Pollution of the aquatic environment is a global concern owing to the devastating effects of contaminants whose levels are growing at an alarming rate, and it has become a major threat for marine organisms, as well as to humans as consumers. This study has been carried out on leaping mullet obtained from Aliaga Bay, which is located on the west coast of the Aegean Sea near Izmir and hosts the world’s fifth largest shipyard, plus a broad range of industrial activities, including an oil refinery and a paper factory. The waste from these industries, combined with municipal sewer discharges, is the main cause of pollution in this region. There is no national documentation or research on the determination of pollution resulting from the industrial activities in this area. In the present study, the degree of induction of CYP4501A-associated 7-ethoxyresorufin O-deethylase (EROD) activity and immunochemical detection of CYP1A1 in the liver of leaping mullet (Lisa saliens) were used as biomarkers for the assessment of polycyclic aromatic hydrocarbon (PAH)-type organic pollutants in Aliaga Bay. Mullet caught from different locations of the bay had approximately 52 times more EROD activity than the feral fish sampled from a clean reference site near Foca, Izmir. The results of this study indicate that Aliaga Bay is highly contaminated with PAH-type organic pollutants.  相似文献

To investigate the environmental safety of waste disposal landfill sites and of land reclaimed from such sites, we evaluated the toxicity of leachate from these sites by a combination of bioassays in the Japanese killifish medaka Oryzias latipes. We tested for lethal toxicity in adult and larval medaka and for hatching inhibition of embryos from eggs. As biochemical evidence of the effects of leachate exposure, CYP1A (EROD activity) and vitellogenin (Vtg) were induced. We also bioassayed water-treated leachate and downstream river water. Leachate solution was lethal to larval and adult medaka. Embryo hatchability was inhibited, and abnormal hatching, spinal deformity and anisophthalmia occurred in embryos exposed to leachate solution. CYP1A was induced by exposure to leachate solution diluted to 1.0%, and EROD activity was significantly higher than in control. Vtg and unknown proteins were induced in the sera of male medaka exposed to the diluted leachate solution. Conventional water treatments worked effectively to remove toxic compounds but did not work well to remove element ions, including heavy metals. Treated leachate produced neither lethal toxicity nor hatching abnormalities during the exposure period. Fish toxicity tests for leachate would be useful for monitoring the environmental safety of landfill sites.  相似文献

Samples of compost-amended soil from waste dumping sites in Lagos Metropolis were extracted with dichloromethane (3 × 20 cm3) and the extract was evaporated at 35 °>C. The residue was extracted with 2,2,4-trimethylpentane, and portions of the solution were applied to a column containing silica gel from which aliphatic and aromatic hydrocarbons were eluted with n-hexane and toluene respectively. Analysis of the n-hexane fraction using gas chromatography showed the presence of a mixture of aliphatic hydrocarbons, ranging from C9 to C25, while ultraviolet analysis of the toluene fraction suggested 1,2-benzanthracene; 2,3-benzphenanthrene, chrysene and pyrene as polyaromatic compounds present in samples analyzed. The crude extracts were highly coloured and viscous. Total extractable organic residues in the 2,2,4-trimethylpentane extracts ranged from 36 to 89 mg g-1 of soil.  相似文献

The ecological risks of polynuclear aromatic hydrocarbons (PAH) in aquatic sediments will vary with both toxicity and bioavailability to aquatic biota. While there are standardized protocols to test the acute toxicity of sediment-borne compounds to aquatic invertebrates, there are none for assessing bioavailability to fish. We found that sediment-borne PAH caused an exposure-dependent induction of cytochrome P450 (CYP1A) enzymes in rainbow trout (Oncorhynchus mykiss) fingerlings exposed in semi-static 96 h bioassays, as shown by increased activity of ethoxyresorufin-o-deethylase (EROD). Assuming that PAH are taken up by trout due to partitioning from organic and inorganic constituents of sediments, we tested the effect of different test variables on bioaccumulation using induction as an index of exposure. EROD activity increased with exposure of fish to increasing volumes of sediments containing PAH, i.e., with increasing ratios of sediment to water. Uptake of single compounds from sandy sediments did not differ from uptake from clay or low organic (7% LOI — loss on ignition) sediments, but decreased when organic content was very high (58% LOI). Maximum induction was observed within 24~h of exposure and at 7.5^C relative to 15, 22, or 28^C. Storage and handling techniques had minor effects on sediment EROD induction potency. Absolute EROD activity was greater in white sucker (Catostomus commersoni) a benthic species, than in trout, a pelagic species. However, when basal (negative control) activity was accounted for, there was no difference in response between the species. Together, these experiments provide a basis for developing a standard protocol to test the bioavailability to fish of sediment-borne PAH.  相似文献

