Fish live in direct contact with their immediate external environment and, therefore, are highly vulnerable to aquatic pollutants. In this study, Oreochromis niloticus were caught at three different sites in Al-Hassa irrigation channels, namely Al-Jawhariya, Um-Sabah and Al-Khadoud. The histological changes in gills and liver were detected microscopically and evaluated with semi-quantitative analyses. Also, heavy metals have been determined in the water samples in these sites. Results showed that all sites were polluted by different kinds of heavy metals. Cd and Pb were mostly detected at concentrations above the WHO reference values. Meanwhile, various histopathological abnormalities were observed in gills and liver of fish specimens. In the gill filaments, cell proliferation, lamellar cell hyperplasia, lamellar fusion, lifting of the respiratory epithelium, and the presence of aneurysmal areas were observed. In the liver, there was vacuolization of the hepatocytes, sinusoidal congestion, necrosis of the parenchyma tissue, nuclear pyknosis, eosinophilic hepatocellular degeneration, pigment accumulation, an increase in the number and size of melanomacrophage centers. Liver tumors with severe chronic inflammation were occasionally found in fish at Al-Khadoud area (first-time report). The histological lesions were comparatively most severe in the liver. Despite heavy metals assessment did not show marked differences among sites, histopathological biomarkers indicated that the surveyed fish are living under stressful environmental conditions. So, we suggest use those biomarkers in future monitoring of aquatic systems.  相似文献

Genotoxicity of freshwater pollution was assessed by measuring DNA damage in haemocytes of caged freshwater crayfish Astacus leptodactylus by the means of Comet assay and micronucleus test, integrated with the measurements of physiological (total protein concentration) and immunological (total haemocyte count) haemolymph parameters as biomarkers of undergone stress. Crayfish were collected at the reference site (River Mre?nica) and exposed in cages for 1 week at three polluted sites along the Sava River (Zagreb, Sisak, Krapje). The long term pollution status of these locations was confirmed by chemical analyses of sediments. Statistically significant increase in DNA damage measured by the Comet assay was observed at all three polluted sites comparing to the crayfish from reference site. In addition, native crayfish from the mildly polluted site (Krapje) cage-exposed on another polluted site (Zagreb) showed lower DNA damage than crayfish from the reference site exposed at the same location indicating adaptation and acclimatisation of crayfish to lower levels of pollution. Micronuclei induction showed similar gradient of DNA damage as Comet assay, but did not reach the statistical significance. Observed increase in total haemocyte count and total protein content in crayfish from polluted environments in the Sava River also confirmed stress caused by exposure to pollution. The results of this study have proved the applicability of caging exposure of freshwater crayfish A. leptodactylus in environmental genotoxicity monitoring using Comet assay and micronucleus test.  相似文献

Environmental monitoring by means of biomarker parameters assessed in different species is a useful tool. It has the advantage of providing a quantitative response as well as valuable information of ecological relevance on the chronic adverse effects caused by water pollution. The aim of this study was to assess the response of biochemical and physiological parameters of Cnesterodon decemmaculatus, a native teleost, simultaneously caught in two sites of Reconquista river, a highly polluted peri-urban river. This study compared the measured parameters with that of specimens of the same species captured in an unpolluted body water, and correlated the detected changes with the physicochemical profile of the water at each site. A comparison was made of selected parameters of gill, brain and liver and of somatic indices of fish collected from polluted and reference sites. The main parameters whose changes allowed to discriminate between sampling sites were gill (Na(+),K(+))-ATPase, brain acetylcholinesterase (AChE) and liver aminotransaminases activities; tissues' protein content and liver somatic index (LSI) were also sensitive biomarkers in brain and liver, respectively. The results showed that the response of the measured biomarkers allowed for the differentiation of sampling sites according to their water quality and confirmed that Cnesterodon decemmaculatus may be a useful test organism for the biomonitoring of freshwater environments. In addition, the simultaneous measurement of the physicochemical parameters of the water samples showed a good correspondence between the biomarkers responses and the environmental chemical stress conditions.  相似文献

The responses of Dicentrarchus labrax and Liza aurata to aquatic pollution were assessed in a contaminated coastal lagoon, using both traditional and novel biomarkers combined. DNA damage, assessed by comet assay, was higher in both fish species from the contaminated sites, whereas levels of cytochrome P450 1A1 gene expression were not significantly altered. The liver histopathological analysis also revealed significant lesions in fish from contaminated sites. Alterations in ras and xpf genes were analysed and additional pollutant-responsive genes were identified. While no alterations were found in ras gene, a downregulation of xpf gene was observed in D. labrax from a contaminated site. Suppression subtractive hybridization applied to D. labrax collected at a contaminated site, revealed altered expression in genes involved in energy metabolism, immune system activity and antioxidant response. The approach and results reported herein demonstrate the utility of anchoring traditional biomarker responses alongside novel biomarker responses.  相似文献

