We examined the influence of macronutrient (nitrate and phosphate) additions on Ni uptake by phytoplankton (Prorocentrum donghaiense and Skeletonema costatum) and its subsequent transfer to marine copepods (Calanus sinicus and Labidocera euchaeta). Ni uptake by phytoplankton after 24h of exposure was markedly dependent on nutrient conditions, with a higher nutrient quota facilitating Ni accumulation in the algae. Trophic transfer was quantified by measurements of the Ni assimilation efficiency in C. sinicus and L. euchaeta, feeding on the algae under different nutrient treatments. Ni assimilation efficiency generally increased with an increase of nutrient concentration in the algae. A significant positive-correlation was found between the Ni assimilation efficiencies of the copepods and the %intracellular Ni in the algal cells. However, ambient nutritional conditions had little effect on the physiological turnover rate constant of Ni by copepods. Thus, nutrient enrichment may lead to an increase in Ni uptake and transfer in marine plankton.  相似文献

A unicellular green microalga, Chlorella vulgaris, entrapped in calcium alginate as algal beads were employed to remove nutrients (N and P) from simulated settled domestic wastewater. A significantly higher nutrient reduction was found in bioreactors containing algal beads (at concentrations ranging from 4 to 20 beads ml(-1) wastewater) than the blank alginate beads (without algae). A complete removal of NH(4)(+)-N and around 95% reduction of PO(4)(3-)-P was achieved within 24 h of treatment in bioreactors having the optimal algal bead concentration (12 beads ml(-1), equivalent to 1:3 algal beads:wastewater, v/v). The NH(4)(+)-N removal was significantly lower at low (around 4 beads ml(-1)) and high (>15 beads ml(-1)) algal bead concentrations. On the other hand, the effect of bead concentration on phosphate removal was less obvious, and bead concentrations ranging from 8 to 20 beads ml(-1) showed comparable percentages of phosphate reduction. Algal uptake and adsorption on alginate gels were found to be the major processes involved in the removal of N and phosphate in the present study. In addition, NH(4)(+)-N could be lost via ammonia volatilization while PO(4)(3-)-P was removed by chemical precipitation, as alkaline pH was recorded in the immobilized microalgal treatment system.  相似文献

The uptake of Cd, Se(IV) and Zn by the freshwater alga Scenedesmus obliquus and the subsequent transfer and release budget in Daphnia magna were investigated under different nutrient additions and cell incubation conditions. An increase in ambient phosphate concentrations from 0.5 micromol l(-1) to 50 micromol l(-1) significantly increased the intracellular accumulation of Cd (by 18x) and Zn (by 5x), but decreased the accumulation of Se (by 126x) in the alga. The percentage of these metals distributing in the intracellular pool of algae also increased substantially with increasing ambient P concentrations. Nitrate addition from 5.0 to 200 micromol l(-1) did not influence the uptake of any of the three metals, although a significant decrease in the intracellular Se distribution was observed. Radiolabeled algae under different nutrient manipulations (semi-continuous culture, starvation, and P-pulse treatments) were used to measure trophic transfer assimilation efficiency (AE) in Daphnia. When the algal cells were grown in a semi-continuous culture, starved for N and P, or were treated with P-pulse, the AEs of Cd and Zn were generally independent of the nutritional conditions, but the Se AE was significantly affected by different P levels. The efflux rate constants, determined during 10 d depuration following 7 days of dietary uptake, decreased significantly for Cd and Zn, but were relatively constant for Se with increasing P concentration. N-addition caused no effect on the metal efflux rate constants. P- or N-additions did not influence the release budget (including molting, neonates, excretion and feces) for all three elements in Daphnia. Our study indicated that phosphate enrichment may substantially increase metal uptake in green alga S. obliquus. Responses of trophic transfer in Daphnia to nutrient enrichment were metal specific. P-enrichment can possibly lead to considerable decrease on Se transfer from algae to zooplankton.  相似文献

