简述了饮用水消毒副产物(DBPs)的基因毒性与致癌性的研究进展。从Ames试验、SOS/umu试验、彗星试验、微核试验及一些新颖的致突变试验结果对DBPs基因的毒性,以及从毒理学实验、流行病学研究和致癌风险评估3个方面对DBPs的致癌性进行了分析和总结,以期为今后饮用水DBPs毒性效应及其致毒机理研究提供参考,进而促进饮用水质量管理与立法的发展。  相似文献

The process of a bioindication of genotoxic effects of complex mixtures on the environment using higher plants is very appropriate and effective. We present the results of an in situ indication of the genotoxic effects of polluted environment near Žilina city. For a more complex monitoring we used: the Tradescantia micronucleus (Trad-MCN) assay, the Tradescantia microspore test and an evaluation of the abortivity of the pollen grains of native plant species. We found significant differences in the frequency of the micronuclei when using the Trad-MCN test in local of Dubeň. The Tradescantia pollen abortivity test showed significant differences in the frequency of the abortive pollen grains between the exposed groups and the control group. By using native plant species in the pollen abortivity test we found significant differences in both of the two locations for the four following species during two consecutive years: Artemisia vulgaris, Melilotus albus, Trifolium pratense, Typha latifolia.  相似文献

Heavy Metal Load Of Soil, Water And Vegetables In Peri-Urban Delhi   总被引:5,自引:0,他引:5  
Peri-urban lands are often used for production of vegetables for better market accessibility and higher prices. But most of these lands are contaminated with heavy metals through industrial effluents, sewage and sludge, and vehicular emission. Vegetables grown in such lands, therefore, are likely to be contaminated with heavy metals and unsafe for consumption. Samples of vegetables i.e., spinach (Spinacia oleracea L.) and okra (Abelmoschus esculentus L.); soil and irrigation water were collected from 5 peri-urban sites of New Delhi to monitor their heavy metal loads. While heavy metal load of the soils were below the maximum allowable limit prescribed by the World Health Organization (WHO), it was higher in irrigation water and vegetable samples. The spinach and okra samples showed Zn, Pb and Cd levels higher than the WHO limits. The levels of Cu, however, were at their safe limits. Metal contamination was higher in spinach than in okra. Spatial variability of metal contamination was also observed in the study. Bio-availability of metals present in soil showed a positive relationship with their total content and organic matter content of soil but no relationship was observed with soil pH. Washing of vegetables with clean water was a very effective and easy way of decontaminating the metal pollution as it reduced the contamination by 75 to 100%.  相似文献

应用小鼠淋巴细胞彗星试验对长治市5个点位地下水水质作遗传毒性研究。结果表明,各水样有机浓集物均可对小鼠淋巴细胞DNA产生不同程度的遗传损伤,随着剂量的增加,Olive尾矩与阴性对照组(0.72±0.09)相比均有显著性差异(P0.05,P0.01)。在试验浓度范围内,以彗星尾部DNA含量、彗星尾长、尾矩和Olive尾矩所指示的DNA损伤程度随剂量增大而逐渐增加,存在显著的剂量-效应关系,其相关系数0.815。  相似文献

An alkaline comet assay and a micronucleus test were carried out on erythrocytes of the European chub, Squalius cephalus L., collected in spring and autumn in 2005 and 2006 at three sampling sites in River Sava, near Zagreb, Croatia. The results of comet assay showed the lowest genotoxic influence at the least polluted site, while higher DNA damage was observed at the polluted sites. Although the basal levels of DNA damage were elevated, a clear gradation of DNA damage was found due to pollution intensity in all sampling periods. The lowest cytogenetic damage as revealed by the micronucleus test (MNT) was observed as well at the least polluted site. High variations in MN frequency were observed between sampling periods, although the number of micronucleated erythrocytes was consistently the highest one at the polluted site. The comet assay as a biomarker of genotoxic effect exhibited higher sensitivity in discriminating the genotoxic capacity of studied polluted sites while the MNT was less sensitive. However, both tests should be used together in biomonitoring studies because they can reveal different aspects of DNA damage; comet assay, the early event of genotoxic exposure, and MNT, its final result as a mutagenic potential.  相似文献