Abamectin is used as an acaricide and insecticide for fruits, vegetables and ornamental plants, as well as a parasiticide for animals. One of the major problems of applying pesticides to crops is the likelihood of contaminating aquatic ecosystems by drift or runoff. Therefore, toxicity tests in the laboratory are important tools to predict the effects of chemical substances in aquatic ecosystems. The aim of this study was to assess the potential hazards of abamectin to the freshwater biota and consequently the possible losses of ecological services in contaminated water bodies. For this purpose, we identified the toxicity of abamectin on daphnids, insects and fish. Abamectin was highly toxic, with an EC50 48 h for Daphnia similis of 5.1 ng L−1, LC50 96 h for Chironomus xanthus of 2.67 μg L−1 and LC50 48 h for Danio rerio of 33 μg L−1.  相似文献

Although the use of tributyltin in antifouling paints has been banned, this compound is still a serious pollutant of the marine environment. This paper describes a unique study in which European flounder (Platichthys flesus) were chronically (8 months) exposed to bis(tri-n-butyltin)oxide (TBTO) in the water under controlled laboratory conditions. Residue levels in selected tissues (liver, muscle) and general health status indices were measured and the effects on several organs (gills, liver, mesonephros, ovary/testis, spleen, and gastrointestinal tract) were examined histopathologically. Additionally, morphometric analysis of the thymus was performed. The major finding is that exposure of flounder to 5 μg TBTO/l over a period of 8 months, resulting in body burdens comparable to high field levels, induced significant reduction of thymus volume, possibly affecting immunocompetence of the animals. Chronic exposure of European flounder to tributyltin is therefore likely to affect the general health status of this species in heavily polluted aquatic environments.  相似文献

Changes in biomarkers of fish captured from stressed environments may represent a reliable tool in revealing sublethal effects of the pollutants found in aquatic ecosystems. The response patterns of selected biochemical and morphological variables biomarkers of effect were assessed in adult females of an indigenous teleost Cnesterodon decemmaculatus, caught at a polluted site (San Francisco) of the Reconquista river (Buenos Aires, Argentina). Combined field-caging experiments with controlled laboratory exposure to clean media were performed. The biochemical parameters measured were specific activities of gill (Na+ + K+)-ATPase, liver AlaAT and AspAT, and brain AChE; LSI and CF were also calculated. The changes in gill (Na+ + K+)-ATPase and liver AlaAT activities of fish captured in the field in most cases were reversible after transfer to clean media. The results were interpreted in association with the physicochemical profile of the water samples taken simultaneously with the capture of the fish. Results suggested the suitability of the test species used as tools in environmental monitoring programs of risk assessment.  相似文献

This experiment was conducted to study the genotoxic potentials of sodium arsenite (NaAsO2) in freshwater fish Oreochromis mossambicus by using alkaline comet assay and micronucleus (MN) test. Fish were exposed to three different concentrations (3 ppm, 28 ppm and 56 ppm) of arsenic and gill, liver and blood tissue samples were collected after 48 h, 96 h and 192 h of exposure. Arsenic exposure induced DNA damage in all tissues examined in a concentration dependent manner. A significant (< 0.05) increase in the comet tail DNA (%) of the exposed fish liver, gill, and blood was observed after 48 h and 96 h of exposure, but a decline in DNA damage was recorded in all the tissues at all the three concentrations studied after 192 h of exposure. Liver tissue exhibited significantly (< 0.05) higher DNA damage at all the concentrations examined, followed by gill and blood. Higher liver tail DNA (51.38 ± 0.21%) refers that it is more prone to injury to arsenic toxicity than the gill and blood. In blood samples arsenic induced micronucleus formation in a concentration dependent manner and highest (5.8 ± 0.46%) value was recorded in 56 ppm after 96 h of exposure, whereas, it was decreased after 192 h of exposure at all the three concentrations of NaAsO2 examined which refers to the DNA repairing ability of fish to arsenic toxicity. The results of this study depict the genotoxic potentials of arsenic to fish which in turns provide insight on advanced study in aquatic toxicology.  相似文献

