The present study was conducted with an aim to gain better insight of brassinosteroid generated response on the effects of cadmium on photosynthetic machinery and active oxygen metabolism in two tomato cultivars (K-25 and Sarvodya). These tomato cultivars were subjected to graded cadmium levels in soil (0, 3, 6, 9 or 12 mg kg−1 soil) with their foliage being sprayed with 0 or 10−8 M of 28-homobrassinolide/24-epibrassinolide (HBL/EBL) at 59 d stage. The results suggested that photosynthetic parameters, leaf water potential and activity of several enzymes (nitrate reductase and carbonic anhydrase) decreased significantly in both the cultivars, to a lesser extent in K-25 than Sarvodya with the increasing levels of cadmium in the soil. However, the activity of antioxidant enzymes and proline content increased in response to metal treatment as well as the application of brassinosteroids (HBL/EBL). Overall, exogenous application of brassinosteroids improved the activity of photosynthetic machinery and that of antioxidant defense system in both the cultivars, and also nullified the damaging effect of metal on these parameters.  相似文献

Mercury toxicity induces oxidative stress in growing cucumber seedlings   总被引:6,自引:0,他引:6  
In this study, the effects of exogenous mercury (HgCl(2)) on time-dependent changes in the activities of antioxidant enzymes (catalase and ascorbate peroxidase), lipid peroxidation, chlorophyll content and protein oxidation in cucumber seedlings (Cucumis sativus L.) were investigated. Cucumber seedlings were exposed to from 0 to 500microM of HgCl(2) during 10 and 15 days. Hg was readily absorbed by growing seedlings, and its content was greater in the roots than the in shoot. Time and concentration-dependent reduction in root and shoot length was observed at all concentrations tested, equally in the roots and shoot, at both 10 and 15 days. At 50microM HgCl(2), root fresh weight of 15-day-old seedlings increased, and at other concentrations, it reduced. For 10-day-old seedlings, reduction in root and shoot fresh biomass was observed. At 15 days, only at 50microM HgCl(2) was there no observed reduction in shoot fresh biomass. Dry weight of roots increased at 500microM both at 10 and 15 days, though at 250microM HgCl(2) there was only an increase at 15 days. There was a significant effect on shoot dry weight at all concentrations tested. Hg-treated seedlings showed elevated levels of lipid peroxides with a concomitant increase in protein oxidation levels, and decreased chlorophyll content when exposed to between 250 and 500microM of HgCl(2). At 10 days, catalase activity increased in seedlings at a moderately toxic level of Hg, whereas at the higher concentration (500microM), there was a marked inhibition. Taken together, our results suggest that Hg induces oxidative stress in cucumber, resulting in plant injury.  相似文献

A hydroponic experiment was carried out to study the effect of cadmium (Cd) on growth, Cd accumulation, lipid peroxidation, reactive oxygen species (ROS) content and antioxidative enzymes in leaves and roots of Arabis paniculata F., a new Cd hyperaccumuator found in China. The results showed that 22-89 microM Cd in solution enhanced the growth of A. paniculata after three weeks, with 21-27% biomass increase compared to the control. Cd concentrations in shoots and roots increased with increasing Cd supply levels, and reached a maximum of 1662 and 8670 mg kg(-1) Cd dry weight at 178 microM Cd treatment, respectively. In roots, 22-89 microM Cd reduced the content of malondialdehyde (MDA), superoxide (O(2)(-1)) and H(2)O(2) as well as the activities of superoxide dismutase (SOD), guaiacol peroxidase (GPX), ascorbate peroxidase (APX) and glutathione reductase (GR). In leaves, the contents of MDA, O(2)(-1) and H(2)O(2) remained unaffected by 22-89 microM Cd, while 178 microM Cd treatment significantly increased the MDA content, 69.5% higher than that of the control; generally, the activities of SOD, catalase (CAT), GPX and APX showed an increasing pattern with increasing Cd supply levels. Our present work concluded that A. paniculata has a great capability of Cd tolerance and accumulation. Moderate Cd treatment (22-89 microM Cd) alleviated the oxidative stress in roots, while higher level of Cd addition (178 microM) could cause an increasing generation of ROS, which was effectively scavenged by the antioxidative system.  相似文献