The input of anthropogenic contaminants to the aquatic environment is a major concern for scientists, regulators and the public. This is especially relevant in areas such as the Tamar valley in SW England, which has a legacy of contamination from industrial activity in the nineteenth and twentieth centuries. Following on from previous laboratory validation studies, this study aimed to assess the relationship between genotoxic and cytotoxic responses and heavy metal concentrations in two bivalve species sampled from locations along the Tamar estuary. Adult cockles, Cerastoderma edule, and blue mussels, Mytilus edulis, were sampled from five locations in the Tamar and one reference location on the south Devon coast. Bivalve haemocytes were processed for comet and neutral red retention (NRR) assays to determine potential genotoxic and cytotoxic effects, respectively. Sediment and soft tissue samples were analysed for metal content by inductively coupled plasma mass spectrometry. Sediment concentrations were consistent with the physico-chemical nature of the Tamar estuary. A significant correlation (P?=?0.05) was found between total metal concentration in sediment and C. edule soft tissues, but no such correlation was found for M. edulis samples. DNA damage was elevated at the site with highest Cr concentrations for M. edulis and at the site with highest Ni and Pb concentrations for C. edule. Analysis of NRR revealed a slight increase in retention time at one site, in contrast to comet data. We conclude that the comet assay is a reliable indicator of genotoxic damage in the field for both M. edulis and C. edule and discuss reasons for the apparent discrepancy with NRR.  相似文献

Existing methods for the comparison of genotoxic effects in the comet assay bear considerable disadvantages such as the problem to link information about concentration dependence and severity of effects. Moreover, given the lack of standardized protocols and the use of various standards, it may be extremely difficult to compare different studies. In order to provide a method for standardized comparative assessment of genotoxic effects, the concept of genotoxicity equivalents (Gene-TEQ) was developed. As potential reference compounds for genotoxic effects, three directly acting (N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), methyl-methanesulfonate, and N-methyl-N-nitrosourea) and three indirectly acting (cyclophosphamide, dimethylnitrosamine, and 4-nitroquinoline-oxide) genotoxic substances were compared with respect to their cytotoxic (neutral red) and genotoxic (comet assay) concentration-response profiles in the permanent fish cell line RTL-W1. For further comparison, two sediment extracts from the upper Danube River were investigated as environmental samples. Based on the results of cytotoxicity and genotoxicity testing, MNNG was selected as the reference compound. At several exposure levels and durations, genotoxic effects of both the other pure substances and the environmental samples were calculated as percentages of the maximum MNNG effect and related to the absolute MNNG effect (EC values). Thus, genotoxicity equivalent factors (Gene-TEQs) relative to MNNG could be calculated. Gene-TEQs can easily be applied to pure substances, mixtures and field samples to provide information about their toxicity relative to the reference compound. Furthermore, the Gene-TEQ concept allows a direct comparison of environmental samples from different laboratories.  相似文献

The present study is to assess a year round change of genotoxicity of surface water in Taihu Lake, Yangtze delta. The water samples were collected in spring, summer, autumn and winter of 2001, respectively. The bacterial L-arabinose resistance forward mutation test (ara test), along with the Ames test, was performed for the evaluation of time profile of mutagenicity of the surface water samples. The strong genotoxic potentials were detected in the near-shore area, especially in the vicinity to the major city. All the samples collected from the lake center and areas distant from dense human activity showed genotoxicity-free in year round time. Intensified human activity along the lake may play an important role for the genotoxic pollution. By comparison of the samples from year round, the winter samples showed the strongest mutagenic potential of all seasons, which indicates that meteorological and hydrological factors also affect the water quality. The impact of dense human activity, improper management and season change should be put into consideration for designing an effective remediation project in the lake basin area.  相似文献

A novel microbiological mutagenicity assay, based on bioluminescence of a marine bacterium Vibrio harveyi mutant strain, potentially suitable for monitoring and assessment of mutagenic pollution of marine environment, has been described recently. Here, we tested the use of this assay, in combination with either mussels (Mytilus sp.) or semipermeable membrane devices (SPMDs), in assessment of accumulation of mutagens in marine water (samples of Baltic Sea water were tested). Either similar results were obtained in both systems or higher signals in the SPMD-based system were detected, depending on the tested water samples. We conclude that the use of both mussels and SPMDs in combination with the V. harveyi bioluminescence mutagenicity assay is a method suitable for monitoring and assessment of accumulation of mutagenic pollutants in marine environment, but in some cases the SPMD-based system may provide a more sensitive test.  相似文献

