The fractionation of carbon and chlorine stable isotopes of dichloromethane(CH_2Cl_2,DCM)upon dechlorination by cells of the aerobic methylotroph Methylobacterium extorquens DM4 and by purified DCM dehalogenases of the glutathione S-transferase family was analyzed.Isotope effects for individual steps of the multi-stage DCM degradation process,including transfer across the cell wall from the aqueous medium to the cell cytoplasm,dehalogenase binding,and catalytic reaction,were considered.The observed carbon and chlorine isotope fractionation accompanying DCM consumption by cell supensions and enzymes was mainly determined by the breaking of C\Cl bonds,and not by inflow of DCM into cells.Chlorine isotope effects of DCM dehalogenation were initially masked in high density cultures,presumably due to inverse isotope effects of non-specific DCM oxidation under conditions of oxygen excess.Glutathione cofactor supply remarkably affected the correlation of variations of DCM carbon and chlorine stable isotopes(Δδ~(13)C/Δδ~(37)Cl),increasing corresponding ratio from 7.2–8.6 to 9.6–10.5 under conditions of glutathione deficiency.This suggests that enzymatic reaction of DCM with glutathione thiolate may involve stepwise breaking and making of bonds with the carbon atom of DCM,unlike the uncatalyzed reaction,which is a one-stage process,as shown by quantum-chemical modeling.  相似文献

Evaluation system can encourage and guide entrepreneurs, and impel them to perform well in environment management. An evaluation method based on advantage structure is established. It is used to analyze entrepreneur environment management behavior in China. Entrepreneur environment management behavior evaluation index system is constructed based on empirical research. Evaluation method of entrepreneurs is put forward, from the point of objective programming-theory to alert entrepreneurs concerned to think much of it, which means to take minimized objective function as comprehensive evaluation result and identify disadvantage structure pattern. Application research shows that overall behavior of Chinese entrepreneurs environmental management are good, specially, environment strategic behavior are best, environmental management behavior are second, cultural behavior ranks last. Humic substances are ubiquitous redox-active organic compounds of environment.In this study,experiments were conducted to determine the reduction capacity of humic acid in the matrix of bromate and Fe(Ⅲ) solutions and the role of Fe(Ⅲ) in this redox process.The results showed that the humic acid regenerated Fe(Ⅱ) and reduced bromate abiotically.The addition of Fe(Ⅲ) could accelerate the bromate reduction rate by forming humic acid-Fe(Ⅲ) complexes.Iron species acts as electron mediator and catalyst for the bromate reduction by humic acid,in which humic acid transfers electrons to the complexed Fe(Ⅲ) to form Fe(Ⅱ),and the regenerated Fe(Ⅱ) donate the electrons to bromate.The kinetics study on bromate reduction further indicated that bromate reduction by humic acid-Fe(Ⅲ) complexes is pH dependent.The rate decreased by 2-fold with the increase in solution pH by one unit.The reduction capacity of Aldrich humic acid was observed to be lower than that of humic acid or natural organic matter of Suwanne River,indicating that such redox process is expected to occur in the environment.  相似文献

Diesel exhaust aerosols(DEAs) can absorb and accumulate toxic metal particulates and bacteria suspended in the atmospheric environment, which impact human health and the environment.The use of acoustic standing waves(ASWs) to aggregate DEA is currently considered to be an efficient particle removal method; however, study of the effect of different temperatures on the acoustic aggregation process is scarce. To explore the method and technology to regulate and optimize the aerosol aggregation process through temperature tuning, an acoustic apparatus integrated with a temperature regulation function was constructed. Using this apparatus, the effect of different characteristic temperatures(CTs) on the aerosol aggregation process was investigated experimentally in the ASW environment. Under constant conditions of acoustic frequency 1.286 kHz, voltage amplitude 17 V and input electric power 16.7 W, the study concentrated on temperature effects on the aggregation process in the CT range of 58–72°C. The DEA opacity was used. The results demonstrate that the aggregation process is quite sensitive to the CT, and that the optimal DEA aggregation can be achieved at 66°C. The aggregated particles of 68.17 μm are composed of small nanoparticles of 13.34–62.15 nm. At CTs higher and lower than 66°C, the apparatus in non-resonance mode reduces the DEA aggregation level. For other instruments, the method for obtaining the optimum temperature for acoustic agglomeration is universal. This preliminary demonstration shows that the use of acoustic technology to regulate the aerosol aggregation process through tuning the operating temperature is feasible and convenient.  相似文献

