Nitrate and ammonium uptake rates were measured for three year-classes of the perennial macrophyte Laminaria groenlandica Rosenvinge, collected from nitrogen-depleted waters in Barkley Sound, British Columbia, Canada, in summer 1981. A time course of uptake rate revealed that ammonium uptake was high during the first hour and then decreased for all three year-classes; the opposite pattern was exhibited for the time course of nitrate uptake rate. Nitrate uptake rate increased linearly with nitrate concentration up to the highest level tested (60 M). The nitrate uptake rate of first-year plants was three times higher than second- and third-year plants; ammonium uptake rates showed similar patterns to those for nitrate. The interaction between nitrate and ammonium was examined for first-year plants. Nitrate and ammonium were taken up simultaneously and uptake rates were identical and equal to uptake rates when only nitrate or ammonium was present in the medium. Therefore, first-year plants are able to take up twice as much inorganic nitrogen per unit time when both nitrate and ammonium are present. First-year plants showed significant diel periodicity in ammonium uptake rates, whereas second- and third-year plants showed no periodicity in nitrate or ammonium uptake rates.  相似文献

Using the marine harpacticoid copepod Tigriopus californicus, the effects of phytoplankton feeding deterrents and toxins were differentiated and measured. Eight compounds were tested for feeding deterrence and toxicity responses: four apo-fucoxanthinoids (apo-10′-fucoxanthinal, apo-12′-fucoxanthinal, apo-12-fucoxanthinal, and apo-13′-fucoxanthinone) and four well-known phycotoxins (domoic acid, okadaic acid, microcystin-LR, and a mixture of PSP-1 toxins). Since several of these compounds exhibited both feeding deterrence and toxicity, a model was developed to deconvolute the observed toxicity response from the observed feeding deterrence response, and to classify these compounds based on the degree of toxicity and/or feeding deterrence they exhibited towards T. californicus. Microcystin-LR, the PSP-1 toxins, and the four apo-fucoxanthinoids behaved only as feeding deterrents at low concentrations. Okadaic acid exhibited both toxicity and feeding deterrence at low concentrations, with the threshold concentration for feeding deterrence at a lower level than the threshold concentration for toxicity. Domoic acid acted only as a toxin at low concentrations, with all decreases in feeding resulting from the death of the copepod. Received: 4 November 1996 / Accepted: 3 December 1996  相似文献

A natural groundwater system modified by pollutant phenols and agricultural nitrate has been modelled in the laboratory by a series of sacrificial microcosm experiments. Samples of aquifer sediment and groundwater from the margin of the phenol plume were used to inoculate anaerobic microcosms enriched in nitrate and pollutant phenols. Rapid degradation of phenol and p-cresol was observed over a 35-day period leading to the generation of inorganic carbon and a number of transient intermediates. O-cresol proved to be recalcitrant on the experimental time-scale. A mass balance calculation shows that, during degradation, carbon was conserved in the aqueous phase. Groundwater-sediment interactions were monitored using carbon stable isotope data. A mass balance for solution TIC indicates thatp-cresol degradation stimulated the dissolution of sedimentary carbonate phases due to the formation of carbonic acid. Compound-specific carbon isotope analysis (GC-IRMS) was used to search for 13C enrichment in residual p-cresol. A slight enrichment trend (epsilon = -2.5/1000) was tentatively identified. The potential of this fractionation effect for obtaining in situ degradation rates is discussed. Results from the microcosm experiments help to explain the observed distribution of nitrate and phenols within the polluted aquifer.  相似文献

Marine phytoplankton have been shown to use chemical feeding deterrents to reduce or inhibit zooplankton grazing. In order to screen phytoplankton species for feeding deterrent production and to isolate and identify feeding deterrent compounds, a new, rapid, and reliable laboratory bioassay was developed. This bioassay used the laboratory-reared harpacticoid copepod Tigriopus californicus and measured inhibition of feeding by measuring the fecal pellet production rate. The bioassay was capable of detecting deterrent compounds: (1) adsorbed onto ground fish food (a normally palatable food); (2) dissolved in a mixture of seawater and live Thalassiosira pseudonana cells (a species of diatom which had no feeding deterrent activity); and (3) present in live cell cultures. Method (2) was recommended for use in bioassay-guided fractionation (isolation of chemical compounds), as it was reliable, rapid, accurate, and easy to perform with large numbers of samples. The total bioassay time was < 48 h, and data collection required only a microscope. Methanolic cell extracts of several phytoplankton species were screened for feeding deterrent activity. Extracts from the diatom Phaeodactylum tricornutum and the dinoflagellate Gonyaulax grindleyi gave feeding deterrent responses, while extracts from the diatom Thalassiosira pseudonana gave no feeding deterrent responses. Live P. tricornutum cells deterred feeding at densities of 6x105 cells ml-1. This bioassay should provide a valuable tool in screening phytoplankton for feeding deterrent compounds and determining the chemical nature of these compounds.  相似文献

In spite of historical and current interest in Ciona intestinalis and its congeners, little is known about evolutionary relationships among the members of the genus Ciona. Here 744-bp sequences of the mitochondrial cytochrome c oxidase subunit I (COI) gene are used to examine phylogenetic relationships among three described species (C. intestinalis, C. roulei, C. savignyi) sampled from multiple coastal sites in the Northeast Pacific (CA, USA), Northwest Atlantic (from New Hampshire to Connecticut, USA), Northeast Atlantic (Sweden and The Netherlands), and Mediterranean (Banyuls-sur-Mer, France). The samples were collected in June–October 2005. The COI sequences of Northeast Pacific/Mediterranean (Type A) and Northwest Atlantic (Type B) C. intestinalis differ by ∼12% and C. roulei is nested within Type B C. intestinalis. Ciona savignyi differs from all other haplotypes by 13–16%. A previously undescribed but morphologically distinct Ciona sp. found at the Banyuls-sur-Mer site was >10% divergent from all other haplotypes. Although these data arise from a single gene study, they indicate that further elucidation of species relationships within the genus and of the species’ distributions will be needed if continuing invasions and potential reproductive isolation are to be investigated.  相似文献