In order to understand the water quality and the genotoxicity of various surfaces in the Guangzhou section of the Pearl River during January to December of 2008, we investigated and studied the current water situation of the surface microlayer (SML) and the subsurface water (SSW) in Guanzhou section (Zhongda Dock and Yuzhu Dock) of the Pearl River by chemical analysis and biological monitoring method (Vicia faba micronucleus test). The results showed that during these months concentrations of the indexes of the two docks water such as total nitrogen (TN), total phosphorus (TP), chemical oxygen demand (COD) exceeded the national III level of surface water quality, and the indexes of SML were much higher than the ones of SSW (P?< 0.05), and the exceeding rate of TN, TP of SML was 100%. According to the eutrophic evaluation standard, the water bodies of SML and SSW in the two docks were in a eutrophication during these months. The eutrophication and pollution of SML was more serious, and the highest index of eutrophication (E value) was up to 81.9, which also had obvious difference with COD and TP (P?< 0.05). The water of SML in the two docks enriched to N, P, and chlorophyll a (Chl. a) seriously, and the enrichment factor of SML in Zhongda Dock to N, P, and Chl. a was 0.71 ?? 2.78, 0.98 ?? 1.18, and 0.49 ?? 13.99, respectively, and the one in the Yuzhu Dock was 1.09 ?? 1.52, 1 ?? 1.14, and 0.72 ?? 4.07, respectively. Through inspecting the water genotoxicity of various layers by V. faba micronucleus test, we could know that the average annual MCN?? of SML and SSW in the two docks was 6.09??, 5.53??, 5.57??, and 5.249??, respectively. In general, the above value of SML was a little higher than the one of SSW, but there was not a remarkable difference (P?> 0.05). This research shows that the water quality in a medium to heavy eutrophication in the Guangzhou section of Pearl River belongs to national III ?? IV level, and SML has the capability of enrichment to the pollutants such as N and P and induces the increase of micronucleus rate of V. faba tip cell. The study also indicates that there may be genotoxicity matters such as N, P in water body.  相似文献

The concentration of heavy metals including Pb, Cu, Zn, Cd, Ni and Fe in different parts of Rosmarinus officinalis medicinal plant grown in Jordan were evaluated. Medicinal plant samples and soil samples were collected from three different zones in Jordan (Irbid, Al-Mafraq and Ma’an). Samples were analyzed by atomic absorption spectrometry (AAS) after chemical treatments using acid digestion procedures. Heavy metal levels in washed and unwashed in each part of R. officinalis were analyzed and compared statistically. Results show that concentrations of investigated heavy metals were varied from plant part to another part of R. officinalis. For example, Pb, Zn, Cu and Cd in most parts of R. officinalis in the three zones were concentrated in the following order: flowers, leaves, stems, whereas Pb, Ni and Fe were concentrated in order as follows: leaves, flowers and stems. Heavy metal concentrations in soil samples was evaluated and correlated with their levels in R. officinalis. Two standard reference materials of plant (SRM 1790a; spinach leaves and CRM 281; rye grass) and one standard reference materials of soil (GBW 07406) were examined to validate the method used. In general, tropical rivers have a great impact on human activities. Bioaccumulation of toxins is a worldwide problem nowadays and has been, historically, overlooked by the supervisory authorities. This study evaluated cytogenotoxic effects of Guaribas river (a Brazilian river) water during dry and rainy seasons of 2014 by using the Allium cepa test system. The toxicogenetic variables, including root growth, mitotic index, and chromosomal aberrations, were analyzed in meristematic cells of A. cepa exposed to water samples taken from the up-, within, and downstream of the city Picos (state: Piauí). The physical-chemical parameters were also analyzed to explain water quality and possible anthropogenic action. Additionally, the presence of heavy metals was also analyzed to explain water quality and possible damaging effects on eukaryotic cells. The results suggest that the river water exerted cytotoxic, mutagenic, and genotoxic effects, regardless of the seasons. In addition, Guaribas river presented physico-chemical values outside the Brazilian laws, which can be a characteristic of human pollution (domestic sewage, industrial, and local agriculture). The genetic damage was positively correlated with higher levels of heavy metals. The pollution of the Guaribas river water may link to the chemical contamination, including the action of heavy metals and their impacts on genetic instability in the aquatic ecosystem. In conclusion, necessary steps should be taken into account for further toxicogenetic studies of the Guaribas river water, as it has an influence in human health of the same region of Brazil.  相似文献

