Two types of inoculated sludges, granular sludge that had been stored at-20°C and activated sludge, were investigated for the domestication of aerobic granular sludges(AGSs in sequencing batch reactors(SBRs). The results showed that using the stored granular sludge as inoculation sludge could effectively shorten the domestication time of AGS and yielded mature granular sludge after 22 days of operation. The AGS domesticated by stored granular sludge had better biomass and sedimentation properties; its MLSS and SVI reached8.55 g/L and 35.27 mL/g, respectively. The removal efficiencies for chemical oxygen demand(COD), ammonium nitrogen(NH_4~+-N) and total phosphorus(TP) reached 90.76%, 97.39% and 96.40%, respectively. By contrast, 54 days were needed to obtain mature granules using activated sludge. The microbial community structure was probed by using scanning electron microscopy(SEM) and high-throughput sequencing. The results showed that the diversity of the microbial community in mature granules was reduced when stored granular sludge rather than activated sludge was employed as inoculation sludge, and the dominant microbes were changed. The dominant species in mature granules domesticated using stored granular sludge were Zoogloea, Acidovorax and Tolumonas at the genus classification level, while the dominant species were Zoogloea and TM7-genera in granules developed from activated sludge.  相似文献

In this study, a full-scale internal circulation(IC) reactor coupled with an external circulation system was developed to treat high-strength leachate from a municipal solid waste(MSW)incineration plant, in which anaerobic sludge granulation was intensively investigated. Results showed that the IC reactor achieved excellent treatment performance under high organic loading rates(OLR) of 21.06–25.16 kg chemical oxygen demand(COD)/(m3? day). The COD removal efficiency and biogas yield respectively reached 89.4%–93.4% and 0.42–0.50 m3/kg COD.The formation of extracellular polymeric substances(EPS) was closely associated with sludge granulation. Protein was the dominant component in sludge EPS, and its content was remarkably increased from 21.6 to 99.7 mg/g Volatile Suspended Solid(VSS) during the reactor operation. The sludge Zeta potential and hydrophobicity positively correlated with the protein/polysaccharide ratio in EPS, and they were respectively increased from-26.2 m V and 30.35% to-10.6 m V and 78.67%, which was beneficial to microbial aggregation. Three-dimensional fluorescence spectroscopy(3 D-EEM) and Fourier transform infrared spectroscopy(FT-IR)analysis further indicated the importance of protein-like EPS substances in the sludge granulation. Moreover, it was also found that the secondary structures of EPS proteins varied during the reactor operation.  相似文献

We examined how long-term operation of anaerobic–oxic and anaerobic–anoxic sequencing batch reactors(SBRs) affects the enhanced biological phosphorus removal(EBPR)performance and sludge characteristics. The microbial characteristics of phosphorus accumulating organism(PAO) and denitrifying PAO(DPAO) sludge were also analyzed through a quantitative analysis of microbial community structure. Compared with the initial stage of operation characterized by unstable EBPR, both PAO and DPAO SBR produced a stable EBPR performance after about 100-day operation. From day 200 days(DPAO SBR)and 250 days(PAO SBR) onward, sludge granulation was observed, and the average granule size of DPAO SBR was approximately 5 times larger than that of PAO SBR. The DPAO granular sludge contained mainly rod-type microbes, whereas the PAO granular sludge contained coccus-type microbes. Fluorescence in situ hybridization analysis revealed that a high ratio of Accumulibacter clade I was found only in DPAO SBR, revealing the important role of this organism in the denitrifying EBPR system. A pyrosequencing analysis showed that Accumulibacter phosphatis was present in PAO sludge at a high proportion of 6%,whereas it rarely observed in DPAO sludge. Dechloromonas was observed in both PAO sludge(3.3%) and DPAO sludge(3.2%), confirming that this organism can use both O_2 and NO_3~- as electron acceptors. Further, Thauera spp. was identified to have a new possibility as denitrifier capable of phosphorous uptake under anoxic condition.  相似文献

