Cypermethrin (CY) is an active cyano pyrethroid effective against a wide range of pests encountered in agriculture and forestry. Although CY is not mutagenic in in vitro assays for gene mutation, in vivo assays showed conflicting results. In vivo genotoxicity of the synthetic pyrethroid CY in erythrocytes of Odontophrynus americanus tadpoles was examined. The frequency of micronuclei (MN) was recorded in blood smears obtained from tadpoles exposed in vivo to four different nominal concentrations 5, 10, 20 or 40?µg?L?1 of the compound and fixed at two sampling times 48 and 96?h. As a positive control larvae were exposed to 40?mg?L?1 of cyclophosphamide (CP). Tadpoles exposed to all CY treatments showed a significant increase in single small MN compared to the negative control group after 48?h and at 5 and 10?µg?L?1 of CY at 96?h. Results obtained here demonstrated the genotoxic effects of the commercial formulation CY in the anuran larvae analyzed. Thus, data suggest that measurements of MN and other erythrocytes morphological aberrations performed in circulating blood samples of O. americanus tadpoles is a method for detecting cytogenetic damage in other native species.  相似文献

Studies were carried out to determine the toxicity of some selected pesticides on fresh water fish in a tropical environment. The uptake of the pesticides lindane, pentachlorophenol (PCP), and propoxur, which are frequently used by farmers and industrialists were studied in concrete ponds at the University of Cape Coast, in Ghana. The fishes used for the study were Oreochromis niloticus, Clarias gariepinus, and Chrysicthys nigrodigitatus, and were obtained from cultured ponds in the Cape Coast District and Mankessim in the Central Region and Weija Dam, in the Greater Accra region of Ghana. Single high lethal concentration (SD) or acute treatment and cumulative/chronic (or multiple minor) lethal concentration (CD) treatment were employed in administering the pesticides to the fishes via water. Gas chromatograph (GC) Electron Capture Detector (ECD) analysis was done on the dead fishes to see the extent of ingestion. The LC50 values obtained for lindane on the three fish samples were as follows: Chrysicthys?–?0.38?mg?L?1; Oreochromis?–?0.42?mg?L?1, and Clarias?–?1.2?mg?L?1. Mortalities occurred in fish within 3–5 days of application. For the PCP on Chrysicthys, Oreochromis, and Clarias species the LC50 values were 0.42, 0.32 and 0.64?mg?L?1, respectively for over a 2–3 day period. For a three-time influx period of propoxur the LC50 for Chrysicthys, Oreochromis, and Clarias, were 22.0, 30.40, and 45.04 (all in mg?L?1), respectively. The results obtained indicated that the pesticides had adverse effects on the general growth and reproduction of fishes. Gonadosomatic indices also showed that the pesticides affected the development of the body, the gonads, and their reproduction.  相似文献

The toxicities of seven chemicals [cadmium (Cd) chloride, potassium dichromate, sodium dodecyl sulfate, potassium chloride (KCl), Triton X-100, zinc (Zn) chloride and copper (Cu) (II) sulfate] were determined using the tropical freshwater cladoceran, Ceriodaphnia rigaudii and the temperate species, Daphnia magna. Data generated was subsequently used to determine the relative sensitivities of both species, using interspecies correlation to compare the 48?h LC50 values for both species. The 48?h LC50 values for C. rigaudii ranged from 0.002?mg?L?1 (potassium dichromate) to 21.1?mg?L?1 (KCl), whereas those for D. magna ranged from 0.3?mg?L?1 for Cu to 418.87?mg?L?1 for KCl. The LC50 values for C. rigaudii were significantly less than that for D. magna for six of the compounds tested. The interspecies correlation also showed a low, positive correlation suggesting that the sensitivities of both species were not similar for the compounds tested. The sensitivity factors for C. rigaudii and D. magna ranged between 0.01 and 12.3. These values further suggested that for six of the compounds tested, C. rigaudii appeared to be more sensitive than D. magna. It is therefore possible that because of these differences in relative sensitivities, toxicity data generated with D. magna may not be appropriate for Trinidad.  相似文献

Ammonia toxicity tests were performed with Labeo bata (bata) larvae of three different size groups. One hundred percent survival of larvae (500.0?±?4.0?mg) was recorded when exposed to ammonia concentrations of 1.0–13.56?mg?L?1 at 96?h of exposure. Bata larvae exposed to ammonia concentrations of 15.8–25?mg?L?1 showed 10–74% mortalities. The 96?h LC50 value for 200 (±5), 250 (±2) and 500 (±4) mg bata larvae were 11.5, 16.8 and 22.5?mg?L?1 un-ionised ammonia concentrations, respectively. When fish were exposed to different doses of ammonia, behavioural changes immediately occurred even at the lowest dose. At first, the fish became hyperexcitable, the skin darkened and they showed an increased ventilation frequency, fish behaviour became normal, 24?h after exposure. A 96?h LC50 value of un-ionised ammonia showed direct relationship with the increasing size of bata larvae.  相似文献

