Abstract

The biotransformation of the nonylphenol isomer [ring-U-14C]-4-(3′,5′-dimethyl-3′-heptyl)-phenol (4-353-NP, consisting of two diastereomers) was studied in soybean and Agrostemma githago cell suspension cultures. With the A. githago cells, a batch two-liquid-phase system (medium/n-hexadecane 200:1, v/v) was used, in order to produce higher concentrations and amounts of 4-353-NP metabolites for their identification; 4-353-NP was applied via the n-hexadecane phase. Initial concentrations of [14C]-4-353-NP were 1 mg L?1 (soybean), and 5 and 10 mg L?1 (A. githago). After 2 (soybean) and 7 days (A. githago) of incubation, the applied 4-353-NP was transformed almost completely by both plant species to four types of products: glycosides of parent 4-353-NP, glycosides of primary 4-353-NP metabolites, nonextractable residues and unknown, possibly polymeric materials detected in the media. The latter two products emerged especially in soybean cultures. Portions of primary metabolites amounted to 19–22% (soybean) and 21–42% of applied 14C (A. githago). After liberation from their glycosides, the primary 4-353-NP metabolites formed by A. githago were isolated by HPLC and examined by GC-EIMS as trimethylsilyl derivatives. In the chromatograms, eight peaks were detected which due to their mass spectra, could be traced back to 4-353-NP. Seven of the compounds were side-chain monohydroxylated 4-353-NP metabolites, while the remaining was a (side-chain) carboxylic acid derivative. Unequivocal identification of the sites of hydroxylation/oxidation of all transformation products was not possible. The main primary metabolites produced by A. githago were supposed to be four diastereomers of 6′-hydroxy-4-353-NP (about 80% of all products identified). It was concluded that plants contribute to the environmental degradation of the xenoestrogen nonylphenol; the toxicological properties of side-chain hydroxylated nonylphenols remain to be examined.  相似文献

为了更好地从基因角度研究亚硝态氮对贝类的生态毒性,需要筛选一个合适的内参基因作为参考,对其毒性相关基因进行定量表达分析。本研究以菲律宾蛤仔(Ruditapes philippinarum)作为研究对象,以亚硝态氮胁迫的鳃组织cDNA作为模板,利用geNorm软件和NormFinder软件对6个候选内参基因进行表达稳定性分析,并对2个结果做了综合赋权分析。结果显示,geNorm分析的基因表达稳定性结果为ActinCyPAEF1αUbi18STubu,NormFinder软件分析的结果为ActinUbiCyPAEF1α18STubu,最后经加权赋值法综合分析获得6个候选基因的表达稳定性为ActinCyPAUbiEF1α18STubu,即Actin为表达最稳定的基因。因此,Actin可以作为衡量亚硝态氮胁迫后菲律宾蛤仔鳃组织中基因表达的内参基因。  相似文献

