The present study was conducted to evaluate the levels of residual DDT in Lebanese soils. A total of 113 surface soil samples were collected for analysis from three major agricultural regions in the country: Mount Lebanon, Beqa'a valley and the coastal plain. The values of residual DDT in soils ranged between 0 and 1190 ng g(-1). The majority of the analyzed soil samples were free or contained very low concentrations of DDE. Only two surface soil samples, which were collected from urban areas with high human activities, contained relatively high concentrations of residual DDT. The levels of residual DDT in all of the analyzed soil samples were within the permissible limits for agricultural soils.  相似文献

A 96-microwell enzyme-linked immunosorbent assay (ELISA) method was evaluated to determine PCDDs/PCDFs in sediment and soil samples from an EPA Superfund site. Samples were prepared and analyzed by both the ELISA and a gas chromatography/high resolution mass spectrometry (GC/HRMS) method. Comparable method precision, accuracy, and detection level (8 ng kg(-1)) were achieved by the ELISA method with respect to GC/HRMS. However, the extraction and cleanup method developed for the ELISA requires refinement for the soil type that yielded a waxy residue after sample processing. Four types of statistical analyses (Pearson correlation coefficient, paired t-test, nonparametric tests, and McNemar's test of association) were performed to determine whether the two methods produced statistically different results. The log-transformed ELISA-derived 2,3,7,8-tetrachlorodibenzo-p-dioxin values and log-transformed GC/HRMS-derived TEQ values were significantly correlated (r=0.79) at the 0.05 level. The median difference in values between ELISA and GC/HRMS was not significant at the 0.05 level. Low false negative and false positive rates (<10%) were observed for the ELISA when compared to the GC/HRMS at 1,000 ng TEQ kg(-1). The findings suggest that immunochemical technology could be a complementary monitoring tool for determining concentrations at the 1,000 ng TEQ kg(-1) action level for contaminated sediment and soil. The ELISA could also be used in an analytical triage approach to screen and rank samples prior to instrumental analysis.  相似文献

It was short of research on the organochlorine pesticides (OCPs) residues in the soils of Hong Kong. Sixty-six representative soil samples were collected from the 46 sites covering five types of land uses in Hong Kong. Hexachlorohexanes (HCH) and 7 Stockholm Convention OCPs were analyzed by gas chromatograph (GC) equipped with a Nickel 63 electronic capture detector (muECD). The results presented that HCH and 5 Stockholm Convention pesticides were detected in Hong Kong soils although the detectable ratio varies to a great extent. The concentration sequence of the five detectable OCPs was HCH > dichlorodiphenyltrichloroethane (DDT) > hexachlorobenzene (HCB) approximately = Endrin > alpha-endosulfan. Among the OCPs and their homologues or isomers, beta-HCH and p,p'-DDE were the two predominant substances according to the concentrations and detectable ratios, concentrations of which in soils were averagely 6.12 microg kg(-1) and 0.41 microg kg(-1) respectively. Soil horizon samples of 0-10 cm, 10-30 cm and >30 cm depth were selected from nine soil profiles to demonstrate the depth distributions of DDT and HCH in soil profiles. Concentrations of HCH tended to increase gradually from the topsoil to bottom layer while the lowest concentration of DDT is usually found in the subsoil (10-30 cm) in most sampling sites. In addition, close correlations of pH(KCl) and total organic carbon (TOC) with HCH indicated an effect on the residues of HCH caused by these two soils properties, but such relationships were not found with DDT or other OCPs.  相似文献

An indirect competitive enzyme-linked immunosorbent assay (ELISA) has been developed and optimized for atrazine determination in soil at different depths (0-10, 10-20, and 20-30 cm) before and after 48 h of application, corn shoot and cow milk samples collected from Dina farm, Egypt. This assay was based on a specific polyclonal antibodies (PAb) raised by immunizing New Zealand rabbits with an immunogen prepared by coupling 3-{4-(ethylamino)-6-(isopropylamino)-1,3,5-triazine-2-yl} thiopropanoic acid to bovine serum albumin (BSA) via N-hydroxysuccinimide (NHS) active ester method. The sensitivity (estimated as IC?? value) was 17.5 μg mL?1 with a detection limit of 0.1 ng mL?1. The maximum atrazine concentration was found in soil especially in the deepest layer (325 and 890 μg kg?1 before and after application, respectively). Atrazine concentration in corn shoot was 333.28, μg kg?1 dry plant, while there was no detectable amount in milk. All samples screened by ELISA were validated by gas chromatography mass spectrometer procedure (GC/MS). Good correlation was achieved between the two methods (r = 0.997 for soil and 0.9814 for plant). This study demonstrates the utility and convenience of the simple, practical and cost-effective ELISA method in the laboratory for analysis of environmental samples. The method is ideal for the rapid screening of large numbers of samples in laboratories where access to GC/MS facilities, is limited or lacking.  相似文献

