Abstract

Male Baladi rabbits were acutely and sub‐chronically intoxicated with cyanofenphos and profenophos. The levels of cholesterol, triglycerides, B‐lipoproteins and total proteins were determined in the serum, brain, spinal cord and sciatic nerve of rabbits. Moreover, the activities of alkaline phosphatase, acid phosphatase and glutamic‐pyruvic transaminase were determined in the liver of the animals. The whole Studie revealed that the biochemical constituents were highly affected by the tested insecticides. Also, the liver function suffered from adverse effects of the tested insecticides.  相似文献

Effect of isoflavone on cypermethrin-induced changes in enzyme activities and free radicals was studied in plasma, liver, brain and testes of male New Zealand White rabbits. Rabbits were orally given sublethal dose of cypermethrin (24 mg/kg BW; 1/100 LD50), while isoflavone (2 mg/kg BW) was given alone or in combination with cypermethrin. The tested doses were given to rabbits every other day for 12 weeks. Results obtained showed that cypermethrin significantly (P < 0.05) induced free radicals in plasma, liver, brain and testes. The activities of glutathione S-transferase (GST) (liver, brain and testes), aspartate aminotransferase (AST) and alanine aminotransferase (ALT) (liver and testes), and alkaline phosphatase (AlP) (liver) were significantly (P < 0.05) decreased due to cypermethrin administration. Contrariwise, the activities of GST, AST, ALT and AIP were increased in plasma. The activity of acetylcholinesterase (AChE) did not change in plasma and brain of treated rabbits with cypermethrin. Isoflavone alone significantly (p < 0.05) decreased the levels of free radicals in plasma, liver, brain and testes, while did not produce any significant effect on the investigated enzymes. However, isoflavone is able to reverse the changes in enzyme activities due to the effect of cypermethrin. Results concluded that isoflavone confers marked protection against cypermethrin-induced oxidative stress in rabbit's plasma, liver, brain and testes.  相似文献

Abstract

A laboratory study was conducted to determine the effect of four experimental insecticides, DOWCO429X, DPX43898, tefluthrin and trimethacarb, on enzyme activities and levels of adenosine 5'‐triphosphate (ATP) in mineral and organic soils. DOWCO429X decreased urease activity in organic soil after 7 days while a stimulatory effect was observed with most treatments after 14 days. No inhibition on acetylene (C2H2) reduction by nitrogenase was evident with any of the insecticides in either soil. With the exception of DOWCO429X and tefluthrin at 7 days in organic soil, none of the insecticide treatments inhibited dehydrogenase activity in either soil. Dehydrogenase activity, measured by formazan formation, was greater in many samples in sandy loam than the control throughout the experiment. No inhibitory effect was observed on amylase activity after 2 or 3 days in sandy soil. A stimulatory effect was apparent in many samples after 2 days in organic soil. All insecticide treatments in sandy soil reduced invertase activity at 2 days. However, none of the experimental insecticides inhibited invertase activity after 3 days. A stimulatory effect in invertase activity was apparent in most cases at 2 days in organic soil and no difference was observed after 3 days. Phosphatase activity in insecticide treated samples was equal to or greater than that of control in sandy soil after 2 h. With the exception of DPX43898, the insecticides depressed phosphatase activity in most organic soil samples. The insecticides did not affect ATP levels in either soil. Results indicated that the chemical treatments at the levels tested did not significantly affect activities of enzymes or level of ATP in both soils.  相似文献