The role of root hairs in Cd acquisition from soil was investigated in three pot experiments using a root hairless mutant (bald root barley, brb) and its wild-type (WT) cultivar of barley (Hordeum vulgare). brb had significantly lower concentrations and lower total amounts of Cd in shoots than WT. The Cd uptake efficiency based on total root length was 8-45% lower in brb than in WT. The difference between brb and WT increased with increasing extractable Cd in soil under the experimental conditions used. Additions of phosphate to soil decreased Cd extractability. Both soil and foliar additions of phosphate decreased root length, and root hair formation in WT. These effects resulted in decreased Cd uptake with increasing P supply. Cd uptake in WT correlated significantly with root length, root hair length and density, and soil extractable Cd. Root hairs contribute significantly to Cd uptake by barley.  相似文献

The effect of arbuscular mycorrhizal fungi (AMF) on growth and element uptake by Ni-hyperaccumulating plant, Berkheya coddii, was studied. Plants were grown under laboratory conditions on ultramafic soil without or with the AM fungi of different origin. The AM colonization, especially with the indigenous strain, significantly enhanced plants growth and their survival. AMF affected also the elemental concentrations that were studied with Particle-induced X-ray emission (PIXE). AMF (i) increased K and Fe in shoots, Zn and Mn in roots, P and Ca both, in roots and shoots; (ii) decreased Mn in shoots, Co and Ni both, in shoots and roots. Due to higher biomass of mycorrhizal plants, total Ni content was up to 20 times higher in mycorrhizal plants compared to the non-mycorrhizal ones. The AMF enhancement of Ni uptake may therefore provide an improvement of a presently used technique of nickel phytomining.  相似文献

The reasons why some cultivars of hot pepper (Capsicum annuum L.) accumulate low levels of Cd are poorly understood. We aimed to compare the characteristics of Cd uptake and translocation in low-Cd and high-Cd hot pepper cultivars by determining the subcellular locations and chemical forms of Cd, and its distribution among different plant organs. We conducted a hydroponic experiment to investigate the subcellular distribution and chemical forms of Cd in roots, stems, and leaves of a low-Cd (Yeshengchaotianjiao, YCT) and a high-Cd cultivar (Jinfuzaohuangjiao, JFZ). The results showed that the concentrations of Cd in almost all subcellular fractions of roots, and in all chemical forms in roots, were higher in YCT than in JFZ. Compared with YCT, JFZ had higher Cd concentrations in almost all subcellular fractions of stems and leaves, and higher Cd concentrations in almost all chemical forms in stems and leaves. Additionally, YCT had significantly higher total Cd accumulation but a lower Cd translocation rate compared with JFZ. In general, the results presented in this study revealed that root-to-shoot Cd translocation via the xylem is the key physiological processes determining the Cd accumulation level in stems and leaves of hot pepper plants. Immobilization of Cd by the cell walls of different organs is important in Cd detoxification and limiting the symplastic movement of Cd.  相似文献

Adverse effects of manufactured nickel oxide nanoparticles on the microalgae Chlorellavulgaris were determined by algal growth-inhibition test and morphological observation via transmission electron microscopy (TEM). Results showed that the NiO nanoparticles had severe impacts on the algae, with 72 h EC(50) values of 32.28 mg NiOL(-1). Under the stress of NiO nanoparticles, C. vulgaris cells showed plasmolysis, cytomembrane breakage and thylakoids disorder. NiO nanoparticles aggregated and deposited in algal culture media. The presence of algal cells accelerated aggregation of nanoparticles. Moreover, about 0.14% ionic Ni was released when NiO NPs were added into seawater. The attachment of aggregates to algal cell surface and the presence of released ionic Ni were likely responsible for the toxic effects. Interestingly, some NiO nanoparticles were reduced to zero valence nickel as determined by X-ray diffraction (XRD) and X-ray photoelectron spectroscopy (XPS) analysis. The maximum ratios of nickel reduction was achieved at 72 h of exposure, in accordance with the time-course of changes in soluble protein content of treated C. vulgaris, implying that some proteins of algae are involved in the process. Our results indicate that the toxicity and bioavailability of NiO nanoparticles to marine algae are reduced by aggregation and reduction of NiO. Thus, marine algae have the potential for usage in nano-pollution bio-remediation in aquatic system.  相似文献

