tongji River. Chlorophytes Dictyosphaerium ehrenbergianum and Chlamydobotrys stellata had an overwhelming superiority during the winter bloom. They were significantly correlated with ammonium, total phosphorus and biochemical oxygen demand. Affected by tail water supply, the diatoms (MP) and euglenoids (W1) dominated in a beheaded river. The multivariate analyses based on the phytoplankton functional groups helped to evaluate the relationships and variations between the urban rivers. The redundancy analysis (RDA) results showed that nitrate nitrogen, water temperature, total nitrogen and total suspended solids were the main influence factors on the phytoplankton community. Except MP, the prevailing groups all showed significant negative correlations with nitrate nitrogen. Availability and utilization of dissolved inorganic nitrogen and hydrodynamic conditions affected the phytoplankton distribution.
