Objective
Genomics Quality Assessment has provided external quality assessments (EQAs) for preimplantation genetic testing (PGT) for 12 years for eight monogenic diseases to identify sub-optimal PGT strategies, testing and reporting of results, which can be shared with the genomics community to aid optimised standards of PGT services for couples.Method
The EQAs were provided in two stages to mimic end-to-end protocols. Stage 1 involved DNA feasibility testing of a couple undergoing PGT and affected proband. Participants were required to report genotyping results and outline their embryo testing strategy. Lymphoblasts were distributed for mock embryo testing for stage 2. Submitted clinical reports and haplotyping results were assessed against peer-ratified criteria. Performance was monitored to identify poor performance.Results
The most common testing methodology was short tandem repeat linkage analysis (59%); however, the adoption of single nucleotide polymorphism-based platforms was observed and a move from blastomere to trophectoderm testing. There was a variation in testing strategies, assigning marker informativity and understanding test limitations, some clinically unsafe. Critical errors were reported for genotyping and interpretation.Conclusion

In order to estimate the introgressive hybridisation propensity (IHP) between genetically modified (GM) oilseed rape (Brassica napus) and certain of its cross-compatible wild/weedy relatives at the landscape level, a conceptual approach was developed. A gene flow index was established enclosing the successive steps to successfully achieve introgressive hybridisation: wild/weedy relatives and oilseed rape should co-occur, have overlapping flowering periods, be compatible, produce viable and fertile progeny, and the transgenes should persist in natural/weedy populations. Each step was described and scored, resulting in an IHP value for each cross-compatible oilseed rape wild/weedy relative. The gene flow index revealed that Brassica rapa has the highest introgressive hybridisation propensity (IHP value = 11.5), followed by Hirschfeldia incana and Raphanus raphanistrum (IHP = 6.7), Brassica juncea (IHP = 5.1), Diplotaxis tenuifolia and Sinapis arvensis (IHP = 4.5) in Flanders. Based on the IHP values, monitoring priorities can be defined within the pool of cross-compatible wild/weedy oilseed rape relatives. Moreover, the developed approach enables to select areas where case-specific monitoring of GM oilseed rape could be done in order to detect potential adverse effects on cross-compatible wild/weedy relatives resulting from vertical gene flow. The implementation of the proposed oilseed rape-wild relative gene flow index revealed that the survey design of existing botanical survey networks does not suit general surveillance needs of GM crops in Belgium. This paper examines the nature of the economic costs and risks involved in brownfield versus greenfield redevelopment in the Greater Toronto Area (Ontario, Canada) from a private sector perspective, and assesses the potential effectiveness of different policies and programmes designed to attenuate associated costs and risks. Through interviews, case-studies and an analysis of hypothetical development scenarios, it has been found that the perception that brownfield redevelopment is less cost-effective and entails greater risks than greenfield development, on the part of the private sector, is true for industrial projects in the province, but not for residential ones, which were found to be feasible, given the assumptions of the present study. Furthermore, the study has found that the attractiveness of residential brownfield projects can increase considerably with minor policy changes, but that promoting industrial redevelopment will require a more vigorous approach that employs a variety of environmental policy and economic development measures. 相似文献

This study presents the laboratory scale results of an extra step in Poly (ethylene terephthalate) – PET mechanical recycling (grinding, washing, drying and reprocessing): a chemical washing after the conventional one. Cooking oil PET bottle flakes were washed in water and then subjected to a reaction with an aqueous solution of sodium hydroxide 5 M at 90 °C for 10 min (chemical washing). After rinsing and drying, the flakes were characterized by thermogravimetry, gas chromatography and elemental analysis tests. The results indicated that the chemically washed material had higher purity than PET washed only with water: 99.3% and 96.7%, respectively, which undoubtedly implies properties, applications and prices closer to those of virgin resin. The production of purified terephthalic acid (TPA) from the chemical washing residue was optimized and reached a purity of 99.6%. Despite the results, the use of chemically washed PET and of TPA obtained is not recommended for direct contact with food, since they still contain some impurities. 相似文献
