Hepatitis A virus (HAV) is currently recognized as an important human food borne pathogen, and it is one of the most resistant enteric RNA viruses, is highly infectious, and may lead to widespread outbreaks. The aim of this study was to optimize the methods to detect HAV from artificially contaminated food. To this end, strawberry and lettuce were experimentally contaminated with HAV suspension containing 6 × 106 copies/ml. After contamination, HAV persistence and washing procedure were evaluated at 0, 1, 3, 7, and 9 days of storage. Five elution buffers (PBS (pH 7.4)/0.1% Tween80; 50 mM glycine/3% (wt/vol) beef extract (pH 9.5); PBS (pH 7, 4); 25 mM glycine/0.1 Tween80; and 1 M sodium bicarbonate) were used to elute the virus, and qualitative and quantitative PCR were used for HAV detection. HAV was detected by qualitative and quantitative PCR using any of the five elution buffers, but PBS was the most effective. Even after washing, HAV was detected up to 9 days after contamination by quantitative PCR. Quantitative PCR was more sensitive than qualitative PCR since samples containing viral load lower than 1.4 × 103 copies/ml could not be detected by qualitative PCR. Quantitative PCR can be used for rapid detection of food borne viruses and will help in the monitoring and control of food borne disease.  相似文献

Different sources were consulted to obtain information on the occurrence of viruses in bivalve molluscs on the European market. Twenty-six peer-reviewed articles were identified reporting on the molecular detection of viral RNA in 4,260 samples in total. The data obtained will be presented geographically on virus types detected, the origin and treatment of the shellfish, and the detection technique applied. The data demonstrate that viral RNA can be detected in shellfish from polluted areas, in depurated shellfish as well as those for human consumption. The European Rapid Alert System for Food and Feed (RASFF) database was consulted as another source. Twenty-eight notifications were identified on the presence of hepatitis A virus or norovirus in shellfish on the European market. The most recent report of the European laboratory network was referred to, to gain insight into the laboratory capability at present for the analyses of shellfish on the presence of viruses. Approximately 67% of 27 participating laboratories obtained intended results for all samples, consisting of lenticules loaded with 103 copies norovirus (genogroup I (GGI) and/or genogroup II (GGII)) and/or 1 × 105–8 × 104 copies of hepatitis A virus. From 1993, there has been a continuous development of molecular detection techniques and tools have been described to ensure quality assurance. End product testing will, however, not be achievable. As depuration has been shown not to be effective for the complete elimination of viruses, shellfish should not be in contact with faecal contaminated water in order to minimise the risk of shellfish-transmittable viral diseases.  相似文献

Climate change presents a major threat to the prospects for sustained economic development in Africa. In spite of this, climate change concerns do not feature prominently in the implementation of national and regional development programmes. The present paper identifies the likely trade-offs and synergies that may emerge from an integrated ‘development-climate’ approach to policy making. Also, the paper presents the case for the formulation and evaluation of an integrated policy approach based on four principle criteria, including; long-term environmental effectiveness, equity considerations, cost-effectiveness and the institutional compatibility of the policy combinations. What is more? The paper suggests specific options for mainstreaming climate change adaptation and mitigation in various sectoral development agenda such as; agricultural intensification, poverty eradication, rural development, urban renewal, energy security of supply and trade. Given the wide divergence of socio-economic systems and the peculiar challenges faced by individual countries in the continent, further research is required on robust country-specific strategies for pursuing an integrated development-climate policy framework.  相似文献

Megacities in developing countries are rapidly transforming places. Under the impetus of global change processes and consequent transformations at the environmental, social, cultural, political and economical scales; factors causing disasters and losses are changing every day. These changes are also altering society’s ability to respond to hazard events. This paper examines the response of slum dwellers who are the most vulnerable and marginal section of urban population and often located in places with high hazard risk with less or no means to reduce the impact of flood events. Marginal population groups in megacities suffer the negative consequences of large scale global change processes and do not benefit from the risk mitigation strategies adopted by city authorities. The paper therefore argues that people living in informal settlements instead have to employ a combination of structural means and complex networks of assistance to recover from floods. Based on the results deduced from data collected with the help of household surveys in the slums of Mumbai, the study demonstrates the types of coping strategies used by slum dwellers and the changing characteristics of these mechanisms under the influence of global change processes in megacities. Furthermore, results show that capacity to respond is not equally distributed among slum dwellers due to underlying socio cultural divisions and emerging economic and political constraints. The paper concludes that to address existing discrepancies in urban societies and within slum settlements, flood mitigation strategies will have to be (1) more inclusive of marginal population (2) sensitive to the limitations and scope of old and new social structures and (3) incorporate innovative networks of support to deal with the consequences of global change.  相似文献