Antimony(Sb) is a toxic metalloid, and its pollution has become a global environmental problem as a result of its extensive use and corresponding Sb-mining activities. The toxicity and mobility of Sb strongly depend on its chemical speciation. In this review, we summarize the current knowledge on the biogeochemical processes(including emission, distribution,speciation, redox, metabolism and toxicity) that trigger the mobilization and transformation of Sb from pollution sources to the surrounding environment. Natural phenomena such as weathering, biological activity and volcanic activity, together with anthropogenic inputs, are responsible for the emission of Sb into the environment. Sb emitted in the environment can adsorb and undergo redox reactions on organic or inorganic environmental media, thus changing its existing form and exerting toxic effects on the ecosystem. This review is based on a careful and systematic collection of the latest papers during 2010–2017 and our research results, and it illustrates the fate and ecological effects of Sb in the environment.  相似文献

研究了代谢调节产物(NaCl)浓度积累对生物法降解二氯甲烷效率的影响。试验表明,一定浓度的NaCl对二氯甲烷降解菌(GD11)的生长有不同程度的抑制作用。摇瓶试验中NaCl对二氯甲烷降解菌的抑制浓度为35.1g/L;当空塔气速91.7m/h、二氯甲烷进口浓度2.02g/m3、温度28.5℃、循环液流量0.06m3/h、pH值6.5~7.5时,生物滴滤塔中NaCl对二氯甲烷降解菌的抑制浓度为23.4g/L,两者相比有一定差异。NaCl浓度对生物膜的厚度有一定影响,这从侧面反映了NaCl浓度对微生物活性的影响。  相似文献

Nitric oxide(NO) is one of the most important air pollutants in atmosphere mainly emitted from combustion source. A biotrickling filter was designed and operated to remove NO from an air stream using bacteria extracted from the sewage sludge of a municipal sewage treatment plant. To obtain the best operation conditions for the biotrickling filter, orthogonal experiments(L9(34)) were designed. Inlet oxygen concentration was found to be the most significant factor of the biotrickling filter and has a significant negative effect on the system. The optimal conditions of the biotrickling filter occurred at a temperature of 40℃, a pH of 8.0 and a chemical oxygen demand of 165 mg/L in the recycled water with no oxygen in the system. The bacteria sample was detected by DNA sequencing technology and showed 93%–98% similarity to Pseudomonas mendocina. Moreover, a full gene sequencing results indicated the bacterium was a brand new strain and named as P. mendocina DLHK. This strain can transfer nitrate to organic nitrogen. The result suggested the assimilation nitrogen process in this system. Through the isotope experimental analysis, two intermediate products(15NO and 15N2O) were found. The results indicated the denitrification function and capability of the biotrickling filter in removing NO.  相似文献

A strain Pandoraea pnomenusa LX-1 that uses dichloromethane(DCM) as sole carbon and energy source has been isolated and identified in our laboratory. The optimum aerobic biodegradation of DCM in batch culture was evaluated by response surface methodology. Maximum biodegradation(5.35 mg/(L·hr)) was achieved under cultivation at 32.8°C, pH 7.3, and 0.66% NaCl. The growth and biodegradation processes were well fitted by Haldane's kinetic model, yielding maximum specific growth and degradation rates of 0.133 hr-1and 0.856 hr-1, respectively. The microorganism efficiently degraded a mixture of DCM and coexisting components(benzene, toluene and chlorobenzene). The carbon recovery(52.80%–94.59%) indicated that the targets were predominantly mineralized and incorporated into cell materials. Electron acceptors increased the DCM biodegradation rate in the following order: mixed oxygen iron sulfate nitrate. The highest dechlorination rate was 0.365 mg Cl-/(hr·mg biomass), obtained in the presence of mixed electron acceptors. Removal was achieved in a continuous biotrickling filter at 56%–85% efficiency, with a mineralization rate of 75.2%. Molecular biology techniques revealed the predominant strain as P. pnomenusa LX-1. These results clearly demonstrated the effectiveness of strain LX-1 in treating DCM-containing industrial effluents. As such, the strain is a strong candidate for remediation of DCM coexisting with other organic compounds.  相似文献