Endophytic bacteria improve phytoremediation of Ni and TCE co-contamination   总被引:1,自引:0,他引:1  
The aim of this work was to investigate if engineered endophytes can improve phytoremediation of co-contaminations by organic pollutants and toxic metals. As a model system, yellow lupine was inoculated with the endophyte Burkholderia cepacia VM1468 possessing (a) the pTOM-Bu61 plasmid, coding for constitutive trichloroethylene (TCE) degradation, and (b) the ncc-nre Ni resistance/sequestration system. Plants were exposed to Ni and TCE and (a) Ni and TCE phytotoxicity, (b) TCE degradation and evapotranspiration, and (c) Ni concentrations in the roots and shoots were determined. Inoculation with B. cepacia VM1468 resulted in decreased Ni and TCE phytotoxicity, as measured by 30% increased root biomass and up to 50% decreased activities of enzymes involved in anti-oxidative defence in the roots. In addition, TCE evapotranspiration showed a decreasing trend and a 5 times higher Ni uptake was observed after inoculation.  相似文献

Subcellular selenium (Se) distributions in the oligochaete Tubifex tubifex and in the insect Chironomus riparius did not vary with Se exposure duration, which was consistent with the observations that the duration of prey Se exposure had little influence on either Se assimilation or loss by a predatory insect (the alderfly Sialis velata). However, these two prey types differed in how Se was distributed in their cells. Overall, the predator assimilated a mean of 66% of the Se present in its prey, which was similar to the mean percentage of Se in prey cells (62%) that was theoretically available for uptake (that is, Se in the protein and organelle fractions). Likewise, data for cadmium, nickel and thallium suggest that predictions of trace element transfer between prey and predator are facilitated by considering the subcellular partitioning of these contaminants in prey cells.  相似文献

This study explored the optimisation of a method of extracting allelochemicals from Pistia stratiotes Linn., identified the optimal dose range for the allelochemicals’ anti-algal effect and investigated their impact on the growth of Microcystis aeruginosa, as well as the production and release of microcystin-LR (MC-LR). Based on measured changes in algal cell density and chlorophyll a (Chl-a) content, the allelochemicals were confirmed to have the strongest anti-algal effect with the lowest half-effect concentration of 65 mg L?1 when they were extracted using ethyl acetate as the extraction solvent, 1:20 g mL?1 as the extraction ratio and 1 h as the extraction time. The allelochemicals extracted from P. stratiotes using this optimal method exhibited the strongest inhibitory effect on the growth of algae when used within a dose range of 60–100 mg L?1; the relative inhibitory ratio reached 50–90 %, and Chl-a content reduced 50–75 % in algae cell cultures within 3–7 days. In addition, the extracted allelochemical compounds demonstrated no significant impact on the extracellular release of MC-LR during the culturing period. The amount of intracellular MC-LR per 106 algal cells increased depending on the increasing dose of allelochemicals from P. stratiotes after 7 days of culturing and maintained stability after 16 days. There was no increase in the total amount of MC-LR in the algal cell culture medium. Therefore, the application of allelochemicals from P. stratiotes to inhibit M. aeruginosa has a high degree of ecological safety and can be adopted in practical applications for treating water subjected to algae blooms because the treatment can effectively inhibit the proliferation of algal cells without increasing the release of cyanotoxin.  相似文献