Deleterious effects of chromium (VI) compounds are diversified affecting almost all the organ systems in a wide variety of animals. Therefore, the present study was carried out to determine the effectiveness of folic acid (FA) in alleviating the toxicity of chromium (VI) on certain biochemical parameters, lipid peroxidation, and enzyme activities of male New Zealand white rabbits. Six rabbits per group were assigned to one of four treatment groups: 0 mg FA and 0 mg Cr(VI)/kg BW (control); 8.3 microg FA/kg BW; 5 mg Cr(VI)/kg BW; 5 mg Cr(VI) plus 8.3 microg FA/kg BW, respectively. Rabbits were orally administered their respective doses every day for 10 weeks. Results obtained showed that Cr(VI) significantly (P < 0.05) increased the levels of free radicals and the activity of glutathione S-transferase (GST), and decreased the content of sulfhydryl groups (SH groups) in liver, testes, brain, kidney, and lung. The activities of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (AlP), acid phosphatase (AcP), and lactate dehydrogenase (LDH) were significantly decreased in liver and testes due to Cr(VI) administration. Also, AlP and AcP activities were significantly decreased in kidney and lung. The activity of acetylcholinesterase (AChE) was significantly decreased in brain and plasma. Contrariwise, the activities of AST and ALT were significantly increased in plasma, while AlP and AcP decreased. Chromium (VI) treatment caused a significant decrease in plasma total protein (TP) and globulin, and increased total lipids (TL), cholesterol, glucose, urea, creatinine, and bilirubin concentrations. Folic acid alone significantly decreased the levels of free radicals in liver, brain, and kidney, and increased the content of SH-group. The activities of AST, ALT, and LDH in liver; AST, ALT, AlP, AcP, and LDH in testes; AcP in kidney; AlP and AcP in lung, and LDH in brain were significantly increased. Plasma TP and albumin were increased, while urea and creatinine were decreased. The presence of FA with Cr(VI) restored the changes in enzyme activities and biochemical parameters. In conclusion, folic acid could be effective in the protection of chromium-induced toxicity.  相似文献

The effects of the herbicide (2,4-D) and six organophosphorus insecticides (Diazinon, Dimethoate, Fenitrothion, Malathion, Phenthoate and Quinalphos) on growth, photosynthesis and chlorophyll a synthesis of the fresh water green alga Chlamydomonas reinhardtii (mt +) were studied. At low concentrations (1 and 5 ppm), the herbicide and all six orgnophosphorus insecticides stimulated photosynthesis of the alga. The stimulating effects on algal growth and chlorophyll a synthesis were only observed in the presence of low concentration (1 ppm) of 2,4-D and Fenitrothion, Growth, photosynthesis and chlorophyll a synthesis of the alga were inhibited in the presence of high concentrations (10, 20 and 40 ppm) of the herbicide and all the six organophosphorus insecticides. Results also indicated that the toxicities of these organophosphorus insecticides on the tested alga were dependent on their chemical structures.  相似文献

The objective of this study was to determine the effect of either 2.5 mg/kg Body Weight or 5 mg/kg Body Weight (BW) doses of isoflavones on semen quality, testosterone levels, lipid peroxidation and semen biochemistry of male New Zealand White rabbits. Animals were given both 2.5 mg/kg BW and 5 mg/kg BW doses of isoflavones. The tested doses were given to rabbits orally every other day for 13 weeks. Treatment with isoflavones caused an increase (p < 0.05) in libido (by decreasing the reaction time), sperm concentration, sperm motility (%), total motile sperm per ejaculate (TMS), packed sperm volume (PSV), total functional sperm fraction (TFSF), total sperm output, initial fructose concentration and normal sperm, while dead sperm was reduced compared to control animals. On the other hand, ejaculate volume, initial hydrogen ion concentration (pH) and plasma testosterone levels did not change in treated animals with both doses of isoflavones as compared to control. Concentrations of thiobarbituric acid-reactive substances (TBARS), total lipids, and low density lipoprotein were significantly (p < 0.05) reduced in seminal plasma of rabbits treated with either 2.5 mg/kg BW or 5 mg/kg BW doses of isoflavones. While, the activities of glutathione S-transferase (GST), lactate dehydrogenase (LDH), aspartate aminotransferase (AST), alanine aminotransferase (ALT), acid phosphatase (AcP), and alkaline phosphatase (AlP) were significantly (p < 0.05) increased in seminal plasma of treated animals. Also, total cholesterol, percentage cholesterol (out of total lipids), and high density lipoprotein were significantly (p < 0.05) increased, while triglyceride did not change in seminal plasma of treated animals. Supplementation at either level of isoflavones did not cause changes in live body weight (LBW), dry matter intake (DMI), and relative weights of testes and epididymis. The present results showed that either 2.5 mg/kg BW or 5 mg/kg BW doses of isoflavones caused an improvement of some semen characteristics and did not have negative effects on male fertility.  相似文献

