Soluble salts, nutrients, and pathogenic bacteria in feedlot-pen runoff have the potential to cause pollution of the environment. A 2-yr study (1998-1999) was conducted at a beef cattle (Bos taurus) feedlot in southern Alberta, Canada, to determine the effect of bedding material [barley (Hordeum vulgare L.) straw versus wood chips] and within-pen location on the chemical and bacterial properties of pen-floor runoff. Runoff was generated with a portable rainfall simulator and analyzed for chemical content (nitrogen [N], phosphorus [P], soluble salts, electrical conductivity [EC], sodium adsorption ratio [SAR], dissolved oxygen [DO], and pH) and populations of three groups of bacteria (Escherichia coli, total coliforms, total aerobic heterotrophs at 27 degrees C) in 1998 and 1999. Bedding had a significant (P < or = 0.05) effect on NH4-N concentration and load in 1999, SO4 load in 1998, SO4 concentration and load in 1999, and total coliforms in both years; where these three variables were higher in wood than straw pens. Location had a significant effect on EC and concentrations of total Kjeldahl nitrogen (TKN), Na, K, SO4, and Cl in 1998, and total coliforms in both years. These seven variables were higher at the bedding pack than pen floor location, indicating that bedding packs were major reservoirs of TKN, soluble salts, and total coliforms. Significantly higher dissolved reactive phosphorus (DRP), total P, and NH4-N concentrations and loads at the bedding pack location in wood pens in 1998, and a similar trend for TKN concentration in 1999, indicated that this bedding-location treatment was a greater source of nutrients to runoff than the other three bedding-location treatments. Bedding, location, and their interaction may therefore be a potential tool to manage nutrients, soluble salts, and bacteria in feedlot runoff.  相似文献

Fresh beef cattle (Bos taurus) manure has traditionally been applied to cropland in southern Alberta, but there has been an increase in application of composted manure to cropland in this region. However, the quality of runoff under fresh manure (FM) versus composted manure (CM) has not been investigated. Our objective was to compare runoff quality under increasing rates (0, 13, 42, 83 Mg ha(-1) dry wt.) of FM and CM applied for two consecutive years to a clay loam soil cropped to irrigated barley (Hordeum vulgare L.). We determined total phosphorus (TP), particulate phosphorus (PP), dissolved reactive phosphorus (DRP), total nitrogen (TN), NH4-N, and NO3-N concentrations and loads in runoff after one (1999) and two (2000) applications of FM and CM. We found significantly (P < or = 0.05) higher TP, DRP, and NH4-N concentrations, and higher DRP and TN loads under FM than CM after 2 yr of manure application. The TP loads were also higher under FM than CM at the 83 Mg ha(-1) rate in 2000, and DRP loads were higher for FM than CM at this high rate when averaged over both years. Application rate had a significant effect on TP and DRP concentrations in runoff. In addition, the slope values of the regressions between TP and DRP in runoff versus application rate were considerably higher for FM in 2000 than for FM in 1999, and CM in both 1999 and 2000. Significant positive relationships were found for TP and DRP in runoff versus soil Kelowna-extractable P and soil water-extractable P for FM and CM in 2000, indicating that interaction of runoff with the soil controlled the release of P. Total P and DRP were the variables most affected by the treatments. Overall, our study found that application of CM rather than FM to cropland may lower certain forms of P and N in surface runoff, but this is dependent on the interaction with year, application rate, or both.  相似文献

In revegetation trials on disturbed riverbank sites in the Montréal region (Québec, Canada), an analysis of their structure by means of their biotic and abiotic components was undertaken. Vegetation sampling and physical surveys, together with soil analyses of 62 stands, grouped in 20 sectors, were carried out in the summer of 1986. The statistical analyses of biotic and abiotic data were executed in parallel by a hierarchical agglomerative cluster analysis (arithmetic average clustering) and by an ordination in reduced space (principal coordinate analysis). The species colonizing these riverbanks are, for the most part, ruderals from the Montréal region, of which more than 50% are introduced species. The pioneering communities characterizing these artificial habitats have had very little impact on the physical conditions of their environment; therefore, the ordination on the plant communities proves to be strongly correlated to the ordination stemming from the abiotic components. The statistical analysis retains the original sectors, underlining the fact that each human intervention generally results in the creation of a particular ecological situation.  相似文献