We evaluated the distribution of 15 metal ions, namely Al, Cd, Cu, Cr, Fe, La, Mn, Ni, Pb, Sc, Ti, V, Y, Zn and Zr, in the soil of a contaminated site in Piedmont (Italy). This area was found to be heavily contaminated with Cu, Cr and Ni. The availability of these metal ions was studied using Tessier’s sequential extraction procedure: the fraction of mobile species, which potentially is the most harmful for the environment, was much higher than that normally present in unpolluted soils. This soil was hence used to evaluate the effectiveness of treatment with vermiculite to reduce the availability of the pollutants to two plants, Lactuca sativa and Spinacia oleracea, by pot experiments. The results indicated that the addition of vermiculite significantly reduces the uptake of metal pollutants by plants, confirming the possibility of using this clay in amendment treatments of metal-contaminated soils. The effect of plant growth on metal fractionation in soils was investigated. Finally, the sum of the metal percentages extracted into the first two fractions of Tessier’s protocol was found to be suitable in predicting the phytoavailability of most of the pollutants present in the investigated soil.  相似文献

Bioaccumulation, subcellular distribution, and acute toxicity of yttrium (Y) were evaluated in Nymphoides peltata. The effects of Y concentrations of 1–5 mg L?1 applied for 4 days were assessed by measuring changes in photosynthetic pigments, nutrient contents, enzymatic and non-enzymatic antioxidants, and ultrastructure. The accumulation of Y in subcellular fractions decreased in the order of cell wall > organelle > soluble fraction. Much more Y was located in cellulose and pectin than in other biomacromolecules. The content of some mineral elements (Mg, Ca, Fe, Mn, and Mo) increased in N. peltata, but there was an opposite effect for P and K. Meanwhile, ascorbate, and catalase activity decreased significantly for all Y concentrations. In contrast, peroxidase activity was induced, while initial rises in superoxide dismutase activity and glutathione content were followed by subsequent declines. Morphological symptoms of senescence, such as chlorosis and damage to chloroplasts and mitochondria, were observed even at the lowest Y concentration. Pigment content decreased as the Y concentration rose and the calculated EC50 and MPC of Y for N. peltata were 2 and 0.2 mg L?1 after 4 days of exposure, respectively. The results showed that exogenous Y was highly available in water and that its high concentration in water bodies might produce harmful effects on aquatic organisms. N. peltata is proposed as a biomonitor for the assessment of metal pollution in aquatic ecosystems.  相似文献

This study investigated the behaviors and mechanisms of phosphate adsorbed onto manganese (Mn) oxide-doped aluminum (Al) oxide (MODAO). The isotherm results demonstrated that the maximum amount of phosphorus (P) adsorbed onto MODAO was 59.8 mg/g at T?=?298 K (pH 6.0). This value was nearly twice the amount of singular AlOOH and could increase with rising temperatures. The kinetic results illustrated that most of the P was adsorbed onto MODAO within 5 h, which was shorter than the equilibrium time of phosphate adsorption onto AlOOH. The Elovich model effectively described the adsorption kinetic data of MODAO because of its heterogeneous surface. The optimal solution pH for phosphate removal was approximately 5.0 because of electrostatic interaction effects. Meanwhile, the decrease in P uptake with increasing ion strength suggested that phosphate adsorption occurred through an outer-sphere complex. Phosphates would compete for adsorption sites on the surface of MODAO in the presence of fluoride ion or sulfate. In addition, the spectroscopic analysis results of Fourier transform infrared spectroscopy and X-ray photoemission spectroscopy indicated that removal mechanisms of phosphate primarily include adhesion to surface hydroxyl groups and ligand exchange.  相似文献

Saxitoxin (STX) is a cyanotoxin, which can cause neurotoxic effects and induce ecological changes in aquatic environments, a potential risk to public and environmental health. Many studies of cytotoxicity on animal cells and algae have been performed, although few compare the toxic effects between the two models. In this sense, we investigated the oxidative stress induced by STX (0.4-3.0 nM) in two different cellular models: Neuro-2A (N2A) cells and Chlamydomonas reinhardtii alga by quantification of malondialdehyde (MDA) levels as indicative of lipid peroxidation (LPO). Also was evaluated the antioxidant defense of these cells systems after exposure to STX by the addition of antioxidants in N2A cells culture, and by the measure of antioxidants enzymes activity in C. reinhardtii cells. The MDA levels of N2A cells increased from 15% to 113% for 0.4 and 3.0 nM of STX, respectively, as compared to control. Superoxide-dismutase and catalase did not appear to protect the cell from STX effect while, in cells treated with vitamin E, the rates of MDA production decreased significantly, except for higher concentrations of STX. No MDA productions were observed in algal cells however some effects on antioxidant enzymes activity were observed when algae were exposed to 3.0 nM STX. Our results indicate that the concentrations of STX that may induce oxidative stress through LPO are different in animal and phytoplankton communities. A combination of algal and animal bioassays should be conducted for reliable assessment of oxidative stress induced by STX.  相似文献