Cadmium toxicity to the green alga Stichococcus bacillaris was investigated in media of pH 3-9. A significant decrease of cadmium toxicity occurred in both the acidic and alkaline ranges of pH. In media of pH 3 and 9, cadmium did not affect the dry mass content substantially. Maximum toxicity of cadmium was noticed at pH 6-7. Voltametric investigations showed a significant effect of pH on electrochemically measured cadmium content in the culture media. Hydrolysis of the medium components and formation of cadmium complexes with OH(-) ions caused a considerable decrease in amounts of electrochemically measured cadmium in the alkaline range of pH.  相似文献

The beet armyworm (Spodoptera exigua) were fed on artificial food contaminated with zinc (200 mg kg(-1) dry mass) or cadmium (66 mg kg(-1) dry mass) for 15 generations. In 15th generation, O2 output and CO2 production of pupae were measured. Exposure to cadmium did not cause any effects whilst exposure to zinc led to a significant increase in the respiration rate of pupae. The average respiratory quotient (RQ) did not differ between treatments (ca. 0.7).  相似文献

The effect of cadmium on stage II naupliar larvae of barnacle (Balanus amphitrite Darwin) was investigated. Barnacle larvae were exposed to 0-15 microM CdCl(2) for 24 h. Apoptotic cells were stained by the terminal deoxyribonucleotidyl transferase mediated dUTP nick end labelling (TUNEL) method. Incidence of apoptosis, as measured by numbers of animals with ectopic pattern of apoptosis as well as numbers of apoptotic cells per animal, was assessed using confocal microscopy. An increase in incidence of apoptosis was observed in the experimental animals with an increase in cadmium concentration. Mortality increased, and motility decreased, when barnacle larvae were exposed to an increasing concentration of cadmium. The relationship between the occurrence of apoptosis and swimming behaviour was investigated in larvae exposed to 10 microM CdCl(2). Significant increases in apoptosis were detected in the non-motile and dead nauplii. This study suggested that whole-mount in situ TUNEL method may be used to study increased occurrence of apoptotic cells in crustacean larvae.  相似文献

The natural background of cadmium content in meat, liver and kidney from 287 cattle from a non polluted area in the subalpine region of Bavaria has been analyzed during the period from 1972 to 1975. The cadmium content in meat was always below the detection limit of 0.005, in liver between < 0.005 to 1.65 (median 0.050) and in kidney 0.008 to 1.95 (median 0.229) ppm Cadmium in fresh weight. The cadmium contents are discussed and compared with preliminary suggestions for cadmium tolerance levels and their consequence for food inspection.  相似文献

Cadmium, like many other pollutants, is nondegradable and can be accumulated by Lymantria dispar at a level that affects fitness components, physiology, and development, which could indicate presence of environment pollution by heavy metals. The cadmium effect on fitness-related traits in the third, fourth, fifth, and sixth instar of L. dispar L. was determined. Furthermore, activities of the following antioxidative defense components after the larvae had been fed on the artificial cadmium-supplemented diet (50 μg Cd/g dry food) were assessed: superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APOX), total glutathione amount (GSH), glutathione-S-transferase (GST), glutathione reductase (GR), and the amount of free sulfhydryl (SH) groups. Statistically significant delay of development in the fourth, fifth, and sixth instar and decrease of the larval mass in the third and fourth instar were estimated after the exposure to cadmium through food in comparison to the control. There were no changes in SOD activity of cadmium-treated larvae. Significantly lower CAT, APOX, and GR activities were recorded in the third, fifth, and in the third instar, respectively. At the same time, higher activity was recorded in the sixth instar, while GST activity was higher in the third. GSH content was significantly lower during all instars after treatment but the amount of SH groups was higher in older larvae. The strategy of antioxidative defense and the adjustment or modulation of fitness-related traits in presence of cadmium was dependent on the age of larvae in L. dispar, which might be used in early metal risk assessment in Lepidoptera and other insects.  相似文献