Detailed analyses of persistent organic pollutants (POPs) such as organochlorine pesticides (OCPs), hexachlorocyclohexane (HCH) isomers (HCHs), dichlorodiphenyltrichloro ethane (DDT) and its metabolites (DDTs) and congeners of polychlorinated biphenyls (PCBs) in soil and surface water from the northeastern São Paulo, Brazil allowed the evaluation of the contamination status, distribution and possible pollution sources. The pesticides and PCBs demonstrated markedly different distributions, reflecting different agricultural, domestic and industrial usage in each region studied. The ranges of HCH, DDT, and PCBs concentrations in the soil samples were 0.05–0.92, 0.12–11.01, 0.02–0.25 ng g−1 dry wt, respectively, and in the surface water samples were 0.02–0.6, 0.02–0.58 and 0.02–0.5 ng l−1, respectively. Overall elevated levels of DDT and PCB were recorded in region 2, a site very close to melting, automotive batteries industries, and agricultural practice regions. High ratios of metabolites of DDT to DDT isomers revealed the recent use of DDT in this environment. The sources of contamination are closely related to human activities, such as domestic and industrial discharge, street runoff, agricultural pesticides and soil erosion, due to deforestation as well as atmospheric transport.  相似文献

Eggplant samples were analyzed for endosulfan residues using ELISA, a technique recognized as a promising tool for screening environmental contaminants. Calibration curve for endosulfan was standardized using kits developed by CFTRI Mysore. Farm gate samples of eggplant from 12 different locations were analyzed. The matrix effect was removed by charcoal clean-up. The results were compared with gas chromatographic (GC) analysis. The residues in different samples were in the range of 5-226 ppb. Correlation coefficient between the two methods was found to be 0.98.  相似文献

Topsoil samples from 56 sites around the Guanting Reservoir, China, were measured for HCH and DDT concentrations. The total soil HCH content (including alpha-, beta-, gamma-, and delta-isomers) in these soil samples ranged from 0 to 7.33 ng x g(-1), with a mean of 0.69 ng x g(-1). These levels were considerably lower than those of the total DDT soil contents (including pp'-DDE, pp'-DDD, op'-DDT, and pp'-DDT), which ranged from 0 to 76.01 ng x g(-1), with a mean of 9.46 ng x g(-1). DDT was also found to be the major pollutant in the soil samples, accounting for approximately 93% of the total organochlorine pesticide (OCP) contents. Several environmental factors including land use, soil texture, soil taxonomy, and microbial biomass were considered to be responsible for the OCP levels observed. The data provide some insight into the effects of environmental conditions such as soil formation, agricultural cultivation, nutrient enrichment, and other anthropogenic activities on the degradation of OCPs in soils. Although the OCP residues currently are below the maximum limits set for use on agricultural land in China, and only rarely would such levels pose significant ecological concern, OCPs are highly persistent in soil and bioaccumulative. The data provided in this study are considered crucial for reservoir remediation, especially since the Guanting Reservoir will serve as one of the main drinking water sources for Beijing in the foreseeable future.  相似文献