It was focused on the effect of different sludge concentrations on the performances of an algal-activated sludge symbiotic system in terms of wastewater treatment, algal-activated sludge characteristics and community structure. The results showed that the highest nutrient removal efficiencies were obtained in the reactor R~2 with soluble chemical oxygen demand(sC OD), ammonia nitrogen(NH_4~+-N) and phosphate(PO_4~(3-)-P) removal efficiencies of(90.6 ± 2.3)%,(97.69 ± 2.6)% and(83.81 ± 2.3)%, respectively. Further investigation exhibited that sludge concentration has a great effect on the dissolved oxygen(DO) concentration, the pH, the growth of algae and the extracellular polymeric substance(EPS) production, which resulted in influencing the settleability and the performance of symbiotic system. The denaturing gradient gel electrophoresis(DGGE) analysis demonstrated that the sludge concentration had a selective power for particular members of algae. Meantime, the stimulated algal population would selectively excite the members of bacteria benefited for the formation of algal-bacterial consortia.The variation of microbial compositions, which was influenced by the different sludge concentrations, might be ultimately responsible for the different treatment performances.  相似文献

In this paper, the inhibition of methanogens by phenol in coal gasification wastewater(CGW)was investigated by both anaerobic toxicity tests and a lab-scale anaerobic biofilter reactor(AF). The anaerobic toxicity tests indicated that keeping the phenol concentration in the influent under 280 mg/L could maintain the methanogenic activity. In the AF treating CGW,the result showed that adding glucose solution as co-substrate could be beneficial for the quick start-up of the reactor. The effluent chemical oxygen demand(COD) and total phenol reached1200 and 100 mg/L, respectively, and the methane production rate was 175 m L CH4/g COD/day.However, if the concentration of phenol was increased, the inhibition of anaerobic micro-organisms was irreversible. The threshold of total phenol for AF operation was 200–250 mg/L. The extracellular polymeric substances(EPS) and particle size distribution of anaerobic granular sludge in the different stages were also examined, and the results indicated that the influence of toxicity in the system was more serious than its effect on flocculation of EPS. Moreover, the proportion of small size anaerobic granular sludge gradually increased from10.2% to 34.6%. The results of high through-put sequencing indicated that the abundance of the Chloroflexi and Planctomycetes was inhibited by the toxicity of the CGW, and some shifts in the microbial community were observed at different stages.  相似文献

Aerobic granules, pre-cultivated at the organic loading rate (OLR) of 3.0 kg COD/(m3 ·day), were used to treat low-strength wastewater in two sequencing batch reactors at low OLRs of 1.2 and 0.6 kg COD/(m3 ·day), respectively. Reactor performance, evolution of granule morphology, structure and microbial community at low OLRs under long-term operation (130 days) were investigated. Results showed that low OLRs did not cause significant damage to granule structure as a dominant granule morphology with size over 540 μm was maintained throughout the operation. Aerobic granules at sizes of about 750 μm were finally obtained at the low OLRs. The granule reactors operated at low OLRs demonstrated effective COD and ammonia removals (above 90%), smaller granule sizes and less biomass. The contents of extracellular polymeric substances in the granules were decreased while the ratios of exopolysaccharide/exoprotein were increased (above 1.0). The granules cultivated at the low OLRs showed a smoother surface and more compact structure than the seeded granules. A significant shift in microbial community was observed but the microbial diversity remained relatively stable. Confocal Laser Scanning Microscopy observation showed that the live cells were spread throughout the whole granule, while the dead cells were mainly concentrated in the outer layer of the granule, and the proteins, polysaccharides and lipids were mainly located in the central regime of the granule. In conclusion, granules cultivated at high OLRs show potential for treating low-strength organic wastewater steadily under long-term operation.  相似文献