Carbamates (CB) are used as insecticides and some of them have been registered as human drugs. The mechanism of CB poisoning involves reversible inhibition of acetylcholine esterase. In the present study, we investigated changes in liver ultrastructure in rabbits (Oryctolagus cuniculus) which were administered bendiocarb for 3, 10, 20, and 30 days. Rabbits in all experimental groups received capsules of bendiocarb (96% Bendiocarb, Bayer, Germany) per os daily at a dose of 5 mg/kg of body weight, and after day 11 received the same dose every 48 h. The observed changes were only moderate, focal, and the effect on the liver was not uniform. On the third day of the experiment, injured hepatocytes had dilated bile capillaries with reduced microvilli. There were no visible alterations in the intercellular contacts. Nuclei of these cells were irregular in shape. Many hepatocytes showed considerable increase in the number of peroxisomes. On day 10 of the experiment, the number of peroxisomes was reduced. Other changes, such as dilated rough endoplasmic reticulum and proliferation of smooth endoplasmic reticulum were observed on day 20. The number of lipid droplets in hepatocytes gradually increased. Usually they were present in low numbers, but on day 30 of the experiment their number increased significantly. They coalesced and formed a single lipid droplet which changed the shape of the nuclei. The results presented in this study indicate that both short and long-term administration of bendiocarb affects the liver ultrastructure. At the same time we also observed rapid onset of regeneration of the damaged tissue through activation of hepatocytes and oval cells.  相似文献

Three insecticides, monocrotophos, quinalphos, and cypermethrin, were applied at 0, 5, 10, and 25 microg g(-1) either singly or in combination to a black clay soil to investigate their effects on the soil microflora and dehydrogenase activity. All three insecticides significantly enhanced the proliferation of bacteria and fungi and the soil dehydrogenase activity even at the highest level of 25 microg g(-1). Monocrotophos or quinalphos in combination with cypermethrin at tested levels interacted significantly to yield additive, synergistic, and antagonistic responses toward bacteria and fungi and dehydrogenase activity in soil. Antagonistic interactions were more pronounced toward soil microflora and dehydrogenase activity when the two (monocrotophos or quinalphos + cypermethrin) insecticides were present together in the soil at highest level (25 + 25 microg g(-1)), whereas synergistic or additive responses occurred at lower level with the same combination of insecticides in soil.  相似文献

Four organophosphorus insecticides and the active metabolites of two phosphorothionate insecticides were tested for their toxic and teratogenic effects on embryos of Xenopus laevis. All compounds caused dose-dependent developmental defects, such as abnormal pigmentation, abnormal gut development, notochordal defects and reduced growth. Malathion, malaoxon, parathion, and paraoxon produced severe defects, while monocrotophos and dicrotophos produced considerably milder defects. All compounds reduced NAD+ levels to a similar extent, regardless of the severity of the defects induced. Thus some commonly used organophosphorus insecticides and their metabolites are teratogenic to Xenopus embryos, but reduced NAD+ level does not appear to be important in causing the developmental defects.  相似文献

The acute toxicity of two organophosphorus (OP) insecticides, azinphosmethyl and acephate, was evaluated in the mummichog, Fundulus heteroclitus. In addition, the effects of certain intrinsic (sex) and extrinsic (salinity and multiple toxicant interraction) variables on the toxic response were also investigated. Azinphosmethyl was by far the most toxic of the two OP insecticides with a 96h LC50 approximately 100,000 x lower than that for acephate. Slight sex differences were observed in the sensitivity of mummichogs to each of the OP insecticides with male fish being marginally more sensitive. Significant effects of low salinity stress were observed only with acephate exposure and, in this case, low salinity appeared to be slightly protective. In general, all of the insecticide mixtures (azinphosmethyl/endosulfan, azinphosmethyl/fenvalerate and acephate/fenvalerate) tested exhibited simple additive toxicity.  相似文献