A high-throughput screening method using selective pressurized liquid extraction (SPLE) and enzyme-linked immunosorbent assay (ELISA) for monitoring dioxins in sediment and soil is described. SPLE conditions were developed by extracting sediment or soil together with alumina, 10% AgNO3 in silica, and sulfuric acid impregnated silica (acid silica) using dichloromethane (DCM) as the solvent at 100 °C and 2000 psi. Post-extraction cleanups were not required for ELISA. Two reference sediments (National Institute of Standards and Technology SRM 1944 and Wellington Laboratories WMS01) were analyzed by the SPLE–ELISA method. The ELISA utilized a polyclonal antibody and 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) as the calibrant. Recoveries of ELISA-derived TCDD equivalents (EQ) relative to the expected gas chromatography/high resolution mass spectrometry (GC/HRMS) derived dioxin toxic equivalent (TEQ) values were 116 ± 11% for SRM 1944 and 102 ± 13% for WMS01. ELISA TCDD EQs were consistent with the dioxin TEQs as measured by GC/HRMS for 25 soil/sediment samples from seven different contaminated sites. The ELISA had an approximate method detection limit of 10 pg g−1 with a precision of 2.6–29% based on the relative percentage difference (%RPD) for duplicate samples. Estimated sample throughput for the SPLE–ELISA was three times or more than that of the GC/HRMS method employing PLE with a multi-column cleanup.  相似文献

Background, aim, and scope  Dicofol is widely used as a pesticide in agriculture applications. Since dicofol is mainly synthesized from dichlorodiphenyltrichlorethane (DDT), it contains DDT as an impurity. The European Community has forced Prohibition Directive 79/117/EEC to reduce DDT in dicofol formulations. Specifically, DDT content in a dicofol formulation cannot exceed 0.1%. The goal of this project was to determine the DDT content in dicofol formulations used in Turkey. Materials and methods  Samples of all the dicofol formulations in Turkey were collected to quantify DDT and DDT-related compounds. Four replicates were used for each sample. GC/MS/MS was used to analyze p,p′ and o,p′ isomers of DDT, DDD, and DDE. A HPLC was used to determine p,p′-Cl-DDT concentrations. Results  The total DDT content of the formulated dicofol was found between 0.3% and 14.3%. The concentration of p,p′-DDE ranged from 167 to 1,042 mg kg−1 in dicofol samples. p,p′-DDT concentrations were found to be 32 to 183 mg kg−1. The o,p’-DDT level ranged from 2 to 34 mg kg−1 in the dicofol formulations analyzed. Discussion  It was estimated that 617.8 kg of DDT was released from dicofol. The main impurity was identified as p,p-Cl-DDT. Based on these results, dicofol serves as a continuing source of DDT contamination. Conclusions  All DDT concentrations in dicofol samples analyzed were higher than the permitted 0.1% level of Prohibition Directive 79/117/EEC. The reduction of dicofol is critical since it serves as a continual source of DDT contamination. Recommendations and perspectives  DDT has been found in soil, water, and air samples. Dicofol has been identified as a contributor to continued DDT contamination in soil and water. More studies are needed to ascertain the source of DDT in the air.  相似文献

A low-cost, high throughput bioanalytical screening method was developed for monitoring cis/trans-permethrin in dust and soil samples. The method consisted of a simple sample preparation procedure [sonication with dichloromethane followed by a solvent exchange into methanol:water (1:1)] with bioanalytical detection using a magnetic particle enzyme-linked immunosorbent assay (ELISA). Quantitative recoveries (83–126 %) of cis/trans-permethrin were obtained for spiked soil and dust samples. The percent difference of duplicate ELISA analyses was within ± 20 % for standards and ± 35 % for samples. Similar sample preparation procedures were used for the conventional gas chromatography/mass spectrometry (GC/MS) analysis except that additional cleanup steps were required. Recoveries of cis/trans-permethrin ranged from 81 to 108 % for spiked soil and dust samples by GC/MS. The ELISA-derived permethrin concentrations were highly correlated with the GC/MS-derived sum of cis/trans-permethrin concentrations with a correlation coefficient (r) of 0.986. The ELISA method provided a rapid qualitative screen for cis/trans-permethrin in soil and dust while providing a higher sample throughput with a lower cost as compared to the GC/MS method. The ELISA can be applied as a complementary, low-cost screening tool to prioritize and rank samples prior to instrumental analysis for exposure studies.  相似文献

