Diabetes mellitus is a worldwide leading metabolic syndrome, associated with profound alterations in carbohydrate, lipids, lipoproteins and protein metabolisms. Worldwide, traditional practitioners for the treatment of diabetes and its complications use a wide variety of medicinal plants. In the present study the aqueous extract of Tephrosia purpurea leaves (TpALet) was evaluated for its antihyperglycemic and antihyperlipidemic effects in streptozotocin induced diabetic rats. Profound alterations in the concentrations of blood glucose, lipids and lipoproteins were observed in diabetic rats. Oral administration of TpALet to diabetic rats at a dose of 600 mg/kg body weight significantly reduced the level of blood glucose and increased the level of plasma insulin as well as normalized the lipids and lipoproteins profile. The present study thus demonstrated that TpALet has prominent antihyperglycemic and antihyperlipidemic effects in streptozotocin induced diabetic rats.  相似文献

Protective effects of quercetin against oxidative stress induced by sodium fluoride intoxication in rat erythrocytes were evaluated. Rats were divided into five groups consisting of 10 in each for this experiment. The animals of group I received water and standard diet to serve as control group, the animals of groups II and III were treated with quercetin (10 and 20?mg?kg?1 body weight), administrated intraperitoneally for 7 days followed by sodium fluoride (600?ppm) in drinking water for the next 7 days. The animals of group IV were treated with vitamin C (10?mg?kg?1) intraperitoneally for 7 days followed by sodium fluoride treatment for next 7 days serving as positive control group. The animals of group V were treated only with sodium fluoride (600?ppm) for the same time and were used as control group. Blood sample were collected via retro-orbital puncture. The antioxidant enzymes, superoxide dismutase, and catalase, as well as the levels of reduced glutathione and lipid peroxidation end products were measured in erythrocytes. There was a significant increase in lipid peroxidation along with a decrease in superoxide dismutase activity in the erythrocytes of sodium fluoride-treated animals. Quercetin treatment prior to fluoride administration normalized the levels of all parameters measured in the rat erythrocytes.  相似文献

In order to study the time-dependent effects of chlordane on thyroid hormone levels and histological structure in rats, 120 healthy 4-week-old SD rats were divided randomly into 12 groups. Rats in treatment groups were treated orally by gavage with chlordane at a dose of 15 mg kg?1 for 5, 10, 15, 20, 30, or 40 days, while rats in control group received an equal volume of corn oil. Serum levels of thyroxine (FT4), triiodothyronine (FT3), and thyroid stimulating hormone (TSH) were measured by radioimmunoassay. Sections of thyroids were dyed with hematoxylin–eosin to detect any pathological alterations. In chlordane treated groups, on day 5, changes in serum levels of FT4, FT3, and TSH were not found. On days 10 and 15, serum levels of FT4 and FT3 decreased, accompanied by increased levels of TSH. On days 20, 30 and 40, serum levels of these hormones returned to control values. As for alterations in histology, on day 10–15, follicular epithelium of the thyroid were found to proliferate into two layers in chlordane treated rats. On day 20, in focal areas the two-layer afollicular epithelia were observed to continuously proliferate into 3–4 layers of follicular epithelium, termed hyperplasia plaques. On days 30 and 40, solid buds and secondary follicles to solid buds were detected. The earliest alteration of thyroid hormones in treated SD rats occurred between 5 and 10 days with corresponding histopathologic changes. As the exposure time increased, the serum levels of hormones and histopathology altered in a time-dependent manner.  相似文献

通过探究iNOS/p38 MAPK信号通路在丙烯腈(acrylonitrile,ACN)诱导脑组织损伤中的作用,为进一步研究ACN的神经毒性作用提供依据。选取50只SPF级健康成年雄性SD大鼠,随机分为5组,每组10只。适应性饲养一周后,以12.5、25.0、50.0 mg·kg~(-1)ACN对大鼠进行灌胃染毒,对照组给予玉米油,另设NAC组(300.0 mg·kg~(-1)NAC+50.0 mg·kg~(-1)ACN),1次·天~(-1),6天·周~(-1),共染毒13周。次日称重并处死大鼠,测定大鼠脑组织NO含量、总NOS水平及iNOS、p-p38和p38蛋白表达水平。结果显示,ACN各剂量组大鼠脑组织脏器系数与对照组比较均显著降低(P0.05),高剂量组大鼠脑脏器系数与NAC组比较降低(P0.05)。高剂量组NO含量和总NOS水平显著高于对照组,与NAC组比较,高剂量组NO含量降低(P0.05),总NOS水平升高(P0.05)。Western blot结果显示,ACN高剂量组大鼠脑组织iNOS、p-p38蛋白表达水平和p-p38/p38比值显著高于对照组和NAC组(P0.05)。ACN可激活iNOS/p38MAPK信号通路,这可能是ACN致大鼠脑组织损伤的机制之一。  相似文献

