The relationship between the expression of extracellular enzymatic system and a metal stress is scarce in fungi, hence limiting the possible use of secretion profiles as tools for metal ecotoxicity assessment. In the present study, we investigated the effect of Zn, Cu, Pb and Cd, tested alone or in equimolar cocktail, on the secretion profiles at enzymatic and protein levels in Trametesversicolor. For that purpose, extracellular hydrolases (acid phosphatase, β-glucosidase, β-galactosidase and N-acetyl-β-glucosaminidase) and ligninolytic oxidases (laccase, Mn-peroxidase) were monitored in liquid cultures. Fungal secretome was analyzed by electrophoresis and laccase secretion was characterized by western-blot and mass spectrometry analyses. Our results showed that all hydrolase activities were inhibited by the metals tested alone or in cocktail, whereas oxidase activities were specifically stimulated by Cu, Cd and metal cocktail. At protein level, metal exposure modified the electrophoretic profiles of fungal secretome and affected the diversity of secreted proteins. Two laccase isoenzymes, LacA and LacB, identified by mass spectrometry were differentially glycosylated according to the metal exposure. The amount of secreted LacA and LacB was strongly correlated with the stimulation of laccase activity by Cu, Cd and metal cocktail. More than 30 years after it was first proposed as a biodiversity conservation strategy, rewilding remains a controversial concept. There is currently little agreement about what the goals of rewilding are, and how these are best achieved, limiting the utility of rewilding in mainstream conservation. Achieving consensus about rewilding requires agreeing about what “wild” means, but many different definitions exist, reflecting the diversity of values in conservation. There are three key debates that must be addressed to find a consensual definition of “wild”: (1) to which extent can people and “wild” nature co-exist?; (2) how much space does “wild” nature need? and (3) what kinds of “wild” nature do we value? This work summarizes the results of a study of atmospheric wet and dry deposition fluxes of Deisopropyl-atrazine (DEA), Desethyl-atrazine (DET), Atrazine, Terbuthylazine, Alachlor, Metolachlor, Diflufenican, Fenoxaprop-p-ethyl, Iprodione, Isoproturon and Cymoxanil pesticides conducted in Strasbourg, France, from August 2000 through August 2001. The primary objective of this work was to calculate the total atmospheric pesticide deposition fluxes induced by atmospheric particles. To do this, a modified one-dimensional cloud water deposition model was used. All precipitation and deposition samples were collected at an urban forested park environment setting away from any direct point pesticide sources. The obtained deposition fluxes induced by atmospheric particles over a forested area showed that the dry deposition flux strongly contributes to the total deposition flux. The dry particle deposition fluxes are shown to contribute from 4% (DET) to 60% (cymoxanil) to the total deposition flux (wet + dry). 相似文献

The occurrence of magnetotactic bacteria (MTB) on a Tunisian marine coast exposed to heavy metals pollution (Sfax, Gulf of Gabès, Mediterranean Sea) was investigated. The MTB population of this Southern Mediterranean coast was compared to the MTB populations previously investigated on the French Northern Mediterranean coast. A dominant MTB coccus morphotype was observed by microscopy analysis. By pyrosequencing technology, the analysis of the 16S ribosomal RNA (rDNA) revealed as much as 33 operational taxonomic sequence units (OTUs) close to sequences of MTB accessible in the databases. The majority were close to MTB sequences of the “Med group” of α-Proteobacteria. Among them, a dominant OTU_001 (99 % of the MTB sequences) affiliated within the Magnetococcales order was highlighted. Investigating the capacities of this novel bacterium to be used in bioremediation and/or depollution processes could be envisaged. 相似文献