The use of organo-chlorine (DDT and HCH) has been banned in China for 20 years. A field survey was carried out during 1999-2000 in the Taihu Lake Region. Organo-chlorine pesticide (OCP) residues in soil, water, fish and sediment samples were investigated. DDT was detected in 5 out of 10 samples with concentration ranging from 0.3 to 5.3 microg/kg in the surface (0-15 cm) layer, 6 out of 10 with 0.5 to 4.0 microg/kg in the subsoil layer (16-30 cm), and 4 of 10 with 0 to 2.7 microg/ kg in the deep soil layer (31-50 cm). Results for HCH residues in soil samples were similar to those of DDT. These results indicate that OCP residues in 0-50 cm profile had been leached out or degraded to safe level. In river water DDT was detected in 10 out of 13 samples ranging from 0.2 to 9.3 microg/l, with an average of 1.0 microg/l. While HCH was detected in 12 out of 13 samples ranging from 0.02 to 36.1 microg/l, with an average 5.6 microg/l. DDT residues in sediment ranged from 0.1 to 8.8 microg/kg, while HCH ranged from 0.3 to 66.5 microg/kg. DDT residues in fish body ranged from 3.7 to 23.5 microg/kg and HCH ranged from 3.7 to 132 microg/kg. These results demonstrate an accumulation through food chain (from soil-water-sediment-microbes-crop-fish-... etc.), also that HCH residues are generally more persistent than DDT residues. High levels of DDT residues and hexachlorocyclohexanes (HCHs) were found in soil, well water, and surface water around a collapsed pesticide storage shed at Vikuge Farm, Tanzania. Residues of DDT and HCHs were found at three soil depths down to 50 cm. Surface soil samples contained up to 28% total DDT and 6% total HCH residues. Water samples had concentrations of up to 30 microg L(-1) of organochlorine pesticides. Other compounds detected were aldrin, azinphos-methyl, carbosulfan, gamma-chlordane, chlorprofam, heptachlor, hexazinone, metamitron, metazachlor, pendimethalin, and thiabendazole. Although the visible remains of pesticides have been removed, the remaining soil is itself hazardous waste and poses a risk to the environment and the inhabitants of the surrounding villages. These findings show the necessity to follow up the environmental situation at former storage sites of obsolete stocks of pesticides, and that the environmental problems are not necessarily solved by removing the visible remains.  相似文献

The analysis of PCBs in fish tissues by immunoassay methods was evaluated using fish collected from a US monitoring program, the National Contaminant Biomonitoring Program of the US Department of Interior, Fish and Wildlife Service. Selected composite whole fish samples, which represented widely varying concentrations and sources of PCBs, were extracted and subjected to congener PCB analysis by gas chromatography (GC) and total PCB analysis using an ELISA (ePCBs) calibrated against technical Aroclor 1248. PCB congener patterns in these fishes were different from the patterns found in commercial Aroclors or their combinations as demonstrated by principal component analysis of normalized GC congener data. The sum of the PCB congeners measured by GC (total-PCBs) ranged from 37 to 4600 ng/g (wet weight). Concentrations of PCBs as determined by the ELISA method were positively correlated with total-PCBs and the ePCBs/total-PCBs ratios for individual samples ranged from 1 to 6. Ratios of ePCBs/total-PCBs for dilutions of Aroclors 1242, 1254, and 1260 and for matrix spikes range from 0.6 for 1242 to 2.5 for 1254 and 1260. These results suggest that higher chlorinated PCB congeners have higher affinity for the anti-PCB antibodies. Partial least squares with latent variable analysis of GC and ELISA data of selected Aroclors and fish samples also support the conclusion that ELISA derived PCB concentrations are dependent on the degree of chlorination.  相似文献

Samples of moss (Scleropodium purum or, in its absence, Hypnum cupressiforme) and of topsoil (0-5 cm) were collected in the spring of 1995, from 75 sites distributed throughout Galicia (northwest Spain) in order to compare the potential usefulness of each in evaluating the atmospheric deposition of contaminants. Analysis was made of the total content of various metals in the moss and of different soil metal fractions (including total metal content, labile metals and metals bound to organic material). We found that the labile fraction most closely resembled the metal content of the moss. Calculation of enrichment factors allowed us to show that the soil was not, except in the case of Cr, the main source of contamination in the moss. By comparing contamination factors of metals in moss and soil fraction samples we found that moss samples were more useful for evaluating the degree of contamination at different sites.  相似文献