This research investigated the feasibility of changing waste into useful materials for water treatment and proposed a coagulation–magnetic separation technique. This technique was rapid and highly effective for clearing up harmful algal blooms in freshwater and mitigating lake eutrophication. A magnetic coagulant was synthesized by compounding acid-modified fly ash with magnetite (Fe3O4). Its removal effects on algal cells and dissolved organics in water were studied. After mixing, coagulation, and magnetic separation, the flocs obtained from the magnet surface were examined by SEM. Treated samples were withdrawn for the content determination of chlorophyll-a, turbidity, chemical oxygen demand (COD), total nitrogen, and total phosphorus. More than 99 % of algal cells were removed within 5 min after the addition of magnetic coagulant at optimal loadings (200 mg L?1). The removal efficiencies of COD, total nitrogen, and phosphorus were 93, 91, and 94 %, respectively. The mechanism of algal removal explored preliminarily showed that the magnetic coagulant played multiple roles in mesoporous adsorption, netting and bridging, as well as high magnetic responsiveness to a magnetic field. The magnetic–coagulation separation method can rapidly and effectively remove algae from water bodies and greatly mitigate eutrophication of freshwater using a new magnetic coagulant. The method has good performance, is low cost, can turn waste into something valuable, and provides reference and directions for future pilot and production scale-ups.  相似文献

Phosphorus (P) is a key biological element and limiting nutrient in aquatic environments. Phosphate (+5) is traditionally associated with the P nutrient supply. However, phosphite (+3) has recently generated a great deal of interest, because of the possibility that it is a P source based on recognition of its vital role in the original life of the early earth. This study investigated whether phosphite can be an alternative P source for Microcystis aeruginosa PCC 7806, one of the predominant bloom species in freshwater systems. The results indicated that M. aeruginosa could not utilize phosphite as a sole P-nutrient directly for cell growth at any concentration, but that phosphite could boost cell numbers and chlorophyll a (Chl-a) content as long as phosphate was provided simultaneously. Specifically, Chl-a production increased sharply when 5.44 mg P L−1 phosphite was added to 0.54 mg P L−1 phosphate medium. Analysis of the maximum yield of PSII indicated that phosphite may stimulate the photosynthesis process of cells in phosphate-phosphite medium. In addition, phosphite failed to support cell growth, even though it more readily permeated the cells in P-deficient medium than in P-sufficient medium. Alkaline phosphatase activity (APA) analysis indicated that, unlike organic P, phosphite inhibits the response of cells to deficient P status, especially under P-deprived conditions.  相似文献

This study investigates biomass, density, photosynthetic activity, and accumulation of nitrogen (N) and phosphorus (P) in three wetland plants (Canna indica, Typha augustifolia, and Phragmites austrail) in response to the introduction of the earthworm Eisenia fetida into a constructed wetland. The removal efficiency of N and P in constructed wetlands were also investigated. Results showed that the photosynthetic rate (P n), transpiration rate (T r), and stomatal conductance (S cond) of C. indica and P. austrail were (p?<?0.05) significantly higher when earthworms were present. The addition of E. fetida increased the N uptake value by above-ground of C. indica, T. augustifolia, and P. australis by 185, 216, and 108 %, respectively; and its P uptake value increased by 300, 355, and 211 %, respectively. Earthworms could enhance photosynthetic activity, density, and biomass of wetland plants in constructed wetland, resulting in the higher N and P uptake. The addition of E. fetida into constructed wetland increased the removal efficiency of TN and TP by 10 and 7 %, respectively. The addition of earthworms into vertical flow constructed wetland increased the removal efficiency of TN and TP, which was related to higher photosynthetic activity and N and P uptake. The addition of earthworms into vertical flow constructed wetland and plant harvests could be the significantly sustainable N and P removal strategy.  相似文献