Arsenic (As) is a ubiquitous element. The current WHO guideline for As in drinking water is 10 μg/L. Furthermore, about 130 million people have only access to drinking water containing more than 10 gAs/L. Although numerous studies have shown the related adverse effects of As, sensitive appropriate biomarkers are still required for studies of environmental epidemiology. A review of the literature has shown that various biomarkers are used for such research. Their limits and advantages are highlighted in this paper: (i) the detection of As or its derivatives in the blood is an indication of the dose ingested but it is not evidence of chronic intoxication. (ii) The detection of As in urine is an indispensible procedure because it is a good marker for internal dose. It has been demonstrated to correlate well for a number of chronic effects related to As levels in drinking water. However confounding factors must be taken into account to avoid misinterpretation and this may require As speciation. (iii) As in the hair and nails reflects the level of long term exposure but it is difficult to relate the level with the dose ingested. (iv) Some studies showed a correlation between urinary As and urinary and blood porphyrins. However, it is difficult to use only porphyrins as a biomarker in a population survey carried out without doing further studies. (v) Genotoxic effects are based on the characterization of these potential effects. Most studies have detected increases in DNA damage, sister chromatid exchange, micronuclei or chromosomal aberrations in populations exposed to As in drinking water. Micronuclei assay is the technique of choice to follow these populations, because it is sensitive and easy to use.To conclude, whatever epidemiological studies are, the urinary and toenail biomarkers are useful to provide indications of internal dose. Among the herbicides used in vineyards, the pre-emergence soil-applied flumioxazin (fmx) is a recently used molecule that inhibits chlorophyll biosynthesis in weed species. The aim of this work is to further characterize the effects of fmx on the non-target grapevine (Vitis vinifera L. cv. Chardonnay) using cutting as a model. Several photosynthesis parameters were estimated during 25 days after treatment with various fmx concentrations (from 0.5mM to 50mM). Measuring chlorophyll fluorescence it appeared that fmx or a by-product penetrated the plant throughout roots and spread throughout vessels. Besides the initial target, protox, fmx affected other functions related to photosynthesis. Fmx induced a simultaneous drop of both P(n), g(s) and T. Fmx caused stomatal closure, which partially explains the decrease of the net photosynthesis. The decline in F(v)/F(m) indicates that the photochemistry of PSII and its ability to reduce the primary acceptor Q(A) are also affected by fmx in grapevine. Fmx leads to a decrease in the coefficients of both photochemical and non-photochemical quenching. Simultaneous stomatal closure and decrease in the quantum yield of CO(2) assimilation indicate a change in energy metabolism following fmx stress. After ten days of fmx treatment, analyses of the response of net carbon assimilation in leaves to different intercellular CO(2) concentrations have shown a decrease of the maximum carboxylation velocity of RuBP. Polyhydroxybutyrate-co-hydroxyvalerate microspheres (PHBV-MS) were prepared as a delivery system for the herbicide atrazine
(ATZ). Characterization of the system included investigation of in vitro release properties and genotoxicity. ATZ − PHBV-MS
particle diameters showed a size distribution range of 1–13 μm. Differential scanning calorimetry analyses indicated that
ATZ was associated with the PHBV microparticles. The release profiles showed a different release behavior for the pure herbicide
in solution, as compared with that containing ATZ-loaded PHBV-MS. Korsmeyer–Peppas model analyses showed that atrazine release
from the microparticles occurred by a combination of diffusion through the matrix and partial diffusion through water-filled
pores of the PHBV microparticles. A Lactuca sativa test result showed that the genotoxicity of ATZ-loaded PHBV-MP was decreased in relation to ATZ alone. The results demonstrate

Blood clinical-chemical parameters (BCCPs) are used to investigate physiological consequences attributed to exposure to anthropogenic stressors, such as exposure to Tris(1,3-dichloro-2-propyl) phosphate (TDCIPP) and Dechlorane Plus (DP), which are flame retardants (FRs), on avian health. Japanese quail (Coturnix japonica) were used as a model species to investigate individual and mixture effects of both FRs by injecting different concentrations (5, 50, or 500 ng/µL) of either of these compounds as well as a 1:1 mixture of both into fertilized eggs. To estimate post-hatching effects, 18 BCCPs were evaluated from 101 chicks at day 14. Albumin levels were found to be significantly elevated in the groups exposed to high doses of TDCIPP alone as well as a mixture of TDCIPP and DP. However, during the course of the study, the initial corn-based feed had to be substituted by a fishmeal-based feed. The 8 days consuming the new feed played a significant role on 12 of 18 BCCPs measured. Consequently, it is recommended that dietary habits need to be considered when investigating the physiological impact of contaminants using BCCPs as biomarkers in growing nestlings both in controlled in vivo and field experiments. 相似文献