In sequentially polyandrous bird species, where females mate with different males in succession during a single breeding season, sperm stored by females can occasionally lead to extrapair fertilisations (EPFs) in clutches cared for by the second and subsequent males. Thus, we predicted that in red phalaropes (Phalaropus fulicarius) – a sex-role-reversed, sequentially polyandrous, arctic breeding shorebird – EPFs would occur more frequently in clutches laid later in the breeding season. We used multilocus DNA profiling to examine the frequency and timing of EPFs in a population of red phalaropes breeding in the Canadian high arctic. Using a technique to determine parentage without maternal DNA, we inferred that 6 of 70 chicks in 18 broods resulted from EPFs – one extrapair chick in each of 6 broods. These results were supported by a further analysis using microsatellite DNA. As predicted, broods containing EPFs hatched from clutches laid significantly later in the season than did broods containing no EPFs. The difference in median hatch dates between broods with and without EPFs was 9.0 days, or 38% of the entire egg-laying period in that season. For the whole breeding season, we estimated that 6.5% of chicks were sired by extrapair males, which is similar to extrapair paternity estimates for other sex-role-reversed birds, but relatively low compared to the majority of socially monogamous species studied so far. Paternity and paternal care in the polygynandrous Smith's longspur   总被引:4,自引:0,他引:4  
In species where females copulate with more than one male during a single breeding attempt, males risk investing in offspring that are not their own. In the polygynandrous Smith's longspur (Calcarius pictus), females copulate sequentially with one to three males for each clutch of eggs and most of these males later assist in feeding the young. Using multilocus DNA profiling, we determined that there was mixed paternity in >75% of broods (n=31) but that few offspring (<1% of 114 nestlings) were sired by males outside the polygynandrous group. Male feeding rate increased significantly with the number of young sired, with males siring four nestlings feeding the brood at double the frequency of males siring only a single nestling. However, male Smith's longspurs appear to show a graded adjustment of paternal care in response to paternity only when other males are available to compensate for reduced care: feeding rate did not vary in relation to paternity when only one male provisioned young at the nest. There was no evidence that males could recognise their own offspring within a brood and feed them preferentially. The number of offspring sired by each male was significantly correlated with the number of days spent copulating with the attending female: on average, a male sired one offspring for every 2 days of copulatory access. If males use their access to females to estimate paternity (and thereby decide on their subsequent level of parental investment), a positive relationship is expected between the amount of female access and the subsequent feeding rate to the nestlings. Nonetheless, male feeding effort was only weakly correlated with female access and more study is needed to determine how males estimate their paternity in a brood. 海洋环境基因组学是功能基因组学的一个分支学科,它将基因组学的最新技术引入到海洋生态环境的研究当中,得到了许多有价值的研究成果,显示出巨大的发展潜力。文章总结了海洋环境基因组学所开展的工作和已取得的成果,指出目前海洋环境基因组学的工作主要集中在三个领域:海洋生物基因资源文库的建立与完善;海洋生物对环境胁迫的应激反应及生理机制的研究;探索不同生物种群的基因表达谱是否与生物地理分布相关联。并着重讨论了利用基因组学技术研究环境胁迫对海洋生物物种的分布、生物物种间的相互作用以及水生物疾病的影响。如海洋环境基因组学通过测定基因表达谱的变化,研究海洋生物对环境温度的生理响应,绘制出生物地理图谱;通过测定基因转录水平的变化,更深入地了解温度变化所造成的生理支出,从而预测热气候漂移可能给海洋生态环境所造成的影响。在水生物疾病的研究方面,海洋环境基因组学用DNA微阵列技术分析染病生物的基因表达谱,筛选出发生变化的基因表达,为理解病毒感染机制提供了大量信息。还可以深入研究环境胁迫因子与微生物种群群落的组成关系,并由此找到通过维持水体的微生态平衡来消除某些病害发生的环境条件。  相似文献

Discrete-depth sampling of inorganic groundwater chemistry is essential for a variety of site characterization activities. Although the mobility and rapid sampling capabilities of direct-push techniques have led to their widespread use for evaluating the distribution of organic contaminants, complementary methods for the characterization of spatial variations in geochemical conditions have not been developed. In this study, a direct-push-based approach for high-resolution inorganic chemical profiling was developed at a site where sharp chemical contrasts and iron-reducing conditions had previously been observed. Existing multilevel samplers (MLSs) that span a fining-upward alluvial sequence were used for comparison with the direct-push profiling. Chemical profiles obtained with a conventional direct-push exposed-screen sampler differed from those obtained with an adjacent MLS because of sampler reactivity and mixing with water from previous sampling levels. The sampler was modified by replacing steel sampling components with stainless-steel and heat-treated parts, and adding an adapter that prevents mixing. Profiles obtained with the modified approach were in excellent agreement with those obtained from an adjacent MLS for all constituents and parameters monitored (Cl, NO(3), Fe, Mn, DO, ORP, specific conductance and pH). Interpretations of site redox conditions based on field-measured parameters were supported by laboratory analysis of dissolved Fe. The discrete-depth capability of this approach allows inorganic chemical variations to be described at a level of detail that has rarely been possible. When combined with the mobility afforded by direct-push rigs and on-site methods of chemical analysis, the new approach is well suited for a variety of interactive site-characterization endeavors.  相似文献