在φ50mm生物滴滤池内进行了二氯甲烷废气净化的实验研究.由工厂活性污泥经驯化培养得到菌种,进一步接种、在填料表面挂膜,接种和挂膜约需30d.滴滤池内装填聚丙烯散堆填料,废气和循环液在滴滤池内逆流操作,循环液pH值为7.0±0.5,温度维持在28.5±2℃的条件下,生物膜系统能较好地适应进口浓度的变化.在气体空床停留时间为15.7s、二氯甲烷进口浓度为0.7~3.12g/m3的范围内,去除效率为72.0%~99.1%.滴滤池中的酸性环境对二氯甲烷的降解具有明显抑制作用.  相似文献

从处理废气的生物滤塔内筛选到1株能高效降解α-蒎烯的菌株PT.通过菌落形态、生理生化特征、16SrRNA基因序列相似性分析及Biolog鉴定等方法,确定该菌株属于荧光假单胞菌(Pseudomonas fluorescens).菌株PT最佳生长条件为: NaCl浓度0.00%、pH7.13、温度25.5℃,降解速率达到最大值5.22mg/(L×h).降解过程符合Haldane’s抑制生长动力学模型,最大比降解速率为0.0364h-1.菌株PT能不同程度地降解一些分子结构较为简单或与α-蒎烯结构相似的工业有机污染物.代谢产物分析表明,菌株PT在降解α-蒎烯的过程中产生柠檬油精、紫苏酸等结构较为简单的物质,它们最终被完全矿化为CO2或合成细胞自身组成物质.碳平衡分析表明,底物有机碳含量完全矿化和转化为细胞生物量的比例分别为64.83%和30.37%.  相似文献

从处理印染废水的厌氧折流板反应器(ABR)系统中分离、纯化并筛选出1株能以苯胺为唯一碳氮源进行代谢的兼性厌氧苯胺降解菌株AN29.经过形态、生理生化特征试验和16S rDNA序列分析结果,鉴定菌株AN29为假单胞菌(Pseudomonas sp.),其特性为:降解苯胺的最适温度为37℃,降解苯胺合适的起始pH值为6.5~8.0,可以利用苯胺的最高浓度为4 000mg/L,合适起始浓度为500~2 000mg/L.  相似文献

在生物滴滤塔中进行低浓度苯乙烯废气的生物净化实验,建立动力学模型,测定苯乙烯实验数据并定性定量检测生物膜微种群。当进口气体浓度为200~1000mg/m3、气体流量0.2~0.25 L/h、液体喷淋量10×10-3~12×10-3m3/h、停留时间50~65s、循环液体pH6.5~7.5时,净化效率为90%~99%。动力学模型的模拟计算值与实验值之间有很好的相关性,相关系数R为0.96~0.99。湿润生物膜微群落的优势菌种群包括恶臭假单胞菌、梭形芽胞杆菌、罗非氏不动杆菌等。恶臭假单胞菌的最大活菌数为5.5×107CFU/g,并随生物滴滤塔运行时间延长有减少趋势。  相似文献

从长期受草甘膦污染的土壤中筛选出10株草甘膦降解菌,其中A5菌株对草甘膦的降解率最高,经鉴定该菌株属于节杆菌属(Arthrobacter)。对A5菌株进行降解特性研究,结表明,该菌株降解草甘膦最适条件为:草甘膦600mg/L,葡萄糖10g/L,NaCl0.5g/L,pH6.5,32℃。在最适降解条件下,该菌株在72h时对草甘膦的降解率达94.2%。  相似文献