Metals have been reported to inhibit organic pollutant biodegradation; however, widely varying degrees and patterns of inhibition have been reported. To investigate the roles of medium composition and metal bioavailability on these different degrees and patterns of inhibition, we assessed the impact of cadmium on naphthalene biodegradation by a newly isolated strain of Comamonas testosteroni in three chemically-defined minimal salts media (MSM): Tris-buffered MSM, PIPES-buffered MSM, and Bushnell-Haas medium. Cadmium (total concentrations of 100 and 500 microM) inhibited biodegradation in each medium. Degrees of inhibition were different in each medium. Cadmium was most inhibitory in PIPES-buffered MSM and least inhibitory in Bushnell-Haas. For example, in Bushnell-Haas medium, 100 microM cadmium reduced the cell yield more than 4-fold compared to controls not containing cadmium. The same concentration of cadmium completely inhibited growth in PIPES-buffered MSM. No difference in inhibition was observed in any medium when cadmium was added 24 h before inoculation rather than when added within one minute of inoculation. Two patterns of inhibition were observed. Inhibition occurred in a dose dependent pattern in Tris- and PIPES-buffered MSM and in a non-dose dependent pattern in Bushnell-Haas. Specifically, in Bushnell-Haas, 100 microM total cadmium extended the lag phase by 23+/-8.66 h, whereas 500 microM did not extend the lag phase. Soluble, ionic cadmium (Cd2+) concentrations were measured and modeled in each medium to assess cadmium bioavailability. In media containing 500 microM total cadmium, bioavailability was highest in Tris- and PIPES-buffered MSM and lowest in Bushnell-Haas. In Bushnell-Haas, cadmium bioavailability was initially higher in the 500 microM treatments (196+/-21.2 microM) than in the 100 microM treatments (78.2+/-2.04 microM); however, after 12 h, bioavailability was higher in the 100 microM treatments (56.4+/-24.8 micro) than the 500 microM treatments (13.3+/-1.2 microM). These data suggest that the type of medium determines the degrees and patterns by which metals inhibit biodegradation and emphasize the importance of coupling metal toxicity and bioavailability data.  相似文献

Experiments were carried out to investigate the ability of water hyacinth (Eichhornia crassipes) to remove five heavy metals (cadmium, chromium, copper, nickel, and lead) commonly found in leachate. All experiments were conducted in batch reactors in a greenhouse. It was found that living biomass of water hyacinth was a good accumulator for copper, chromium, and cadmium. The plants accumulated copper, chromium, and cadmium up to 0.96, 0.83, and 0.50%, respectively, of their dry root mass. However, lead and nickel were poorly accumulated in water hyacinth. Also, nonliving biomass of water hyacinth dry roots showed ability to accumulate all metals, except Cr(VI), which was added in anionic form. The highest total metal sorption by nonliving dry water hyacinth roots was found to take place at pH 6.4. The current research demonstrates the potential of using water hyacinth for the treatment of landfill leachate containing heavy metals.  相似文献

The adverse impacts of lead and cadmium exposure on health outcomes have been reported in the past. Few studies have been conducted on the relationship between lead and cadmium exposures and disability. We evaluated whether lead and cadmium exposures were associated with functional dependence including the total number of disabilities, activities of daily living (ADL), instrumental activities of daily living (IADL), leisure and social activities (LSA), lower extremity mobility (LEM), and general physical activities (GPA) in an elderly population. A total of 5513 eligible subjects were enrolled in the study from the National Health and Nutrition Examination Survey 2001–2006. Serum lead and cadmium exposure assessments were performed using atomic absorption spectrometry. Functional dependence was assessed by 19 structured questions. The relationships between lead and cadmium exposures and functional dependence were investigated using by multivariable linear regression models. Q2, Q3, and Q4 of lead exposure were significantly associated with the total number of disabilities, with β coefficients of ??0.62 (95% CI ??0.99, ??0.24), ??0.64 (95% CI ??1.02, ??0.26), and ??0.81 (95% CI ??1.19, ??0.42), respectively. This relationship remained significant in males. Furthermore, we analyzed the relationships between lead and cadmium exposure quartiles and various functional dependence metrics, and we determined that lead content was significantly associated with decreased ADL, LEM, and GPA (p?<?0.05) and cadmium content was inversely associated with ADL (p?<?0.05). Our study demonstrated a strong relationship between exposure to lead and cadmium and functional dependence in an elderly population.