Climate change associated global warming, rise in carbon dioxide concentration and uncertainties in precipitation has profound implications on Indian agriculture. Maize (Zea mays L.), the third most important cereal crop in India, has a major role to play in country’s food security. Thus, it is important to analyze the consequence of climate change on maize productivity in major maize producing regions in India and elucidate potential adaptive strategy to minimize the adverse effects. Calibrated and validated InfoCrop-MAIZE model was used for analyzing the impacts of increase in temperature, carbon dioxide (CO2) and change in rainfall apart from HadCM3 A2a scenario for 2020, 2050 and 2080. The main insights from the analysis are threefold. First, maize yields in monsoon are projected to be adversely affected due to rise in atmospheric temperature; but increased rainfall can partly offset those loses. During winter, maize grain yield is projected to reduced with increase in temperature in two of the regions (Mid Indo-Gangetic Plains or MIGP, and Southern Plateau or SP), but in the Upper Indo-Gangetic Plain (UIGP), where relatively low temperatures prevail during winter, yield increased up to a 2.7°C rise in temperature. Variation in rainfall may not have a major impact on winter yields, as the crop is already well irrigated. Secondly, the spatio-temporal variations in projected changes in temperature and rainfall are likely to lead to differential impacts in the different regions. In particular, monsoon yield is reduced most in SP (up to 35%), winter yield is reduced most in MIGP (up to 55%), while UIGP yields are relatively unaffected. Third, developing new cultivars with growth pattern in changed climate scenarios similar to that of current varieties in present conditions could be an advantageous adaptation strategy for minimizing the vulnerability of maize production in India.  相似文献

This paper describes how relatively detailed knowledge about probabilities of natural hazards can be used to make decisions to develop areas and control the risk within hazard zones. The assessment serves two purposes. First, it shows how information can support decisions. Second, decision criteria put leads on what information is required. This is helpful to identify unavailable information. We show by an example from a land-slide prone area in Norway how a relatively reliable estimate of the probability of slides ends up in a rather uncertain estimate of the risk. Uncertainty about the risk represented by natural hazards imply great challenges to the development of adaptation policies to meet climate change, but they are required. We develop a simplified criterion for optimal adaptation, and estimate the added social value required to defend development in hazard prone areas instead of developing a risk-free alternative. The value is estimated between 0 and 0.40 Euros per Euro invested in the case area, depending on type of slide, category of asset and other costs that occur in the wake of slides.  相似文献

A novel biphenyl-degrading bacterial strain LA-4 was isolated from activated sludge. It was identified as Dyella ginsengisoli according to phylogenetic similarity of 16S rRNA gene sequence. This isolate could utilize biphenyl as sole source of carbon and energy, which degraded over 95 mg/L biphenyl within 36 h. The major metabolites formed from biphenyl, such as 2-hydroxy-6-oxo-6-phenylhexa-2,4-dienoic acid (HOPDA) and benzoic acid, were identified by LC-MS. The crude cell extract of strain LA-4 exhibited the activity of 2,3-dihydroxybiphenyl 1,2-dioxygenase (2,3-DHBD) and the kinetic parameters were Km= 26.48 μmol/L and Vmax= 8.12 μmol/mg protein. A conserved region of the biphenyl dioxygenase gene bphA1 of strain LA-4 was amplified by PCR and confirmed by DNA sequencing.  相似文献

Apart from the liver disruption, embryotoxicity and genotoxicity, microcystin (MC)-LR also could cause neurotoxicity. Nematode Caenorhabditis elegans was explored as a model to study the neurotoxicity. In the present study, we provided evidence to indicate the neurotoxicity on chemotaxis to NaCl and diacetyl, and thermotaxis from MC-LR exposure to C. elegans. As a result, higher concentrations of MC-LR caused significantly severe defects of chemotaxis to NaC1 and diacetyl, and thermotaxis. The neurotoxicity on chemotaxis to NaCl and diacetyl, and thennotaxis from MC-LR exposure might be largely mediated by the damage on the corresponding sensory neurons (ASE, AWA, and AFD) and interneuron AIY. The expression levels of che-1 and odr-7 were significantly decreased (P<0.01) in animals exposed to MC-LR at concentrations lower than 10 μg/L, whereas the expression levels of ttx-1 and ttx-3 could be significantly (P<0.01) lowered in animals even exposed to 1 μg/L of MC-LR. Moreover, both the chemotaxis to NaCl and diacetyl and the thermotaxis were more significantly reduced m MC-LR exposed mutants of che-1(p674), odr-7(ky4), ttx-1(p767), and ttx-3(ks5) than those in exposed wild-type N2 animals at the same concentrations.  相似文献

Chemotaxis to water-soluble attractants is mainly controlled by ASE sensory neuron whose specification is regulated by che-1 in Caenorhabditis elegans. Our data suggested that exposure to high concentrations of metals, such as Pb, Cu, Ag, and Cr, would result in severe defects of chemotaxis to water-soluble attractants of NaCl, cAMP, and biotin. Moreover, the morphology of ASE neuron structures as observed by relative fluorescent intensities and relative size of fluorescent puncta of cell bodies, relative lengths of sensory endings in ASE neurons, and the expression patterns of che-1 were obviously altered in metal exposed animals when they meanwhile exhibited obvious chemotaxis defects to water-soluble attractants. In addition, the dendrite morphology could be noticeably changed in animals exposed to 150 mol/L of Pb, Cu, and Ag. Furthermore, we observed significant decreases of chemotaxis to water-soluble attractants in Pb exposed che-1 mutant at concentrations more than 2.5 mol/L, and in Cu, Ag, and Cr exposed che-1 mutant at concentrations more than 50 mol/L. Therefore, impairment of the ASE neuron structures and functions may largely contribute to the appearance of chemotaxis defects to water-soluble attractants in metal exposed nematodes.  相似文献