In the present study, an attempt has been made to quantify the fenvalerate accumulated in different tissues (gill, muscle and liver) and observe changes involved in the levels of sodium, potassium and calcium ions and Na+–K+, Mg2+ and Ca2+ adenosine triphosphatase (ATPase) activities in the freshwater fish, Cirrhinus mrigala on short-term and long-term exposure to the median lethal and sublethal concentration of fenvalerate. Residue analysis using gas–liquid chromatography (GLC) technique revealed that fenvalerate accumulated in highest quantity in gill followed by liver and muscle under median lethal concentration (6?µg?L?1). Whereas in sublethal concentration (0.6?µg?L?1), muscle accumulated highest quantity followed by gill and liver, which might be due to the fact that fenvalerate is highly lyphophilic. The ion concentration and ATPase activity were found effected in fish exposed to lethal and sublethal concentrations of fenvalerate. Concentration of Na+, K+ and Ca2+ ions decreased in gill, muscle and liver on being exposed to median lethal concentration to a significant level. Whereas the changes were not highly pronounced at sub lethal level indicating low concentration of fenvalerate and its non-toxic effect at chronic exposure. Na+–K+, Mg2+ and Ca2+ ATPases activity were also found decreased in correspondence to the ionic change under median lethal and sub lethal concentrations in target tissues. This might have lead to behavioural changes and create wide-spread disturbance in the normal physiology, ultimately causing the death of the fish. The results suggest that in biomonitoring programmes, ions and associated ATPases can be a good diagnostic tool for fenvalerate toxicity.  相似文献

The toxic potency of three locally formulated drilling mud lubricants against Paleomonetes africanus, a brackish water crustacean commonly found in the oil producing Niger Delta (Nigeria), was evaluated. A 96?h exposure to quantal (lethal) doses of the mud lubricants gave LC50 (median lethal concentration) of 350, 620 and 210?mg?L?1, respectively. The mud lubricant with the highest molecular weight of 40,000?g?mol?1 (lubricant 2) had the least toxicity on the organisms, attributable to the reduced rate of diffusion into the vital cells of the test organism. The higher toxicity of lubricant 3 over lubricant 1, despite its higher molecular weight, might be as a result of its higher acid value of 21.60?mg?KOH?g?1. The results show that the test lubricants would be environmentally friendly within the baseline concentrations and lethal times (LT50) measured.  相似文献

The toxic effects of an organophosphorous insecticide, monochrotophos (MCP) were investigated on Paramecium caudatum in static acute toxicity tests (10?min and 2?h). The lethal concentrations (50%) were determined by probit method, for technical monocrotophos as 60 and 40.6?mg?L?1, respectively. We have combined conventional light microscopy and a computerized video tracking system to determine behavioural and morphological changes in paramecium on exposure to MCP. Paramecia exposed to highest concentrations (90–100?mg?L?1) used for 10?min exposure, exhibited initial increase with subsequent decrease in mobility with enormous blebbing, leading to lysis of cells. In the second set of experiments, the cells exposed to lethal concentration (40.6?mg?L?1) for 2?h were under stress, and reduced their locomotor behaviour, i.e. distance travelled per unit time (mm in 6?min) and swimming speed (mm?s?1) with increased time of exposure. In the third set of experiments, the number of generations and generation time in 24?h was evaluated with respect to the different sub lethal concentrations (5, 10, 15 and 20?mg?L?1) of toxicant. The number of generations decreased and generation time extended significantly in a concentration dependent manner. The results indicate that the Paramecium toxicity assay could be used as a complimentary system to rapidly elucidate the cytotoxic potential of insecticides.  相似文献