Cadmium accumulation in Allium schoenoprasum L. grown in an aqueous medium   总被引:1,自引:0,他引:1  
The ability of Allium schoenoprasum L. (chives) to accumulate and tolerate cadmium in aqueous Hoagland medium at 50microM and 250microM was tested under continuous growth or several successive harvests of shoots. After 28 days of continuous growth, chives accumulated the metal up to 0.2% and 0.5% of its dry weight, when grown in 50microM and 250microM, respectively. In experiments that the leaves were successively harvested every 16 days, there were no obvious stress symptoms after six harvests during a period of 96 days at 50microM Cd. At 250microM, after 64 days and four harvests, inhibition of growth occurred. In each treatment, a total of 1.2g kg(-1) DW and 2.4g kg(-1) DW was accumulated in the leaves, respectively. Total SH compounds concentration in leaf was found significantly higher by 3 and 7.4 times in plants treated with Cd at 50microM and 250microM in comparison to the control, respectively, while no difference in the concentration of glutathione (GSH+GSSG) was found. Thus, it is assumed that sulphur-containing compounds, yet unknown, are involved in defensive mechanisms against heavy metals in chives. The results presented, point to chives phytoremediation potential, but also on the potential risk in accumulation of heavy metals in a commonly edible plant.  相似文献

The effect of cadmium on sediment processing by laboratory-cultured specimens of Capitella sp I and Capitella sp B was investigated. Specimens were exposed to 0, 60, 90 and 140 microg Cd g(-1) dry weight sediment for a 10 day period (two census days) to observe effects of cadmium on their feeding activity. Under unstressed conditions, Capitella sp I specimens were larger, had higher biomass and produced more fecal pellets than Capitella sp B. Cadmium concentrations had no effect on pellet production and body mass of the studied populations. However, cadmium exposure time significantly affected Capitella sp B pellet production and body mass of both populations following different trends: after 10 days of exposure, the average processed sediment per unit worm mass decreased in Capitella sp I but increased in Capitella sp B. The latter population did not reduce its feeding activity in the presence of cadmium maybe related to the fact that it is derived from a highly polluted environment. This study has shown an unimpaired response of Capitella sp I and Capitella sp B to cadmium in their sediment processing activity, which indicates the potential importance of this species complex in the trophic transfer of metals from sediments to other marine organisms.  相似文献

Fourteen cultivars of bai cai (Brassica campestris L. ssp. chinensis var. communis) were grown in the nutrient solutions containing 0-0.5 microg mL(-1) of cadmium (Cd) to investigate genotypic differences in the effects of Cd exposure on the plant growth and uptake and distribution of Cd in bai cai plants. The Cd exposure significantly reduced the dry and fresh weights of roots and shoots, the dry weight ratio of shoot/root (S/R), total biomass, and chlorophyll content (SPAD value). Cd concentrations in bai cai ranged from 13.3 to 74.9 microg g(-1) DW in shoots and from 163.1 to 574.7 microg g(-1) DW in roots under Cd exposure, respectively. The considerable genotypic differences of Cd concentrations and accumulations in both shoots and roots were observed among 14 bai cai cultivars. Moreover, Cd mainly accumulated in the roots. Cd also caused the changes of uptake and distribution of nutrients in bai cai and under the influence of cadmium, the concentration of potassium (K) decreased in shoot and increased in root. However, the concentrations of magnesium (Mg), phosphorus (P), manganese (Mn), boron (B), and iron (Fe) increased in shoots and decreased in roots. In addition, Cd exposure resulted in an increase in calcium (Ca), sulphur (S), and zinc (Zn) concentrations in both shoots and roots but had no significant effects on the whole uptake of the examined mineral nutrients except for S.  相似文献