Profenofos showed maximum effect on protein synthesis in both tissues at a dose of 0.2 microgram/mL, while the maximum effect on RNA synthesis occurred at 0.2 microgram/mL, while the maximum effect on RNA synthesis occurred at 0.2 microgram mL in the liver and at 2 micrograms/mL in the muscle. Fenvalerate caused maximum stimulation in both liver protein and RNA synthesis at a dose of 2 micrograms/mL, and maximum inhibition in the muscle at 10 and 0.2 micrograms/mL for protein and RNA synthesis respectively. The maximum effect of Dimilin on both tissues was reached at 5 micrograms/mL for protein synthesis and at 0.2 microgram/mL for RNA synthesis. The effect of Dimilin on RNA synthesis was more pronounced in both tissues than its effect on protein synthesis, but this trend was reversed in the case of profenofos and fenvalerate. Present data also showed antagonism between these insecticides on the rate of protein and RNA synthesis.  相似文献

Abstract

A laboratory study was conducted to examine the effects of five insecticides on microbial and enzymatic activities important to fertility in sandy soil. Cyfluthrin significantly increased bacterial populations after 2 wks. Imidacloprid showed an inhibitory effect on fungal numbers after 2 wks incubation while the others did not affect fungal population. No inhibitory effect was observed on nitrification of soil indigenous nitrogen. All treatments stimulated S‐oxidation after 4 wks. With the exception of cyfluthrin and imidacloprid after 2 wks, denitrification in sandy soil indicated that all treatment inhibited denitrification throughout the experiment. No inhibitory effects on biomass‐c were observed during 2‐wk periods. An inhibitory effect was observed on amylase after 1 wk while significant recovery was observed after 3 wks. With the exception of HgCl2, no effect was observed on reducing sugar formation for 2 wks with all treatments. Formazan formation resulting from dehydrogenase activity was significantly greater with tebupirimphos and Aztec for 1 wk. All treatments supressed phosphatase activity for 1 wk, while none of the treatments suppressed phosphatase activity after 2 wks. Amitraz, tebupirimphos and Aztec inhibited urease activity for 1 wk. With the exception of tebupirimphos, no treatments affected N2‐fixation in soil. Although short‐lived inhibitory effects on activities of microbes and enzymes were caused by the insecticides, the soil indigenous microbes can tolerate the chemicals used for control of soil pests.  相似文献

This study was undertaken to investigate the effects of sublethal concentration of three different classes of insecticides (carbamate, organophosphate, and pyrethroid compounds) on the freshwater fish Corydoras paleatus. For this purpose, fish were exposed for 96 hours to commercial pesticides. Different biomarkers were analyzed as levels of lipid peroxidation (LPO), piscine micronucleus test, and enzymatic activities of catalase (CAT), glutathione S-transferase (GST), and acetylcholinesterase (AChE). The brain AChE was inhibited with carbaryl and methyl parathion, but no inhibition was observed with deltamethrin. The insecticides did not cause oxidative stress or genotoxic effects at the tested concentrations. Further studies are needed to elucidate the biotransformation of Corydoras paleatus insecticides and a possible resistance mechanism.  相似文献