Scanning electron microscopic (SEM) study of gills of Catla catla catla (17-day-old) exposed to UV-B radiation (145?µW?cm?2 at the water surface) for three different exposure times: 5, 10 and 15?min was conducted. Fish without UV-B exposure served as control. UV-B radiation damaged both gill filaments and lamellae. The intensity of damage was minimal in 5?min exposed fish, followed by 10?min exposed fish and maximal in 15?min. The gill epithelium was severely damaged in 15?min irradiated fish compared to control. Pavement cells (PVCs) were damaged and the numbers of microridges within PVCs decreased. The deep boundary of PVC was not clear. In some area of gill epithelium, PVCs were destroyed and mitochondrion-rich cells (MRCs) were exposed. The 5?min exposure reduced the number of microridges in the PVCs, but the boundary of PVCs was still visible. MRCs in the gill epithelium were not exposed in 5?min exposed fish. The damage to PVCs and subsequent exposure of MRCs in UV-B irradiated fish may hamper respiratory functions and disturb osmoregulation in catla.  相似文献

Fish of species Parachanna obscura are exposed to four concentrations of cadmium (Cd) (0.02, 0.06, 0.18, or 0.54?mg?L?1). Acute toxicity test results after 96?h showed marked hematological, histological, and behavioral changes. Quantitative blood analysis revealed that white blood cell (WBC), packed cell volume (PCV), and platelets counts were linearly correlated with increasing Cd concentrations; however, blood haemoglobin (Hb) showed an inverse relationship, with higher Hb levels at lower Cd concentrations. Histological changes of the liver showed hepatic separation, bloody appearance, and deposition of a brownish-yellow substance, which increased with rising concentrations of metal. Behavioral changes observed included reduced activity, avoidance response, and deposition of a slimy-whitish film on the body of the fish, which increased with concentration and exposure time.  相似文献

A study was conducted on the Chironomus plumosus larvae to determine initiation of resistance to four pesticides – chlorfenvinphos, chlorpyrifos, chlorpyrifos-methyl, and malathion. First generational LC50 values were well within the threshold value for chironomids based upon the literature. Subsequent LC50 values were observed to increase, indicating a lessening of the toxicity of the pesticides to the chironomid. In the case of chlorfenvinphos, the 96 h LC50 for generations 1–23 was 6 µg L?1, in generations 3–7 was 8.57 µg L?1 and 11.14 µg L?1 for generations 8–9. Generations 10–12 had an LC50 value of 22.58 µg L?1 and generation 13 had an LC50 value of 35.08 µg L?1. Generation 14 had an LC50 value of 47.58 µg L?1. Generations 15–19 and 20–24 had 96 h LC50 values of 60.68 µg L?1, 72.58 µg L?1, 85.08 µg L?1, 97.58 µg L?1 and 110.08 µg L?1, respectively. Generations 25, 26 to 30, 31 to 38 and 39 to 45 had 96 h LC50 values of 160.42 µg L?1, 210.7 µg L?1, 262.24 µg L?1 and 274.36 µg L?1, respectively. The variation between LC50 values was found to be statistically significant. This was observed for most pesticides tested. Larval size and life cycle duration was observed to change from generation to generation with the body size decreasing markedly from 1 to 0.3 cm with life cycle increasing from 7 to 39 days.  相似文献

Bisphenol A (BPA), the plastic monomer and plasticizer with well-known endocrine-disrupting chemical properties, is widely present in the aquatic environment, but little is known regarding hepatic toxicity in fish. This study assessed the effects of chronic exposure to BPA on the antioxidant defense system of Japanese medaka liver. Fish were exposed to sub-lethal concentrations of BPA (0.1, 1, 10, 100, or 1000?µg?L?1) for 60 days. Antioxidant parameters were determined and compared with controls. The results showed that the activities of catalase (CAT), superoxide dismutase, glutathione peroxidase, glutathione S-transferase, and the content of reduced glutathione were all induced at 10?µg?L?1 BPA after 60 days. Further, hepatic CAT was sensitive to BPA at 1?µg?L?1 concentration. In conclusion, CAT may serve as a potential early biomarker of BPA exposure in aquatic environment.  相似文献

