The symbiotic co-evolution of plants and microbes leads to difficulties in understanding which of the two components is responsible for a given environmental response. Plant-microbe studies greatly benefit from the ability to grow plants in axenic (sterile) culture. Several studies have used axenic plant culture systems, but experimental procedures are often poorly documented, the plant growth environment is not optimal, and axenic conditions are not rigorously verified. We developed a unique axenic system using inert components that promotes plant health and can be kept sterile for at least 70 d. Crested wheatgrass (Agropyron cristatum cv. CDII) plants were grown in sand within flow-through glass columns that were positively pressured with filtered air. Plant health was optimized by regulating temperature, light level, CO2 concentration, humidity, and nutrients. The design incorporates several novel aspects, such as pretreatment of the sand with Fe, graduated sand layers to optimize the air-water balance of the root zone, and modification of a laminar flow hood to serve as a plant growth chamber. Adaptations of several sterile techniques were necessary for maintenance of axenic conditions. Axenic conditions were verified by plating and staining leachates as well as a rhizoplane stain. This system was designed to study nutrient and water stress effects on root exudates, but is useful for assessing a broad range of plant-microbe-environment interactions. Based on total organic C analysis, 74% of exudates was recovered in the leachate, 6% was recovered in the bulk sand, and 17% was recovered in the rhizosphere sand. Carbon in the leachate after 70 d reached 255 microg d(-1). Fumaric, malic, malonic, oxalic, and succinic acids were measured as components of the root exudates. 相似文献

Any screening approach, including with cell-free DNA, will have an inferior detection rate compared with 100% diagnostic testing with chromosomal microarrays. Cell-free DNA-based screening, however, should not be seen as a threat to informed choice or maximising the benefits of diagnostic testing. Screening methods have become so much better that more women are now comfortable relying on such screening and do not need the certainty of a diagnostic test. This has not lead to a decline in detection of fetal chromosome abnormalities—in fact, we are now seeing historically high yields from prenatal screening. There are both economic and ethical consequences of offering universal diagnostic testing and abandoning the presumption of a normal infant in otherwise uncomplicated pregnancies. However, for some women, comprehensive information and diagnostic accuracy are important. Offering these women all options, with a careful and comprehensive explanation of the risks and benefits of each, results in outcomes that are best aligned with woman's preferences while at the same time requiring fewer diagnostic tests and lowering costs. Objective
There is increasing evidence supporting the clinical utility of next generation sequencing for identifying fetal genetic disorders. However, there are limited data on the demand for and accessibility of these tests, as well as payer coverage in the prenatal context. We sought to identify clinician perspectives on the utility of prenatal exome sequencing (ES) and on equitable access to genomic technologies for the care of pregnancies complicated by fetal structural anomalies.Method
We conducted two focus group discussions and six interviews with a total of 13 clinicians (11 genetic counselors; 2 Maternal Fetal Medicine/Geneticists) from U.S. academic centers and community clinics.Results
Participants strongly supported ES for prenatal diagnostic testing in pregnancies with fetal structural anomalies. Participants emphasized the value of prenatal ES as an opportunity for a continuum of care before, during, and after a pregnancy, not solely as informing decisions about abortions. Cost and coverage of the test was the main access barrier, and research was the main pathway to access ES in academic centers.Conclusion
Further integrating the perspectives of additional key stakeholders are important for understanding clinical utility, developing policies and practices to address access barriers, and assuring equitable provision of prenatal diagnostic testing. 相似文献75.