Thane district is one of the most industrialized districts in Maharashtra. The heavy industrialization and the increasing urbanization are responsible for the rapidly increasing stress on the water and soil environment of the area. Therefore, an attempt has been made through comprehensive study on the groundwater contamination and soil contamination due to heavy metals in Thane region of Maharashtra. The area undertaken for the study was Thane and its suburbans Kalwa, Divajunction, Dombivali, Kalyan, and Ulhasnagar. Industrialization and urbanization lead to generation of large volumes of wastewater from domestic, commercial, industrial, and other sources, which discharged in to natural water bodies like river and creek in this region. Groundwater samples and soil samples were collected from residential, commercial, agriculture, and industrial areas. Groundwater samples were analyzed for various water quality parameters. The analytical data shows very high concentration of total dissolved solids, total hardness, total alkalinity, chemical oxygen demand, chloride etc. Groundwater and soil samples were analyzed for ten heavy metals by inductively coupled plasma (ICPE-9000) atomic emission spectroscopy. The analytical data reveal that, very high concentration level of arsenic, cadmium, mercury, and nickel throughout the industrial area. The random dumping of hazardous waste in the industrial area could be the main cause of the groundwater and soil contamination spreading by rainwater and wind. In the residential areas the local dumping is expected to be the main source for heavy metals. A comparison of the results of groundwater with WHO guidelines show that most of the groundwater sampling station are heavily contaminated with organic matter and heavy metals. Groundwater samples are heavily contaminated by arsenic, cadmium, mercury, and nickel. Similarly, the results of heavy metals in soil compared with Swedish soil guideline values for polluted soil show that soil samples collected from residential, commercial and industrial areas are heavily contaminated by arsenic, cadmium, mercury, and nickel.  相似文献

The green alga Selenastrum capricornutum expresses a uniqueascorbate peroxidase, that responds to copper and lead. Attemptswere made to test if this peroxidase could be used to monitor thelevels of copper and lead in natural waters. When S.capricornutum was exposed to a stormwater sample, the specificactivity of the peroxidase in the cell extract was commensuratewith the combined copper and lead contents in the sample. Theperoxidase responses were also correlated with the 96 hr biomasstoxicity assay of S. capricornutum. However, unlike thebiomass toxicity assay, the peroxidase activity was not affectedby the anions in the samples. The use of this peroxidase can beused as a marker for testing heavy metal toxicity in the water.  相似文献

Previous studies have proposed that Pardosa astrigera L. Koch (Lycosidae) can be used as a biological indicator of heavy metal contamination in soil. In this study, we estimated the bioaccumulation levels and the bioconcentration factors (BCF) of four heavy metals (Cd, Cu, Pb, and Zn) in adult female P. astrigera collected from various field sites according to heavy metal content gradient and broods. The relationship between heavy metal content in the soil and that in spiders was different depending on the heavy metals and the broods. However, heavy metal content in P. astrigera increased with increasing heavy metal content in the soil. While the heavy metal content in the soil was in the order of Zn > Pb > Cu > Cd, its content in P. astrigera was in the order Zn > Cu > Cd > Pb. The BCF for Cd in both of the broods was distinctly higher than those of the other heavy metals evaluated. These results indicate that P. astrigera may be useful as a biological indicator of Cd soil contamination.  相似文献

Contaminated water are recognized as a potential source of Cryptosporidium oocysts. In Italy studies on the environmentalspread of the parasite are still scarce even if a high frequencyof serological responses could suggest high rates of endemic Cryptosporidium infections. In this investigation theoccurrence of the parasite in water samples from a municipalwastewater treatment plant was monitored with the aim to evaluatepossible correlation between oocysts and bacterial indicators,pathogens and physico-chemical parameters using a multivariateanalysis. Data collected showed high numbers of oocysts, with ahigh percentage of positive samples and a significant correlationwith pH, redox potential and total organic carbon. No correlationwas found between Cryptosporidium concentration and that of the other microrganisms. The results, through a probing statistical analysis, show that none of the selected microrganisms was a reliable predictor of the presence of theparasite.  相似文献

