Expression of the stress biomarkers 70-kDa heat shock proteins (Hsp70) and manganese superoxide dismutase (MnSOD) was measured as the molecular basis of adaptive response against increased experimental temperatures (32–40 °C for a span of 24–72 h) on the fresh water molluscan species, Bellamya bengalensis (Lamark 1882). The experimental snail specimens were collected during summer and winter seasons from two contrasting wetlands: an ecorestored (free from human interference) site (SI) and other experiencing anthropogenic stresses (SII). The mortality rate of the B. bengalensis and the immunoblotting of MnSOD and Hsp70 of their digestive glands were performed at regular intervals during the period of heat stress. The SI provided a lower stress environment based on physicochemical parameters such as pH, dissolved oxygen (DO), biological oxygen demand (BOD), chemical oxygen demand (COD), and alkalinity for the survival of test species, although both sites experienced mortality due to thermal stresses. The parity in protein expressions displayed a uniform mode of adaptive impact to temperature elevations in both field and laboratory exposure. The Hsp70 expression was minimal at lower thermal stress, but increased with a rise in temperature. It is very likely that higher Hsp70 levels are not directly related to survival or adaptation. In contrast, MnSOD levels appeared to be an indicator of adaptive responses vis-a-vis survival of the animals. So, the expression levels of a universal free radical scavenger like MnSOD are recognized as a potential biomarker in a bioindicator species like Bellamya.  相似文献

Endosulfan, malathion, and phosphamidon are widely used pesticides. Subchronic exposure to these contaminants commonly affects the central nervous system, immune, gastrointestinal, renal, and reproductive system. There effects have been attributed to increased oxidative stress. This study was conducted to examine the role of oxidative stress in genotoxicity following pesticide exposure using peripheral blood mononuclear cells (PBMC) in vitro. Further possible attenuation of genotoxicity was studied using N-acetylcysteine (NAC) and curcumin as known modulators of oxidative stress. Cultured mononuclear cells was isolated from peripheral blood of healthy volunteers, and exposed to varying concentrations of different pesticides: endosulfan, malathion, and phosphamidon for 6, 12, and 24 h. Lipid peroxidation was assessed by cellular malondialdehyde (MDA) level and DNA damage was quantified by measuring 8-hydroxy-2'-deoxyguanosine (8-OH-dG) using ELISA. Both MDA and 8-OH-dG were significantly increased in a dose-dependent manner following treatment with these pesticides. There was a significant decrease in MDA and 8-OH-dG levels in PBMC when co-treated with NAC or/and curcumin as compared to pesticide alone. These results indicate that pesticide-induced oxidative stress is probably responsible for the DNA damage, and NAC or curcumin attenuate this effect by counteracting the oxidative stress.  相似文献

An exploratory study of the area surrounding a historical Pb?CZn mining and smelting area in Zawar, India, detected significant contamination of the terrestrial environment by heavy metals. Soils (n?=?87) were analyzed for pH, EC, total organic matter (TOM), Pb, Zn, Mn, and Cd levels. The statistical analysis indicated that the frequency distribution of the analyzed parameters for these soils was not normal. The median concentrations of metals in surface soils were: Pb 420.21 ?? g/g, Zn 870.25 ?? g/g, Mn 696.70 ?? g/g, and Cd 2.09 ?? g/g. Zn concentrations were significantly correlated with Cd (r?=?0.867), indicating that levels of Cd are dependent on Zn. However, pH, electrical conductivity and total organic matter were not correlated significantly with Cd, Pb, Zn, and Mn. To assess the potential mobility of Cd, Pb, and Zn in soils, single (EDTA) as well as sequential extraction scheme (modified BCR) were applied to representative (n?=?23) soil samples. The amount of Cd, Pb, and Zn extracted by EDTA and their total concentrations showed linear positive correlation, which are statistically significant (r values for Cd, Pb, and Zn being 0.901, 0.971, and 0.795, respectively, and P values being <0.001). The correlation coefficients indicate a strong relation between EDTA-extractable metal and total metal. These results appear to justify the use of ??total?? metal contents as a useful preliminary indicator of areas where the risks of metal excess or deficiency are high. The EDTA extractability was maximum for Cd followed by Pb and Zn in soils from all the locations. As indicated by single extraction, the apparent mobility and potential bioavailability of metals in soils followed the order: Cd ?? Pb >?> Zn. Soil samples were sequentially extracted (modified BCR) so that solid pools of Cd, Zn, and Pb could be partitioned into four operationally defined fractions viz. acid-soluble, reducible, oxidizable, and residual. Cadmium was present appreciably (39.41%) in the acid-soluble fraction and zinc was predominantly associated (32.42%) with residual fraction. Pb (66.86%) and Zn (30.44%) were present mainly in the reducible fraction. Assuming that the mobility and bioavailability are related to solubility of geochemical forms of metals and decrease in the order of extraction, the apparent mobility and potential metal bioavailability for these contaminated soil samples is Cd > Zn > Pb.  相似文献