Aquatic macrophytes play an important role in the structural and functional aspects of aquatic ecosystems by altering water movement regimes, providing shelter to fish and aquatic invertebrates, serving as a food source, and altering water quality by regulating oxygen balance, nutrient cycles, and accumulating heavy metals. The ability to hyperaccumulate heavy metals makes them interesting research candidates, especially for the treatment of industrial effluents and sewage waste water. The use of aquatic macrophytes, such as Azolla with hyper accumulating ability is known to be an environmentally friendly option to restore polluted aquatic resources. The present review highlights the phytoaccumulation potential of macrophytes with emphasis on utilization of Azolla as a promising candidate for phytoremediation. The impact of uptake of heavy metals on morphology and metabolic processes of Azolla has also been discussed for a better understanding and utilization of this symbiotic association in the field of phytoremediation.  相似文献

Hepatic MFO components (cytochrome P-450, cytochrome b(5), and ethoxyresorufin O-deethylase, EROD) were measured in brown bullheads (Ictalurus nebulosus) inhabiting a creek receiving a complex mixture of organics and trace metals. The activities of these same enzymes were also measured in bullheads from an uncontaminated reference site to assess the relative ability of MFO parameters to serve as a biomarker of aquatic pollution. Bullheads analyzed from the polluted site had lower hepatic microsomal P-450 (p < 0.01) concentrations and similar EROD activities per mg protein as compared to bullheads from the reference site. However, analysis of enzyme turnover ratios revealed greater EROD activity per mg cytochrome P-450 (p < 0.05) in the fish from the polluted site. No difference in cytochrome b(5) activities were observed between the two groups. As compared to the reference site, bullheads collected from the polluted creek had an increased occurrence of lip and lower jaw lesions and liver damage, including elevated liver/body weight ratios. Accordingly, the monooxygenase activities measured in this study were not reliable indicators of chemical pollution or contaminant stress in bullheads in the polluted creek. Further research is needed concerning contaminant interactions, particularly among organic pollutants and metals and their effects on monooxygenase activities.  相似文献

Liver lesions in wild fish have been associated with xenobiotic exposure. Facing reports of pollution in the Douro River estuary (north of Portugal), we have been making field surveys using fishes and targeting histopathological biomarkers of exposure and effect. Herein, we intended to better characterize and report the rate of one poorly understood lesion—hepatocellular fibrillar inclusions (HFI)—found in European flounder (Platichthys flesus). With this report, we aimed to establish sound baseline data that could be viewed as a starting point for future biomonitoring, while offering the world's second only pool of field data on such a liver toxicopathic lesion, which could be compared with data available from the UK estuaries. Sampling was done in the Douro River estuary over 1 year. A total of 72 animals were fished with nets, in spring–summer (SS) and autumn–winter (AW) campaigns. Livers were processed for histopathology and both routine and special staining procedures (alcian blue, periodic acid Schiff (PAS), tetrazonium coupling reaction). Immunohistochemistry targeted AE1/AE3 (pan cytokeratins). The severity of the HFI extent was graded using a system with four levels, varying from 0 (absence of HFI) to 3 (high relative density of cells with HFI). Cells (isolated/groups) with HFI appeared in 35 % or more of the fish, in the total samples of each season, and over 40 % in more homogeneous sub-samples. There were no significant differences when comparing samples versus sub-samples or SS versus AW. When merging the data sets from the two seasons, the frequency of fish with HFI was ≈36 % for the total sample and ≈49 % for the sub-sample. The extreme group (biggest and smallest fish) revealed a HFI frequency of only 16 %, which differed significantly from the total and sub-sampled groups. Immunostaining and PAS were negative for the HFI, and alcian blue could, at times, faintly stain the inclusions. These were positive with the tetrazonium reaction. We showed the presence of HFI in European flounder from the Douro River estuary, proving that they are essentially protein in nature, that no seasonal changes existed in the HFI frequency, and that it was rarer in the smallest and biggest fish groups. Within the ranges of weight/size of our total sample, we estimate that the frequency of HFI in the local flounder is ≈35 %. That rate stands as a baseline value for future assessments, namely for biomonitoring purposes targeting correlations with the estuary pollution status.  相似文献

The study reports the accumulation, distribution and metabolism of technical endosulfan in Jenynsia multidentata. Adult females were exposed to acute sublethal concentrations (0.072, 0.288 and 1.4 μg L−1). After 24 h, fish were sacrificed and gills, liver, brain, intestine and muscle were removed. Results show that both isomers of technical-grade endosulfan (α- and β-) are accumulated in fish tissues and biotransformation to endosulfan sulfate occurs at all concentrations tested. Significantly differences in endosulfan accumulation were only found at 1.4 μg L−1 but not between the lowest concentrations. However a similar distribution pattern was observed at all exposure levels where liver, intestine and brain had the highest levels of α-, β-endosulfan and endosulfan sulfate. Moreover, liver and brain showed the highest endosulfan sulfate:α-endosulfan ratios due to high biotransfomation capacity. J. multidentata demonstrated to be a sensitive species under exposure to technical endosulfan and, therefore, could be used to assess aquatic pollution.  相似文献