In order to examine the time-dependent effects of pentabrominated diphenyl ether (BDE-99) on thyroid hormones levels and histological structure in rats, 4-week-old SD rats were divided into 10 groups randomly, according to their body weight. Rats in treatment groups were orally gavaged with BDE-99 at the dose of 60 mg kg?1 for 10, 15, 20, 30 or 40 days, while rats in control group received equal volume of corn oil. Serum levels of thyroxine (FT4), triiodothyronine (FT3), and thyroid stimulating hormone (TSH) were measured by radioimmunoassay. Sections of thyroids were dyed with hematoxylin-eosin (HE) to detect any pathologically alterations. In BDE-99 treated groups, on day 10, changes in serum levels of FT4, FT3 and TSH were not found. The effects of lithium (Li) supplementation on lipid profile and fluidity of cerebrum and cerebellum membranes in aluminum (Al)-treated rats were investigated. A significant decrease in the levels of total lipids and cholesterol was observed in both the regions following Al exposure, which however were significantly increased following Li supplementation. Further, glycolipids and gangliosides contents were significantly decreased in cerebrum, but increased in cerebellum following Al treatment. Interestingly, Li supplementation reversed the trends and regulated the levels of glycolipids and gangliosides. Al treatment also elevated conjugated diene formation and phospholipid levels in both cerebrum and cerebellum membranes, which however were decreased upon Li supplementation. The cholesterol/phospholipid ratio however was decreased after Al treatment and Li supplementation was able to enhance the ratio in both cerebrum and cerebellum. Further, Al treatment significantly increased the fluorescence polarization, anisotropy and order parameter, which however were normalized following Li supplementation. The levels of Al were also found to be significantly elevated in cerebrum and cerebellum after Al treatment and normalized in cerebellum following Li treatment. Therefore, the present study shows the potential of Li in regulating the changes produced by Al on membrane composition and fluidity in rat brain.  相似文献

Chemical mixtures are an important area of research as individuals are exposed to low doses of persistent chemical agents known as environmental pollutants throughout their life time. Polychlorinated biphenyls (PCBs) and diethyl phthalate (DEP) are ubiquitous environmental pollutants that could be present in the same environmental compartment; hence organisms may get simultaneously exposed to both. Therefore, a study was undertaken to see whether PCB and DEP together show interactive chronic mixture toxicity in male Wistar rats. Healthy male Wistar rats weighing 70–100?g were randomly assigned to four groups of six each. Control rats were fed on normal diet and water ad libitum. Oil control rats were maintained on a normal diet mixed with corn oil. Rats were given Clophen A60 (PCB) and DEP dissolved individually in corn oil mixed with the diet at 50?mg?kg?1 of the diet/day, as well as a mixture in corn oil mixed with the diet both at 50?mg?kg?1 of the diet/day. After 150 days of treatment animals were sacrificed and enzymes and other biochemical parameters in the serum and liver were assessed. Liver weight to body weight ratio showed a significant increase in Clophen A60 and in Clophen A60?+?DEP treated rats. In the DEP, Clophen A60 and Clophen A60?+?DEP treated groups there was significant increase in liver and serum alanine aminotransferase (ALT) and acid phosphatase (ACP) activity. Aspartate aminotransferase (AST) was significantly increased in the liver and serum of DEP treated rats only. Cholesterol levels were significantly increased only in the serum and the liver of DEP treated rats. Triglyceride levels were significantly increased in the serum of treated rats and only in the liver of Clophen A60 and Clophen A60?+?DEP treated rats. Liver glycogen levels were significantly increased in DEP and Clophen A60?+?DEP treated rats. In all treated animals, there was a significant decrease in liver glutathione reductase (GR). Histology of liver showed severe vacuolations, fatty degeneration and loss of hepatic architecture in Clophen A60 and Clophen A60?+?DEP treated rats, whereas in DEP treated rats only loss of hepatic architecture and granular deposits in the hepatocytes was predominant with mild vacuolations of centrilobular and periportal area. It is evident from this study of mixture toxicity of Clophen A60 and DEP that there is no significantly enhanced toxicity due to the interaction of these two compounds. On the other hand, to some extent there is alleviation in toxicity as evidenced by enzyme ACP and AST levels in the liver. The hepatocellular damage and biliary congestion caused by these two compounds, which can be confirmed by significantly increased liver weights and elevated serum and liver enzyme levels as well as histology, was almost the same between individual and mixture treated group.  相似文献