In present study, we measured the activities of some selected cytochrome P450 isozymes like ethoxyresorufin-o-deethylase (EROD), pentoxyresorufin-o-deethylase (PROD), and N-nitrosodimethylamine demethylase (NDMA-d) treating experimental rats with different environmental toxicants, namely trichloroethylene (TCE), heavy metal mixture, and ethyl alcohol individually or in combination and by exposing the Allium cepa bulbs to increasing concentration of TCE for 48 h. In animal system, NDMA-d activity displayed a remarkable amplification by 4.2-fold in the liver of alcohol ingested rats compared with control animals. TCE intake also resulted in a marked increase of around 3.8-fold and 1.2-fold in rats’ NDMA-d activity in the liver and kidney, respectively. In onion bulbs, an amazing rise in the activity of test enzymes was recorded in a dose-dependent manner. Among the said enzymes, PROD showed maximum increase up to the extent of 22-fold in comparison with control at 20 ppm of TCE exposure, whereas NDMA-d and EROD showed 11- and 9-fold increase in enzymatic activity, respectively, compared with the control. Based on this study, we conclude and advocate that all the selected isozymes of CYP450 system, viz. PROD, NDMA-d, and EROD can act as potent biomarkers in plant system for assessing the TCE pollution.  相似文献

Incineration has become one of the principal methods for municipal solid waste disposal particularly in all large cities throughout the world. Currently, the municipal solid waste incinerator fly ashes (MSWIF) are disposed of by landfill. The metal speciation of cadmium (Cd), chromium (Cr), copper (Cu), lead (Pb), and zinc (Zn) in MSWIF after been extracted with water at different pH values were examined using a sequential extraction procedure. The extraction sequence was as follows: (1) Exchangeable (NaOAc, pH 8.2), (2) Bound to Carbonates (NaOAc, pH 5.0), (3) Bound to metal oxides (HONH3Cl), (4) Bound to organic matters (HNO3, H2O2), and (5) Residual (HNO3, HCl, H2O2, HF, 1:3:1:3). The heavy metal contents in the extraction solutions were determined by inductively coupled plasma atomic emission spectrometry. The heavy metal concentrations in the different fractions obtained by sequential extraction show distinct distribution trends. The extractable fraction ranges from 25.5 to 88% of the total element content. With the pH of the extractant fall below the neutral and acidic ranges, the concentrations of heavy metals rise substantially due to the released of metals bound to carbonate fraction.  相似文献

The present study reports the first analysis of water pollutants in Sri Lankan waters using a suite of biomarkers in Nile tilapia (Oreochromis niloticus) residing in Bolgoda Lake which receives urban, industrial and domestic wastes from multiple sources. The fish were collected from the lake in the dry period (April 2005) and wet periods (September 2005, October 2006) and the levels of biomarkers viz. hepatic ethoxyresorufin O-deethylase (EROD), glutathione S-transferase (GST), metallothioneins, biliary fluorescent aromatic compounds, brain and muscle cholinesterases (ChE) were compared with those of the laboratory reared control fish and the fish obtained from a less polluted water body, Bathalagoda reservoir (reference site). The results revealed that biomarker levels of the fish collected from the reference site were not significantly different from the controls. Hepatic EROD and GST activities in fish from Bolgoda Lake were induced 4.2-16.6 folds and 1.4-3.3 folds respectively compared with the control fish. Analysis of bile in the lake fish revealed recent uptake of naphthalene, pyrene and benzo(a)pyrene type polycyclic aromatic hydrocarbons (PAHs). The induction of EROD activities in feral fish reflects the exposure of fish to aryl hydrocarbon receptor agonists including PAHs present as pollutants in the Bolgoda Lake. Cholinesterase activity in the fish inhabiting one sampling site of Bolgoda Lake was lower (22-40% inhibition) than the activity measured in the control fish indicating the presence of anticholinesterase pollutants in the area. Hepatic metallothionein levels in the lake fish were higher (1.9-3.2 folds) in comparison to the controls indicating metal exposure. The results support the potential use of these biomarkers in Nile tilapia in assessing pollution in tropical water bodies.  相似文献