The current study determined behavioral and electrophysiological photosensitivities for three species of mesopelagic crustaceans: Pasiphaea multidentata Esmark, 1866 (Decapoda: Pasiphaeidae), Sergestes arcticus Kröyer, 1855 (Decapoda: Sergestidae), and Meganyctiphanes norvegica M. Sars, 1857 (Euphausiacea: Euphausiidae). In addition, in situ quantifications of the species vertical distributions in relation to downwelling irradiances were also determined in two locations in the northwest Atlantic Ocean, Wilkinson Basin (WB) and Oceanographer Canyon (OC). Data are from six 2-week cruises between June and September from 1995 to 2001. P. multidentata and M. norvegica were the most abundant large crustaceans in WB, and S. arcticus and M. norvegica were the most abundant large crustaceans in OC. The behavioral light sensitivity thresholds of P. multidentata and M. norvegica from WB were both 107 photons cm–2 s–1 and those of S. arcticus and M. norvegica from OC were both 108 photons cm–2 s–1. Electrophysiologically, P. multidentata was significantly more sensitive than M. norvegica from either location, S. arcticus was significantly more sensitive than M. norvegica from OC, and M. norvegica from WB was significantly more sensitive than M. norvegica from OC. A correlation was found between electrophysiologically measured photosensitivity and downwelling irradiance, with the most sensitive species, P. multidentata and S. arcticus, associated with the lowest irradiance at daytime depths. The photosensitivities of M. norvegica collected from the clearer waters of OC were significantly lower than those of individuals collected from the more turbid WB waters. These results indicate that downwelling irradiance has a significant impact on interspecies and intraspecies vertical distribution patterns in the mesopelagic realm.Communicated by J.P. We have previously detected chromosome abnormalities in human embryos whilst identifying the sex for preimplantation diagnosis of X-linked disease. In this study we assess the incidence of these abnormalities, both for sex chromosomes and autosomes 1 and 17, using dual fluorescent in situ hybridization (FISH). Sixty-nine normally fertilized embryos of good morphology at the 6–10 cell stage (day 3 post-insemination) were examined. The embryos were spread whole using HCl and Tween 20 to dissolve the cytoplasm. Thirty-four embryos were analysed for the sex chromosomes and 35 for autosomes 1 and 17. All probes were directly labelled with fluorochromes allowing analysis in 2 h. Control lymphocytes demonstrated that the probes were of high specificity. For the sex chromosomes, five embryos were mosaic (15 per cent) with the remaining 29 being uniformly XX or XY. In no case was an XX nucleus found in an otherwise XY embryo, indicating that even though mosaicism for the sex chromosomes is present, such abnormalities would not lead to a misdiagnosis of sex. For the autosomes, 16 embryos were abnormal (46 per cent); one embryo was triploid, one was monosomic for chromosome 1, and ten others were diploid mosaics (three diploid/aneuploid, three diploid/polyploid, and four diploid/haploid). A further four embryos had variable chromosome numbers in the majority of nuclei which appeared to be the result of uncontrolled mitotic division. The presence of haploidy or double monosomy, which occurred in 15 per cent of nuclei, has important implications for the diagnosis of trisomies and dominant disorders.  相似文献

In freshwater ecosystems, a large number of chemical substances are able to disturb homeostasis of fish by modulating one or more physiological functions including the immune system. The aim of this study was to assess multi-biomarker responses including immunotoxicity induced by urban and agricultural pressure in European bullheads living in a small French river basin. For this purpose, a set of biochemical, immunological, physiological and histological parameters was measured in wild bullheads from five locations characterized by various environmental pressures. Moreover, to address effects of physiological status and contamination level variation on biomarker responses, fish were sampled during three periods (April, July and October). Results revealed a clear impact of environmental pressure on fish health and particularly on immunological status. An increase of EROD activity was recorded between upstream and downstream sites. Upstream sites were also characterized by neurotoxicological effects. Fish exhibited upstream/downstream variations of immunological status but strong differences were observed according to sampling season. Conversely, regarding biochemical and immunological effects, no significant response of physiological indexes was recorded related to environmental pressures. According to these results, the European bullhead appears as a valuable model fish species to assess adverse effects in wildlife due to urban and agricultural pressures.  相似文献