Adequate temporal trend analysis of mercury (Hg) in freshwater ecosystems is critical to evaluate if actions from the human society have affected Hg concentrations ([Hg]) in fresh water biota. This study examined temporal change in [Hg] in Northern pike (Esox lucius L.) in Swedish freshwater lakes between 1994 and 2006. To achieve this were lake-specific, multiple-linear-regression models used to estimate pike [Hg], including indicator variables representing time and fish weight and their interactions. This approach permitted estimation of the direction and magnitude of temporal changes in 25 lakes selected from the Swedish national database on Hg in freshwater biota. A significant increase was found in 36% of the studied lakes with an average increase in pike [Hg] of 3.7 ± 6.7% per year that was found to be positively correlated with total organic carbon. For lakes with a significant temporal change the dataset was based on a mean of 30 fish, while for lakes with no temporal change it was based on a mean of 13 fish.  相似文献

Purpose

The aim of this work was to assess the levels of copper and zinc in fish from the main freshwater ecosystems of Moldova, in relation with species, habitat, age, sex, season, and development stage.

Methods

Fish from Cyprinidae and Percidae families (Cyprinus carpio, Carassius auratus gibelio, Rutilus rutilus heckeli, Abramis brama, Aristichthys nobilis, Hypophtalmichthys molitrix, Sander lucioperca) were collected from Prut and Dniester rivers, Cuciurgan, Dubasari, and Costesti-Stanca reservoirs, and ponds of farms in the Dniester delta. The Cu and Zn content of fish tissues (skeletal muscles, liver, gonads, gills, skin, and scales) was determined by flame atomic absorption spectrophotometer AAS-3, of water by graphite furnace HGA 900 of AAnalist 400.

Results

The level of heavy metals accumulation in muscles of immature fish follows their dynamics in water. The highest concentration of zinc was registered in the gonads of mature fish, and of copper??in the liver. The lowest Cu and Zn contents were recorded in the muscles and are in the United Nations Food and Agriculture Organization safety-permissible levels for human consumption. Cu and Zn contents in muscles of fish depend on specimen age. Their level in fish gonads was sharply increasing during pre-spawning period. During the early developmental stages, the metal concentration in fish eggs and larvae varies within wide limits, but the accumulation pattern is similar in the investigated species.

Conclusions

The fish represent one of the most indicative factors for the estimation of trace metals pollution in freshwater systems and this is important not only for monitoring purposes, but also for the fish culture ones.  相似文献

Since the 1990s, the Lake Maggiore (Northern Italy) has been recognized as an aquatic environment contaminated by DDTs and other persistent organic pollutants, but to date just few studies were carried out to investigate the effects of pollution to aquatic organisms. The aim of this study was the application of a stepwise approach based on chemical data, a suite of biomarkers and the integration of their responses into a biomarker response index (BRI) to evaluate the site-specific quality assessment in different sampling stations of Lake Maggiore, one of the largest European lakes. We used as biological model the freshwater bivalve Zebra mussel (Dreissena polymorpha). Several hundred bivalve specimens were sampled on May 2011 from eight sampling sites located along the lake shoreline. We measured levels of DDTs, PCBs, HCHs, HCB, and PAHs accumulated in D. polymorpha soft tissues by GC/MSn, while the activities of catalase, superoxide dismutase, glutathione peroxidase, and glutathione S-transferase, as well as the lipid peroxidation and protein carbonyl content were evaluated in homogenates from native bivalves as oxidative stress indices. Moreover, DNA damage was investigated by the alkaline precipitation assay. Significant imbalances of enzymatic activity were found in mussels from most of the sampling sites, as well as notable increases of damage to macromolecules. Health status of mussels from Baveno was greatly affected by lake pollution, probably due to high levels of DDTs measured in this site, while a wide variability in biomarker responses was found in all the other stations. The application of a BRI allowed distinguishing impacts of pollution to bivalves, confirming mussels from Baveno as the most threatened and revealing that also the health status of bivalves from Suna, Brissago, Pallanza, and Laveno is affected. These evidences suggest the usefulness of a specific index to integrate all the biomarker endpoints in order to provide a correct environmental risk assessment.  相似文献