The pollution is increasing in the environment by different kinds of human activities, which results in the accumulation of heavy metals including cadmium in the soil and water and it causes different types of problems to living beings. As the plants are utilized by human being as food and medicine, therefore, it is mandatory to see the effect of metals on plants. In this context, efforts have been made to observe the effect of different concentration of Cadmium (Cd) on Phyllanthus amarus Schum. and Thonn., because Cd is the widespread metal and the plants response to low and high level of exposure is a complex phenomenon. P. amarus is mostly grown as weed in agricultural and waste lands. It is a reputed plant used in Indian indigenous systems of medicine with hepatoprotective, diuretic, stomachic properties and is recently being used for the treatment of hepatitis B. The study revealed that Cd causes significant decrease in fresh and dry weight, length of root and shoot, protein, chlorophyll, carotenoids and sugar and increase in starch content. It is interesting to note that the therapeutically active compounds—phyllanthin and hypophyllanthin, enhanced at certain levels of Cd due to abiotic stress. Besides, the ultramorpholical changes were also observed in stomatal opening and wax deposition on both the surfaces of leaves.  相似文献

Effects of cadmium (Cd) on the growth and photosynthesis of the bloom-forming cyanobacterium Microcystis aeruginosa Kütz 854 were investigated. The growth was markedly inhibited when it was treated with 4 microM Cd. However, the biomass production was almost not influenced after a prolonged exposure at Cd concentrations < or = 2 microM. Chlorophyll content was more sensitive to Cd toxicity than phycobiliproteins at 0.5 microM Cd. However, the decrease of phycobiliproteins was larger than chlorophyll at the highest Cd concentration. A significant increase of F(v)/F(m) value was observed at Cd concentrations < or = 2 microM. On the other hand, when cells were treated with 4 microM Cd, F(v)/F(m) was significantly increased after 12 h of treatment but decreased after 48 h. The true photosynthesis was decreased with the increase of Cd concentration at 2 h. However, we noticed a recovery when the treatment was prolonged. After 48 h of exposure at the highest Cd concentration, photosynthetic oxygen evolution was markedly inhibited but dark respiration increased by 67%. Cellular Cd contents were augmented with the increase of Cd concentration. To our knowledge, we have demonstrated for the first time that the inhibitory site of Cd in M. aeruginosa is not located at the PSII or PSI level, but is probably situated on the ferredoxin/NADP(+)-oxidoreductase enzyme at the terminal of whole electron transport chain. We noticed also an increase of PSI activity, which is probably linked to the enhancement of cyclic electron transport around PSI. We can conclude that the increase of cyclic electron transport and dark respiration activities, and the decrease of phycobiliproteins might be adaptive mechanisms of M. aeruginosa 854 under high Cd conditions.  相似文献

The response of tobacco plants (Nicotiana tabacum L.)--non-transformed and transformed with a metallothionein gene MThis from Silene vulgaris L.--to increase cadmium supply in the nutrient solution was compared. The transgenic plants accumulated significantly more Cd both in the roots and the leaves. Visual toxicity symptoms and disturbance in water balance were correlated with Cd tissue content. Treatment with 300 microM CdCl(2) resulted in inhibition of photosynthesis and mobilization of the ascorbate-glutathione cycle. Treatment with 500 microM CdCl(2) led to irreversible damage of photosynthesis and oxidative stress. An appearance of a new peroxidase isoform and changes in the leaf polypeptide pattern were observed at the highest Cd concentration. The level of non-protein thiols gradually increased following the Cd treatment both in transgenic and non-transformed plants.  相似文献