Natural contamination of drinking water with arsenic results in the exposure of millions of people world-wide to unacceptable levels of this metalloid. This is a serious global health problem because arsenic is a Group 1 (proven) human carcinogen and chronic exposure is known to cause skin, lung, and bladder tumors. Furthermore, arsenic exposure can result in a myriad of other adverse health effects including diseases of the cardiovascular, respiratory, neurological, reproductive, and endocrine systems. In addition to chronic environmental exposure to arsenic, arsenic trioxide is approved for the clinical treatment of acute promyelocytic leukemia, and is in clinical trials for other hematological malignancies as well as solid tumors. Considerable inter-individual variability in susceptibility to arsenic-induced disease and toxicity exists, and the reasons for such differences are incompletely understood. Transport pathways that influence the cellular uptake and export of arsenic contribute to regulating its cellular, tissue, and ultimately body levels. In the current review, membrane proteins (including phosphate transporters, aquaglyceroporin channels, solute carrier proteins, and ATP-binding cassette transporters) shown experimentally to contribute to the passage of inorganic, methylated, and/or glutathionylated arsenic species across cellular membranes are discussed. Furthermore, what is known about arsenic transporters in organs involved in absorption, istribution, and metabolism and how transport pathways contribute to arsenic elimination are described.  相似文献

Objectives: We examined associations among race/ethnicity, socioeconomic factors, and driving status in a nationally representative sample of >26,000 U.S. high school seniors.

Methods: Weighted data from the 2012 and 2013 Monitoring the Future surveys were combined and analyzed. We imputed missing values using fully conditional specification multiple imputation methods. Multivariate logistic regression modeling was conducted to explore associations among race/ethnicity, socioeconomic factors, and driving status, while accounting for selected student behaviors and location. Lastly, odds ratios were converted to prevalence ratios.

Results: 23% of high school seniors did not drive during an average week; 14% of white students were nondrivers compared to 40% of black students. Multivariate analysis revealed that minority students were 1.8 to 2.5 times more likely to be nondrivers than their white counterparts, and students who had no earned income were 2.8 times more likely to be nondrivers than those earning an average of ≥$36 a week. Driving status also varied considerably by student academic performance, number of parents in the household, parental education, census region, and urbanicity.

Conclusions: Our findings suggest that resources—both financial and time—influence when or whether a teen will learn to drive. Many young people from minority or lower socioeconomic families who learn to drive may be doing so after their 18th birthday and therefore would not take advantage of the safety benefits provided by graduated driver licensing. Innovative approaches may be needed to improve safety for these young novice drivers.  相似文献

An assessment of the household wastes as larval habitats of the dengue vectors was made considering Kolkata, India, as geographical area. Wastes of four major categories, namely, earthen, porcelain, plastic and coconut shells were monitored for positive with immature of either Aedes aegypti or Aedes albopictus. Twenty six types of wastes with varying size and shape, resembling containers, were identified that hosted mosquito immature. The number of waste containers positive for Aedes immature varied significantly (P < 0.05) with respect to location, type and month. The relative density of Aedes immature in the waste containers varied significantly (P < 0.05) with the types and months. The significant interaction between the month, waste container types and density of Aedes immature suggest that the household wastes are important contributors to the maintenance of the population of Aedes mosquito in the city. Based on the relative density of mosquito immature in the wastes, cluster analysis allowed segregation and classification of the wastes and their importance as mosquito larval habitats. Apparently, the containers that are most frequently disposed off contributed largely to the sustenance of Aedes mosquito population in the city. This calls for a strict legislation towards disposal as well as enhanced management of the household wastes. A link between the wastes disposed and subsequent conversion to the mosquito larval habitats cautions for continuance of Aedes population and possibility of dengue epidemics if the existing management practices are not improved.  相似文献

Correlation equations relating aqueous solubility to molar volume and aqueous solubility to total molecular surface area were derived by regression analysis for several chemical classes which included both liquids and solids. These were similar to equations relating solubility with total molecular surface area and had similar correlation coefficients. Scirpus littoralis is a wetland plant commonly found in Yamuna flood plains of Delhi, India. The ability of Scirpus littoralis to take up and translocate five metals- Mn, Ni, Cu, Zn and Pb from fly ash dosed and metal spiked soils were studied under waterlogged and field conditions for 90 days. Scirpus littoralis accumulated Mn, Ni, Cu, Zn and Pb upto a maximum of 494.92, 56.37, 144.98, 207.95 and 93.08 ppm dry wt., respectively in below ground organs (BO) in 90 days time. The metal content ratios BO/soil (B/S) were higher than shoot/soil ratios (T/S) for all the metals, the highest being for Ni. Metal ratios BO/water (B/W) were also higher than shoot/water (T/W) ratios but the B/W ratio was maximum for Zn. The changes in nutrient status (N, P) in soil water and plants were also studied at interval of 30 days. The Pearson's correlation between metal uptake and N, P uptake were calculated. All the metals except Ni showed negative correlation with nitrogen but they were all non-significant. However, P uptake showed positive correlations with all the metals and all were significant at 1% confidence limit.  相似文献

A preliminary study of inorganic elements in the river bank soil and plants from the River Yamuna in Delhi has been carried out. Soil at Okhla, near the Water Works plant has a high zinc content (1,215 g g–1 and this is reflected in the high concentration of zinc (2,029 g g–1) in Eichhornia plants growing in the river at this point. Although in general the aluminium content of the soils is low in comparison with the world mean, soil near the effluent from the ash settling basins of the Indraprathsa Power Station contains elevated aluminium.Eichhornia plants growing near this effluent are stunted and unhealthy, and is suggested that, taking into account the high pH, possible causes are toxicity from aluminium or boron.  相似文献