Poaching can have devastating impacts on animal and plant numbers, and in many countries has reached crisis levels, with illegal hunters employing increasingly sophisticated techniques. We used data from an 8‐year study in Savé Valley Conservancy, Zimbabwe, to show how geographic profiling—a mathematical technique originally developed in criminology and recently applied to animal foraging and epidemiology—can be adapted for use in investigations of wildlife crime. The data set contained information on over 10,000 incidents of illegal hunting and the deaths of 6,454 wild animals. We used a subset of data for which the illegal hunters’ identities were known. Our model identified the illegal hunters’ home villages based on the spatial locations of the hunting incidences (e.g., snares). Identification of the villages was improved by manipulating the probability surface inside the conservancy to reflect the fact that although the illegal hunters mostly live outside the conservancy, the majority of hunting occurs inside the conservancy (in criminology terms, commuter crime). These results combined with rigorous simulations showed for the first time how geographic profiling can be combined with GIS data and applied to situations with more complex spatial patterns, for example, where landscape heterogeneity means some parts of the study area are less likely to be used (e.g., aquatic areas for terrestrial animals) or where landscape permeability differs (e.g., forest bats tend not to fly over open areas). More broadly, these results show how geographic profiling can be used to target antipoaching interventions more effectively and more efficiently and to develop management strategies and conservation plans in a range of conservation scenarios.  相似文献

Forty‐five flood control reservoirs, authorized in the Watershed Protection and Flood Prevention Act 1954, were installed by United States Department of Agriculture (USDA) between 1969 and 1982 in the Little Washita River Experimental Watershed (LWREW), located in central Oklahoma. Over time, these reservoirs have lost sediment and flood storage capacity due to sedimentation, with rates dependent on upstream land use and climate variability. In this study, sedimentation rates for 12 reservoirs representing three major land use categories within LWREW were measured based on bathymetric surveys that used acoustic profiling system. Physiographic and climate attributes of drainage area of surveyed reservoirs were extracted from publicly available data sources including topographic maps, digital elevation models, USDA Natural Resource Conservation Service soils, and weather station databases. Correlation, principal component analysis, and stepwise regression were utilized to analyze the relationship between normalized reservoir sedimentation rates (ReSRa) and the drainage area characteristics to determine the major variables controlling sedimentation within the LWREW. Percent of drainage area with extreme slopes, saturated hydraulic conductivity, and maximum daily rainfall event recorded in spring explained most of the variability in ReSRa. It was also found that percent reduction in reservoir surface area can be used as a surrogate for estimating ReSRa. The implications of the results are discussed.  相似文献

The biochemical consequences induced by xenobiotic stress are featured in dose-response and time-resolved landscapes. Understanding the dynamic process of cellular adaptations is crucial in conducting the risk assessment for chemical exposure. As one of the most phenotype-related omics, metabolome in response to environmental stress can vary from seconds to days. Up to now, very few dynamic metabolomics studies have been conducted to provide time-dependent mechanistic interpretations in understanding xenobiotics-induced cellular adaptations. This study aims to explore the time-resolved metabolite dysregulation manner and dynamically perturbed biological functions in MCF-7 cells exposed to bisphenol A (BPA), a well-known endocrine-disrupting chemical. By sampling at 11 time points from several minutes to hours, thirty seven significantly dysregulated metabolites were identified, ranging from amino acids, fatty acids, carboxylic acids and nucleoside phosphate compounds. The metabolites in different pathways basically showed distinct time-resolved changing patterns, while those within the common class or same pathways showed similar and synchronized dysregulation behaviors. The pathway enrichment analysis suggested that purine metabolism, pyrimidine metabolism, aminoacyl-tRNA biosynthesis as well as glutamine/glutamate (GABA) metabolism pathways were heavily disturbed. As exposure event continued, MCF-7 cells went through multiple sequential metabolic adaptations from cell proliferation to energy metabolism, which indicated an enhancing cellular requirement for elevated energy homeostasis, oxidative stress response and ER-α mediated cell growth. We further focused on the time-dependent metabolite dysregulation behavior in purine and pyrimidine metabolism, and identified the impaired glycolysis and oxidative phosphorylation by redox imbalance. Lastly, we established a restricted cubic spline-based model to fit and predict metabolite’s full range dysregulation cartography, with metabolite’ sensitivity comparisons retrieved and novel biomarkers suggested. Overall, the results indicated that 8 h BPA exposure leaded to global dynamic metabolome adaptions including amino acid, nucleoside and sugar metabolism disorders, and the dysregulated metabolites with interfered pathways at different stages are of significant temporal distinctions.  相似文献