Aluminum flocculant can enhance the flocculating performance of activated sludge.However,the binding mechanism of aluminum ion(Al 3+) and extracellular polymeric substances(EPS) in activated sludge is unclear due to the complexity of EPS.In this work,threedimensional excitation emission matrix fluorescence spectroscopy(3DEEM),fluorescence quenching titration and Fourier transform infrared spectroscopy(FT-IR) were used to explore the binding behavior and mechanism between Al 3+ and EPS.The results showed that two fluorescence peaks of tyrosineand tryptophan-like substances were identified in the loosely bound-extracellular polymeric substances(LB-EPS),and three peaks of tyrosine-,tryptophanand humic-like substances were identified in the tightly boundextracellular polymeric substances(TB-EPS).It was found that these fluorescence peaks could be quenched with Al 3+ at the dosage of 3.0 mg/L,which demonstrated that strong interactions took place between the EPS and Al 3+.The conditional stability constants for Al 3+ and EPS were determined by the Stern-Volmer equation.As to the binding mechanism,the-OH,N-H,C=O,C-N groups and the sulfurand phosphorus-containing groups showed complexation action,although the groups in the LB-EPS and TB-EPS showed different behavior.The TB-EPS have stronger binding ability to Al 3+ than the LB-EPS,and TB-EPS play an important role in the interaction with Al 3+.  相似文献

The effects of tourmaline on nitrogen removal performance and biofilm structures were comparatively investigated in two identical laboratory-scale sequencing batch biofilm reactors(SBBRs)(denoted SBBR1 and SBBR2) at different nitrogen loading rates(NLRs) varying from(0.24 ± 0.01) to(1.26 ± 0.02) g N/(L·day). SBBR1 was operated in parallel with SBBR2, but SBBR1 was filled with polyurethane foam loaded tourmaline(TPU) carriers and another(SBBR2) filled with polyurethane foam(PU) carriers. Results obtained from this study showed that the excellent and stable performance of SBBR1 was obtained. Ammonia nitrogen removal and total nitrogen removal were higher in SBBR1 than that in SBBR2 with increase of NLR. At an NLR of(0.24 ± 0.01) g N/(L·day), the majority of the spherical and elliptical bacteria were surrounded by the extracellular polymeric substance(EPS) and bacillus or filamentous bacteria in two SBBRs biofilms. When NLR increased to(1.26 ± 0.02) g N/(L·day), the clusters were more obvious in the SBBR1 biofilm than that in the SBBR2 biofilm. Bacteria in SBBR1 were inclined to synthesis more EPS, and the formed EPS could protect the bacteria from free ammonia(FA) under extreme condition NLR(1.26 ± 0.02) g N/(L·day). The results of polymerase chain reaction-denaturing gradient gel electrophoresis analysis showed that the microbial community similarity in SBBR2 decreased more obviously than that in SBBR1 with the increase of NLR, which the microbial community in SBBR1 was relatively stable.  相似文献

The bacterial strain Paracoccus denitrificans W12, which could utilize pyridine as its sole source of carbon and nitrogen, was added into a membrane bioreactor (MBR) to enhance the treatment of a pharmaceutical wastewater. The treatment efliciencies investigated showed that the removal of chemical oxygen demand, total nitrogen, and total phosphorus were similar between bioaugmented and non-bioaugmented MBRs, however, significant removal of pyridine was obtained in the bioaugmented reactor. When the hydraulic retention time was 60 hr and the influent concentration of pyridine was 250-500 mg/L, the mean effluent concentration of pyridine without adding W12 was 57.2 mg/L, while the pyridine was degraded to an average of 10.2 mg/L with addition of W12. The bacterial community structure of activated sludge during the bioaugmented treatment was analyzed using polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE). The results showed that the W12 inoculum reversed the decline of microbial community diversity, however, the similarity between bacterial community structure of the original sludge and that of the sludge after bioaugmentation decreased steadily during the wastewater treatment. Sequencing of the DNA recovered from DGGE gel indicated that sp., Sphingobium sp., Comamonas sp., and Hyphomicrobium sp. were the dominant organisms in time sequence in the bacterial community in the bioaugmented MBR. This implied that the bioaugmentation was affected by the adjustment of whole bacterial community structure in the inhospitable environment, rather than being due solely to the degradation performance of the bacterium added.  相似文献