Two living Chlorella species were used to remove nickel from solution containing 30 micrograms Ni ml-1 in 10 successive cycles. The present study also examined the continued viability of these two algal species after repeated exposure to nickel. The two species of Chlorella were Chlorella vulgaris (commercially available) and WW1 (indigenous species isolated from domestic sewage and was tentatively identified as Chlorella miniata). The nickel removal percentage of WW1 cells was maintained at around 85% in the first five cycles, then declined slightly from the fifth cycle onwards, and finally achieved around 70% removal at the end of the 10th cycle. On the contrary, the removal efficiency of C. vulgaris declined from 50 to 30% during the 10 cycles of nickel bisorption. At the end of these 10 successive cycles, WW1 accumulated a substantial amount of Ni2+ (the cumulative cellular Ni concentration was 0.92% dry w.), while the value was only 0.17% in the case of C. vulgaris. These results suggest that the local isolate, WW1, had more consistent and satisfactory ability for removing Ni than the commercial C. vulgaris. Both algal species were still capable of dividing after each nickel treatment cycle, suggesting that the cells were not killed even when significant amounts of nickel were adsorbed/absorbed. However, Ni exposure adversely affected the physiological activity of algal cells as reflected by the decline in division rate and chlorophyll-a activity in both species. Such negative effects became more obvious as the number of cyclic treatments was increased. Nevertheless, WW1 cells appeared to recover from nickel treatment when re-cultivated in commercial medium for 2 weeks.  相似文献

The accumulation and fraction distribution of Ni(II) in sludge was determined, and their effect on the performance of sequencing batch reactor (SBR) systems was evaluated at laboratory scale. The results showed that the removal efficiencies of substrates decreased significantly with increasing feeding concentration of Ni(II) into SBRs. The concentration of Ni(II) fed into the SBRs was significantly positively correlated with the Ni(II) contents accumulated in the sludge, while it was negatively correlated with the biomass in the SBRs. The accumulated Ni(II) in the sludge was distributed mainly in the available fraction, accounting for 75.8–90.0 % of the total Ni(II) content. The accumulated content of Ni(II) in each sludge fraction could be predicted satisfactorily by the feeding Ni(II) concentrations in the solutions. As compared with the total contents and other chemical fractions, Ni(II) in the oxidizable fraction in sludge exhibited more important inhibition effects on sludge microorganisms in the SBRs.  相似文献

The uptake of the organophosphates tris(2-chloroethyl) phosphate (TCEP), tris(1-chloro-2-propyl) phosphate (TCPP), tributyl phosphate (TBP), the insect repellant N,N-diethyl toluamide (DEET), and the plasticizer n-butyl benzenesulfonamide (NBBS) into plants was studied in greenhouse experiments and simulated with a dynamic physiological plant uptake model. The calibrated model was coupled to a tipping buckets soil transport model and a field scenario with sewage sludge application was simulated. High uptake of the polar, low-volatile compounds TCEP, TCPP, and DEET into plants was found, with highest concentrations in straw (leaves and stem). Uptake into carrot roots was high for TCPP and TBP. NBBS showed no high uptake but was rapidly degraded. Uptake into barley seeds was small. The pattern and levels of uptake could be reproduced by the model simulations, which indicates mainly passive uptake and transport (i.e., by the transpiration stream, with the water) into and within the plants. Also the field simulations predicted a high uptake from soil into plants of TCEP, TCPP, and DEET, while TBP is more likely taken up from air. The BCF values measured and calculated in the greenhouse study are in most cases comparable to the calculated values of the field scenario, which demonstrates that greenhouse studies can be suitable for predicting the behavior of chemicals in the field. Organophosphates have a high potential for bioaccumulation in crops and reach agricultural fields both via sewage sludge and by atmospheric deposition.  相似文献