The aim of this study was to investigate the effects of curcumin (CUR) on antioxidant status, body weight (BW) gains, and some reproductive parameters in male rats exposed to subchronic doses of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). Thirty-two rats were divided into four groups. The first group was kept as control. The second group (TCDD group) was given TCDD at a dose of 50 ng·kg?1 BW per day; the third group (CUR group) was treated with CUR at a dose of 80 mg·kg?1 BW per day. The fourth group (TCDD + CUR group) was given TCDD and CUR at the same doses simultaneously. Malondialdehyde (MDA) levels were significantly increased in the TCDD group. In addition, TCDD exposure decreased liver superoxide dismutase (SOD) activity, catalase (CAT) activities of kidney and brain, glutathione peroxidase (GSH-Px) activities of liver, kidney, and brain, and glutathione levels of liver, kidney, and heart. However, CUR treatment with TCDD exposure decreased MDA levels in all tissues and increased SOD activities of liver, kidney, and brain, CAT activity of heart, and GSH-Px activities of heart and brain. TCDD caused a decrease in BW gain, and CUR partially eliminated this effect of TCDD. In addition, while reproductive organ weights, sperm concentration, and sperm motility tended to decrease with TCDD exposure, these effects tended to be close to normal levels by CUR treatment. In conclusion, CUR was seen to be effective in the treatment and prevention of toxicity induced by subchronic TCDD exposure.  相似文献

为探讨2,3,7,8-四氯二苯并二噁英(TCDD)和多氯联苯(Aroclor 1254)联合染毒对大鼠外周血淋巴细胞DNA的损伤效应,将20只雄性Sprague-Dawley(SD)大鼠随机均分为4组,即对照组(橄榄油)、Aroclor 1254单独染毒组(10mg·kg-1)、TCDD单独染毒组(10μg·kg-1)和联合染毒组(TCDD 10μg·kg-1+Aroclor 1254 10 mg·kg-1).大鼠每天灌胃染毒1次,连续染毒6d.染毒过程中记录大鼠的体征和体重.末次染毒后24h取外周血,分离淋巴细胞,采用碱性单细胞凝胶电泳技术(彗星试验)检测外周血淋巴细胞DNA损伤,并采用CASP软件分析各组彗星的尾部DNA百分含量(Tail DNA%)、尾长(Tail Length)和尾矩(Tail Moment).结果表明,染毒结束时TCDD单独染毒和联合染毒组大鼠体重显著低于对照组,且联合染毒组表现更为明显.TCDD单独染毒组和联合染毒组大鼠外周血淋巴细胞DNA可见明显损伤,TailDNA%、Tail Length和Tail Moment均显著高于对照组(p<0.05),且联合染毒组大鼠细胞DNA损伤更为严重,但Arolor 1254单独染毒组大鼠未见明显DNA损伤.析因分析提示TCDD与Aroclor 1254对大鼠外周血淋巴细胞DNA损伤的联合毒性效应为协同作用.本研究结果表明TCDD与Aroclor 1254联合染毒可加重大鼠外周血淋巴细胞DNA损伤,在对PCBs与二噁英的混合暴露进行危险性评估时要注意这种联合毒性作用.  相似文献

An attempt has been made to study the influence of taurine on mercury intoxicated rats. The animals were treated with sublethal dose of mercuric chloride (2 mg/kg body wt.) for 30 days. During the mercury treatment, the level ofAspartate transaminase(AST), Alanine transaminase (ALT) and Alkaline phosphatase(ALP) in serum and lipid peroxidation (LPO) in liver tissue significantly increased whereas Glutathione (GSH), Glutathione peroxidase(GPx), Catalase (CAT) and Superoxide dismutase (SOD) were simultaneously decreased in the liver tissue. Present results indicate that the liver tissue was completely damaged, after mercury treatment. In another group of animals, taurine (5 mg/kg body wt.) was administrated for another 15 days. Taurine administration was observed to improve the liver function in mercury intoxicated animal as indicated by the decline in increased levels of AST, ALT and ALP in serum and LPO content in liver tissue. The decreased level of antioxidant system (GSH, GPx, CATand SOD) has been promoted Results suggested that taurine played a vital role in reducing the mercury toxicity in intoxicated animals.  相似文献