The present research focused on using spatial analysis to determine relationships among land ownership property sizes and landscape structure, with a focus on conservation management implications. Indices and metrics of ownership property sizes and landscape structure were calculated for 20 km buffer areas around 31 North American Breeding Bird Survey transects, 12 located within the Edwards Plateau ecoregion and 18 in contiguous ecoregions. The number of bird species observed at each transect provided a measure of avian species richness associated with land cover classes for each respective transect (González in Urban influence on diversity of avifauna in the Edwards Plateau of Texas: effect of property sizes on rural landscape structure, Texas A&M University, 2005). Spatial correlations were calculated between each pair of the landscape indices. Spatial analysis identified a “threshold of habitat fragmentation” for the 500 acre (ac) ownership property size. Significant spatial correlations among variables showed that property sizes lower than 500 ac produced habitat fragmentation represented by a decrease in mean patch size (MN) and proximity among habitat patches (Index PROX). Spatial analysis also made possible the prioritization of ecological sub-regions of the Edwards Plateau for conservation or restoration. The Live Oak-Mesquite Savannah showed the highest average ownership property size (7305 ac) and the highest values of patch richness. Based on the results, management in the Live Oak-Mesquite Savannah sub-region should focus on the conservation of land mosaic diversity to assure native avian species turnover (Whittaker 1972). In Balcones Canyon Lands, 64 % of land was covered by farms smaller than 500 ac and the overall average ownership property size was above the threshold of fragmentation (1440 ac), implying that management policies there should focus both on habitat conservation and on restoration. In contrast, 71 % of land in the Lampasas Cut Plains was covered by farms smaller than 500 ac, and average ownership property size was very close to the fragmentation threshold (625 ac). Consequently, the results indicate that management in the Lampasas Cut Plains sub-region should focus on habitat restoration (e.g., corridors that connect isolated habitat patches). In general, the threshold of ownership property size, 500 ac, is important for conservation planning because below that threshold of property size, habitat patch size begins to decrease and the distance between equivalent patches of habitat increases. Isolated patches act as islands within a sea of less suitable habitat which produce negative effects on biodiversity. Identifying the spatial characteristics indicative of habitat fragmentation, or the likelihood thereof, is an important issue for conservation planning in places with urban sprawl influence.  相似文献

In recent decades, many toxicological tests based on in vivo or in vitro models, mainly from mammalian (rat–mouse) and fish species, were used to assess the risks raised by contact or ingestion of molecules of pharmaceutical, agricultural, or natural origin. But no, or few, in vitro tests using other non-mammalian models such as bird have been explored despite their advantages: the embryonic gonads of birds have a high plasticity of development sensitive to estrogen, and sperm production is nearly two times faster than in rodents. Hence, we have established an in vitro culture of germ cells and somatic cells from chicken post-natal testis, and we have evaluated the sensitivity against the endocrine disruptor compound mono-(2-ethylhexyl) phthalate (MEHP) in comparison to previous studies using rodent and human models. After 96 h of exposure in presence of 10 μM MEHP, chicken seminiferous tubules cultures present a structural alteration, a reduction in cell proliferation and in germ cells population. Apoptosis of germ and somatic cells increases in presence of 1 μM MEHP. Furthermore, MEHP does not affect inhibin B and lactate production by Sertoli cells. These results are in accordance with previous studies using rat, mice, or human culture of testicular cells and in similar range of exposures or even better sensitivity for some “end-points” (biological parameters). In conclusion, the establishment of this postnatal testicular cells culture could be considered as an alternative method to in vivo experiments frequently used for evaluating the impact on the terrestrial wildlife species. This method could be also complementary to mammal model due to the limiting number of animals used and its elevated sensitivity.  相似文献

An oxidative metabolism of Collembolan Proisotoma minuta was determined with a model compound of aldrin and dieldrin in this paper. The seven-day LD(50) values for aldrin, dieldrin, and piperonyl butoxide in salt solution were 0.496, 0.367, and 8.346 mg L(-1), respectively. When P. minuta were exposed to aldrin, dieldrin was the sole metabolite. The conversion of aldrin to dieldrin was known to be catalyzed by P450 monooxygenases system. It has been shown that the synergist piperonyl butoxide inhibited the metabolism of aldrin in P. minuta.  相似文献

In addition to obviating the use of synthetic agrochemicals and emphasizing farming in accordance with agro-ecological guidelines, organic farming acknowledges the integrity of plants as an essential element of its natural approaches to crop production. For cultivated plants, integrity refers to their inherent nature, wholeness, completeness, species-specific characteristics, and their being in balance with their (organically farmed) environment, while accomplishing their “natural aim.” We argue that this integrity of plants has ethical value, distinguishing integrity of life, plant-typic integrity, genotypic integrity, and phenotypic integrity. We have developed qualitative criteria to ethically evaluate existing practices and have applied these criteria to assess whether current plant breeding and propagation techniques violate the integrity of crop plants. This process has resulted in a design of a holistic, scientific approach of organic plant breeding and seed production. Our evaluation has met considerable criticism from mainstream (crop) scientists. We respond to the following questions: (1). Can ethics be incorporated into objective crop sciences? (2). What is the nature of the intrinsic value of plants in organic farming? We argue that criteria to take integrity into account can only be assessed from a holistic perspective and we show that a holistic approach is needed to design such ethical notions in a consistent way. The ethical notions have been further elaborated by formulating human responsibility and respect towards crop plants. Responsibility and respect can only be shown by providing crop plants the right to be nurtured and to express natural behavior at all levels of integrity.  相似文献