Both regulated and unregulated air pollutants were detected during an intensive seasonal sampling campaign in a mixed industrial/semi-rural area on the outskirts of Rome, Italy, at two sites located opposite a hospital waste incinerator, downwind according to the direction of the prevailing local winds. Concentrations of pollutants were significantly lower than in urban atmospheres. The composition of particulate organic material indicated a heavy biogenic impact, accompanied by a lower contribution from petroleum-related processes. Both PAH and nitro-PAH group compositions of particulates were used to assess the nature and relative importance of sources. Both sites showed that different and diffuse sources contributed to local pollution with a significant contribution from traffic, proving that the hospital waste incinerator was not the main pollution source in this area. Among unregulated compounds, a series of positional isomers of nitro-PAHs and other organic compounds associated with particulate matter were investigated. In particular, 1- and 3-nitrophenanthrene identification was carried out, and they proved to be the most abundant nitro-PAHs.  相似文献

A survey of 120 coffee farmers in the Portland watershed revealed that they lacked training in pesticide application, and had no concept of the transport of residues in the environment and their impact on non-target organisms.Residues of organochlorine (OC) and organophosphorous compounds (OP) were monitored monthly for over a year in plantation soil, and water, sediment and fauna of three rivers and coastal waters of Portland watershed by gas chromatography. OP residues were not detected in any sample while OC residues were below detection levels in Rio Grande. The mean concentration ± standard error of residues detected in water (g L_1), sediment (ng g_1) and fauna (ng g_1 in wet weight) were: -endosulfan 2.7 ± 1.29, 3.8 ± 0.15 and 15.9 ± 1.61, respectively, in Spanish River, 1.56 ± 0.43, 24.3 ± 16.44 and 9.0 ± 1.86, respectively, in Swift River; 0.40 ± 0.02, 1.77 ± 0.68 ± 12.63, respectively, in sea coast; -endosulfan, 1.2 ± 0.48, 0 and 8.1 ± 1.99, respectively, in Spanish River, 1.9 ± 0.49, 0.75 ± 0.32 and 11. ± 4.32, respectively, in Swift River; 0, 5.1 ± 0.30 and 30.9 ± 15.96, respectively, in sea coast; endosulfan sulphate, 0.12 ± 0.12, 4.8 ± 1.62 and 10.0 ± 2.02, respectively, in Spanish River, 3.6 ± 0.95, 3.1 ± 0.56 and 7.9 ± 1.29, respectively, in Swift River and 0, 3.9 ± 2.17 and 24.0 ± 14.67, respectively, in sea coast.Dieldrin residues were detected only once in water (0.2 ± 0.03) and sediment (0.02 ± 0.003) of Spanish River, water (0.76 ± 0.09) of Swift River and sediment (0.1 ± 0.005) of sea coast; pp DDE was found twice in water (3.1 ± 1.53) and sediment (0.1 ± 0.007) of Swift River and water (0.8 ± 0.22) and sediment (6.14 ± 0.41) of sea coast. Considering that diesel oil is one of the most common aquatic contaminants, we compare the oxidative stress between two species of fish with different habitats (Pterygoplichthys anisitsi, benthic and Oreochromis niloticus, nektonic) exposed to diesel oil. Malondialdehyde concentrations (MDA) and the activities of ethoxyresorufin-O-deethylase (EROD), glutathione S-transferase (GST), catalase (CAT), superoxide dismutase (SOD), and glutathione peroxidase were all analyzed in the fishes' livers and gills after 2 and 7 days of exposure to different concentrations of diesel (0.1 and 0.5 mL/L). In the tilapia, MDA levels and the activities of EROD and GST activity in the liver, as well as MDA levels and the activities of GST and SOD in the gill had statistically significant differences between the treatments and between the times of exposure. For the catfish, the same occurred in the case of MDA, EROD, and SOD in the liver and in CAT and SOD in the gills. There were significant differences in the enzyme activity and lipid peroxidation between the species. Although the activity of most enzymes seemed to be more expressive and responsive to diesel in O. niloticus, diesel oil also caused significant effects on oxidative stress parameters in P. anisitsi, even though this species is benthic and thus has less access to insoluble fractions of diesel oil. Therefore, both species can be used as sentinel organisms in environmental biomonitoring of diesel contamination.  相似文献