The glyphosate-based herbicide, Roundup®, is one of the most used pesticides worldwide. In concert with the advent of transgenic crops resistant to glyphosate, the use of this pesticide has led to an increase in agricultural yields. The objective of this study was to evaluate the genotoxic effect that the herbicide Roundup® (at a concentration of 6.67 μg/L, corresponding to 3.20 μg/L glyphosate) can have on the fish Corydoras paleatus. Treatment groups were exposed for 3, 6, and 9 days, and effects were analyzed using the piscine micronucleus test (PMT) and comet assay. A group subjected to filtered water only was used as a negative control. The PMT did not show differences between the control and exposed groups for any of the treatment times. In contrast, the comet assay showed a high rate of DNA damage in group exposed to Roundup® for all treatment times, both for blood and hepatic cells. We conclude that for the low concentration used in this research, the herbicide shows potential genotoxic effects. Future research will be important in evaluating the effects of this substance, whose presence in the environment is ever-increasing.  相似文献

Faecal contamination is a major causative factor for incidence of water borne infectious diseases. Certain hydrogen sulfide (H2S) producing enteric bacteria such as Salmonella sp. and Citrobacter sp., assosiated with coliforms, have been considered for rapid detection of recent faecal contamination in water. The basic H2S test medium, modified by adding 0.25 gm/L of L-cystine HCl, was examined for its efficiency with 90 water samples collected from 40 pipe supplies, 20 open wells, 15 hand pumps and 15 different surface water bodies (river, streams and ponds). Sterilized modified culture medium in glass vials was inoculated with 100 mL of each sample and incubated at 20, 25, 30, 35 and 44 C for 18, 24, 42, 48, 66 and 72 h. Blackening of content in incubated vials was considered positive. For comparison, most probable number (MPN) of coliform and faecal coliform per 100 mL was also estimated in each sample by multiple tube fermentation (MTF) method. H2S positive result was exhibited by 78% of samples. Coliform (> 10) and faecal coliform/100 mL were also detected in 59% of samples. Maximum H2S positive results (100%) were found with well and surface water samples incubated at 30, 35 and 44 C for 18 h. Coliform (> 10) and faecal coliform/100 mL were also detected in most of these samples. Pipe supplies (60%) and hand pumps (73%) also exhibited considerable H2S production. Coliforms and faecal coliforms were also found in significant number of these samples. Thus, the modified H2S test may prove a useful alternative indicator of faecal contamination for water quality surveillance and screening of large number of water samples in short duration, particularly during any outbreak of water borne disease among rural population.  相似文献

In order to assess the chemical properties of groundwater and soil in ophiolitic zone of Firuzabad, in east of Shahrood, Iran, 10 soil samples with regard to sensitive points (vicinity to mine, ophiolitic rocks, and villages) and 10 groundwater samples including nine samples from springs, and also one sample from a well in a village of the study area were taken. These samples were analyzed in laboratories using inductively coupled plasma method. The soil samples were also evaluated for grain size. The obtained results show that most of heavy and major elements were exceeding the permissible levels in soil and water samples in the study area. On the subject of soil quality, concentrations of elements Cr, Mn, Fe, Ca, Mg, Ca, Ni, and Zn are above permissible levels. Enrichment factor and index of geoaccumulation have been calculated for heavy and major elements of all soil samples. According to the obtained results, it may be argued that soil samples are contaminated in relation to the above-mentioned indices. Comparing the concentrations of elements with results of grain size analysis illustrates that the concentrations of Cr, Ni, Fe, Mg, and Co are positively correlated with sand fraction and the concentrations of Al, P, Mn, and Pb are directly proportional with clay fraction in soil samples. The study on water contamination suggests that concentrations of elements Cr, Ni, and Mg in groundwater samples of the study area are above the permissible levels. Some indices like metal index and heavy metal pollution index show that most of the water samples include heavy metal contamination.  相似文献
