Knowledge about the prevalence and diversity of antibiotic resistance genes in soil bacteria communities is required to evaluate the possibility and ecological consequences of the transfer of these genes carried by genetically modified (GM) plants to soil bacteria. The neomycin phosphotransferase gene (nptII) conferring resistance to kanamycin and neomycin is one of the antibiotic resistance genes commonly present in GM plants. In this study, we investigated kanamycin-resistant (Km(R)) and neomycin-resistant (Nm(R)) soil bacterial populations in a 3-year field trial using a commercial GM corn (Zea mays L.) carrying the nptII gene and its near isogenic line. The results showed that a portion (2.3 - 15.6 %) of cultivable soil bacteria was naturally resistant to kanamycin or neomycin. However, no significant difference in the population level of Km(R) or Nm(R) soil bacteria was observed between the GM and non-GM corn fields. The nptII gene was not detected in any of the total 3000 Km(R) or Nm(R) isolates screened by PCR. Further, total soil bacterial cells were collected through Nycodenz gradient centrifugation and bacterial community DNA was subjected to PCR. Detection limit was about 500 cells per gram of fresh soil. Our study suggests that the nptII gene was relatively rare in the soil bacterial populations and there was no evidence of gene transfer from a GM corn plant to soil bacteria based on the data from total soil bacterial communities. 相似文献
Chemical analyses were performed in nine fish species that are popular on the Polish market. These included Baltic fish (cod, herring, salmon), fish farmed in Poland (carp, trout), marine fish imported from China (Alaska pollock, sole), and farmed fish imported from Vietnam and China (sutchi catfish, tilapia). The nutritional composition (amino acid, micro- and macronutrients, fat-soluble vitamins - A1, D3, E) and certain contaminants (organochlorine pesticides, OCPs; indicator polychlorinated biphenyl, PCB6; polychlorinated dibenzo-paradioxins and polychlorinated dibenzofurans, PCDD/Fs; dioxin-like polychlorinated biphenyls, dl-PCBs; organotin compounds, OCTs; dyes, malachite green and crystal violet; veterinary drug residues, nitrofurans and chloramphenicol; toxic metals, Cd, Pb, Hg) in the muscle tissues of fish were determined. It was confirmed that the fish species analyzed were excellent sources of amino acids, and were rich in phosphorous and selenium. Baltic Sea fish (salmon, herring), fish farmed in Poland (carp and trout), and tilapia were also rich in vitamin D3.Traces of OCP, PCB6, OCT, dyes, veterinary drug residues, and heavy metals were detected in concentrations which do not pose a threat to consumers at the current rate of fish consumption in Poland. However, the problem might arise from the content of PCDD/Fs and dl-PCBs in fatty Baltic fish. The fish species analyzed, differed in their nutritional values and degrees of contamination. We suggest that for optimum health and safety, it is advisable that consumers include a variety of different fish species in their diets. 相似文献
This study has applied the concept of the hybrid PAC-UF process in the treatment of the final effluent of the palm oil industry for reuse as feedwater for low-pressure boilers. In a bench-scale set-up, a low-cost empty fruit bunch-based powdered activated carbon (PAC) was employed for upstream adsorption of biotreated palm oil mill effluent (BPOME) with the process conditions: 60 g/L dose of PAC, 68 min of mixing time and 200 rpm of mixing speed, to reduce the feedwater strength, alleviate probable fouling of the membranes and thus improve the process flux (productivity). Three polyethersulfone ultrafiltration membranes of molecular weight cut-off (MWCO) of 1, 5 and 10 kDa were investigated in a cross-flow filtration mode, and under constant transmembrane pressures of 40, 80, and 120 kPa. The permeate qualities of the hybrid processes were evaluated, and it was found that the integrated process with the 1 kDa MWCO UF membrane yielded the best water quality that falls within the US EPA reuse standard for boiler-feed and cooling water. It was also observed that the permeate quality is fit for extended reuse as process water in the cement, petroleum and coal industries. In addition, the hybrid system’s operation consumed 37.13 Wh m−3 of energy at the highest applied pressure of 120 kPa, which is far lesser than the typical energy requirement range (0.8–1.0 kWh m−3) for such wastewater reclamation.
The persistence of pesticides in soils has both economic and environmental significance and is often used as a key parameter in pesticide risk assessment. Persistence of acetochlor [2'-ethyl-6'-methyl-N-(ethoxymethyl)-2-chloroacetylanilide] in two New Zealand field soils was measured over two years and the data were used to identify models that adequately describe acetochlor persistence in the field. Acetochlor was sprayed onto six fallow plots (3 x 9 m each) at each site at the recommended rate (2.5 kg a.i. ha(-1)) and at twice that rate. Acetochlor concentrations were measured in soil cores. Simple first-order kinetics (Model 1) adequately described acetochlor persistence in Hamilton clay loam soil (Humic Hapludull, Illuvial Spadic) at the high application rate, but overestimated it at the low application rate. A quadratic model (Model 2), a first-order double-exponential model (Model 3), a first-order biphasic model (Model 4), or a two-compartment model (Model 5) better described acetochlor persistence at the low application rate. The time for 50% (DT50) and 90% (DT90) of initial acetochlor loss was approximately 9 and 56 d, and 18 and 63 d at low and high application rates, respectively. The more complex Models 2 through 5 also better described the biphasic dissipation of acetochlor in Horotiu sandy loam soil (Typic Orthic Allophanic) than Model 1, with Model 1 significantly underestimating acetochlor concentrations on the day of application at both application rates. The DT50 and DT90 values were 5 and 29 d and 7 and 31 d at low and high application rates, respectively. Overall, application rate significantly affected the DT50 and DT90 values in the Hamilton soil, but not in the Horotiu soil. Faster acetochlor loss in the Horotiu soil possibly resulted from the higher soil organic carbon content that retained more acetochlor near the soil surface where higher temperature and photolysis accelerated the loss. 相似文献
Compost stability and maturity are important parameters of compost quality. To date, nearly all compost characterization has been performed using samples freshly collected because sample storage can affect compost stability and maturity evaluation. However, sample preservation is sometimes necessary, especially for scientific research purposes. There is little information available on the effects of sample storage on compost stability and maturity. Samples of biosolids compost with different levels of stability and maturity were collected from four compost facilities in Florida (referred to as Register, Winslow, Sunset, and Meadow). Comparisons of CO2 evolution, seed germination rate, and water-soluble organic carbon (WSOC) were made between fresh samples with short storage at 4 degrees C for less than 1 wk and air-dried or frozen compost samples stored for 1 yr. The effects of storage (air-dry or frozen) on the measured parameters depended on compost stability and maturity and on the compost material source. Frozen storage reduced the peak CO2 evolution of Register samples by 12 to 29%, while accumulated CO2 evolution was reduced by 43 to 64% and 110 to 277% with air-dry and frozen storage, respectively. The storage effect on CO2 evolution with more stable compost was inconsistent. Storage did not affect compost phytotoxicity, except for samples from the Sunset facility. Air-drying reduced the WSOC by up to 35%, and freezing increased it by up to 34%, while both storage methods had no significant effect on samples of low WSOC. Despite all these variations, WSOC had a significant and consistent relation to CO2 evolution and seed germination rates with R2 of 0.78 and 0.57, respectively, regardless of storage methods. 相似文献