Free fatty acids and sterols were assessed in fresh manure and anaerobic lagoon sludge from swine production facilities in North Carolina. Eight free fatty acids and five sterols were identified and quantified in both manure and sludge samples. Compound identification was performed by gas chromatography/mass spectroscopy (GC-MS), and compound quantities were determined by gas chromatography after solid phase extraction with a 50:50 mixture of diethyl ether and hexane. The free fatty acids occurring in greatest abundance in both fresh manure and lagoon sludge were palmitic, oleic, and stearic. Free fatty acid content in fresh manure ranged from approximately 3 microg g(-1) dry weight (dw) to over 45 microg g(-1) dw. In lagoon sludge, free fatty acid content ranged from about 0.8 microg g(-1) dw to nearly 4 microg g(-1) dw. Coprostanol and epicoprostanol were the sterols in largest concentrations in fresh manure and lagoon sludge samples. Total sterol content ranged from approximately 0.5 microg g(-1) dw to around 11 microg g(-1) dw in fresh manure and from 3.5 microg g(-1) dw to almost 9 microg g(-1) dw in lagoon sludge. Fresh manure and lagoon sludge both had high levels of inorganic cations (e.g., Ca, Mg, Fe) capable of binding free fatty acids and forming insoluble complexes, thereby potentially reducing fatty acid biodegradation. In anaerobic lagoons, sterols are an organic fraction of sludge that are resistant to bacterial degradation. In the case of fresh manure, fatty acids could represent a potential source of energy via the manufacture of biodiesel fuel, if efficient means for their extraction and transesterification can be devised.  相似文献

The distributions of copper, zinc, iron, and cadmium among the tissues of Dorippe granulata were determined. The highest copper concentrations were found in the haemolymph (c. 53 microg ml(-1)) while the highest iron concentrations occurred in the gills (c. 720 microg g(-1) dry weight) and the highest zinc concentrations in the exoskeleton (c. 200 microg g(-1) dry weight). By comparison, concentrations of the non-essential metal, cadmium, were low in all tissues (mean = 10 microg g(-1) dry weight). The highest value was recorded from the midgut gland of a female crab (18.5 microg Cd g(-1) dry weight). Concentrations of copper, zinc, and iron were positively correlated with tissue-hydration levels. Such a relationship was not found for cadmium. The findings are discussed with regard to trace-metal levels found in temperate and tropical brachyurans from clean and polluted localities.  相似文献

This study focuses on the screening of cadmium-resistance bacterial strains from heavy metal-polluted soils to examine their plant growth promotion and cadmium uptake in rape (Brassica napus). A large number of bacteria were isolated from heavy metal-polluted soil in Nanjing, China. Thirty isolates showing cadmium-resistance on Cd-amended medium were selected and evaluated for their potential to solubilize cadmium carbonate in solution culture. Atomic absorption spectrometer analysis showed variable amounts of water-soluble Cd (ranging from 24 to 117 mg l(-1)) released by the cadmium-resistant bacterial strains from cadmium carbonate. Qualitative analysis confirmed the presence of indole acetic acid as the auxin in the culture of these cadmium-resistant bacterial strains. Root elongation assay conducted on rape under gnotobiotic conditions demonstrated increases (up to 31%) in root elongation of inoculated rape seedlings compared to the control plants. Based upon cadmium-resistance, bio-activation of CdCO3 and growth-promoting activity, three isolates were selected for promoting plant growth and uptake of cadmium from cadmium-amended soil in pot experiments. Inoculation with the isolates was found to increase root dry weight (ranging from 8% to 20%) and shoot dry weight (ranging from 6% to 25%) of rape. An increase in cadmium content varying from 16 to 74%, compared to the non-inoculated control, was observed in rape plants cultivated in soil treated with 100 mgCd kg(-1) (as CdCl2) and inoculated with the isolates. The bacterial isolates were also able to colonize and develop in the rhizosphere soil of rape after root inoculation.  相似文献