The article presents the results of screening research on acute toxicity of groundwater contaminated by leachates to the bacterium Vibrio fischeri (ToxAlert 100® test) and crustacean Daphnia magna (Daphtoxkit F? magna test according to PN-EN ISO 6341), as well as on chronic toxicity to D. magna (test according to ISO 10706). The groundwater samples were taken from seven large city landfills in the Pomeranian province (Poland), which were open before 1990 and designed without proper attention to being leak-tight. Due to the lack of either European or Polish classification of ecotoxicological qualities of surface and underground waters, an attempt was made to evaluate the ecotoxicological quality of the examined waters on the basis of the Helsinki Committee's recommendations for industrial effluents, the classification of sediment according to Krebs (Krebs F. Coastal marine ecosystems in many parts of the world are under unrelenting stress caused by urban development, pollutants and other ecological impacts such as building of infrastructure, land reclamation for port and industrial development, habitat modification, tourism and recreational activities. The present work is a first extensive field study using the marine sponge, Sigmadocia fibulata as a bioindicator to detect metal pollution between inshore and offshore environment of the ‘Gulf of Mannar’, India. Samples of S. fibulata were collected from different places of inshore (0.1–0.5?km) and offshore (3–5?kms) locations. Metal concentrations in water and bioaccumulation in sponges were determined by ICP-MS (Inductively coupled plasma-mass spectrometry). Enrichment of metal contamination was more in the inshore, when compared to offshore environment. Higher concentrations of metals Fe, Al, Ni, Mn, Cu, Cr, Co, Ba, Zn, V, Pb and Cd in inshore waters (3–6.4 times than offshore) may be due to the discharge of monsoonal rains, carrying a discharge of industrial and agricultural wastes and sewage directly into the coast. This is substantiated by a highly significant positive correlation between concentrations of metals in the water and accumulation in sponges. The bioaccumulation of metals in sponge tissue were in order of Fe?>?Al?>?Ni?>?Mn?>?Cu?> Cr?>?Co?>?Ba?>?Zn?>?V?>?Cd?>?Pb in both the inshore and offshore regions. Apparently, the macromolecule composition (sugars, proteins and lipids) was significantly reduced by the accumulation of metals in inshore sponges. The current findings indicated that the frequent monitoring is necessary to assess the eco-health of the marine environment by choosing bio-indicator species like S. fibulata, which provide accurate, reliable measurements of environmental quality.  相似文献

We investigated the leaching characteristics of bisphenol A (BPA) from two kinds of epoxy-resin pavement materials, one containing epoxy resins (EPs) and the other containing epoxy-acrylate resins (EPAs). Both samples contained residual BPA monomer, at levels of 9.0?µg?g?1 for the EP resin sample and 4.4?µg?g?1 for the EPA resin sample. These amounts were larger than amounts previously measured for polycarbonate samples. The amount of BPA leached from the samples increased with temperature. The leaching of BPA from EP was more strongly affected by temperature than the leaching from EPA. The pH also affected the amount of leached BPA. The maximum leached amount was observed under alkaline conditions (pH 10.8) for both sample types. The amounts of BPA that might leach from pavement materials during 1?h of heavy rain were estimated to be 0.9?µg?m?2 for EP and 3.5?µg?m?2 for EPA. Our results indicate that EPs disposed of in waste landfills without any treatment may be a source of BPA in leachate at landfill sites.  相似文献

The sublethal effect of silver and chromium on some biochemical constituents was studied in the green mussel Perna viridis. A study on the toxicokinetic behavior, metabolism of chlorpropham, and its effect on cytochrome P450 from liver microsomes was carried out in albino rats after a single and consecutive oral administration at 500?mg?kg?1 body weight for 10 and 20 days. Chlorpropham was detected in the blood at 0.08?h (11.43?±?1.72?µg?mL?1) reaching a maximum concentration at 2?h (30.90?±?2.55?µg?mL?1) and a minimum at 48?h (1.95?±?0.20?µg?mL?1) after a single oral administration of 500?mg?kg?1. The absorption rate constant (K a) was 0.66?±?0.48?h?1. The Vd area (18.01?±?2.78?L?kg?1) and t 1/2 β (12.23?±?1.96?h) values suggested a wide distribution and long persistence of the compound in the body, respectively. The higher ClR (0.82?±?0.00?L?kg?1?h?1) compared to ClH (0.18?±?0.02?L?kg?1?h?1) value indicated that a major portion of chlorpropham was excreted through the urine (30%) compared to the faeces (2.81%). Chlorpropham residue was detected in all tissues of rat at 0.25?h while its metabolite, meta-chloroaniline was detected in liver, kidney, heart, lung, and spleen tissue at 0.25?h. Meta-chloroaniline was not detected in skeletal muscle, brain, fat, and stomach tissue at any time of the observation period. Maximum concentrations of chlorpropham and meta-chloroaniline were detected at 2?h (except in the spleen), and minimum concentrations of chlorpropham at 24 (heart, lung, spleen, skeletal muscle, and stomach) and 48?h (liver, kidney, brain, and fat tissue) respectively; and meta-chloroaniline at 24?h (except heart and spleen). The tissue half-life of chlorpropham in rat varied from 3.80 to 11.60?h. Repeated oral administration of chlorpropham at 500?mg?kg?1 for 10 and 20 days caused an induction of the liver microsomal pellet of rat.  相似文献