Abstract

The objective of this research was to investigate the potential damage caused by the residual concentrations of the insecticides Regent® WS 800 and Curbix® SC 200, containing fipronil and ethiprole, respectively as active ingredients, on the liver of Oreochromis niloticus. The analyses of HSP70 shock protein labelling and cell death process by TUNEL method were performed in order to measure the effects of the exposure of cell repair system of fish to both insecticides. Statistical analyses showed no significant molecular damage to the hepatic tissue of animals. Nevertheless, variations in HSP70 and DNA fragmentation levels, endpoint of cell repair system response and cellular death, respectively, were observed in several groups. These results indicate that the cell repair machinery was efficient when in contact with residual concentrations of insecticides. However, the DNA fragmentation detected by the TUNEL method suggests that even in face of the cytoprotective action of the HSP70 protein, there are damages that become irreparable. To finish, it is worth mentioning that given the results obtained from residual concentrations, use in the field should be with caution.  相似文献

The effect of intratracheal administration of DDT (5 mg/100 g body weight) or endosulfan (1 mg/100 g body weight) to rats for three consecutive days, has been studied on liver lipid metabolism. The administration of DDT but not endosulfan significantly increased the liver weight and the microsomal protein contents. Both DDT and endosulfan treatments significantly increased the contents of microsomal phosphatidylcholine (PC), total-free- and esterified cholesterol. The distribution of unsaturated fatty acids of microsomal PC and PE was increased by DDT treatment. The intratracheal administration of DDT caused fatty infiltration of liver which was probably due to increased synthesis of triglycerides (TG). This is supported by the increased incorporation of radioactive palmitate-1-14C into microsomal TG. However, the increased incorporation of palmitate-1-14C into microsomal PC and phosphatidylethanolamine (PE) after the DDT treatment, was due to the increased transacylation reaction supported by the decreased activity of microsomal phospholipase A. The intratracheal administration of endosulfan did not have pronounced effect on liver fatty infiltration, or transacylation reaction in microsomal PC and PE. However, the results have shown that the treatments of DDT or endosulfan increased the PC contents and the incorporation of radioactive [methyl-3H]choline into PC of microsomes, resulting the increased synthesis of PC via CDPcholine pathway. Thus, the intratracheally administered DDT or endosulfan to rats showed that both the insecticides cause manifestations in the biochemistry of microsomal membrane lipids, although the effects of DDT being more pronounced. Therefore, the translocation effects of these insecticides or metabolites from lung to liver is established.  相似文献

The acute toxicity of permethrin, resmethrin, and cypermethrin to four species of aquatic non-target invertebrate organisms, found in estuarine and freshwater ecosystems, was evaluated. Artemia franciscana and Brachionus plicatilis larvae, as estuarine organisms, and Brachionus calyciflorus and Thamnocephalus platyurus larvae, as freshwater organisms, were exposed for 24 h to concentrations of these pyrethroids, and the LC(50) values were compared. The freshwater organisms were more sensitive to these pyrethroids than estuarine organisms tested. A. franciscana larvae were more tolerant organisms than B. plicatilis larvae. The freshwater organisms tested have demonstrated to be a good alternative to the standard acute toxicity assays using Daphnia, although Brachionus plycatilis larvae were more sensitive to these pyrethroid insecticides than T. platyurus. The behavior of chlorantraniliprole (CAP) and dinotefuran (DNF) insecticides was investigated in clay loam soil, a common type of the Egyptian soil. Effect of temperature, pH and particle size of the soil on the adsorption process was studied. Adsorption isotherm by bulk soil and its constituents; humic acid (HA), clay, silt and sand fractions was measured using batch equilibration technique. The results showed that the adsorption of the insecticides tested was significantly affected by the temperature and was a spontaneous interfacial process in the soil. Freundlich model accurately predicted the adsorption behavior of both insecticides. The interaction between soil and insecticides was endothermic and the highest adsorption for CAP and DNF was obtained at pH 9. However, the effect of pH on the adsorption of DNF was lower than that of CAP. Sorption of CAP and DNF on HA fraction was significantly greater than on clay fraction and bulk soil. In addition, the adsorption was significantly increased with particle size decrease. It could be inferred that the adsorption of CAP and DNF on clay loam soil was physical in nature and greatly influenced by the soil components, pH and temperature.  相似文献