Throughfall and bulk precipitation samples were collected for two watersheds at Acadia National Park, Maine, from 3 May to
16 November 2000, to determine which landscape factors affected mercury (Hg) deposition. One of these watersheds, Cadillac
Brook, burned in 1947, providing a natural experimental design to study the effects of forest type on deposition to forested
watersheds. Sites that face southwest received the highest Hg deposition, which may be due to the interception of cross-continental
movement of contaminated air masses. Sites covered with softwood vegetation also received higher Hg deposition than other
vegetation types because of the higher scavenging efficiency of the canopy structure. Methyl mercury (MeHg) deposition was
not affected by these factors. Hg deposition, as bulk precipitation and throughfall was lower in Cadillac Brook watershed
(burned) than in Hadlock Brook watershed (unburned) because of vegetation type and watershed aspect. Hg and MeHg inputs were
weighted by season and vegetation type because these two factors had the most influence on deposition. Hg volatilization was
not determined. The total Hg deposition via throughfall and bulk precipitation was 9.4 μg/m2/year in Cadillac Brook watershed and 10.2 μg/m2/year in Hadlock Brook watershed. The Bear Brook Watershed in Maine (BBWM), USA, and the Fernow Experimental Forest in West Virginia, USA, represent unique, long-term, paired, whole watershed, experimental manipulations focusing on the effects of nitrogen (N) and sulfur (S) deposition on temperate forests. Both watersheds began whole-ecosystem additions of N and S as (NH4)2SO4 in the fall of 1989, and both are entering their third decade of chronic enrichment of the treated watersheds, while the reference watersheds offer unique opportunities to evaluate forest watershed responses to recovery. Differences between BBWM and Fernow in the history of atmospheric deposition, soil properties, and forest composition all contribute to different response trajectories in stream chemical exports over time. The four watersheds represent a spectrum of N enrichment and retention, ranging from ≈98% N retention in the reference watershed in Maine, to ≈20% N retention in the treated watershed in West Virginia. Despite these differences, there is evidence that mechanisms of response in base cation leaching and other processes are similar among all four watersheds. In both cases, the history to date of two decades of research and monitoring has provided new insights into ecosystem response not evident in more traditional short-term research. 相似文献

The ecosystem services concept is used to make explicit the diverse benefits ecosystems provide to people, with the goal of improving assessment and, ultimately, decision-making. Alongside material benefits such as natural resources (e.g., clean water, timber), this concept includes—through the ‘cultural’ category of ecosystem services—diverse non-material benefits that people obtain through interactions with ecosystems (e.g., spiritual inspiration, cultural identity, recreation). Despite the longstanding focus of ecosystem services research on measurement, most cultural ecosystem services have defined measurement and inclusion alongside other more ‘material’ services. This gap in measurement of cultural ecosystem services is a product of several perceived problems, some of which are not real problems and some of which can be mitigated or even solved without undue difficulty. Because of the fractured nature of the literature, these problems continue to plague the discussion of cultural services. In this paper we discuss several such problems, which although they have been addressed singly, have not been brought together in a single discussion. Linking community knowledge with modern techniques to record and analyse risk related data is one way of engaging and mobilising community capacity. This paper discusses the use of the Geographic Information System (GIS) at the local level and the need for integrating modern technology and indigenous knowledge into disaster management. It suggests a way to mobilise available human and technical resources in order to strengthen a good partnership between local communities and local and national institutions. The paper also analyses the current vulnerability of two communes by correlating hazard risk and loss/damage caused by disasters and the contribution that domestic risk maps in the community can make to reduce this risk. Biodiversity offsets are increasingly being used for securing biodiversity conservation outcomes as part of sustainable economic
development to compensate for the residual unavoidable impacts of projects. Two recent New Zealand examples of biodiversity
offsets are reviewed—while both are positive for biodiversity conservation, the process by which they were developed and approved
was based more on the precautionary principal than on any formal framework. Based on this review and the broader offset literature,
an environmental framework for developing and approving biodiversity offsets, comprising six principles, is outlined: (1)
biodiversity offsets should only be used as part of an hierarchy of actions that first seeks to avoid impacts and then minimizes
the impacts that do occur; (2) a guarantee is provided that the offset proposed will occur; (3) biodiversity offsets are inappropriate
for certain ecosystem (or habitat) types because of their rarity or the presence of threatened species within them; (4) offsets
most often involve the creation of new habitat, but can include protection of existing habitat where there is currently no
protection; (5) a clear currency is required that allows transparent quantification of values to be lost and gained in order
to ensure ecological equivalency between cleared and offset areas; (6) offsets must take into account both the uncertainty
involved in obtaining the desired outcome for the offset area and the time-lag that is involved in reaching that point. 相似文献