为了探讨双酚A(bisphenol A,BPA)对海洋微藻的生态毒性效应,实验选择了以青岛大扁藻(Platymonas helgolanidica)作为受试物种,设置6个实验浓度(即0、2、4、6、8、10 mg·L-1)对微藻进行了96 h暴露处理,测定了不同浓度暴露下对青岛大扁藻的生长以及抗氧化系统酶活性等指标。研究结果表明,BPA对青岛大扁藻的96h-EC50为9.32 mg·L-1,属高毒类污染物。青岛大扁藻经过BPA暴露处理后,细胞密度下降,细胞色素含量降低,并且呈现明显的剂量-效应关系;细胞抗氧化系统中超氧化物歧化酶(SOD)、抗坏血酸过氧化物酶(APX)、单脱氢抗坏血酸还原酶(MDHAR)、脱氢抗坏血酸还原酶(DHAR)、谷胱甘肽还原酶(GR)活性均受到干扰。  相似文献

In order to study the relation of the mobility and distribution of metals between Tamarix parts and their crystal salts – soil system, different experiments were conducted with plant-rich and plant-free soils at the banks of river Nile, Aswan, Egypt. For these purposes, Tamarix top and subsoil samples near and far from the plant were collected from six different locations at the banks of river Nile. Elemental analysis of Fe, Mn, Ca, Mg, Cr, Cu, Ni, Zn, Cd and Pb in soil, and in different parts of Tamarix and their crystal salt samples was carried out by atomic absorption spectrometry, whereas Na and K were measured by atomic emission spectrophotometry. The bioavailability of metals in the soil samples was evaluated by determining their contents using sequential extraction single-step, providing higher metal concentrations in occluded Fe/Mn oxide fraction. The ratio of heavy metal concentrations in topsoil to that in subsoil enhancement (RTE) ranged from 0.67 to 4.4 and 0.22 to 4.61 for soil Tamarix and soil free from plant, respectively for all measured total element concentrations. Data indicate that the mobility expressed as transfer factor (TF) was obtained as Fe?>?Mg?>?Cr?>?Mn?>?Ca?>?K?>?Na, for Tamarix leaves. The major part of accumulated Na in Tamarix is retained in the plant leaves, while roots accumulated high concentration of Co.  相似文献

Okadaic acid (OA) is a dinoflagellate toxin which accumulates in shellfish producing diarrhetic shellfish poisoning (DSP) in humans. It was found that OA is a highly selective inhibitor of protein phosphatase types 1 (PP1) and 2A (PP2A) which produces a marked increase in phosphorylation of several proteins, including p38 mitogen-activated protein (MAP) kinase. The cytotoxicity attributed to OA and the effects on p38 MAP kinase and calcium current were examined in the oyster Crassostrea gigas in this study. Data showed that p38 MAP kinase is strongly expressed in oyster heart and that OA bioaccumulated in cultured heart cells. Hence the effects of OA was tested in vitro and in vivo on oysters. OA was found to (i) exert a positive chronotropic effect on cultured atrial cardiomyocytes which is related to an increase in calcium current via PKC as shown by patch clamp measurements, (ii) produce an activation/phosphorylation of MAP kinase as shown by Westernblot while the non-phosphorylated p38 remained constant during treatment, (iii) did not induce a pro-apoptotic effect. Data suggest that OA may also stimulate the anti-apoptotic pathway by phosphatase inhibition.  相似文献

Given the wide industrial use of chromium (Cr) and its environmental contamination, chromium represents a risk to humans exposed to the metal. Considering that Cr(VI) is a potent oxidizing agent that increases intracellular oxidation and DNA damage, it would be worth considering the pretreatment of cells with antioxidants as a means of preventing Cr(VI)-induced toxicity. The objective of this study was to pretreat yeast cells with the water-soluble vitamin E analogue Trolox in an effort to increase cell tolerance against reactive chromium and reactive oxygen species formed during Cr(VI) reduction. Results revealed a decrease in Cr(VI)-induced cytotoxicity and mitotic gene conversions in Trolox-pretreated cells. The protective effect of Trolox in Cr(VI) induced genotoxicity was confirmed also with the prokaryotic Salmonella typhimurium SOS/umu test. Pretreatment of cells with Trolox (1) increased total Cr bioaccumulation, (2) decreased Cr(VI)-induced intracellular oxidation, (3) decreased Cr(V) persistence and (4) increased OH? formation in yeast extracts. These findings might be useful in directing future investigations concerning the use of Trolox as a human antioxidant supplement, and in clinical applications related to Cr-induced genotoxicity in occupational and environmental situations where chromium is a problem.  相似文献