The kinetic response of ammonium- or silicate-limited and ammonium- or silicatestarved populations of Chaetoceros debilis, Skeletonema costatum, and Thalassiosira gravida was determined by a single addition of the limiting nutrient to a steady-state culture and subsequent monitoring of the nutrient disappearance of the limiting and non-limiting nutrients at frequent time intervals. The kinetic response of nonlimited (nutrient) populations of these three species was also determined. Three distinct modes of the uptake of the limiting nutrient were observed for ammonium-or silicate-limited populations of these three species, surge uptake (V s ), internally (cellular) controlled uptake (V i ), and externally (ambient limiting nutrient concentration) controlled uptake (V e ). Non-limited populations did not exhibit the three distinct segments of uptake, V s , V i and V e . Estimates of the maximal uptake rate (V max) and the Michaelis constant (K s ) were obtained from nutrient-limited populations during the V e segment of the uptake curve. Pooled values of V e for the three ammonium-limited populations yielded V max and K s estimates of 0.16 h-1 and 0.5 g-at NH4–N l-1. Kinetic data derived from the V e segment of the uptake curve for silicate-limited populations yielded different values of V max and K s for each of the three species. In a number of parameters that were measured, T. gravida was clearly different from C. debilis and S. costatum and its recovery from nutrient starvation was the slowest. Recovery of all species from silicate limitation or starvation was slower than from ammonium limitation or starvation. Ammonium-starved populations maintained a maximal uptake rate at a substrate concentration an order of magnitude lower (0.1 g-at NH4–N l-1) than that observed for NH4-limited populations (1.0 g-at NH4–N l-1). Adaptation to the severity of the nutrient limitation occurred as changes in the magnitude of cellular characteristics, such as short-term uptake potential (V s ) and affinity for the substrate (K s ). Use of the internet as a trade platform has resulted in a shift in the illegal wildlife trade. Increased scrutiny of illegal wildlife trade has led to concerns that online trade of wildlife will move onto the dark web. To provide a baseline of illegal wildlife trade on the dark web, we downloaded and archived 9852 items (individual posts) from the dark web, then searched these based on a list of 121 keywords associated with illegal online wildlife trade, including 30 keywords associated with illegally traded elephant ivory on the surface web. Results were compared with items known to be illegally traded on the dark web, specifically cannabis, cocaine, and heroin, to compare the extent of the trade. Of these 121 keywords, 4 resulted in hits, of which only one was potentially linked to illegal wildlife trade. This sole case was the sale and discussion of Echinopsis pachanoi (San Pedro cactus), which has hallucinogenic properties. This negligible level of activity related to the illegal trade of wildlife on the dark web relative to the open and increasing trade on the surface web may indicate a lack of successful enforcement against illegal wildlife trade on the surface web.  相似文献

The results of a 21 month sampling program measuring tritium in tree transpirate with respect to local sources are reported. The aim was to assess the potential of tree transpirate to indicate the presence of sub-surface seepage plumes.Transpirate gathered from trees near low-level nuclear waste disposal trenches contained activity concentrations of 3H that were significantly higher (up to ∼700 Bq L−1) than local background levels (0-10 Bq L−1). The effects of the waste source declined rapidly with distance to be at background levels within 10s of metres. A research reactor 1.6 km south of the site contributed significant (p < 0.01) local fallout 3H but its influence did not reach as far as the disposal trenches.The elevated 3H levels in transpirate were, however, substantially lower than groundwater concentrations measured across the site (ranging from 0 to 91% with a median of 2%). Temporal patterns of tree transpirate 3H, together with local meteorological observations, indicate that soil water within the active root zones comprised a mixture of seepage and rainfall infiltration. The degree of mixing was variable given that the soil water activity concentrations were heterogeneous at a scale equivalent to the effective rooting volume of the trees. In addition, water taken up by roots was not well mixed within the trees. Based on correlation modelling, net rainfall less evaporation (a surrogate for infiltration) over a period of from 2 to 3 weeks prior to sampling seems to be the optimum predictor of transpirate 3H variability for any sampled tree at this site.The results demonstrate successful use of 3H in transpirate from trees to indicate the presence and general extent of sub-surface contamination at a low-level nuclear waste site.  相似文献

Traffic-generated air pollutant emissions can be classified into exhaust and non-exhaust emissions. Increased attention is focussing on non-exhaust emissions as exhaust emissions are progressively limited by regulations. To characterise metal-rich emission from abrasion processes, size-segregated analysis of atmospheric aerosol particles sampled with micro-orifice uniform deposit impactors (MOUDI) in March 2007 in London was performed. The samples were collected at a roadside and a background site and were analysed for Al, Ba, Cu, Fe, Sb, Ti, V, Zn, Ca2+, K+, Mg2+, Na+, and NH4+. Most components showed a clear roadside increment, which was evident as a higher mass concentration and a change in the size distribution. In particular, Fe, Cu, Ba, and Sb correlated highly, indicative of a common traffic-related source. Using complementary information on the fleet composition, vehicle number and average speed, the brake wear emission was calculated using the EMEP/CORINAIR emission database. The total PM10 and barium emission of the traffic was determined by ratio to NOx whose source strength was estimated from published emission factors. Barium was found to comprise 1.1% of brake wear (PM10) particles from the traffic fleet as a whole, allowing its use as a quantitative tracer of brake wear emissions at other traffic-influenced sites.  相似文献