BACKGROUND: Triggered by the requirement of Water Framework Directive for a good ecological status for European river systems till 2015 and by still existing lacks in tools for cause identification of insufficient ecological status MODELKEY (http:// www.modelkey.org), an Integrated Project with 26 partners from 14 European countries, was started in 2005. MODELKEY is the acronym for 'Models for assessing and forecasting the impact of environmental key pollutants on freshwater and marine ecosystems and biodiversity'. The project is funded by the European Commission within the Sixth Framework Programme. OBJECTIVES: MODELKEY comprises a multidisciplinary approach aiming at developing interlinked tools for an enhanced understanding of cause-effect-relationships between insufficient ecological status and environmental pollution as causative factor and for the assessment and forecasting of the risks of key pollutants on fresh water and marine ecosystems at a river basin and adjacent marine environment scale. New modelling tools for risk assessment including generic exposure assessment models, mechanistic models of toxic effects in simplified food chains, integrated diagnostic effect models based on community patterns, predictive component effect models applying artificial neural networks and GIS-based analysis of integrated risk indexes will be developed and linked to a user-friendly decision support system for the prioritisation of risks, contamination sources and contaminated sites. APPROACH: Modelling will be closely interlinked with extensive laboratory and field investigations. Early warning strategies on the basis of sub-lethal effects in vitro and in vivo are provided and combined with fractionation and analytical tools for effect-directed analysis of key toxicants. Integrated assessment of exposure and effects on biofilms, invertebrate and fish communities linking chemical analysis in water, sediment and biota with in vitro, in vivo and community level effect analysis is designed to provide data and conceptual understanding for risk arising from key toxicants in aquatic ecosystems and will be used for verification of various modelling approaches. CONCLUSION AND PERSPECTIVE: The developed tools will be verified in case studies representing European key areas including Mediterranean, Western and Central European river basins. An end-user-directed decision support system will be provided for cost-effective tool selection and appropriate risk and site prioritisation.  相似文献

Matching the trend seen among the major large rivers of the globe, the Amazon River and its tributaries are facing aquatic ecosystem disruption that is affecting freshwater habitats and their associated biodiversity, including trends for decline in fishery resources. The Amazon’s aquatic ecosystems, linked natural resources, and human communities that depend on them are increasingly at risk from a number of identified threats, including expansion of agriculture; cattle pastures; infrastructure such as hydroelectric dams, logging, mining; and overfishing. The forest, which regulates the hydrological pulse, guaranteeing the distribution of rainfall and stabilizing seasonal flooding, has been affected by deforestation. Flooding dynamics of the Amazon Rivers are a major factor in regulating the intensity and timing of aquatic organisms. This study’s objective was to identify threats to the integrity of freshwater ecosystems, and to seek instruments for conservation and sustainable use, taking principally fish diversity and fisheries as factors for analysis.  相似文献

We evaluated whether growth rates of six fish species correlated with PCB concentrations in a moderately-to-heavily polluted freshwater ecosystem. Using a large dataset (n = 984 individuals), and after accounting for growth effects related to fish age, habitat, sex, and lipids, growth correlated significantly, but positively with lipid-corrected PCB concentrations for 4 of 6 species. Remaining species showed no correlations between growth and PCBs. Comparisons with regional, lentic growth averages for four species confirmed growth was on par and in three of four cases higher than regional averages in the PCB-polluted ecosystem. We conclude that for these species, at the range of concentrations examined, these PCBs do not exert negative impacts on growth. Rather, factors often cited as influential to growth were also driving growth trends in this study. Future studies that evaluate whether pollution affects growth must account for major growth drivers prior to attributing growth differentials to pollution alone.  相似文献

A new simple and sensitive method to distinguish chemically polluted from unpolluted situations in freshwater ecosystems is reported. For this purpose, Chironomus gr thumni larvae were collected from a polluted urban river downstream a sewage treatment plant. For the first time, ELISA assay was used to semi-quantify the multixenobiotic resistance transporters (MXR) in these small pertinent bioindicators. The use of samples immediately fixed in the field gives a delay to isolate larvae and allows multi-sampling along a longitudinal transect in a river at a given time. Results exhibit an induction of MXR proteins in larvae from the polluted river and a deinduction in larvae maintained 11days in unpolluted water. They show new evidences to use midge larvae in biomonitoring environmental programs. They answer to first biomarker calibration steps for the ongoing development of MXR transporters as a detection tool of xenobiotic impacts on bioindicator invertebrates in their freshwater habitats.  相似文献