Elevated levels of heavy metals in soils are a result of industrial activities, atmospheric deposition, and the land application of sewage sludges and industrial by-products. Their persistence in the soil environment has created interest in the possible changes in solubility. In this study, total dissolved concentrations of Pb, Zn, and Cu were monitored in seven metal-amended soils (a calcareous and six acid mineral soils). Single metal solutions were added to soils and equilibrated (aged) for 40 days. During the 40 days the soil was allowed to air-dry and was rewetted in cycles of about 5 days. At the end of this reaction period, metal solubility was measured (by atomic absorption spectrometry and direct current plasma spectrometry) at the initial soil pH and at decreased pH values which were induced by addition of small aliquots of acid. As expected, solubility of added Pb, Zn, and Cu increased with a decrease in pH. Furthermore, the results showed that the solubility relationship with pH was similar in all non-calcareous soils. This suggests that metal solubility may be controlled by similar soil components, presumably involving soil characteristics such as pH, organic matter content, and soil mineralogy. For each metal, an approximate pH value was found at which solubility deviated from the solubility of metals when they occur in soils at typical (natural) values. This pH was about (pH+/-0.2): 5.2 for Pb, 6.2 for Zn, and 5.5 for Cu. Thus, pH values below these thresholds may enhance metal mobility, biological availability and toxicity in soils. Metals dissolved at higher pH in the calcareous soil (18.8 g kg(-1) inorganic carbon, initial pH 8.2). In a calcareous soil, a significant fraction of these metals react with carbonates, and decreased pH results in much higher metal dissolution. Yet, metal solubility in soils is not determined by the formation and dissolution of single metal compounds.  相似文献

The aim of this study was to determine the dietary exposure of DDT in foodstuffs consumed in Hong Kong by secondary school students in Hong Kong in 2005. Food samples of domestic or imported food were collected randomly from local markets for the determination of DDT. The analytical method for this survey was based on isotope dilution technique. DDT isomers were determined at parts per billion (mugkg(-1)) levels in foods by pressured fluid extraction followed by gel permeation column cleanup and gas chromatography with mass spectrometry detection. Among the 98 composite samples, 25 were found positive with DDT levels greater than the limit of quantitation (LOQ) (1mugkg(-1) for each isomer of DDT). Only two of the meat products were found to contain low levels of the metabolites of DDT. A number of fish and shellfish were found to contain not just the metabolites of DDT, but also parent DDT. The main contribution to DDT was from seafood. The dietary intake of DDT for an average and high secondary school student consumers were estimated to be 0.145 and 0.291mugkg(-1)bwd(-1), respectively. Both levels fell well below the Provisional Tolerable Daily Intake (PTDI) of 10mugkg(-1)bwd(-1) established by the Joint Food Agriculture Organization/World Health Organization Meeting on Pesticide Residues (JMPR). It was concluded that both the average and high secondary school student consumers were unlikely to experience major toxicological effects of DDT.  相似文献

Leaves of Eucalyptus sp., Prunus domestica (plum), and Anacardium occidentale (cashew), as well as roots and leaves of Manihot esculenta (cassava) were used to study the local distribution of pesticides and metabolites from a point source, an old storage site at Vikuge farm in Tanzania. The GPC-cleaned extracts were analyzed by GC-ECD and GC/MS. Eleven organochlorine pesticide residues namely pentachloroanisole, p,p'-DDT, o,p'-DDT, p,p'-DDE, o,p'-DDE, p,p'-DDD, o,p'-DDD, alpha-HCH, beta-HCH, delta-HCH and epsilon-HCH were detected in the samples. The concentrations of total DDT were 818 ng/g fw in Eucalyptus sp., 16 ng/g fw in A. occidentale and 4 ng/g fw in P. domestica. In M. esculenta, total DDT ranged from 191 to 586 ng/g fw in roots and 7 to 425 ng/g fw in leaves. The concentrations of total HCH were up to 15 ng/g fw in Eucalyptus sp., while the concentrations of pentachloroanisole were up to 2 ng/g fw in leaves of M. esculenta. There are very strong positive correlations in the concentrations of the detected compounds, suggesting that they have a common source. The low DDE/DDT ratios (0.02-0.07) in all samples indicate input of non-degraded DDT from the source. The high alpha-HCH/gamma-HCH ratios in some samples (>3.1-10) indicate input of technical HCH. The concentrations of total DDT in cassava roots were either very close to, or greater than, the FAO/WHO limit, indicating risks and concern to public health.  相似文献
